Matteucinol combined with temozolomide inhibits glioblastoma proliferation, invasion, and progression: an in vitro, in silico, and in vivo study

Glioblastoma is the most prevalent and malignant brain tumor identified in adults. Surgical resection followed by radiotherapy and chemotherapy, mainly with temozolomide (TMZ), is the chosen treatment for this type of tumor. However, the average survival of patients is around 15 months. Novel approaches to glioblastoma treatment are greatly needed. Here, we aimed to investigate the anti-glioblastoma effect of the combination of matteucinol (Mat) (dihydroxyflavanone derived from Miconia chamissois Naudin) with the chemotherapeutic TMZ in vitro using tumor (U-251MG) and normal astrocyte (NHA) cell lines and in vivo using the chick embryo chorioallantoic membrane (CAM) assay. The combination was cytotoxic and selective for tumor cells (28 μg/mL Mat and 9.71 μg/mL TMZ). Additionally, the combination did not alter cell adhesion but caused morphological changes characteristic of apoptosis in vitro. Notably, the combination was also able to reduce tumor growth in the chick embryo model (CAM assay). The docking results showed that Mat was the best ligand to the cell death membrane receptor TNFR1 and to TNFR1/TMZ complex, suggesting that these two molecules may be working together increasing their potential. In conclusion, Mat-TMZ can be a good candidate for pharmacokinetic studies in view of clinical use for the treatment of glioblastoma.

Aspectos econômicos do uso da parte aérea in natura de mandioca (Manihot esculenta Crantz) para nutrição de cordeiros confinados e semiconfinados

RESUMO O objetivo deste trabalho foi avaliar a influência da inclusão da rama de mandioca na dieta de cordeiros confinados e semiconfinados, em substituição parcial de alimentos concentrados, como forma de redução de custos de produção. O experimento foi conduzido no Centro de Tecnologia de Ovinos (CTO), município de Campo Grande, MS. Foram utilizados 56 cordeiros do grupo genético Pantaneiro, sendo 28 machos e 28 fêmeas, com pesos médios de 18,84kg±2,02kg. As dietas recebidas foram: tratamento controle com ração comercial e tratamentos contendo a parte aérea de mandioca (PAM) em substituição de 10% dessa ração. Os machos confinados obtiveram ganho de peso médio diário de 210g e os semiconfinados, de 178g. No semiconfinamento, as fêmeas controle tiveram GMD de 120g e menor desempenho entre os grupos experimentais. A inclusão PAM acarretou uma redução de 31,29% para a mesma categoria animal. Entre os sistemas de produção, o semiconfinamento apresentou maior lucratividade média, 6,4%, e uma rentabilidade de 7,99% em relação ao sistema de confinamento, que obteve 0,04% de lucratividade e 0,87% de rentabilidade. Conclui-se que a inclusão da parte aérea da mandioca aumenta a eficiência econômica dos sistemas produtivos sem afetar o desenvolvimento dos animais em terminação.
ABSTRACT The scope of this study was to evaluate the inclusion of raw cassava aerial parts in the diet of lambs in partial substitution of rations, in order to reduce production costs. The experiment was conducted at Sheep Technology Centre (CTO) in Campo Grande City, MS. A sample of 56 lambs from Pantaneiro genetic group was used, 28 males and 28 females, average weight of 18.84kg±2.02kg. The inclusion of PAM decreased the costs by 31.29% for the same category of animal. Among the production systems the semi-feedlot, showed the highest average profitability of 6.4% and a return of 7.99%, compared to the feedlot system, which obtained 0.04% 0.87% profitability and return. In conclusion, the inclusion of the aerial part of cassava increases the economic efficiency of production systems without affecting the development of finishing animals.

Growth and reproduction of the mangrove crab Goniopsis cruentata (Latreille, 1803) (Crustacea: Decapoda: Grapsidae) in southeastern Brazil

Goniopsis cruentata é um caranguejo semi-terrestre comum em manguezais brasileiros e um importante recurso pesqueiro para comunidades tradicionais no nordeste do Brasil. Com o objetivo de contribuir com o conhecimento sobre a espécie, o presente estudo avaliou as curvas de crescimento em largura de carapaça e em peso, o crescimento relativo do peso pela largura da carapaça e a variação temporal dos índices gonadossomático e hepatossomático para a espécie. Um total de 524 caranguejos foram coletados em área de manguezal no município de Ubatuba, estado de São Paulo. Os parâmetros das curvas de crescimento e a longevidade (tmax) foram estimados para machos (CW∞=50,6 mm, WE∞=56,4 g, k=2,24, t0=0,003631502 ano-1, tmax=1,3 anos) e fêmeas (CW∞=50,7 mm, WE∞=58,8 g, k=2,50, t0=0,003247209 ano-1, tmax=1,2 anos). A idade no início da maturidade sexual foi de 0,23 anos para ambos os sexos. O modelo de crescimento em peso foi isométrico para indivíduos em estágios de desenvolvimento imaturos e alométrico negativo para adultos. A espécie exibiu reprodução contínua com picos reprodutivos nos meses de primavera e verão. A dinâmica de peso das gônadas e hepatopâncreas não estiveram claramente relacionadas. Os padrões de crescimento e reprodução indicaram que Goniopsis cruentata possui uma história de vida que prioriza a reprodução em detrimento da sobrevivência. A espécie exibiu algumas das maiores taxas de cresci mento e menores estimativas de longevidade relatados para espécies de braquiúros no Brasil.
Goniopsis cruentata is a common semi-terrestrial crab in Brazilian mangroves and an important fishery resource for traditional communities in the northeastern Brazilian coast. Aiming to contribute to the knowledge about the species, this study evaluated the carapace width and weight growth curves, the relative growth of weight versus carapace width, and the temporal variation of gonadosomatic and hepatosomatic indices for the species. A total of 524 crabs were collected in a mangrove area of Ubatuba municipality, state of São Paulo. The growth-curves parameters and longevity (tmax) were estimated for males (CW∞=50.6 mm, WE=56.4 g, k=2.24, t0=0.003631502 year-1, tmax=1.3 years) and females (CW∞=50.7 mm, WE∞=58.8 g, k=2.50, t0=0.003247209 year-1, tmax=1.2 years). The age at onset of sexual maturity was 0.23 years for both genders. The weight-growth model was isometric for the immature developmental stages and allometric negative for adults. The species exhibited a continuous reproduction, with breeding peaks in spring and summer months. The weight dynamics of gonads and hepatopancreas were not clearly related. The growth and reproductive patterns indicated that Goniopsis cruentata has a life-history that prioritizes reproduction instead of survival. The species exhibited some of the highest growth rates and lowest longevity estimates reported for brachyuran species in Brazil.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

O gênero Heteropterys é um dos maiores gêneros entre as Malpighiaceae. Entretanto, poucos estudos citológicos e palinológicos foram relatados. O presente trabalho descreveu pela primeira vez o número cromossômico, o padrão heterocromático, o comportamento meiótico, a viabilidade polínica e aspectos palinológicos de Heteropterys umbellata, uma espécie muito disseminada. Uma grande população brasileira foi estudada utilizando técnicas convencionais para o estudo de cromossomos meióticos e acetólise para acessar a morfologia polínica. A espécie apresentou 2n = 20 cromossomos, desenvolvimento meiótico normal e pólens viáveis. Grandes blocos de heterocromatina foram observados ao redor dos cen-trômeros. A coloração com DAPI foi positiva para a hetero-cromatina centromérica, enquanto marcas com CMA3 foram observadas somente em um sítio terminal de um par de cromossomos homólogos. Este resultado e a presença de um par de cromossomos associados a um nucléolo durante o paquíteno e diacinese sugerem a presença de somente um par de RONs. A análise palinológica revelou que os grãos de pólen são apolar, com 6 poros e colpóides associados a todos eles. O conteúdo polínico foi positivo para o teste de amido e a exina mostrou-se rugulada com poucas regiões psiladas.
The genus Heteropterys is one of the major genera in Malpighiaceae. However, few cytological and palynological studies were reported. The present work described for the first time the chromosome number, heterochromatin pattern, meiotic behavior, pollen viability and palynological aspects of Heteropterys ubellata, a very spread species. One large Brazilian population was evaluated using conventional techniques for meiotic studies and acetolyse to access the pollen morphology. The species showed 2n = 20 chromosomes, normal meiotic development and viable pollens. Great blocks of heterochromatin were observed around the centromers. DAPI staining was positive for centroeric heterochroatin, hile CMA3 ark as observed just at terinal regions of one pair of hoologues chromosomes. This result and the presence of one chromosome pair attached to the nucleoli during the pachytene and diakinesis suggested the presence of only one pair of NORs. Palynological analysis revealed that pollen grains are apolar, 6 porate and with colpoids associated to all pores. The pollen content was positive for the starch test, and the exine was rugulate with little psilate regions.

Composição química da cera epicuticular de biótipos de azevém resistente e suscetível ao glyphosate

Objetivou-se com este trabalho determinar a composição química da cera epicuticular dos biótipos de azevém (Lolium multiflorum) resistente e suscetível ao glyphosate, buscando relações entre suas características e a resistência dos biótipos ao herbicida. A cera epicuticular foi extraída e quantificada e os seus constituintes analisados por cromatografia em fase gasosa, acoplada a espectrômetro de massa (CG-EM). Para determinação da composição química, amostras de lâmina foliar foram retiradas 30 dias após a emergência das plantas, coletando-se a primeira folha com lígula totalmente visível. A quantidade de cera epicuticular extraída não diferiu entre os biótipos. Entre os compostos que constituem a cera epicuticular, os álcoois são os mais abundantes, sendo representados por apenas um composto: o hexacosan-1-ol (46,80% no biótipo resistente e 52,20% no biótipo suscetível). Ao comparar a polaridade da cera epicuticular dos biótipos de azevém, constatou-se que tanto no biótipo resistente quanto no suscetível a cera epicuticular apresentou mais de 50% de componentes polares (álcoois e aldeídos) em sua constituição, sendo esse valor igual a 69,80% no biótipo resistente e 64,94% no biótipo suscetível. Por meio da caracterização apresentada, pode-se afirmar que existem pequenas diferenças na cera epicuticular dos biótipos de azevém resistente e suscetível ao glyphosate; o biótipo resistente apresentou grau de polaridade pouco superior ao do biótipo suscetível, porém essa diferença não pode ser considerada marcante a ponto de determinar maior ou menor tolerância de um biótipo ou outro ao herbicida glyphosate.
The objective of this work was to determine the chemical composition of epicuticular wax of Italian ryegrass (Lolium multiflorum) biotypes resistant and susceptible to glyphosate, searching for relationships between their characteristics and the resistance of the biotypes to the product. The epicuticular wax was extracted and quantified, and its constituents analyzed by gas chromatography, coupled to mass spectrometer (CG-EM). To determine the chemical composition, leaf blade samples were obtained 30 days after plant emergence, the first leaf being collected with totally visible ligule. The amount of epicuticular wax did not differ between the biotypes. Alcohols are the most abundant among the compounds constituting the epiticular wax, represented by only one compound, hexacosan-1-ol (46.80% in the resistant biotype and 52.20% in the susceptible). When comparing the polarity of the epicuticular wax of the Italian ryegrass biotypes, it was verified that both in the resistant and susceptible biotypes, the epicuticular wax presented more than 50% of polar compounds (alcohols and aldehydes) in its constitution, with that value being equal to 69.80% in the resistant biotype and 64.94% in the susceptible. Thus, it can be stated that small differences exist in the epicuticular wax of Italian ryegrass biotypes resistant and susceptible to glyphosate. The resistant biotype presented a slightly higher polarity degree than the susceptible biotype but such difference cannot be considered sufficiently relevant to determine a greater or smaller tolerance of either biotype to glyphosate.

Objetivou-se com este trabalho avaliar a capacidade competitiva entre biótipos de azevém resistente e suscetível ao glyphosate, bem como a interferência deles, em diferentes densidades, sobre o crescimento de plantas de trigo. No momento da colheita, aos 50 dias após a emergência do azevém, avaliaram-se o número de perfilhos, a altura de plantas e a área foliar. Nessa mesma ocasião, coletou-se a parte aérea e as raízes das plantas de trigo e de azevém resistente e suscetível, determinando-se a seguir a massa seca desse material em partes separadas (raiz, caule e folhas). Com base nos dados avaliados, as seguintes variáveis para o trigo e para os biótipos de azevém foram calculadas: taxa de crescimento da cultura (TCC = MS A/Ndias), em que MS A é a massa seca da parte aérea e Ndias é o período em dias entre a emergência e a colheita das plantas; área foliar específica (SLA = Af/MSf), em que Af é a área foliar e MSf é a massa seca foliar; e o índice de área (IAF = Af/St), sendo St a superfície de solo, indicando qual a área de folhas por m² de solo. As características avaliadas altura de planta, massa seca e área foliar dos biótipos de azevém suscetível apresentaram menor tendência de redução e maior plasticidade fenotípica, com o incremento da densidade de plantas por área em relação aos biótipos resistentes. Com relação à competição dos biótipos de azevém com plantas de trigo, efeito negativo sobre a cultura também foi observado quando esta se encontrava sob interferência do biótipo suscetível. Conclui-se que o biótipo suscetível de azevém é mais competitivo que o resistente.
The objective of this work was to evaluate the competitiveness of ryegrass biotypes resistant and susceptible to glyphosate, as well as their interference in wheat growth at different densities. At harvesting, 50 days after emergence, tillering, height, and leaf area of ryegrass plants were evaluated and shoot and roots of ryegrass and wheat were collected and their root, stalk and leaf dry mass was determined. Based on the data, the following variables were evaluated for ryegrass and wheat: crop growth rate (TCC = MS A/Ndays), with MS A being shoot dry mass and Ndays the number of days between emergence and plant harvesting; specific leaf area (SLA = Af /MSf),with Af being the leaf area and MSf leaf dry mass; and leaf area index (IAF = Af/St), St being soil area, indicating leaf area per soil area. Plant height, dry mass and leaf area of the susceptible ryegrass showed smaller reductions and better phenotypic plasticity, in function of the increasing density per soil area, compared to the resistant one. The susceptible biotype showed to be more competitive and its interference in wheat development was more visible at lower densities. It was concluded that the susceptible ryegrass biotype is more competitive than the resistant one.

Comparison of behavioral and clinical effects of intravenous amitraz or romifidine administration in horses

Compararam-se os efeitos sedativos e clínicos em seis eqüinos tratados com romifidina (60mcg/kg) ou amitraz (0,4mg/kg), pela via intravenosa, durante 120 minutos. A romifidina promoveu ptose da cabeça (PC), reduziu a freqüência respiratória (FR) e o movimento da válvula íleo-cecal (MC) durante 120 minutos, provocou alteração postural (AP) por 90 minutos, induziu ptose palpebral (PP) e sedação (SD) por 60 minutos, diminuiu a motilidade intestinal (MI) e causou ptose labial (PL) durante 45 minutos. O amitraz reduziu a freqüência cardíaca (FR), FR, MI e MC e provocou PC, PP, PL, prolapso peniano (PP) e sedação com alteração postural por 120 minutos. Aos 90 minutos, o amitraz induziu a PC e a PP mais intensamente que a romifidina; o mesmo ocorreu entre 45 e 60 minutos para PL. A romifidina produziu sedação maior aos 10 minutos; para o amitraz, a sedação foi mais intensa entre 90 e 120 minutos. Conclui-se que a romifidina e o amitraz produzem sedação intensa por 60 e 120 minutos, respectivamente. A sedação e as alterações posturais induzidas pelo amitraz foram de início tardio, porém tiveram maior duração.
Clinical and behavioral effects of intravenous injection of romifidine (60mcg/kg) or amitraz (0.4mg/kg) were compared in six horses, over 120 min. Romifidine caused head ptosis (HP), reduced respiratory rate (RR) and ileo-cecal sphincter activity (IC) until 120 minutes; produced posture changes (PC) until 90 minutes, upper eyelid drop (ED) and sedation (SD) until 60 minutes, intestinal motion (IM) reduction and lower lip drop (LD) until 45 minutes. Amitraz reduced heart rate (HR), RR, IM and IC, and promoted HP, ED, LD, penis drop (PD) and sedation with posture changes until 120min. At 90min amitraz produced HP and ED more intensively than romifidine. The LD was similar between 45 and 60 minutes. Romifidine produced intense sedation at 10min while amitraz sedation was more significant between 90 and 120 minutes. It is concluded that both romifidine and amitraz can produce strong sedation until 60 and 120 minutes, respectively. Though amitraz sedation and posture changes had a later onset related to romifidine, these effects lasted longer.
