ABSTRACT Aedes (Stegomyia) albopictus (Skuse, 1854) was reported in Brazil for the first time in 1986 and has shown marked expansion throughout the Brazilian territory. During a routine activity to control dengue fever conducted by the Division of Entomology of the Municipal Health Department in Rio Branco city, adults and immatures of Culicidae were collected in a peri-urban area. The identified Culicidae forms indicated that they belonged to the species Ae. albopictus. This is the first official record of the presence of Ae. albopictus in the Acre State, confirming its current presence in all Brazilian states.

The development of new strategies for the functionalization of the historically inert C—H bond arises as an excellent way to create new carbon—carbon and carbon— heteroatom bonds. With increasingly chemo- and site-selective methods that enables the late-stage functionalization of C—H bonds, the modification of specific sites in natural products and pharmaceuticals without altering their scaffold emerges as a powerful means for the diversification of complex molecules. In this review, we will introduce concepts of late-stage modifications, the use of C—H bond functionalization to forge new carbon—carbon and carbon—heteroatom bonds using metal catalysis and photochemistry in simple examples and their applications in natural products and pharmaceuticals. The aim of the review is to update and display to the reader the contributions and implications of this methodology to organic synthesis. Antioxidant Properties of Croton zehntneri Pax et Hoffm. Essential Oil and Its Inclusion Complex with β-Cyclodextrin Prepared by Spray Drying

Croton zehntneri Pax et Hoffm. is a plant from Northeastern Brazil, whose main component of its essential oil is estragole. Its thermal instability and low aqueous solubility prevent its technological application. This study aimed to prepare and characterize the Croton zehntneri essential oil (CZEO) complexed with β-cyclodextrin (β-CD), and to evaluate antioxidant activity of free and complexed-CZEO in vitro systems by the elimination of 2,2-diphenyl-1-picrylhydrazyl radicals (DPPH•), 2,2’-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS•+), hydroxyl and nitric oxide, as well as their ability to transfer electrons by the reducing potential. The infrared spectroscopy and thermal techniques demonstrated the formation of the inclusion complex (Est/β-CD) obtained by spray drying method. The free CZEO and its Est/β-CD at 0.9, 1.8, 3.6, 7.2, 14.4 and 28.8 µg mL-1 showed the ability to remove DPPH• (EC50 (half maximal effective concentration) 26.06 and 9.46 µg mL-1, respectively) and ABTS•+ (EC50 22.73 and 4.47 µg mL-1, respectively), nitric oxide inhibition (EC50 17.65 and 2.68 µg mL-1, respectively), hydroxyl radical sequestration (EC50 23.42 and 2.34 µg mL-1, respectively) and reducing potential (EC50 46.48 and 12.47 µg mL-1, respectively). The formation of the inclusion complex significantly increases its antioxidant potential.

In this study, we assessed the cytotoxic effect of 1,3-diphenyl-2-allyl-1,3-propanedione (DPAP) on B16F10 and Tm5 melanoma cells and investigated its interaction with DNA (deoxyribonucleic acid) using optical tweezers. The compound showed to be effective against B16F10 cells with selectivity index (SI) of 7.92 and interaction to the outer surface of the double helix, probably at the minor groove of DNA. This likely induced a bending deformation of the polymer chain, decreasing persistence length, and indicated that each DPAP molecule occupies a single base pair upon binding to the double helix.

In this paper, we have applied state-of-the-art technologies and new investigations were developed focusing on obtaining simple and inexpensive molecules for treating cardiovascular diseases, as they are considered the leading cause of death in the world. For the first time, the inclusion complex of sulcatone (SU) and β-cyclodextrin (β-CD) was achieved (using a spray drier) and characterized making use of spectroscopic, thermal and computational methods. In addition, the system was evaluated regarding its vasorelaxant properties. Evidence of the inclusion complex formation was provided using different techniques, such as X-ray diffraction (XRD), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), Fourier transform infrared (FTIR), and 1H nuclear magnetic resonance (NMR) spectroscopies and then deduced by pharmacological experiments, which showed significant changes in relation to the initial material.

Drying of microalga Scenedesmus obliquus BR003 in a gas dryer at low temperatures

ABSTRACT: Development of efficient and specific equipment to dry microalgae is essential for commercial use of these microorganisms that show high growth rates and biotechnological potential. Drying costs of microalgae biomass are still high and they should be reduced for the production of compounds with low added value. Therefore, we evaluated the drying process of the microalga Scenedesmus obliquus BR003 using low temperatures. S. obliquus BR003 is a robust microalga that shows high lipid productivity. At laboratory scale, it was observed that the biomass of S. obliquus BR003 was rapidly dried at low temperatures between 50 and 60 ° C. A gas dryer was used to evaluate the drying of the biomass of S. obliquus BR003 on a pilot-scale. The biomass was dried in less than 24 h using the gas dryer; however, the high moisture of the microalga biomass required a higher air renovation in the drying chamber. Computational fluid dynamics analysis of the gas dryer showed two important parameters to achieve greater effectiveness of heat and mass transfer rates during the drying process of the microalga biomass. It was concluded that a gas dryer suitable for the microalgae biomass should have multiple air injection points, and an efficient circulation and renovation system of air inside the drying chamber.
RESUMO: O desenvolvimento de equipamentos eficientes e específicos para a secagem de microalgas é essencial para a exploração comercial destes microrganismos que apresentam alta taxa de crescimento e grande potencial biotecnológico. Os custos de secagem da biomassa de microalgas ainda são elevados e precisam ser reduzidos para a produção de compostos com baixo valor agregado. Portanto, realizou-se o estudo da secagem da microalga Scenedesmus obliquus BR003 utilizando baixas temperaturas. S. obliquus BR003 é uma microalga robusta que apresenta alta produtividade de lipídeos. Em escala laboratorial, observou-se que a biomassa de S. obliquus BR003 foi rapidamente seca em baixas temperaturas entre 50 e 60 ºC. Um secador a gás foi utilizado para avaliar a secagem da biomassa de S. obliquus BR003 em escala piloto. A biomassa foi seca em menos de 24 h utilizando o secador a gás, entretanto, a elevada umidade da biomassa da microalga requereu uma maior renovação de ar na câmara do secador. A análise de fluidodinâmica computacional do secador a gás mostrou dois parâmetros importantes para se obter uma maior efetividade de transferência de calor e massa durante o processo de secagem da biomassa de microalga. Concluiu-se que um secador a gás adequado, para a biomassa de microalgas, deve possuir múltiplos pontos de injeção de ar, e um eficiente sistema de circulação e renovação de ar no interior da câmara de secagem.

ABSTRACT The maturation process of Sesbania virgata seeds was investigated through fruits and seeds at different maturation stages, where they were described morphologically. For the seeds, fresh and dry mass and moisture were determined and structural analysis of the tegument under a light microscope was performed. Germination tests in the presence and absence of drying and the electrophoretic profile of heat resistant proteins were also performed. The results showed that the seeds reached physiological maturity at 44 days after anthesis (DAA), evidencing a secondary cell wall. The acquisition of desiccation tolerance was observed at 44 DAA, with the seeds responding to the drying treatment. Protein bands were observed from 32 DAA, which remained constant until the end of maturation. These results allowed the characterization of the development and acquisition of desiccation tolerance in S. virgata seeds.

RESUMO As exigências de culturas agrícolas para o macronutriente enxofre (S) são similares ou superiores às de fósforo. Apesar disso, muitas vezes, o S não é incorporado ao manejo da adubação de culturas. Objetivou-se avaliar, em condições de campo, o efeito de fontes e doses de S na cultura da soja. O delineamento foi em blocos ao acaso, com 4 repetições. Os tratamentos foram dispostos em esquema fatorial com um tratamento adicional (3 x 5 + 1), totalizando 64 unidades experimentais. Foram utilizadas fontes (enxofre elementar, superfosfato simples e gesso agrícola) e doses (20 kg ha-1, 40 kg ha-1, 60 kg ha-1, 80 kg ha-1 e 100 kg ha-1) de S, mais um controle (sem fertilizante). Foram avaliados os seguintes parâmetros: altura da planta no florescimento, massa de 1.000 grãos, produtividade e eficiência agronômica, as quais foram influenciadas pelas doses e fontes de S; número de vagens por planta, que somente foi influenciado pelas fontes; e número de vagens com 3 grãos, que não se diferenciou estatisticamente em nenhum tratamento. A maior produtividade foi de 1.927 kg ha-1, na dose 80 kg ha-1 de S para o gesso, e a maior eficiência agronômica foi observada para o superfosfato simples, na dose de 20 kg ha-1 de S.
ABSTRACT Crop demands for the macronutrient sulfur (S) are similar or greater than those for phosphorous. However, S is often overlooked in the management of crop fertilization. This study aimed to evaluate the effect of sources and doses of S on soybean cultivation under field conditions. A randomized block design, with 4 replications, was used. The treatments were arranged in a factorial scheme with one additional treatment (3 x 5 + 1), totaling 64 experimental units. Sources (elemental sulfur, single superphosphate and agricultural gypsum) and doses (20 kg ha-1, 40 kg ha-1, 60 kg ha-1, 80 kg ha-1 and 100 kg ha-1) of S were used, in addition to one control (without fertilizer). The following parameters were assessed: plant height at flowering, 1,000-seed weight, yield and agronomic efficiency, which were affected by the doses and sources of S; number of pods per plant, only affected by the sources of S; and number of pods with 3 seeds, with no statistical difference for any of the treatments. The highest yield recorded was 1,927 kg ha-1, at the dose of 80 kg ha-1 of S for gypsum, and the greatest agronomic efficiency was observed for the single superphosphate, at the dose of 20 kg ha-1 of S.

Resumo: Em sementes, tolerância à dessecação (TD) e longevidade são necessárias para a melhor dispersão da espécie e para o estabelecimento de um banco de sementes no solo. Ambas são adquiridas durante fase de maturação do desenvolvimento das sementes e perdidas durante a germinação. A TD pode ser re-induzida em sementes germinadas por um tratamento osmótico e/ou com ácido abscísico. Entretanto, não há informações sobre como esses tratamentos afetam a longevidade das sementes. No presente estudo, utilizou-se sementes germinadas de Sesbania virgata como modelo experimental para investigar os efeitos na longevidade de um tratamento osmótico para re-induzir TD. A longevidade de sementes germinadas de S. virgata submetidas ou não a um tratamento osmótico foi analisada após armazenamento ou envelhecimento acelerado. A radícula é o órgão mais sensível e os cotilédones são o órgão mais resistente. A habilidade de produzir raízes laterais é imprescindível para a sobrevivência das sementes. Sementes germinadas de S. virgata com radícula de 1mm de comprimento toleram dessecação e armazenamento por até três meses sem redução significativa na longevidade. O tratamento osmótico melhora a TD nessas sementes, mas não a longevidade. Sementes germinadas de S. virgata são um bom modelo para estudar a TD desacoplada da longevidade.
Abstract: Seed desiccation tolerance (DT) and longevity are necessary for better dissemination of plant species and establishment of soil seed bank. They are acquired by orthodox seeds during the maturation phase of development and lost upon germination. DT can be re-induced in germinated seeds by an osmotic and/or abscisic acid treatment. However, there is no information on how these treatments affect seed longevity. Germinated Sesbania virgata seeds were used as a model system to investigate the effects of an osmotic treatment to re-establish DT on seed longevity. Longevity of germinated S. virgata seeds treated and non-treated by an osmoticum was analysed after storage or artificial ageing. The radicle is the most sensitive organ, the cotyledons are the most resistant, and the ability to produce lateral roots is the key for whole seed survival. Germinated S. virgata seeds with 1mm protruded radicle tolerate desiccation and storage for up to three months without significant losses in viability. An osmotic treatment can improve DT in these seeds, but not longevity. Germinated S. virgata seeds are a good model to study DT uncoupled from longevity. Further studies are necessary to unveil the molecular mechanisms involved in both DT and longevity.

Syagrus romanzoffiana (Arecaceae) es una palma nativa y ampliamente distribuida de América del Sur. El presente estudio investigó la longevidad y la germinación de las semillas enterradas de esta especie en un banco experimental. Se realizaron pruebas de germinación y viabilidad de laboratorio para hacer una comparación con los resultados de campo. Pirenos (semillas encerradas por el endocarpio) fueron enterrados en el borde de un fragmento de bosque en julio (época seca) y exhumados mensualmente por un año, para la evaluación del contenido de agua y del porcentaje de semillas germinadas y viables. Las pruebas de germinación se realizaron en un germinador tipo Mangelsdorf a 30°C bajo luz constante y la evaluación de la viabilidad se realizó con la prueba de tetrazólio. Una muestra adicional de pirenos fue enterrada para evaluar el porcentaje de emergencia de las plántulas. Se tomaron los datos climáticos y se determinó la humedad del suelo. En el laboratorio, los pirenos se almacenaron durante un año en una sala de temperatura controlada a 20°C y 75% (±10%) de humedad relativa del aire, para evaluar los cambios en el porcentaje de germinación y la viabilidad en el tiempo. En el campo, se observó una reducción de la viabilidad de las semillas durante el período de estudio, con una pérdida total de la viabilidad de las semillas no germinadas a los siete meses después del entierro. La germinación máxima (cerca de 26%) se observó en las muestras que fueron exhumadas entre cinco y siete meses después del entierro. En el campo, la emergencia de las plántulas no superó el 10% y no se observó mortalidad de plántulas. Los porcentajes de germinación y de semillas viables disminuyeron tanto en el suelo como en el laboratorio. Las semillas almacenadas mantuvieron la viabilidad hasta por seis meses de almacenamiento, con una reducción marcada a partir de entonces. Después de las pruebas de germinación (cuatro meses) en el laboratorio, todas las semillas restantes eran inviables. Discutimos las características ecológicas, fisiológicas y reproductivas de la especie, y llegamos a la conclusión de que las semillas de S. romanzoffiana tienen corta duración después de la imbibición y bajo potencial de formación del banco de semillas en el suelo.
Syagrus romanzoffiana is a palm tree native and widely distributed of South America. The present study investigated the longevity and germination of the buried seeds of this species in an experimental seed bank. Laboratory germination and viability tests were performed for comparison with field results. Pyrenes (seeds enclosed by the endocarp) were buried in a forest fragment edge in July 2012 (dry season) and exhumed monthly during one year, for the assessment of water content and percentage of germinated and viable seeds. Germination tests were conducted in a Mangelsdorf-type germinator at 30°C under constant light and the viability was assessed by the tetrazolium test. An additional sample of pyrenes was buried to evaluate the percentage of seedling emergence and survival. Climatic and soil moisture data were recorded. In the laboratory, the pyrenes were stored for one year in a temperature-controlled room at 20ºC and 75% (±10%) relative air humidity to assess changes in the percentage of germination and viability over time. In the field, a reduction in seed viability was observed over the study period, with a total loss of viability of non-germinated seeds at seven months after burial. The maximum germination (close to 26%) was observed in the samples that were exhumed between five and seven months after burial. In the field, seedling emergence did not exceed 10% and seedling mortality was not observed. The percentages of germination and of viable seeds decreased both in burial and stored seeds. The stored seeds maintained viability at up to six months, with marked reduction thereafter. After the germination tests (four months) in the laboratory, all of the remaining seeds were nonviable. The ecological, physiological and reproductive characteristics of the species are discussed, and we concluded that S. romanzoffiana seeds have short longevity after imbibition, and low potential for soil seed bank formation. Isolation, characterization and evaluation of antimicrobial and cytotoxic activity of estragole, obtained from the essential oil of croton zehntneri (euphorbiaceae)

Croton zehntneri (Euphorbiaceae) é uma planta aromática nativa da região Nordeste do Brasil. O monoterpenóide Estragol (ESL) foi isolado por métodos cromatográficos clássicos a partir do óleo essencial (OE) das folhas de C. zehntneri e caracterizado por CG-DIC e CG-EM, tendo seu potencial antimicrobiano e citotóxico avaliado. A análise do OE permitiu a identificação de 100% dos constituintes integrados, cujo rendimento foi cerca de 1.8%. Os principais componentes identificados foram: eucaliptol, estragol (84.7%) e espatulenol. A dosagem de 50 μg/disco de ESL apresentou halos de inibição bastante expressivos frente a bactérias gram positiva e fungos. O ESL apresentou toxicidade frente a Artemia salina com CL50 e CL90 de 4,54 e 8,47 μg mL-1. Entretanto, nos testes de inibição tumoral (células humanas), não houve inibição satisfatória em nenhuma das linhagens de células humanas cancerígenas (MCF-7, HEP-2 e NCIH292).
Croton zehntneri (Euphorbiaceae) is a native aromatic plant from Northeast region of Brazil. The monoterpenoid estragole (ESL) has been isolated by classical chromatographic methods from the essential oil (EO) of C. zehnteneri leaves and characterized by GC-FID and GC-MS, its antimicrobial and cytotoxic potentials being assessed. The analysis of the EO enabled the identification of 100% of the integrated constituents, of which yield was about 1.8%. The main components identified were: eucalyptol, estragole (84.7%) and spathulenol. The dosage of 50 μg/disk of ESL presented fairly significant zones of inhibition against Gram-positive bacteria and fungi. The ESL presented toxicity against Artemia salina with LC50 and LC90 of 4,54 and 8,47 μg mL-1. However, in tumor inhibition assays (human cells), there were no rewarding inhibition in any of the human cancer cell lines (MCF-7, HEP-2 and NCI-H292).

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Direct detection of Mycobacterium tuberculosis complex in bovine and bubaline tissues through nested-PCR

Post-mortem bacterial culture and specific biochemical tests are currently performed to characterize the etiologic agent of bovine tuberculosis. Cultures take up to 90 days to develop. A diagnosis by molecular tests such as PCR can provide fast and reliable results while significantly decreasing the time of confirmation. In the present study, a nested-PCR system, targeting rv2807, with conventional PCR followed by real-time PCR, was developed to detect Mycobacterium tuberculosis complex (MTC) organisms directly from bovine and bubaline tissue homogenates. The sensitivity and specificity of the reactions were assessed with DNA samples extracted from tuberculous and non-tuberculous mycobacteria, as well as other Actinomycetales species and DNA samples extracted directly from bovine and bubaline tissue homogenates. Regarding the analytical sensitivity, DNA of the M. bovis AN5 strain was detected up to 1.5 pg by nested-PCR, whereas DNA of M. tuberculosis H37Rv strain was detected up to 6.1 pg. The nested-PCR system showed 100% analytical specificity for MTC when tested with DNA of reference strains of non-tuberculous mycobacteria and closely-related Actinomycetales. A clinical sensitivity level of 76.7% was detected with tissues samples positive for MTC by means of the culture and conventional PCR. A clinical specificity of 100% was detected with DNA from tissue samples of cattle with negative results in the comparative intradermal tuberculin test. These cattle exhibited no visible lesions and were negative in the culture for MTC. The use of the nested-PCR assay to detect M. tuberculosis complex in tissue homogenates provided a rapid diagnosis of bovine and bubaline tuberculosis.

Croton zehntneri, a plant native to northeastern Brazil, is widely used in folk medicine to treat gastrointestinal problems and has rich essential oil content. The essential oil of C. Zehntneri was analyzed by GC-MS, and its inclusion complex with β-cyclodextrin (β-CD) was characterized by both vibrational spectroscopy and differential scanning calorimetry (DSC). Estragol was the major component identified in the essential oil by the study. IR spectra indicated an interaction of β-CD with essential oil from C. zehntneri, a finding corroborated by the stability constant and scanning calorimetry. Microencapsulation within β-CD has the potential to mask sensory attributes and increase aqueous solubility of oils, thereby improving their applicability as drugs.
