Biochemical and hormonal parameters of goats kept in a controlled environment consuming water with different levels of salinity

RESUMO O objetivo do trabalho foi avaliar as variáveis bioquímicas e hormonais de caprinos das raças Moxotó e Canindé, submetidos a duas temperaturas (26,0±0,6ºC e 32,0±1,2ºC), termoneutra e acima da zona de conforto térmico, respectivamente), consumindo água com três níveis de salinidade (1,0, 6,0 e 12,0dSm-1), utilizando-se 36 animais (18 de cada raça), com idade média de 5,0±0,6 meses e peso médio de 20,0±2,3kg, alojados em gaiolas metabólicas no interior de uma câmara climática. Os animais foram distribuídos em um delineamento inteiramente ao acaso, com esquema fatorial de 2 x 2 x 3 (2 raças, 2 temperaturas e 3 níveis de salinidade) e três repetições. A glicose e a ureia apresentaram efeito significativo (P<0,05) em função da salinidade da água. Glicose, colesterol, proteína, albumina, globulina, AST e hormônios (T4, T3 e cortisol) variaram conforme as temperaturas (P<0,05). Observou-se efeito significativo do horário sobre as variáveis hormonais (P<0,05). As variáveis bioquímicas e hormonais sofrem alterações em função da temperatura e do turno do dia, de modo que o metabolismo é reduzido em animais sob estresse térmico e acelerado quando os animais encontram-se na zona de conforto térmico.
ABSTRACT The objective of this work was to evaluate the biochemical and hormonal variables of Moxotó and Canindé goats submitted to two temperatures - 26.0±0.6 (thermoneutral) and 32.0±1.2°C (above thermal comfort zone) - and consuming water with three levels of salinity (1.0, 6.0 and 12.0 dSm-1). Thirty-six animals (18 of each breed) were used, with an average age of 5.0±0.6months and an average weight of 20.0±2.3kg, housed in metabolic cages inside a climate chamber. The animals were distributed in a completely randomized design with a 2 × 2 × 3 factorial scheme (2 breeds, 2 temperatures and 3 levels of salinity) and three replications. The glucose and urea had a significant effect (P>0.05) according to water salinity. Glucose, cholesterol, protein, albumin, globulin, aspartate aminotransferase and hormones (T4, T3 and cortisol) varied according to temperature (P<0.05). There was a significant effect of time on hormonal variables (P<0.05). Biochemical and hormonal variables changed according to temperature and day shift, so that metabolism was reduced in the animals under thermal stress and accelerated when animals were in the thermal comfort zone.

Hyptis crenata, commonly known as “salva-do-Marajó”, “hortelã-do-campo”, and “hortelãzinha”, is used in folk medicine in Northeast Brazil as tea or infusion to treat inflammatory diseases. Due to the pharmacological efficacy and the low toxicity of the essential oil of Hyptis crenata (EOHc), we decided to investigate the EOHc antiedematogenic effect in experimental models of inflammation. EOHc was administrated orally at doses of 10-300 mg/kg to male Swiss albino mice. Paw edema was induced by subcutaneous injection in the right hind paw of inflammatory stimuli (carrageenan, dextran, histamine, serotonin, and bradykinin) 60 min after administration of EOHc. EOHc significantly inhibited the induced edema. The inhibitory effect of EOHc on dextran-induced edema extended throughout the experimental time. For the 30, 100, and 300 mg/kg doses of EOHc, the inhibition was of 40.28±1.70, 51.18±2.69, and 59.24±2.13%, respectively. The EOHc inhibitory effect on carrageenan-induced edema started at 10 mg/kg at the second hour (h) and was maintained throughout the observation period. At 30, 100, and 300 mg/kg doses the inhibition started earlier, from 30 min. At the edema peak of 180 min, 56, 76, and 82% inhibition was observed for 30, 100, and 300 mg/kg doses, respectively. Additionally, the effect of EOHc on carrageenan-induced paw edema was influenced by the time of administration. The EOHc also inhibited myeloperoxidase activity. In conclusion, the EOHc showed a potent effect, both preventing and reversing the edema, consistent with its anti-inflammatory use in folk medicine.

RESUMO O presente estudo avaliou a influência do regime hídrico sobre efeitos do propofol em Phrynops geoffroanus, bem como a qualidade da contenção farmacológica produzida. Foram utilizados 10 animais, anestesiados em dois momentos com propofol, na dose de 35mg/kg, por via intracelomática, com intervalo de 15 dias. O grupo 1 fora retirado da água 12 horas antes do procedimento e o grupo 2 permaneceu dentro da água até o momento da indução anestésica. Foram mensuradas frequência cardíaca, pressão arterial sistólica, temperatura, frequência respiratória e saturação de oxi-hemoglobina, e avaliados duração do período de latência, período hábil, período de recuperação e qualidade da contenção farmacológica. Houve diferença significativa entre grupos apenas quanto à frequência cardíaca. No grupo 1, as médias dos períodos de latência, hábil anestésico e de recuperação foram de 16,8±8,4, 86,5±79,4 e 1,5±3,8 minutos, respectivamente. Já no grupo 2, as médias foram de 19,9±9,8, 110,9±104,7 e 28,8±58,2 minutos, respectivamente. Concluiu-se que o regime hídrico de 12 horas não influenciou os parâmetros anestésicos e fisiológicos dos animais e que a qualidade da contenção farmacológica foi considerada boa em ambos os grupos.
ABSTRACT The present study evaluated the influence of the water regime on the effects of propofol on Phrynops geoffroanus, as well as the quality of the pharmacological containment produced. Ten animals, anesthetized at two times with propofol at a dose of 35mg / kg, were used intracelomatically with a 15 day interval. Group 1 was withdrawn from the water 12 hours prior to the procedure and Group 2 remained in the water until the time of anesthetic induction. Heart rate, systolic blood pressure, temperature, respiratory rate and oxyhemoglobin saturation were measured and the duration of the latency period, skill period, recovery period and quality of pharmacological containment were measured. There was a significant difference between groups only regarding heart rate. In Group 1, the means of the latency, skillful anesthesia and recovery periods were 16.8±8.4, 86.5±79.4 and 1.5±3.8 minutes, respectively. In Group 2, the mean values were 19.9±9.8, 110.9±104.7 and 28.8±58.2 minutes, respectively. It was concluded that the 12-hour water regime did not influence the anesthetic and physiological parameters of the animals, and the quality of the pharmacological restraint was considered good in both groups.

Abstract: This study evaluated the antibacterial, antifungal, and antioxidant effect of 7-hydroxy-4’,6-dimethoxy-isoflavone and essential oil of Myroxylon peruiferum. The compound was isolated and its structure elucidated by NMR. The chemical composition of essential oil determined by GC-MS analysis. To evaluation of antimicrobial activity, the Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC) and Minimum Fungicidal Concentration (MFC) were performed. In addition to analysis of antioxidant activity, DPPH radical scavenging tests, iron chelating assay (FIC), antioxidant reducing power assay (FRAP) and β-carotene bleaching assay (BCB) were performed. For the essential oil were identified 24 organized compounds having as main constituents; Germacrene D (17.2%), α-pinene (14.8%) and E-caryophyllene (10.8%). The results showed that isoflavone (2000 to 156 μg/mL) and essential oil (5.0 to 1.25%) present antibacterial and antifungal activity against Gram-positive bacteria and filamentous fungi. The isoflavone and the essential oil also presented antioxidant activity in all the tests, mainly on inhibition of the oxidation of β-carotene test concentrations ranging from 60 to 100%. In conclusion, isoflavone and essential oil from M. peruiferum present an antimicrobial alternative against Gram-positive bacteria, especially of the genus Staphylococcus and dermatophyte fungi of the genus Trichophyton, as well as a natural compound antioxidant.

RESUMO: A mastite é um problema sanitário importante em ovelhas da raça Santa Inês, ocasionando prejuízo ao produtor em virtude do descarte de matrizes e da queda no ganho de peso dos cordeiros. O objetivo deste trabalho foi avaliar a saúde da glândula mamária de ovelhas da raça Santa Inês na secagem e no puerpério e pesquisar a eficácia da terapia intramamária com gentamicina na secagem. Sessenta e quatro ovelhas foram divididas em grupos controle (GC) e tratamento (GT), cada um contendo 32 animais, e a saúde da glândula mamária avaliada na secagem e no puerpério. As ovelhas do GT receberam 250mg de gentamicina (Gentocin® Mastite Vaca Seca/ Schering-Plough Veterinária, produto indicado pela empresa para utilização em vacas de leite) em cada metade mamária. Para o diagnóstico, foram realizados exame físico da glândula mamária, California Mastitis Test, contagem de células somáticas e cultura do leite. No GC, das 45 (70,3%) metades mamárias sadias na secagem, 12 desenvolveram mastite subclínica e nove mastite clínica no puerpério. No GT, das 51 (79,7%) metades mamárias sadias na secagem, seis desenvolveram mastite subclínica e 11 mastite clínica no puerpério. Não houve associação entre o tratamento e a ocorrência de mastite no puerpério. Das 19 (29,7%) metades mamárias do GC que apresentaram mastite subclínica na secagem, três permaneceram com mastite subclínica e cinco desenvolveram mastite clínica no puerpério. No GT, das 13 (20,3%) metades mamárias com mastite subclínica na secagem, quatro permaneceram com mastite subclínica e quatro desenvolveram mastite clínica. Não houve associação entre o tratamento e a cura ou persistência da mastite no puerpério. Os principais micro-organismos isolados, na secagem e puerpério, de animais com mastite subclínica ou clínica foram Staphylococcus spp., com predominância de Staphylococcus Coagulase Negativa (SCN). No puerpério, ocorreram 29 casos de mastite clínica, sendo 19 com isolamento, 10 com SCN e seis com S. aureus. Mannheimia haemolytica foi isolado em um caso de mastite subclínica e um caso de mastite clínica. Novos protocolos e diferentes formas de manejo na secagem e no puerpério devem ser pesquisados
ABSTRACT: Mastitis represents an important health problem for Santa Inês breed, causing losses to the producer, due to loss of ewes or the decrease in weight gain of lambs. The aim of this work was to assess the health of the mammary gland of Santa Inês ewes at the drying and puerperium and to investigate the efficacy of a dry-off therapy with gentamicin. In this study, 64 ewes were divided in a control group (GC) and treatment group (GT), and the health of the mammary gland was assessed at the drying and puerperium. The GT ewes received 250mg of gentamicin (Gentocin® DryCow/Schering-Plough, product indicated for use in dairy cows) in each mammary half. For diagnosis, clinical examination, California Mastitis Test, somatic cell count and milk culture was performed. In the GC, of the 45 (70.3%) healthy mammary halves at the drying, 12 developed subclinical mastitis and nine clinical mastitis at the puerperium. In the GT, among 51 (79.7%) healthy mammary halves at the drying, six developed subclinical mastitis and 11 clinical mastitis at the puerperium. No association was observed between treatment and the occurrence of mastitis at puerperium. Of the 19 (29.7%) mammary halves of the GC that presented subclinical mastitis at the drying, three remained with subclinical mastitis and five developed clinical mastitis at the puerperium. In the GT, of the 13 (20.3%) mammary halves that had subclinical mastitis at the drying, four remained with subclinical mastitis and four developed clinical mastitis. No association was observed between treatment and cure or persistence of mastitis at the puerperium. The main microorganisms isolated, at the drying and puerperium, from animals with subclinical or clinical mastitis were Staphylococcus spp., predominantly coagulase negative Staphylococcus (CSN). At the puerperium, 29 cases of clinical mastitis occurred, 19 with isolation, where 10 were CNS and six S. aureus. Mannheimia haemolytica was isolated in one case of subclinical mastitis and other of clinical mastitis. News protocols and different ways of handling at drying and at puerperium must be investigated.

Histophilus somni-associated syndromes in sheep from Southern Brazil

Abstract Histophilus somni is a Gram-negative bacterium that is associated with a disease complex (termed histophilosis) that can produce several clinical syndromes predominantly in cattle, but also in sheep. Histophilosis is well described in North America, Canada, and in some European countries. In Brazil, histophilosis has been described in cattle with respiratory, reproductive, and systemic disease, with only one case described in sheep. This report describes the occurrence of Histophilus somni-associated disease in sheep from Southern Brazil. Eight sheep with different clinical manifestations from five farms were investigated by a combination of pathological and molecular diagnostic methods to identify additional cases of histophilosis in sheep from Brazil. The principal pathological lesions were thrombotic meningoencephalitis, fibrinous bronchopneumonia, pulmonary abscesses, and necrotizing myocarditis. The main clinical syndromes associated with H. somni were thrombotic meningoencephalitis (n = 4), septicemia (n = 4), bronchopneumonia (n = 4), and myocarditis (n = 3). H. somni DNA was amplified from multiple tissues of all sheep with clinical syndromes of histophilosis; sequencing confirmed the PCR results. Further, PCR assays to detect Pasteurella multocida and Mannheimia haemolytica were negative. These findings confirmed the participation of H. somni in the clinical syndromes investigated during this study, and adds to the previous report of histophilosis in sheep from Brazil.

RESUMO: A excreção fracionada de eletrólitos é calculada para verificar a função renal e auxiliar na interpretação de distúrbios eletrolíticos e ácido-base. O deslocamento de abomaso à esquerda é frequente em vacas leiteiras, ocasionado alcalose hipoclorêmica e hipocalêmica. Há pouca informação na literatura sobre excreção fracionada de eletrólitos em vacas com deslocamento de abomaso. Este estudo objetivou mensurar a excreção fracionada de sódio, potássio e cloro e a acidúria paradoxal em vacas leiteiras com deslocamento abomasal. Amostras sanguíneas e urinárias foram coletadas de 30 vacas antes e 24, 48 e 72 horas após operação. As vacas foram divididas em dois grupos (G1: laparoscopia e G2: laparotomia) com 15 animais cada. As concentrações de cloro, sódio, potássio e creatinina foram dosadas no soro e urina. Mensurou-se pH urinário e o hematócrito. A excreção fracionada e diferença de íons fortes urinário [SID]urina foram calculados utilizando fórmulas publicadas. Vacas de ambos os grupos apresentaram alcalose hipocalêmica antes da operação. Não houve alterações significativas na excreção fracionada de sódio, potássio e cloro, no pH urinário e na [SID]urina entre os tratamentos e momentos. Acidúria paradoxal foi observada no G1 antes e 24h após operação. A excreção fracionada e [SID]urina são ferramentas importantes para interpretar a alcalose hipoclorêmica e hipocalêmica em vacas leiteiras com deslocamento de abomaso, como também a acidúria paradoxal e o retorno do fluxo abomasal.
ABSTRACT: The fractional excretion of electrolytes is used to assess renal function and interpret electrolyte and acid-base imbalances. Left displaced abomasum is a common disorder in dairy cows, which causes hypokalemic, hypochloremic metabolic alkalosis. There is limited information on fractional excretion of electrolytes in cows with displaced abomasum. This study aimed to measure the fractional excretion of sodium, potassium, and chloride and paradoxical aciduria in dairy cows with displaced abomasum. Blood and urine samples were collected from 30 dairy cows before and 24, 48, and 72 h after surgery. The cows were divided into two groups (G1: laparoscopy and G2: laparotomy) with 15 cows each. The concentrations of chloride, sodium, potassium, and creatinine were measured in serum and urine. Urinary pH and packed cell volume were measured. Fractional excretion of sodium, potassium, and chloride and urinary strong ion difference [SID]urine were calculated using published formulas. Cows in both groups showed hypokalemic, metabolic alkalosis before surgery; however, hypochloremia was observed only in G2. Potassium concentration significantly increased 24, 48, and 72 h after surgery in G1 and 48 and 72 h after surgery in G2. There were no significant changes in fractional excretion of sodium, chloride, and potassium and urinary pH and [SID]urine between treatments and time points. Paradoxical aciduria was observed before and 24 h following surgery in G1. Fractional excretion and urinary SID are valuable tools to understand hypochloremic, hypokalemic alkalosis in dairy cows with displaced abomasum, as well as paradoxical aciduria and return of abomasal flux.

RESUMO: A urolitíase é uma doença importante de cordeiros confinados. A acidificação da urina, pela ingestão de cloreto de amônio, é o método preventivo mais frequentemente empregado. Devido à falta de informação específica em ovinos, este estudo foi realizado para avaliar as alterações que ocorrem nos eletrólitos urinários de cordeiros, que receberam cloreto de amônio na dieta. Foram utilizados 100 cordeiros, com 3 meses de idade, que foram mantidos em confinamento durante 56 dias, e distribuídos em 3 grupos: G1 (n=40) que receberam 400mg/kg de peso vivo (PV) de cloreto de amônio/dia, durante 21 dias; G2 (n=40) que receberam 400mg/kg de PV de cloreto de amônio/dia durante 42 dias; e G3 (n=20) que não receberam cloreto de amônio. Os cordeiros foram examinados e as amostras de sangue e urina foram colhidas a cada 7 dias: 0 (antes do início da ingestão de cloreto de amônio), 7, 14, 21, 28, 35, e 42 dias. As concentrações séricas e urinárias de sódio (Na+), potássio (K+), cloreto (Cl-), e de creatinina foram mensuradas em todos os momentos de colheita. A excreção fracionada urinária (EFu) de eletrólitos e a diferença de íons fortes (SID) na urina [(Na+ + K+) - Cl] foram calculadas. A EFu de Na+, K+ e Cl- não variou ao longo do tempo em G3, provando que a dieta de confinamento, por si só, não influenciou a excreção urinária destes eletrólitos. A ingestão de cloreto de amônio, pelo grupo G1 e G2, influenciou a EFu sobre o tempo de confinamento. A SID urinária foi mais precisa do que a EFu de Cl- para demonstrar que a concentração de Cl- aumentou na urina, o que destacou a relevância desta variável.
ABSTRACT: Urolithiasis is an important disease of lambs confined. The urine acidification, by ammonium chloride intake, is the preventive method most frequently employed. Due to the lack of specific information in sheep, this study was performed to evaluate the electrolyte changes that occur in the urine of lambs receiving ammonium chloride in the diet. One hundred male lambs, 3 months old, were kept in a feedlot during 56 days, and distributed in 3 groups: G1 (n=40) receiving 400mg/kg BW of ammonium chloride/day during 21 days; G2 (n=40) receiving 400mg/kg BW of ammonium chloride/day during 42 days; and G3 (n=20) that did not receive ammonium chloride. The lambs were examined and blood and urine samples were collected every 7 days: 0 (the beginning of ammonium chloride intake), 7, 14, 21, 28, 35, and 42 days. Serum and urine sodium (Na+), potassium (K+), chloride (Cl-), and creatinine concentrations were measured. The urinary fractional excretion (FE) of electrolytes and the urine strong ion difference [(Na+ + K+) - Cl-] were calculated. FEs of Na+, K+, and Cl- did not vary over time in G3, proving that the feedlot diet, by itself, did not influence the urinary excretion of these electrolytes. The ingestion of ammonium chloride, instead, influenced FEs over the time of feedlot. The urinary SID was more accurate than the FE of Cl- to demonstrate that the concentration of Cl- increased in the urine. It highlights the relevance of this variable.

RESUMO: O objetivo deste trabalho foi avaliar a importância da transfaunação no tratamento e recuperação de cabras e ovelhas com acidose lática ruminal aguda (ALRA) induzida experimentalmente. Seis cabras (41,2±5,6kg) e seis ovelhas (46,8±4,57kg), não gestantes e não lactantes, receberam 15g de sacarose por quilo de peso corporal para indução da enfermidade. A ALRA foi induzida duas vezes em cada animal, com intervalo de 30 dias após recuperação total da primeira indução. Os procedimentos terapêuticos consistiram na remoção do conteúdo ruminal líquido por lavagem e sifonamento com auxílio de sondagem esofágica, e na correção da acidose metabólica com soluções eletrolíticas, contendo lactato ou bicarbonato de sódio, infundidas por via intravenosa. A transfaunação fez parte de apenas um dos tratamentos de cada animal e consistiu na administração por sondagem esofágica de 2L de suco ruminal de um bovino sadio. A recuperação completa foi avaliada por exames físicos e exames do suco ruminal realizados até quatro dias após os procedimentos terapêuticos. A eficácia dos protocolos de tratamento, com ou sem transfaunação, foi comparada. O protocolo de indução foi efetivo em induzir a enfermidade e as cabras e ovelhas apresentaram sinais clínicos de intensidade máxima (apatia, atonia ruminal, distensão abdominal, diarreia de consistência pastosa a líquida e desidratação moderada) 16 horas após a administração intrarruminal de sacarose, sem distinção entre as espécies. Neste momento, as características do suco ruminal mostraram-se semelhantes aos quadros típicos de ALRA. A frequência de movimentos ruminais se normalizou no terceiro dia após os procedimentos terapêuticos, sem diferença entre as espécies, e independente de terem recebido a transfaunação ou não. A transferência de suco ruminal também não acelerou a recuperação do apetite, que foi considerado normal somente no quarto dia após os procedimentos terapêuticos, em ambas as espécies. Em relação ao líquido ruminal, quando receberam a transfaunação, os caprinos e ovinos apresentaram recuperação das características de cor, odor e consistência mais rapidamente do que quando não receberam. A atividade fermentativa da microbiota ruminal não sofreu influência da transfaunação e se normalizou dois e três dias após os procedimentos terapêuticos nas ovelhas e cabras, respectivamente. A transfaunação promoveu o retorno mais rápido da população de protozoários ruminais, que já foram observados 24 horas após os procedimentos terapêuticos, em ambas as espécies. Quando os animais não receberam a transfaunação, o retorno dos protozoários só ocorreu no segundo dia após os procedimentos terapêuticos. A recuperação completa dos animais estudados ocorreu em até quatro dias, independente da realização da transfaunação ou não. Conclui-se que a transferência de suco ruminal não pode ser considerada medida crucial para o tratamento e convalescença de caprinos e ovinos acometidos por de ALRA.
ABSTRACT: The aim of this study was to evaluate the value of transfaunation in the treatment and recovery of goats and sheep with acute rumen lactic acidosis (ARLA) experimentally induced. Six goats (41.2±5.6kg) and six sheep (46.8±4.57kg), non-pregnant and non-lactating, received 15g of sucrose per kilogram of body weight for the disease induction. The ARLA was induced in each animal twice with an interval of 30 days after full recovery of the first induction. Therapeutic procedures consisted in removal of the rumen fluid content by washing and siphoning with an esophageal tube and correction of metabolic acidosis with intravenous electrolyte solutions containing sodium lactate or sodium bicarbonate. The transfaunation was part of only one of each animal treatment and consisted in administration of two liters of rumen fluid colleted from a healthy cow. Complete recovery was assessed by physical examinations and rumen fluid examinations up to four days after therapeutic procedures. The efficacy of treatment protocols, with or without transfaunation was compared. The protocol used was effective in induce the disease and goats and sheep showed clinical signs of maximum intensity (apathy, rumen stasis, abdominal distension, diarrhea and moderate dehydration) 16 hours after the intraruminal administration of sucrose. At this moment, the characteristics of ruminal fluid were similar to the typical ARLA. The frequency of ruminal movements became normal on the third day after therapeutic procedures, with no difference between species, and regardless transfaunation. The ruminal fluid transfer did not accelerate the recovery of appetite, considered normal only on the fourth day after treatment in both species. When transfauntation was received, goats and sheep showed recovery of characteristics of color, odor and consistency faster than when they did not receive. The fermentative activity of the rumen microbiota was not impacted by transfaunation and normalized two and three days after therapeutic procedures in sheep and goats, respectively. The transfaunation promoted faster return of the protozoa population, which was observed 24 hours after therapeutic procedures in both species. When the animals did not receive transfaunation, the protozoa returns occurred only on the second day after therapeutic procedures. Full recovery of the animals occurred within four days, regardless of transfaunation. In conclusion, the ruminal fluid transfer can not be considered crucial for the treatment and convalescence of goats and sheep affected by ARLA.

RESUMO Os manguezais representam zonas de elevada produtividade biológica sendo uma das regiões mais ricas em substâncias bioativas utilizadas na produção de medicamentos. Conocarpus erectus (Combretaceae) conhecida como mangue botão é uma das espécies encontrada nos manguezais e é utilizada na medicina popular no tratamento de anemia, catarro, conjuntivite, diabetes, diarréia, febre, gonorréia, dor de cabeça, hemorragia, orquite, brotoeja, inchaços e sífilis. O presente estudo objetivou investigar a toxicidade aguda de extrato aquoso de folhas de C. erectus em camundongos albinos suíços. O material vegetal foi coletado no manguezal de Vila Velha, localizado em Itamaracá (PE). O material foi submetido a um screening fitoquímico onde foram usados protocolos extrativos de identificação de moléculas majoritárias presentes nas folhas. A avaliação da toxicidade aguda do extrato aquoso de C. erectus seguiu o modelo da Toxicidade Aguda de Classe baseado no Guia da OECD 423, 2001. As moléculas majoritárias identificadas foram: flavonóides, taninos e saponinas. A DL50 foi estimada em 2.000 mg/Kg p.c. Portanto, o extrato apresentou baixa toxicidade aguda classificada na categoria 5.
ABSTRACT Mangroves represent areas of high biological productivity and it is a region rich in bioactive substances used in medicine production. Conocarpus erectus (Combretaceae) known as button mangrove is one of the species found in mangroves and it is used in folk medicine in the treatment of anemia, catarrh, conjunctivitis, diabetes, diarrhea, fever, gonorrhea, headache, hemorrhage, orchitis, rash, bumps and syphilis. The present study aimed to investigate the acute toxicity of aqueous extract of leaves of C. erectus in Swiss albino mice. The plant material was collected in Vila Velha mangroves, located in Itamaracá (PE). The material was subjected to a phytochemical screening where extractive protocols to identify majority molecules present in leaves were used. The evaluation of acute toxicity of aqueous extract of C. erectus followed the model of Acute Toxicity Class based on OECD 423 Guideline, 2001. The majority molecules were identified: flavonoids, tannins and saponins. The LD50 was estimated at 2,000 mg/kg bw. Therefore, the aqueous extract showed low acute toxicity classified in category 5.

Azole resistance in Candida spp. isolated from Catú Lake, Ceará, Brazil: an efflux-pump-mediated mechanism

Abstract Since, there is no study reporting the mechanism of azole resistance among yeasts isolated from aquatic environments; the present study aims to investigate the occurrence of antifungal resistance among yeasts isolated from an aquatic environment, and assess the efflux-pump activity of the azole-resistant strains to better understand the mechanism of resistance for this group of drugs. For this purpose, monthly water and sediment samples were collected from Catú Lake, Ceará, Brazil, from March 2011 to February 2012. The obtained yeasts were identified based on morphological and biochemical characteristics. Of the 46 isolates, 37 were Candida spp., 4 were Trichosporon asahii, 3 were Cryptococcus laurentii, 1 Rhodotorula mucilaginosa, and 1 was Kodamaea ohmeri. These isolates were subjected to broth microdilution assay with amphotericin B, itraconazole, and fluconazole, according to the methodology standardized by the Clinical and Laboratory Standards Institute (CLSI). The minimum inhibitory concentrations (MICs) of amphotericin B, itraconazole, and fluconazole were 0.03125–2 µg/mL, 0.0625 to ≥16 µg/mL, and 0.5 to ≥64 µg/mL, respectively, and 13 resistant azole-resistant Candida isolates were detected. A reduction in the azole MICs leading to the phenotypical reversal of the azole resistance was observed upon addition of efflux-pump inhibitors. These findings suggest that the azole resistance among environmental Candida spp. is most likely associated with the overexpression of efflux-pumps.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Avaliou-se a eficácia do protocolo radioterápico hipofracionado no tratamento de felinos portadores de carcinoma epidermóide facial. Um protocolo hipofracionado de radioterapia foi aplicado em cinco gatos portadores de um ou mais carcinomas epidermóides faciais, em um total de 10 lesões neoplásicas, confirmadas por meio de análise histológica. Duas lesões foram classificadas como T1, quatro como T2, duas como T3 e duas como T4. Os animais foram submetidos a quatro frações radioterápicas de 7,6 a 10gy, com intervalo de uma semana entre elas, utilizando-se um acelerador linear com feixe de elétrons. O acompanhamento dos animais foi realizado semanalmente durante o tratamento e aos 30 e 60 dias após o término da radioterapia. Neste estudo, 40% das lesões resultaram em remissão completa, 40% em remissão parcial e 20% não apresentaram resposta ao tratamento. A taxa de resposta encontrada nessa pesquisa foi baixa, porém o protocolo de hipofracionamento radioterápico foi seguro para gatos com carcinoma epidermóide facial e resultou em efeitos colaterais leves/moderados
The efficacy of hypofractionated radiation protocol for feline facial squamous cell carcinoma was evaluated. Hypofractionated radiation therapy was applied to five cats showing single or multiple facial squamous cell carcinomas, in a total of ten histologically confirmed neoplastic lesions. Of the lesions, two were staged as T1, four as T2, two as T3, and two as T4. The animals were submitted to four radiation fractions from 7.6 to 10 grays each, with one week intervals. The equipment was a linear accelerator with electrons beam. The cats were evaluated weekly during the treatment and 30 and 60 days after the end of the radiation therapy. In this study, 40% of the lesions had complete remission, 40% partial remission, and 20% did not respond to the treatment. Response rates were lower as compared to other protocols previously used. However, hypofractionated radiation protocol was considered safe for feline facial squamous cell carcinoma.

O objetivo deste estudo foi avaliar o efeito da sibutramina (S) 15 mg/dia, fluoxetina (F) 60 mg/dia, e metformina (M) 1.700 mg/dia, associadas a uma dieta de 1.500 kcal/dia, na redução de parâmetros antropométricos e metabólicos. S (n= 8), F (n= 9) e M (n= 8) foram comparadas ao placebo (n= 10) em 35 pacientes obesos durante 90 dias de tratamento. As reações adversas também foram avaliadas durante o tratamento. O grupo F demonstrou uma redução no IMC (11,0%), peso (10,0%), circunferência abdominal (11,0%) e % de tecido adiposo (12,8). Também foram observados um aumento nos níveis de HDL-colesterol (25,8%) e uma redução nos níveis de triglicérides (28,3%), no grupo F. O grupo S apresentou uma redução de 7,91% na circunferência abdominal e de 9,65 na % de tecido adiposo. Já o grupo M apresentou reduções no IMC (4,03%), circunferência abdominal (6,92%), HOMA (23,5%) e pressão arterial (6,08% na sistólica, 2,08% na diastólica). Os três fármacos analisados foram bem tolerados durante o tratamento. Concluímos que a F e a S demonstraram maior eficácia na redução dos parâmetros antropométricos e metabólicos dos pacientes obesos quando comparadas à M, entretanto todas podem ser prescritas para essa finalidade, desde que sejam consideradas as características individuais dos pacientes.
The aim of this study is to assess the effects of sibutramine (S) 15 mg/day, fluoxetine (F) 60 mg/day, and metformin (M) 1,700 mg/day, as an adjunct therapy to a 1,500 kcal/day diet, in reducing anthropometric and metabolic parameters. S (n= 8), F (n= 9), and M (n= 8) were compared to placebo (n= 10) in 35 obese patients in a 90-day trial. Side effects were also studied during the treatment. The data demonstrated that F therapy resulted in a greater average reduction in BMI (11.0%), weight (10.0%), abdominal circumference (11.0%) and %fatty-tissue (12.8). An elevation in HDL-cholesterol (25.8%) and a reduction in average triglyceride levels (28.3%) were also shown. S presented a 7.91% reduction in the abdominal circumference and a 9.65 reduction in %fatty-tissue was also found. M group presented reductions in BMI (4.03%), waist circumference (6.92%), HOMA (23.5%) and blood pressure (6.08% in systolic and 2.08% in diastolic). In general, the three drugs can be considered well tolerated. We concluded that F and S demonstrated a greater mean reduction in anthropometric and metabolic parameters when compared to M, however all of them are useful for that purpose, when the subjects’ characteristics are considered.
