RESUMO: Na modelagem de curvas de crescimento deve-se considerar que dados longitudinais podem apresentar autocorrelação residual, sendo que, se tal característica não é considerada, os resultados e inferências podem ser comprometidos. A abordagem bayesiana, que considera informações à priori sobre o fenômeno em estudo tem se mostrado eficiente na estimação de parâmetros. No entanto, como geralmente não é possível obter as distribuições marginais de forma analítica, faz-se necessário a utilização de algum método, como o método de reamostragem ponderada, para gerar amostras dessas distribuições e assim obter uma aproximação para as mesmas. Dentre as vantagens desse método, destaca-se a geração de amostras independentes e o fato de não ser necessário avaliar convergência. Diante desse contexto, o objetivo deste trabalho foi apresentar a modelagem não linear bayesiana do crescimento em altura de plantas do cafeeiro, irrigadas e não irrigadas (NI), considerando a autocorrelação residual e os modelos não lineares Logístico, Brody, von Bertalanffy e Richards. Em vista dos resultados, verificou-se que, para as plantas NI, o DIC e CPOc, indicaram que, dentre os modelos avaliados, o modelo Logístico é o que melhor descreve o crescimento em altura do cafeeiro ao longo do tempo. E, para as plantas irrigadas, esses mesmos critérios indicaram o modelo Brody. Assim, o crescimento da planta do cafeeiro não irrigado e irrigado seguiram padrões de crescimento distintos, a altura do cafeeiro não irrigado apresentou crescimento sigmoidal com taxa máxima de crescimento aos 726 dias após o plantio, já o cafeeiro irrigado inicia seu desenvolvimento com altas taxas de crescimento que vão diminuindo aos poucos com o tempo.

ABSTRACT: When modeling growth curves, it should be considered that longitudinal data may show residual autocorrelation, and, if this characteristic is not considered, the results and inferences may be compromised. The Bayesian approach, which considers priori information about studied phenomenon has been shown to be efficient in estimating parameters. However, as it is generally not possible to obtain marginal distributions analytically, it is necessary to use some method, such as the weighted resampling method, to generate samples of these distributions and thus obtain an approximation. Among the advantages of this method, stand out the generation of independent samples and the fact that it is not necessary to evaluate convergence. In this context, the objective of this work research was: to present the Bayesian nonlinear modeling of the coffee tree height growth, irrigated and non-irrigated (NI), considering the residual autocorrelation and the nonlinear Logistic, Brody, von Bertalanffy and Richard models. Among the results, it was found that, for NI plants, the Deviance Information Criterion (DIC) and the Criterion of density Predictive Ordered (CPO), indicated that, among the evaluated models, the Logistic model is the one that best describes the height growth of the coffee tree over time. For irrigated plants, these same criteria indicated the Brody model. Thus, the growth of the non-irrigated and irrigated coffee tree followed different growth patterns, the height of the non-irrigated coffee tree showed sigmoidal growth with maximum growth rate at 726 days after planting and the irrigated coffee tree starts its development with high growth rates that gradually decrease over time.

Abstract Background: Cryptoccocal meningitis continues to present high incidence among AIDS patients. The treatment of choice is the synergistic combination of flucytosine (5-FC) with amphotericin B deoxycholate (AmBd) or its lipid formulations. However, 5-FC is unavailable in many countries and AmB demands hospitalization. The combination of AmB with the fungistatic fluconazole (FLC) or the use of high FLC daily doses alone became the choice. Nonetheless, sterilization of cerebrospinal fluid is delayed with FLC monotherapy, mainly with high fungal burden. These findings suggest the search for new antifungal compounds, such as liriodenine. Methods: Liriodenine antifungal activity was evaluated by three procedures: determining the minimum inhibitory concentration (MIC) on 30 strains of the Cryptococcus neoformans (C. neoformans) complex and 30 of the Cryptococcus gattii (C. gattii) complex, using EUCAST methodology and amphotericin B deoxycholate as control; performing the time-kill methodology in two strains of the C. neoformans complex and one of the C. gattii complex; and injury to cryptococcal cells, evaluated by transmission electron microscopy (TEM). Liriodenine absorption and safety at 0.75 and 1.50 mg.kg-1 doses were evaluated in BALB/c mice. Results: Liriodenine MICs ranged from 3.9 to 62.5 μg.mL-1 for both species complexes, with no differences between them. Time-kill methodology confirmed its concentration-dependent fungicidal effect, killing all the strains below the limit of detection (33 CFU.mL-1) at the highest liriodenine concentration (32-fold MIC), with predominant activity during the first 48 hours. Liriodenine induced severe Cryptococcus alterations - cytoplasm with intense rarefaction and/or degradation, injury of organelles, and presence of vacuoles. Liriodenine was better absorbed at lower doses, with no histopathological alterations on the digestive tract. Conclusion: The fungicidal activity confirmed by time-kill methodology, the intense Cryptococcus injury observed by TEM, the absorption after gavage administration, and the safety at the tested doses indicate that the liriodenine molecule is a promising drug lead for development of anticryptococcal agents.

Resumo: O objetivo desde trabalho foi avaliar como a parametrização e a aplicação de diferentes valores alométricos afetam a obtenção do ajuste mais adequado do modelo de von Bertalanffy, na descrição da curva de crescimento de mamíferos produtores de carne (bovinos, porcos, coelhos e ovelhas). Entre os modelos não lineares, o de von Bertalanffy tem sido frequentemente aplicado em diversas áreas, com diferentes parametrizações. Em geral, esse modelo tem sido utilizado com um valor alométrico de m = 2/3; no entanto, para mamíferos, acredita-se que o valor possa ser m = 3/4. Os dados analisados quanto à massa dos mamíferos de corte de acordo com a sua idade foram obtidos de instituições de pesquisa e da literatura. Os resultados mostraram que o modelo de von Bertalanffy, com o valor alométrico de m = 3/4 e com a parametrização utilizada, forneceu melhores ajustes aos avaliadores de qualidade. Além disso, o modelo suavizou a superestimativa do parâmetro a, o que possibilitou interpretação direta ao parâmetro b, com menores valores das medidas de curvatura, principalmente as paramétricas, e forneceu ajustes mais confiáveis. O modelo de von Bertalanffy pode ser utilizado na descrição da curva de crescimento de mamíferos produtores de carne.

Abstract: The objective of this work was to evaluate how the parameterization and the application of different allometric values affect the obtention of the most adequate fit of von Bertalanffy’s model, in the description of the growth curve of meat-producing mammals (bovine, pigs, rabbits, and sheep). Among the nonlinear models, von Bertalanffy’s has been very often applied in several areas, with different parameterizations. This model has been commonly used with an allometric value of m = 2/3; however, for mammals, it is believed that this value can be m = 3/4. The analyzed data referring to the mass of meat-producing mammals according to their age were obtained from research institutions and from the literature. The results showed that von Bertalanffy’s model, with the allometric value of m = 3/4 and the used parameterization, provided better adjustments to quality evaluators. Besides, the model softened the overestimation of parameter a, giving a direct interpretation of parameter b, with the lowest values for curvature measurements, mainly for the parametric ones, and provided more reliable adjustments. Von Bertalanffy’s model can be used in the description of the growth curves of meat-producing mammals.

RESUMO: O objetivo deste trabalho foi verificar se o padrão de crescimento do fruto do cafeeiro, considerando acúmulo de massa fresca em função do tempo, é realmente duplo sigmoidal e selecionar o modelo não linear mais indicado para descrever tal comportamento. Os dados utilizados são quatorze observações longitudinais de massa fresca média de frutos do cafeeiro obtidos em um experimento com a cultivar Obatã IAC 1669-20. Foram comparados os ajustes fornecidos pelos modelos Logístico e Gompertz em suas versões simples e duplo. A estimação dos parâmetros foi feita pelo método dos mínimos quadrados utilizando o algoritmo de Gauss-Newton implementado na função nls do software R. Pode-se concluir que o padrão de crescimento do fruto do cafeeiro, em acúmulo de massa fresca, é duplo sigmoidal. Os modelos duplo Gompertz e duplo Logístico se mostraram adequados para descrever tal curva de crescimento, com uma superioridade do modelo duplo Logístico.

ABSTRACT: This study aimed to verify if the growth pattern of coffee berries, considering fresh mass accumulation over time, is double sigmoid and to select the most suitable nonlinear model to describe such behavior. Data used consisted of fourteen longitudinal observations of average fresh mass of coffee berries obtained in an experiment with the cultivar Obatã IAC 1669-20. The fits provided by the Logistic and Gompertz models were compared in their single and double versions. Parameters were estimated using the least squares method using the Gauss-Newton algorithm implemented in the nls function of the R software. It can be concluded that the growth pattern of the coffee fruit, in fresh mass accumulation, is double sigmoid. The double Gompertz and double Logistic models were adequate to describe such a growth curve, with a superiority of the double Logistic model.
