Immunopathological evaluation of oral squamous cell carcinomas: a case series

Resumen La evaluación de la expresión del ligando 1 de muerte programada (PD-L1) es un criterio de elegibilidad para la inmunoterapia anti-PD-L1 en el carcinoma oral de células escamosas (COCE). También deben evaluarse otros componentes del microambiente tisular. Aquí se informaron cuatro casos de carcinoma de células escamosas orales. Se realizaron análisis histopatológicos del grado tumoral y TIL (infiltrado inflamatorio asociado a tumor) e inmunohistoquímica para CD4, CD8 y PD-L1. Los casos se clasificaron según el microambiente tisular en resistencia inmunitaria adaptativa, ignorancia inmunológica, tolerancia o inducción intrínseca. Este análisis destaca la importancia de comprender el microambiente inmunológico que puede presentar el COCE para la identificación de dianas terapéuticas más eficaces contra la enfermedad.
Abstract The evaluation of the expression of programmed cell death protein (PD-L1) is an eligibility criterion for anti-PD-L1 immunotherapy in oral squamous cell carcinoma (OSCC). Other components of tissue microenvironment should also be evaluated. Here, four cases of oral squamous cells carcinoma were reported. Histopathological analysis of tumor grade and TILs and immunohistochemistry for CD4, CD8, and PD-L1 was done. The cases were classified according to the tissue microenvironment in adaptive immune resistance, immunological ignorance, tolerance, or intrinsic induction. This analysis highlights the importance of understanding the immunological microenvironment that the OSCC can present for the identification of more effective therapeutic targets against the disease.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

This guideline aimed to update the concepts and formulate the standards of conduct and scientific evidence that support them, regarding the diagnosis and treatment of the Cardiomyopathy of Chagas disease, with special emphasis on the rationality base that supported it.nbsp;
Chagas disease in the 21st century maintains an epidemiological pattern of endemicity in 21 Latin American countries. Researchers and managers from endemic and non-endemic countries point to the need to adopt comprehensive public health policies to effectively control the interhuman transmission of T. cruzi infection, and to obtain an optimized level of care for already infected individuals, focusing on diagnostic and therapeutic opportunistic opportunities.

Pathogenic and pathophysiological mechanisms of the Cardiomyopathy of Chagas disease were revisited after in-depth updating and the notion that necrosis and fibrosis are stimulated by tissue parasitic persistence and adverse immune reaction, as fundamental mechanisms, assisted by autonomic and microvascular disorders, was well established. Some of them have recently formed potential targets of therapies.nbsp;
The natural history of the acute and chronic phases was reviewed, with enhancement for oral transmission, indeterminate form and chronic syndromes. Recent meta-analyses of observational studies have estimated the risk of evolution from acute and indeterminate forms and mortality after chronic cardiomyopathy. Therapeutic approaches applicable to individuals with Indeterminate form of Chagas disease were specifically addressed. All methods to detect structural and/or functional alterations with various cardiac imaging techniques were also reviewed, with recommendations for use in various clinical scenarios. Mortality risk stratification based on the Rassi score, with recent studies of its application, was complemented by methods that detect myocardial fibrosis.nbsp;
The current methodology for etiological diagnosis and the consequent implications of trypanonomic treatment deserved a comprehensive and in-depth approach. Also the treatment of patients at risk or with heart failure, arrhythmias and thromboembolic events, based on pharmacological and complementary resources, received special attention. Additional chapters supported the conducts applicable to several special contexts, including t. cruzi/HIV co-infection, risk during surgeries, in pregnant women, in the reactivation of infection after heart transplantation, and others.nbsp; nbsp;nbsp;
Finally, two chapters of great social significance, addressing the structuring of specialized services to care for individuals with the Cardiomyopathy of Chagas disease, and reviewing the concepts of severe heart disease and its medical-labor implications completed this guideline.
Esta diretriz teve como objetivo principal atualizar os conceitos e formular as normas de conduta e evidências científicas que as suportam, quanto ao diagnóstico e tratamento da CDC, com especial ênfase na base de racionalidade que a embasou.
A DC no século XXI mantém padrão epidemiológico de endemicidade em 21 países da América Latina. Investigadores e gestores de países endêmicos e não endêmicos indigitam a necessidade de se adotarem políticas abrangentes, de saúde pública, para controle eficaz da transmissão inter-humanos da infecção pelo T. cruzi, e obter-se nível otimizado de atendimento aos indivíduos já infectados, com foco em oportunização diagnóstica e terapêutica.
Mecanismos patogênicos e fisiopatológicos da CDC foram revisitados após atualização aprofundada e ficou bem consolidada a noção de que necrose e fibrose sejam estimuladas pela persistência parasitária tissular e reação imune adversa, como mecanismos fundamentais, coadjuvados por distúrbios autonômicos e microvasculares. Alguns deles recentemente constituíram alvos potenciais de terapêuticas.
A história natural das fases aguda e crônica foi revista, com realce para a transmissão oral, a forma indeterminada e as síndromes crônicas. Metanálises recentes de estudos observacionais estimaram o risco de evolução a partir das formas aguda e indeterminada e de mortalidade após instalação da cardiomiopatia crônica. Condutas terapêuticas aplicáveis aos indivíduos com a FIDC foram abordadas especificamente. Todos os métodos para detectar alterações estruturais e/ou funcionais com variadas técnicas de imageamento cardíaco também foram revisados, com recomendações de uso nos vários cenários clínicos. Estratificação de risco de mortalidade fundamentada no escore de Rassi, com estudos recentes de sua aplicação, foi complementada por métodos que detectam fibrose miocárdica.
A metodologia atual para diagnóstico etiológico e as consequentes implicações do tratamento tripanossomicida mereceram enfoque abrangente e aprofundado. Também o tratamento de pacientes em risco ou com insuficiência cardíaca, arritmias e eventos tromboembólicos, baseado em recursos farmacológicos e complementares, recebeu especial atenção. Capítulos suplementares subsidiaram as condutas aplicáveis a diversos contextos especiais, entre eles o da co-infecção por T. cruzi/HIV, risco durante cirurgias, em grávidas, na reativação da infecção após transplante cardíacos, e outros.nbsp;nbsp;nbsp;
Por fim, dois capítulos de grande significado social, abordando a estruturação de serviços especializados para atendimento aos indivíduos com a CDC, e revisando os conceitos de cardiopatia grave e suas implicações médico-trabalhistas completaram esta diretriz.nbsp;

RESUMO Objetivo: Descrever as práticas de ressuscitação volêmica em unidades de terapia intensiva brasileiras e compará-las com as de outros países participantes do estudo Fluid-TRIPS. Métodos: Este foi um estudo observacional transversal, prospectivo e internacional, de uma amostra de conveniência de unidades de terapia intensiva de 27 países (inclusive o Brasil), com utilização da base de dados Fluid-TRIPS compilada em 2014. Descrevemos os padrões de ressuscitação volêmica utilizados no Brasil em comparação com os de outros países e identificamos os fatores associados com a escolha dos fluidos. Resultados: No dia do estudo, foram incluídos 3.214 pacientes do Brasil e 3.493 pacientes de outros países, dos quais, respectivamente, 16,1% e 26,8% (p < 0,001) receberam fluidos. A principal indicação para ressuscitação volêmica foi comprometimento da perfusão e/ou baixo débito cardíaco (Brasil 71,7% versus outros países 56,4%; p < 0,001). No Brasil, a percentagem de pacientes que receberam soluções cristaloides foi mais elevada (97,7% versus 76,8%; p < 0,001), e solução de cloreto de sódio a 0,9% foi o cristaloide mais comumente utilizado (62,5% versus 27,1%; p < 0,001). A análise multivariada sugeriu que os níveis de albumina se associaram com o uso tanto de cristaloides quanto de coloides, enquanto o tipo de prescritor dos fluidos se associou apenas com o uso de cristaloides. Conclusão: Nossos resultados sugerem que cristaloides são usados mais frequentemente do que coloides para ressuscitação no Brasil, e essa discrepância, em termos de frequências, é mais elevada do que em outros países. A solução de cloreto de sódio 0,9% foi o cristaloide mais frequentemente prescrito. Os níveis de albumina sérica e o tipo de prescritor de fluidos foram os fatores associados com a escolha de cristaloides ou coloides para a prescrição de fluidos.
Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.

OBJECTIVES: Craniofacial resection (CFR) procedures for craniofacial tumors with cranial extension are often extensive. Although CFRs may yield good oncological results, there are concerns about high perioperative morbidity and mortality. This study aimed to determine risk factors for perioperative mortality after open CFR in terms of deaths occurring during index hospitalizations. METHODS: We conducted a retrospective analysis of CFRs conducted at a tertiary oncology hospital from May 2009 through December 2018. RESULTS: Our analysis included data from the medical records of 102 patients, the majority of whom were male (n=74, 72.5%). The mean age was 61 years (±18.3 years). Skin malignancies (n=64, 63.4%) accounted for nearly two-thirds of the treated tumors, and most of these were squamous cell carcinoma. Postoperative medical complications occurred in 33 patients (33%), and surgical complications occurred in 48 (47%). Multivariate analysis revealed the only independent risk factors for perioperative deaths to be the presence of intracranial tumor extension on preoperative imaging (hazard ratio [HR]=4.56; 95% confidence interval [CI]: 1.74-11.97; p=0.002) and the unexpected emergence of postoperative neurological dysfunction (HR=10.9; 95% CI: 2.21-54.3; p=0.003). CONCLUSIONS: In our study, factors related to tumor extension were associated with a higher risk of perioperative death.

Resumo Fundamento A síndrome de takotsubo (takotsubo) é uma forma de cardiomiopatia adquirida. Dados nacionais sobre essa condição são escassos. O Registro REMUTA é o primeiro a incluir dados multicêntricos dessa condição no nosso país. Objetivo Descrever as características clínicas, prognóstico, tratamento intra-hospitalar e mortalidade hospitalar e em 1 ano de seguimento. Métodos Estudo observacional, retrospectivo, tipo registro. Incluídos pacientes internados com diagnóstico de takotsubo ou que desenvolveram esta condição durante internação por outra causa. Os desfechos avaliados incluíram fator desencadeador, análise dos exames, uso de medicações, complicações e óbito intra-hospitalar e em 1 ano de seguimento. O nível de significância adotado foi de 5%. Resultados Foram incluídos 169 pacientes, em 12 centros no Estado do Rio de Janeiro. A idade média foi de 70,9 ± 14,1 anos e 90,5% eram do sexo feminino; 63% dos casos foram de takotsubo primário e 37% secundário. Troponina I foi positiva em 92,5% dos pacientes e a mediana de BNP foi de 395 (176,5; 1725). Supradesnivelamento do segmento ST esteve presente em 28% dos pacientes. A fração de ejeção do ventrículo esquerdo teve mediana de 40 (35; 48)%. Observamos taxa de 25,7% de ventilação mecânica invasiva e 17,4% de choque. Suporte circulatório mecânico foi utilizado em 7,7%. A mortalidade intra-hospitalar foi de 10,6% e a mortalidade ao final de 1 ano foi de 16,5%. Takotsubo secundário e choque cardiogênico foram preditores independentes de mortalidade. Conclusão Os resultados do REMUTA mostram que takotsubo não se trata de patologia benigna como se pensava, especialmente no grupo de takotsubo secundário que acarreta elevada taxa de complicações e de mortalidade. Troponin I was positive in 92.5% of patients, and median BNP was 395 (176.5; 1725). ST-segment elevation was present in 28% of patients. Median left ventricular ejection fraction was 40 (35; 48)%. We observed invasive mechanical ventilation in 25.7% of cases and shock in 17.4%. Mechanical circulatory support was used in 7.7%. In-hospital mortality was 10.6%, and mortality at 1 year of follow-up was 16.5%. Secondary TTS and cardiogenic shock were independent predictors of mortality. Conclusion The results of the REMUTA show that TTS is not a benign pathology, as was once thought, especially regarding the secondary TTS group, which has a high rate of complications and mortality. OBJECTIVES: We designed a cohort study to describe characteristics and outcomes of patients with coronavirus disease (COVID-19) admitted to the intensive care unit (ICU) in the largest public hospital in Sao Paulo, Brazil, as Latin America becomes the epicenter of the pandemic. METHODS: This is the protocol for a study being conducted at an academic hospital in Brazil with 300 adult ICU beds dedicated to COVID-19 patients. We will include adult patients admitted to the ICU with suspected or confirmed COVID-19 during the study period. The main outcome is ICU survival at 28 days. Data will be collected prospectively and retrospectively by trained investigators from the hospital’s electronic medical records, using an electronic data capture tool. We will collect data on demographics, comorbidities, severity of disease, and laboratorial test results at admission. Information on the need for advanced life support and ventilator parameters will be collected during ICU stay. Patients will be followed up for 28 days in the ICU and 60 days in the hospital. We will plot Kaplan-Meier curves to estimate ICU and hospital survival and perform survival analysis using the Cox proportional hazards model to identify the main risk factors for mortality. RESULTS: We expect to include a large sample of patients with COVID-19 admitted to the ICU and to be able to provide data on admission characteristics, use of advanced life support, ICU survival at 28 days, and hospital survival at 60 days. CONCLUSIONS: This study will provide epidemiological data about critically ill patients with COVID-19 in Brazil, which could inform health policy and resource allocation in low- and middle-income countries.

Abstract: Several microbiological indicators of soil quality present high sensitivity, but little is known about the influence of topographic factors on them. This work aimed to evaluate variability of biological indicators of soil quality across a hillslope under native forest and the influence of topographic factors on them. Four positions on a hillslope were evaluated. Activity of the enzymes β-glucosidase, acid phosphatase, urease and fluorescein diacetate (FDA) hydrolysis were determined, as well as basal and substrate-induced respiration, and density of microorganisms: total bacteria, total fungi, actinobacteria, phosphate solubilizers, ammonifiers, native rhizobia, free-living N2-fixing bacteria, spores of arbuscular mycorrhizal fungi and percentage of root colonization by arbuscular mycorrhizal fungi. Activity and density of microorganisms were correlated with topographic factors. The relation of these factors to the variations of the evaluated indicators was determined using the random forest algorithm. Microbiological indicators varied according to the hillslope positions. The indicators urease, basal respiration, spore density, mycorrhizal colonization, total bacteria and fungi, phosphate solubilizers, and free-living N2-fixing bacteria detected in JNFB and FAM culture medium did not vary with terrain attributes and were therefore more indicated in cases of topographic variations. This and future studies can help to select the best microbiological indicators for different conditions.

Serum immunoglobulin from Nellore cattle produced by in vitro fertilization and treated for umbilical diseases

RESUMO: Os objetivos desse estudo foram mensurar as concentrações séricas das imunoglobulinas de bovinos Nelore, produzidos por fertilização in vitro (FIV), acometidos por onfalopatias e avaliar a excisão cirúrgica como método de tratamento. Utilizou-se 16 animais, com idade variando de 1 a 15 meses, machos e fêmeas, acometidos por afecções umbilicais. Amostras de sangue foram colhidas para realização de hemogramas e avaliação das imunoglobulinas, por meio de eletroforese e turbidimetria com sulfato de zinco. Foram observados quatro casos de hérnias umbilicais, dois de hérnia umbilical associadas à persistência de úraco, dois casos de hérnia umbilical com persistência de úraco e onfaloarterite, três de hérnia umbilical com divertículo de úraco, três casos de persistência de úraco, um de divertículo de úraco e um de onfalite. O hemograma revelou diferença significativa para os valores de Volume Globular e Leucócitos, nos momentos pré e pós-tratamento cirúrgico dos animais. A eletroforese evidenciou valores de imunoglobulina abaixo da normalidade na maioria dos animais, enquanto que a turbidimetria apresentou níveis normais na maioria dos bovinos, quando comparados com valores de referência. A maioria dos bovinos estudados, acometidos de onfalopatias e concebidos por FIV, apresentaram hipoglobulinemia. Os resultados obtidos não permitem afirmar que existe relação entre as onfalopatias, falha de transferência de imunidade passiva e FIV.
ABSTRACT: The aim of this study was to measure serum immunoglobulin concentrations of Nellore cattle produced by in vitro fertilization (IVF) with umbilical diseases and to evaluate surgical excision as a method of treatment. Sixteen cattle with ages ranging from 1 to 15 months, males and females, affected by umbilical diseases were enrolled in the study. Blood samples were collected for cell counts and the determination of immunoglobulin concentrations by electrophoresis and zinc sulphate turbidimetry (ZST). Four calves were presented with umbilical herniation, two with an umbilical herniation associated with a persistent urachus, two with an umbilical herniation with a persistent urachus and omphaloarteritis, three with an umbilical herniation and an urachal diverticulum, three with a persistent urachus, one with an urachal diverticulum, and one with omphalitis. The blood cell counts pre- and post-surgical revealed differences in cell volume and the number of leukocytes. The immunoglobulin values measured by electrophoresis values were below normal in most animals, whereas the ZST showed normal levels in most of them. Most of the calves affected by umbilical diseases and produced by IVF presented hypoglobulinaemia. Correlations between umbilical diseases, failure of passive transfer of immunity and IVF could not be demonstrated.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

FUNDAMENTO: O sistema de mapeamento eletro anatômico Ensite Navx é muito usado na ablação por radiofrequência (RF) da fibrilação atrial, ajudando na confecção de lesões lineares. Contudo, a correspondência da linha virtual criada pelo Ensite com a lesão patológica ainda não foi avaliada. OBJETIVO: Avaliar a continuidade da linha virtual criada pelo Ensite em modelo suíno. MÉTODOS: Realizamos ablação linear por RF (cateter de 8 mm e irrigado) em ambos os átrios de 14 suínos (35 kg) guiada pelo EnSite. Os animais foram sacrificados 14 dias pós-ablação para análise macroscópica e histológica. RESULTADOS: Foram confeccionadas 23 lesões lineares em átrio direito e 21 em átrio esquerdo dos 14 animais. A potência, temperatura e impedância médias das aplicações foram de 56 W, 54 ºC e 231 Ω para o cateter de 8mm, e de 39 W, 37 ºC e 194 Ω para o cateter irrigado. Todas (100%) as linhas foram identificadas nas faces epicárdica e endocárdica, denotando transmuralidade. À macroscopia, as lesões eram extensas e pálidas, com 3,61 cm de comprimento e 0,71 cm de profundidade e contínuas. A transmuralidade das lesões foi confirmada pela microscopia. Houve correspondência na localização das linhas do mapa virtual com as da peça anatômica em 21 das 23 (91,3%) das linhas do átrio direito e 19/21 (90,4%) do átrio esquerdo. CONCLUSÃO: Nesse modelo, as linhas criadas no mapa virtual pelo sistema EnSiteNavX se correlacionam a lesões lineares transmurais contínuas na peça anatômica, sugerindo que esse método é adequado para a ablação linear da fibrilação atrial.
BACKGROUND: EnSiteNavx electroanatomic mapping system is widely used in radiofrequency (RF) atrial fibrillation ablation, helping the creation of linear lesions. However, the correspondence of the virtual line created by EnSite with the pathological lesion has not yet been evaluated. OBJECTIVE: to assess the continuousness of Ensite-guided virtual lines in a swine model. METHODS: we performed RF ablation linear lesions (8mm and irrigated catheters tips) in both atria of 14 pigs (35Kg) guided by the EnSite. The animals were sacrificed 14 days post-ablation for macroscopic and histological analysis. RESULTS: a total of 23 lines in the right atrium and 21 lines in the left atrium were created in 14 animals. The medium power, impedance and temperature applications were 56 W, 54 ºC and 231 Ω for the 8mm tip, and 39W, 37ºC, 194 Ω for the irrigated tip catheter, respectively. All (100%) lines were identified on the epicardial and endocardial surfaces, denoting transmurality. At macroscopic examination, lesions were extensive and pale, continuous, with 3.61 cm long and 0.71 cm deep. The transmurality of the lesions was confirmed by microscopy. There was a correlation in the location of the lines at the virtual map and the anatomical lesions in 21 of 23 (91.3%) of the right atrium and 19/21 (90.4%) of the left atrium. CONCLUSION: In this model, the lines created in the virtual map by EnSiteNavX system correspond to continuous transmural linear lesions in anatomical specimen, suggesting that this method is suitable for linear ablation of atrial fibrillation.

Neste trabalho caracterizou-se a qualidade dos frutos em diferentes pontos de colheita para armazenamento das cultivares de maracujá: BRS Gigante Amarelo, BRS Ouro Vermelho e BRS Sol do Cerrado. Foram avaliados as características físicas dos frutos (aparência, perda de massa, coloração da casca) e os parâmetros químicos do suco (conteúdo de acidez, pH, ácido ascórbico, sólidos solúveis totais, açúcares solúveis totais e açúcares redutores). Utilizou-se uma Amostragem Simples ao Acaso (ASA), com 0,05 de significância e as médias foram comparadas pelo Teste "t" por Intervalo de Confiança. Os frutos colhidos nos estádios iniciais de amadurecimento apresentaram maior perda de massa e alteração da qualidade do suco. Conclui-se que os frutos das diferentes cultivares colhidos na safra de verão (novembro/dezembro) devem ter pelo menos 55% de casca com coloração amarela para proporcionar menor perda de massa, manter a qualidade do suco e aumentar a sua vida útil.
In this study the fruit quality was characterized at different harvest points for storage of varieties of passion fruits: BRS Gigante Amarelo, BRS Ouro Vermelho and BRS Sol do Cerrado. The physical characteristics of fruits (appearance, mass loss and peel color) and chemical parameters of juice (content of acidity, pH, ascorbic acid, total soluble solids, total soluble sugars and reducing sugars) were characterized. The method of Simple Random Sampling (SRS) was applied, with 0.05 level of significance, and means were compared using the "t" Test for Confidence Intervals. Fruits harvested in the early ripening stages showed a higher mass loss as well as the worst juice quality. It was concluded that fruits of different cultivars harvested in summer seasons must present at least 55% of yellow peel color to render the lower mass loss, the best quality of juice and to extend the storage time.

We extended our previous exploration of sulfur bridges as bioisosteric replacements for atoms forming the bridge between the aromatic rings of combretastatin A-4. Employing coupling reactions between 5-iodo-1,2,3-trimethoxybenzene and substituted thiols, followed by oxidation to sulfones with m-CPBA, different locations for attaching the sulfur atom to ring A through the synthesis of nine compounds were examined. Antitubulin activity was performed with electrophoretically homogenous bovine brain tubulin, and activity occurred with the 1,2,3-trimethoxy-4-[(4-methoxyphenyl)thio]benzene (12), while the other compounds were inactive. The compounds were also tested for leishmanicidal activity using promastigote forms of Leishmania braziliensis (MHOM/BR175/M2904), and the greatest activity was observed with 1,2,3-trimethoxy-4-(phenylthio)benzene (10) and 1,2,3-trimethoxy-4-[(4-methoxyphenyl) sulfinyl]benzene (15).

Soil fauna as an indicator of soil quality in forest stands, pasture and secondary forest

As interações entre os invertebrados edáficos e as variações ambientais, para avaliação da qualidade do solo, são pouco conhecidas. Nesse sentido, o objetivo deste estudo foi avaliar a qualidade do solo em áreas com diferentes manejos, utilizando a fauna edáfica como indicador. O trabalho foi realizado em Além Paraíba, MG, em cinco áreas com diferentes coberturas vegetais (acácia, mimosa, eucalipt, pastagem e floresta secundária). Para a amostragem da comunidade de invertebrados do solo, foram utilizadas armadilhas "pit fall", nas estações seca e chuvosa. Foram determinados a abundância da fauna, as riquezas média e total, o índice de diversidade de Shannon, o índice de uniformidade de Pielou e o índice de mudança V. As áreas de floresta secundária e plantio de mimosa apresentaram a maior abundância da fauna no período seco (111,3 e 31,7 indivíduos por armadilha por dia, respectivamente). No período chuvoso, as áreas de floresta secundária, de mimosa e de acácia não diferiram. Os maiores valores de riqueza média e total foram quantificados na área de floresta secundária no período seco e na área de mimosa no período chuvoso. O índice de Shannon variou de 1,57 nas áreas de acácia e eucalipto, no período chuvoso, a 3,19 na área de eucalipto, no período seco. O índice de uniformidade de Pielou foi maior para os povoamentos florestais (eucalipto, acácia e mimosa), na estação seca. Já na estação chuvosa, as áreas de pastagem e floresta secundária superaram os povoamentos florestais. Pela aplicação do índice de mudança V, a área de mimosa foi a que apresentou a menor percentagem de grupos extremamente inibidos, tanto no período seco quanto no chuvoso (36 e 23 %, respectivamente). Entre os povoamentos florestais, a área de mimosa foi a que proporcionou a maior abundância de fauna edáfica.
The interactions between soil invertebrates and environmental variations are relatively unknown in the assessment of soil quality. The objective of this study was to evaluate soil quality in areas with different soil management systems, based on soil fauna as indicator, in Além Paraíba, Minas Gerais, Brazil. The soil invertebrate community was sampled using pitfall traps, in the dry and rainy seasons, from areas with five vegetation types (acacia, mimosa, eucalyptus, pasture, and secondary forest). The abundance of organisms and the total and average richness, Shannon's diversity index, the Pielou uniformity index, and change index V were determined. The fauna was most abundant in the areas of secondary forest and mimosa plantations in the dry season (111.3 and 31.7 individuals per trap per day, respectively). In the rainy season, the abundance of organisms in the three vegetation types did not differ. The highest values of average and total richness were recorded in the secondary forest in the dry season and in the mimosa stand in the rainy season. Shannon's index ranged from 1.57 in areas with acacia and eucalyptus in the rainy season to 3.19 in the eucalyptus area in the dry season. The uniformity index was highest in forest stands (eucalyptus, acacia and mimosa) in the dry season, but higher in the rainy season in the pasture and secondary forest than in the forest stands. The change index V indicated that the percentage of extremely inhibited groups was lowest in the area with mimosa, both in the dry and rainy season (36 and 23 %, respectively). Of all forest stands, the mimosa area had the most abundant soil fauna.
