Five cycles of intrapopulation recurrent selection in half-sib progenies of fresh corn

ABSTRACT Fresh corn is a highly appreciated vegetable in Brazil; this crop has high added value and profitability when compared to dry corn. Despite the consumer market demand for quality, cultivars are not launched every crop season and there is a small number of cultivars available to growers. This denotes a need for breeding programs focused on generating information and genotypes that can occupy this market niche. The objective of this work was to evaluate the progress of five cycles of intrapopulation recurrent selection in half-sib progenies of fresh corn, in the southwest region of Goiás. A randomized block experimental design was used, with evaluation of 64 progenies of corn half-sib progenies and selection intensity of 30% per cycle, with three replications, in the 2011-2012, 2013-2014, 2015-2016, 2017-2018, and 2020-2021 summer crop seasons. The half-sib progenies of fresh corn exhibited genetic variability through intrapopulation recurrent selection in the different selection cycles, which is still promising for further selections after five cycles, with evaluation of 64 progenies per cycle. The base population from crossings between hybrids for tropical and temperate climates showed to be promising for increasing the genetic base of fresh corn populations, with satisfactory gains over the five cycles, presenting mean heritability between the cycles of approximately 78.9% for relative chlorophyll index, 62.2% for plant height, 75.1% for ear insertion height, 70% for ear diameter, 56.6% for ear length, 71.4% for unhusked ear yield, and 71% for marketable ear yield.
RESUMO O milho fresco é uma hortaliça muito apreciada pelos brasileiros e, além disso, possui alto valor agregado e maior lucratividade, quando comparado aos grãos secos. O mercado consumidor apesar de exigente na qualidade, não é abastecido com novas cultivares a cada safra e com um número ínfimo sendo disponível aos produtores. Assim, existe a necessidade da criação de programas de melhoramento, com o intuito de gerar informações e genótipos capazes de suprir esse nicho de mercado. O objetivo do estudo foi avaliar o progresso da seleção em progênies de meiosirmãos de milho fresco, em cinco ciclos de seleção recorrente intrapopulacional, na região sudoeste de Goiás. O delineamento experimental utilizado foi o de blocos casualizados, com a avaliação de 64 progênies de meios-irmãos e intensidade de seleção de 30% por ciclo, com três repetições, em diferentes safras de verão (2011-2012, 2013-2014, 2015-2016, 2017-2018 e 2020-2021). As progênies de famílias de meios-irmãos de milho fresco apresentaram variabilidade genética por meio de seleção recorrente intrapopulacional nos diferentes ciclos de seleção, com avaliação de 64 progênies por ciclo. A população base de cruzamentos entre híbridos para climas tropicais e temperados mostrou-se promissora para aumentar a base genética de populações de milho fresco, com ganhos satisfatórios ao longo dos cinco ciclos, com herdabilidade média de aproximadamente 78,9% para o índice relativo de clorofila, 62,2% para altura de planta, 75% para inserção da espiga, 70% para diâmetro da espiga, 56,6% para comprimento da espiga, 71,4% para espigas empalhadas e 71% para espigas comerciais entre os ciclos.

RESUMO O objetivo deste trabalho foi estimar e comparar parâmetros fenotípicos e genéticos após três ciclos de seleção recorrente intrapopulacional em milho fresco sob três disponibilidades nutricionais. Três experimentos derivados de uma mesma população foram realizados para avaliar a capacidade das progênies em adquirir os nutrientes, um sob fertilização adequada (controle – MV-006), outro sob baixa disponibilidade de nitrogênio (N – MV-006N) e outro sob baixa disponibilidade de fósforo (P – MV-006P). Os experimentos foram realizados em delineamento de blocos casualizados, com 64 progênies de meios-irmãos, com três repetições. Foram avaliadas as características de índice relativo de clorofila, altura da planta e inserção da espiga, diâmetro e comprimento da espiga, produtividade de espigas empalhadas e despalhadas. As populações apresentam variabilidade para as três condições de disponibilidade nutricional. A variabilidade foi muito reduzida com apenas três ciclos de seleção, sendo indicado avaliar um número maior de progênies de meiosirmãos, acima de 64, sob baixa disponibilidade de N e P. A dose a ser usada para identificar as progênies mais eficientes no programa de melhoramento deve estar próxima a recomendada, com uma redução lenta após os ciclos seletivos. Dessa forma, a variabilidade será mantida e com maior possibilidade de identificação de progênies mais promissoras. Além disso, a frequência de alelos favoráveis aumenta de forma mais gradual e eficaz, sendo que a dose de 60 kg ha-1 de N e P não é indicada na fase inicial do programa, como nível crítico.
ABSTRACT The objective of this work was to estimate and compare phenotypic and genetic parameters after three cycles of intrapopulation recurrent selection for fresh corn grown under three nutrient availability conditions. Three experiments derived from the same population (MV-003) were conducted to assess the ability of progenies to absorb nutrients, one under adequate fertilization (control; MV-006), one under low nitrogen availability (N; MV-006N), and one under low phosphorus availability (P; MV-006P). The experiments were carried out in randomized blocks, with 64 half-sib progenies and three replications for each nutritional condition. Chlorophyll index, plant height, ear insertion, ear diameter, ear length, unhusked ear yield, and marketable husked ear were evaluated. The populations showed variability for the three nutrient availability conditions. The variability reduced after only three selection cycles; thus, evaluating a higher number of half-sib progenies, above 64, under low N and P availability is indicated. The rates used to identify the most efficient progenies in breeding programs should be close to the appropriate one, with a slow reduction after the selection cycles. The variability will be thus maintained, with a greater possibility of identifying more promising progenies. In addition, the frequency of favorable alleles increases more gradually and effectively. The N and P rate of 60 kg ha-1 is not indicated as a critical level for the initial phase of the program.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

Nutritional status of rice plants supplied with silicon in response to Pyricularia oryzae infection

ABSTRACT Considering the importance of silicon (Si) to increase rice resistance to blast, this study investigated the nutritional status of rice plants supplied with this element in response to Pyricularia oryzae infection. Plants were grown in nutrient solutions containing 0 and 2 mmol·L-1 of Si (referred to -Si and +Si plants, respectively). The area under blast progress curve decreased for +Si plants due to a significant increase of 714% on the foliar Si concentration in contrast to -Si plants. For +Si plants, the foliar N, P, K, Ca, Mg, Cu, and B concentrations were significantly lower in comparison to -Si plants. On infected plants, the foliar N, K, and Cu concentrations were significantly lower in comparison to noninfected ones. The foliar Zn, Fe, and Mn concentrations were significantly lower for -Si inoculated plants in comparison to -Si noninoculated ones. The foliar Mn and S concentrations were significantly lower for +Si inoculated plants in comparison to +Si noninoculated ones. The foliar Fe concentration was significantly lower for +Si inoculated plants in comparison to +Si noninoculated ones. Foliar Zn and Mn concentrations were significantly lower for +Si plants in comparison to -Si plants regardless of their inoculation with P. oryzae. Foliar Fe and S concentrations were significantly lower for +Si plants in comparison to -Si plants, respectively, for noninoculated and inoculated plants. Taken together, Si stood for itself in reducing blast symptoms without the need of high foliar concentrations of either macro or micronutrients that could affect plantgrowth.

Asymptomatic Plasmodium infection in a residual malaria transmission area in the Atlantic Forest region: Implications for elimination

Abstract INTRODUCTION: Elimination of malaria in areas of interrupted transmission warrants careful case assessment to avoid the reintroduction of this disease. Occasional malaria cases are reported among visitors of the Atlantic Forest area of Brazil, while data on residents of this area are scarce. METHODS: A sectional study was carried out to examine 324 individuals living in a municipality where autochthonous cases were detected. RESULTS: Asymptomatic Plasmodium infections were detected in 2.8% of the individuals by polymerase chain reaction (PCR), with one case of P. falciparum (0.3%), two cases of P. vivax (0.6%), and six cases of P. malariae (1.9%). The thick blood smears were negative in all individuals. Serological tests performed in 314 subjects were reactive in 11.1%, with 3.5% for P. falciparum and 7.7% for P. vivax. A subsample of 42 reactive individuals for any Plasmodium species showed P. malariae in 30.9% of specimens. Individuals who entered the Atlantic Forest region were 2.7 times more likely to exhibit reactive serology for P. vivax compared with individuals who did not enter this region (p<0.05). Children <15 years had a higher chance of reactive serology for P. falciparum and P. vivax than individuals ≥15 years of age (p<0.05). Individuals living in the Paraiso district had a higher chance of reactive serology for P. vivax compared to other districts (p<0.05). No associations were found between sex, past exposure to malaria, or serological response to antibodies of any Plasmodium species. CONCLUSIONS: The implications of these results for the elimination of malaria were discussed.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The lethality of malaria in the extra-Amazonian region is more than 70 times higher than in Amazonia itself. Recently, several studies have shown that autochthonous malaria is not a rare event in the Brazilian southeastern states in the Atlantic Forest biome. Information about autochthonous malaria in the state of Rio de Janeiro (RJ) is scarce. This study aims to assess malaria cases reported to the Health Surveillance System of the State of Rio de Janeiro between 2000-2010. An average of 90 cases per year had parasitological malaria confirmation by thick smear. The number of malaria notifications due to Plasmodium falciparum increased over time. Imported cases reported during the period studied were spread among 51% of the municipalities (counties) of the state. Only 35 cases (4.3%) were autochthonous, which represents an average of 3.8 new cases per year. Eleven municipalities reported autochthonous cases; within these, six could be characterised as areas of residual or new foci of malaria from the Atlantic Forest system. The other 28 municipalities could become receptive for transmission reintroduction. Cases occurred during all periods of the year, but 62.9% of cases were in the first semester of each year. Assessing vulnerability and receptivity conditions and vector ecology is imperative to establish the real risk of malaria reintroduction in RJ.

Anti-glycosylphosphatidylinositol (GPI) antibodies (Abs) may reflect and mediate, at least partially, anti-disease immunity in malaria by neutralising the toxic effect of parasitic GPI. Thus, we assessed the anti-GPI Ab response in asymptomatic individuals living in an area of the Brazilian Amazon that has a high level of malaria transmission. For comparative purposes, we also investigated the Ab response to a crude extract prepared from Plasmodium falciparum, the merozoite surface protein (MSP)3 antigen of P. falciparum and the MSP 1 antigen of Plasmodium vivax (PvMSP1-19) in these individuals and in Angolan patients with acute malaria. Our data suggest that the Ab response against P. falciparum GPI is not associated with P. falciparum asymptomatic infection in individuals who have been chronically exposed to malaria in the Brazilian Amazon. However, this Ab response could be related to ongoing parasitaemia (as was previously shown) in the Angolan patients. In addition, our data show that PvMSP1-19may be a good marker antigen to reflect previous exposure to Plasmodium in areas that have a high transmission rate of P. vivax.

Infecção assintomática por Plasmodium é um novo desafio para a saúde pública no Brasil. A reação em cadeia da polimerase (PCR) é o melhor método para detectar baixas parasitemias presentes em pacientes com infecção assintomática. Nas áreas endêmicas, a coleta de sangue total é dificultada pela distancia geográfica, transporte e adequada armazenagem das amostras. A coleta de sangue em papel de filtro pode ser uma alternativa nessas áreas de difícil acesso. Neste estudo foram comparados três diferentes métodos de extração de ADN a partir de papel de filtro usando como controle extração a partir de sangue total. O protocolo Chelex®-Saponina foi o que obteve o melhor resultado quando comparado com os outros três protocolos. No entanto a sensibilidade foi de 66,7% para o P. falciparum e 31,6% para o P. vivax. Conclui-se que em caso de infecção assintomática o papel de filtro não é ainda uma boa alternativa para coleta de amostras.
Asymptomatic Plasmodium infection is a new challenge for public health in the American region. The polymerase chain reaction (PCR) is the best method for diagnosing subpatent parasitemias. In endemic areas, blood collection is hampered by geographical distances and deficient transport and storage conditions of the samples. Because DNA extraction from blood collected on filter paper is an efficient method for molecular studies in high parasitemic individuals, we investigated whether the technique could be an alternative for Plasmodium diagnosis among asymptomatic and pauciparasitemic subjects. In this report we compared three different methods (Chelex®-saponin, methanol and TRIS-EDTA) of DNA extraction from blood collected on filter paper from asymptomatic Plasmodium-infected individuals. Polymerase chain reaction assays for detection of Plasmodium species showed the best results when the Chelex®-saponin method was used. Even though the sensitivity of detection was approximately 66% and 31% for P. falciparum and P. vivax, respectively, this method did not show the effectiveness in DNA extraction required for molecular diagnosis of Plasmodium. The development of better methods for extracting DNA from blood collected on filter paper is important for the diagnosis of subpatent malarial infections in remote areas and would contribute to establishing the epidemiology of this form of infection.

Different levels of taxonomic resolution in bioassessment: a case study of oligochaeta in lowland streams

OBJETIVO: Este estudo analisou as uso de oligoquetos em diferentes níveis de resolução taxonômica como indicadores ambientais em rios de planície Argentina afetadas por diferentes usos da terra. MÉTODOS: Os sítios de amostragem foram agrupados com base nas características físico-químicas e habitat (perturbação baixa, moderada ou alta). Amostras de oligoquetos foram coletadas sazonalmente em habitats de sedimentos e vegetação. RESULTADOS: Os aumentos de nutrientes e matéria orgânica resultaram em densidades elevadas de Oligochaeta, mas quando a perturbação também envolveu mudanças no habitat físico ou incrementos em substâncias tóxicas, a abundância diminuiu de forma significativa para valores ainda mais baixos que os de ambientes não perturbados. As respostas dos Naidinae e Tubificinae foram semelhantes. A densidade de Pristininae diminuiu com o aumento da perturbação, mas as densidades de Enchytraeidae e Rhyacodrilinae aumentaram nos locais mais altamente perturbados. Os Opistocystidae não ocorreram em locais de alta perturbação. A riqueza de espécies e a diversidade (H') foram menores em locais de perturbação elevada e ainda mais baixos nos sedimentos. Algumas espécies não apresentaram restrições no tipo de habitat ou com o nível de contaminação: Limnodrilus hoffmeisteri, Dero furcatus, D. digitata, D. pectinata, Pristina longiseta e P. aequiseta. Além disso, Trieminentia corderoi, Slavina appendiculata e Aulodrilus pigueti exibiram uma maior abundância em locais de baixa perturbação e não foram registrados em locais com elevadas perturbações. CONCLUSÕES: Os Oligochaeta apresentaram uma valência ecológica relativamente ampla, através de seu extensivo número de espécies. Embora os níveis taxonômicos mais baixos podem dar informações sobre o status ambiental, testes com espécies com diferentes sesibilidades para diferentes tipos e graus de contaminação serão da maior relevância para a avaliação da qualidade ecológica.
AIM: This study evaluated the use of oligochaetes at different levels of taxonomic resolution as environmental indicators in Argentine lowland streams affected by different land uses. METHODS: Sampling sites were grouped based on the physicochemical and habitat characteristics (low-, moderate-, and high-impact disturbance). Collection of the oligochaetes samples was carried out seasonally in sediment and vegetation habitats. RESULTS: The increases in nutrients and organic matter produced elevated densities of the Oligochaeta, but when the disturbance also involved changes in the physical habitat or enhancements in toxic substances, the abundance decreased significantly to values even lower than those of non-impacted environments. The responses of Naidinae and Tubificinae were similar. The density of the Pristininae decreased with increasing impact, but those of the Enchytraeidae and Rhyacodrilinae increased at the most highly impacted sites. The Opistocystidae were not recorded in high-impact sites. Species richness and diversity (H') were lower in high-impact sites and even lower in sediments. Some species presented no restrictions in the habitat type or with the contamination level: Limnodrilus hoffmeisteri, Dero furcatus, D. digitata, D. pectinata, Pristina longiseta, and P. aequiseta. Moreover, Trieminentia corderoi, Slavina appendiculata, and Aulodrilus pigueti exhibited the highest abundances at low-impact sites and were not registered in high-impact sites. CONCLUSIONS: The Oligochaeta show a relatively wide ecological valence through their extensive number of species. Although lower taxonomic levels can give information about environmental status, test-species' sensitivities to different types and degrees of contamination will be of utmost relevance to the evaluation of ecological quality.

Mudanças no padrão epidemiológico da malária em área rural do médio Rio Negro, Amazônia brasileira: análise retrospectiva

Foi realizado um estudo retrospectivo dos casos de malária ocorridos entre 1992 e 2004 no Município de Barcelos, Amazonas, Brasil, aprofundando as informações existentes na área altamente endêmica do Rio Padauri, afluente do Rio Negro. Foram identificados 16.795 casos de malária, sendo 61,4% (10.318) da área rural e 38,6% (6.477) da área urbana, com uma incidência parasitária anual média de 136,7 por mil habitantes na área urbana e 613,6 na área rural. A incidência parasitária anual média no Rio Padauri foi de 708,9/1.000. Nesse rio, foram diferenciados dois períodos epidemiológicos, sendo um denominado epidêmico (1992-1998) e outro pós-epidêmico (1999-2004). Comparando os dois períodos, a proporção homem/mulher mudou de 1,8 para 1,14, a média de idade dos pacientes passou de 17,9 para 14,8, o percentual de casos por Plasmodium falciparum reduziu-se de 51,9% para 23,7, e o percentual de lâminas com baixa densidade parasitária de P. falciparum passou de 35,3% para 44,9%, e as de P. vivax de 24% para 35%. Todas essas diferenças foram estatisticamente significativas (p < 0,05). Tais mudanças no padrão epidemiológico da malária no rio Padauiri devem ser esclarecidas mediante estudos prospectivos.

Inactivation of Escherichia coli in water by TiO2-assisted disinfection using solar light

Estudou-se a desinfecção de águas de abastecimento por fotocatálise heterogênea usando um reator de fluxo em um sistema composto por uma placa de vidro com TiO2 P 25 (Degussa) imobilizado e luz solar como fonte de radiação. Foram utilizados dois modos de operação do reator: passagem única e recirculação. Os experimentos foram conduzidos utilizando inicialmente uma água preparada em laboratório e posteriormente água coletada em poços e lagos de uma região próxima à Campinas, SP. Estudou-se a influência de fatores, tais como, o modo de operação do reator, a cor e turbidez da água, os quais influenciam significativamente na eficiência fotocatalítica de descontaminação e, portanto, na viabilidade da aplicação do processo. Em dias ensolarados, alcançou-se uma redução na carga bacteriológica de cerca de 100% do valor inicial de Escherichia coli (2 <FONT FACE=Symbol>´</FONT> 10³ NMP per 100 mL) para soluções de água sintética, e, 80% do valor inicial de Escherichia coli (16.6 to 22.2 <FONT FACE=Symbol>´</FONT> 10³ MPN per 100 mL) para água in natura, através da fotocatálise heterogênea usando TiO2.
TiO2-assisted heterogeneous photocatalysis and photolysis were evaluated for the disinfection of water samples using a glass reactor with immobilized TiO2 (catalyst), solar light and E. coli as an indicator microorganism of the efficiency of disinfection. Parameters such as color and turbidity of the water, level of coliform bacteria (by the Colilert® method), inclination angle of the solar reactor, solar light intensity, flow rate and retention time were controlled during the experiments. Two different operational modes were used for the solar reactor: single pass mode and recirculation mode. First, synthetic water was used in the disinfection tests as a model system; second, tests were conducted using natural samples specifically groundwater collected from a lake and a well. In bacterial suspensions in synthetic water in the absence of color and turbidity, heterogeneous photocatalysis was responsible for the reduction of approximately 100% of the initial concentration of E. coli. Only a 56.5% reduction was obtained by photolysis during the same solution recirculation time, which indicated a better efficiency using the catalyst. From the natural samples, total inactivation was not achieved in the studied cases. However, photocatalysis using TiO2/solar light was shown to be quantitatively efficient in the destruction of the total coliforms in water, reaching values up to around 80% inactivation in natural waters with initial levels of total coliforms ranging from 16.6 to 22.2<FONT FACE=Symbol>´</FONT>10³ MPN per 100 mL.
