Rheumatoid arthritis (RA), psoriatic arthritis (PsA), and ankylosing spondylitis (AS) are inflammatory diseases with different bone remodeling patterns. Fibroblast-like synoviocytes (FLS) are cells involved in the transition from an acute and reparable phase to a chronic and persistent stage in these diseases. The distinction of joint phenotypes involves inflammatory cytokines such as tumor necrosis factor alpha (TNF-α), interleukin (IL)-17, and IL-22 directly or through key signaling pathways such as Wnt. To evaluate the role of FLS as the source of Wnt antagonists (sFRP3/FRZB and Dkk1) in the synovia, levels of TNF- α, IL-17, IL-22, Dkk1, and sFRP3 were measured by ELISA directly in the synovial fluid of patients with RA, PsA, or AS. Dkk1 and sFRP3 were also measured in the FLS culture supernatants after different inflammatory stimulus. sFRP3 and Dkk1 are constitutively expressed by FLS. IL-22 and sFRP3 were positively correlated (r=0.76; P<0.01) in synovial fluid. The stimulation of FLS with IL-22, but not TNF-alpha and IL-17, increased the production of sFRP3. No stimulus altered the basal expression of Dkk1. These results showed, for the first time, the ability of IL-22 to increase the expression of sFRP3/FRZB by human FLS in both in vitro and ex vivo models. This finding linked IL-22 to local inhibition of Wnt signaling and possibly to blockade of osteogenesis. Furthermore, FLS presented as a source of this inhibitor in synovial fluid, assigning to this cell a bone injury mechanism.

Resumo Objetivo: Este estudo com acompanhamento de longo prazo visou a avaliar o quadro clínico, os achados laboratoriais, o perfil histológico, os tratamentos e os resultados de crianças e adolescentes com hepatite autoimune. Métodos: Foram analisados os prontuários médicos de 828 crianças e adolescentes com HAI. Foi usado um questionário para coletar os dados anônimos sobre o quadro clínico, os achados bioquímicos e histológicos e os tratamentos. Resultados: De todos os pacientes, 89,6% tinham hepatite autoimune-1 e 10,4% hepatite autoimune-2. O sexo feminino foi predominante nos dois grupos. A idade média no início dos sintomas foi 111,5 (6; 210) e 53,5 (8; 165) meses nos pacientes com hepatite autoimune-1 e hepatite autoimune-2, respectivamente. Foi observado início clínico agudo em 56,1% e 58,8% e sintomas insidiosos em 43,9% e 41,2% dos pacientes com hepatite autoimune-1 e hepatite autoimune-2, respectivamente. A probabilidade de insuficiência hepática foi 1,6 vezes maior para hepatite autoimune-2; 3,6% e 10,6% dos pacientes com hepatite autoimune-1 e hepatite autoimune-2, respectivamente, apresentaram insuficiência hepática fulminante; o risco foi 3,1 vezes maior para hepatite autoimune-2. Os níveis de gamaglobulina e imunoglobulina G foram significativamente maiores nos pacientes com hepatite autoimune-1, ao passo que os níveis de imunoglobulina A e C3 foram menores em pacientes com hepatite autoimune-2; 22,4% dos pacientes apresentaram cirrose e a remissão bioquímica foi atingida em 76,2%. A taxa de sobrevida atuarial foi de 93,0%. Um total de 4,6% pacientes foram submetidos a transplante de fígado e 6,9% morreram (hepatite autoimune-1: 7,5%; hepatite autoimune-2: 2,4%). Conclusões: Nesta grande série clínica de crianças e adolescentes brasileiros, a hepatite autoimune-1 foi mais frequente e os pacientes com hepatite autoimune-2 mostraram maiores taxas de remissão da doença com respostas mais rápidas aos tratamentos. Os pacientes com hepatite autoimune-1 apresentaram maior risco de óbito.

Abstract Objective: This large study with a long-term follow-up aimed to evaluate the clinical presentation, laboratory findings, histological profile, treatments, and outcomes of children and adolescents with autoimmune hepatitis. Methods: The medical records of 828 children and adolescents with autoimmune hepatitis were reviewed. A questionnaire was used to collect anonymous data on clinical presentation, biochemical and histological findings, and treatments. Results: Of all patients, 89.6% had autoimmune hepatitis-1 and 10.4% had autoimmune hepatitis-2. The female sex was predominant in both groups. The median age at symptom onset was 111.5 (6; 210) and 53.5 (8; 165) months in the patients with autoimmune hepatitis 1 and autoimmune hepatitis-2, respectively. Acute clinical onset was observed in 56.1% and 58.8% and insidious symptoms in 43.9% and 41.2% of the patients with autoimmune hepatitis-1 and autoimmune hepatitis-2, respectively. The risk of hepatic failure was 1.6-fold higher for autoimmune hepatitis-2. Fulminant hepatic failure occurred in 3.6% and 10.6% of the patients with autoimmune hepatitis-1 and autoimmune hepatitis-2, respectively; the risk was 3.1-fold higher for autoimmune hepatitis-2. The gamma globulin and immunoglobulin G levels were significantly higher in autoimmune hepatitis-1, while the immunoglobulin A and C3 levels were lower in autoimmune hepatitis-2. Cirrhosis was observed in 22.4% of the patients; biochemical remission was achieved in 76.2%. The actuarial survival rate was 93.0%. A total of 4.6% underwent liver transplantation, and 6.9% died (autoimmune hepatitis-1: 7.5%; autoimmune hepatitis-2: 2.4%). Conclusions: In this large clinical series of Brazilian children and adolescents, autoimmune hepatitis-1 was more frequent, and patients with autoimmune hepatitis-2 exhibited higher disease remission rates with earlier response to treatment. Patients with autoimmune hepatitis-1 had a higher risk of death.

The aim of this study was to evaluate the effects of sodium hyaluronate (HY), single-walled carbon nanotubes (SWCNTs) and HY-functionalized SWCNTs (HY-SWCNTs) on the behavior of primary osteoblasts, as well as to investigate the deposition of inorganic crystals on titanium surfaces coated with these biocomposites. Primary osteoblasts were obtained from the calvarial bones of male newborn Wistar rats (5 rats for each cell extraction). We assessed cell viability using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide assay and by double-staining with propidium iodide and Hoechst. We also assessed the formation of mineralized bone nodules by von Kossa staining, the mRNA expression of bone repair proteins, and the deposition of inorganic crystals on titanium surfaces coated with HY, SWCNTs, or HY-SWCNTs. The results showed that treatment with these biocomposites did not alter the viability of primary osteoblasts. Furthermore, deposition of mineralized bone nodules was significantly increased by cells treated with HY and HY-SWCNTs. This can be partly explained by an increase in the mRNA expression of type I and III collagen, osteocalcin, and bone morphogenetic proteins 2 and 4. Additionally, the titanium surface treated with HY-SWCNTs showed a significant increase in the deposition of inorganic crystals. Thus, our data indicate that HY, SWCNTs, and HY-SWCNTs are potentially useful for the development of new strategies for bone tissue engineering.

A estomatite protética é uma condição inflamatória que ocorre em usuários de prótese total e está frequentemente associada a leveduras do gênero Candida, Os perfis de suscetibilidade a antifúngicos têm sido extensivamente estudados em pacientes com candidíase ou em indivíduos imunossuprimidos, mas não em portadores sadios de Candida. No presente estudo, fluconazol, itraconazol, voriconazol, terbinafina e 5-flucitosina foram testados contra 109 isolados orais de Candida spp. Todos os agentes antifúngicos mostraram-se eficazes contra as amostras avaliadas, exceto a Terbinafina. O presente trabalho pode fornecer dados epidemiológicos com relação à susceptibilidade a antifúngicos de Candida spp em indivíduos com saúde oral.

Denture stomatitis is an inflammatory condition that occurs in denture wearers and is frequently associated with Candida yeasts. Antifungal susceptibility profiles have been extensively evaluated for candidiasis patients or immunosupressed individuals, but not for healthy Candida carriers. In the present study, fluconazole, itraconazole, voriconazole, terbinafine and 5-flucytosin were tested against 109 oral Candida spp. isolates. All antifungal agents were effective against the samples tested except for terbinafine. This work might provide epidemiological information about Candida spp. drug susceptibility in oral healthy individuals.

Foram determinados os índices de infestação por moscas-das-frutas em seis cultivares de Coffea arabica L. nos sistemas com e sem arborização, sob manejo orgânico. O delineamento experimental foi inteiramente casualizado com parcelas sub-subdivididas e quatro repetições. Uma amostra de 250 g de frutos maduros por parcela foi colhida em maio de 2005. As cultivares ‘Icatu Amarelo’ e ‘Catucaí Amarelo’ mostraram menor susceptibilidade aos tefritídeos, nos dois sistemas de cultivo. Quanto aos lonqueídeos, ‘Oeiras’, ‘Catucaí Amarelo’ e ‘Catuaí Vermelho’ foram as cultivares menos susceptíveis no sistema arborizado, não havendo diferença entre as cultivares no sistema sem arborização. Dos frutos foram obtidas as seguintes espécies de Tephritidae: Ceratitis capitata (Wiedemann), Anastrepha fraterculus (Wiedemann) e A. sororcula Zucchi (Tephritidae). Os lonqueídeos foram representados por: Neosilba bifida Strikis & Prado, N. certa (Walker), N. glaberrima (Wiedemann), N. pendula (Bezzi), N. pseudopendula (Korytkowski & Ojeda), Dasiops rugifrons Hennig, Neosilba n.sp.10 e Neosilba n.sp.14.

The infestation indices by fruit flies were determined for six cultivars of Coffea arabica L. in shaded and unshaded systems under organic management. The experiment was set in a completely randomized design with a split-split-plot arrangement and four replicates. A 250g-sample of maturing fruits per plot was harvested in May 2005. The cultivars ‘Icatu Amarelo’ and ‘Catucaí Amarelo’ were the least susceptible to attack by tephritids in both systems. As for lonchaeids, ‘Oeiras’, ‘Catucaí Amarelo’ and ‘Catuaí Vermelho’ were the least susceptible cultivars in the shaded system, and there was no difference among the cultivars in the unshaded system. The following tephritid species were obtained: Ceratitis capitata (Wiedemann), Anastrepha fraterculus (Wiedemann) and A. sororcula Zucchi (Tephritidae). Lonchaeids were represented by Neosilba bifida Strikis & Prado, N. certa (Walker), N. glaberrima (Wiedemann), N. pendula (Bezzi), N. pseudopendula (Korytkowski and Ojeda), Dasiops rugifrons Hennig, Neosilba n.sp.10 and Neosilba n.sp.14.

Desde 2000, diversos casos de infecção e doença por Macrorhabdus ornithogaster (megabacteria) foram diagnosticados no Setor de Doenças das Aves (Escola de Veterinária da UFMG). A doença clínica foi caracterizada por emagrecimento, prostração, perda do apetite, caquexia e morte, em curso crônico, embora com forma mais aguda em canários e periquitos. O microrganismo grande, em forma de bastão, visível a partir de 100 aumentos sem e com coloração, pode também ser detectado em aves de aspecto clínico normal, principalmente galinhas, perus, codornas e pombos. Em emas (Rhea), avestruzes (Struthio camelus), canários, mandarins, galinhas da Angola (Numida meleagris) e periquitos Australianos (Melopsittacus undulatus), a severidade da doença foi sempre maior, ocasionando até 100% de mortalidade em alguns plantéis. Na maioria das espécies a doença foi detectada em aves com endo e/ou ectoparasitismo. O cultivo de M. ornithogaster foi obtido em meio sólido (ágar para fungos patogênicos) e subcultivado em meio líquido (thioglicolato), do proventriculo de galinha, galinha da Angola, perdiz de chuckar e canário. O resultado mais surpreendente na microscopia de M. ornithogaster foi a presença de motilidade, detectada tanto de cultivos in vitro quanto de preparações úmidas de in vivo. Diferenças nos aspectos das colônias foram notadas entre os isolados. Infecções experimentais em galinha (SPF) e codorna japonesa permitiram a detecção do organismo nos proventrículos das aves de aspecto normal. Nas codornas, à necropsia notaram-se hemorragias hepáticas. A infecção experimental em camundongos via intraperitoneal resultou em 100% de mortalidade, também com lesões hepáticas. Aspectos do cultivo, a importância da doença, as espécies de aves susceptíveis e seu papel na epidemiologia são discutidos.

Since 2000, Macrorhabdus ornithogaster "megabacteriosis" has been diagnosed in the avian diseases laboratory in a diversity of avian species and varied spectrum of disease. The disease in some species (chickens, turkeys, guinea fowls) was clinically characterized by emaciation, prostration, loss of appetite, cachexia and death, with a typically chronic course. A more acute disease was observed in finches (canary-Serinus and zebra-Taeniopygia) and budgerigars (Melopsittacus undulatus). The large rod shaped organism, visible from 100 times magnification, with and without staining, could be detected in sick and also in reasonably normal individuals of some species, such as chickens, turkeys, quails and pigeons. In rheas (Rhea americana), ostriches (Struthio camelus), canaries, zebra-finches, guinea-fowl (Numida meleagris) and budgerigars. The disease was severe, causing to up to 100% mortality. The infection could be detected in some species along with other infectious or disease problems, such as endoparasites (helminths, coccidia) and ectoparasitism (order Mallophaga or/and order Acarina). The cultivation of M. ornithogaster was successfully achieved in solid and liquid media, originated from chickens (four isolates), guinea fowl (1 isolate), chuckar partridge (1 isolate) and canary (1 isolate). A very interesting finding at microscopy was motility of M. ornithogaster, as detected both in cultures obtained on agar for pathogenic fungi and passaged into thioglycolate broth, as well as on samples observed in wet preparations from in vivo. Differences in colony aspects were noted among the isolates. Experimental infections were attempted in chicken and japanese quail, using a chicken isolate, allowing the detection of the organism in the proventriculus and liver in apparently normal birds. One chicken isolate was injected intraperitoneally in Balb/c mice and resulted in 100% mortality.

A virulência das amostras de vacinas lentogênicas La Sota, Ulster e VG-GA do vírus da doença de Newcastle (VDN) foi determinada por análise morfométrica da espessura da traquéia de galinhas (n=12) de 45 dias de idade, livres de anticorpos anti-VDN, vacinadas por via intra-traqueal. As traquéias foram avaliadas 1, 2, 3, 4, 6 e 8 dias pós-vacinação. A amostra La Sota induziu maior espessamento da traquéia no decorrer de todo período experimental. Ao terceiro dia pós-vacinação, período em que as traquéias se apresentaram mais espessas, as amostras La Sota e Ulster não diferiram em sua virulência, sendo ambas mais virulentas do que a amostra VG-GA, induzindo maior espessamento de traquéia e causando lesões histológicas mais severas.

Virulence of three vaccinal lentogenic strains (La Sota, Ulster and VG-GA) of the Newcastle disease virus (NDV) was determined by morphometric analysis of tracheal thickness, in 45-day-old SPF chickens (n=12), free from NDV antibodies. Tracheal thickness was evaluated 1, 2, 3, 4, 6, and 8 days after intratracheal vaccination. La Sota strain induced higher tracheal swelling than the others, during most of the time of the experiment. Maximum swelling of tracheal mucosa occurred at the thirdday after vaccination. At that time, La Sota and Ulster had the same virulence, and both caused higher swelling of tracheal mucosa than VG-GA strain.

Tracheal organ cultures (TOC) were prepared and used for evaluating four Brazilian isolates of infectious bronchitis virus (IBV). IBV field isolates and vaccine strains were titrated in TOC and results compared to those from chicken embrionated eggs. Serum neutralization (SN) employing IBV strain-specific serum was performed for evaluating relationships between isolates. Titration results of tests performed in TOC or eggs were in mutual agreement and were considered for validating the adapted TOC methodology as alternative for virological studies in our laboratory. Sera specific to M41 (Massachusetts) or A5968 (Connecticut) did neutralize their respective IBV strains only. Field strains 208 and 29-78 specific sera did neutralize Massachusetts serotype strains M41 and H120, but PM2 serum did only M41. Strain PM4 specific serum did not neutralize any of the reference IBV analyzed, including M41, A5968 and H120 and may indicate that the isolate is serologically different from the Massachusetts serotype, currently adopted for vaccine strains in Brazil.

Relata-se um caso fatal de sarcosporidiose em pássaro-preto (melro), Gnorimopsar chopi chopi, caracterizado por intensa prostração, respiração superficial e decúbito lateral. Embora não tenham sido observadas alterações significativas no sistema respiratório, o quadro clínico confundia-se com doença nesse sistema, em decorrência do comprometimento dos músculos ligados à respiração.

Foram produzidos anticorpos monoclonais (AcM) da subclasse IgG1 contra as proteínas N (53KDa) e S2 (82KDa) do vírus da bronquite infecciosa das galinhas (VBIG) amostra M41. Os híbridos secretores originaram-se da fusão entre células de mielomas da linhagem Sp2/0-Ag14 e linfócitos B de camundongos Balb/c previamente imunizados com o vírus completo. O primeiro critério de seleção foi por ELISA, no qual 36 híbridos originados de duas fusões reagiram positivamente; destes, foram selecionados dois AcM que reagiram contra as proteínas N (N3D4) e S2 (S12B2) do VBIG da amostra M41 (sorotipo Massachusetts) no western blotting. Os mesmos AcM foram também capazes de reconhecer as estirpes Ark-99 (sorotipo Arkansas) e A5968 (sorotipo Connecticut) do VBIG no western blotting. No ELISA, o AcM contra S2 (S12b2) reconheceu 11 estirpes das 11 estudadas (M41, SE-17, H52, 297, 283, PM-1, PM-2, PM-3, 351, 29-78 E 327), enquanto o AcM contra a proteína N reconheceu apenas seis estirpes (M41, SE-17, H52, 283, 327 e 297). O anticorpo monoclonal contra S2 comportou-se como uma boa ferramenta de diagnóstico do VBIG, independentemente do sorotipo que pode ser aplicado em ensaios de rotina laboratorial, especialmente no diagnóstico das doenças que se confundem com a BIG nas galinhas, enquanto o AcM contra N (N3F10) demonstrou reconhecer uma região menos conservada entre as estirpes de VBIG e pode ser útil em estudos comparativos entre isolados de VBIG.

Murine hybridomas producing IgG1 monoclonal antibodies (Mabs) against N and S2 proteins (53KDa and 82KDa, respectively) from avian infection bronchitis virus (IBV) strain M41 were generated by the fusion of a myeloma cell line (Sp2/0-Ag14) with spleen cells from Balb/c mice previously immunized with whole virus IBV M41. Post-fusion screening criterion was by ELISA and 36 positive hybrids were generated after fusions. Two hybrids specific to N (N3F10) and S2 (S12B2) proteins from M41 (serotype Massachusetts) were selected by western blotting. These Mabs recognized the Ark-99 (serotype Arkansas) and A5968 (serotype Connecticut) IBV strains in addition to M41. By ELISA, the Mab against the S2 (S12B2) recognized all reference and Brazilian strains (M41, SE-17, H52, 297, 283, PM-1, PM-2, PM-3, 351, 29-78 E 327) studied, while the Mab against N recognized only six (M41, SE-17, H52, 283, 327 e 297) strains. The Mab against S2 may become a useful tool for IBV detection on the routine diagnosis of infectious bronchitis, especially for helping the differential diagnosis of clinically and pathologically confusing diseases, while the Mab against N (N3F10) recognized a probably less conserved region among the strains and may be interesting to comparing IBV isolates.

Um painel de anticorpos monoclonais (AcM) específicos contra o vírus da bronquite infecciosa das galinhas (VBIG) amostra Massachusetts M41 foi desenvolvido com a finalidade de possibilitar ferramentas para futuros estudos de amostras locais de VBIG. Os clones produtores de AcM foram detectados e caracterizados por ELISA, e para a determinação quanto à especificidade ao componente de VBIG foi utilizado western blotting (WB). Os híbridos produtivos foram clonados por diluição limitante, expandidos in vitro e mantidos em nitrogênio líquido. As bandas protéicas reconhecidas em WB pelos AcM apresentaram pesos moleculares que variaram de 180 a 30 kDa. Oito AcM reconheceram apenas um polipeptídeo e quatro ligaram-se ao polipeptídeo S (3, 18, 52 e 57) inteiro, com a banda de peso molecular aproximado de 180 kDa. Cinco AcM (5, 12, 41, 70 e 72) ligaram-se em mais de uma banda de VBIG. Outros sete AcM não se ligaram a nenhum polipeptídeo de VBIG. Entre os AcM que apresentam perspectivas de utilização em pesquisa e diagnóstico de VBIG incluem-se os produzidos pelo clone 42, dirigido contra S1, os produzidos pelos clones 34, 43 e 71, dirigidos contra S2, pelos clones 7 e 22, dirigidos contra M e pelos clones 15 e 50, cujos AcM reagiram contra a nucleoproteína N.

Monoclonal antibodies (Mab) specific to IBV Massachusetts M41 were produced aiming to the development of assays for IBV diagnosis and research, such as for rapidly detecting or typing IBV isolates. Mabs were detected by ELISA and characterized based on the isotype (ELISA) or IBV structural specificity (western blotting) respectively as IgA, IgG1, IgG2a, IgG2b or IgM or as specific to S1 (42), S2 (34, 43 and 71), M (7 and 22), N (15 and 50) and S whole molecule (3, 18, 52 and 57), the major IBV structural proteins

Relata-se um surto de malária por Haemoproteus columbae em pombos-correio (Columba livia), caracterizado por um quadro agudo com mortalidade de 3% ao dia em aves de aspecto saudável ou crônico com prostração e fraqueza. O exame clínico das aves doentes evidenciou sinusite e conjuntivite e à necropsia traqueíte hemorrágica, aerossaculite, esplenomegalia, nefromegalia e hepatomegalia com hemorragias. O díptero hematófago da família Hippoboscidae Pseudolynchia canariensis foi encontrado inserido entre as penas dos indivíduos. Esfregaços de sangue periférico e cardíaco e impressões de baço e fígado corados por Giemsa permitiram a visualização de inclusões intraeritrocitárias características de Haemoproteus columbae

Descreve-se a ocorrência de filariose em passeriformes da espécie Oryzoborus maximiliani (bicudo) mantidos em cativeiro. Os sinais clínicos incluiram insuficiência respiratória e prostração, evoluindo para decúbito lateral e morte. Todos os indivíduos capturados adoeceram e morreram em poucos dias. As lesões mais significativas foram encontradas nos pulmões, que estavam acinzentados na região adjacente ao saco aéreo abdominal. Impressões do pulmão observadas ao microscópio em 100 aumentos permitiram a visualização de grande número de formas alongadas típicas de nematódeo. Considerando suas dimensões e os relatos da literatura consultada, especulou-se a possibilidade de filariose. As condições de estresse de captura e cativeiro podem ser determinantes do quadro agudo observado, o que permite sugerir esta suspeita em casos semelhantes. Considera-se importante, entretanto, a possibilidade de a manifestação clínica na forma crônica ou assintomática poder ser mais comum que a aguda.

Soros de aves experimentalmente infectadas, contendo espiroquetas viáveis, foram submetidos a dois procedimentos antes da criopreservação: glicerol na diluição de 1/2 (v/v), designado como soro com glicerol a 50% (GS), e dimetilsulfóxido na proporção de 1/10 (v/v), designado como soro com DMSO a 10% (DS). Após 15 meses de estocagem em nitrogênio líquido, amostras dos tratamentos GS e DS foram descongeladas e suas infectividades foram testadas em frangos susceptíveis. Apesar de ambos os procedimentos terem mantidos a infectividade da bactéria, DMSO a 10% no soro de frango apresentou-se mais satisfatório como criopreservante.
