Abstract The total cost of processes that use lipases can be reduced by using the mycelium or not purified crude extract with lipolytic activity. These catalysts can be used without complex purification and can show desirable properties to emerging applications like hydrolysis and biotransformation of vegetable oils. To explore the potential of multiple forms of lipases (produced by the same filamentous fungus species), this work investigated intracellular and extracellular lipases produced by Trichoderma koningii (intracellular lipases were never studied), Penicillium spp. F02 (isolated from industrial grease trap) and Aspergillus niger. For this, batch fermentations were conducted for 96h using olive oil as a carbon source. The submerged culture was interrupted with vacuum filtration to isolate the mycelium and crude extract. The lipolytic activities were determined by the hydrolysis method of olive oil. The best results were obtained for intracellular activity with T. koningii (81.5 U/g of dry biomass) and for extracellular activity with A. niger (10.6 U/mL of crude extract). Both were attained in 96h of fermentation. T. koningii showed an accumulation of intracellular lipases and a depreciated extracellular activity in the initial 24 hours, which can be reasonable indications that there is strong retention of lipases in the mycelium. Statistical analysis within the range of 37°C to 44°C associated with pH from 7.8 to 8.0 favored the activity of lipases from T. koningii. All these findings are promising for future studies in the enzymatic synthesis of biodiesel with lipases (intracellular and extracellular) from T. koningii.

RESUMO Introdução: A pré-eclâmpsia (PE) é definida pelo desenvolvimento de hipertensão arterial associada à proteinúria após a semana de gestação em mulheres previamente normotensas. A interleucina 17A (IL17A) é um potente indutor de inflamação tecidual, e polimorfismos no gene IL17A podem modular a expressão gênica e afetar o funcionamento das células Th17, contribuindo para a suscetibilidade à PE. Objetivo: Investigar os polimorfismos rs4711998 A>G, rs8193036 C>T e rs2275913 A>G no gene IL17A em mulheres com PE. Métodos: Trata-se de um estudo do tipo caso-controle, composto por 263 mulheres, sendo 89 diagnosticadas com PE e 174 do grupo-controle. Os polimorfismos investigados foram avaliados a partir do ácido desoxirribonucleico (DNA) genômico extraído do sangue periférico pela técnica de discriminação alélica por reação em cadeia da polimerase (PCR) em tempo real. O risco de os polimorfismos do gene IL17A contribuírem com a PE foi avaliado pelo modelo de herança através da regressão logística. O poder estatístico apresentou 99,5% para a detecção de associação. A significância estatística foi definida como p < 0,05. Resultados: As frequências genotípicas, assim como a análise de regressão logística múltipla, não foram estatisticamente significativas para os polimorfismos rs3761549 C>T, rs3761548 A>C e rs2232365 A>G do gene IL17A. Não foi observada associação entre nenhum dos haplótipos dos polimorfismos investigados e o risco de desenvolvimento de PE. Conclusão: Não há associação entre as frequências alélicas e genotípicas, os modelos de herança e os haplótipos dos polimorfismos rs4711998 A>G, rs8193036 C>T e rs2275913 A>G do gene IL17A e a PE.
ABSTRACT Introduction: Preeclampsia is defined by the development of hypertension associated with proteinuria after the 20th week of gestation in previously normotensive women. IL17A is a potent inducer of tissue inflammation and polymorphisms in the IL17A gene can modulate gene expression and affect the functioning of Th17 cells, strengthening susceptibility to preeclampsia. Objective: To investigate the polymorphisms rs4711998 A>G, rs8193036 C>T and rs2275913 A>G in the IL17A gene in women with preeclampsia. Methods: This is a control case study, composed of 263 women, 89 with preeclampsia and 174 of the control group. The polymorphisms investigated by real time polymerase chain reaction (PCR) allele discrimination technique. The risk of IL17A polymorphisms contributing to preeclampsia was assessed by the inheritance model through logistic regression. Statistical power presented 99.5% for association detection. Statistical significance was defined as p < 0.05. Results: Genotype frequencies as well as multiple logistic regression analysis were not statistically significant for the rs4711998 A>G, rs8193036 C>T and rs2275913 A>G polymorphisms of the IL17A gene. No association was found between any haplotypes of the polymorphisms investigated and the risk of developing PE. Conclusion: There is no association between the allele frequencies, genotype, inheritance models and haplotypes of the rs4711998 A>G, rs8193036 C>T and rs2275913 A>G polymorphisms of the IL17A gene and PE.
RESUMEN Introducción: La preeclampsia (PE) se define como hipertensión arterial asociada a proteinuria después de la semana 20 de gestación en mujeres anteriormente normotensas. La interleucina 17ª (IL17A) es un inductor potente de inflamación tisular y polimorfismos del gen IL17A pueden modular la expresión génica y afectar las funciones de las células Th17, aumentando la susceptibilidad a la PE. Objetivo: Investigar los polimorfismos rs4711998 A>G, rs8193036 C>T y rs2275913 A>G del gen IL17A en pacientes con PE. Métodos: Se realizó un estudio de casos y controles, compuesto por 263 mujeres: 89 diagnosticadas con PE y 174 del grupo de control. Se evaluaron los polimorfismos investigados a partir del ácido desoxirribonucleico (ADN) genómico aislado de la sangre periférica por reacción en cadena de la polimerasa (PCR) en tiempo real para discriminación alélica. El riesgo de los polimorfismos del gen IL17A para la PE fue evaluado según el modelo de herencia empleando regresión logística. El poder estadístico presentó 99,5% para la detección de asociación. Resultados: Las frecuencias genotípicas, así como el análisis de regresión logística múltiple, no fueron estadísticamente significativas para los polimorfismos rs3761549 C>T, rs3761548 A>C y rs2232365 A>G del gen IL17A. Ningún haplotipo de los polimorfismos investigados mostró asociación con el riesgo de desarrollo de PE. Conclusión: No hay asociación entre las frecuencias alélicas y genotípicas, los modelos de herencia y los haplotipos de los polimorfismos rs4711998 A>G, rs8193036 C>T y rs2275913 A>G del gen IL17A y la PE.

This study aims to assess a new approach for concomitant valorization of two industrial wastes - raw glycerin and spent brewer’s yeast - for the bioproduction of valuable carotenoids. Microbial pigments have numerous applications in the food and cosmetic industries. First, four cultures of yeasts were screened using pure glycerol and either (NH4)2SO4 or urea as carbon and nitrogen sources, respectively. The ability of the best performing culture to accumulate carotenoids was investigated in a medium in which only wastes were supplemented as carbon and nitrogen sources. All the fermentations were carried out in 500 mL-Erlenmeyer flasks containing 150 mL of the medium. Microbial culture was incubated at 30 °C and 150 rpm for 120 h. Particularly, R. marina was the strain with the biggest potential to produce total carotenoids (up to 420 μg g-1). The four major carotenoid pigments identified by LCAPCI-MS were β-carotene, γ-carotene, torulene, and torularhodin. All fermentation assays and all the determinations were performed in triplicate. Results show that the bioprocess proposed in this work is technically and environmentally feasible, and sustainable. The simultaneous use of raw glycerin and spent brewer’s yeast for the production of carotenoids by R. marina is reported for the first time.

RESUMO: As cutias são animais silvestres de pequeno porte, cujo peso corpóreo pode chegar até 4kg, e existem em todo território brasileiro. São considerados importantes dispersores de sementes, especialmente para árvores de grande porte, existindo espécies que dependem quase que exclusivamente destas para sua distribuição territorial. Este trabalho teve por objetivo a caracterização ultrassonográfica modo B e Doppler dos órgãos abdominais de cutias hígidas criadas em cativeiro. Foram utilizadas 15 cutias, contidas quimicamente, oriundas do Núcleo de Estudos e Preservação de Animais Silvestres - NEPAS, CCA-UFPI, submetidas a exame ultrassonográfico em modo B e Doppler. A parede da vesícula urinária presentou-se hiperecogênica, fina, lisa e regular em todo seu trajeto anatômico, com espessura média de 0,09±0,03cm. Os rins demonstraram ecotextura fina e homogênea, ecogenicidade global preservada, hipoecogênico em relação ao baço e isoecogênico ou discretamente hipoecogênico em relação ao fígado. O traçado em Doppler espectral mostrou pico sistólico e diastólico, amplo e afilado, exibindo baixa resistência de fluxo, com uma porção diastólica contínua e cheia, que diminui gradativamente no decorrer da diástole (75,83±1,42cm/s para o rim direito e 80,43±1,22 cm/s para o esquerdo. A adrenal direita apresentou uma variação de comprimento entre 0,61 a 1,18cm e diâmetro variando entre 0,17 a 0,32cm, enquanto a adrenal esquerda evidenciou comprimento de 0,62 a 1,16 e diâmetro de 0,14 a 0,25cm. O baço das cutias mostrou formato filiforme, com polos pontiagudos e diâmetro de 1,02±0,18cm. O fígado da cutia ocupa todo o espaço cranial da cavidade abdominal, em contato direto com o diafragma. O fluxo vascular intrahepático permitiu individualizar as veias porta (VP) e veias hepáticas (VH). As veias porta foram distinguidas, particularmente pelo padrão ecogênico de suas paredes, quando comparadas com as veias hepáticas. A espessura do pâncreas foi de 0,51±0,1cm e o ducto pancreático mediu 0,12±0,02cm. O estômago relaciona-se à esquerda com o baço e a direita com o duodeno proximal e colón transverso. Sua espessura de parede mensurada foi de 0,16±0,05cm. A aorta abdominal possui diâmetro de 0,43±0,04cm e fluxo vascular de 95,2±2,16cm/s. Este estudo caracterizou os órgãos e vasos sanguíneos abdominais de cutias, por meio de ultrassonografia modo B e Doppler, o que permitiu definir o tamanho, formato, posição, ecogenicidade, ecotextura dos constituintes anatômicos, além de estabelecer valores de referência para a rede vascular e fluxo sanguíneo na espécie.
ABSTRACT: Agoutis are small-sized wild animals whose body weight can reach up to 4kg, and are found throughout Brazil. They are considered important seed dispersers, especially for big trees and there are species that rely almost exclusively on these animals for their territorial distribution. The objective of the present study was B scan and Doppler ultrasound characterization of the abdominal organs of healthy agoutis reared in captivity. Fifteen agoutis, chemically restrained, were used from the Nucleus for Wild Animal Studies and Conservation (Núcleo de Estudos e Preservação de Animais Silvestres - NEPAS), CCA-UFPI, submitted to B scan and Doppler ultrasound examination. The urinary bladder wall was hyperechogenic, thin, smooth and regular throughout its anatomic path, with 0.09±0.03cm mean thickness. The kidneys showed fine and homogeneous echotexture, preserved global echogenicity, hyperechogenic in relation to the spleen and isoechogenic or discreetly hyperechogenic in relation to the liver. The spectral Doppler trace showed systolic and diastolic peaks, wide and thread-like, with low flow resistance and a continuous and full diastolic portion that decreased gradually during the diastole (75.83±1.42cm/s, for the right kidney and 80.43±1.22cm/s, for the left kidney). The right adrenal gland was 0.61-1.18cm long and 0.17-0.32cm in diameter, while the left adrenal gland was 0.62-1.16 long with 0.14-0.25cm diameter. The agouti spleen was filiform in shape, with pointed poles and 1.02±0.18cm in diameter. The agouti liver occupied all the abdominal cavity cranial space in direct contact with the diaphragm. The intrahepatic vascular flow allowed individualization of the portal vein (PV) and hepatic vein (HV). The portal veins were distinguished from the hepatic veins mainly by their wall echogenic pattern. The pancreas was 0.51±0.1 cm thick and the pancreatic duct measured 0.12±0.02cm. The stomach was placed to the left the spleen and to the right of the proximal intestine and the transversal colon and the walls were 0.16±0.05cm thick. The abdominal aorta was 0.43±0.04cm in diameter and showed 95.2±2.16cm/s vascular flow. This study characterized agouti organs and abdominal blood vessels by B scan and Doppler ultrasound, that permitted definition of the size, shape, position, echogenicity and echotexture of the anatomic constituents and established reference values for the vascular network and blood flow in the species.

Metabolomics is an emerging and promising omics approach used to understand biological mechanisms. By untargeted and targeted metabolomics analyses, metabolites are determined in biological samples (fluids, cells, tissues, etc.) by comparison of control groups with altered groups, undergoing different therapies, submitted to differing stress levels, dietary modulation, or promoted by a disease, or specific condition, etc., using sophisticated analytical techniques, and advanced data treatment and statistical analyses. In this review, the concepts involved in metabolomics studies were presented, describing in details all steps involved in the metabolomics workflow, for untargeted and targeted strategies. Finally, the potential of metabolomics is illustrated by applications in representative areas: clinical, environmental, food and nutrition, forensic toxicology, microbiology, parasitology, plants, and sports. Relevant reviews were compiled to characterize each of these areas, and a corresponding application of untargeted and targeted metabolomics were described.

Abstract Aims: This article is a methodological description of a randomized clinical trial (ClinicalTrials.gov U1111-1181-4455) aiming to evaluate the time-course (monthly) and associations between blood pressure changes and other health-related adaptations in response to exercise training in hypertensive elderly. Methods: The patients will be randomized to a control or combined training group interventions (aerobic and resistance exercise), with monthly assessments in four months. Although, the changes in baseline blood pressure is the primary clinical outcome, the secondary outcomes include: body composition, cardiorespiratory fitness, muscle strength, arterial stiffness, baroreceptor sensitivity, cardiovascular autonomic modulation, inflammatory markers, oxidative stress, growth factors, tissue remodeling markers, metabolic profile, renal function, cognitive function and quality of life. Results: To support the understanding of the blood pressure changes in hypertensive elderly, a time-course of exercise-induced adaptations including cardiovascular and immunological adaptations are fundamental for research in this field. Conclusion: To investigate the time-course of combined training-induced adaptations including all the diverse aspects of health in hypertensive elderly a well-controlled protocol design is necessary, mainly to clarify the relationship between cardiovascular and immunological exercise-induced adaptations.

ABSTRACT In 2012, the first-generation protease inhibitors telaprevir (TVR) and boceprevir (BOC) were introduced in the Brazilian health system for treatment of chronic hepatitis C, after their approval by the National Committee for Health Technology Incorporation (CONITEC). However, these medicines were discontinued in 2015. The short period of use in therapy and their high cost require a discussion about the consequences for patients and for the health system of the early incorporation of new therapies. The article presents a qualitative analysis of the incorporation process of both medications in Brazil and the results of a multicenter study that included patients treated with BOC or TVR between January 2011 and December 2015 in five Brazilian cities. The study included 855 patients (BOC: n=247) and (TVR: n=608). The document analysis showed that CONITEC’s decision to incorporate BOC and TVR was based on results of phase III clinical trials that compared sustained virologic response (SVR) rates of patients treated with BOC and TVR with rates of those that received placebo. However, these studies included a low percentage of cirrhotic patients. The SVR rates observed in this multicenter study were worse than clinical trials pointed out (BOC: 45.6%; TVR: 51.8%), but similar to those achieved with previously adopted therapies. The discontinuation rate due to adverse events was (BOC: 15.4%; TVR: 12.7%). Based on these unsatisfactory results, the study brings a discussion that goes beyond the therapy outcomes, exploring the incorporation of these high-cost medicines and the related decision-making process, contributing to future decisions in medicine policies and in the treatment of chronic hepatitis C.

RESUMO: O eletrocardiograma computadorizado é um dos meios de diagnóstico utilizado para avaliação do coração e vem sendo cada vez mais presente na rotina veterinária. Este trabalho teve por objetivo definir o padrão eletrocardiográfico de cutias (D. prymnolopha) anestesiadas com cetamina e midazolam. Foram utilizadas 18 cutias clinicamente saudáveis, provenientes do Núcleo de Estudos e Preservação de Animais Silvestres (NEPAS) da Universidade Federal do Piauí. Os animais foram submetidos à contenção química com cloridrato de cetamina a 5% na dosagem de 15mg/kg associado ao midazolam, na dosagem de 1mg/kg, por via intramuscular. Os exames eletrocardiográficos foram realizados pelo método computadorizado, com o eletrocardiógrafo veterinário (Módulo de Aquisição de ECG Para Computador (ECG - PC versão Windows 95) Tecnologia Eletrônica Brasileira (TEB) composto por um circuito eletrônico ligado externamente a um notebook, e de um software instalado no disco rígido do computador. Os valores de duração do complexo QRS, intervalos PR e QT, comparados entre machos e fêmeas, apresentaram diferença significativa. Em milivoltagem a onda R foi o único parâmetro que apresentou diferença significativa entre machos e fêmeas. O peso dos animais também foi significativamente diferente entre os gêneros. O protocolo anestésico mostrou-se bem tolerado pelos animais deste experimento, não ocorrendo quadros de arritmias durante o tempo de monitoramento dos animais.
ABSTRACT: An electrocardiogram is a test that assesses heart electrical activity and is applied more frequently in the veterinary care of wild animals. The present study aimed to define the electrocardiogram pattern of agoutis (Dasyprocta prymnolopha Wagler, 1831) anesthetized with ketamine and midazolam. Eighteen clinically healthy agoutis (D. prymnolopha) were used from the Nucleus for Wild Animal Studies and Conservation (NEPAS) of the Federal University of Piauí, Brazil. The animals were chemically restrained with 5% ketamine hydrochloride at a dose of 15mg/kg and midazolam at a dose of 1mg/kg by intramuscular injection. Electrocardiogram tests were carried out by a computerized method with the veterinary electrocardiogram [Acquisition Model for Computer (ECG - PC version Windows 95) Brazilian Electronic Technology (TEB) consisting of an electronic circuit externally connected to a notebook computer with ECGPC-VET (TEB) software installed on the hard disc. In analysing the EKG results, significant differences were observed for QRS complex duration, PR and QT intervals and for R wave millivoltage between the genders; but we observed a significant influence of weight despite the gender. In the present experiment, the anaesthetic protocol was shown to be well tolerated by the agoutis, and no arrhythmias occurred during the time the animals were monitored. The reference values obtained should be used to better understand the cardiac electrophysiology of the species and for its clinical and surgical management.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.
