BACKGROUND Key genes control the infectivity of the Schistosoma haematobium causing schistosomiasis. A method for understanding the regulation of these genes might help in developing new disease strategies to control schistosomiasis, such as the silencing mediated by microRNAs (miRNAs). The miRNAs have been studied in schistosome species and they play important roles in the post-transcriptional regulation of genes, and in parasite-host interactions. However, genome-wide identification and characterisation of novel miRNAs and their pathway genes and their gene expression have not been explored deeply in the genome and transcriptome of S. haematobium. OBJECTIVES Identify and characterise mature and precursor miRNAs and their pathway genes in the S. haematobium genome. METHODS Computational prediction and characterisation of miRNAs and genes involved in miRNA pathway from S. haematobium genome on SchistoDB. Conserved domain analysis was performed using PFAM and CDD databases. A robust algorithm was applied to identify mature miRNAs and their precursors. The characterisation of the precursor miRNAs was performed using RNAfold, RNAalifold and Perl scripts. FINDINGS We identified and characterised 14 putative proteins involved in miRNA pathway including ARGONAUTE and DICER in S. haematobium. Besides that, 149 mature miRNAs and 131 precursor miRNAs were identified in the genome including novel miRNAs. MAIN CONCLUSIONS miRNA pathway occurs in the S. haematobium, including endogenous miRNAs and miRNA pathway components, suggesting a role of this type of non-coding RNAs in gene regulation in the parasite. The results found in this work will open up a new avenue for studying miRNAs in the S. haematobium biology in helping to understand the mechanism of gene silencing in the human parasite Schistosome.

Abstract Objective The present study aimed to investigate the participation of focal adhesion kinases (FAK) in interactions between osteoblastic cells and titanium (Ti) surfaces with three different topographies, namely, untreated (US), microstructured (MS), and nanostructured (NS). Methodology Osteoblasts harvested from the calvarial bones of 3-day-old rats were cultured on US, MS and NS discs in the presence of PF-573228 (FAK inhibitor) to evaluate osteoblastic differentiation. After 24 h, we evaluated osteoblast morphology and vinculin expression, and on day 10, the following parameters: gene expression of osteoblastic markers and integrin signaling components, FAK protein expression and alkaline phosphatase (ALP) activity. A smooth surface, porosities at the microscale level, and nanocavities were observed in US, MS, and NS, respectively. Results FAK inhibition decreased the number of filopodia in cells grown on US and MS compared with that in NS. FAK inhibition decreased the gene expression of Alp, bone sialoprotein, osteocalcin, and ALP activity in cells grown on all evaluated surfaces. FAK inhibition did not affect the gene expression of Fak, integrin alpha 1 ( Itga1 ) and integrin beta 1 ( Itgb1 ) in cells grown on MS, increased the gene expression of Fak in cells grown on NS, and increased the gene expression of Itga1 and Itgb1 in cells grown on US and NS. Moreover, FAK protein expression decreased in cells cultured on US but increased in cells cultured on MS and NS after FAK inhibition; no difference in the expression of vinculin was observed among cells grown on all surfaces. Conclusions Our data demonstrate the relevance of FAK in the interactions between osteoblastic cells and Ti surfaces regardless of surface topography. Nanotopography positively regulated FAK expression and integrin signaling pathway components during osteoblast differentiation. In this context, the development of Ti surfaces with the ability to upregulate FAK activity could positively impact the process of implant osseointegration.

In occult hepatitis B infection (OBI), hepatitis B virus DNA (HBV DNA) can be detected in serum samples; however, oral fluid collection for detection of HBV DNA has not yet been explored, despite the availability of collection devices. Serum and oral fluid samples from 45 hepatitis B core antibody (anti-HBc)-positive patients were collected for the amplification of the HBV polymerase gene. HBV DNA was detected in five serum and four oral fluid samples (the detection limit for oral fluid was 1.656 log IU/mL in paired serum). In conclusion, simple methodologies of sample collection and in-house polymerase chain reaction (PCR) allowed detection of HBV DNA, and these could be used to improve the diagnosis of OBI, especially in locations with limited resources.

Resumo A artéria tibial posterior e a artéria fibular se originam do tronco tibiofibular, na fossa poplítea. A trajetória clássica da artéria tibial posterior é correr entre o tríceps sural e os músculos do compartimento posterior da perna, e, então, seguir posteriormente ao maléolo medial. Já a artéria fibular corre na margem lateral da perna, seguindo profundamente aos músculos. O estudo dessas artérias é relevante para o campo da angiologia, cirurgia vascular e cirurgia plástica. O presente trabalho é o primeiro relato de caso de uma anastomose entre ambas artérias, na porção distal de suas trajetórias. Tais artérias se anastomosaram em formato de “X”, antes da divisão da artéria tibial posterior em ramos plantares. Foi feita uma revisão de literatura das variações de tais artérias, dando ênfase ao aspecto clínico e embriológico, de modo a contribuir para novas investigações sobre esses vasos.

Abstract The posterior tibial artery normally arises from tibial-fibular trunk at the popliteal fossa, together with the fibular artery. The classic course of the posterior tibial artery is to run between the triceps surae muscle and muscles of the posterior compartment of the leg before continuing its course posteriorly to the medial malleolus, while the fibular artery runs through the lateral margin of the leg. Studies of both arteries are relevant to the fields of angiology, vascular surgery and plastic surgery. To the best of our knowledge, we report the first case of an anastomosis between the posterior tibial artery and the fibular artery in their distal course. The two arteries joined in an unusual “X” format, before division of the posterior tibial artery into plantar branches. We also provide a literature review of unusual variations and assess the clinical and embryological aspects of both arteries in order to contribute to further investigations regarding these vessels.

O objetivo deste trabalho foi desenvolver um sistema de transformação genética para genótipos tropicais de milho pelo bombardeamento de embriões imaturos com micropartículas. O bombardeamento com partículas foi realizado utilizando-se uma construção gênica contendo os genes bar e uidA sob o controle do promotor CaMV35S. As melhores condições para transformação das linhagens de milho tropical L3 e L1345 foram obtidas após o cultivo de embriões imaturos durante 4 horas, antes do bombardeamento, em alta osmolaridade, e bombardeados com a pressão de 1.100 psi de gás hélio, dois tiros por placa e uma distância de vôo da micropartícula de 6,6 cm. As freqüências de transformação obtidas variaram entre 0,9 e 2,31%. A integração dos genes heterólogos no genoma das plantas transgênicas de milho foi confirmada por análises de Southern blot e expressão dos genes bar e uidA. O protocolo de transformação genética de milho desenvolvido neste estudo irá aumentar, provavelmente, a eficiência de produção de novas linhagens tropicais de milho expressando características agronômicas desejáveis.

The objective of this work was to develop a genetic transformation system for tropical maize genotypes via particle bombardment of immature zygotic embryos. Particle bombardment was carried out using a genetic construct with bar and uidA genes under control of CaMV35S promoter. The best conditions to transform maize tropical inbred lines L3 and L1345 were obtained when immature embryos were cultivated, prior to the bombardment, in higher osmolarity during 4 hours and bombarded at an acceleration helium gas pressure of 1,100 psi, two shots per plate, and a microcarrier flying distance of 6.6 cm. Transformation frequencies obtained using these conditions ranged from 0.9 to 2.31%. Integration of foreign genes into the genome of maize plants was confirmed by Southern blot analysis as well as bar and uidA gene expressions. The maize genetic transformation protocol developed in this work will possibly improve the efficiency to produce new transgenic tropical maize lines expressing desirable agronomic characteristics.

The main objective of this work was to determine the effect of the use of hypophyseal raw extract to induce fish spawning in curimbatá specie. An election of females of frog bull (Rana castesbeiana) was carried through for extraction of hyphophysys, where the female donors had been those that presented an increment of their visceral socket, which was provoked by ovaries in mature phase or advanced maturation. Twenty curimbatá breeders (Prochilodus affinis) had been used for spawning. The males had an initial average weight of 170.0 g and an average length of 21.72 cm and the females with an initial weight average of 181.0 g and an average length of 6.28 cm. The experiment was carried through with 2 treatments: Treatment 1: 0.5 mg/kg and Treatment 2: 0.7mg/kg. The animals were stocked in aquarium with 1000 L capacity with constant aeration. The females received two intramuscular injections of frog bull raw extract, being the first dose of 0.5 mg/kg and the second 0.7 dose of mg/kg, with interval of 12 hours. The females with dosages of 0.5 mg/kg had obtained a superior number of larvae than those with 0,7 mg/kg. The difference was statistical significant (P>0,06). This result was probably due to different stadiums of gonadal maturation. Both dosages (0.5 and 0.7 mg/kg) of frog bull hypophyseal raw extract had allowed to induce reproduction of curimbatá females, showing an alternative in the reproductive handling.

La objetivou-se com este trabalho determinar o efeito do uso do extracto bruto de hipófisis (EBH) na indução da desova em curimbatá. Foi realizado uma seleção de fêmeas de rã touro (Rana castesbeiana) para extração da hipófise, onde as fêmeas doadoras foram aquelas que apresentavam aumento da cavidade visceral, provocado pelo ovários em estádio maduro ou de maturação avançada. Para desova foram utilizados 20 reprodutores de curimbatá (Prochilodus affinis). Os machos com peso médio inicial de 170,0 g e comprimento médio de 21,72 cm e as fêmeas com peso médios iniciais de 181,0 g e comprimento médio de 6,28 cm. O experimento foi realizado com dois tratamentos: Tratamento 1: 0,5 mg/kg e Tratamento 2: 0,7 mg/kg. Os animais foram alojados em aquário com 1000 L de água, aeração constante. Vinte fêmeas receberam duas injeções intramusculares de extrato bruto de rã touro, sendo a primeira dose de 0,5 mg/kg e a segunda dose de 0,7 mg/kg, com intervalo de 12 horas. Os curimbatás submetidos a dosagens de 0,5 mg/kg obtiveram números de larvas superior e estatisticamente significativas (P>0,06) ao tratamento 2 utilizando 0,7 mg /kg, provavelmente esse resultado pode ser em função de umas fêmeas estarem em diferentes estádios de maturação gonadal. Em curimbatá duas de doses de extrato bruto de hipófise de rã touro nas concentrações de 5 e 7 mg/kg, permitiram a indução reprodutiva das fêmeas, mostrando uma alternativa no manejo reprodutivo.

INTRODUÇÃO: A compartimentação atrial intra-operatória foi realizada em 27 pacientes, utilizando ultra-som (US). Esta forma de energia parece ser mais efetiva na criação de linhas de lesão nos átrios, mais profundas e mais uniformes, sem causar carbonização. CASUÍSTICA E MÉTODOS: Foram operados, de março de 1999 e junho de 2000, 27 pacientes, com média de idade de 36 anos. Destes, 19 eram mulheres, 23 eram portadores de doença reumática na valva mitral (5 eram casos de reoperação), 2 apresentavam insuficiência mitral por degeneração mixomatosa, 1 era portador de cardiopatia congênita e 1 apresentava fibrilação atrial isolada. RESULTADOS: Os tempos operatórios foram em média de 166,6 minutos para a operação, 69,2 minutos de circulação extracorpórea, 39,7 minutos de parada cardíaca pelo pinçamento da aorta, e 12,5 minutos e 14 minutos para efetuar as linhas de ablação nos átrios direito e esquerdo, respectivamente. Houve reversão ao ritmo sinusal em 24 pacientes, durante o ato operatório. Em 2 pacientes a reversão não foi obtida e 1 paciente apresentou bloqueio atrioventricular transitório, não havendo uma explicação plausível para os 2 casos de não reversão, pois os 2 pacientes foram submetidos a operação para correção da valvopatia pela primeira vez e o átrio não estava muito dilatado. Foram observadas duas recorrências e dois óbitos ocorreram por insuficiência respiratória e choque cardiogênico, no pós-operatório imediato, sem relação com a técnica. A porcentagem de sucesso foi de 81,4%, por ocasião da alta hospitalar. Todos os pacientes fizeram uso de verapamil ou amiodarona, para remodelação atrial. CONCLUSÃO: O uso do ultra-som, para criar linhas de ablação no átrio, durante a cirurgia cardíaca, é efetivo, modifica e torna mais fácil o procedimento do labirinto, causa menos dano aos tecidos, diminui o tempo de cirurgia e as possibilidades de complicações pós-operatórias.

INTRODUCTION: The intraoperative atrial compartmentalization was done in 27 patients, utilizing ultrasound energy (US). This source has been more effective in creating deeper and uniform atrial line lesions, without causing carbonization. MATERIAL AND METHODS: From March 1999 to June 2000, 27 patients (19 females) averaging 36 years old were submitted to this operation. Five were operated for the second time due to mitral valve disorders, 2 for mixomatose valve degenerations, 1 for congenital defect and 1 for lone atrial fibrillation. RESULTS: The procedure takes 166.6 minutes on average, in each surgery, that is 69.2 minutes for the cardiopulmonary bypass, 39.7 minutes for cardiac arrest by aorta clamping, 12.5 minutes for the right atrial lesion lines and 14 minutes for the left one. The reversion to sinus rhythm was achieved in 24 patients. In 2 the reversion was not obtained and in 1 transient total atrium-ventricular block was observed. No explanation was found for the failure to revert the 2 patients, since both were primarily phase submitted to valve repair operation and the atrium was not very enlarged. In the postoperative phase, atrial fibrillation recurrence was observed in 2 patients and two deaths occurred cuased by pulmonary problem and cardiogenic shock unrelated to the technique. The success rate at the hospital discharge was 81.4%. To all patients were given verapamil or amiodarone for electrical atrium remodeling. CONCLUSION: The use of US during mitral valve operation to compartmentalize the atrium, in patients with CAF, makes the reversion to sinus rhythm so easy and fast that is possible to consider employing this technique in patients having CAF associated to other surgical cardiac diseases. The use of US, during cardiac operation, to create the atrial line lesion, modifies and makes easier the Maze procedure, causes less tissue damage, decreases the surgical time and the chance for postoperative complications.

A miocardiopatia provocada pela doença de Chagas cria, freqüentemente, circuitos elétricos de reentrada, possibilitando o desencadeamento de taquicardia ventricular, geralmente refratária (TVR) às drogas antiarrítmicas. Muitas vezes, este quadro desorganiza eletricamente os ventrículos, provocando a morte do paciente. Nova proposta técnica para tratamento da (TVR) foi empregada em 9 pacientes, portadores da doença de Chagas com esta arritimia, sendo 8 com aneurisma de ponta e 1 da região infero-basal. Na maioria dos casos, o foco da taquicardia encontrava-se fora da borda do aneurisma ou da área de fibrose, geralmente na região basal ou póstero-lateral do ventrículo esquerdo. A idade variou entre 34 e 62 anos, com média de 48. Quatro eram do sexo masculino e 5 do feminino. Todos encontravam-se no grau funcional III e IV e a maioria apresentava episódios freqüentes de síncope, provocados pela taquicardia. Em 2 dos pacientes havia relato de AVC prévio e foi encontrado aneurisma de ponta em 8, aneurisma póstero-basal com extensa fibrose em 1 e trombo intracavitário em 6. Durante o ato cirúrgico foram induzidas as taquicardias clínicas em todos os pacientes. Na região em que o toque do instrumental cirúrgico conseguiu interrompê-las, foram realizadas aplicações de radiofreqüência, através de cateteres de ablação, no centro e nas bordas do foco. Logo após, e no sétimo dia de pós-operatório, nenhuma taquicardia pôde ser induzida com os protocolos de estimulação ventricular programada. A evolução de 13 +/- 7 meses, sem uso de drogas antiarrítmicas, mostra que 8 estão assintomáticos e em classe funcional I e II. Um paciente, com doença pulmonar obstrutiva crônica, faleceu de insuficiência respiratória no pós-operatório tardio (3 meses), sem ter apresentado taquicardia. Em conclusão, esta técnica é facilmente reprodutível, podendo ser realizada com simplicidade, sem necessidade de aparelhagem sofisticada de eletrofisiologia, e apresenta alto grau de sucesso na cura da TVR. Em nossa casuística, até o momento, o índice foi de 100%.

The ventricular reentry circuits commonly found in Chagas'Disease(CD) frequently originate ventricular tachycardia(VT) that can lead to the patient's death. Most of this arrhythmia is drug-resistant. A new surgical technique was used on 9 patients, with VT and CD. 8 had aneurysms in the apical and 1 in the infero-basal portion of the left ventricle. Ages ranged from 34-62 (mean 48), with five females. All patients were classified as functional class III and IV (NYHA), and most presented syncope associated with VT. During surgery, clinical tachycardias were induced by programmed ventricular pacing and were interrupted by endocardial touch mapping, using a surgical instrument in the region of probable origin of tachycardia. Previous morphology registered on the 12 leads surface EKG oriented by its. In that and its surrounding area, radiofrequence was applied using a 7f ablation catheter. The patients did not require any antiarrhythmic drugs and were followed up for 13+/-7 months; eight are in good status, asymptomatic and in functional class I and II, without any tachycardia; one died from lung disease three months later without tachycardia during this period. This technique is simple, reproducible and achieves a high rate of success (100% in our series) and does not require sophisticated electrophysiological personnel and facilities.
