RESUMEN Dada la importancia ecológica y susceptibilidad de los elasmobranquios a frecuentes capturas incidentales, el seguimiento de sus pesquerías tiene gran relevancia. Considerando la ausencia de información acerca de la diversidad de elasmobranquios asociada con la pesquería de peces de pico en el puerto pesquero de Robledal, en la isla de Margarita, Venezuela, un estudio de seguimiento fue llevado a cabo entre enero de 2014 y enero de 2015. Tres ejemplares de Pseudocarcharias kamoharai fueron capturados a 100 km al suroeste de la isla, a una profundidad aproximada de 150 m. Se determinaron algunos caracteres anatómicos, y morfométricos relevantes para la especie, con los que fueron identificados. Estos registros confirman la presencia de la especie en aguas venezolanas, ampliando su rango de distribución y proporcionando nuevos datos sobre patrones cromáticos y aspectos reproductivos.

ABSTRACT Given the ecological importance and elasmobranch susceptibility to frequent bycatch, the surveillance of their fisheries has high relevance. Considering the lack of information about elasmobranch diversity associated with billfish fishery in the fishing port of Robledal, Margarita Island, Venezuela, a monitoring study was achieved between January 2014 and January 2015. Three specimens of Pseudocarcharias kamoharai were caught about 100 km southwest of the island, at a depth of about 150 m. Some anatomical, meristic and morphometric characters relevant to the species were determined, which allowed taxonomical identification. These records confirm the presence of the species in Venezuelan waters, extending its distribution range; and providing new data on chromatic patterns and reproductive aspects.

El género Pterois contiene nueve especies válidas, nativas del Mar Rojo y el Océano Índico en el Pacífico occidental. P. volitans y P. miles son nativas del Indo-Pacífico, y fueron introducidas en las aguas de Florida como resultado de la liberación de peces confinados en acuario y han sido reconocidas recientemente como invasoras en el Atlántico Occidental y Mar Caribe (Costa Rica hasta Venezuela). Los estudios citogenéticos realizados hasta ahora en el género Pterois cubren solamente aspectos básicos de tres especies que incluyen a P. volitans del océano Indo-Pacífico. Debido a la ausencia de información detallada sobre las características cromosómicas de esta especie invasora, el objetivo del presente estudio fue investigar las características citogenéticas en ejemplares de Venezuela mediante técnicas convencionales y moleculares y comparar los resultados con los reportados para el área de distribución original. Para ello, se investigaron las características cariotípicas mediante tinción con Giemsa, bandeo-C, impregnación con Nitrato de Plata (Ag-NOR) e hibridación fluorescente in situ (FISH) dual para localizar los genes ribosomales 18S rDNA y 5S rDNA en cuatro ejemplares de pez león capturados en la Isla Margarita, Venezuela. La comparación de secuencias del gen 16S de los especímenes analizados con secuencias ya incluidas en el Genbank permitieron corroborar la identificación de P. volitans excluyendo así la posibilidad de una identificación errónea de P. miles. El número diploide fue 2n=48 (2m+10sm+36a) con un FN=60. Los cromosomas presentaron tamaños que disminuyen de manera uniforme dificultando la identificación de homólogos, excepto el único par metacéntrico y el par cromosómico número 2. El bandeo-C reveló tres pares de cromosomas bandas-C negativos, mientras que los restantes presentaron bloques bandas-C positivos en posición telomérica y, en algunos casos, intersticial. Sólo dos cromosomas mostraron bandas-C pericentroméricas. La tinción secuencial reveló las Ag-NOR localizadas en los extremos de los brazos cortos del par número dos y el ensayo FISH demostró que los genes 18S rDNA y 5S rDNA se localizan en ese mismo par. Se discute la co-localización de los genes 5S rDNA y 18S rDNA. La distribución de la heterocromatina constitutiva y localización de las NORs en los peces examinados difirió de la reportada para ejemplares de P. volitans del Océano Índico (Java), sugiriendo que durante la propagación de P. volitans han ocurrido reorganizaciones cromosómicas que involucran la heterocromatina.

The genus Pterois includes nine valid species, native to the Red Sea and Indian Ocean throughout the Western Pacific. P. volitans and P. miles are native to the Indo-Pacific, and were introduced into Florida waters as a result of aquarium releases, and have been recently recognized as invaders of the Western Atlantic and Caribbean Sea (Costa Rica to Venezuela). Thus far, cytogenetic studies of the genus Pterois only cover basic aspects of three species, including P. volitans from Indo-Pacific Ocean. Considering the lack of more detailed information about cytogenetic characteristics of this invasive species, the objective of the present study was to investigate the basic and molecular cytogenetic characteristics of P. volitans in Venezuela, and compare the results with those from the original distribution area. For this, the karyotypic characteristics of four lionfish caught in Margarita Island, Venezuela, were investigated by examining metaphase chromosomes by Giemsa staining, C-banding, Ag-NOR, and two-colour-Fluorescent in situ hybridization (FISH) for mapping of 18S and 5S ribosomal genes. Comparing the sequences of the 16S gene of the specimens analyzed, with sequences already included in the Genbank, we corroborated that our specimens identified as P. volitans are in fact this species, and hence exclude the possibility of a misidentification of P. miles. The diploid number was 2n=48 (2m+10sm+36a) with FN=60. Chromosomes uniformly decreased in size, making it difficult to clearly identify the homologues except for the only metacentric pair, and the pairs number two, the largest of the submetacentric series. C-banding revealed only three pairs of chromosomes negative for C-band, whereas all remaining chromosomes presented telomeric and some interstitial C-positive blocks. Only two chromosomes were C-banding positive at the pericentromeric regions. Sequential staining revealed Ag-NOR on the tips of the short arms of chromosome pair number two and the FISH assay revealed that 18S rDNA and 5S rDNA genes are co-located on this chromosome pair. The co-localization of 5S rDNA and 45S rDNA is discussed. Both constitutive heterochromatin and NOR location detected in samples examined in this study, differ from those reported for P. volitans in previous analysis of specimens collected in Indian Ocean (Java), suggesting the occurrence of chromosome microrearrangements involving heterochromatin during the spread of P. volitans.Rev. The objectives of this study were to estimate future climatic scenarios for the Spring-Summer cycle in the Ciénega de Chapala region in Jalisco, Mexico; estimate the impact of the scenarios on agro-climatic and phenological parameters of the maize crop; and climatic scenarios on water requirements of maize in the studied area. Databases series were available of 1977-2006 series from the stations of the National Water Commission (CONAGUA) and the database of average temperature anomalies and, the precipitation for the period 2007-2099, in the A2 greenhouse gases emission scenario. Using trigonometric models, the future values of maximum and minimum temperatures and thermal oscillation were estimated. An adjustment of the Hargreaves model was performed to the pan evaporation model in order to estimate the evapotranspiration. For the estimation of the phenological variables, the average thermal requirements of some commercial varieties were considered and, Gompertz models were used to estimate the days with temperatures beyond the threshold of the maize crop. In the late XXI century there will be an increase of 6.5 °C of maximum temperatures, 3.6 °C of mean temperatures and, 0.09 °C of minimum temperatures, with respect to the current values. The thermal oscillation will have an increase in its values with a slight slope, indicating that, in the future there will be more extreme thermal conditions than today. The growing season in the wet period will experience a shortening and, the heat units required for the maize crop will be covered faster; day and nighttime temperatures will be increased as well, impacting the processes of photosynthesis and respiration in this species; an increase of the number of days with temperatures above the crop threshold is expected, causing the plants to experience greater periods of time under heat stress; due to the shortening of the crop cycle, the plants' water demand will also decrease.

Los objetivos de este estudio fueron estimar los escenarios climáticos futuros para el ciclo primavera-verano en la región Ciénega de Chapala en Jalisco, México; estimar el impacto de los escenarios sobre los parámetros agroclimáticos y fenológicos del cultivo de maíz; y los escenarios climáticos sobre los requerimientos hídricos del cultivo de maíz en la zona de estudio. Se dispuso de bases de datos climáticos serie 1977-2006 de las estaciones de la Comisión Nacional del Agua (CONAGUA), y la base de datos de anomalías de temperatura media y precipitación para el periodo 2007-2099, en el escenario de emisión de gases efecto invernadero A2. Mediante el uso de modelos trigonométricos se estimaron los valores futuros de temperaturas máximas, mínimas, y oscilación térmica. Se realizó un ajuste del modelo Hargreaves, al modelo del tanque evaporímetro para la estimación de evapotranspiración. Para estimar las variables fenológicas, se consideraron los requerimientos térmicos promedio de algunas variedades comerciales y se utilizaron modelos Gompertz, para estimar días con temperatura fuera de los umbrales del cultivo de maíz. A finales del siglo XXI se tendrá un incremento de 6.5 °C en temperaturas máximas, 3.6 °C en temperaturas medias y de 0.09 °C en temperaturas mínimas, con respecto a los valores actuales. La oscilación térmica presentará un incremento en sus valores con una pendiente ligera, indicando que en el futuro se tendrán condiciones térmicas más extremosas que las actuales. La estación de crecimiento en el periodo húmedo experimentará un acortamiento y las unidades calor requeridas por el cultivo de maíz serán cubiertas más rápidamente, las temperaturas diurnas y nocturnas se verán incrementadas, que impactará los procesos de fotosíntesis y de respiración en dicha especie agrícola; se espera un incremento del número de días con temperaturas mayores al umbral de este cultivo, provocando que las plantas experimenten mayores lapsos de tiempo bajo estrés térmico; debido al acortamiento en la duración del ciclo de cultivo se tendrá una disminución en la demanda hídrica de las plantas.

The objectives of this paper were to quantify the trend of the climatic variables for the Autumn-Winter cycle during 1961-2006 periods in order to establish future climatic and agroclimatic scenarios for the Autumn-Winter cycle and to assess the impact of climate change on wheat development in Ciénega region of Chapala, Jalisco, Mexico. The data from 10 weather stations were used to retrospectively analyze the climatic change and general circulation models in order to estimate future climatic change. The standard normal homogeneity test was applied to detect trend changes in the observed values. Future scenarios were generated for five climatic parameters and five agroclimatic parameters, in order to assess climatic change impacts on wheat. The results showed a change in the trend of maximum and minimum temperatures around the year 1983. In future scenarios, the maximum temperature will increase up to 6.4 °C by the end of the XXI century, causing increases in average temperature and temperature variation. The minimum temperature is maintained between 9 and 10 °C throughout the century. Evapotranspiration will increase, there will be acceleration in the heat units' accumulation, day and night temperatures will increase and the number of chilling hours will decrease. Wheat cultivation will suffer unfavorable conditions for vernalization and a shortening of phenological stages, so that the use of varieties with lower vernalization requirements and greater tolerance to hot conditions, will be one adaptation measurement to continue the wheat cultivation in the studied area.

Los objetivos de este trabajo fueron cuantificar la tendencia de variables climáticas para el ciclo otoño-invierno durante el periodo 1961-2006, establecer escenarios climáticos y agroclimáticos futuros, para el ciclo otoño-invierno y evaluar el impacto del cambio climático sobre el desarrollo del trigo en la región Ciénega de Chapala, Jalisco, México. Se utilizaron datos de 10 estaciones climatológicas para analizar el cambio climático con visión retrospectiva, y modelos de circulación general para estimar el cambio climático a futuro. Se aplicó la prueba de homogeneidad normal estándar, para detectar cambio de tendencia en los valores observados. Se generaron los escenarios futuros para cinco parámetros climáticos y cinco agroclimáticos, para evaluar los impactos del cambio climático sobre el trigo. Los resultados mostraron un cambio en la tendencia de las temperaturas máxima y mínima alrededor del año 1983. En los escenarios futuros, la temperatura máxima se incrementará hasta 6.4 °C a finales del siglo XXI, provocando incrementos en temperatura media y oscilación térmica. La temperatura mínima se mantiene entre 9 y 10 °C durante todo el siglo. Se tendrán incrementos en la evapotranspiración, aceleración en la acumulación de unidades calor, incrementos en las temperaturas diurnas y nocturnas y disminución en el número de horas frío. El cultivo de trigo sufrirá condiciones desfavorables para la vernalización y acortamiento en sus etapas fenológicas, por lo que el uso de variedades con menor requerimiento de vernalización y con mayor tolerancia a condiciones cálidas, será una de las medidas de adaptación a seguir para continuar con el trigo en la zona de estudio.

Sharks are suffering from intensive exploitation by worldwide fisheries leading to a severe decline in several populations in the last decades. The lack of biological data on a species-specific basis, associated with a k-strategist life history make it difficult to correctly manage and conserve these animals. The aim of the present study was to develop a DNA-based procedure to discriminate shark species by means of a rapid, low cost and easily applicable PCR analysis based on 5S rDNA repeat units amplification, in order to contribute conservation management of these animals. The generated agarose electrophoresis band patterns allowed to unequivocally distinguish eight shark species. The data showed for the first time that a simple PCR is able to discriminate elasmobranch species. The described 5S rDNA PCR approach generated species-specific genetic markers that should find broad application in fishery management and trade of sharks and their subproducts.

The karyotype and chromosomal characteristics of the characid fish Triportheus venezuelensis were investigated using differential staining techniques (C-banding, Ag-NOR staining) and fluorescent in situ hybridization (FISH) with an 18S rDNA probe. The diploid chromosome number (2n = 52), karyotype composition and sex chromosome determination system of the ZZ/ZW type were the same as previously described in other species of the genus Triportheus. However, extensive variation regarding nucleolus organizer regions (NOR) different from other species was observed. 18S rDNA sequences were distributed on nine chromosome pairs, but the number of chromosomes with Ag-NORs was usually lower, reaching a maximum of four chromosomes. When sequential staining experiments were performed, it was demonstrated that: 1.) active NORs usually corresponded to segments with 18S rDNA genes identified in FISH experiments; 2.) several 18S rDNA sequences were not silver-stained, suggesting that they do not correspond to active NORs; and 3.) some chromosomes with silver-stained regions did not display any 18S rDNA signals. These findings characterize an extensive polymorphism associated with the NOR-bearing chromosomes of T. venezuelensis and emphasize the importance of combining traditional and molecular techniques in chromosome studies.

This paper describes the karyotype of Odontesthes regia by means of Giemsa staining, C-banding, to reveal the distribution of the constitutive heterochromatin, and by Ag-staining and fluorescent in situ hybridization (FISH), to locate ribosomal genes (rDNA). The chromosome diploid modal count in the species was 2n = 48. The karyotype is composed of one submetacentric pair (pair 1), 16 subtelocentric pairs (pairs 2 to 17), and 7 acrocentric pairs (pairs 18 to 24). With the exception of pair 1 it was not possible to classify the homologous chromosomes accurately because differences in chromosome size were too slight between adjacent pairs. The distribution of C-banded heterochromatin allowed for a more accurate matching of the majority of chromosomes of the subtelocentric series. Silver staining of metaphase spreads allowed for the identification of Nucleolus Organizer Regions (Ag-NOR) on pair 1. FISH experiments showed that 18S rDNA sequences were located, as expected, in the same chromosome pair identified as the Ag-NOR-bearing one

Este trabajo describe el cariotipo de Odonthestes regia, por medio de tinción Giemsa, bandeo C, para revelar la distribución de la heterocromatina constitutiva, y por medio de tinción con nitrato de plata e hibridación fluorescente in situ (FISH), para localizar genes ribosomales (rADN). El recuento modal cromosómico diploide en la especie fue de 2n = 48. El cariotipo está compuesto por un par submetacéntrico (par 1), 16 pares subtelocéntricos (pares 2 al 17) y 7 pares acrocéntricos (pares 18 al 24). Con excepción del par 1, no fue posible clasificar con exactitud a los cromosomas homólogos, ya que las diferencias en el tamaño fueron muy pequeñas entre pares adyacentes. La distribución de la heterocromatina por bandeo C permitió aparear a la mayoría de los cromosomas de la serie subtelocéntrica. La tinción con plata de preparaciones metafásicas permitió la identificación de las regiones organizadoras del nucléolo (Ag-RON) en el par 1. Los experimentos FISH mostraron que las secuencias ADNr 18S estaban localizadas, como era de esperar, en el mismo par cromosómico identificado como los portadores de Ag-RON

Resumen Se estudió el cariotipo de Serrasalmus rhombeus de Caicara del Orinoco, Venezuela. El número diploide de cromosomas 2n=60 estuvo compuesto de 30 metacéntricos, 16 submetacéntricos, 4 subtelocéntricos y 10 acrocéntricos (NF=110). Fueron detectados cuatro pares de cromosomas portadores de NOR. Las NOR están localizadas en la región terminal de los brazos cortos de un par de cromosomas subtelocéntricos de mediano tamaño y en la región terminal de tres pares acrocéntricos pequeños. La diversidad del cariotipo en esta especie sugiere que la formula del cariotipo y el número de NOR podrían ser de utilidad para la caracterización de stocks o poblaciones intraespecíficas.

Summary Cytogenetic studies were conducted on Serrasalmus rhombeus from Caicara del Orinoco, Venezuela. The diploid chromosome number 2n=60 was composed of 30 metacentric, 16 submetacentric, 4 subtelocentric and 10 acrocentric chromosomes (NF= 110). Four pairs of NOR-bearing chromosomes were detected. NOR are located terminally on the short arms of one pair of medium sized subtelocentric chromosomes, and on three small sized acrocentric chromosome pairs. The karyotype diversity in this species suggests that karyotype formulae and NOR number could be useful for characterization of intraspecific stocks or populations.

Resumo Estudou-se o cariotipo de Serrasalmus rhombeus de Caicara del Orinoco, Venezuela. O número diploide de cromossomas 2n=60 esteve composto de 30 metacéntricos, 16 submetacéntricos, 4 subtelocéntricos e 10 acrocéntricos (NF=110). Foram detectados quatro pares de cromossomas portadores de NOR. As NOR estão localizadas na região terminal dos brazos curtos de um par de cromossomas subtelocéntricos de tamanho medio e na região terminal de três pares acrocéntricos pequenos. A diversidade do cariotipo nesta espécie sugere que a fórmula do cariotipo e o número de NOR poderiam ser de utilidade para a caracterização de stocks ou povoações intraespecíficas.
