RESUMO A síndrome de granuloma leproide canino (SGLC), também conhecida como lepra canina, é uma doença infecciosa cutânea nodular causada por Mycobacterium sp. Apesar de ser relatada mundialmente, ainda é bastante desconhecida e subdiagnosticada. O diagnóstico pode ser conseguido por citopatologia ou histopatologia de lesões cutâneas, mas a identificação do agente infeccioso é complexa, uma vez que o crescimento in vitro bacteriano não é possível, dependendo de técnicas moleculares como a PCR para confirmar o DNA de Mycobacterium na amostra. Relatou-se um caso da SGLC em Niterói, estado do Rio de Janeiro, Brasil, diagnosticado por citopatologia e submetido à identificação molecular do agente. Foi realizada amplificação por PCR do gene hsp65, que revelou 100% de homologia genética com a cepa M. murphy. Este é o primeiro relato da SGLC com identificação molecular no estado do Rio de Janeiro, o que mostra a importância de se acrescentar a SGLC ao diagnóstico diferencial das doenças granulomatosas de pele nessa região.

ABSTRACT Canine Leproid Granuloma Syndrome (CLGS), also known as canine leprosy, is a cutaneous nodular infectious disease caused by Mycobacterium sp.. Despite being reported worldwide, it is still quite unknown and underdiagnosed. Diagnosis may be achieved by cytopathology or histopathology of skin lesions, but identification of the infectious agent is complex, since bacterial in vitro growth is not possible, relying upon molecular techniques such as PCR to confirm Mycobacterium DNA in the sample. We report a CLGS case in Niteroi, Rio de Janeiro state, Brazil, diagnosed by cytopathology and submitted to molecular identification of the agent. PCR amplification of hsp65 gene was performed and revealed 100% genetic homology to M. murphy strain. This is the first CLGS report with molecular identification in Rio de Janeiro state, and this finding should raise awareness about CLGS as a differential diagnosis among granulomatous skin diseases in this region.

No estágio inicial do trauma encefálico, o emprego de fração inspirada de oxigênio (FiO2) de 1,0 é indicado. Todavia, altas FiO2 têm sido correlacionadas com atelectasia. Assim, avaliaram-se os efeitos das FiO2= 1.0 e FiO2= 0.6 sobre a função cardiopulmonar em cães com pressão intracraniana (PIC) elevada e anestesiados com propofol. Oito animais foram anestesiados em duas ocasiões e receberam, durante a ventilação controlada, FiO2 = 1(G100) ou FiO2 = 0,6 (G60). Propofol foi usado para indução (10mg.kg-1) e seguido por infusão contínua (0,6mg.kg-1minuto-1). O aumento da PIC foi induzido pela obliteração temporária da veia jugular (OJv). As mensurações foram realizadas 20 minutos após OJv (T0) e em intervalos de 15 minutos (de T15 a T60). A pressão parcial de oxigênio alveolar no G60 foi menor do que no G100 durante todo o procedimento. O gradiente alveolar-arterial no G100 foi maior do que no G60, em T0 e T60. Não foram observadas diferenças para: relação pressão parcial de oxigênio/fração inspirada de oxigênio, relação arterioalveolar, índice respiratório, mistura arteriovenosa, oferta de oxigênio, consumo de oxigênio, taxa de extração de oxigênio, frequência cardíaca, pressão da artéria pulmonar média, pressão de oclusão da artéria pulmonar, índice cardíaco, índice sistólico e índice de resistência vascular sistêmica. No G100, a pressão arterial média em T0 foi maior do que em T45. Em cães com alta PIC, a função cardiopulmonar não foi influenciada pelas diferentes FiO2empregadas.

In the initial stage of traumatic brain injury, the use of 1.0 inspired oxygen fraction (FiO2) is indicated. However, high FiO2 has been correlated with atelectasis. Thus, the effects of FiO2= 1.0 and FiO2= 0.6 on the cardiopulmonary function in propofol-anesthetized dogs with high intracranial pressure (ICP) were evaluated. Eight dogs were anesthetized on two occasions, receiving, during controlled ventilation, an FiO2= 1 (G100) or an FiO2= 0.6 (G60). Propofol was used for induction (10mg.kg-1) followed by a continuous rate infusion (0.6mg.kg-1.minute-1). An increase in the ICP was induced by temporary obliteration of the right jugular vein (OJv) 50 minutes after induction of anesthesia. The measurement was taken twenty minutes after OJv (T0) and then at 15-minute intervals (T15 to T60). Alveolar oxygen partial pressure in G60 was lower than in G100 during the whole procedure. Alveolar-arterial oxygen gradient in G100 was greater than in G60 at T0 and at T60. No differences were observed for arterial oxygen partial pressure/inspired oxygen fraction ratio, arterial-to-alveolar oxygen pressure ratio, respiratory index, venous admixture, oxygen delivery, oxygen consumption, oxygen extraction, heart rate, mean pulmonary arterial pressure, pulmonary arterial occlusion pressure, cardiac index, stroke index and systemic vascular resistance index. In G100, mean arterial pressure at T0 was higher than at T45. In dogs with high ICP, the cardiopulmonary function was not influenced by the different FiO2 used.

A presença do limiar anaeróbio (LAn) em exercício resistido tem sido objeto de estudo nos últimos anos. O objetivo do estudo foi identificar e comparar o LAn em uma população idosa no leg press (LP) e supino reto (SR). Catorze homens idosos (68.9 ± 4.0 anos, 170.5 ± 0.05 cm e 76.1 ± 7.8 kg) realizaram dois testes incrementais máximos baseados em porcentagens de uma repetição máxima (1RM) para a determinação do LAn no LP e SR. As variáveis lactacidemia, frequência cardíaca (FC) e percepção subjetiva de esforço (PSE) foram coletadas ao final de cada estágio e comparadas entre os exercícios na intensidade referente ao LAn. O nível de significância adotado foi p = .05. O LAn no LP ocorreu em 27.9 ± 5.0% de 1RM, estatisticamente superior ao SR, que foi identificado em 21.5 ± 3.1% de 1RM. No entanto, a lactacidemia foi estatisticamente inferior no LP (1.36 ± 0.40 mmol.L-1) em relação ao SR (1.92 ± 0.38 mmol.L-1). Apesar de intensidades diferentes do LAn entre os exercícios, as variáveis FC e PSE não apresentaram diferenças significativas. Foi possível identificar o LAn em idosos nos aparelhos LP e SR, ocorrendo em intensidades inferiores no SR.

The presence of anaerobic threshold (AT) in resistance exercise has been studied in recent years. The aim of this study was to identify and to compare the AT in elderly population during leg press (LP) and bench press (BP). Fourteen elderly men (68.9 ± 4.0 years, 170.5 ± 0.05 cm e 76.1 ± 7.8 kg) performed two maximal incremental tests based on percentages of one repetition maximum (1RM) for the determination of AT during LP and BP. Blood lactate, heart rate (HR) and rating of perceived exertion (RPE) were measured at the final of each stage and it were compared between the exercises at the intensity of the AT. The level of significance was p = .05. The AT in the LP was 27.9 ± 5.0% of 1RM and in the BP was 21.5 ± 3.1% of 1RM, with statistical differences between the exercises. However, blood lactate was significantly lower on LP (1.36 ± 0.40 mmol.L-1) when compared with BP (1.92 ± 0.38 mmol.L-1). Although the intensity of AT has been different between the exercises, HR and RPE did not presented statistical significant differences. It was possible to identify the AT on elderly population during LP and BP exercises and it occurred in lower intensities for BP.

A enzima quitinase de Moniliophthora perniciosa, o fungo causador da doença vassoura de bruxa em Theobroma cacao, foi parcialmente purificada por precipitação com sulfato de amônio e filtração em coluna de Sephacryl S-200 usando fosfato de sódio como tampão de extração. A metodologia de Superfície de resposta (MSR) foi usada para determinar o pH e temperatura ótimos. Foram isoladas quatro quitinases diferentes: ChitMp I, ChitMp II, ChitMp III e ChitMp IV. A ChitMp I mostrou temperatura ótima de 44-73ºC e pH ótimo de 7,0-8,4. A ChitMp II apresentou temperatura ótima entre 45-73ºC e pH ótimo de 7,0-8,4. ChitMp III apresentou temperatura ótima de 54-67ºC e pH ótimo de 7,3-8,8. ChitMp IV apresentou temperatura ótima de 60ºC e pH ótimo em 7,0. Em relação à biologia computacional a sequência primária foi feita in silico usando banco de dados do Projeto Genoma/Proteoma do M. perniciosa, obtendo-se uma sequência com 564 pb e 188 aminoácidos, que foi utilizada para a obtenção da estrutura tridimensional por metodologia de modelagem comparativa. Os modelos gerados foram submetidos à validação usando Procheck 3.0 and ANOLEA. O modelo proposto para a quitinase foi submetido à análise dinâmica por 1 ns, resultando em um modelo com 91,7% de seus resíduos em locais regiões favoráveis no gráfico de Ramachandran e RMS de 2,68.

The enzyme chitinase from Moniliophthora perniciosa the causative agent of the witches' broom disease in Theobroma cacao, was partially purified with ammonium sulfate and filtration by Sephacryl S-200 using sodium phosphate as an extraction buffer. Response surface methodology (RSM) was used to determine the optimum pH and temperature conditions. Four different isoenzymes were obtained: ChitMp I, ChitMp II, ChitMp III and ChitMp IV. ChitMp I had an optimum temperature at 44-73ºC and an optimum pH at 7.0-8.4. ChitMp II had an optimum temperature at 45-73ºC and an optimum pH at 7.0-8.4. ChitMp III had an optimum temperature at 54-67ºC and an optimum pH at 7.3-8.8. ChitMp IV had an optimum temperature at 60ºC and an optimum pH at 7.0. For the computational biology, the primary sequence was determined in silico from the database of the Genome/Proteome Project of M. perniciosa, yielding a sequence with 564 bp and 188 amino acids that was used for the three-dimensional design in a comparative modeling methodology. The generated models were submitted to validation using Procheck 3.0 and ANOLEA. The model proposed for the chitinase was subjected to a dynamic analysis over a 1 ns interval, resulting in a model with 91.7% of the residues occupying favorable places on the Ramachandran plot and an RMS of 2.68.

Avaliaram-se os efeitos das frações inspiradas de oxigênio (FiO2) de 1 e 0,6 sobre o índice biespectral (BIS) em cães com pressão intracraniana elevada e submetidos a infusão contínua de propofol. Oito animais foram anestesiados em duas ocasiões, recebendo durante a ventilação controlada FiO2 = 1(G100) ou FiO2 = 0,6 (G60). Propofol foi usado para indução (10mg.kg-1) e seguido por infusão contínua (0,6mg.kg-1minuto-1). Após vinte minutos da implantação do cateter de fibra óptica do monitor de pressão intracraniana, na superfície do córtex cerebral direito, realizaram-se as mensurações basais de BIS, índice de qualidade de sinal, taxa de supressão, eletromiografia, pressão parcial de dióxido de carbono ao final da expiração, pressão arterial média, pressão intracraniana e pressão de perfusão cerebral. Em seguida, interrompeu-se o fluxo sanguíneo da veia jugular direita, para o aumento da pressão intracraniana e depois de 20 minutos, novas mensurações foram realizadas em intervalos de 15 minutos (T0, T15, T30, T45 e T60). A pressão parcial de oxigênio no sangue arterial variou conforme a FiO2. Para os outros parâmetros não foram registradas diferenças significativas. Conclui-se que o monitoramento pelo BIS não foi afetado pelo emprego de diferentes FiO2.

The effects of inspired oxygen fractions (FiO2) of 1 and 0.6 on bispectral index (BIS) in dogs with high intracranial pressure, submitted to a continuous rate infusion of propofol were evaluated. Eight dogs were anesthetized on two occasions, receiving, during controlled ventilation, an FiO2 = 1 (G100) or an FiO2 = 0.6 (G60). Propofol was used for induction (10mg.kg-1, IV), followed by a continuous rate infusion (0.6mg.kg-1.minute-1). After 20 minutes, a fiber-optic catheter was implanted on the surface of the right cerebral cortex to monitor the intracranial pressure, the baseline measurements of BIS values, signal quality index, suppression ratio number, electromyogram indicator, end-tidal carbon dioxide partial pressure, mean arterial pressure, intracranial pressure and cerebral perfusion pressure were taken. Then, the blood flow from the right jugular vein was interrupted in order to increase intracranial pressure and after 20 minutes additional recordings were performed at 15-minute intervals (T0, T15, T30, T45 and T60). The arterial oxygen partial pressure varied according to the changes in oxygen. For the other parameters, no significant differences were recorded. The BIS monitoring was not influenced by different FiO2.

Insertional mutagenesis is an important tool for functional genomics in Drosophila melanogaster. The insertion site in the KG00562 mutant fly line has been mapped to the CG8709 (herein named DmLpin) locus and to the 3’ of kermit (also called dGIPC). This mutant line presents a high lethality rate resulting from a gain of function. To obtain some insight into the biological role of the mutated locus, we have characterized the mutation and its relation to the high mortality of the KG00562 fly line. In this mutant, we did not detect one of the DmLpin transcripts, namely DmLpinK, but we did detect an unusual 2.3-kb mRNA (LpinK-w). Further investigation revealed that the LpinK-w transcript results from an aberrant splicing between the untranslated first exon of DmLpinK and the mini-white marker gene. Lack of DmLpinK or LpinK-w expression does not contribute to lethality, since heterozygous KG00562/Def7860 animals presented lethality rates comparable to those of the wild type. In contrast, the overexpression of kermit was associated with lethality of the KG00562 fly line. Significantly higher levels of kermit were detected in the Malpighian tubules of KG00562/+ flies that presented higher lethality rates than wild-type or KG00562/Def7860 animals, in which the lethality was rescued. In agreement with a recently reported study, our data support the hypothesis that misexpression of kermit/dGIPC could interfere with Drosophila development, with further investigations being needed in this direction.

Estudaram-se os parâmetros eletrocardiográficos utilizando-se os métodos convencional, computadorizado e computadorizado impresso de 30 cães da raça Beagle clinicamente normais. As medidas eletrocardiográficas dos três diferentes traçados foram analisadas na derivação DII. A comparação entre os métodos revelou diferença nos valores da onda P, onda R, complexo QRS, intervalo QT e intervalo PR e na polaridade da onda T. Este fato deve ser levado em consideração, na dependência do método a ser utilizado.

Electrocardiographic parameters were studied in thirty healthy adult Beagles, using conventional and computerized methods and reading the computerized registration printed on paper. The electrocardiographic measurements of the three different tracings were analyzed in lead II. The results obtained showed that there are differences among the three methods for P wave, R wave, QRS complex, QT interval, PR interval and polarity of the T wave. Special attention should be given to these differences, depending on the chosen method.

O estudo consistiu na identificação precoce da ocorrência de arritmias em cães com cardiomiopatia dilatada experimental induzida pela doxorrubicina (DOX). Utilizaram-se 10 cães adultos, sadios, distribuídos nos grupos A (n=5) e B (n=5). O grupo B recebeu 30mg/m² de DOX, via intravenosa, a cada 21 dias, até a dose cumulativa de 180 ou 240mg/m². No grupo A (controle), administrou-se solução salina 0,9%, via intravenosa, nos mesmos intervalos do grupo B. Ao se evidenciar o quadro de disfunção miocárdica nos cães do grupo B, caracterizado pela fração de encurtamento menor que 20%, aumento da separação septal do ponto E acima de 0,7cm e aumento do índice volumétrico do ventrículo esquerdo ao final da sístole (61,4ml/m²), realizaram-se os eletrocardiogramas por 24 horas. Os resultados demonstraram aumentos de 44,6% e 41,7% nas freqüências cardíacas mínima e média, respectivamente, e presença, com maior freqüência, de arritmias supraventriculares do que ventriculares nos animais do grupo B. Concluiu-se que o Holter é eficaz e demonstra, com precocidade e melhor definição, as alterações da freqüência e do ritmo cardíaco de cães com disfunção miocárdica induzida pela doxorrubicina.

Early identification of arrhythmias in dogs showing doxorubicin-induced cardiomyopathy was studied. Ten healthy dogs were assigned to groups A (n=5) and B (n=5). Dogs from group B were given doxorubicin 30mg/m² intravenously, every 21 days, until a cumulative dose of 180mg/m² or 240mg/m² was reached. Dogs from group A (used as control) were administered saline intravenously at the same group B intervals. As soon as myocardium dysfunction was observed in dogs from group B, determined by a shortening fraction below 20%, increased E-point to septal separation above 0.7cm, and increased end-systolic left ventricular volume index (61.4ml/m²), a 24-hour Holter was recorded in all dogs from both groups. There was an increase of minimum heart rate (44.6%) and mean heart rate (41.7%) in animals from group B in comparison with the control animals. Either supraventricular or ventricular arrhythmias were observed, despite group B dogs showed higher occurrence of supraventricular arrhytmias. Holter monitoring is efficient in early determination of heart rate and cardiac rhythm alterations in dogs showing doxorubicin-induced myocardial dysfunction.

Este artigo visa a comunicar o desenvolvimento de um software para tratamento de dados experimentais, denominado LAB Fit. Na verdade, trata-se de um pacote de programas envolvendo ajuste de curvas, gráficos 2D e 3D, estatística básica, propagação de erros e algumas ferramentas matemáticas. O software foi inicialmente publicado em inglês, buscando atingir um público alvo maior que o brasileiro, há cerca de dois anos. Devido ao sucesso alcançado, verificado pelo grande número de downloads e pelo número crescente de licenciamentos, está sendo disponibilizada, também, uma versão em português. Exemplos típicos de uso do LAB Fit mostram parte das potencialidades do software tanto no ensino como na pesquisa experimental. O seu desempenho foi verificado utilizando o ''Statistical Reference Datasets Project (SRDP)'' do ''National Institute of Standards and Technology (NIST)'', que apresenta 27 conjuntos de dados e respectivas funções com valores certificados para os parâmetros. Todos os 27 resultados obtidos pelo LAB Fit são estatisticamente idênticos aos valores certificados.

This paper aims to communicate the development of a software for treatment of experimental data called LAB Fit (curve fitting, 2D and 3D graphs, basic statistics, error propagation and mathematical tools). The english version was released two years ago and, due to the achieved success, verified by the great number of downloads, it is also being released a portuguese version. Typical examples of using of the LAB Fit show part of the potentialities of the program in the experimental teaching and research. The performance of the software has been verified with the Statistical Reference Datasets Project (SRDP) of the National Institute of Standards and Technology (NIST), which presents 27 datasets (and functions) with certified values for the parameters. All the 27 LAB Fit results are statistically identical to the certified values.
