Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

Genetics of Parkinson’s disease in Brazil: a systematic review of monogenic forms

RESUMO Introdução: Um número crescente de mutações causando formas monogênicas de doença de Parkinson (DP) tem sido descrito, principalmente entre pacientes da Europa e da América do Norte. Como a arquitetura genética varia entre diferentes populações, entender os perfis genéticos específicos de pacientes brasileiros é essencial para um melhor aconselhamento genético e para a seleção de participantes para ensaios clínicos. Objetivo: Revisão sistemática para identificar estudos genéticos brasileiros na área e definir o cenário para estudos futuros das formas monogênicas de DP no Brasil. Métodos: Nós pesquisamos as bases de dados MEDLINE, EMBASE e Web of Science desde a criação até dezembro de 2019, usando termos para “Parkinson’s disease”, “genetics” e “Brazil”. A extração de dados foi feita por dois revisores independentes. Para os genes LRRK2 e PRKN, calculamos a prevalência estimada para cada estudo e realizamos uma meta-análise. Resultados: Um total de 32 estudos foram incluídos e 94 pacientes brasileiros com DP com mutações causativas foram identificados em 2872 pacientes avaliados (3.2%). As mutações no PRKN causaram DP em 48 de 576 pacientes (8.3%). As mutações no LRRK2 foram identificadas em 40 de 1566 pacientes (2.5%), sendo a mutação mais comum a p.G2019S (2.2%). Conclusões: As mutações na PRKN são a causa mais comum de DP autossômica recessiva, e as mutações no LRRK2 a causa mais comum de DP autossômica dominante. Nós observamos uma falta de estudos epidemiológicos robustos em genética de DP, especialmente por não levar em conta a diversidade de nossa população.
ABSTRACT Background: Increasing numbers of mutations causing monogenic forms of Parkinson's disease (PD) have been described, mostly among patients in Europe and North America. Since genetic architecture varies between different populations, studying the specific genetic profile of Brazilian patients is essential for improving genetic counseling and for selecting patients for clinical trials. Objective: We conducted a systematic review to identify genetic studies on Brazilian patients and to set a background for future studies on monogenic forms of PD in Brazil. Methods: We searched MEDLINE, EMBASE and Web of Science from inception to December 2019 using terms for "Parkinson's disease", "genetics" and "Brazil". Two independent reviewers extracted the data. For the genes LRRK2 and PRKN, the estimated prevalence was calculated for each study, and a meta-analysis was performed. Results: A total of 32 studies were included, comprising 94 Brazilian patients with PD with a causative mutation, identified from among 2,872 screened patients (3.2%). PRKN mutations were causative of PD in 48 patients out of 576 (8.3%). LRRK2 mutations were identified in 40 out of 1,556 patients (2.5%), and p.G2019S was the most common mutation (2.2%). Conclusions: PRKN is the most common autosomal recessive cause of PD, and LRRK2 is the most common autosomal dominant form. We observed that there was a lack of robust epidemiological studies on PD genetics in Brazil and, especially, that the diversity of Brazil’s population had not been considered.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A direct dynamics study of protonated alcohol dehydration and the Diels-Alder reaction

As dinâmicas da desidratação do álcool protonado (R)-3,3-dimetilbutan-2-ol (álcool pinacolil), [(CH3)3C-CH(OH2)CH3]+, e da cicloadição eteno + 1,3-butadieno foram estudadas com a técnica de dinâmica molecular direta de Born-Oppenheimer (BOMD) usando o métodoAM1. Mais de 10.000 trajetórias foram geradas, em que a maioria foi utilizada no procedimento ainda não explorado de recozimento simulado/fragmentação (SA/F). A energia potencial obtida com o método AM1 (PES-AM1) para o álcool protonado apresenta dois estados de transição associados ao intermediário complexo [(CH3)3C-CHCH3]+hhhOH2 e a migração do grupo CH3 levando ao produto complexo [(CH3)2C-CH(CH3)2]+hhhOH2. A dinâmica direta forneceu um número desprezível de trajetórias envolvendo estes complexos, pois o momento adquirido pelo fragmento H2O leva à dissociação completa. Portanto, o rearranjo do carbocátion secundário [(CH3)3C-CHCH3]+ foi praticamente inexistente durante a dinâmica. Apesar do caminho concertado (dissociação do fragmento H2O e migração do grupo CH3) não ser uma coordenada de reação intrínseca (IRC) no método PESAM1, um número significativo de trajetórias envolveu este caminho. Para a reação de Diels-Alder, mesmo partindo-se do estado de transição simétrico, e utilizando uma função de ondaAM1 restrita, a dinâmica forneceu um número significativo de trajetórias que seguiram caminhos assimétricos, isto é, não-IRC, em direção ao ciclohexeno, independentemente do procedimento utilizado na inicialização. E notável ainda que todas estas trajetórias que seguiram caminhos assimétricos envolveram um caminho de reação concertado.
The dynamics of dehydration of the protonated (R)-3,3-dimethylbutan-2-ol (pinacolyl alcohol), [(CH3)3C-CH(OH2)CH3]+, and of ethene + 1,3-butadiene cycloaddition were studied with the Born-Oppenheimer molecular dynamics (BOMD) technique for direct dynamics using the AM1 method. More than 10,000 trajectories were generated, most of them related to the unexplored simulated annealing/fragmentation approach. The AM1 potential energy surface (PES) for the protonated pinacolyl alcohol presents two transition states related to the [(CH3)3C-CHCH3]+hhhOH2 intermediate complex and to CH3 migration leading to the [(CH3)2C-CH(CH)3]2+hhhOH2 product complex. Direct dynamics yielded negligible trajectories involving these complexes, since the momentum acquired by the H2O fragment led to a complete dissociation. Thus, rearrangement of the secondary carbocation [(CH3)3C-CHCH3]+ was practically inexistent during the dynamics. Despite the concerted path (H2O dissociation and CH3 migration) not being an IRC (intrinsic reaction coordinate) path in AM1-PES, a statistically significant number of trajectories involved this path. As for the Diels-Alder reaction, even when started from a symmetric transition state using the spin restricted AM1 wavefunction, the dynamics yielded a significant number of trajectories that followed asymmetric, i.e.non-IRC, paths toward cyclohexene, independent of the initialization approach. It is noteworthy that all these asymmetric path trajectories led to a concerted reaction mechanism.

Biflavonoids and other phenolics from Caesalpinia pyramidalis (Fabaceae)

O reestudo do extrato clorofórmico das folhas de Caesalpinia pyramidalis (Caesalpinioidea, Fabaceae) forneceu, além do novo biflavonóide denominado caesalflavona, podocarpusflavona A, agathisflavona, apigenina, kaempferol, sitosterol e lupeol. Por outro lado, a partir do extrato clorofórmico do caule foram obtidos 4, 4'-diidroxi-2'-metoxi-chalcona, (-)-siringaresinol e galato de metila. Não foram encontrados biflavonóides nesta parte da planta. Até o presente, C. pyramidalis é a primeira espécie do gênero que contém biflavonóides. As estruturas das substâncias foram estabelecidas através da análise dos seus dados espectrométricos utilizando-se técnicas de EM, IV, UV, RMN 1D e 2D.
The chloroform extract of the leaves of Caesalpinia pyramidalis (Caesalpinioidea, Fabaceae) yielded the new biflavonoid named caesalflavone, as well as podocarpusflavone A, agathisflavone, apigenin and kaempferol. The chloroform extract of the trunk wood gave 4,4'-dihydroxy-2'-methoxychalcone, (-)-syringaresinol, and methyl gallate. Biflavonoids were not found in trunk wood. Until now, C. pyramidalis is the first species in the genus to present biflavonoids. The structural elucidation of the isolated compounds and their derivatives were based on MS, IR, UV, 1D and 2D NMR spectral analyses.
