Lolium multiflorum grass is the major pollen allergen source in the southern region of Brazil, but most of its allergens remain poorly characterized. The aim of this study was to investigate antibody reactivity to L. multiflorum crude and carboxymethyl-ligand extracts in allergic patients and healthy individuals. Ion exchange carboxymethyl (CM) chromatography (CM-Sepharose) was used to isolate proteins (S2) from L. multiflorum crude extract (S1), which were assessed by SDS-PAGE. S1- and S2-specific IgE and IgG4 levels were measured by ELISA using sera from 55 atopic and 16 non-atopic subjects. Reactive polypeptide bands in S1 and S2 were detected by immunoblotting, and the most prominent bands in S2 were analyzed by mass spectrometry (MS-MS). Similar IgE and IgG4 levels were observed to both S1 (IgE median absorbance: 1.22; IgG4 median absorbance: 0.68) and S2 (IgE median absorbance: 1.26; IgG4 median absorbance: 0.85) in atopic subjects. S1 and S2 had positive correlations for IgE and IgG4 (IgE: r=0.9567; IgG4: r=0.9229; P<0.0001) levels. Homology between S1 and S2 was confirmed by IgE (84%) and IgG4 (83%) inhibition. Immunoblotting revealed that the 29-32 kDa band was recognized by 100% of atopic subjects in both S1 and S2. MS-MS analysis identified similarity profile to groups 1 and 5 grass allergens. This study revealed that carboxymethyl-ligand fraction played an important role for pollen allergy diagnosis by containing clinically relevant allergens and constituted a promising candidate for allergen-specific immunotherapy.

RESUMO: Avaliou-se neste trabalho o efeito da presença do capim-braquiarão (Urochloa brizantha) sobre o crescimento de cinco espécies arbóreas do bioma Mata Atlântica, em Seropédica-RJ. O estudo abrangeu cinco experimentos em delineamento inteiramente casualizado. Em cada experimento, testou-se em vasos o efeito da gramínea sobre o crescimento das espécies Cedrela fissilis, Guazuma ulmifolia, Schinus terebinthifolius, Sapindus saponaria e Hymenaea courbaril. Foram medidos mensalmente a altura, o diâmetro do coleto e a mortalidade das plantas. Aos 180 dias, avaliou-se também a massa seca de parte aérea e radicular das espécies arbóreas e da gramínea. Houve forte interferência do capim-braquiarão sobre o crescimento das cinco espécies estudadas. A redução de crescimento em altura e diâmetro do coleto chegou, respectivamente, a 32% e 29% em S. saponaria e a 26% e 44% em G. ulmifolia. A interferência da gramínea foi ainda maior sobre o acúmulo de massa seca, promovendo reduções na parte aérea que variaram de 48% em H. courbaril a 90% em G. ulmifolia e em S. terebinthifolius. A massa seca de raízes das espécies arbóreas reduziu entre 28% e 84% em relação ao controle sem a presença da gramínea. C. fissilis apresentou taxa de mortalidade de 83%, quando em convivência com U. brizantha. As demais espécies, à exceção de S. saponaria, também apresentaram mortalidade quando sob competição, variando de 15% a 30%. Este estudo evidencia a importância do controle de gramíneas em projetos de reflorestamento visando maiores ganhos em crescimento e sobrevivência das espécies arbóreas.

ABSTRACT: The effect of palisade grass (Urochloa brizantha) was evaluated on the growth of five tree species from the Atlantic Forest biome in Seropédica, RJ, Brasil. This study consisted of five experiments in a completely randomized design. The effect of grass on the growth of Cedrela fissilis, Guazuma ulmifolia, Schinus terebinthifolius, Sapindus saponaria, and Hymenaea courbaril was tested in pots at each experiment. Height, collar diameter, and plant mortality were measured monthly. Shoot and root dry matter of tree species and grass were evaluated at 180 days. A strong interference of palisade grass was observed on the growth of the five studied species. The reduction of growth in height and collar diameter reached 32 and 29% in S. saponaria and 26 and 44% in G. ulmifolia, respectively. Grass interference was even higher on dry matter accumulation, promoting reductions in the shoot that ranged from 48% in H. courbaril to 90% in G. ulmifolia and S. terebinthifolius. Root dry matter of tree species was reduced between 28 and 84% in relation to the control without the presence of grass. The species C. fissilis had a mortality rate of 83% in coexistence with U. brizantha. The other species, except S. saponaria, presented mortality from 15 to 30% under competition. This study evidences the importance of controlling grasses in reforestation projects aiming higher gains in growth and survival of tree species.

RESUMO: Gramíneas invasoras são um dos maiores obstáculos a serem superados em plantios de restauração. Assim, desenvolver técnicas eficientes e de baixo custo para superar esse obstáculo é um desafio para a ciência e a prática da restauração ecológica. O objetivo deste trabalho foi avaliar o efeito do coroamento com papelão sobre o crescimento de espécies arbóreas e os custos operacionais dessa técnica em um reflorestamento implantado no município de Seropédica-RJ. A vegetação predominante do local era de Andropogon bicornis L. Foi instalado um experimento em blocos casualizados com dois tratamentos e 12 repetições. Os tratamentos consistiram de coroamento com papelão e coroamento com enxada. Os papelões utilizados (tampa de embalagem de papelão para pizza de 50 cm x 50 cm) foram previamente tratados com solução preservativa à base de CuSO4. Os tratamentos foram aplicados a 11 espécies florestais do bioma Mata Atlântica: Anadenanthera colubrina var. cebil (Griseb.) Altschul, Citharexylum myrianthum Cham., Enterolobium contortisiliquum (Vell.) Morong, Eugenia brasiliensis Lam, Eugenia uniflora Lam, Handroanthus chrysotrichus (Mart. ex DC.) Mattos, Hymenaea courbaril L., Inga laurina (Sw.) Willd., Plinia cauliflora (Mart.) Kausel, Plathymenia reticulata Benth. e Triplaris americana L. Foram feitas avaliações de altura, diâmetro à altura do coleto (DAC) e taxa de sobrevivência aos 3, 9 e 15 meses após o plantio. Também foi realizada a quantificação dos custos de cada técnica de coroamento. Em geral, altura, DAC e taxas de crescimento não diferiram significativamente entre tratamentos de coroamento para todas as espécies avaliadas. O tratamento de coroamento com papelão apresentou maior taxa de sobrevivência de plantas (80%) em relação ao tratamento com coroamento com enxada (73%). O custo do coroamento com papelão durante 12 meses foi 40% inferior ao do coroamento com enxada. Esses resultados demonstram que a técnica de coroamento com papelão pode ser uma alternativa viável e de menor custo para substituir o coroamento com enxada em áreas de reflorestamento com espécies do bioma Mata Atlântica.

ABSTRACT: Invasive grasses are one of the biggest obstacles to be overcome in restoration plantations. Thus, developing efficient and low-cost techniques to overcome this obstacle is a challenge for science and the ecological restoration practice. The objective of this study was to evaluate the effect of the use of cardboard for the crowning operation on the growth of forest tree species from the Atlantic Forest biome and on the operating costs of this technique in a reforestation implemented in Seropédica, Rio de Janeiro state. The predominant vegetation of the area was of Andropogon bicornis L. An experiment was installed in randomized blocks, with two treatments and 12 replications. The treatments consisted of crowning with cardboard and crowning with hoe. The used cardboard (50 x 50 cm) were pre-treated with a preservative solution with CuSO4. Treatments were applied to 11 forest species from the Atlantic Forest biome: Anadenanthera colubrina var. cebil (Griseb.) Altschul, Citharexylum myrianthum Cham., Enterolobium contortisiliquum (Vell.) Morong, Eugenia brasiliensis Lam, Eugenia uniflora Lam, Handroanthus chrysotrichus (Mart. ex DC.) Mattos, Hymenaea courbaril L., Inga laurina (Sw.) Willd., Plinia cauliflora (Mart.) Kausel, Plathymenia reticulata Benth. e Triplaris americana L. Evaluations were carried out as for height, diameter at collar height (DCH) and survival rate, 6, 12 and 18 months after planting. The costs of each crowning method were also quantified. Overall, the height, DCH and growth rates did not differ between the crowning treatments for the species evaluated. However, the cardboard crowning treatment showed a higher survival rate (80%) of plants, compared with the hoe crowning (73%). The cost of the crowning with cardboard over a 12-month period was 40% lower than that of the crowning with hoe. These results show that the cardboard crowning technique can be a viable and a cost effective alternative to replace the crowning with hoe in reforestation areas with species from the Atlantic Forest biome.

Estimates of occult hepatitis B virus (HBV) infection prevalence varies among different studies depending on the prevalence of HBV infection in the study population and on the sensitivity of the assay used to detect HBV DNA. We investigated the prevalence of occult HBV infection in cirrhotic patients undergoing liver transplantation in a Brazilian referral center. Frozen liver samples from 68 adults were analyzed using a nested polymerase chain reaction assay for HBV DNA. The specificity of the amplified HBV sequences was confirmed by direct sequencing of the amplicons. The patient population comprised 49 (72.1%) males and 19 (27.9%) females with a median age of 53 years (range=18-67 years). Occult HBV infection was diagnosed in three (4.4%) patients. The etiologies of the underlying chronic liver disease in these cases were alcohol abuse, HBV infection, and cryptogenic cirrhosis. Two of the patients with cryptic HBV infection also presented hepatocellular carcinoma. Markers of previous HBV infection were available in two patients with occult HBV infection and were negative in both. In conclusion, using a sensitive nested polymerase chain reaction assay to detect HBV DNA in frozen liver tissue, we found a low prevalence of occult HBV infection in cirrhotic patients undergoing liver transplant, probably due to the low prevalence of HBV infection in our population.

O potencial evocado auditivo de tronco encefálico (BAEP) é um teste eletrodiagnóstico que detecta e registra a atividade elétrica do sistema auditivo desde a cóclea até o tronco encefálico, gerada após a emissão de um estímulo acústico na orelha externa. O objetivo deste estudo é obter dados normativos para o BAEP em cães da raça Dálmata para usá-lo para detecção de surdez e de outras alterações neurológicas. BAEP foi obtido de 30 cães da raça Dálmata para um estudo normativo no Brasil. As latências médias para as ondas I, III e V foram 1,14ms (±0,09); 2,62ms (±0,10) e 3,46ms (±0,14), respectivamente. A média das latências dos intervalos I-III, III-V e I-V foi 1,48ms (±0,17); 0,84ms (±0,12) e 2,31ms (±0,18), respectivamente. Alteração unilateral foi observada em 16,7% dos animais, e surdez bilateral foi observada em um cão. Os dados normativos obtidos neste trabalho são compatíveis com outros dados já publicados. Segundo a revisão realizada, este é o primeiro relato da ocorrência de surdez em cães da raça Dálmata no Brasil.

The brain stem auditory-evoked potential (BAEP) is an electrophysiologic test that detects and records the electrical activity in the auditory system from cochlea to midbrain, generated after an acoustic stimulus applied to the external ear. The aim of this study is to obtain normative data for BAEP in Dalmatian dogs in order to apply this to the evaluation of deafness and other neurologic disorders. BAEP were recorded from 30 Dalmatian dogs for a normative Brazilian study. Mean latencies for waves I, III, and V were 1.14 (±0.09), 2.62 (±0.10), and 3.46 (±0.14) ms, respectively. Mean inter-peak latencies for I-III, III-V, and I-V intervals were 1.48 (±0.17), 0.84 (±0.12), and 2.31 (±0.18) ms, respectively. Unilateral abnormalities were found in 16.7% of animals and bilateral deafness was seen in one dog. The normative data obtained in this paper is compatible with other published data. As far as we know this is the first report of deafness occurrence in Dalmatian dogs in Brazil.

Potencial evocado auditivo é um método eletrodiagnóstico que permite avaliação funcional das vias auditivas desde a orelha média até o tronco encefálico. O uso desse teste em medicina veterinária é pouco difundido no Brasil. O presente artigo relata o uso do potencial evocado auditivo para detecção de deficiência auditiva unilateral em um gato com síndrome vestibular periférica secundária a otite média.

The brainstem auditory evoked potential is an electrodiagnostic test that allows a functional assessment of the auditory pathways from the middle ear to the brainstem. This test, in veterinary medicine, is not commonly used in Brazil. This paper reports the use of auditory evoked potential for deafness detection in a cat with unilateral peripheral vestibular syndrome secondary to otitis media.

O objetivo deste trabalho foi realizar estudo normativo dos potenciais evocados auditivos obtidos de 34 cães sem raça definida e avaliar a influência da idade nos resultados obtidos. Os animais foram divididos em dois grupos de diferentes faixas etárias e o potencial evocado auditivo foi realizado com um estímulo de 85dB. O grupo 1 incluiu 16 cães com idades entre um e oito anos e o grupo 2, 18 animais com idades acima de oito anos. O comprimento e o diâmetro da cabeça foram mensurados e não houve diferença estatística entre os dois grupos. No grupo 1, as médias das latências das ondas I, III e V foram: 1,13; 2,64 e 3,45ms; e dos intervalos I-III, III-V e I-V foram 1,51; 0,81 e 2,32ms, respectivamente. No grupo 2, as médias das latências das ondas I, III e V foram 1,15; 2,62 e 3,55ms; e dos intervalos I-III, III-V e I-V foram 1,47; 0,93 e 2,40ms, respectivamente. As latências observadas neste estudo foram semelhantes a estudos prévios realizados por outros autores. Nos grupos etários analisados e nas condições preestabelecidas deste estudo, a idade influenciou a latência da onda V e, consequentemente, modificou os intervalos III-V e I-V, portanto deve-se considerar essa variante na interpretação dos resultados do BAEP em cães.

The aim of this paper was to obtain normative data of auditory evoked potentials from 34 mixed breed dogs and evaluate the age influence. The animals were divided in two groups of different ages and auditory evoked potential was performed with a 85dB stimulus intensity. Group 1 included 16 dogs between 1 and 8 years of age, and group 2 included 18 dogs with over 8 years of age. The length and head diameter were measured and there was no statistical difference between the two groups. In group 1, mean latencies of waves I, III, and V were 1.13; 2.64, and 3.45ms, and the intervals I-III, III-V, and I-V were 1.51; 0.81, and 2.32 ms, respectively. In group 2, the mean latencies of waves I, III and V were 1.15, 2.62, and 3.55ms, and the intervals I-III, III-V, and I-V were 1.47, 0.93, and 2.40ms, respectively. The latencies observed in this study were similar to previous studies conducted by other authors. It was observed that significant differences were present for wave V and intervals III-V and I-V latencies when comparing groups with different ages, consequently this characteristic must be considered during BAEP result interpretation.

We investigated whether liver injury by dual exposure to ethanol and carbon tetrachloride (EtOH + CCl4) for 15 weeks would persist after hepatotoxic agents were removed (EtOH + CCl4/8wR). After 15 weeks of hepatic injury with ethanol (5.5%, m/v) and carbon tetrachloride (0.05, mL/kg, ip), 5 of 11 female Wistar rats were sacrificed. The other 6 rats were maintained for an additional 8 weeks without hepatotoxic agents. Ultrasonography showed increased liver echogenicity and dilation of portal vein caliber in both groups (EtOH + CCl4: 0.22 ± 0.01 cm, P < 0.001; EtOH + CCl4/8wR: 0.21 ± 0.02 cm, P < 0.01) vs control (0.16 ± 0.02 cm). Histopathology showed regenerative nodules in both experimental groups. Histomorphometry revealed increased fibrosis content in both groups (EtOH + CCl4: 12.6 ± 2.64%, P < 0.001; EtOH + CCl4/8wR: 10.4 ± 1.36%, P < 0.05) vs control (2.2 ± 1.21%). Collagen types I and III were increased in groups EtOH + CCl4 (collagen I: 2.5 ± 1.3%, P < 0.01; collagen III: 1.3 ± 0.2%, P < 0.05) and EtOH + CCl4/8wR (collagen I: 1.8 ± 0.06%, P < 0.05; collagen III: 1.5 ± 0.8%, P < 0.01) vs control (collagen I: 0.38 ± 0.11%; collagen III: 0.25 ± 0.06%). Tissue transglutaminase increased in both groups (EtOH + CCl4: 66.4 ± 8%, P < 0.01; EtOH + CCl4/8wR: 58.8 ± 21%, P < 0.01) vs control (7.9 ± 0.8%). Cirrhosis caused by the association of CCl4-EtOH remained for at least 8 weeks after removal of these hepatotoxic agents. Ultrasound images can be a useful tool to evaluate advanced hepatic alterations.

Vinte e nove pintos SPF de um dia foram inoculados com o vírus da doença infecciosa da bursa de Fabricius (VDIB) para avaliar a ocorrência precoce de apoptose e a expressão da proteína viral 2 (VP2) e da enzima gliceraldeído fosfato dehidrogenase (GAPDH). Os animais foram distribuídos em cinco grupos: 1-controle; e 2 a 5- com 24, 48, 72 e 96 horas pós-inoculação, respectivamente. Fragmentos da bursa de Fabricius foram colhidos para processamento histológico e extração de RNA. Lâminas coradas em HE e TUNEL (marcação in situ da fragmentação do genoma com transferase terminal de deoxinucleotídeo) foram utilizadas na morfometria do índice apoptótico. Amostras de mRNA foram testadas para a expressão dos genes VP2 e GAPDH utilizando-se transcrição reversa e RT-PCR. Utilizou-se um kit SYBR GREEN PCR, e a reação foi desenvolvida em ABI Prism 7000 SDS. Os índices apoptóticos cresceram progressivamente indicando uma relação na atrofia bursal causada pelo VDIB. Paralelamente, os resultados da PCR em tempo real demonstraram queda da carga viral nas células linfóides da bursa nos diferentes intervalos de tempo do experimento. Esses resultados sugerem um papel protetor da apoptose na diminuição da replicação viral.

Twenty-nine SPF 1-day-old chicks were inoculated with infectious bursal disease virus (IBDV) to evaluate early apoptosis and the expression of viral protein 2 (VP2) and glyceraldehyde-3-phosphate dehydrogenease (GAPDH). Five groups were formed: G1-control -and G2 to G5, - 24, 48, 72 and 96 hours post inoculation, respectively. Half of each BF was fixed and processed by routine techniques. To quantify apoptosis, 5µm-thick sections were stained with HE and submitted to TUNEL (terminal transferase UDP nick end labeling) technique. mRNA was extracted from pooled samples of 3 animals/group and used for the expression of VP2 and GADPH genes using the reverse transcription and real-time polymerase chain reaction (RT-PCR). A SYBR GREEN PCR kit was used and the reaction was carried out in an ABI Prism 7000 SDS. Apoptotic indexes progressively increased indicating a role of IBDV in inducing hypotrophy of the BF. Also, it was showed that as long as apoptosis increased, viral protein expression decreased, which suggests that apoptosis plays a role as a defense mechanism against viral replication.

Foram determinados os índices de infestação por moscas-das-frutas em seis cultivares de Coffea arabica L. nos sistemas com e sem arborização, sob manejo orgânico. O delineamento experimental foi inteiramente casualizado com parcelas sub-subdivididas e quatro repetições. Uma amostra de 250 g de frutos maduros por parcela foi colhida em maio de 2005. As cultivares ‘Icatu Amarelo’ e ‘Catucaí Amarelo’ mostraram menor susceptibilidade aos tefritídeos, nos dois sistemas de cultivo. Quanto aos lonqueídeos, ‘Oeiras’, ‘Catucaí Amarelo’ e ‘Catuaí Vermelho’ foram as cultivares menos susceptíveis no sistema arborizado, não havendo diferença entre as cultivares no sistema sem arborização. Dos frutos foram obtidas as seguintes espécies de Tephritidae: Ceratitis capitata (Wiedemann), Anastrepha fraterculus (Wiedemann) e A. sororcula Zucchi (Tephritidae). Os lonqueídeos foram representados por: Neosilba bifida Strikis & Prado, N. certa (Walker), N. glaberrima (Wiedemann), N. pendula (Bezzi), N. pseudopendula (Korytkowski & Ojeda), Dasiops rugifrons Hennig, Neosilba n.sp.10 e Neosilba n.sp.14.

The infestation indices by fruit flies were determined for six cultivars of Coffea arabica L. in shaded and unshaded systems under organic management. The experiment was set in a completely randomized design with a split-split-plot arrangement and four replicates. A 250g-sample of maturing fruits per plot was harvested in May 2005. The cultivars ‘Icatu Amarelo’ and ‘Catucaí Amarelo’ were the least susceptible to attack by tephritids in both systems. As for lonchaeids, ‘Oeiras’, ‘Catucaí Amarelo’ and ‘Catuaí Vermelho’ were the least susceptible cultivars in the shaded system, and there was no difference among the cultivars in the unshaded system. The following tephritid species were obtained: Ceratitis capitata (Wiedemann), Anastrepha fraterculus (Wiedemann) and A. sororcula Zucchi (Tephritidae). Lonchaeids were represented by Neosilba bifida Strikis & Prado, N. certa (Walker), N. glaberrima (Wiedemann), N. pendula (Bezzi), N. pseudopendula (Korytkowski and Ojeda), Dasiops rugifrons Hennig, Neosilba n.sp.10 and Neosilba n.sp.14.

As moscas frugívoras vêm assumindo grande importância econômica em cafezais por provocarem queda prematura de frutos e redução significativa da qualidade da bebida. O café é um repositório natural de espécies de Tephritidae que infestam frutas de importância econômica. Este estudo avaliou a influência dos sistemas de café sombreado e a pleno sol, sob manejo orgânico, na infestação natural de frutos de Coffea arabica L. Icatu Amarelo por moscas frugívoras. Um experimento inteiramente casualizado foi instalado em Valença, RJ, com dois tratamentos (café sombreado com Musa sp. e Erithrina verna Vell. e solteiro a pleno sol) e quatro repetições. Uma amostra de 1 kg de frutos maduros por parcela foi colhida em março de 2004 para avaliar a infestação e identificar as espécies infestantes. O índice médio de infestação dos frutos foi significativamente maior no sistema sombreado. Quatro espécies de Tephritidae (Ceratitis capitata (Wiedemann), Anastrepha fraterculus (Wiedemann), Anastrepha obliqua (Macquart) e Anastrepha sororcula Zucchi) e seis de Lonchaeidae (Neosilba pendula (Bezzi), Neosilba pseudopendula (Korytkowski and Ojeda), Neosilba certa (Walker), Neosilba glaberrima (Wiedemann), Neosilba n.sp.9 e Neosilba n.sp.10) foram obtidas dos frutos de café. Nove espécies de parasitóides foram identificadas, das quais seis pertencem a Braconidae (Asobara anastrephae (Muesebeck), Doryctobracon areolatus (Szépligeti), Doryctobracon brasiliensis (Szépligeti), Utetes anastrephae (Viereck), Opius bellus Gahan e Opius sp.) e três a Figitidae (Aganaspis pelleranoi (Brèthes), Dicerataspis flavipes (Kieffer) e Odontosema anastrephae Borgmeier). A percentagem de parasitismo total foi de 8,8% e 12,4% em café sombreado e a pleno sol, respectivamente.

Frugivorous flies have been acquiring great economic importance in coffee crop because they cause premature dropping of coffee beans and significantly decrease the quality of the beverage. The coffee plant is also a natural reservoir of Tephritidae species that infest fruits of economic importance. This study evaluated the influence of the shaded and unshaded coffee systems, under organic management, on the natural infestation of fruits of Coffea arabica L. var. Icatu Amarelo by frugivorous flies. An experiment in completely randomized design was carried out in Valença, RJ, Brazil, with two treatments (shaded coffee with Musa sp. and Erithrina verna Vell. and unshaded coffee monoculture) and four replicates. A 1kg-sample of maturing fruits per plot was harvested in Mach 2004 to evaluate infestation and identify the flies. The mean infestation index was significantly higher in the shaded coffee system. Four species of Tephritidae (Ceratitis capitata (Wiedemann), Anastrepha fraterculus (Wiedemann), Anastrepha obliqua (Macquart) and Anastrepha sororcula Zucchi), and six of Lonchaeidae (Neosilba pendula (Bezzi), Neosilba pseudopendula (Korytkowski and Ojeda), Neosilba certa (Walker), Neosilba glaberrima (Wiedemann), Neosilba n.sp.9 and Neosilba n.sp.10) were recovered from coffee fruits. Nine parasitoid species were obtained, six belonging to Braconidae (Asobara anastrephae (Muesebeck), Doryctobracon areolatus (Szépligeti), Doryctobracon brasiliensis (Szépligeti), Utetes anastrephae (Viereck), Opius bellus Gahan and Opius sp.), and three to Figitidae (Aganaspis pelleranoi (Brèthes), Dicerataspis flavipes (Kieffer) and Odontosema anastrephae Borgmeier). The total percent parasitism was 8.8% and 12.4% in the shaded and unshaded coffee systems, respectively.

Desenvolveu-se a técnica de transcrição reversa - reação em cadeia pela polimerase para detectar o vírus da hepatite do camundongo em tecido hepático. Para se eliminar possíveis falhas na reação de amplificação RT-PCR usou-se um segundo ciclo de amplificação com primers internos para confirmar a especificidade e aumentar a sensibilidade do teste. Após a optimização do protocolo, descreve-se a detecção da amplificação específica da seqüência-alvo em camundongos de 18 das 20 colônias examinadas.

A reverse transcriptase polymerase chain reaction (RT-PCR) to detect mouse hepatitis virus (MHV) in hepatic tissue was developed. To circumvent possible failures in RT-PCR amplifications, a second round of PCR with internal primers was used to confirm the specificity and increase the sensitivity of the test. Using this method specific amplification of MHV sequences was observed in 18 out of 20 mouse colonies examined.

Um sistema de ELISA foi utilizado para se estudar a prevalência do herpesvirus eqüino 1 (HVE-1) em plantéis de cavalos brasileiros vacinados e não vacinados. Amostras de soros foram coletadas de éguas gestantes assintomáticas, antes e após o parto e de seus respectivos potros recém-nascidos. O antígeno foi preparado de uma fração viral purificada (HVE-1 amostra Ab1). O ponto de corte (absorvância de 0,3516) foi determinado considerando o limite máximo para os valores negativos. Esse valor determinou sensibilidade de 100% e especificidade de 94,7%, utilizando um banco de soros de éguas gestantes e fetos. No estudo observou-se alta correlação entre o ELISA e a soro-neutralização para detecção de anticorpos específicos contra o EHV-1.

An ELISA system was applied to study the prevalence of EHV-1 in vaccinated and non-vaccinated Brazilian horses. Serum samples were taken from non-symptomatic pregnant mares, before and after the parturition, and from their neonatal foals. The antigen was prepared from a purified virus fraction (HVE-1 AB1 strain). The ELISA absorbance of 0.3516 was determined as the threshold of negative values. This cut-off level was defined from a selected interval, which determined a sensibility of 100% and a specificity of 94.7%, from a serum collection obtained from pregnant mares and fetuses. In this study, a strong correlation was found between the ELISA and serum neutralization for specific EHV-1 antibody detection.

A técnica da reação em cadeia pela DNA polimerase (PCR) foi usada para a amplificação rápida de um fragmento de 456bp da região única curta (Us) do genoma do BHV-1. Iniciadores de 18pb do gene da ORF1 foram usados para a amplificação das amostras-padrão e brasileiras. Uma amplificação clássica não foi bem sucedida. A amplificação foi obtida quando se escolheu uma região com baixa concentração de GC no DNA do BHV-1 e através da desnaturação térmica (95° C para 5min) seguida de ciclos térmicos (94° C por 1min e 30seg; 52° C por 1min; 72° C por 1min e 30seg; e ainda por 35 ciclos de 94° C por 1min; 52° C por 1min; 72° C por 1min e 30seg; usando um tempo de extensão final de 72° C por 5min). A clivagem com Pst I confirmou a especificidade do fragmento da ORF 1 do BHV-1. A amplificação do fragmento em todas as amostras testadas sugere que a região é fortemente conservada no genoma do BHV-1. Este PCR poderá detectar rapidamente amostras clínicas, sendo sensível e específico para diagnosticar infecções pelo BHV-1.
