Abstract Healthcare workers present an increased risk of contagion for the SARS-CoV-2 virus due to their labor exposure. Here, we describe the clinical, laboratory, and immunological characteristics of healthcare workers, before vaccine application, exposed to SARS-CoV-2-infected patients. We collected sociodemographic, clinical, and laboratory information from 50 professionals who worked during the COVID-19 pandemic at the Clinical Hospital of the Northwest in Brazil. The results showed that most workers are women, over 50 years old, and worked as nursing technicians. Approximately 56% of workers were positive for a previous infection by RT-PCR and/or anti-SARS-CoV-2-immunoglobulin tests. Increased levels of hematocrit, neutrophils, NK lymphocytes, and fibrinogen, were found in positive healthcare workers, suggesting a light inflammatory status. The immunological findings showed an increase in IL-17 production and a Th2/Th17/Th22 profile followed by high serology for anti-SARS-CoV-2 IgM and IgG. Those data reveal the importance of studies with healthcare workers to investigate if the continuous exposition to the virus may result in chronic activation of the immune system and/or pulmonary inflammation in this target group.

Analytical method validation for the identification and quantification of dissolved gold and gold nanoparticles in cosmetics products by single particle inductively coupled plasma mass spectrometry

Nanoparticle (NP) applications have recently gained more space in the development of many products, especially in cosmetology. The aim of the present study was to validate a single particle inductively coupled plasma mass spectrometry technique for the detection and quantification of gold nanoparticles (AuNPs) and gold ions (Au (i)) in different cosmetics currently marketed in Brazil. The size limit of detection for pure AuNPs was 22 nm, and a concentration limit of detection of 2.3×106 particles g−1 was achieved. The limit of detection for total gold was 0.02 μg L−1 obtained in standard mode conditions. The recovery achieved was 98% for dissolved Au and 109% for AuNPs; the relative standard deviation was <5%. This validated method was applied to cosmetic products (n= 10) from different national and international industrial producers. Most cosmetics analysed contained quantifiable amounts of dissolved gold (< 0.005-2.1 μg g−1) and only four contained quantifiable amounts AuNPs the variability of the results detected (< LOD – 9.3 × 108particles g−1). The products that contained the greatest amount of these metallic NPs were the artisanal products. The results demonstrate the importance of adequate quality control for cosmetics to ensure consumer safety.

DETECTION AND QUANTIFICATION OF SILVER NANOPARTICLES BY spICP-MS. The growing interest in nanotechnology has led to an increase in the production and application of nanoparticles worldwide. Due to the unique functional properties of nanoparticles, these materials are being used by many industries, including the agricultural and food sectors. Among the commercially available nanomaterials, it is possible to highlight those produced with silver nanoparticles. One of the most promising techniques for the analysis of metallic nanoparticles is the Inductively coupled plasma mass spectrometry (ICP-MS) performed in single particle mode (spICP-MS). However, the use of the technique in the detection and measurement of nanoparticles requires that the equipment be operated differently than when used to analyze dissolved metal solutions. Thus, this article presents the use of the spICP-MS technique for the measurement and quantification of nanoparticles, as well as their validation. The method proved to be adequate for the purpose, presenting a satisfactory result for the selectivity test and recovery of 83.7 (40 nm) and 77.6% (80 nm). The detection limits, determined for the most frequent size, dissolved silver concentration and particle concentration, were 17.5 nm, 0.736 ng mL-1;, 146 particles mL-1;, respectively. Thus, the results obtained indicated the possibility of using the technique to determine the size and concentration of silver nanoparticles.

Abstract: Currently, the research of new natural compounds with biological potential demonstrates great ethnopharmacological importance. In this study, we evaluated the biological properties promoted by saline extract from Malpighia emarginata DC leaves, whose objective is to evaluate the antioxidant, antimicrobial and cytotoxicity potential. Phytochemical characterization was performed by UPLC-MS chromatography to identify the chemical compounds. For the antioxidant potential, DPPH, ATT and FRAP methods were used. The antibacterial and antifungal tests were performed evaluating the MIC50, MIC90, CMB and CMF parameters. Moreover, antibiofilm action was evaluated. Cytotoxicity and proliferation were performed using splenocytes from Balb/c mice and were evaluated by cytometry. We found a list of phenolic compounds among other bioactive compounds in the M. emarginata saline extract. In addition, higher antioxidant profile and antifungal activity against different strains of Candida spp. was promoted by the saline extract. Splenocytes showed greater cell viability (more than 90%) and showed higher proliferate index in 24 and 48 hours of incubation with the extract. Saline extract from Malpighia emarginata DC has potential action like antioxidant and antifungal agent without promote animal cell damage.

ABSTRACT Leishmaniasis and trypanosomiasis are globally widespread parasitic diseases which have been responsible for high mortality rates. Since drugs available for their treatment are highly hepatotoxic, nephrotoxic and cardiotoxic, adherence to therapy has been affected. Thus, the search for new, more effective and safer drugs for the treatment of these diseases is necessary. Natural products have stood out as an alternative to searching for new bioactive molecules with therapeutic potential. In this study, the chemical composition and antiparasitic activity of the essential oil from Protium ovatum leaves against trypomastigote forms of Trypanosoma cruzi and the promastigote forms of Leishmania amazonensis were evaluated. The essential oil was promising against trypomastigote forms of T. cruzi (IC50= 28.55 μg.mL-1) and L. amazonensis promastigotes (IC50 = 2.28 μg.mL-1). Eighteen chemical constituents were identified by Gas Chromatography coupled to Mass Spectrometry (GC-MS) in the essential oil, whose major constituents were spathulenol (17.6 %), caryophyllene oxide (16.4 %), β-caryophyllene (14.0 %) and myrcene (8.4 %). In addition, the essential oil from P. ovatum leaves had moderate cytotoxicity against LLCMK2 adherent epithelial cell at the concentration range under analysis (CC50 = 150.9 μg.mL-1). It should be highlighted that this is the first report of the chemical composition and anti-Trypanosoma cruzi and anti-Leishmania amazonensis activities of the essential oil from Protium ovatum leaves.

Resumo Objetivo: Verificar se lactentes com alergia à proteína do leite de vaca (APLV) apresentam níveis inadequados de vitamina D. Métodos: Estudo transversal, envolveu 120 crianças de até dois anos, um grupo com APLV e outro de comparação, captadas dos ambulatórios de Gastroenterologia Pediátrica, Alergologia Pediátrica e Puericultura de um hospital universitário, no Nordeste brasileiro. Foi aplicado um formulário e foram coletadas amostras sanguíneas para a análise da vitamina D, foram considerados inadequados os níveis < 30 ng/mL. Níveis de vitamina D foram expressos em média e desvio padrão e a frequência dos graus de suficiência e demais variáveis, em proporções. Resultados: Lactentes com APLV, quando comparados com os saudáveis, apresentaram uma menor média do nível da vitamina D (30,93 vs. 35,29 ng/mL) (p = 0,041) e maior frequência de deficiência (20,3% vs. 8,2) (p = 0,049). Maior frequência de níveis inadequados de vitamina D foi observada nas crianças com APLV que estavam em aleitamento materno exclusivo/predominante (p = 0,002). Independentemente do período de exposição solar, a frequência de um status inadequado de vitamina D foi semelhante entre os grupos (p = 0,972). Conclusões: Menores níveis de vitamina D foram observados em lactentes com APLV, especialmente naqueles em aleitamento materno exclusivo/predominante, que configura esse como um possível grupo de risco para essa deficiência.
Abstract Objective: To verify whether infants with cow's milk protein allergy have inadequate vitamin D levels. Methods: This cross-sectional study included 120 children aged 2 years or younger, one group with cow's milk protein allergy and a control group. The children were recruited at the pediatric gastroenterology, allergology, and pediatric outpatient clinics of a university hospital in the Northeast of Brazil. A questionnaire was administered to the caregiver and blood samples were collected for vitamin D quantification. Vitamin D levels <30 ng/mL were considered inadequate. Vitamin D level was expressed as mean and standard deviation, and the frequency of the degrees of sufficiency and other variables, as proportions. Results: Infants with cow's milk protein allergy had lower mean vitamin D levels (30.93 vs.35.29 ng/mL; p = 0.041) and higher deficiency frequency (20.3% vs.8.2; p = 0.049) than the healthy controls. Exclusively or predominantly breastfed infants with cow's milk protein allergy had higher frequency of inadequate vitamin D levels (p = 0.002). Regardless of sun exposure time, the groups had similar frequencies of inadequate vitamin D levels (p = 0.972). Conclusions: Lower vitamin D levels were found in infants with CMPA, especially those who were exclusively or predominantly breastfed, making these infants a possible risk group for vitamin D deficiency.

Isotermas de dessorção de pimenta-de-macaco foram determinadas pelo método gravimétrico estático nas temperaturas de 35, 45 e 55 ºC, com umidade relativa variando de 5,5-81%. Três modelos matemáticos foram aplicados para analisar os dados experimentais. O modelo de GAB modificado apresentou o melhor ajuste aos dados experimentais. O calor isostérico e a entropia diferencial foram determinados pela aplicação das equações de Clausius-Clapeyron e Gibbs-Helmholtz, respectivamente. O calor isostérico e a entropia da isoterma de dessorção apresentaram comportamento similar. A teoria da compensação entalpia-entropia foi aplicada às isotermas indicando que o mecanismo de dessorção de umidade das partes aéreas de pimenta-de-macaco é controlado pela entalpia.
Desorption isotherms of monkey pepper were determined by static gravimetric method at temperatures 35, 45, and 55 ºC, over a relative humidity range of 5.5-81%. Three mathematical models were applied to analyse the experimental data. The modified GAB model showed the best adjustment of the experimental data. The isosteric heat and differential entropy were determined by applying Clausius-Clapeyron and Gibbs-Helmholtz equations, respectively. The isosteric heat and the entropy of desorption isotherm presented similar behavior. The enthalpy-entropy compensation theory was applied to the isotherms, indicating that the aerial parts desorption mechanism are enthalpy-controlled.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Nesse estudo um planejamento experimental de misturas de biodieseis é utilizado para avaliar e otimizar a estabilidade oxidativa de amostras de biodiesel de soja contendo diferentes teores de biodiesel de sebo e de mamona. As misturas de biodieseis foram preparadas com teores que variaram entre 80-86% (v/v) para a soja, 5-15% (v/v) para o sebo e 1-10% (v/v) para a mamona. O período de indução da amostra de biodiesel de soja puro serviu de base para a comparação com a estabilidade das misturas. A estratégia de planejamento experimental de misturas mostrou-se adequada para a formulação de misturas de biodiesel com ótima estabilidade oxidativa. O modelo quadrático obtido na etapa de otimização ajustou-se adequadamente aos dados experimentais, sendo capaz de prever as proporções ótimas para misturas com maior estabilidade oxidativa. O melhor período de indução foi obtido com as seguintes proporções: 82,5% (v/v) de soja, 7,5% (v/v) de sebo e 10,0% (v/v) de mamona.
This study proposes an experimental design of biodiesel constrained mixtures to evaluate and optimize the oxidative stability of soybean biodiesel mixed with different amounts of tallow and castor bean biodiesels. To prepare the biodiesel blends, the proportions of each type of biodiesel were varied, producing mixtures that contained 80-86% (v/v) soybean, 5-15% (v/v) tallow, and 1-10% (v/v) castor bean biodiesel. The induction period of pure soybean biodiesel served as the baseline for comparing the stability of the mixtures. The experimental design strategy was capable of producing the best oxidative stability for biodiesel mixtures. The quadratic model obtained in the optimization stage fits the experimental data well. The higher induction period occurred at the following composition: 82.5% (v/v) soybean, 7.5% (v/v) tallow and 10.0% (v/v) castor bean.

Uma nova espécie de Culicoides do subgênero Mataemyia Vargas, Culicoides huaynacapaci Felippe-Bauer, é descrita e ilustrada baseada em exemplares fêmeas e machos coletados picando humanos no Estado de Cajamarca, na Amazônia Peruana. A nova espécie é comparada com a espécie afim, C. albuquerquei Wirth & Blanton.
A new species of Culicoides of the subgenus Mataemyia Vargas, Culicoides huaynacapaci Felippe-Bauer, is described and illustrated based on female and male specimens collected biting humans in Department of Cajamarca, in Peruvian Amazonia. The new species is compared with its similar congener C. albuquerquei Wirth & Blanton.

OBJETIVO: Avaliar o pico de pressão inspiratória e a freqüência ventilatória obtidos por médicos quando da utilização de balão auto-inflável neonatal em modelo de pulmão. MÉTODOS: Quinze médicos ventilaram simuladores de pulmão de recém-nascidos a termo e pré-termo, sendo os desfechos captados através de um monitor de ventilação. RESULTADOS: As medianas dos picos de pressão foram 23 (intervalo interquartil, 15-47) e 26 (intervalo interquartil, 14-51) cmH2O, sendo menor que 20 em 41,2 e 35,8% das vezes; maior que 40 em 29,7 e 33,6% das vezes e entre 27 e 33 cmH2O em 8,2 e 6,5% das vezes, para o pulmão de termo e de pré-termo, respectivamente. As medianas das freqüências ventilatórias foram de 45 (intervalo interquartil, 36-57) e 48 (intervalo interquartil, 39-55,5) ciclos por minuto, sendo menor que 30 em 9,3 e 6,7% das vezes e maior que 60 em 12 e 13,3% das vezes, respectivamente, para o pulmão de termo e de pré-termo. As diferenças nas medianas não foram estatisticamente significantes. CONCLUSÃO: A ventilação utilizando o balão auto-inflável promoveu adequação para a freqüência ventilatória em aproximadamente 80% das vezes, e os médicos não conseguiram promover ventilação com mínima variabilidade na pressão, alcançando níveis diferentes daqueles definidos no curso de reanimação neonatal em 70% das vezes. Essa situação independeu da ventilação do pulmão-teste análogo ao sistema respiratório de recém-nascidos a termo ou pré-termo.
OBJECTIVE: To evaluate the peak inspiratory pressure and ventilation rate achieved by physicians when using a neonatal self-inflating bag on a lung model. METHODS: Fifteen physicians ventilated full term and preterm infant lung simulators while the outcomes were captured by a ventilation monitor. RESULTS: Median peak pressures in cmH2O for full term and preterm lungs were 23 (interquartile range: 15-47) and 26 (interquartile range: 14-51), being less than 20 in 41.2 and 35.8% of the pressure curves analyzed, more than 40 in 29.7 and 33.6%, and between 27 and 33 cmH2O in 8.2 and 6.5% of the curves, respectively. Median ventilation rates were 45 (interquartile range: 36-57) and 48 (interquartile range: 39-55.5) cycles per minute, being more than 30 in 9.3 and 6.7% of pressure curves and more than 60 in 12 and 13.3% of pressure curves, for the full term and preterm lungs, respectively. The differences between these medians were not statistically significant. CONCLUSIONS: Ventilation rates achieved with the self-inflating bag were adequate in approximately 80% of pressure curves analyzed, but the physicians were unable to provide ventilation with minimal pressure variation, producing pressures that diverged from those defined by the neonatal resuscitation training course in 70% of the curves. This was irrespective of whether they were ventilating the lung model analogous to preterm or full term infant lungs.

A análise da presença de auto-anticorpos feita por imunofluorescência indireta em células HEp-2 constitui-se em um método de triagem escolhido na maioria dos laboratórios clínicos. A ausência de uma nomenclatura definida para a descrição dos laudos tem trazido problemas na utilização clínica do teste, pelas dificuldades no controle de qualidade e na padronização dos resultados, que, por sua vez, embora similares, recebiam denominações diferentes. O I Consenso Brasileiro para Padronização dos Laudos de FAN HEp-2 reuniu em agosto de 2000, em Goiânia, diversos especialistas de todo o Brasil. Esses emitiram pareceres em consenso para os distintos padrões: nucleares, nucleolares, citoplasmáticos e aparelho mitótico. Foram feitas recomendações sobre os critérios para a leitura de uma lâmina, bem como para relação entre a diluição de triagem e o sistema óptico utilizado.
The technique of immunofluorescence using HEp-2 cells as substrate is the screening method of choice for the presence of autoantibodies in many clinical laboratories. The lack of a specific terminology for reporting results brings problems in quality control, clinical utility of the test, and standardization attempts. The first Brazilian Consensus for Standardization of ANA in HEp-2 Cells took place in Goiânia in August 2000. Several laboratory specialists with experience in the methodology showed up. They established guidelines for the description of ANA patterns in the Portuguese language, encompassing distinct descriptions for nuclear, nucleolar, cytoplasmic and mitotic apparatus patterns of fluorescence. Recommendations were also established regarding screening titers, final dilution titer, and on morphological criteria for reading the slides.

Starch levels in refrigerated and frozen chicken based meat products

Os teores de amido foram determinados em produtos a base de frango comercializados em Belo Horizonte, MG, no período de dezembro de 1996 a março de 1997. As amostras foram analisadas quanto aos teores de umidade e amido. O amido foi submetido a hidrolise ácida e a glicose resultante foi determinada a 620 nm após reação com antrona. O método apresentou exatidão (92,7% de recuperação), precisão (CV = 3,0%), sensibilidade (limite de quantificação = 1,25 g/100 g) e simplicidade na determinação de amido em produtos cárneos. Dentre os produtos analisados, amido foi detectado em 100% das amostras de almôndega e empanado, 60% das de lingüiça, 50% das de mortadela e 30% das de salsicha. Amido não foi detectado nas amostras de hambúrguer analisadas. Teores médios mais elevados de amido foram encontrados em empanado (14,85 g/100 g) seguido de almôndegas (4,45 g/100 g), lingüiça (1,73 g/100 g), mortadela (1,14 g/100 g) e salsicha (0,57 g/100 g). O teor médio de umidade variou de 35,68 em lingüiça a 46,24 g/100 g em empanado. Todas as amostras de mortadela e 90% das de salsicha atenderam a legislação vigente quanto aos teores de amido. Foi observada variação nos teores de amido entre produtos de diferentes marcas e de uma mesma marca.
The levels of starch were determined in chicken based meat products commercialized in Belo Horizonte, MG, Brazil, from December 1996 to March 1997. Samples were analyzed for moisture and starch contents. Starch was acid hydrolyzed and the resulting glucose was determined at 620 nm after reaction with anthrone. The method was observed to be accurate (92.7% recovery), precise (CV = 3.0%), sensitive (quantification limit = 1.25 g/100 g) and simple in the determination of starch in meat products. Among products analyzed, starch was detected in 100% of meat balls and nuggets samples, in 60% of the sausage, 50% of the bologna and 30% of the frankfurter. Starch was not detected in hamburger samples analyzed. Higher mean starch levels were found in nuggets (14.85 g/100 g) followed by meat balls (4.45 g/100 g), sausage (1.73 g/100 g), bologna (1.14 g/100 g) and frankfurter (0.57 g/100 g). Mean moisture content varied from 35.68 in sausage to 46.24 g/100 g in nuggets. No significant correlation was observed between moisture and starch contents. Every sample of bologna and 90% of the frankfurter contained starch levels according to the Brazilian legislation. Starch levels varied considerably within brands as well as within lots of the same brand.
