Pyrazolyl-Tetrazoles and Imidazolyl-Pyrazoles as Potential Anticoagulants and their Integrated Multiplex Analysis Virtual Screening

This article reports a novel virtual screening algorithm seeking the rational identification of novel lead anticoagulants. Seven 5-(3-methyl-1-aryl-1H-pyrazol-4-yl)-1H-tetrazoles and seven novel 1-aryl-4-(4,5-dihydro-1H-imidazol-2-yl)-3-methyl-1H-pyrazoles were obtained in three steps starting from arylhydrazine hydrochlorides as raw materials in good yields: 50-72% and 50-85%, respectively. All compounds were submitted to an in silico target-base pipeline named integrated multiplex analysis virtual screening (IMA-VS), which comprises the evaluation of their (i) fitting physicochemical properties to the chemical environment of the target enzyme; (ii) active-site homing electrostatic potential to the target enzyme; (iii) structural fitting to the target active site through molecular docking; and (iv) overall absorption, distribution, metabolism, excretion and toxicity (ADMET) profile. After the virtual selection of potential anticoagulant hits, all molecules were synthesized and candidates were evaluated in vitro for their anticoagulant and hemolytic profile. The most promising candidate pointed out by IMA-VS was compound 1-(3',4'-dichlorophenyl)-4-(4,5-dihydro-1H-imidazol-2-yl)-3-methyl-1H-pyrazole that shown to display factor Xa (FXa)-specific inhibitory activity in vitro, acting as an uncompetitive inhibitor with an inhibition constant (Ki) = 61.16 ± 12.96 µM, in addition to the lowest hemolytic activity of the series. Further experiments revealed the antithrombotic activity of this compound in an in vivo model of arterial thrombosis induced by FeCl3.

A method based on QuEChERS (quick, easy, cheap, effective, rugged, and safe) extraction and gas chromatography-mass spectrometry (GC-MS) detection was described for the pesticides dichlorvos, disulfoton, ethoprophos, parathion methyl, fenchlorphos, chlorpyrifos, azinphos methyl and prothiofos in sapodilla. For all compounds studies, it was observed a strong matrix effect showing the need to use matrix matched calibration strategy. Method was validated, and good linearity (R > 0.99) was obtained for all pesticides studied with limits of detection (LODs) and quantification (LOQs) ranging from 0.01 to 0.06 mg kg-1 and 0.03 to 0.2 mg kg-1, respectively. Recovery studies were performed at different levels (0.08, 0.10, 0.14, 0.20, 0.35 and 1.17 mg kg-1) and showed good results (between 70 and 120% with relative standard deviation (RSD) < 20%). A statistical test was applied to the coefficients of the analytical curves obtained in the sapodilla matrix. Analyses of commercial samples showed chlorpyrifos were detected in about 70 and 33% for fruit and pulps samples, respectively. It should be noted that chlorpyrifos is not permitted in sapodilla crops by ANVISA and EC guidelines.

Spatial Distribution of Eggs of Alabama argillacea Hübner and Heliothis virescens Fabricius (Lepidoptera: Noctuidae) on Bt and non-BtCotton

RESUMO Dentre as opções para o controle de lagartas de Alabama argillacea(Hubner, 1818) e Heliothis virescens (Fabricius, 1781) na cultura algodoeira, aplicações de inseticidas e o controle biológico têm sido amplamente usados. O algodão transgênico 'Bt' tem se inserido como uma alternativa extremamente viável, porem não se sabe com exatidão como as plantas transgênicas afetam as populações de organismos que se inter-relacionam em um agroecossistema. Nesse sentido, é importante saber como a disposição espacial de insetos pragas ou benéficos é afetada, pois pode alterar o método de amostragem dessas espécies na cultura, além de sua forma de controle. Este estudo foi conduzido buscando investigar o padrão da distribuição espacial de ovos de A. argillacea e de H. virescens em cultivar DeltaOpalR (convencional) e DP90BR (algodão-Bt)A coleta dos dados ocorreu durante o ano agrícola 2006/2007 em duas áreas de 5.000 m2 localizada na região do distrito de Nova América, município de Caarapó. Em cada área amostral, composta por 100 parcelas de 50 m2, foram realizadas 15 avaliações com contagens dos ovos das pragas em analise, em duas plantas por parcela. Os índices de agregação (razão variância/media, índice de Morisita e Expoente k da Distribuição Binomial Negativa) e o teste de qui-quadrado, com o ajuste dos valores encontrados e esperados as distribuições teóricas de frequência (Poisson, Binomial Negativa e Binomial Positiva), mostraram que em ambas cultivares a distribuição espacial de posturas dessas espécies estão distribuídas de acordo com o modelo de distribuição espacial agregada, ajustando-se ao padrão da distribuição binomial negativa.
ABSTRACT Among the options to control Alabama argillacea (Hübner, 1818) and Heliothis virescens (Fabricius, 1781) on cotton, insecticide spraying and biological control have been extensively used. The GM'Bt' cotton has been introduced as an extremely viable alternative, but it is yet not known how transgenic plants affect populations of organisms that are interrelated in an agroecosystem. For this reason, it is important to know how the spatial arrangement of pests and beneficial insect are affected, which may call for changes in the methods used for sampling these species. This study was conducted with the goal to investigate the pattern of spatial distribution of eggs of A. argillacea and H. virescens in DeltaOpalTM (non-Bt) and DP90BTMBt cotton cultivars. Data were collected during the agricultural year 2006/2007 in two areas of 5,000 m2, located in in the district of Nova América, Caarapó municipality. In each sampling area, comprising 100 plots of 50 m2, 15 evaluations were performed on two plants per plot. The sampling consisted in counting the eggs. The aggregation index (variance/mean ratio, Morisita index and exponent k of the negative binomial distribution) and chi-square fit of the observed and expected values to the theoretical frequency distribution (Poisson, Binomial and Negative Binomial Positive), showed that in both cultivars, the eggs of these species are distributed according to the aggregate distribution model, fitting the pattern of negative binomial distribution.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Para identificar alternativas para a fumigação de solo em processos de biorremediação, foi avaliada a adição de benomil e de óleo vegetal no crescimento de Lentinus crinitus e fungos mitospóricos. Benomil pode ser usado como alternativa ao brometo de metila. A adição de óleo vegetal favoreceu o crescimento de L. crinitus.
In order to identify alternatives for soil fumigation in bioremediation process, the addition of benomyl and vegetable oil on the growth of L. crinitus and mitosporic fungi were evaluated. Benomyl can be used as an alternative to methyl bromide. Addition of vegetable oil favors the growth of L. crinitus.

Avaliou-se o efeito de concentrações de P e épocas de coleta sobre o crescimento, nutrição mineral e teor de óleo essencial da menta (Mentha piperita L.). O delineamento experimental foi em blocos casualizados com parcelas subdivididas, em esquema fatorial 2x5, sendo 2 épocas de coleta (65 e 95 dias de cultivo (DC)) e 5 concentrações de P na solução nutritiva (6; 12; 18; 24 e 30 mg L-1), com 4 repetições. Foi determinada a massa seca de folhas (MSF), caules (MSC), raízes (MSR) e total (MST) e as relações folha:caule (F:C) e parte aérea:raiz (PA:R). Também, foi determinado o teor de óleo essencial em folhas frescas. Os níveis críticos foliares e acúmulo de nutrientes na parte aérea da menta foram estimados por meio da análise química das folhas e caules. As concentrações de P na solução influenciaram o crescimento e a produção de óleo essencial pela menta. Na coleta realizada aos 65 DC, apenas a MSF foi influenciada pelo P, não se observando efeito sobre os teores de óleo essencial. Aos 95 DC, o aumento da concentração de P promoveu aumentos significativos na massa seca das partes e do total da menta e, também, nos teores de óleo essencial, sendo o teor máximo (2,192 dag kg-1) obtido com 19,50 mg L-1 de P. As maiores concentrações de P avaliadas (24 e 30 mg L-1), promoveram aumentos na produção de matéria fresca e seca da parte aérea da menta, mas, reduziram os teores de óleo essencial, devido ao efeito de diluição, não significando, portanto, ganhos de produção de óleo por planta. Os teores foliares dos nutrientes associados ao teor máximo de óleo aos 95 DC foram em g kg-1: N=37,2; =3,9; K=21,2; Ca=9,3; Mg=3,8; S=3,0; e, em mg kg-1: B=35; Cu=8; Fe=323; Mn=145 e Zn=22. A exigência nutricional aos 95 DC relacionada ao teor máximo de óleo essencial seguiu a seqüência: N>K>Ca>P>Mg>S>Fe>Mn>B>Zn>Cu.
The effect of P concentrations and harvest period upon the growth, mineral nutrition and essential oil content of mint (Mentha piperita L.) was evaluated. The experimental design was of randomized blocks in a split-plot scheme, in a 2x5 factorial with four replications, being two harvest periods (65 and 95 days of cultivation (DC)) and five concentrations of P in the nutrient solution (6; 12; 18; 24 and 30 mg L-1) evaluated. The leaves dry weight (MSF), stems dry weight (MRS), roots dry weight (MSR), total dry weight (MST), leaf:stem ratio (F:C) and aerial part:root ratio (PA:R) were evaluated. The essential oil content in fresh leaves was determined. The critical levels and nutrient accumulation in leaves and stems were estimated through leaves and stems chemical analysis. The level of P increased the leaf dry matter production 65 DC. At 95 DC, the P concentration in the nutrient solution, did significantly increase plant growth and essential oil content, the maximum essential oil content being (2.192 dag kg-1) obtained with 19.48 mg L-1 of P. The higher P concentration in nutrient solution (24 and 30 mg L-1), increased the fresh and dry matter of the aerial portion of the plants, but, decreased the essential oil content. Due to the dilution effect, the increase of fresh and dry matter production did not result in an increase in the oil yield per plant. The critical levels of nutrients in the leaves corresponding to the maximum essential oil content at 95 DC in g kg-1 were: N=37,2; P=3,9; K=21,2; Ca=9,3; Mg=3,8; S=3,0 and, in mg kg-1: B=35; Cu=8; Fe=323; Mn=145 and Zn=22. The nutrients requested at 95 DC related to the maximum essential oil content was: N>K>Ca>P>Mg>S>Fe>Mn>B>Zn>Cu.

Da casca do caule de Picramnia teapensis Tul. foram isolados dois novos benzoatos triterpênicos, o 3alfa,7beta-dibenzoato do ácido lup-20(29)-en-28-óico e o 7beta-benzoato do ácido 3alfa-hidroxi-lup-20(29)-en-28-óico. A elucidação estrutural desses compostos foi fundamentada na análise dos dados espectroscópicos. Outros compostos conhecidos, beta-sitosterol, estigmasterol, lupeol e epilupeol, foram identificados em mistura por CG-EM. Nenhum dos triterpenos benzoilados mostrou efeito inibitório no crescimento in-vitro do Leucoagaricus gongylophorus (Fisher), também citado como Leucocoprinus gongylophorus (Heim), syn Rozites gongylophora (Möller), o fungo simbionte cultivado pelas formigas cortadeiras de folhas Atta sexdens L.
Two new benzoic acid esters of triterpene alcohols [lup-20(29)-en-28-oic acid 3alpha,7beta-dibenzoate and 3alpha-hydroxy-lup-20(29)-en-28-oic acid 7beta-benzoate] were isolated from the stem bark of Picramnia teapensis Tul. The structures of these compounds were established on the basis of spectral analyses. Other known compounds, beta-sitosterol, estigmasterol, lupeol and epilupeol, were identified in mixture by GC-MS. The triterpene esters have not shown in-vitro inhibitory effect on the growth of Leucoagaricus gongylophorus (Fisher), referred also as Leucocoprinus gongylophorus (Heim), syn Rozites gongylophora (Möller), the symbiotic fungus cultivated by the leaf-cutting ant Atta sexdens L.
