ABSTRACT This study investigated the effects of dietary supplementation with Mansoa alliacea hydroalcoholic extracts on growth, blood and immune parameters of Arapaima gigas. Fish were fed for 30 days with diets enriched with 0, 4, 8, and 12 g kg-1 of M. alliacea hydroalcoholic extract and subjected to infection with Aeromonas hydrophila and handling stress. Fish fed with 8 g kg-1 of extract showed significant increase in final weight, specific growth rate and feed efficiency when compared to the other groups. Glucose, triglycerides, total proteins, and globulins increased significantly in fish fed with 8 g kg-1 of extract, whereas albumin decreased. The number of thrombocytes increased significantly with the dietary supplementation of 8 and 12 g kg-1 of extract. After the challenge with A. hydrophila and handling stress, fish fed with 8 g kg-1 of extract had significantly higher levels of glucose, globulins, and albumins, and fish fed with 8 and 12 g kg-1 of extract showed an increment of respiratory burst. Triglyceride levels dropped significantly in fish fed with 4, 8, and 12 g kg-1 of extract, whereas the number of neutrophils increased, and total thrombocytes, leukocytes and lymphocytes were higher in fish fed with 12 g kg-1 of extract. Dietary supplementation with M. alliacea extract at 8 g kg-1 was efficient in improving the growth and innate immunity of A. gigas, being potentially useful in fish farming to control the development of A. hydrophila infections.

Abstract Sampling deep-sea biota is a significant challenge because of the logistics required, in terms of vessels and equipment, to obtain minimally preserved specimens. Traditional methods (trawls, nets, and dredges) cause physical damage, stress, and even contamination during the process of removal from the seabed and their displacement through the water column to the surface. Preserving conditions similar to those found in situ is particularly important when the sampling strives to maintain living organisms and for analyses where contamination or degradation by stress or damage may interfere with the results. Therefore, for the sampling and storage of this biota with less interference, a polypropylene box was designed based on the model of Kellogg et al. (2009) incorporating adaptations to be used by a Remotely Operated Vehicle (ROV). This new device has been successfully used in eight oceanographic campaigns, adequately performing for sediment and biota sampling, including coral reef forming or framework species (Scleractinia), octocorals, associated fauna, and rhodoliths, at depths between 50 and 900 m.

ABSTRACT The objective of this study was to evaluate genotypes of Cucurbita moschata and Cucurbita maxima for resistance to the fungus Macrophomina phaseolina (Taissi) Goid. The experiment evaluated 17 accessions of Cucurbita moschata Duchesne and 20 accessions of Cucurbita maxima Duchesne preserved in the Cucurbit Germplasm Collection of the Federal Rural University of the Semi-Arid Region (UFERSA). Four commercial controls were also used, two for each species. In both studies, three isolates of M. phaseolina were inoculated, which are kept at the Fungal Culture Collection of UFERSA: MM1531 (GenBank identificatio: MM1531 (GenBank identification: MN136199), ME249 and ME250. Plants received inoculum of the pathogen in pieces of colonized toothpick and were evaluated for the incidence and severity of the disease. Both species showed resistant plants, but with varied frequencies for the accessions. Considering the species, the frequencies observed for C. moschata were higher than those observed for C. maxima. The accessions of C. moschata ABPUN 206 F1, P114-1, P160-2, P11-2, P114-6, P14-02 and P97-1 show different levels of resistance to the Macrophomina phaseolina isolates MM1531, ME-249 and ME-250.Thus, for an efficient selection in a breeding program for the characteristic, the selection strategies adopted must consider each source of resistance in isolationand not each species.

ABSTRACT Shallow flows correspond to turbulent flows whose horizontal dimensions are considerably larger than the vertical one. In Hydraulic Engineering, they refer, e.g., to stormwater basins, stabilization ponds for wastewater treatment and aquaculture tanks. Since they involve low flow velocities, a continuous settling process often affects such shallow reservoirs. Therefore, it is important to expand the knowledge about the influence of its geometry on the hydrodynamic behavior and the sedimentation tendency. This paper aims to analyze flow patterns in a rectangular reservoir with symmetrically positioned upstream and downstream channels, taking into account three different flow rates under steady flow regime (0.50, 1.25 and 3.40 L/s). Experimental tests were performed in a laboratory prototype, consisted of a 3.0 m long and 2.0 m wide reservoir, with a maximum depth of 0.30 m. Also, it was applied the WOLF 2D software for numerical modeling of all variants. Experimentally, a symmetrical hydrodynamic behavior was observed only for the lowest flow rate, while the flow pattern was asymmetrical for the other cases. On the other hand, the numerical model indicated hydrodynamic symmetry for all scenarios.

OBJECTIVE: MicroRNAs (miRNAs) are small non-coding RNAs that regulate gene expression at the posttranscriptional level. Some miRNAs, including let-7a and miR-195, have been described as tumor suppressors. However, the roles of these microRNAs in breast cancer progression remain controversial. The aim of this study is to evaluate miR-195 and let-7a expression as potential biomarkers of invasive breast cancer. METHODS: In the present study, 200 individuals were separated into three groups: (i) 72 women constituting the control group who were selected according to rigorous and well-established criteria; (ii) 56 patients with benign breast tumors; and (iii) 72 patients with malignant breast cancers of different clinical stages. The miR-195 and let-7a expression levels in serum were evaluated by real-time PCR. The results were assessed alone and in combination, and the analysis included an estimation of sensitivity and specificity in ROC curves. RESULTS: Compared with the benign and control groups, both microRNAs were downregulated in the malignant breast cancer patient group. Compared with the malignant group, the combination of both biomarkers in the control and benign groups showed good sensitivity and specificity in the serum with AUCs of 0.75 and 0.72, respectively. The biomarker combination for the control group versus the malignant group exhibited a better sensitivity and specificity than for the benign group versus the malignant group. CONCLUSION: These findings support the evidence that the analysis of miR-195 and let-7a can be used as a non-invasive biomarker for breast cancer detection.

ABSTRACT: The use of animal-origin residues as a nutrient source without prior and proper investigations on their positive and negative impacts can affect the quality of agricultural soils and the environment into which they are inserted. In this sense, the aim of this study was to assess the impact of swine wastewater and mineral fertilization application on soil, soybean crop, and leachate characteristics. The experiment was developed in drainage lysimeters under field conditions. The following doses of swine wastewater were applied: 0, 100, 200, and 300 m3 ha−1, both with and without mineral fertilization. Swine wastewater application contributed to increasing soil nutrients; however, a special attention should be paid to Cu+ and Zn+ accumulation, which may interfere with soil quality in the long-term. In addition, Na+, K+, and N can be leached into groundwater and cause impacts on water quality. Wastewater supplied soybean crop requirements with some nutrients such as K+ and Zn2+, without the need for an additional mineral supplementation. Thus, once the above conditions are monitored, swine wastewater can be applied to soil.

ABSTRACT: The working equid population in Corumbá, Southern Pantanal, is very large and has a crucial role in the main economic activity of the State of Mato Grosso do Sul, the beef cattle industry. The aim of the present study was to estimate the prevalence of equine infectious anaemia (EIA) in working equids of ranches in the municipality of Corumbá, by the official agar gel immunodiffusion (AGID) test, and evaluate the adoption of the Programme for the Prevention and Control of Equine Infectious Anaemia proposed by Embrapa Pantanal and official entities in the 1990s. From September to November 2009, forty ranches distributed through the area of the municipality were visited, and serum samples were obtained from 721 equines and 232 mules. According to previous publications and the present data, it was concluded that the prevalence of EIA in this population has increased from 18.17% to 38.60%, which represents at this time approximately 13,000 infected animals. There was no significant difference between the apparent prevalence of equines and mules. It was also verified that the control programme was not known by the greater part of the interviewed ranch owners, managers and foremen and, in their perception, EIA is not a primary threat to address. Among the studied variables, the serologic testing practice significantly reduced the risk for the presence of EIA seropositivity, as well as the separation of riding equipment and segregation of seropositives.

Abstract The aim was to describe the morphology of the tongue of the Marsh deer (Blastocerus dichotomus), the largest deer of the Brazilian fauna, since there is little detailed information on their morphology. Tongues of eight adult Marsh deers, belonging to Projeto cervo-do-pantanal de Porto Primavera were analyzed for their macroscopic aspects and by light microscopy. The Marsh deer tongue occupies most of the oral cavity, where the root and body are fixed caudally by the hyoid bone, and in its middle portion by the frenulum linguae; its free apex is flat and plane, has little prominent torus lingae and there is no demarcation of fossa linguae. The lateral and ventral surfaces are covered by a thin mucosa; however the dorsal surface is covered by thick mucosa, although soft. On the dorsal surface are the papillae: filiform, conical, lenticular, fungiform and vallate. Histologically was found that the mucosa has a stratified squamous epithelium, the lamina propria consists of connective tissue rich in collagen fibers arranged in several directions, where there is abundant vascularization and accumulation of lymphoid tissue.

OBJECTIVE: Exercise is a protective factor for cardiovascular morbidity and mortality, with unclear mechanisms. Changing the myocardial metabolism causes harmful consequences for heart function and exercise contributes to metabolic adjustment modulation. Peroxisome proliferator-activated receptors (PPARs) are also myocardium metabolism regulators capable of decreasing the inflammatory response. We hypothesized that PPAR-α is involved in the beneficial effects of previous exercise on myocardial infarction (MI) and cardiac function, changing the expression of metabolic and inflammatory response regulators and reducing myocardial apoptosis, which partially explains the better outcome. METHODS AND RESULTS: Exercised rats engaged in swimming sessions for 60 min/day, 5 days/week, for 8 weeks. Both the exercised rats and sedentary rats were randomized to MI surgery and followed for 1 week (EI1 or SI1) or 4 weeks (EI4 or SI4) of healing or to sham groups. Echocardiography was employed to detect left ventricular function and the infarct size. Additionally, the TUNEL technique was used to assess apoptosis and immunohistochemistry was used to quantitatively analyze the PPAR-α, TNF-α and NF-κB antigens in the infarcted and non-infarcted myocardium. MI-related mortality was higher in SI4 than in EI4 (25% vs 12%), without a difference in MI size. SI4 exhibited a lower shortening fraction than EI4 did (24% vs 35%) and a higher apoptosis/area rate (3.97±0.61 vs 1.90±1.82) in infarcted areas (both p=0.001). Immunohistochemistry also revealed higher TNF-α levels in SI1 than in EI1 (9.59 vs 4.09, p<0.001) in infarcted areas. In non-infarcted areas, EI4 showed higher levels of TNF-α and positive correlations between PPAR-α and NF-κB (r=0.75, p=0.02), in contrast to SI4 (r=0.05, p=0.87). CONCLUSION: Previously exercised animals had better long-term ventricular function post-MI, in addition to lower levels of local inflammatory markers and less myocardial apoptosis, which seemed to be related to the presence of PPAR-α.

The phosphatidylinositol 3-kinase and mitogen-activated protein kinase pathways mediate β cell growth, proliferation, survival and death. We investigated whether protein restriction during pregnancy alters islet morphometry or the expression and phosphorylation of several proteins involved in the phosphatidylinositol 3-kinase and mitogen-activated protein kinase pathways. As controls, adult pregnant and non-pregnant rats were fed a normal-protein diet (17%). Pregnant and non-pregnant rats in the experimental groups were fed a low-protein diet (6%) for 15 days. Low protein diet during pregnancy increased serum prolactin level, reduced serum corticosterone concentration and the expression of both protein kinase B/AKT1 (AKT1) and p70 ribosomal protein S6 kinase (p70S6K), as well as the islets area, but did not alter the insulin content of pancreatic islets. Pregnancy increased the expression of the Src homology/collagen (SHC) protein and the extracellular signal-regulated kinases 1/2 (ERK1/2) independent of diet. ERK1/2 phosphorylation (pERK1/2) was similar in islets from pregnant and non-pregnant rats fed a low-protein diet, and was higher in islets from pregnant rats than in islets from non-pregnant rats fed a normal-protein diet. Thus, a short-term, low-protein diet during pregnancy was sufficient to reduce the levels of proteins in the phosphatidylinositol 3-kinase pathway and affect islet morphometry.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The objective of this work was to evaluate the parasitic fauna of hybrid tambacu (Colossoma macropomum x Piaractus mesopotamicus) from fish farms and the host-parasite relationship. A hundred and fourteen fish were collected from four fish farms in Macapá, in the state of Amapá, Brazil, 80.7% of which were infected by: Ichthyophthirius multifiliis (Ciliophora); Piscinoodinium pillulare (Dinoflagellida); Anacanthorus spatulatus, Notozothecium janauachensis, and Mymarothecium viatorum (Monogenoidea); Neoechinorhynchus buttnerae (Acanthocephala); Cucullanus colossomi (Nematoda); Perulernaea gamitanae (Lernaeidae); and Proteocephalidae larvae (Cestoda). A total of 8,136,252 parasites were collected from the examined fish. This is the first record of N. buttnerae, C. colossomi, N. janauachensis, M. viatorum, and Proteocephalidae for hybrid tambacu in Brazil. Ichthyophthirius multifiliis was the most prevalent parasite, whereas endohelminths were the less. A positive correlation was observed between number of I. multifiliis and total length and weight of fish, as well as between number of P. gamitanae and total length. The infection by I. multifiliis had association with the parasitism by Monogenoidea. Low water quality contributes to high parasitism of hybrid tambacu by ectoparasites, which, however, does not influence the relative condition factor of fish.
