OBJECTIVES: To investigate the expression levels of surface markers of activation (CD38 and HLA-DR), inhibition (PD-1, TIGIT and CD57) and co-stimulation (CD28 and CD127) on CD4+ T cells of children/adolescents with vertical HIV infection (HI patients) and HIV-uninfected (HU) controls vaccinated with the meningococcal C conjugate vaccine (MCC). METHODS: HI patients (n=12), aged 8-17 years, were immunized with two MCC injections, while HU controls (n=9), aged 5.3-10.7 years, received a single MCC dose (as per national recommendation at the time of this study, a single MCC vaccine dose should be given for healthy children and youth aged 1-18 years). The HI patients were categorized according to the combined antiretroviral therapy (cART) treatment. Blood samples were obtained before vaccination, after priming, and after the administration of a booster dose of vaccine to determine the serum bactericidal antibody (SBA) titers and the expression levels of surface markers on CD4+ T cells by flow cytometry. The levels of serum cytokines, IL-4 and CXCL-13 were also measured using Luminex kits. RESULTS: The co-expression of the TIGIT-HLA-DR-CD38 molecules increased in the CD4+ T cells of HI patients/no-cART who also showed a lower frequency of CD127+CD28+ CD4+ T cells than HI patients/cART and HU group subjects. There were significant negative correlations between the frequency of exhausted CD4+ T cells and the SBA response. IL-4 levels were higher in HI patients/cART and positively correlated with SBA titers but negatively associated with the expression of exhaustion markers. Moreover, the CXCL-13 levels were positively correlated with the exhausted CD4+ T cells. CONCLUSION: The results of our study suggest that the co-expression of exhaustion markers and/or loss of co-stimulatory molecules influence the SBA response in HI patients.

Abstract Background: New evidence demonstrates that aging and dyslipidemia are closely associated with oxidative stress, DNA damage and apoptosis in some cells and extravascular tissues. However, in monocytes, which are naturally involved in progression and/or resolution of plaque in atherosclerosis, this concurrence has not yet been fully investigated. In this study, we evaluated the influence of aging and hypercholesterolemia on serum pro-inflammatory cytokines, oxidative stress, DNA damage and apoptosis in monocytes from apolipoprotein E-deficient (apoE-/-) mice compared with age-matched wild-type C57BL/6 (WT) mice. Experiments were performed in young (2-months) and in old (18-months) male wild-type (WT) and apoE-/- mice. Results: Besides the expected differences in serum lipid profile and plaque formation, we observed that atherosclerotic mice exhibited a significant increase in monocytosis and in serum levels of pro-inflammatory cytokines compared to WT mice. Moreover, it was observed that the overproduction of ROS, led to an increased DNA fragmentation and, consequently, apoptosis in monocytes from normocholesterolemic old mice, which was aggravated in age-matched atherosclerotic mice. Conclusions: In this study, we demonstrate that a pro-inflammatory systemic status is associated with an impairment of functionality of monocytes during aging and that these parameters are fundamental extra-arterial contributors to the aggravation of atherosclerosis. The present data open new avenues for the development of future strategies with the purpose of treating atherosclerosis.

O cálcio é um nutriente essencial necessário em diversas funções biológicas. Estudos têm demonstrado a associação entre o baixo consumo de cálcio e doenças crônicas, entre elas osteoporose, câncer de colón, hipertensão arterial e obesidade. Entretanto, grande parte da população brasileira apresenta consumo de cálcio abaixo do recomendado. Este artigo objetiva revisar os fatores endógenos (idade e estado hormonal) e exógenos (fitatos, oxalatos, sódio, compostos bioativos e vitamina D) que influenciam a absorção do cálcio, bem como as principais metodologias utilizadas para avaliar a absorção e biodisponibilidade desse nutriente. Discorre-se sobre os possíveis fatores para o baixo consumo de cálcio: 1) Hábito alimentar - substituição de leite por bebidas com baixo teor de cálcio como o refrigerante, refeições realizadas fora de casa e a não realização de refeições como o café da manhã; 2) Alto custo dos alimentos fontes de cálcio. Além disso, este artigo discute as estratégias para otimizar o consumo do cálcio, que incluem: 1) Aumentar o conhecimento sobre a importância do consumo de cálcio para a saúde e as principais fontes alimentares desse nutriente; 2) Aumentar a disponibilidade de alimentos fortificados com cálcio; 3) Uso de suplementos em grupos específicos - quando e como administrar os sais de cálcio.

Calcium is an essential nutrient required for numerous biological functions. Studies have demonstrated an association between low calcium intake and chronic diseases, such as osteoporosis, colon cancer, hypertension, and obesity. However, most Brazilians do not meet the adequate intake for calcium. This review focuses on the endogenous (age, hormonal state) and exogenous (phytate, oxalate, sodium, bioactive compounds and vitamin D) factors that can influence calcium absorption. The main methods used for evaluating calcium absorption and bioavailability. The potential factors for the low calcium intake: 1) Food habits - substitution of milk for soft drinks, eating away from home and skipping meals specially breakfast; 2) High cost of dairy food. Besides, this article discuss strategies to optimize calcium intake: 1) Increase knowledge of the relationship between calcium and health and the main food sources; 2) Increase availability of calcium-fortified foods; 3) Supplement use for target groups - when and how administrate calcium salts.

Uncaria tomentosa (Willd.) DC. e Uncaria guianensis (Aubl.) Gmel., conhecidas comercialmente como unha-de-gato, são trepadeiras lenhosas nativas das florestas tropicais amazônica e central-americana. Essas espécies de Rubiaceae enfrentam ultimamente um eminente risco de extinção devido tanto ao extrativismo indiscriminado quanto ao desflorestamento de seus habitats naturais. Esse trabalho descreve métodos de cultivo in vitro para as duas espécies, analisando o perfil e o teor dos alcalóides oxindólicos pentacíclicos marcadores por CLAE no material vegetal obtido. Os resultados mostraram que os métodos desenvolvidos foram capazes de produzir plantas com perfil e teor dos alcalóides alvos similares à plantas nativas e cultivadas in vivo, com a vantagem adicional de produção em menor tempo de mudas próprias para plantio no campo. Os dados evidenciaram que o cultivo in vitro pode ser uma ferramenta viável para a produção das duas espécies com impacto na sua comercialização e na preservação das florestas.

Uncaria tomentosa (Willd.) DC. and Uncaria guianensis (Aubl.) Gmel., commercially known as cat's claw, are large woody vines native of the Amazon and Central America rainforests. These Rubiaceae species face nowadays an imminent risk of extinction due to indiscriminate harvesting in the wild as well as to increasing deforestation of their natural habitat. This work describes in vitro cultivation methods for both species with determination, by HPLC, of the pentacyclic oxindole alkaloid contents and profiles of the plant material. The results have proved that the methods we developed were able to produce plants with alkaloid profiles and contents similar to the wild and in vivo cultivated species, and with the additional advantage of producing suitable young plants in a shorter period of time. The data showed that the in vitro technique can be a feasible tool for the growth of the species, and may thus be important for their commercialization and for their conservation as a forest resource.
