Exogenous amylase increases gas production and improves in vitro ruminal digestion kinetics of sorghum and corn grains

ABSTRACT The aim of this study was to evaluate the effect of exogenous amylase on gas production, in vitro dry matter digestibility (IVDMD), and in vitro digestion kinetics of sorghum (Sorghum vulgaris) and two corn hybrids of different grain textures. Ruminal fluid was collected from two rumen-fistulated cows receiving or not exogenous amylase (0.7g kg-1 of dry matter (DM basis)), provided to achieve 396 kilo Novo units kg-1 for amylase activity (DM basis). Gas production was measured after 1, 3, 6, 9, 12, 15, 18, 21, 24, 30, 36, 42 e 48 hours of incubation. Amylase increased gas production (mL) by 5.4%. Corn hybrids have higher in vitro dry matter digestibility than sorghum. Exogenous amylase increased the potential of gas production (A) (P=0.01). There was an effect of hybrid for IVDMD (P<0.01). The addition of exogenous amylase increases the in vitro gas production, improves fermentation kinetics, and increases the production of the ammonia nitrogen of corn and sorghum grains, but does not affect in vitro and dry matter digestibility or the short-chain fatty acids production.
RESUMO O objetivo deste estudo foi avaliar o efeito da amilase exógena na produção de gases, a digestibilidade in vitro da matéria seca (DIVMS) e a cinética de digestão in vitro de sorgo (Sorghum vulgaris) e de dois híbridos de milho de diferentes texturas de grãos. O líquido ruminal foi coletado de duas vacas fistuladas no rúmen recebendo ou não amilase exógena (0,7g kg-1 de matéria seca (MS)), fornecida para atingir 396 kg Novo unidades kg-1 para atividade de amilase (base na MS). A produção de gás foi medida após uma, três, seis, nove, 12, 15, 18, 21, 24, 30, 36, 42 e 48 horas de incubação. A amilase aumentou a produção de gás (mL) em 5,4%. Híbridos de milho apresentam maior DIVMS que o sorgo. A amilase exógena aumentou o potencial de produção de gás (A) (P=0,01). Houve efeito de híbrido para DIVMS (P<0,01). Amilase exógena aumenta a produção de gás in vitro, melhora a cinética da fermentação e aumenta a produção de nitrogênio amoniacal de grãos de milho e sorgo, mas não afeta a digestibilidade in vitro da matéria seca ou a produção de ácidos graxos de cadeia curta.

Epithelial cell migration is an essential response to enteric pathogens such as enteropathogenic Escherichia coli (EPEC). This study aimed to investigate the effects of EPEC infection on intestinal epithelial cell migration in vitro, as well as the involvement of type III secretion system (T3SS) and Rho GTPases. Crypt intestinal epithelial cells (IEC-6) were infected with EPEC strains (E2348/69, ΔescF, and the LDI001 strain isolated from a malnourished Brazilian child) and commensal E. coli HS. Wound migration and cell death assays were performed at different time-points. Transcription and expression of Rho GTPases were evaluated using real-time PCR and western blotting. Overall, EPEC E2348/69 reduced migration and increased apoptosis and necrosis levels compared to EPEC LDI001 and E. coli HS strains. Moreover, EPEC LDI001 impaired cell migration at a higher level than E. coli HS and increased necrosis after 24 hours compared to the control group. The different profiles of virulence genes between the two wild-type EPEC strains, characterized by the absence of espL and nleE genes in the LDI001, might explain the phenotypic results, playing significant roles on cell migration impairment and cell death-related events. Moreover, the type III secretion system is determinant for the inhibition of intestinal epithelial cell migration by EPEC 2348/69, as its deletion prevented the effect. Active Rac1 concentrations were increased in E2348/69 and LDI001-infected cells, while the T3SS-deficient strain did not demonstrate this activation. This study contributes with valuable insight to characterize the mechanisms involved in the impairment of intestinal cell migration induced by EPEC.

ABSTRACT The aim of this study was to verify the parasitologycal aspects of fish (Hoplias malabaricus), coming from São Bento city, Maranhão state. For this, 70 fish were used, verifying the parasitism indexes and coefficients. For the parasitological results nematodeos were identified, with zoonotical potential of the Contracaecum spp, Pseudoterranova spp, Eustrongylides spp; monogenetics Gyrodactirus spp, Dactylogirus spp and larva Acanthella of Acanthocephalo. Of the 70 (100%) fishes, 64 (91.4%) were sponged, with prevalence of 82.8% (58) for the nematodeo Contracaecum spp, 65.7% (46), Pseudoterranova spp, 7.1% (5), Eustrongylides spp, 71.4% (50), monogenetics and larva Acanthella of Acantocephalo, respectively. A maior amplitude de intensidade (1-30), coeficiente de variação foi verificado para o nematódeo Pseudoterranova spp., a intensidade média (7,3), índice de abundância (6,7), coeficiente de dominância (36,4) foi ao Contacaecum spp., todos os parasitos estavam no mesentério da cavidade abdominal, encistados em forma larval.The largest intensity width (1-30), variation coefficient was verified for the nematodeo Pseudoterranova spp, the medium intensity (7,3), abundance index (6,7), dominance coefficient (36,4) it went to the Contacaecum spp, all of the parasites were in the mesenterio of the abdominal cavity, encysted in larval form. The results show that fish from this region presented unsatisfactory sanitary conditions in parasitological aspect and can expose consumers to the risks of foodborne illnesses.

Antioxidant effect of simvastatin throught oxidative imbalance caused by lisdexamfetamine dimesylate

O presente estudo tem o objetivo de investigar diretamente os efeitos comportamentais e antioxidantes da sinvastatina em um modelo de mania bipolar induzida por dimesilato de lisdexanfetamina. Ratos Wistar foram tratados durante 30 dias com simvastatina. No 24º dia após o início do tratamento, foi administrada dimesilato de lisdexanfetamina durante 7 dias em cada rato. Os resultados sugerem que sinvastatina combinada com dimesilato de lisdexanfetamina induziu um aumento significativo na locomoção, e administração de dimesilato de lisdexanfetamina provoca um desequilíbrio oxidativo determinado por um aumento na peroxidação lipídica, oxidação de proteínas e alterações nas atividades de enzimas antioxidantes em áreas do cérebro; adicionalmente, na presença de sinvastatina a maioria destes efeitos foram bloqueados. Estes resultados contribuem para um melhor entendimento dos papeis críticos do tratamento dos distúrbios neuropsiquiátricos com dimesilato de lisdexanfetamina, associado com o aumento do estresse oxidativo e com alterações das enzimas antioxidantes. Tendo em vista o papel central exercido pelo dimesilato de lisdexanfetamina, o efeito antioxidante estabelecido pela terapia com sinvastatina é de grande interesse.
The present study aims to directly investigate the behavioral and antioxidant effects of simvastatin in a model of bipolar mania induced by lisdexamfetamine dimesylate. Wistar rats were treated for 30 days with simvastatin. On the 24th day after the start of treatment, each rat was administered lisdexamfetamine dimesylate for 7 days. The results suggest that simvastatin combined with lisdexamfetamine dimesylate induced a significant increased locomotion and lisdexamfetamine dimesylate administration causes an oxidative imbalance determined by an increment in lipid peroxidation, protein oxidation and alterations in the activities of antioxidant enzymes in brain areas; moreover, in the presence of simvastatin, most of these effects were prevented. These findings contribute to a better understanding of the critical roles of lisdexamfetamine dimesylate in the treatment of neuropsychiatric disorders, associated with increased oxidative stress and changes in antioxidant enzymatic defense. In view of the central role played by lisdexamfetamine dimesylate, the established antioxidant effect of simvastatin therapy is of major interest.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A artrite-encefalite caprina (CAE) é diagnosticada rotineiramente pela técnica de imunodifusão em gel de agarose (IDGA), que é considerada pouco sensível. Objetivou-se com este estudo padronizar testes de Elisa-i e Western Blot (WB) para diagnóstico precoce de anticorpos em caprinos contra CAEV e comparar os resultados obtidos nesses testes com a prova de IDGA. Para a padronização dos testes Elisa-i e WB, utilizaram-se diferentes concentrações e diluições de antígeno, soros e conjugado. No Elisa-i, adotaram-se microplacas rígidas com 96 poços, sendo a combinação de concentração de 0,5µg/poço de antígeno e diluições de 1:100 de soro e 1:1500 de conjugado a que apresentou melhor resultado. No WB foram utilizadas membranas de nitrocelulose, definindo-se as diluições de 1:50 de soro e 1:15000 de conjugado. Para avaliar o desempenho das técnicas, 222 amostras de soro caprino foram testadas e os dados obtidos foram comparados com o IDGA. A sensibilidade e a especificidade do Elisa-i/IDGA, WB/IDGA e WB/Elisa-i foram de 70% e 91%, 100% e 72,6%, 84,6% e 76,5%, concomitantemente. O índice Kappa desses testes foi de 0,35, 0,2 e 0,36, respectivamente. As técnicas de Elisa-i e WB apresentaram-se mais sensíveis que a IDGA, podendo ser utilizadas como ferramentas para o diagnóstico precoce da CAE.
Caprine arthritis-encephalitis (CAE) is routinely diagnosed with the Agarose Gel Immunodiffusion (AGID) technique, which is considered to have low sensitivity. The objective of this study was to standardize testing i-Elisa and Western Blot for early detection of antibodies against CAEV in goats and compare the results obtained in these tests with proof of AGID. For standardization of i-Elisa and WB, different concentrations and dilutions of antigen, sera and conjugate were used. In the i-Elisa, rigid microplate with 96 wells was adopted, and the combination that showed the best result was a concentration of 0.5µg/ well of antigen and dilutions of the serum of 1:100 and conjugate of 1:1500. In the WB nitrocellulose membranes were used, and the dilutions of the serum were defined at 1:50 and conjugate at 1:15000. To evaluate the performance of the techniques, 222 goat serum samples were tested and the data were compared with the AGID. The sensitivity and specificity of Elisa-i/IDGA, WB/AGID and WB/Elisa-i were 70% and 91%, 100% and 72.6%, 84.6% and 76.5%, concomitantly. The Kappa index of these tests was 0.35, 0.2 and 0.36, respectively. The i-Elisa and WB techniques were more sensitive than the AGID and can be used as tools for early diagnosis of CAE.

Métodos de secagem e rendimento dos extratos de folhas de Plectranthus barbatus (boldo-da-terra) e P. ornatus (boldo-miúdo)

Plectranthus sp. pertence à família Lamiaceae e inclui cerca de 300 espécies. Duas delas, P. barbatus (PB) e P. ornatus (PO) destacam-se pelo interesse medicinal. As principais substâncias encontradas em Plectranthus sp. são os diterpenóides, óleos essenciais e compostos fenólicos. A melhor forma de utilizar as folhas de PB e PO é a fresco; porém, na comercialização há necessidade de recorrer a processos de secagem. Entretanto, isto pode interferir no princípio ativo, desta forma, estudos para determinar o melhor método gravimétrico são importantes. O objetivo deste estudo foi avaliar qual o melhor método de secagem, onde haja maior perda de água possível, e calcular os rendimentos dos extratos visando avaliar se o método gravimétrico interfere na quantidade de extrato produzido. Desta forma, utilizou-se quatro métodos de secagem e após evaporação do solvente, fez-se o cálculo do rendimento dos extratos secos. O percentual de água foi superior em PB e PO (média geral de 90,12±8,22%). Dentre as secagens, em estufa de ventilação forçada (EVF) foi mais eficiente em PB (88,87%) e em estufa convencional (ECO) foi melhor em PO (95,13%). O maior rendimento do extrato seco em PB foi na secagem em camada delgada (CD) (20,96%), enquanto que em PO foi ECO (41,34%). Não houve correlação entre o método de secagem e o rendimento de extrato obtido.
Plectranthus sp. belongs to the family Lamiaceae and includes about 300 species. Two of them, P. barbatus (PB) and P. ornatus (PO), have medicinal interest. The main substances found in Plectranthus sp. are the diterpenoids, essential oils and phenolic compounds. The best way to use the leaves of PB and PO is cool; but for marketing there is a need of resort to drying processes. However, this may interfere on the values of active principle; thus, studies to determine the best gravimetric methods are important. The objective of this study was to evaluate the best method of drying, where there the greater loss of water possible, and calculate the percentage of the extracts to assess whether the gravimetric method interferes with the amount of extract produced. To this was done by drying four different methods ventilated dry (EVF), conventional drying (ECO), microwave (MCO) and layer thing (CD) and after evaporation of the solvent, was made the calculation the percentage of the dried extract. The water content was higher in PB and PO (overall average of 90.12±8.22%). Among the drying, EVF was more efficient for PB (88.87%) and ECO was better for PO (95.13%). The highest content of dry extract was for PB in to CD (20.96%), whereas for PO was in to ECO (41.34%). There is no relationship between the drying method and in the quantity of extract obtained.

O alto polimorfismo encontrado no lócus do gene da αS1-caseína em caprinos, classificado em quatro níveis de expressão - alto, médio, baixo e nulo -, está associado à produção de 3,6; 1,6; 0,6 e 0g/L/alelo, respectivamente. O estudo foi realizado para investigar possíveis variações na produção de leite e seus constituintes, no perfil de caseínas e na lipólise da gordura. Quarenta e quatro cabras foram distribuídas em cinco genótipos: dois homozigotos, um para alta (AA) e outro para produção intermediária (EE), e três heterozigotos chamados AE, AF e EF, para αs1-caseína. Para a lipólise, o leite foi subamostrado em quatro alíquotas que sofreram tratamento térmico no momento da ordenha e após 24h de resfriamento. Diferenças entre genótipos foram observadas para a produção de caseína e de suas frações. As demais variáveis não diferiram entre genótipos. O genótipo AA apresentou os maiores conteúdos de caseína (28,6g/L) e de αS1-cn (22,3%). Os demais genótipos apresentaram média de 20,4g/L. Os grupos AE e AF apresentaram média de 12,1, EE-10,1 e EF-9,1% de αS1-cn. O resfriamento do leite por 24 horas aumentou a taxa de lipólise no leite. A genotipagem das cabras para αS1-cn pode ser usada como ferramenta de seleção com objetivo de obter produtos lácteos com distintos perfis de proteínas.
A high polymorphism is found in the locus of goat αS1-casein gene and it is classified in four levels of expression, named high, medium, and low, associated with production of 3.6, 1.6, 0.6, and 0 g/L/allele, respectively. The study was conducted to investigate possible variations on milk yield and components, profile of casein, and lipolysis of fat. Forty-four goats were assigned to five distinct genotypes as two homozygous, one for high (AA) and the other for intermediate yield (EE); and three heterozygous named AE, AF, and EF for the αs1-casein. For lipolysis, milk was sampled in four aliquots which were treated soon after milking and 24 hours after cooling. Differences were observed for both casein yield and its fractions. No difference was found for other variables. The AA genotype presented the higher content of both casein (28.6g/L) and αS1-cn (22.3%). Other genotypes averaged 20.4g/L for casein content. Values of αS1-cn were 12.1% for heterozygous and 10.1 and 9.1% for both EE and EF genotype respectively. Cooling the milk for 24 hours increased the rate of lipolysis. Genotyping goats for the αS1-cn can be used as a tool for selecting animal targeting milk products with distinct profiles of proteins.

OBJETIVO: O objetivo deste trabalho é o de verificar a aplicabilidade do método imunohistoquímico na quantificação das diferentes formas das fibras elásticas em prega vocal humana. FORMA DE ESTUDO: coorte transversal. MATERIAL E MÉTODO: Foram seguidos os seguintes critérios de inclusão: idade entre 25 e 40 anos, gênero masculino, cor branca, morte causada por ferimento de arma de fogo, menos de doze horas de morte, ausência de intubação traqueal e de trauma na região cervical e que, por análise microscópica, não apresentassem qualquer alteração da mucosa das pregas vocais. Por estes critérios dez pregas vocais foram obtidas e selecionou-se, aleatoriamente, uma prega vocal que pertencia a um indivíduo de 28 anos. A prega vocal foi transversalmente seccionada em nove regiões e três cortes de cada fragmento foram utilizados para a realização das colorações Verhoeff e resorcina-fuchsina de Weighert e para a realização do estudo imunohistoquímico. Realizou-se quantificação colorimétrica das fibras elásticas. RESULTADO: As camadas intermediária e profunda da prega vocal apresentam valores muito superiores aos da camada superficial, nas colorações histológicas. A quantidade de tropoelastina identificada pelos anticorpos não apresentou grandes diferenças entre os valores da camada superficial e os da camada intermediária e profunda. CONCLUSÃO: A imunohistoquímica é uma técnica que identifica, em prega vocal humana, todas as formas de fibras elásticas e que também possibilita a realização de medidas objetivas.
AIM: Verify the use of immunohistochemistry as a method to measure all forms of elastic fibers at human vocal folds. STUDY DESIGN: transversal cohort. MATERIAL AND METHOD: We collected vocal folds following these criterion: age between 25 and 40, Caucasian men, dead by gun shot, within 12 hours of death, without instrumentation of the larynx or suspicion of neck injury and without mucosal lesions noted by microscopy. Ten vocal folds were collected and one, of a man aged 28 years, was selected to study. The vocal fold was transversely cut in 9 regions and in each segment three slides were made. These slides were stained by Verhoeff and Weighert's resorcin-fuchsin and used for immunohistochemistry. The elastic compound was measured by colorimetric software analysis. RESULTS: In Verhoeff and Weighert's resorcin-fuchsin, the intermediate and deep layer showed values higher than those of the superficial layer. The amount of tropoelastin identified by the antibody at the superficial layer was close to those of intermediate and deep layer. CONCLUSION: Immunohistochemistry is a method that can identify and measure all forms of elastic fibers at human vocal fold.

Deficiência de hormônio do crescimento após radioterapia por meduloblastoma na infância: relato de caso

A radiação do crânio para tratamento das neoplasias do sistema nervoso central na infância pode evoluir com sequelas neuroendócrinas, sendo a deficiência de hormônio do crescimento (GH) com retardo do crescimento linear, uma das mais frequentes. Relatamos o caso de menino de 10 anos com cefaléia occipital associada a vertigem, náuseas e vômitos. A tomografia do crânio demonstrou processo expansivo no hemisfério cerebelar esquerdo, que foi retirado cirurgicamente. O exame histopatológico revelou meduloblastoma e o paciente foi submetido a radioterapia crânio-espinhal. Evoluiu sem recidiva da neoplasia e sem déficits neurológicos durante 4 anos. Apresentou retardo do crescimento estatural, sendo confirmada a hipótese de deficiência de GH. Atualmente, encontra-se em uso de GH 0,1 U/kg/dia, tendo apresentado incremento de 4cm na estatura em 6 meses. O presente caso destaca a importância do acompanhamento criterioso de pacientes submetidos à radiação do crânio para tratamento oncológico na infância, visto que podem evoluir com deficiências neuroendócrinas e serem beneficiados com reposição hormonal.
Craniospinal radiation therapy for treatment of brain tumors may result in growth hormone (GH) insufficiency with resultant linear growth retardation, one of the most common complications. We report the case of a 10-year-old boy presenting headache associated to vertigo, nausea and vomiting. A CT scan showed an homogeneous mass in the left cerebelar hemisphere, that was surgically removed. The histopathological examination revealed medulloblastoma and the patient was submitted to craniospinal radiation. He did not present tumor recurrence nor neurological or cognitive deficits during 4 years, but evolved to short stature due to GH deficiency. Nowadays, he is being receiving GH 0,1 U/kg/day and has presented 4cm stature increment after 6 months. The present case highlights the importance in carefully monitoring of children after cranial radiation for oncologic treatment, because they may develop hormonal deficiencies, that can be successfully replaced.
