El estudio de la estructura genética de poblaciones de plantas silvestres es esencial para su manejo y conservación. Varios marcadores de ADN e isoenzimas se han utilizado en este tipo de análisis. Con el fin de proporcionar una mejor comprensión de los resultados obtenidos y saber que marcador codominante elegir para futuros estudios en poblaciones naturales de Oryza glumaepatula, este trabajo busco evaluar y comparar dos marcadores de ADN, isoenzimas y microsatélites, en la diversidad y estructura genética de 13 poblaciones, destacando las similitudes y divergencias de cada marcador, así como la importancia relativa de los resultados en genética de poblaciones y conservación. Para los SSR, ocho loci SSR fueron evaluados, y los fragmentos se visualizaron utilizando el procedimiento de coloración con plata. Los análisis de isoenzimas se realizaron en geles de poliacrilamida, en los seis loci enzimáticos. Los loci SSR mostraron mayores niveles de diversidad genética que los loci isoenzimáticos, en promedio. La diferenciación genética entre los loci SSR (R ST=0.631, equivalente a F ST=0.533) fue inferior a la obtenida con las isoenzimas (F ST=0.772). Ambos marcadores mostraron alta desviación del equilibrio de Hardy-Weinberg (F IS=0.744 y 0.899, respectivamente, para SSR e isoenzimas). La tasa media aparente de cruzamiento para SSR ( =0.14) fue mayor que la obtenida con isoenzimas ( =0.043), aunque ambos marcadores detectaron niveles más bajos en la tasa de fecundación cruzada para la Amazonia, en comparación con la región del Pantanal. La estimación de número de migrantes también fue mayor para los SSR (Nm=0.219) que en isoenzimas (Nm=0.074). No se obtuvo ninguna correlación entre las distancias genéticas y geográficas para los SSR, y para las isoenzimas se obtuvo una correlación positiva entre las distancias genéticas y geográficas. Llegamos a la conclusión de que estos marcadores son divergentes en la detección de los parámetros de la diversidad genética en O. glumaepatula y que los microsatélites son más eficientes para detectar la información a nivel intra-poblacional, mientras que las isoenzimas son más potentes para detectar la diversidad entre poblaciones.

The study of the genetic structure of wild plant populations is essential for their management and conservation. Several DNA markers have been used in such studies, as well as isozyme markers. In order to provide a better comprehension of the results obtained and a comparison between markers which will help choose tools for future studies in natural populations of Oryza glumaepatula, a predominantly autogamous species, this study used both isozymes and microsatellites to assess the genetic diversity and genetic structure of 13 populations, pointing to similarities and divergences of each marker, and evaluating the relative importance of the results for studies of population genetics and conservation. A bulk sample for each population was obtained, by sampling two to three seeds of each plant, up to a set of 50 seeds. Amplified products of eight SSR loci were electrophoresed on non-denaturing polyacrylamide gels, and the fragments were visualized using silver staining procedure. Isozyme analyses were conducted in polyacrylamide gels, under a discontinuous system, using six enzymatic loci. SSR loci showed higher mean levels of genetic diversity (A=2.83, p=0.71, A P=3.17, Ho=0.081, He=0.351) than isozyme loci (A=1.20, p=0.20, A P=1.38, Ho=0.006, He=0.056). Interpopulation genetic differentiation detected by SSR loci (R ST=0.631, equivalent to F ST=0.533) was lower than that obtained with isozymes (F ST=0.772). However, both markers showed high deviation from Hardy-Weinberg expectations (F IS=0.744 and 0.899, respectively for SSR and isozymes). The mean apparent outcrossing rate for SSR ( =0.14) was higher than that obtained using isozymes ( =0.043), although both markers detected lower levels of outcrossing in Amazonia compared to the Pantanal. The migrant number estimation was also higher for SSR (Nm=0.219) than isozymes (Nm=0.074), although a small number for both markers was expected due to the mode of reproduction of this species, defined as mixed with predominance of self fertilization. No correlation was obtained between genetic and geographic distances with SSR, but a positive correlation was found between genetic and geographic distances with isozymes. We conclude that these markers are divergent in detecting genetic diversity parameters in O. glumaepatula and that microsatellites are powerful for detecting information at the intra-population level, while isozymes are more powerful for inter-population diversity, since clustering of populations agreed with the expectations based on the geographic distribution of the populations using this marker

Embora apenas uma pequena quantidade de água da Terra esteja reservada em corpos d'água da superfície continental, esses ambientes desempenham papel importante nos ciclos biogeoquímicos, conectando a superfície à atmosfera. O território brasileiro é recortado por uma densa rede fluvial e exibe um enorme número de lagos rasos. Impactos de natureza humana têm sido intensos modificadores de ecossistemas límnicos. A biodiversidade e os processos ecossistêmicos são fortemente modulados por forças sazonais fluvial e/ou precipitação. Essas macroforçantes ecológicas respondem às mudanças climáticas. As águas interiores são ecossistemas com elevada biodiversidade, promovem transferências de energia dentro da paisagem, conectando os ecossistemas, e atuam na manutenção de fluxos de matérias - animais, vegetais, nutrientes e inóculos. Esses ecossistemas controlam o clima numa escala regional. Neste capítulo, são descritas algumas respostas dos ecossistemas aquáticos às alterações climáticas, tanto conceitualmente como analisando os possíveis cenários de mudanças climáticas em diferentes regiões no Brasil. Potenciais sinais biogeoquímicos em diferentes ecossistemas límnicos brasileiros foram identificados. Os ecossistemas límnicos são pressionados pelas atividades do uso do solo, pela fragmentação da paisagem, pelo represamento e pelo desvio de rios, pela urbanização, pela carga de águas residuais e do nível de poluentes. Essas ações perturbadoras podem alterar os padrões biogeoquímicos nas águas interiores numa escala temporal mais curta quando comparada às mudanças climáticas. A manutenção da sustentabilidade das ecossistemas aquáticos brasileiros é urgente de modo a prevenir futuros eventos catastróficos.

Although only a small amount of the Earth's water exists as continental surface water bodies, this compartment plays an important role in the biogeochemical cycles connecting the land to the atmosphere. The territory of Brazil encompasses a dense river net and enormous number of shallow lakes. Human actions have been heavily influenced by the inland waters across the country. Both biodiversity and processes in the water are strongly driven by seasonal fluvial forces and/or precipitation. These macro drivers are sensitive to climate changes. In addition to their crucial importance to humans, inland waters are extremely rich ecosystems, harboring high biodiversity, promoting landscape equilibrium (connecting ecosystems, maintaining animal and plant flows in the landscape, and transferring mass, nutrients and inocula), and controlling regional climates through hydrological-cycle feedback. In this contribution, we describe the aquatic ecological responses to climate change in a conceptual perspective, and we then analyze the possible climate-change scenarios in different regions in Brazil. We also indentify some potential biogeochemical signals in running waters, natural lakes and man-made impoundments. The possible future changes in climate and aquatic ecosystems in Brazil are highly uncertain. Inland waters are pressured by local environmental changes because of land uses, landscape fragmentation, damming and diversion of water bodies, urbanization, wastewater load, and level of pollutants can alter biogeochemical patterns in inland waters over a shorter term than can climate changes. In fact, many intense environmental changes may enhance the effects of changes in climate. Therefore, the maintenance of key elements within the landscape and avoiding extreme perturbation in the systems are urgent to maintain the sustainability of Brazilian inland waters, in order to prevent more catastrophic future events.

OBJECTIVES: Helicobacter pylori-associated infection is common in South Africa, as in other developing countries. Antibiotic resistance is recognised as a major cause of treatment failure. We studied the susceptibility and resistance patterns of H. pylori to guide empiric treatment and prevent the emergence of resistance. METHODS: Two hundred H. pylori strains obtained from gastric biopsies of patients presenting with gastric-related morbidities attending Livingstone Hospital, Port Elizabeth, were evaluated for their susceptibility to seven antibiotics - metronidazole, clarithromycin, tetracycline, amoxicillin, gentamicin, ciprofloxacin and erythromycin. H. pylori was isolated following standard microbiology procedures, and susceptibility determined using the Kirby-Bauer disc diffusion and agar dilution methods. Comparisons of antimicrobial resistance rates with sex of the patients were determined using the chi-square test; a p-value of <0.05 was considered significant. RESULTS: Marked susceptibility was observed for ciprofloxacin (100%) and amoxicillin (97.5%), and good activity for clarithromycin (80%) and gentamicin (72.5%). However, marked resistance (95.5%) was observed for metronidazole. The minimal inhibitory concentration (MIC) ranged from 0.0625 µg/ml to 8 µg/ml. The lowest MIC, with a range of 0.0625 - 1 µg/ml, was recorded for ciprofloxacin, while the highest (5 - 8 µg/ml) was noted for gentamicin. CONCLUSION: Multidrug resistance was commonly encountered - a finding of clinical significance that calls for continuous surveillance of antibiograms to guide empiric treatment. We advocate the inclusion of ciprofloxacin in the treatment regimen of H. pylori infection in our study environment.

Knowledge of the genetic structure and diversity of natural populations is important in developing strategies for in situ and ex situ conservation. We used eight microsatellite loci to estimate genetic structure and investigate within and between population genetic variation in eleven Brazilian wild rice (Oryza glumaepatula) populations. The study showed the following genetic diversity parameters: average number of 3.1 alleles per locus; 77.3% polymorphic loci; 0.091 observed heterozygosity and 0.393 gene diversity. F-statistics detected by microsatellite loci were: F ST = 0.491 (and R ST = 0.608), F IS = 0.780 and F IT = 0.888. No population was in Hardy-Weinberg equilibrium. The estimated apparent outcrossing rate (0.143) indicated a predominance of self-fertilization. The gene flow values were low (Nm = 0.259 and 0.161 for F ST and R ST, respectively). Populations were spatially structured but without a correlation between genetic and geographic distances. Five populations (PG-4, PG-2, PU-1, SO-4, NE-18) were identified as priorities for conservation strategies. Populations from the Amazon biome showed heterogeneity with respect to intrapopulation diversity. The high level of genetic differentiation between populations and the high number of private alleles suggested that sampling should be carried out on a large number of O. glumaepatula populations for ex situ conservation purposes.

O estudo da estrutura genética de populações naturais é muito importante para a conservação e o uso da variabilidade genética disponível na natureza. Esta pesquisa relaciona-se com a análise da estrutura genética de populações a partir de dados moleculares reais e simulados. Visando estimar variâncias de estimativas de parâmetros pertinentes, o método de reamostragem bootstrap foi aplicado levando em conta diferentes unidades amostrais, a saber: indivíduos dentro de populações (I), populações (P) e indivíduos e populações concomitantemente (I, P). Os parâmetros considerados foram: o índice de fixação total (F ou F IT), o indice de fixação intrapopulacional (f ou F IS) e a divergência interpopulacional (teta ou F ST). O trabalho objetivou estimar a variância amostral das estimativas destes parâmetros para verificar se as variâncias de <IMG SRC="/img/fbpe/sa/v60n1/14549x09.gif">, <IMG SRC="/img/fbpe/sa/v60n1/14549x10.gif">e <IMG SRC="/img/fbpe/sa/v60n1/14549x11.gif">, obtidas pela reamostragem de indivíduos e populações concomitantemente (I, P) são equivalentes às obtidas pela soma (I+P) das variâncias estimadas reamostrando-se I e P separadamente. A equivalência foi verificada em todos os casos investigados, mostrando ser possível estimar as variâncias das estimativas de <IMG SRC="/img/fbpe/sa/v60n1/14549x09.gif">, <IMG SRC="/img/fbpe/sa/v60n1/14549x10.gif">e <IMG SRC="/img/fbpe/sa/v60n1/14549x11.gif">, para cada fonte de variação (unidade amostral) somando-as depois para estimar a variância total. O procedimento facilita o uso do método bootstrap em dados com estrutura hierárquica e permite mensurar a importância relativa de cada fonte de variação sobre a variância amostral total das estimativas dos parâmetros.

Studying the genetic structure of natural populations is very important for conservation and use of the genetic variability available in nature. This research is related to genetic population structure analysis using real and simulated molecular data. To obtain variance estimates of pertinent parameters, the bootstrap resampling procedure was applied over different sampling units, namely: individuals within populations (I), populations (P), and individuals and populations simultaneously (I, P). The considered parameters were: the total fixation index (F or F IT), the fixation index within populations (f or F IS) and the divergence among populations or intrapopulation coancestry (theta or F ST). The aim of this research was to verify if the variance estimates of <IMG SRC="/img/fbpe/sa/v60n1/14549x09.gif">, <IMG SRC="/img/fbpe/sa/v60n1/14549x10.gif">and <IMG SRC="/img/fbpe/sa/v60n1/14549x11.gif">, found through the resampling over individuals and populations simultaneously (I, P), correspond to the sum of the respective variance estimates obtained from separated resampling over individuals and populations (I+P). This equivalence was verified in all cases, showing that the total variance estimate of <IMG SRC="/img/fbpe/sa/v60n1/14549x09.gif">, <IMG SRC="/img/fbpe/sa/v60n1/14549x10.gif">and <IMG SRC="/img/fbpe/sa/v60n1/14549x11.gif">can be obtained summing up the variances estimated for each source of variation separately. Results also showed that this facilitates the use of the bootstrap method on data with hierarchical structure and opens the possibility of obtaining the relative contribution of each source of variation to the total variation of estimated parameters.

O fato de que os valores do índice de fixação intrapopulacional ( f ) e os das taxas de fecundação cruzada (t) mantém uma estreita relação entre si é amplamente conhecido. Resultados das análises de marcadores moleculares em populações naturais têm demonstrado uma elevada variação destes valores para locos diferentes avaliados em uma mesma população, sugerindo que outros fatores, além daqueles descritos no modelo de equilíbrio genético de Wright devem estar operando. Pelo uso de simulações, demonstra-se neste trabalho que a condição finita de uma população é suficiente para que os valores estimados de f passem a se estabilizar ao longo de um intervalo de variação e não mais em um único ponto. Demonstra-se ainda que variações nas taxas de fecundação cruzada ao longo das gerações amplificam substancialmente a magnitude deste intervalo. A correlação entre os valores estimados de f obtidos de locos diferentes nestas condições mostrou-se dependente dos valores das taxas de fecundação cruzada e da magnitude da variância destas taxas entre gerações, podendo ser nula sob condições de panmixia e taxas constantes. A condição finita das populações associada a pequenas flutuações na taxa média de fecundação cruzada de diferentes gerações, condições tipicamente encontradas na natureza, podem explicar as discrepâncias entre os valores estimados de f de locos diferentes, comumente reportadas na literatura. Há possibilidade de que a magnitude da variância entre os valores estimados de f de diferentes locos forneça uma estimativa do número efetivo de indivíduos reprodutivamente ativos em uma dada população.

The intrapopulation fixation index ( f ) is inversely related to the outcrossing rate (t). Results obtained from data on molecular markers of natural populations have shown that these values are highly variable, even when measured in the same group of individuals. It is thus suggested that factors besides those described in Wright's genetic equilibrium must be operating. Using simulated data sets this study shows that the finite size condition of a population is sufficient to spread the estimated f values along a range at equilibrium, as opposed to keeping them at the theoretical equilibrium point. The variation in outcrossing rates can amplify this range considerably. Correlation between estimated f values obtained from different loci in this condition showed to be negatively related to the outcrossing rates, and positively related to the variance of these rates along generations. The finite size of populations associated to small fluctuations in t mean values over time may explain the usually reported high variation among estimated f values of different loci.

Marcadores isoenzimáticos ou moleculares têm sido empregados em estudos da estrutura genética e do sistema reprodutivo de populações naturais. Parâmetros genéticos populacionais de interesse são estimados, porém, há pouca informação sobre o erro associado a essas estimativas em função dos diferentes níveis de amostragem. Neste trabalho, o método de reamostragem bootstrap foi aplicado sobre locos, indivíduos, populações e indivíduos e populações concomitantemente, em dados de populações naturais. Para os parâmetros índice de fixação total (F ), índice de fixação intrapopulacional ( f ), diversidade entre populações (teta) e taxa aparente de cruzamento (t a) foram obtidos, em função das fontes de reamostragem, os erros associados às estimativas desses parâmetros, a distribuição de empírica dessas estimativas e intervalos de confiança para tais parâmetros. Geralmente, os menores erros estão associados a <img SRC="http:/img/fbpe/sa/v58n4/6299s3.gif">, e verificou-se que apenas as distribuições empíricas de <img SRC="http:/img/fbpe/sa/v58n4/6299s1.gif">e <img SRC="http:/img/fbpe/sa/v58n4/6299s2.gif">tendem à normalidade quando indivíduos estão envolvidos na reamostragem. Foram feitas reamostragens com tamanho variável de amostras bootstrap, visando obter o número necessário de locos, indivíduos e populações para atingir um dado nível de precisão na estimação de F, f e teta. Em geral, os tamanhos amostrais utilizados nas pesquisas com populações naturais foram suficientes apenas para estimar teta, com a precisão estabelecida. A fonte de variação de locos foi responsável pelos maiores erros associados a <img SRC="http:/img/fbpe/sa/v58n4/6299s1.gif">e <img SRC="http:/img/fbpe/sa/v58n4/6299s2.gif">, sendo recomendável aumentar o número de locos em pesquisas dessa natureza. A amostragem de populações também deverá merecer atenção no planejamento de pesquisas futuras.

Genetic structure and reproductive system of natural populations have often been studied using molecular markers and isozymes. Specific genetic parameters are therefore being estimated for this purpose. There is little information about errors associated with these estimates due to different possible sampling levels. Some real datasets were used in this study and bootstrapping was performed over loci, individuals, populations and both individuals and populations simultaneously. These different resampling strategies produced associated errors of the estimates and their empiric distributions and confidence intervals for the following parameters: total fixation index (F ), fixation index within populations (f ), diversity between populations (theta ) and apparent outcrossing rate (t a). Generally, smaller errors were associated with <img SRC="http:/img/fbpe/sa/v58n4/6299s3.gif">. The only empirical distribution of estimates approaching normality were for <img SRC="http:/img/fbpe/sa/v58n4/6299s1.gif">and <img SRC="http:/img/fbpe/sa/v58n4/6299s2.gif">when individuals were envolved in the resampling process. Bootstrap samples of variable sizes were used to obtain the number of loci, individuals and populations necessary to achieve a given precision of F, f and theta. Generally, the magnitude of errors of the estimates in most datasets indicated that sample sizes currently used in research involving natural populations were suitable only for the estimation of theta. The largest errors of <img SRC="http:/img/fbpe/sa/v58n4/6299s1.gif">and <img SRC="http:/img/fbpe/sa/v58n4/6299s2.gif">, were associated to sampling errors due to loci. Therefore, future studies should use larger numbers of loci. Sampling errors due to population was also important and should not be neglected.

Através da análise de 18 locos isoenzimáticos polimórficos, foram estimadas as freqüências alélicas referentes a 214 indivíduos adultos de quatro populações naturais de Cryptocarya moschata de duas regiões do estado de São Paulo. Com base nas heterozigosidades observadas e esperadas, foram obtidas estimativas das estatísticas F de Wright. Para fins de comparação, utilizou-se também o método da análise da variância para estimação dos parâmetros correspondentes F = FIT, <FONT FACE="Symbol">q</font>P= FST e f = FIS. Os dois métodos forneceram resultados bastante concordantes: <img src="http:/img/fbpe/rbb/v22s2/f.gif" alt="f.gif (76 bytes)">IT = 0,142; <img src="http:/img/fbpe/rbb/v22s2/f.gif" alt="f.gif (76 bytes)">ST = 0,140; <img src="http:/img/fbpe/rbb/v22s2/f.gif" alt="f.gif (76 bytes)">IS = 0,002 e <img src="http:/img/fbpe/rbb/v22s2/f.gif" alt="f.gif (76 bytes)">= 0,116; <img src="http:/img/fbpe/rbb/v22s2/a3form05.gif" alt="a3form05.gif (151 bytes)">P = 0,123 e <img src="http:/img/fbpe/rbb/v22s2/a3form06.gif" alt="a3form06.gif (144 bytes)">= 0,008, indicando que os indivíduos dentro das populações devem ser panmíticos e que a diversidade entre populações é bastante alta, sendo similar à que se espera para famílias com estruturação de meios-irmãos. Calculando <img src="http:/img/fbpe/rbb/v22s2/a3form05.gif" alt="a3form05.gif (151 bytes)">P com as populações tomadas duas a duas, notou-se tendência de <img src="http:/img/fbpe/rbb/v22s2/a3form05.gif" alt="a3form05.gif (151 bytes)">P crescer com a distância geográfica o que sugere tendência de isolamento pela distância. O fluxo gênico foi estimado em 0,9 indivíduos por geração, o que corrobora a pronunciada diferenciação populacional encontrada. Devido ao valor negligível encontrado de <img src="http:/img/fbpe/rbb/v22s2/a3form07.gif" alt="a3form07.gif (245 bytes)">IS, o tamanho efetivo de variância de cada população é equivalente ao número de indivíduos amostrados. As estratégias de manejo e conservação necessárias para a preservação da alta variabilidade genética intrapopulacional de C. moschata implicam na manutenção de populações com número grande de indivíduos. Além disso, para a preservação da espécie como um todo, a manutenção de muitas populações provavelmente é necessária.

This paper was aimed at studying the genetic structure of populations of Cryptocarya moschata and proposing conservation strategies for the species. Allozyme data of 18 polymorphic loci were collected from 214 trees sampled from four natural populations located in two regions of the State of São Paulo, Brazil. Estimates of Wright's F statistics were computed on the basis of observed and expected heterozygosities. The analysis of variance, pooled over loci and alleles, was also used to compare corresponding estimates of F (= FIT), <FONT FACE="Symbol">q</font>P (= FST) and f (= FIS). Both methods provided similar results with <img src="http:/img/fbpe/rbb/v22s2/f.gif" alt="f.gif (76 bytes)">IT = 0.142, <img src="http:/img/fbpe/rbb/v22s2/f.gif" alt="f.gif (76 bytes)">ST = 0.140, <img src="http:/img/fbpe/rbb/v22s2/f.gif" alt="f.gif (76 bytes)">IS = 0.002 and <img src="http:/img/fbpe/rbb/v22s2/f.gif" alt="f.gif (76 bytes)">= 0.116, <img src="http:/img/fbpe/rbb/v22s2/a3form05.gif" alt="a3form05.gif (151 bytes)">P = 0.123 and <img src="http:/img/fbpe/rbb/v22s2/a3form06.gif" alt="a3form06.gif (144 bytes)">= 0.008. Results indicated that individuals within populations are probably panmitic. Diversity among populations was very high, similar to what would be expected for groups of plants with a half-sib family structure. Estimates of <img src="http:/img/fbpe/rbb/v22s2/a3form05.gif" alt="a3form05.gif (151 bytes)">P, obtained for populations taken two at a time, indicated that diversity tended to increase with distance and suggested that isolation by distance is a factor to be considered. An estimated gene flow of 0.9 migrants per generation corroborated to the pronounced divergence found among populations. Due to the negligible <img src="http:/img/fbpe/rbb/v22s2/a3form07.gif" alt="a3form07.gif (245 bytes)">IS value found, the variance effective size for each population is equivalent to its sampling number. In this case, management and conservation strategies aimed at preserving high intrapopulation genetic variation, will imply in the maintenance of large populations. In addition, due to the relatively large divergence detected among populations, the preservation of the species as a whole will also require the maintenance of many such populations.
