Effects of autolyzed yeast on leukocyte oxidative metabolism and pneumonia occurrence in feedlot steers

ABSTRACT: Although yeast supplementation has shown an immunostimulant effect in feedlot cattle, the question remains whether it influences the occurrence of the bovine respiratory disease complex (BRD) in feedlot cattle. Therefore, our objective was to evaluate whether dietary supplementation with autolyzed yeast affects the efficiency of blood phagocytes, reducing inflammation and the occurrence of BRD in feedlot cattle. A randomized experimental trial was conducted with whole steers with half Angus blood for 106 days: control (n=10): diet without yeast; Y4 (n=12): yeast diet (4g per animal per day); and Y7 (n=7): yeast diet (7g per animal per day). On Days 0, 14, 42, 70, and 105, blood count, leukocyte oxidative metabolism, serum haptoglobin, frequency of nasal secretion and orbital temperature were evaluated. On slaughter day, a lung evaluation was performed. On Days 42 and 70 of feedlot finishing, the treated animals showed increased leukocyte oxidative metabolism (D42 P=0.04; D70 P=0.02) compared to the control group. These animals had a lower frequency of mucopurulent nasal secretion, lower orbital temperature and reduced pneumonia occurrence than the treated groups (P=0.05) with less lung lesion severity (P=0.01), allowing us to infer that the autolyzed yeast improves blood phagocytic function and decreases the inflammation and BRD of feedlot steers, especially at a dose of 4g.
RESUMO: Embora a suplementação com leveduras tenha demonstrado efeito imunoestimulante em bovinos, permanece-se a dúvida se ela influencia na ocorrência do complexo doença respiratória bovina (CDRB) em bovinos confinados. Portanto, objetivou-se avaliar se a suplementação dietética com levedura autolisada influencia a eficiência de fagócitos sanguíneos, reduzindo inflamações e a ocorrência de CDRB em bovinos confinados. Realizou-se um ensaio experimental randomizado com 36 novilhos inteiros 1/2 sangue Angus por 106 dias: controle (n=10): dieta sem levedura; Y4 (n=12): dieta com levedura (4g por animal por dia); e Y7 (n=7): dieta com levedura (7g por animal por dia). Nos dias 0, 14, 42, 70 e 105, hemograma, metabolismo oxidativo de leucócitos, haptoglobina sérica, frequência de secreção nasal e temperatura orbital foram avaliados. No dia do abate, foi realizada avaliação pulmonar. Nos dias 42, e 70 de terminação em confinamento, os animais tratados apresentaram aumento do metabolismo oxidativo dos leucócitos (D42 P=0,04; D70 P=0,02) em relação ao grupo controle. Esses animais apresentaram menor frequência de secreção nasal mucopurulenta, menor temperatura orbital, menor ocorrência de pneumonias (P=0,05) com menor gravidade das lesões pulmonares (P=0,01), permitindo inferir que a levedura autolisada na dieta melhorou a eficiência dos fagócitos sanguíneos, reduzindo inflamações e a ocorrência de CDRB, especialmente na dosagem de 4g.

Abstract Objective To analyze the effect of combined training (CT) in postural control and gait parameters in postmenopausal women. Methods A parallel-group, randomized, control study was conducted with 16 weeks of combined training (n = 16) versus a non-training control group (n = 12) in postmenopausal women (aged 59.3 ± 8.0). Pre and postintervention assessments included postural control (using an AMTI force platform – Advanced Mechanical Technology, Inc., Watertown, MA, USA) and gait impairments (using baropodometry). In addition, the upper limb strength and abdominal tests, as well as aerobic capacity, assessed functional indicators. Results The CT intervention in postmenopausal women resulted in improved gait (stride length (p = 0.006); speed (p = 0.013); double support time (p = 0.045); and improved postural control (displacement area of postural sway in a normal base of support with eyes open (p = 0.006). Combined training increased functional indicators (abdominal - p = 0.031; aerobic capacity - p = 0.002). Conclusion In conclusion, combined aerobic plus strength training effectively improved gait and balance control in older women. The postmenopausal women from the CT group walked faster and with bigger steps after the intervention than the control group. In addition, they presented decreased postural sway in standing and decreased the percentage of double support time while walking, which means improved static and dynamic balance control and functional indicators.
Resumo Objetivo Analisar o efeito do treinamento combinado (TC) no controle postural e nos parâmetros da marcha em mulheres na pós-menopausa. Métodos Foi realizado um estudo controlado randomizado de grupos paralelos com 16 semanas de treinamento combinado (n = 16) versus um grupo controle sem treinamento (n = 12) em mulheres na pós-menopausa (59,3 ± 8,0 anos). As avaliações pré e pós-intervenção incluíram controle postural (usando a plataforma de força AMTI) e deficiências da marcha (usando baropodometria). Além disso, os testes de força de membros superiors e abdominal, bem como a capacidade aeróbica, avaliaram indicadores funcionais. Resultados A intervenção do TC em mulheres na pós-menopausa resultou em melhora da marcha (comprimento da passada (p = 0,006), velocidade (p = 0,013), tempo de apoio duplo (p = 0,045) e controle postural aprimorado (área de deslocamento da oscilação postural em base de apoio normal com olhos abertos (p = 0,006). O TC aumentou os indicadores funcionais (abdominal - p = 0,031; capacidade aeróbia - p = 0,002). Conclusão Em conclusão, o TC de força e aeróbico melhorou efetivamente o controle da marcha e do equilíbrio em mulheres idosas. As mulheres na pós-menopausa do grupo CT caminharam mais rápido e com passos maiores após a intervenção do que o grupo controle. Além disso, elas apresentaram redução da oscilação postural em pé e do percentual de tempo de apoio duplo durante a caminhada, o que significa melhora no controle do equilíbrio estático e dinâmico e dos indicadores funcionais.

Anabolic steroids and their effects of on neuronal density in cortical areas and hippocampus of mice

Resumo As substancias anabólicas tem sido cada vez mais utilizadas por fisiculturistas e atletas com o objetivo de melhorar o desempenho e a estética. No entanto, essa prática tem causado algumas preocupações aos médicos e pesquisadores, devido ao desconhecimento das consequencias que o uso indiscriminado e ilícito dessas substâncias podem causar. Diante disso, este estudo analisou os efeitos de dois esteroides anabolizantes (EA) comercialmente disponíveis, Winstrol Depot® (Stanozolol) e Deposteron® (cipionato de testosterona), na densidade neuronal das regiões corticais límbica, motora e sensitive bem como das áreas CA1, CA2, CA3 do hipocampo. Foram utilizados 60 camundongos Swiss (30 machos e 30 fêmeas), separados em três grupos: controle e dois grupos experimentais, que receberam o EA. De cada cérebro, foram coletadas amostras homotípicas e semi-seriadas em cortes frontais das áreas estabelecidas para o estudo. Os resultados mostraram que as fêmeas tratadas com cipionato de testosterona apresentaram uma redução em todas as regiões analisadas já as fêmeas tratadas com Stanozolol mostraram uma diminuição em algumas áreas do hipocampo. Em relação aos animais machos, o stanozolol levou a uma diminuição na densidade neuronal em uma região do hipocampo. Estes dados nos permitem concluir que doses supra fisiológicas de esteroides utilizadas neste estudo podem causar danos consideráveis ao tecido nervoso com comprometimento ultraestrutural e consequentemente comportamental. Essas alterações podem interferir na perda de rendimento físico e no desempenho de atletas e não atletas e podem causar danos irreparáveis a indivíduos que fazem uso irresponsável destes EA.
Abstract Anabolic substances have been increasingly used by bodybuilders and athletes with the goal of improving performance and aesthetics. However, this practice has caused some concern to physicians and researchers because of unknowledge of consequences that the indiscriminate and illicit use of these substances can cause. Thus, this study analyzed the effects of two commercially available anabolic steroids (AS), Winstrol Depot® (Stanozolol) and Deposteron® (Testosterone Cypionate), in the neuronal density of limbic, motor and sensory regions on the cerebral cortex and in CA1, CA2, CA3 regions of the hippocampus. A total of 60 Swiss mice were used (30 males and 30 females), separated into three groups: control and two experimental groups, which received the AAS. From each brain, homotypic and semi-serial samples were taken in frontal sections from areas established for the study. The results showed that females treated with testosterone cypionate presented a reduction in all regions tested and the ones treated with Stanozolol showed a decrease in some hippocampal areas. Regarding male animals, stanozolol led to a decrease in neuron number in one hippocampal region. These data allow us to conclude that supra-physiological doses of steroids used in this study, can cause considerable damage to nervous tissue with ultrastructural and consequently behavioral impairment. These changes could interfere with the loss of physical yield and performance of athletes and non-athletes and may cause irreparable damage to individuals making irresponsible use of anabolic steroids.

Resumo O objetivo deste estudo foi analisar o comportamento biológico de microesferas de hidroxiapatita (HA) substituída com magnésio (Mg) durante o reparo de defeito ósseo crítico, tendo em vista que este íon é de grande interesse clínico, pois está intimamente associado à homeostasia e à mineralização óssea. Para tanto, utilizou-se 30 ratos para compor três grupos experimentais: GI - defeito ósseo preenchido com microesferas de HA; GII - defeito ósseo preenchido com microesferas de HA substituída com Mg; GIII (controle) - defeito ósseo vazio; avaliados nos pontos biológicos de 15 e 45 dias. Os resultados histológicos evidenciaram, aos 15 dias, discreto infiltrado inflamatório crônico e neoformação óssea restrita às bordas, em todos os grupos. Nos grupos GI e GII, os biomateriais mantiveram-se íntegros e circundados por tecido conjuntivo frouxo. Aos 45 dias, notou-se resposta inflamatória do tipo crônica granulomatosa discreta nos grupos GI e GII, e no GIII presença escassa de células inflamatórias mononucleares. As microesferas implantadas no GI e GII mantiveram-se, em sua maioria, íntegras e envolvidas por tecido conjuntivo fibroso. Notou-se discreta neoformação óssea próxima às bordas e circunjacente a algumas microesferas. No GIII, a mineralização limitou-se às bordas e a área remanescente foi preenchida por tecido conjuntivo fibroso. Conclui-se que os biomateriais foram biocompatíveis, bioativos, osteocondutores e apresentaram biodegradação lenta, indicando seu grande potencial para em aplicações clínica como biomaterial de preenchimento.
Abstract The aim of this study was to analyze the biological behavior and osteogenic potential of magnesium (Mg) substituted hydroxyapatite (HA) microspheres, implanted in a critical bone defect, considering that this ion is of great clinical interest, since it is closely associated with homeostasis and bone mineralization. For the purpose of this study, 30 rats were used to compose three experimental groups: GI - bone defect filled with HA microspheres; GII - bone defect filled with HA microspheres replaced with Mg; GIII - empty bone defect; evaluated at biological points of 15 and 45 days. The histological results, at 15 days, showed, in all the groups, a discrete chronic inflammatory infiltrate; biomaterials intact and surrounded by connective tissue; and bone neoformation restricted to the borders. At 45 days, in the GI and GII groups, an inflammatory response of discrete granulomatous chronic type was observed, and in the GIII there was a scarce presence of mononuclear inflammatory cells; in GI and GII, the microspheres were seen to be either intact or fragmented, surrounded by fibrous connective tissue rich in blood vessels; and discrete bone neoformation near the edges and surrounding some microspheres. In GIII, the mineralization was limited to the borders and the remaining area was filled by fibrous connective tissue. It was concluded that the biomaterials were biocompatible and osteoconductive, and the percentage of Mg used as replacement ion in the HA did not favor a greater bone neoformation in relation to the HA without the metal.

RESUMO: Salmonella Infantis é frequentemente associada a infecções humanas no mundo todo sendo transmitida pelo consumo de alimentos contaminados, principalmente aqueles de origem animal, com destaque para a carne de frango. Objetivou-se avaliar características de virulência, resistência antimicrobiana e a similaridade genética de 51 estirpes de S. Infantis isoladas em amostras de origem avícola. As estirpes foram isoladas no período de 2009 a 2010 em uma empresa com ciclo completo de produção de frango de corte, localizada no estado de São Paulo, Brasil. Foi realizado o teste de susceptibilidade antimicrobiana e pela técnica de PCR, foi avaliada a presença dos genes lpfA (fímbria-adesão), agfA (fímbria-biofilme) e sefA (fímbria-adesão) e os genes de resistência aos beta-lactâmicos (bla TEM, blaSHV, blaCTX-M e blaAmpC ). A relação filogenética foi determinada pelo método de RAPD-PCR. Dentre as drogas testadas, os maiores percentuais de resistência foram para amoxacilina com 35,3% e sulfonamida com 15,7%. Onze perfis de resistência aos antimicrobianos foram identificados (A1 a A11), sendo que nenhum deles apresentou perfil de multirresistência (>3 classes de antimicrobianos). Houve 100% de positividade para o gene agfA, 92,2% para o gene lpfA e nenhuma estirpe apresentou o gene sefA. A maioria dos isolados apresentaram semelhanças no potencial de virulência, pois foram positivos simultaneamente para dois genes estudados, agfA e lpfA (92,2% - 47/51). Das 18 (35,3%) estirpes resistentes aos antimicrobianos da classe dos β-lactâmicos, 10 (55,5%) foram positivas para o gene blaAmpC , cinco (27,8%) para blaCTX-M , duas (11,1%) para blaSHV e nenhuma estirpe apresentou o gene bla TEM . A avaliação filogenética demonstrou a presença de cinco clusters (A, B, C, D e E) com similaridade superior a 80%, e três estirpes distintas que não foram agrupadas em nenhum dos clusters. O cluster B agrupou 33 estirpes, todas positivas para os genes lpfA e agfA, provenientes tanto do aviário quanto do matadouro frigorífico, persistentes durante todo o período do estudo. Este cluster ainda agrupou 18 estirpes clones com similaridade genética superior a 99%, todas isoladas no matadouro frigorífico. A presença dos genes de virulência, associada à persistência das estirpes clones durante um longo período do estudo, alertam para a possibilidade de S. Infantis em formar biofilme, devendo ser constantemente monitorada na cadeia de produção avícola, especialmente no ambiente de abate, de forma a conhecer o perfil das estirpes que podem contaminar o produto final e assim avaliar os perigos que representam para a saúde pública.
ABSTRACT: Salmonella Infantis is frequently associated with human infections worldwide and is transmitted by consumption of contaminated foods, particularly those of animal origin, especially the chicken meat. We aimed to evaluate virulence characteristics, antimicrobial resistance and the genetic similarity of 51 strains of S. Infantis isolated from samples of poultry origin. The strains were isolated from 2009 to 2010 in a company with full cycle of broiler’s production in the state of São Paulo, Brazil. The antimicrobial susceptibility test was performed and, by PCR, we evaluated the presence of the genes lpfA (hem-adhesion), agfA (hem-biofilm) and sefA (hem-adhesion) and resistance genes to beta-lactams (blaTEM, blaSHV, bla CTX-M and blaAmpC ). The phylogenetic relationship was determined by RAPD-PCR method. Among the drugs tested, the highest percentages of resistance were to amoxicillin (35.3%) and to sulfonamide (15.7%). Eleven antimicrobial resistance patterns were identified (A1 to A11), none of them presented a multiresistance profile (> 3 antimicrobials classes). There was 100% of positivity for the agfA gene, 92.2% for the lpfA gene, and no strain presented the sefA gene. Most of the isolates showed similarities in virulence potential, since they were simultaneously positive for two studied genes, agfA and lpfA (92.2%, 47/51). Of the 18 (35.3%) strains resistant to antimicrobials of the β-lactam class, 10 (55.5%) were positive to blaAmpC gene, five (27.8%) for blaCTX-M , two (11.1%) to blaSHV and no strain presented the blaTEM gene. The phylogenetic evaluation has shown the presence of five clusters (A, B, C, D and E) with similarity greater than 80%, and three distinct strains which were not grouped in any cluster. Cluster B grouped 33 strains, all positive for lpfA and agfA genes, from both, the broiler farming facility and the slaughterhouse, persistent throughout all the study period. This cluster also grouped 18 strains clones with genetic similarity greater than 99%, all isolated in the slaughterhouse. The presence of virulence genes associated with persistent strains clones for a long period, warns to the possibility of S. Infantis to form biofilm, and should be constantly monitored in broilers’ production chain, in order to know the profile of the strains that may contaminate the final product and evaluate the hazards that represents to public health.

THERMAL INSULATION WITH RECYCLED MATERIAL IN CREEPS FOR PIGLETS

ABSTRACT In the swine intensive farming, one of the biggest problems is to meet the thermal requirements of piglets in heated creeps, which demand high consumption of electricity and increase in production costs. The aim of this study was to evaluate the thermal efficiency and the consumption of electric energy in heated and internal insulated creeps, made of recycled material, when compared to creeps without thermal insulation. The treatments were: control (TCON): creep heated through suspended electric resistance, controlled by thermostat; thermal insulation with recycled material (TPK). The experiment was carried out for 21 days. In the treatments, the temperature (°C) and relative humidity (RH) of the air, consumption of electric energy (kWh), specific consumption (kWh kg-1), specific cost (R$ kg-1), weight gain (kg) and daily weight gain (kg day-1) were measured. The indoor insulation creep kept the upper air temperature (33.05°C) to the control treatment without thermal insulation (32.6°C) and was more efficient in the use of electric energy (2.46 kWh) in comparison to the non-insulated creep (3.48 kWh). The heat-insulated creep shelter consumed the equivalent of 1.02 kWh more than the insulation treatment to maintain the thermal comfort condition of piglets between 30 and 34 ° C. The conclusion is that the use of recycled material as thermal insulation in heated creeps reduces heat losses and economy electricity.

Leaves from the Tree Poincianella pluviosa as a Renewable Source of Antiplasmodial Compounds against Chloroquine-Resistant Plasmodium falciparum

Poincianella pluviosa is a tree whose stem bark is reputed to have antidiarrheal, wound healing, antitumoral and antimalarial properties. Considering that the leaves of the tree are a more abundant and renewable source than the stem bark, we chose to study the antimalarial potential of this organ part. The aqueous leaf extract exhibited a rich phenolic profile. It was active (IC50= 6.88 ± 1.64 µg mL-1) against chloroquine resistant Plasmodium falciparum (W2) without toxicity to HepG2 A16 cells. The organic fraction was more active (IC50= 1.83 µg mL-1) than the aqueous leaf extract itself. Ten phenolic compounds were obtained from the latter. This is the first report on the phenolic composition of P. pluviosa leaves. Ellagic acid (IC50= 0.215 ± 0.007 µg mL-1) proved to be 32 fold more active than the aqueous leaf extract itself. Considering their renewable aspect, leaves of P. pluviosa appear to be a more interesting source of antimalarial compounds than stem bark.

Effect of entomopathogens on Africanized Apis mellifera L. (Hymenoptera: Apidae)

ABSTRACT This study aimed to evaluate the effect of commercially used entomopathogens on Africanized Apis mellifera L. (Hymenoptera: Apidae). Four bioassays were performed: 1) pulverized entomopathogens on A. mellifera; 2) entomopathogens sprayed on a smooth surface; 3) entomopathogens sprayed on soy leaves; and 4) entomopathogens mixed with candy paste (sugar syrup). Five treatments were prepared: sterile distilled water (control), distilled water sterilized with Tween® 80 (0.01%), and the commercial entomopathogens Metarhizium anisopliae E9 (1.0 × 109 conidia mL−1), Beauveria bassiana PL63 (1.0 × 108 conidia mL−1) and Bacillus thuringiensis var. kurstaki HD-1 (3.0 × 108 spores mL−1). Each treatment consisted of five repetitions, with 20 workers per repetition, which were stored in a plastic box and, later, in a biological oxygen demand (B.O.D.) incubator (27 ± 2 ºC, RH of 60% ± 10%, 12-h photophase). The mortality of the workers was evaluated from 1 h to 240 h, and the data were analyzed using Bayesian inference. The workers killed by the ingestion of candy paste contaminated with the pathogens (products) were randomly separated and selected for the removal of the midgut. Each midgut was fixed in Bouin's solution and prepared for histology. B. bassiana was verified to reduce the survival of A. mellifera workers in all bioassays. Moreover, M. anisopliae reduced the survival of A. mellifera workers directly sprayed, on a smooth surface and mixed with candy. B. thuringiensis reduced A. mellifera survival on a smooth surface and mixed with candy paste. However, its effects were lower than that observed by B. bassiana. The treatments with the biological products did not induce morphometric alterations in the midgut of A. mellifera.

Resumo Foram analisadas raízes e folhas de Panicum maximum Tanzânia, Mombaça e Massai; Urochloa brizantha Piatã, Marandu e Xaraés; Urochloa humidicola Llanero; Urochloa ruziziensis Ruzizienses; Urochloa hybrida Mulato II e Cynodon nlemfuensis Estrela-roxa, procurando identificar caracteres relacionados à melhor adaptação ao ambiente e à qualidade nutritiva das forrageiras. As dez cultivares foram semeadas em blocos inteiramente casualizados com três repetições. Após estabelecidas foi coletado material vegetativo, do qual lâminas histológicas foram confeccionadas a partir de secções do terço médio de raízes e folhas. Foram observadas diferenças significativas (p>0,05) nas raízes: maior volume de células epidérmicas (28,62 µm) e diâmetro total (1926,41 µm) de Llanero; em Estrela-roxa maior espessura do cilindro vascular (975,09 µm) e número maior de protoxilemas (42,25) e formação de aerênquimas nas cultivares Piatã, Mulato II, Xaraés, Massai, Llanero, e Estrela-roxa; Nas folhas, foram constatadas maiores proporções de células buliformes (121,07 µm) e mesofilo foliar mais espesso (263,63 µm) em Llanero; em Xaraés e Marandu maiores proporções de fibras esclerenquimáticas; em Massai menores resultados para quantidade de fibras. Conclui-se que as cultivares Estrela-roxa, Llanero e Massai apresentam maior adaptabilidade ao ambiente e melhor qualidade nutritiva.
Abstract Roots and leaves of Panicum maximum Tanzânia, Mombaça and Massai; Urochloa brizantha Piatã, Marandu and Xaraés; Urochloa humidicola Llanero; Urochloa ruziziensis Ruzizienses; Urochloa hybrida Mulato II and Cynodon nlemfuensis Estrela-roxa were analyzed, seeking to identify characters for better adaptation to the environment that may interfere with digestibility of tissue from the point of view of the rumen in cattle. Were planted ten cultivars in a completely randomized blocks with three repetitions. Was collected vegetative material, which histological slides were prepared from middle third of the sections of roots and leaves. Were observed differences (p>0.05) in the roots: higher volume of epidermal cells (28.62 µm) and overall diameter (1926.41 µm) of Llanero; thicker vascular cylinder (975.09 µm) and more protoxylem (42.25) in Estrela-roxa and occurrence of aerenchyma in cultivars Piatã, Mulato II, Xaraés, Massai, Llanero and Estrela-roxa; Were found higher proportions of bulliform cells in the leaves (121.07 µm) and thicker leaf mesophyll in U. humidicola Llanero (263.63 µm); higher proportion of sclerenchyma fibers in Xaraés and Marandu; lower results for amount of fibers in P. maximum Massai. We conclude that the cultivars Estrela-roxa, Llanero and Massai have greater adaptability to the environment and better nutritional quality.

Anatomy and micromorphometric analysis of leaf Catasetum x apolloi Benelli & Grade with addition of potassium silicate under different light sources

Resumo O objetivo desse trabalho foi comparar vitroplantas de Catasetum x apolloi cultivadas sob luz natural e luz artificial e diferentes concentrações de silicato de potássio, fornecendo dados sobre diferenciação anatômica que auxiliem no processo de aclimatação dessa espécie. Utilizou-se plantas provenientes da semeadura in vitro, 5 protocormos de aproximadamente 0,5 cm foram inoculados em frascos com capacidade para 500 mL contendo 100 mL de meio de cultura alternativo, acrescido de silicato de potássio (0,0; 0,5; 1,0 mL L–1), pH ajustado para 5,5 ±0,5 e gelificado com 4gL–1 de ágar antes do processo de autoclavagem. As culturas foram mantidas sob luz natural (TAA) e luz artificial (TAN) por 90 dias, e feitas análises micromorfométricas (diâmetro polar e equatorial, densidade e índice estomático, espessura do limbo na nervura central e nervuras secundárias). As aplicações de K2SiO4 em meio alternativo, propiciaram: alongamento da hipoderme; mesofilo mais espesso e nervura central mais proeminente; células guardas elípticas; formação de câmaras supraestomáticas; menor densidade estomática e estômatos com menores diâmetros equatorial e polar. As condições que podem favorecer a aclimatação são menores intensidades de luz e menores doses de silicato de potássio.
Abstract The aim of this study was to compare vitroplants Catasetum x apolloi grown under natural light and artificial light and different concentrations of potassium silicate, providing data on the anatomical differentiation that aids the acclimatization process of this species. Plants from in vitro seeding were used; 5 protocorms of approximately 0.5 cm were inoculated into vials with a capacity of 500 mL containing 100 mL of alternative culture medium plus potassium silicate (0.0, 0.5; 1.0 mL L–1), pH adjusted to 5.5 ± 0.5 and gelated with 4GL–1 agar before the autoclaving process. Cultures were maintained under natural light (TNE) and artificial light (TAE) for 90 days, and micromorphometric analysis was performed for polar and equatorial diameter, density and stomatal index, blade thickness in the central rib, and secondary veins. Applications in K2SiO4 alternative medium provided the following: elongation of the hypodermis, thicker mesophyll, and more prominent midrib; elipptical guard cells; formation of epistomatal chamber; and lower stomatal density and stomatal with lower equatorial and polar diameters. The conditions that favored the acclimatization were lower light intensities and lower potassium silicate doses.

Resumo O objetivo do presente estudo foi avaliar propriedades físico-químicas da pasta à base de hidróxido de cálcio (Calen(r)) combinado com diferentes proporções de óxido de zinco (OZ) (1:0,5 e 1:0,65, 1:0,8 e 1:1) (Calen(r) /OZ). Os materiais foram comparados quanto ao tempo de endurecimento, variação do pH, radiopacidade, solubilidade, alterações dimensionais, escoamento e liberação de elementos químicos. Os dados obtidos foram submetidos à análise de variância (nível de 5% de significância). Maior grau de endurecimento, valores de alterações dimensionais, e solubilidade foram encontrados para Calen(r) /OZ 1:0,65 e 1:0,5. Calen(r) /OZ 1:0,5 e 1:0,65, apresentaram os maiores valores de pH em todos os momentos. Todos os materiais apresentaram valores elevados de radiopacidade com diferenças significativas (p>.05). Foram encontradas diferenças significativas apenas entre Calen(r) /OZ de 1:0,5 e 1:1 na liberação de cálcio e zinco (p< 0,05), enquanto que a quantidade de zircônia foi semelhante entre todos os grupos analisados (p>.05). Em relação às propriedades avaliadas no presente estudo, as combinações de Calen(r) com óxido de zinco nas proporções 1:0,5 e 1:0,65 são mais adequadas como material obturador para os canais radiculares de dentes decíduos.
Abstract This study evaluated physiochemical proprieties of a calcium hydroxide-based paste (Calen(r)) combined with a zinc oxide cement at different ratios (1:0.5, 1:0.65, 1:0.8 and 1:1). Materials were compared regarding setting time, pH variation, radiopacity, solubility, dimensional changes, flow and release of chemical elements. Data were analyzed statistically by ANOVA and Tukey's test (α=0.05). Longer setting time and higher dimensional changes and solubility values were exhibited by 1:0.65 and 1:0.5 ratios (p<0.05). The 1:0.5 and 1:0.65 ratios exhibited the highest pH values at all time points. All materials exhibited high radiopacity values. Significant differences were found only between 1:0.5 and 1:1 ratios for calcium and zinc release (p<0.05), whereas the amount of zirconium was similar among all groups (p>0.05). Considering the evaluated proprieties, combinations of Calen(r) paste with ZO at 1:0.5 and 1:0.65 ratios had the best results as root canal filling materials for use in primary teeth.

Este estudo avaliou a ação inseticida sistêmica e o efeito residual de nanoformulações à base de derivados de nim (Azadirachta indicaA. Juss, Meliaceae) sobre ninfas de Bemisia tabaci biótipo B (Hemiptera: Aleyrodidae), espécie-praga de diversas culturas de importância econômica. Suspensões coloidais contendo nanocápsulas carregadas com produtos derivados de A. indica foram preparadas utilizando polímeros biodegradáveis como poli-ε-caprolactona (PCL) e poli-β-hidroxibutirato (PHB). A matriz encapsulada era composta por uma formulação comercial contendo uma quantidade enriquecida e conhecida dos limonoides azadiractina e 3-tigloilazadiractol (Azamax® 1,2 CE). Inicialmente foi estimada a CL50, para ninfas de B. tabaci, de soluções contendo óleo de nim, que foram aplicadas via água de irrigação (solo) em tomateiro. Posteriormente, dois lotes de nanoformulações, com variação no tipo e quantidade dos polímeros e na quantidade do óleo comercial, foram testados quanto à ação sistêmica. As nanoformulações NC L5-2 (com nanocápsulas de PCL) e NC L6-1 (com nanocápsulas de PHB), as mais eficazes entre as testadas, foram avaliadas quanto ao efeito residual. A CL50 estimada foi de 180,67 mg L–1 de azadiractina. Constatou-se também que a ação sistêmica de óleos e nanoformulações contendo compostos de nim dependem das condições ambientais em que são aplicados. O Azamax® não apresenta efeito deterrente sobre a oviposição, nem age sobre o desenvolvimento embrionário da mosca-branca quando aplicado via água de irrigação no solo no momento da infestação. As nanoformulações selecionadas são bioativas mesmo cerca de 30 dias após a aplicação, não diferindo do produto comercial.
This study evaluated the systemic insecticidal action and the residual effect of neem-based nanoformulations (Azadirachta indica A. Juss, Meliaceae) on Bemisia tabaci biotype B (Hemiptera: Aleyrodidae) nymphs, a pest of several economic important crops. Colloidal suspensions containing nanocapsules loaded with derived products from A. indica were prepared by using biodegradable polymers such as poly-ε-caprolactone (PCL) and poly-β-hydroxybutyrate (PHB).The encapsulated matrix was a commercial formulation, which contained not only enriched amount of limonoids azadirachtin and 3-tigloilazadiractol, but also in known quantities (Azamax® 1.2 CE). Initially, it was estimated the LC50 of solutions containing neem oil, which were applied through irrigation water (soil) in tomato in order to control nymphs of B. tabaci. Afterwards, two lots of nanoformulations, varying in type and amount of polymers and in quantity of commercial oil, were evaluated for systemic action. The nanoformulations NC L5-2 (nanocapules in PCL) and NC L6-1 (nanocapsules in PHB) caused higher mortality among others and for this reason were assayed for residual effect. The estimated value for LC50 was 180.67 mg L–1 of azadirachtin. The results showed that systemic action of oils and nanoformulations containing neem compounds depends on environmental conditions. Azamax® did not show either deterrent effect of oviposition or effect on whitefly embryogenesis when applied in soil irrigation at the time of infestation. The nanoformulations selected are bioactives even about 30 days after application and do not differ of commercial product.

RESUMO Objetivou-se caracterizar fenotípica e genotipicamente isolados clínicos de enterobactérias resistentes aos carbapenêmicos (CRE) provenientes do Hospital Universitário de Santa Maria (RS). Entre os isolados clínicos submetidos ao teste modificado de Hodge (MHT), 62,5% apresentaram positividade, indicando possível produção de carbapenemase. A reação em cadeia da polimerase (PCR) demonstrou que o blaKPC foi o gene mais encontrado (31%), seguido de blaIMP (12,5%). O uso conjunto de distintas metodologias faz-se necessário para identificar a resistência aos carbapenêmicos produzida pelas enterobactérias, de modo a auxiliar o controle de infecção prevenindo a disseminação desses microrganismos.
ABSTRACT The present study aimed to genotypically and phenotypically characterize clinical isolates of carbapenem-resistant Enterobacteriaceae collected from inpatients at the University Hospital of Santa Maria, during seven months. Among the clinical isolates subjected to the modified Hodge test (MHT), 62.5% were positive, indicating possible production of carbapenemase. Polymerase chain reaction (PCR) demonstrated that blaKPC was the most frequently found gene (31%), followed by blaIMP (12.5%). Combined use of the methods is needed to identify carbapenem resistance in enterobacteria to prevent their spread and control the infections caused by these organisms.

Occurrence and characterization of Campylobacter spp.isolates in dogs, cats and children

Com o objetivo de melhorar o entendimento das infecções por Campylobacterspp. em cães, gatos e crianças no Brasil, foram avaliadas 160 amostras fecais de crianças e 120 swabs retais de pets (103 cães e 17 gatos). Do total das amostras das crianças, 6,87% foram positivas para Campylobacter spp. e em cães e gatos a positividade foi de 18,3%. Das 33 amostras positivas para Campylobacter spp., 57,6% foram identificadas como C. jejunie 33,4% foram identificadas como C. coli. Mais de 50% das amostras isoladas de pets foram resistentes a ceftiofur, sulphazotrim, norfloxacina e tetraciclina. Em crianças, a maioria das amostras foi resistente a amoxilina, cefazolina, ceftiofur, eritromicina e norfloxacina. De 19 isolados de C. jejuni, 11 isolados de crianças e cinco (5) de cães tinham dois (2) dos quatro (4) genes de virulência flaA, pldA, cadFor ciaB. Associação positiva entre a presença de Campylobacter spp. e diarreia em cães e gatos foi observada em animais desverminados e com hemograma sugestivo de infecção bacteriana. Também houve associação positiva entre a presença dos genes de virulência e a ocorrência de diarreia, e entre o uso de antibióticos e a positividade para Campylobacterspp. em suabes fecais de pets. Os dados desse trabalho indicam que cepas virulentas de Campylobacter spp. são fatores de risco para diarreia em cães e a resistência antimicrobiana é comum em isolados de cães
To improve the understanding of implications of Campylobacterspp. infections in pets and children of different environments were analysed 160 faecal samples from children and 120 from pets (103 dogs and 17 cats). Campylobacter spp. were detected in 6.87% of the children and in 18.3% of the dogs and cats. From 33 stool samples positive for Campylobacter spp., 57.6% were identified as C. jejuni, and 33.4% were identified as C. coli. More than 50% of the isolates in pets were resistant to ceftiofur, sulphazotrim, norfloxacin and tetracycline. In humans, most of the isolates were resistant to amoxicillin, cefazolin, ceftiofur, erythromycin and norfloxacin. From 19 isolates of C. jejuni, 11 isolates from children and 5 from dogs contained two to four of the virulence genes flaA, pldA, cadF or ciaB.We found an association between the presence of virulence genes and diarrhoea. Furthermore, an association was observed between the presence of Campylobacter spp. and diarrhoea in dewormed pets with blood picture suggestive of bacterial infection, and the therapeutic use of antibiotics was associated with more positive detection of Campylobacterspp. in the faeces of pets. Our data indicate that virulent strains of Campylobacter spp. can be risk factor to diarrhoea in animals, and that high resistance to antimicrobial agents is common in pets.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.
