RESUMO Nesta pesquisa foram realizados experimentos com duas classes de hormônios vegetais (giberelina e citocinina); com o objetivo de verificar o efeito do ácido giberélico e 6-benzilaminopurina (GA3 e BAP) sobre a lignificação e o crescimento longitudinal em mudas de Eucalyptus grandis. O experimento foi realizado em casa de vegetação, na qual foram aplicados os reguladores de crescimento combinados ou isolados, nas seguintes concentrações: GA3 (49,13 μM e 98,26 μM); BAP (111 μM e 222 μM), com cinco repetições para cada tratamento. As comparações entre as médias da altura das plantas aos 6 meses mostraram que os alongamentos mais pronunciados do caule corresponderam aos tratamentos com a maior concentração de GA3. No tratamento em que foi combinada a menor concentração de GA3 com a menor concentração de BAP (49,13 μM de GA3 + 111 μM de BAP), ocorreu o menor teor de lignina 23,37%.

ABSTRACT In this research experiments were accomplished with two classes of vegetable hormones (gibberellin and cytokinin); with the objective of verifying the effect of gibberellic acid and 6-Benzylaminopurine (GA3 and BAP) on the lignification and the longitudinal growth in seedlings of Eucalyptus grandis. The experiment was accomplished greenhouse, where were the growth regulators combined or isolated applied, in the following concentrations: GA3 (49.13 μM e 98.26 μM); BAP (111 μM e 222 μM), with five repetitions for each treatment. The comparisons among the averages of the height of the plants to the 6 months showed that the most pronounced prolongations of the stem corresponded to the treatments G[2], G[2]C[1] and G[2]C[2] where the largest concentration of GA3 was used. In the treatment G[1]C[1] where it was combined to smallest concentration of GA3 with to smallest concentration of BAP (49,13 μM de GA3 + 111 μM de BAP), it happened the smallest lignin tenor 23,37%.

A calogênese foi efetuada utilizando-se reguladores de crescimento em diferentes concentrações para obtenção das melhores características em relação ao aspecto da textura dos calos, com o objetivo principal de produzir calos friáveis de explantes caulinares de Eucalyptus grandis. Segmentos caulinares de aproximadamente um centímetro foram excisados de mudas de 30 dias. Os tratamentos foram elaborados tendo como base o meio MS adicionado de TDZ isolado ou combinado com AIA, 2,4-D, ANA. Cada tratamento constou de cinco repetições, com cinco explantes por placa. Os calos foram avaliados visualmente e por manipulação quanto à textura, após 210 dias de cultivo in vitro. Os explantes que não produziram calos foram aqueles expostos a tratamentos com 50,0 µM de 2,4-D e 3,0 µM de TDZ, e em ausência de regulador de crescimento (controle). Os melhores protocolos para manutenção de calos friáveis de explantes caulinares de Eucalyptus grandis, nas condições realizadas neste trabalho, foram os que utilizaram apenas TDZ.

Callogenesis was achieved using growth regulators at different concentrations to obtain the best characteristics regarding the calluses texture aspect with the main objective to produce friable calluses from stem explants of Eucalyptus grandis. Stem segments of approximately 1 cm were excised from 30-day seedlings. The treatments were prepared based on the MS medium added with TDZ alone or combined with AIA; 2,4-D; ANA. Each treatment consisted of five repetitions with five explants per plate. After 210 days of in vitro culture, the calluses were evaluated visually and by manipulation for texture. The explants that did not produce calluses were those exposed to treatments with 2,4-D 50.0 µM; TDZ 3.0 µM and in absence of a growth regulator (control).The best protocols for maintenance of friable calluses of stem explants of Eucalyptus grandis under the conditions performed in this work were those using only TDZ.

Neste trabalho, objetivou-se analisar a relação dos reguladores de crescimento GA3 e BAP na lignificação e, consequentemente no diâmetro das fibras de plantas de Eucalyptus grandis aos seis meses de idade. Estudos anatômicos, bioquímicos e genéticos em decorrência da aplicação exógena de reguladores de crescimento na fase de desenvolvimento do vegetal, têm sido cada vez mais enfatizados. Alguns desses estudos têm obtido avanço no que se refere à interação dos hormônios vegetais em relação à anatomia e ao processo de lignificação. A aplicação dos reguladores de crescimento GA3 e BAP foi realizada isoladamente ou combinados nas respectivas concentrações: GA3 (49,13 µM e 98,26 µM); BAP (111 µM e 222 µM), em mudas de Eucalyptus grandis até seis meses de idade. Análises químicas e anatômicas foram realizadas com o segmento caulinar das plantas tratadas. Para quantificar o teor de lignina pelo método Klason, as amostras foram livres para a retirada de extrativos e proteínas. Para a mensuração do diâmetro das fibras, as amostras, após sua dissociação, foram analisadas por microscopia ótica. O teor de lignina variou entre 23,37 a 29,64% e o diâmetro de fibra variou entre 13,33 a 20,39 µm. Também, pelos resultados, foi possível visualizar, com o tratamento G[1] (49,13 de GA3 com 111µM de BAP) que o teor de lignina reduziu em 21,2% em relação ao controle, e o C[1] diâmetro de fibra obteve o menor aumento em relação aos outros tratamentos.

The aim of this study was to analyze the influence of growth regulators GA3 and BAP on lignification and consequently on the diameter of fibers in Eucalyptus grandis plants at age six months. Anatomical, biochemical and genetic studies after exogenous application of growth regulators during the development phase have been increasingly explored. Some of these studies have achieved progress regarding the interaction of plant hormones with the anatomy and the lignification process in plants. The application of growth regulators GA3 and BAP was done separately or combined in the following concentrations: GA3 (49.13 µM and 98.26 µM), BAP (111 µM and 222 µM), using seedlings of Eucalyptus grandis at age up to six months. Chemical and anatomical analyses were performed on the stem segment of plants subjected to treatment. To quantify lignin content by the Klason method, samples were subjected to removal of extractives and proteins. To measure fiber diameter, once dissociated, samples were analyzed by optical microscopy. The lignin content ranged between 23.37% and 29.64% and fiber diameter ranged between 13.33 µm and 20.39 µm. From results it was possible to visualize, with the G[1]treatment (49.13 µM of GA3 with 111µM of BAP) that lignin content dropped C[1] by 21.2% in relation to the control treatment, and that fiber diameter had the smallest increase in relation to other treatments.

Lignina é uma biomacromolécula indispensável a diversos processos biológicos das plantas, dos quais o mais importante é conferir rigidez necessária à manutenção da verticalidade do caule e principalmente assegurar a existência de vias rápidas de circulação da água e minerais. Esta pesquisa foi realizada com plantas de Eucalyptus urophylla e os resultados podem viabilizar outros estudos na área da biotecnologia. Nesta pesquisa foram aplicadas diferentes concentrações de reguladores de crescimento [ácido jasmónico (1μM e 2 μM) e ácido 2,4-diclorofenoxiacético (18,12 μM e 36,24 μM)]. O experimento foi factorial completo com delineamento inteiramente casualizado. Os resultados mostraram que os tratamentos com o ácido jasmónico na sua primeira concentração activou o processo de formação da lignina enquanto a segunda concentração desactivou o processo de lignificação. No tratamento com 2,4-D (A1- 18,12 μM) o processo de lignificação contribuiu para reduzir a deposição de lignina, enquanto (A2 - 36,24 μM) teve uma redução menor comparado com a testemunha. Os tratamentos conjugados J1A1 (1μM de ácido jasmónico + e 18,12 μM de 2,4-D) e J1A2 (2μM de ácido jasmónico + e 36,24 μM de 2,4-D) apresentaram efeito sinergístico com predomínio do ácido jasmónico, sobre a biossíntese da lignina. Os resultados apontaram para modificação do processo de lignificação assim como na constituição proteica e nas características anatómicas.

La lignine est une biomacromolécule indispensable à divers processus biologiques des végétaux, dont le plus important est de conférer de la rigidité nécessaire au maintien de la verticalité de la tige principale et d'assurer l'existence des voies rapides de circulation de l'eau et de minéraux. Cette recherche a été réalisée avec des jeunes plantes de Eucalytpus urophylla et les résultats peuvent viabiliser d'autres études dans le domaine de biotechnologie. Dans cette étude, différentes concentrations de régulateurs de croissance ont été appliquées [acide jasmonique ((1μM et 2 μM) et acide 2,4-diclorephenoxiacétique (18,12 μM et 36,24 μM)]. Le dispositif expérimental a été le factoriel complet avec un design complètement aléatoire. Les résultats ont montré que les traitements avec l'acide jasmonique dans sa première concentration ont activé le processus de formation de la lignine pendant que la deuxième concentration a désactivé ce processus. Dans le traitement avec l'acide 2,4-D (A1- 18,12 μM), le processus de lignification a contribué pour la réduction du dépôt de lignine, alors que (A2 - 36,24 μM) a eu une réduction plus petite lorsque comparée avec le témoin. Les traitements combinés J1A1 (1μM d'acide jasmonique et 18,12 μM de 2,4-D) e J1A2 (2μM d'acide jasmonique et 36,24 μM de 2,4-D) ont présenté un effet synergique avec une prédominance de l'acide jasmonique sur la biosynthèse de la lignine. Les résultats indiquent une modification du processus de lignification ainsi que de la constitution des protéines et des caractéristiques anatomiques.

Lignin is an indispensable biomacromolecule for various biological processes in plants. One of the most important lignin functions is to maintain the rigidity needed for the verticality of the main stem and to ensure the water and mineral movement. This research was carried out with seedlings of Eucalyptus urophylla and the results may facilitate further studies in biotechnology. In this research were applied different concentrations of plant growth regulators jasmonic acid (1 and 2 μM) and 2,4-dichlorophenoxyacetic acid (18.12 and 36.24 μM)]. The experiment was full factorial with a completely randomized design. The results showed that the treatment with jasmonic acid in its first dosage enabled the process of lignin formation and in a second dosage disabled the process of lignification. In the treatment with 2,4-D (A1-18.12 μ M) the process of lignification contributed to reduce the deposition of lignin, while (A2 - 36.24 μM) had a smaller reduction compared with the standard. The combined treatments J1A1 (1 μM jasmonic acid and 18.12 + μ M 2,4-D) and J1A2 (2 μ M jasmonic acid and (36.24 + μ M 2,4-D) showed synergistic effect with a predominance of jasmonic acid on lignin formation. The results pointed out for change in the lignification process as well as protein and anatomical features.

Abstract The objective of this research was to suggest a systematic model of wood identification through metabolomic analysis by 13C NMR, toward a data bank formation. In this work cyclohexane extracts from wood of E. urograndis were used. Four specimes with aproximately 15 years old were colected from differents regions. The data of 13C NMR showed that spectra of hexane extractes from heartwood (DAP), were considered the most representative for the study for the E. urograndis, due to the largest number of existing spectrum signals. After the data it was possible to identify a common spectral profile for each anatomic region of wood (sapwood, transition zone and heartwood) of E. urograndis.

Resumo O objetivo da pesquisa foi propor um modelo sistemático de identificação de madeiras através de análise metabolômica por Ressonância Magnética Nuclear do 13C (RMN 13C) constituindo um banco de dados de 13C. Neste trabalho foram utilizados os extratos obtidos com ciclohexano de madeiras do híbrido E. urograndis. Quatro espécimes com aproximadamente 15 anos de idades foram coletados em diferentes localidades. Os dados de RMN 13C mostraram que os espectros dos extratos ciclohexano da madeira do cerne (DAP) foram os mais representativos para o estudo do E. urograndis devido ao número de absorções verificadas. Com os dados foi possível traçar o perfil espectral comum para o híbrido do E. urograndis de cada região anatômica (alburno, zona de transição e cerne).

As plantas lenhosas, de maneira geral, possuem alto teor de lignina, podendo alcançar valores entre 25 a 35% em relação a massa seca. Valores normalmente encontrados nas plantas arbóreas. Algumas plantas jovens ou de hábitos de crescimentos inferiores aos arbustivos possuem baixo teor de lignina, neste sentido, buscando superar os atuais níveis de lignificação, foram testados em plantas fatores exógenos de estimulação. Um sistema enzimático complexo é responsável pela formação biossintética dos precursores intermediários da lignina, entretanto as peroxidases são responsáveis pela fase determinante da polimerização na parede celular, cuja atividade foi pressupostamente modificada através da aplicação de cálcio e boro, visando aumentar a resistência da planta aos fatores bióticos e abióticos, entre outros. Para tal, utilizou-se a espécie Solanum gilo (família Solanaceae) cultivar Português como planta experimental sob vários tratamentos, monitorando as aplicações quinzenal e semanalmente, nas formas simples e múltiplas de aplicação de CaSO4, H3BO3, CaCl2 e H3BO3/CaSO4, CaSO4/CaCl2, CaCl2/ H3BO3 e CaSO4/H3BO3/CaCl 2. O tratamento CaSO4/CaCl2, estatisticamente avaliado, induziu às plantas a uma taxa de lignificação equivalente ao teor de lignina de 19%, superando a testemunha que apresentou 14 %. Modificações anatômicas foram também observadas nos elementos de vaso e fibras no que tange ao comprimento, diâmetro e espessura da parede celular. A quantificação da lignina foi realizada segundo o método de Klason. A espectroscopia no infravermelho foi utilizada para análise composicional, revelando a existência de modificação da composição de lignina guaiacílica da planta testemunha (G:S), 1,2:1 para lignina siringílica (G:S), 0,8:1 nas plantas que receberam o tratamento (CaSO4/CaCl2) quinzenal e semanalmente, respectivamente.

Normally woody plants present high lignin content, even overcoming 35% from dry weight. Such values are almost found in arboreal plants. However, young plants or plants with lower growth form than tree show low lignin content. In this term, current levels of lignification could be increased by exogenous factor of stimulation. There are complex enzymatic systems responsible for the lignin intermediary precursors formation, however, the peroxidases enzymes have been pointed out as responsible for the lignin polymerization in the cellular wall, whose activity was modified supposedly by calcium and boron application, seeking to increase the plant resistance to the biotic and non biotic factors. The Solanum gilo (Solanaceae family), (Portuguese cultivars) was used as experimental plant according to the several treatments, installed twice (15 days) and once (7 days) for month, in the simple and multiple form to CaSO4, H3BO3, CaCl2 and H3BO3/CaSO4, CaSO4/CaCl2, CaCl2/H3BO3 and CaSO4 /H3BO3/CaCl2 treatments, respectively. The treatment (CaSO4/CaCl2) appraised statistically showed plants with 19% of lignin, overcoming the standard plants that presented 14%. Anatomical modifications were also observed in the vessels and fibers according to the length, diameter and thickness of the cellular wall. The lignin was quantified by Klason method and the compositional analysis was carried out by infrared spectroscopy, (free extractives samples), revealing the existence of compositional modification from guaiacyl lignin in the standard plants (G:S) 1.2:1 to syringyl lignin (G:S) 0.8:1 in the plants that received (CaSO4/CaCl2) twice for month and weekly, respectively.

ABSTRACT This communication turn on the use of Solanun gilo (Solanaceae) for lignification test on the nutritional point of view. This plant was chosen due its fast growth, high calcium mobility and low lignin content (15,51%). The experiment was carried out using calcium sulfate (CaSO4) and calcium chloride (CaCl2) for weekly and biweekly application. The treated plants with CaSO4/CaCl2 (biweekly application) showed 4% of additional lignin content. High syringyl unit concentration was also verified at Guaiacyl: Syringyl ratio (G: S) of 0.8:1 and 1.2:1, for weekly and biweekly application, respectively. The estimation of the lignin content and its composition were performed by Klason and infrared spectroscopy methods.

ABSTRACT The vascular evolution date since 450 million years ago. During the evolution occured several botanical events. The lignification for example is one of the last steps of vascular differentiation, that is a characteristic of plants evolution. The understanding about vascular development, physiological, biochemical and molecular approaches must be considered. Those approaches have been possible due plants as Arabidopsis, Coleus, Zinnia and Populus, which present easy visualization, good adaptability, as well as great laboratory condition of surviving. With the available of the molecular technology will be possible to advance in the knowledge of the mechanisms control to distinguish the type of vascular organization, as well as the gens expression during the differentiation.

RESUMO Aevolução do tecido vascular de plantas data de 450 milhões anos atrás. Durante a evolução aconteceram várias ocorrências de natureza botânica. Uma das ocorrências na diferenciação nas plantas vasculares é, por exemplo, a lignificação, processo que caracteriza a fase final da diferenciação celular. Para o entendimento do desenvolvimento vascular, são usadas abordagens de cunho fisiológico, bioquímico e molecular. Estas abordagens têm sido possíveis em decorrência do uso de plantas que apresentam boa visualização, adaptabilidade em condições de laboratório e crescimento rápido como é o caso do Arabidopsis, Coleus, Zinnia e Populus, que são modelos padrões para o estudo de diferenciação vascular. Com a disponibilidade de tecnologia molecular será possível avançar no conhecimento dos mecanismos de controle que dirigem e definem o tipo de organização do tecido vascular, assim como expressões de genes participativos da fase de diferenciação.
