ABSTRACT Some native species of the Cerrado have great ecological and economic potential. An alternative that aims to contribute to the good performance of post-planting plants is the incorporation of biochar into the soil. The objective of this work was to evaluate the effect of filter cake biochar as a soil conditioner and its influence on the growth and physiology of macaúba, araçá and cajuzinho do cerrado. The experiments were set up at Fazenda Hamilton de Abreu Navarro, Federal University of Minas Gerais, where the effect of four doses of filter cake biochar (1%, 2%, 4% and 8% v/v) and two treatments were evaluated. control: one with soil correction (7.5 g of GEOX) and mineral fertilization (380 g NPK 4:14:8 per plant) and the other without fertilization. To monitor the performance of the seedlings, evaluations were carried out at 30, 60 and 120 days after transplanting, the physiological parameters (photosynthesis, transpiration, stomatal conductance, CO2 consumed, internal carbon and electron transport rate), and morphological parameters (height and diameter), at 30, 60, 90 and 120 days after transplanting. At 120 days, the production of shoot, root and total dry biomass was evaluated. For macaúba, the application of biochar, at a dose of 2%, favored its photosynthesis. As for araçá, at 30 days after transplanting, the treatment without fertilization was better for most variables, however the treatments with biochar also had satisfactory results. The cajuzinho from the cerrado also responded positively to the application of biochar at all doses. The physiological and morphological variables responded well to treatments without fertilization and with application of biochar to the soil. Therefore, filter cake biochar can be indicated in the planting of the studied species, as well as the non-application of corrective and mineral fertilizer.

RESUMO O objetivo deste trabalho foi avaliar o efeito do biochar de torta de filtro como condicionador de solo e sua influência no crescimento e na fisiologia de macaúba, araçá e cajuzinho do cerrado. Os experimentos foram montados na Fazenda Hamilton de Abreu Navarro da UFMG, onde foram avaliados o efeito de quatro doses de biochar de torta de filtro (1%, 2%, 4% e 8% v/v) e dois tratamentos controle: um com correção do solo e adubação mineral, e o outro sem adubação. Para acompanhar o desempenho das mudas, foram realizadas avaliações aos 30, 60 e 120 dias após o transplantio, dos parâmetros fisiológicos (fotossíntese, transpiração, condutância estomática, CO2 consumido, carbono interno e taxa de transporte de elétrons), e morfológicos (altura e diâmetro), aos 30, 60, 90 e 120 dias após o transplantio. Aos 120 dias, avaliou-se a produção de biomassa seca da parte aérea, de raízes e total. Para a macaúba, a aplicação de biochar, na dose de 2%, favoreceu a sua fotossíntese. Já para o araçá, aos 30 dias após o transplantio, o tratamento sem adubação foi melhor para a maioria das variáveis, entretanto os tratamentos com biochar também tiveram resultados satisfatórios. O cajuzinho do cerrado também respondeu positivamente à aplicação de biochar em todas as doses. As variáveis fisiológicas e morfológicas responderam bem aos tratamentos sem adubação e com aplicação de biochar ao solo. Portanto, o biochar de torta de filtro pode ser indicado no plantio das espécies estudadas, assim como a não aplicação de corretivo e fertilizante mineral.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

Abstract The present study was based on the analysis of 10,467 trawls of the industrial piramutaba (Brachyplatystoma vaillantii) fishing fleet of the Brazilian state of Pará, which were mapped by onboard GPS loggers (between February 2008 and September 2011) and the PREPS data from 40 vessels which were tracked by this system between 2008 and 2011. The variation in the mean monthly CPUE, based on Lomb’s periodogram, revealed a well-defined and constant cycle with a duration of approximately one year. Three environmental factors influenced this cycle. The El Niño 3.4 index had a negative correlation with the CPUE of the piramutaba fishery, with a time lag of 15 months, while monthly rainfall and the mean discharge of the Amazon River correlated strongly (r=0.89 and 0.87, respectively; p<0.001) with the CPUE, with time lags of 12 and 11 months, respectively. The spatiotemporal analysis of the distribution of the activity of the piramutaba fishing fleet indicated that the most intense area of operation of the fleet lies between latitudes 00º N and 02° N, and longitudes 047º40’ W and 049º40’ W. This area was divided into four geographic quadrants, although fishery operations were concentrated in only three of these quadrants. The study proposed a quadrimester fishing cycle with zoning in three of the quadrants, where fishing would be permitted for four months (occupation period), followed by an 8-month rest period for the recuperation of stocks, aiming at the sustainability of this fishing exploration.

Resumo Parasitos do gênero Oesophagostomum causam doenças entéricas e podem afetar a criação de animais, como os suínos. O objetivo deste estudo foi identificar as espécies e explorar a diversidade genética de Oesophagostomum spp. infectando suínos em contato próximo com humanos, no estado do Piauí, Brasil. Oitenta e sete amostras fecais foram coletadas para testes parasitológicos, análise morfométrica dos ovos e análises moleculares. A taxa geral de positividade para ovos estrongiliformes foi de 81,6%. Quarenta e duas amostras de ovos estrongiliformes foram submetidas à PCR e seis sequências cox1 (637 bp) foram identificadas para o gênero Oesophagostomum. As sequências foram identificadas como Oesophagostomum dentatum, O. quadrispinulatum e O. columbianum. Na árvore filogenética e na rede haplotípica, 89 sequências foram separadas em sete clusters, incluindo sequências de referência do GenBank. Oesophagostomum dentatum e O. quadrispinulatum são espécies estreitamente relacionadas e formaram um grupo monofilético com O. aculeatum. Oesophagostomum columbianum apresentou semelhança com sequências de parasitas obtidos de pequenos ruminantes e o clado foi posicionado mais próximo de O. bifurcum. Alta diversidade interespecífica foi encontrada e a diversidade intraespecífica variou de acordo com as espécies. Esse foi o primeiro estudo a caracterizar sequências de DNA de Oesophagostomum isoladas de suínos no Brasil.

Abstract Helminths of the genus Oesophagostomum cause enteric diseases and affect domestic animals such as pigs. The aim of this study was to explore the species composition and genetic diversity of Oesophagostomum spp. infecting pigs in close contact with humans in the state of Piauí, Brazil. Eighty-seven fecal samples were collected for parasitological tests and molecular analysis. Through microscopy, the overall positivity rate for strongyliform eggs was 81.6% among the pigs studied. Forty-two strongyliform egg samples were subjected to PCR and six cox1 sequences (637 bp) were identified for the genus Oesophagostomum. The sequences were identified as Oesophagostomum dentatum, O. quadrispinulatum and O. columbianum. In the phylogenetic tree and haplotype network, 89 sequences were separated into seven clusters, which also included reference sequences from GenBank. Oesophagostomum dentatum and O. quadrispinulatum were seen to be closely related species and formed a monophyletic group related to O. aculeatum. Oesophagostomum columbianum showed similarity with sequences from parasites infecting small ruminants and the clade was positioned closer to O. bifurcum. High interspecific diversity was found and intraspecific diversity varied according to the species. This was the first study to characterize Oesophagostomum DNA sequences obtained from pigs in Brazil.

Considering the increasing number of low cost vegetable oil product adulterations, it is necessary to assess the authenticity of the lipid sources used in the dog food manufacture, consequently different brands and classifications of feed were analyzed to verify the product authenticity by gas chromatography (GC) with flame ionization detector (FID) and by electrospray ionization mass spectrometry (ESI-MS). Fatty acid (FA) composition analysis was performed to compare the FAs amount in samples with the respective guarantee levels stated by the manufacturers on its product label. Gas chromatography with flame ionization detector (GC-FID) results for omega-3 and 6 percentages are within the limits presented by each manufacturer, while the timnodonic + cervonic acids amount information presented on feed sample labels is not in accordance with GC-FID results. Consequently, the samples were analyzed by ESI-MS, the results exposed the chicken oil presence in the feed and eicosapentaenoic (EPA) + docosahexaenoic (DHA) acids absence, since no lipid profiles similar to fish oils or triacylglycerols (TAGs) containing these two FAs were found. Thus, the information presented on labels are in disagreement with the results obtained both by GC-FID FA composition analysis and by ESI-MS lipid profile analysis. Principal component analysis (PCA) confirmed that the main contribution was from FAs found by GC-FID.

The present study investigated the activity of pyrimidine derivatives against Aedes aegypti. Two compounds, 3c and 3d showed excellent larvicide activity. Additionally, quantitative structure-activity relationship (QSAR) models were built using multiple-linear regression and partial least squares with descriptors generated from Dragon and VolSurf+ software, respectively. The best model is obtained with multiple linear regression (MLR), leading to a robust model. Moreover, the QSAR model is validated by means of some internal validation techniques in order to check its reliability, quality and robustness for predicting the larvicidal activity against A. aegypti. The models confirmed that the three-dimensional structure of molecules, steric properties, hydrophobic polar surface area, log partition (logP) and a simple pattern of substituent groups as methyl, methoxy, and succinimide in the pyrimidine derivatives are responsible for the larvicidal activity of the pyrimidine derivatives. Even more, the activity decreases by an electron-withdrawing group in R1 and increases when it is replaced by an aromatic ring activator group. These findings will aid in further studies of new pyrimidine derivatives active against Aedes aegypti.

Human milk (HM) lipids provide about 50% energy and facilitate the transport and absorption of fat-soluble vitamins for infants, but their composition should be studied when the milk is processed. This study evaluated the lipid quality of lyophilized and vacuum-packed colostrum (C), transitional (T), and mature (M) HM for a period of 1, 30, 60, 90, 120, 150, and 180 days of storage at –18 ºC. From the acidity (ºDornic), lipid content, and fatty acid (FA) composition results, no significant differences in the analyzed period were observed. The relative percentage of major triacylglycerols (TAG) of the HM was not altered, showing that the lyophilization process in HM is a good alternative that could be applied in human milk banks (HMB), increasing HM preservation and facilitating its transport and storage.

Coconut oil has several domestic uses and health benefits, which can be used in food, pharmaceuticals and cosmetics products. However, it has been the target of adulteration with lower price oils and fats, such as soybean oil. In this study, a fast, easy and simple methodology was used to detect low quantities of intentionally adulterated coconut oil with soybean oil by direct infusion electrospray ionization mass spectrometry (ESI-MS) at different levels (0, 2, 5, 10, 15, 20, 30, 50, 70 and 100%). In the oil industry, intentional adulterations usually occur with the addition of low quantities of soybean oil to coconut oil. Therefore, the suggested ESI-MS method is promising for routine analysis to guarantee the quality control of coconut oil since it is possible to detect adulteration with a minimal of 2% soybean oil addition.

Abstract Osteogenesis Imperfecta (OI) is a heterogeneous genetic disorder characterized by bone fragility and fracture. Mutations in 20 distinct genes can cause OI, and therefore, the genetic diagnosis of OI is frequently difficult to obtain because of the great number of genes that can be related with this disease. Studies that report the most frequently mutated genes in OI patients can help to improve molecular strategies for diagnosis of the disease. In order to characterize the mutation profile of OI in Brazilian patients, we analyzed 30 unrelated patients through SSCP screening, NGS gene panel, and/or Sanger sequencing for the 11 most frequently mutated genes in the database of mutations, including COL1A1, COL1A2, P3H1, CRTAP, PPIB, SERPINH1, SERPINF1, FKBP10, SP7, WNT1 and IFITM5. Disease-causing variants were identified in COL1A1, COL1A2, FKBP10, P3H1, and IFITM5. A total of 28 distinct mutations were identified, including seven novel changes. Our data show that the analysis of these five genes is able to detect at least 95% of causative mutations in OI disorder from Brazilian population. However, it has to be taken into considerations that distinct populations can have different frequencies of disease-causing variants. Hence, it is important to replicate this study in other groups.

Morinda citrifolia (noni) is found in the Atlantic and Amazonian Forest in Brazil and with recognized uses in folk medicine. However, few studies exist in its chemical characterization. The present work aims to investigate the efficiency of the sonication for extracting bioactive compounds from fruits and leaves of noni, using an experimental design of 23, whose variables were time, temperature and mass of the plant. Parallel extractions were performed using two extraction solvents (hexane and ethanol) and the same experimental design. The compounds were identified by gas chromatography coupled to mass spectrometry. The independent variable was the mass yield and all the variables studied in the sonication process were significant (p < 0.05%) for the process. Ethanol was more effective than hexane and the leaves produced higher yield and diversity of compounds than fruits. Vitamin E (tocopherol), octanoic acid, hydroxymethylfurfural, phytol and squalene were the main compounds identified.

For the accomplishment of Olympic and Paralympic Games in Rio de Janeiro city (Rio 2016), the government of the Rio de Janeiro state has undertaken to monitor air quality before and during the events. In Beijing, China, and Athens, Greece, the air quality was monitored in Olympic venues in order to evaluate the athletes’ performance in relation to the environment in which they were exposed. This study has the same proposal to Rio de Janeiro, Brazil. The air quality scenario of the three previous years (2013, 2014, 2015) of Rio 2016 was considered. Coarse (PM10) and fine (PM2.5) particles and O3 were monitored continuously on the stations located near to competition venues, as required by International Olympic Committee (IOC). The levels registered ranged from 6 to 96 μg m-3 for PM10, 1 to 44 μg m-3 for PM2.5 and 121 to 269 μg m-3 for O3. These concentrations exceeded the national and international air quality standards. These high concentrations are associated with uncountable civil works to build Olympic arenas and the urban mobility´s improvement. However, the concentrations for all the pollutants monitored in Rio de Janeiro city were in lower concentrations than in Beijing Olympic Games 2008.

Vegetable oils have properties that are beneficial to the human skin, and so, they are being used increasingly as ingredients of many cosmetic preparations; however, they are targets of adulteration. This study will evaluate the authenticity of Brazilian cosmetics based on vegetable oils (sesame, peanut, sunflower, and almonds) to determine the composition of fatty acids by gas chromatography with flame ionization detection (GC-FID) and the percentage of fatty acids in conjunction with chemometric treatments (principal component analysis), and lipid profiles using direct infusion electrospray ionization mass spectrometry (ESI-MS). The results obtained were compared with the respective pure vegetable oils. Of the seven brands analyzed, three were revealed as authentic, three were found to be adulterated with the addition of large amounts of soybean oil, and one showed no vegetable oils. These results demonstrate the relevance of quality control for cosmetics based on vegetable oils, which are more susceptible to adulteration owing to their higher cost when compared with vegetable oils such as soybean.

RESUMO Introdução: Trombofilia é a suscetibilidade à trombose, de natureza genética, adquirida ou mista. Entre as causas adquiridas, destaca-se a síndrome do anticorpo antifosfolípide (SAF) - doença autoimune caracterizada por anticorpos antifosfolípides, eventos trombóticos ou perda gestacional recorrente. O diagnóstico laboratorial baseia-se na detecção do anticoagulante lúpico (ACL), do anti-β2-glicoproteína 1 e da anticardiolipina; entretanto a execução do ACL ainda demanda uniformização. A última diretriz publicada pelo Clinical and Laboratory Standards Institute (CLSI) prioriza as etapas de triagem e confirmatória, em detrimento da mistura. Objetivos: Comparar as formas de liberação do ACL e adotar um protocolo de investigação em anuência às normas internacionais. Métodos: Trinta e seis amostras com resultados prolongados na etapa de triagem pelo ensaio do tempo do veneno da víbora de Russel (dRVVT) ou tempo de tromboplastina parcial ativada (TTPA) foram submetidas às etapas de mistura (1:1) e confirmatórias com altas concentrações de fosfolipídios. Resultados: Para o TTPA, foram considerados positivos os valores cujo cálculo do índice de circulação de anticoagulante (ICA) resultasse superior a 15%. Para o dRVVT, utilizou-se também o valor da razão entre triagem e confirmatória. Das amostras testadas, 14 revelaram correção na etapa da mistura, mas somente uma resultou em pesquisa negativa. Conclusão: O cálculo do ICA auxiliou na identificação dos anticorpos fracos que possivelmente sofreram diluição na etapa da mistura. Não há um exame padrão-ouro para o diagnóstico da SAF, e a pesquisa do ACL ainda demanda uniformização da técnica e da interpretação.

ABSTRACT Introduction: Thrombophilia is a thrombosis susceptibility of genetic, acquired or mixed nature. Among acquired causes, the antiphospholipid syndrome (APS) stands out as an autoimmune disease characterized by antiphospholipid antibodies, thrombotic events or recurrent gestational loss. Laboratory diagnosis is based on the detection of lupus anticoagulant (LAC), anti-β2-glycoprotein 1 and anticardiolipin; however the determination of LAC still demands uniformity. The last guideline published by the Clinical and Laboratory Standards Institute (CLSI) prioritizes the screening and confirmatory steps, to the detriment of the mixing phase. Objectives: To compare the forms of releasing the LAC and to adopt an investigation protocol in agreement with the international guidelines. Methods: Thirty-six samples with prolonged results in the screening step by the dilute Russell viper venom time (dRVVT) or activated partial thromboplastin time (APTT) were subjected to the mixing steps (1:1) and to the confirmatory steps with high concentrations of phospholipids. Results: For APTT, values whose indexes of circulating anticoagulant (ICA) were greater than 15% were considered positive. For dRVVT, the ratio between screening and confirmation was also used. Of the 36 tested samples, 14 showed correction in the mixing step, but only one resulted negative. Conclusion: ICA aided in identifying the weak antibodies that were probably diluted in the mixing step. There is no gold standard test for the diagnosis of APS, and LAC detection still requires standardization of technique and interpretation.

Omega-3 fish oil supplements are widely consumed as source of eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids, presenting beneficial effects on human health. This study aimed to evaluate fifteen brands of omega-3 fish oil supplements available in Brazilian market in order to estimate the Brazilian reality regarding those supplements. Twelve fatty acids were quantified by gas chromatography with a flame ionization detector (GC-FID), and lipid profile were obtained via mass spectrometry fingerprinting using direct electrospray ionization mass spectrometry (ESI-MS) to assess the form in which fatty acids are present as well as the possible fraud existence. Among all analyzed samples, thirteen brands were revealed as EPA and DHA sources (90.2-440.3 and 77.8-302.3 mg g-1lipid, respectively) in triacylglycerols (TAG) or ethyl esters (EE) form. However, two brands were discovered with addition of large amounts of soybean oil, leading the final consumer to ingest this low-cost oil believing that they are consuming adequate doses of EPA and DHA.

OBJETIVO: Validar um modelo de obesidade induzida por dieta hiperlipídica, de baixo custo, fácil reprodutibilidade, que mimetizasse características observadas no humano e viabilizasse posteriores proposições terapêuticas. MATERIAIS E MÉTODOS: Dezesseis camundongos Swiss receberam dieta padrão (DP) ou dieta hiperlipídica (DH), durante 10 semanas. RESULTADOS: Embora o grupo DP tenha apresentado maior consumo de água (p < 0,01) e ração (p < 0,001), o grupo DH apresentou maior ganho de peso corpóreo (p < 0,5) e aumento de coxins adiposos (p < 0,001), favorecendo maior índice de adiposidade (p < 0,001), glicemia (p < 0,01) e área sob a curva nos testes de tolerância à insulina (p < 0,001) e à glicose (p < 0,01). CONCLUSÃO: Validou-se um modelo de obesidade induzida por dieta hiperlipídica associada à resistência à ação da insulina e à intolerância à glicose, em um período de 10 semanas.

OBJECTIVE: Validate a model of high-fat diet-induced obesity, of low cost, easy reproducibility, that could express characteristics observed in human, and would enable subsequent therapy proposals. MATERIALS AND METHODS: Sixteen Swiss mice received a standard diet (DP) or high-fat diet (DH) for 10 weeks. RESULTS: Although the DP group had greater water (p < 0.01) and feed (p < 0.001) consumption, the DH group had greater body weight (p < 0.5) and adipose tissue gain (p < 0.001), favoring higher adiposity index (p < 0.001), glucose (p < 0.01), and area under the curve in the insulin (p < 0.001) and glucose (p < 0.01) tolerance tests. CONCLUSION: A high-fat diet-induced obesity model has been validated, which was also associated with insulin resistance and glucose intolerance after a period of 10 weeks.
