Abstract This study aimed to use the petiole residues of miriti to partially replace pine wood and evaluate the effects of this incorporation on the physical and mechanical properties of the particleboards. Panels were produced with 0, 25, 50, and 75% of miriti in partial replacement of wood and 12% of urea-formaldehyde adhesive. The chemical-physical properties of miriti residues showed potential particleboard production as they were similar to those of pinewood. For the physical properties, the panels were classified as low density. There was an increase in the compression ratio and, consequently, in the hygroscopicity . For the mechanical properties, it was observed that the modulus of elasticity (MOE) and modulus of rupture (MOR) did not show any statistical differences. For perpendicular traction (PT), there was a significant reduction in 50%. Considering a general analysis of the physical and mechanical properties, up to 40% of mirti can be substituted, as in this situation they meet the properties mentioned in CS-236-66, except for PT for commercialization of the panels.

Resumo O objetivo deste trabalho foi utilizar os resíduos do pecíolo do miriti em substituição parcial da madeira de pinus e avaliar os efeitos dessa incorporação nas propriedades físicas e mecânicas dos painéis aglomerados. Os painéis foram produzidos com 0, 25, 50 e 75% de miriti em substituição parcial de madeira e 12% de adesivo uréia-formaldeído. As propriedades físico-químicas dos resíduos de miriti mostraram potencial de produção de aglomerados, uma vez que foram semelhantes às do pinus. Quanto às propriedades físicas, os painéis foram classificados como de baixa densidade. Houve aumento na taxa de compressão e, consequentemente, na higroscopicidade. Para as propriedades mecânicas, observou-se que o módulo de elasticidade (MOE) e o módulo de ruptura (MOR) não houve diferença estatística. Para tração perpendicular (TP) houve redução significativa de 50%. Considerando uma análise geral das propriedades físicas e mecânicas, é possível uma substituição em até 40% de miriti, pois nessa situação atendem as propriedades mencionadas na CS-236-66, com exceção da TP para a comercialização dos painéis.

ABSTRACT Objective: This randomized crossover trial evaluated periodontal indexes of two types of 3 x 3 retainers (a modified 0.032-in SS V-loop retainer and a conventional 0.0215-in SS coaxial wire retainer) after bonded for six months. Also, bonded failure rate, and a questionnaire about comfort, ease of cleaning and overall preference were recorded. Material and Methods: 15 patients were enrolled in this study who used both retainers for six months each, having a 15-day wash-out interval between each bonded retainer usage. The following periodontal index were recorded: Plaque Index (PI), Calculus Index (CI) and Gingival Index (GI). Patients answered a questionnaire to assess comfort, ease of cleaning and overall retainer-type preference. Rate of bonding failure was also evaluated. Results: V-Loop retainer showed higher PI (P<0.05) as compared to conventional 0.0215-in coaxial wire retainer. However, CI and GI presented no statistically significant differences between both types of retainers. The conventional 0.0215-in coaxial wire retainer was chosen as the most comfortable (p<0.05), although no statistically significant differences were found for all other questionnaire answers. Bonding failure events were more observed in the 3x3 V-Loop retainer (p<0.002), as compared to the conventional 0.0215-in coaxial retainer. Conclusion: V-Loop retainer showed higher PI (p<0.05), higher bonding failure rate and less comfortable, as compared to conventional 0.0215-in coaxial wire.

RESUMO Objetivo: Este estudo cruzado e randomizado avaliou os índices periodontais de dois tipos de contenções 3x3 (uma contenção aço V-Loop de 0,032” modificada e uma contenção convencional de fio coaxial aço de 0,0215”) após colagem, por seis meses. Além disso, foram registradas a taxa de falha na colagem e um questionário sobre conforto, facilidade de limpeza e preferência geral. Material e Métodos: Foram incluídos nesse estudo 15 pacientes que usaram ambas as contenções por seis meses cada, com intervalo de quinze dias entre cada contenção fixa. Foram registrados os seguintes índices periodontais: Índice de Placa (IP), Índice de Cálculo (IC) e Índice Gengival (IG). Os pacientes responderam a um questionário para avaliar o conforto, a facilidade de limpeza e a preferência geral pelo tipo de contenção. A taxa de falha de colagem também foi avaliada. Resultados: A contenção V-Loop apresentou maior IP (p<0,05) em comparação ao fio coaxial convencional. Entretanto, IC e IG não apresentaram diferenças estatisticamente significativas entre as contenções testadas. A contenção convencional de fio coaxial 0,0215” foi escolhida como a mais confortável (p<0,05), embora não tenham sido encontradas diferenças estatisticamente significativas para todas as outras respostas do questionário. Eventos de falha de colagem foram mais observados na contenção V-Loop 3 x 3 (p<0,002) em comparação com a contenção coaxial convencional de 0,0215”. Conclusão: A contenção V-Loop apresentou maior IP (p<0,05), maior taxa de falha de colagem e foi menos confortável em comparação ao fio coaxial convencional 0,0215”.

Abstract The aim of this study is to report an original case series of synchronous jawbone diseases. Data of patients seen over 13 years were extracted from the files of three Oral Radiology and Pathology diagnostic centers in Brazil. The clinical, radiographic, and laboratory characteristics were tabulated and analyzed by the authors; the patients were described according to lesion type. Seventy-two synchronous jawbone diseases were included in this study. Florid osseous dysplasia, Gorlin-Goltz syndrome, and cherubism were the most frequent disorders reported in this case series. In addition, the posterior mandible area was the main site of manifestation. Florid osseous dysplasia and Gorlin-Goltz syndrome represented two-thirds of our samples. With the utilization of adequate demographic, clinical, and radiologic information, it is possible to diagnose most of the synchronous lesions of jawbones. Sometimes, however, we need complementary exams, such as histopathologic and biochemical analysis or dosing of calcium, phosphorus, and alkaline phosphatase.

ABSTRACT Low-temperature storage is one of the most effective techniques to maintain the quality and durability of cut flowers. Both quality maintenance and durability are dependent on plant metabolism factors such as antioxidant system. The objective was to evaluate the effect of low-temperature on precooling and storage after harvest, on the metabolism of calla lily [Zantedeschia aethiopica (L.) K. Spreng] concerning the antioxidant system, levels of hydrogen peroxide (H2O2), and malondialdehyde (MDA). Therefore, different storage temperatures (4 ºC and 21 ºC) were tested as well as the effect of precooling in different periods (0 to 24 hours). It was observed that the activity of catalase (CAT) and superoxide dismutase (SOD) was increased at 4 °C, as well as inflorescence durability in the commercial standard (around 12 days). At 21 °C, the content of H2O2 and MDA was higher and the visual quality reduction in the inflorescence was accelerated. Low-temperature leads to a higher activity of the enzymes of the antioxidant system (CAT and SOD) and increases postharvest longevity for calla lily. Precooling did not influence the visual quality and longevity of calla lily. Long-term exposure periods at low temperatures, with 12 and 24 hours of precooling caused stress, evidenced higher MDA levels and higher CAT activity. SOD activity was related to low temperature and shorter precooling periods resulted in better antioxidant system activity and lower MDA levels.

RESUMO O armazenamento em baixa temperatura é uma das técnicas mais eficazes para manter a qualidade e a durabilidade das flores cortadas. Tanto a manutenção da qualidade quanto a durabilidade são dependentes de fatores do metabolismo da planta como o sistema antioxidante. O objetivo foi avaliar o efeito da baixa temperatura no pré-resfriamento e armazenamento pós-colheita, no metabolismo do copo-de-leite [Zantedeschia aethiopica (L.) K. Spreng] quanto ao sistema antioxidante, níveis de peróxido de hidrogênio (H2O2) e malondialdeído (MDA). Portanto, diferentes temperaturas de armazenamento (4 ºC e 21 ºC) foram testadas assim como o efeito do pré-resfriamento em diferentes períodos (0 a 24 horas). Foi observado que a atividade da catalase (CAT) e da superóxido dismutase (SOD) aumentou no armazenamento à 4 °C, assim como a durabilidade da inflorescência no padrão comercial (aproximadamente 12 dias). À 21 °C, o teor de H2O2 e MDA foi maior e a redução da qualidade visual na inflorescência foi acelerada. A baixa temperatura leva a uma maior atividade das enzimas do sistema antioxidante (CAT e SOD) e aumenta a longevidade pós-colheita do copo-de-leite. O pré-resfriamento não influenciou na qualidade visual e longevidade do copo-de-leite. Períodos de exposição de longa duração a baixas temperaturas, com 12 e 24 horas de pré-resfriamento causaram estresse, evidenciados por maiores níveis de MDA e maior atividade de CAT. A atividade da SOD foi relacionada à baixa temperatura e períodos mais curtos de pré-resfriamento resultaram em melhor atividade do sistema antioxidante e menores níveis de MDA.

ABSTRACT Genipa americana L. is a forest species of high socioeconomic potential. However, predatory extractivism actions threaten its existence, making it necessary to adopt conservationist practices. G. americana seeds show sensitivity to desiccation and cooling, making it unfeasible for conservation by conventional methods. Thus, cryopreservation is an promising alternative for the long-term conservation of species that produce unorthodox seeds, such as the genipap. In this sense, the objective was to cryopreservation of seeds of G. americana and to evaluate the effects of desiccation and freezing on germination and establishment of seedlings. Initially, seeds were dehydrated in silica gel 0, 16, 18, 20, 22 and 24 h, and then were cryopreserved in liquid nitrogen (LN) (-196 °C) during 24 h. After thawing, the viability and germination were analyzed. Dehydrated and non-cryopreserved seeds were also analyzed. The silica gel desiccation caused a reduction in viability and germination of the seeds of G. americana. The initial seed water content was so high (47%) that storage in LN (+LN) without prior dehydration treatment resulted in seed mortality. It was verified that, the dehydration in silica gel for the minimum time 20 h (corresponding to 14% water content) provides greater freezing tolerance, allowing the successful cryopreservation. Silica gel dehydration followed by immersion in LN was shown to be highly efficient for cryopreservation of seeds of G. americana, besides germinatio after thawing, high survival rates (100%) were obtained, with growth and normal establishment of the seedlings after acclimatization.

Resumo Strelitzia ou ave-do-paraíso, é uma importante planta ornamental tropical de difícil propagação devido aos problemas de dormência das sementes e ao reduzido número de mudas obtidas nas divisões das plantas. Com o maior conhecimento de sua estrutura, é possível desenvolver estratégias para melhorar o processo de propagação dessa espécie, uma vez que as informações sobre estes aspectos são limitadas. Assim, o objetivo foi caracterizar sementes e embriões de Strelitzia reginae. A caracterização das sementes foi realizada por meio de análise de imagens em equipamento GroundEye, Raio-X e análises anatômicas e bioquímicas. Adicionalmente, foram obtidas as seguintes características biométricas das sementes: área de 0,33 cm2, diâmetro máximo de 0,736 cm, diâmetro lateral máximo de 0,59 cm, diâmetro mínimo de 0,58 cm e perímetro de 2,47 cm. As sementes de S. reginae podem ser consideradas amiláceas por conterem 15% de amido. Pela análise anatômica do tegumento, observou-se a presença de uma exotesta (ex) e uma camada parenquimática posterior que foi dividida em mesotesta (m) e endotesta (en). A caracterização de sementes e embriões de estrelícia apresentou observações relevantes para a taxonomia e fisiologia desta espécie. As sementes são aleuro-amiláceas e apresentam área média de 0,33 cm2, com diâmetro lateral máximo de 0,59 cm. Como resultado de estudos anatômicos, foi possível identificar o tegumento responsável pela dormência.

Abstract Strelitzia or bird-of-paradise, is an important tropical ornamental plant that is difficult to propagate because of seed dormancy issues and the low number of seedlings obtained from plant divisions. With greater knowledge of its structure, it is possible to develop strategies to improve the propagation process for this species, since information on this subject is currently limited. Thus, the objective was to carry out an anatomical, morphological and biochemical characterization of Strelitzia reginae seeds and embryos. The characterization of the seeds was performed through image analysis using GroundEye equipment, X-rays, and anatomical and biochemical analyses. Additionally, the following biometric characteristics of the seeds were obtained: an area of 0.33 cm2, a maximum diameter of 0.736 cm, a maximum lateral diameter of 0.59 cm, a minimum diameter of 0.58 cm, and a perimeter of 2.47 cm. The seeds of S. reginae can be considered starchy since they contain 15% starch. By integument anatomical analysis, we observed the presence of an exotesta (ex) and a posterior parenchymatic layer that was divided into a mesotest (m) and endotest (en). The characterization of Strelitzia reginae seeds and embryos showed relevant observations for the taxonomy and physiology of this species. The seeds are aleurostarches and present an area of 0.33 cm2 on average, with maximum lateral diameter of 0.59 cm. As result of anatomical studies, it was possible to determine coat responsible for integument dormancy.

SUMMARY: Adverse events (AE) contribute significantly to postoperative morbidities and comorbidities. Many AEs occur due to a lack of anatomical knowledge and its variants. Latrogenic bile duct injuries, for instance, represent a serious surgical complication of laparoscopic cholecystectomy. Anatomical knowledge for the identification and adequate drainage of all ducts is relevant and fundamental in order to avoid future errors. The objective of the study was to morphometrically analyze the bile ducts in adult human corpses. 13 livers were extracted from adult human corpses to obtain the ducts: choledochal, common hepatic and cystic. After morphological analysis, duct measurements (length and diameter) were continued using a digital caliper. The data obtained were tabulated in SPSS 21 program, performing descriptive analysis with mean and standard deviation. The averages of bile ducts were 61.05 (± 16.43) mm in length and 3.86 (± 0.72) mm in diameter. The cystic duct length and diameter averages were 33.59 (± 12.29) mm and 3.40 (± 0.79) mm, respectively. The common hepatic ducts had an average of 30.02 (± 7.19) mm in length and 3.74 (± 1.18) mm in diameter. The analyzed samples presented different values ?? from those already described in the literature, where the length of the cystic ducts was greater, while the length of the common hepatic ducts was numerically smaller. This work is very significant, as the morphometric variability of the bile ducts allows for varying morphological situations that can compromise the hepatobiliar physiology.

RESUMEN: Los eventos adversos (EA) contribuyen significativamente a las morbilidades y comorbilidades postoperatorias. Muchos EA se deben a la falta de conocimiento de la anatomía y sus variaciones. Por ejemplo, las lesiones iatrogénicas de las vías biliares representan una complicación quirúrgica grave de la colecistectomía laparoscópica. El conocimiento anatómico para la identificación y drenaje adecuado de todos los conductos es relevante y fundamental para evitar futuros errores. El objetivo del estudio fue analizar morfométricamente las vías biliares en cadáveres humanos adultos. Se extrajeron 13 hígados de cadáveres humanos adultos y se retiraron los conductos: colédoco, hepático común y cístico. Después del análisis morfológico, se continuó con las mediciones de los conductos (longitud y diámetro) utilizando un calibrador digital. Los datos fueron tabulados en el programa SPSS 21, mediante análisis descriptivos con media y desviación estándar. Los promedios de las vías biliares fueron de 61,05 (± 16,43) mm de longitud y 3,86 (± 0,72) mm de diámetro. Los promedios de longitud y diámetro del conducto cístico fueron 33,59 (± 12,29) mm y 3,40 (± 0,79) mm, respectivamente. Los conductos hepáticos comunes tenían un promedio de 30,02 (± 7,19) mm de longitud y 3,74 (± 1,18) mm de diámetro. Las muestras analizadas presentaron valores diferentes a los ya descritos en la literatura, donde la longitud de los conductos císticos era mayor, mientras que la longitud de los conductos hepáticos comunes fue numéricamente menor. Este trabajo es significativo, debido a que la variabilidad morfométrica de las vías biliares y permite identificar situaciones morfológicas que pueden comprometer la fisiología hapatobiliar.

Resumo O processo de introdução de espécies exóticas é reconhecido como uma das maiores ameaças à ecologia e economia do planeta, devido às mudanças nas interações em comunidades biológicas nativas. Nesse sentido, relatar as bioinvasões é o primeiro passo para entender seu processo de implantação e criar estratégias para mitigar possíveis danos socioambientais. Aqui relatamos os primeiros registros de P. unifilis introduzidos no Pantanal Norte, Mato Grosso, Brasil. Obtivemos os primeiros registros das espécies ocasionalmente durante 2014 e 2015. Nos anos seguintes, de 2016 a 2019, realizamos observações sistemáticas por meio de incursões náuticas no rio Paraguai, entre a foz do rio Sepotuba e a Estação Ecológica Taiamã (TES). Incluímos também registros de terceiros, quando foi possível comprovar os locais de identificação e registro das espécies. Adultos de Podcnemis unifilis, filhotes e ninhos foram registrados em diferentes pontos ao longo do rio Paraguai. Nossos resultados indicam que P. unifilis foi introduzido próximo ao núcleo urbano de Cáceres há cerca de 30 anos. Atualmente, ela tem uma população estabelecida, registrando ninhos e filhotes por anos consecutivos. Uma em particular é a sua ocorrência no TES, importante viveiro natural de peixes e refúgio da fauna pantaneira. Assim, sugere-se o monitoramento contínuo de P.unifilis, uma vez que o ambiente do Pantanal com diversos ambientes aquáticos distintos, oferece condições favoráveis para o aumento da população da espécie no Pantanal Norte.

Abstract The process of exotic species introduction is recognized as one of the greatest threats to the ecology and economy of the planet, due to changes in interactions in native biological communities. In this sense, reporting bioinvasions is the first step to understanding its introduction process and creating strategies to mitigate possible socio-environmental damages. Here we report the first records of Podocnemis unifilis introduced in the Northern Pantanal, Mato Grosso, Brazil. We obtained the species’ first records occasionally during 2014 and 2015. In the following years from 2016 to 2019, we carried out systematic observations through nautical incursions on the Paraguay River, between the Sepotuba River mouth and the Taiamã Ecological Station (TES). We also included third part records, when it was possible to prove the species identification and registration sites. Podocnemis unifilis adults, hatchling and nests were recorded at different points along the Paraguay River. Our results indicate that P. unifilis was introduced close to the urban nucleus of Cáceres about 30 years ago. Currently, it has an established population, recording nests and hatchling for years in a row. A particular one is its occurrence at the TES, an important natural fish nursery and refuge for the wetland fauna. Thus, continuous monitoring is suggested from P.unifilis, since the Pantanal’s environment with several different aquatic environments, offers favorable conditions for the species population increase in the Northern Pantanal.

RESUMO O presente trabalho tem como objetivo analisar o efeito de diferentes espécies de madeiras e de bambu (Pinus taeda, Cereus jamacaru e Bambusa vulgaris) na hidratação de matrizes cimentícias. Para tanto, os materiais lignocelulósicos foram moídos e caracterizados antes e depois de tratamento em água quente. Posteriormente, o efeito da adição de diferentes níveis (5, 10 e 33,3%) de madeiras, pré-tratadas ou não, em pastas de cimento foram analisados a partir dos ensaios de calorimetria isotérmica e termogravimetria. Os resultados demonstraram que as partículas de cada espécie lignocelulósica apresentaram um aumento das propriedades de umidade e absorção concomitante a diminuição de suas densidades e níveis de extrativos a partir do pré-tratamento aplicado. Os outros componentes químicos destes elementos, majoritariamente, todavia, não foram afetados. Por outro lado, foi observado que quanto maior o teor de tais materiais lignocelulósicos, pré-tratados ou não, nas matrizes de cimento, maior o impacto na hidratação do cimento, segundo o ensaio de calorimetria. Já com relação ao resultado do ensaio de termogravimetria observou-se uma tendência geral, com ou sem pré-tratamento de partículas ou adição de água de compensação, de aumento dos produtos hidratados, fato atribuído, principalmente, a presença de água livre e gases nos materiais lignocelulósicos.

ABSTRACT The present work aims to analyze the effect of different species of wood and bamboo (Pinus taeda, Cereus jamacaru and Bambusa vulgaris) on the hydration of cementitious matrices. For this, the lignocellulosic materials were ground and characterized before and after treatment in hot water. Subsequently, the effect of adding different levels (5, 10 and 33.3%) of wood, pre-treated or not, in cement pastes were analyzed using isothermal calorimetry and thermogravimetry tests. The results showed that the particles of each lignocellulosic species showed an increase in moisture and absorption properties concomitant with a decrease in their densities and extractive levels from the pre-treatment applied. The other chemical components of these elements, however, were mostly unaffected. On the other hand, it was observed that the higher the content of such lignocellulosic materials, pre-treated or not, in the cement matrices, the greater the impact on cement hydration, according to the calorimetry test. Regarding the result of the thermogravimetry test, a general trend was observed, with or without pre-treatment of particles or addition of make-up water, of an increase in hydrated products, a fact mainly attributed to the presence of free water and gases in the lignocellulosic materials.

RESUMO As espécies tropicais podem exigir temperaturas mais altas e maiores concentração de reguladores de crescimento para o desenvolvimento in vitro. Em função dessas condições afetarem o metabolismo vegetal, o objetivo foi identificar como diferentes temperaturas na sala de crescimento e concentrações de giberelinas podem afetar os embriões no desenvolvimento in vitro de estrelícia, analisando o efeito sobre EROS e a estabilidade citogenética. Os embriões zigóticos foram cultivados em meio MS suplementado com 0, 5, 10 e 20 µM de GA3 e mantidos em sala de crescimento sob temperaturas de 25 °C, 30/25 °C e 30 °C. Após 60 dias, observou-se maior germinação dos embriões (72%) e comprimento das brotações (3,14 cm) em meio adicionado com 20 µM de GA3. Nessa concentração, houve um aumento na atividade da redutase do nitrato sem alterar a estabilidade citogenética. A temperatura influenciou apenas o comprimento da parte aérea e da raiz, que foram maiores a 25 °C. A 30 °C, a atividade de SOD e APX aumentou em comparação com o tratamento a 25 °C. Assim, a adição de 20 µM de GA3 ao meio de cultura e temperatura de 25 °C na sala de crescimento deve ser utilizada para cultura de embriões zigóticos de estrelitzia.

ABSTRACT Tropical species may require higher temperatures as well as higher growth regulator concentrations for in vitro development. Since these conditions may affect plant metabolism, the objective of this study was to identify how different temperatures and gibberellin concentrations may affect the in vitro development of strelitzia embryos, analyzing the effect on ROS and cytogenetic stability. Zygotic embryos were cultivated on MS medium supplemented with 5, 10 and 20 µM GA3 under temperatures of 25 °C, 30/25 °C and 30 °C. After 60 days, higher embryonic germination rate (72%) and shoot length of plantlets (3.14 cm) were observed on medium containing 20 µM gibberellic acid (GA3). At this concentration, there was an increase in nitrate reductase activity with no change in the cytogenetic stability. The temperature influenced only shoot and root lengths, which were highest at 25 °C. At 30 °C, superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities increased compared with those at 25 °C. Thus, the addition of 20 µM GA3 to the culture medium and a temperature of 25 °C in the growth room should be used for zygotic embryo culture of strelitzia.

Resumo A classificação precisa da qualidade é um dos requisitos do mercado de flores, que está se tornando cada vez mais sofisticado e exigente. No entanto, há uma falta de informações sobre como essa classificação é realizada, e muitas vezes é conduzida de maneira menos empírica e mais subjetiva. Portanto, este trabalho teve como objetivo comparar a eficiência da classificação pessoal (in-person) e usando imagens digitais para determinar a qualidade de flores de antúrio ‘Tropical’. Foram realizados três experimentos quanto ao registro das imagens e ao modo de exibição das imagens e comparando três métodos de classificação da qualidade pós-colheita do antúrio. Concluiu-se que as medidas dos parâmetros colorimétricos podem ser realizadas em qualquer lugar da espata, mas o período em que os parâmetros medidos são afetados pela senescência. Os avaliadores diferem na precisão com que atribuem notas e não há necessidade de conhecimento prévio para avaliar a qualidade usando imagens. O parâmetro L* medido pelo colorímetro apresentou maior correlação com a avaliação da qualidade pós-colheita de Anthurium andraeanum Lind. ‘Tropical’. A classificação por imagens pode ser realizada usando diferentes dispositivos eletrônicos, e a classificação in-person é mais rigorosa do que a classificação realizada com imagens digitais.

Abstract Accurate quality classification is one of the requirements of the flower market, which is becoming increasing more sophisticated and demanding. However, there is a lack of information regarding how such classification is performed, and it is often conducted in a less empirical and more subjective way. Therefore, this work aimed to compare the efficiency of personal classification (in-person) and using digital images to determine the quality of anthurium cv. Tropical (Anthurium andraeanum Lind.). Three experiments were performed regarding the recording of the images and the display mode of the images and comparing three methods of classifying postharvest quality of anthurium. It was concluded that measurements of colorimetric parameters can be performed anywhere on the spathe, but the period in which the parameters are measured does affect. Evaluators differ in the thoroughness with which they assign grades, and there is no need for prior knowledge to assess quality using images. The L* parameter measured by the colorimeter presented higher correlation with the evaluation of postharvest quality of Anthurium andraeanum Lind. ‘Tropical’. Classification by images can be performed using different electronic devices, and in-person classification is more rigorous than classification conducted using digital images.

RESUMO A Campomanesia rufa é uma espécie frutífera nativa do Brasil considerada como vunerável pela IUCN (International Union for Conservation of Nature) e poucas informações sobre a propagação são encontradas. Neste contexto, o objetivo do presente estudo foi desenvolver protocolos de germinação in vitro, micropropagação e calogênese para a espécie. Para germinação in vitro, as sementes foram inoculadas em meio MS suplementado com GA3 (Ácido Giberelico), para a indução da parte aérea, o meio de cultura foi suplementado com BA (benzilamina), BAP (6-Benzilamina Purina) ou TDZ (Thidiazuron). Para o crescimento da parte aérea, o meio de cultura contendo BA, BAP e TDZ foi mantido sob lâmpadas fluorescentes brancas e sob diodos emissores de luz (LED). O GA3 também foi testado para elongação de brotações in vitro. Para o controle da oxidação, o meio foi suplementado com PVP (polivinilpirrolidona) e para a indução de calos, 2,4-D (Ácido 2,4-diclorofenoxiacético) . Os resultados mostraram 68% de germinação, independentemente da concentração de GA3. No estágio de propagação, BAP a 4,5 µM induziu maior número de brotações (4,53) e lâmpadas de LED como fonte de luz combinada com o meio de cultura com 1 μM de BAP induziu maior número de brotações (4,08). A maior média de comprimento de C. rufa (31,9 mm) foi obtida usando 8,8 µM de GA3. O uso de PVP (584,3 µM) controla até 27,3% de oxidação em explantes de folhas jovens. O uso de 10 µM de 2,4-D leva a uma maior formação de calos (58,7%). Portanto, pode-se concluir que o uso de BAP é eficiente na indução de brotações, o PVP controla a oxidação de segmentos foliares e o 2,4-D induz calos em C. rufa.

ABSTRACT The Campomanesia rufa is a fruitful species native from Brazil considered as endangered by the IUC (International Union for Conservation of Nature) and low information regarding its propagation is available. In this context, the aim of the present study was to develop in vitro germination, micropropagation and callogenesis protocols for the species. For in vitro germination, seeds were inoculated in MS medium supplemented with GA3 (gibberellic acid) and for shoot induction, the medium was supplemented with three different cytokinins BA (benzyladenine), BAP (6-Benzylaminopurine) or TDZ (Thidiazuron). For shoot growth, culture medium containing BA, BAP, TDZ were maintained under white fluorescent lamps and blue:red light-emitting diodes (LED). GA3 was also tested on in vitro shoot elongation. For oxidation control, the medium was supplemented with PVP (polyvinylpyrrolidone) and for callus induction, 2.4-D (dichlorophenoxyacetic acid). The results showed 68% germination regardless of GA3 concentration. In the propagation stage, BAP at 4.5 µM induced a higher number of shoots (4.53) and LED lamps as the light source combined with the culture medium with 1 μM BAP induced a higher number of shoots (4.08). The highest average of C. rufa length (31.9 mm) was obtained using 8.8 µM GA3. The use of PVP (584.3 uM) controls up to 27.3% oxidation in young leaf explants. The use of 10 µM 2.4-D leads to a higher callus formation (58.7 %). Therefore, it can be concluded that the use of BAP is efficient in the induction of shoots, PVP controls oxidation leaf segments, and 2.4-D induces callus in C. rufa.

ABSTRACT The market of flowers and ornamental plants is dependent on the diversification of species and the availability of high quality propagation materials. Actually, in vitro culture techniques performance a prominent role in the multiplication and maintenance of commercially propagated ornamental plant species, and are promising for the production of thousands of high quality plants in relatively short term. In addition, when market demand for a particular species is low or zero in a specific period of the year, in vitro culture techniques allow the conservation of cultures under aseptic conditions, by Slow Growth Storage (SGS), from a few weeks to one year (or more), without affecting their viability and potential regrowth. This can be achieved by modifying the constitution of the culture medium and the maintenance conditions of in vitro cultures. Obviously, the success of the technique depends on greatly on the physiological characteristics of the species to be conserved, as well. Once a SGS protocol is optimized, the expenses labor, the possibility of contamination and the probability of somaclonal variation can be reduced markedly.

ABSTRACT This study aimed to impute the genetic makeup of individual fishes of Serrasalmidae family on the basis of body weight and morphometric measurements. Eighty-three juveniles, belonging to the genetic groups Pacu, Pirapitinga, Tambaqui, Tambacu, Tambatinga, Patinga, Paqui and Piraqui, were separated into 16 water tanks in a recirculation system, with two tanks per genetic group, where they remained until they reached 495 days of age. They were then weighed and analyzed according to the following morphometric parameters: Standard Length (SL), Head Length (HL), Body Height (BH), and Body Width (BW). The identity of each fish was confirmed with two SNPs and two mitochondrial markers. Two analyses were performed: one for the validating the imputation and another for imputing a genetic composition of animals considered to be advanced hybrids (post F1). In both analyses, we used linear mixed models with a mixture of normal distributions to impute the genetic makeup of the fish based on phenotype. We applied the mixed models method, whereby the environmental effects were estimated by the Empirical Best Linear Unbiased Estimator (EBLUE) and genetic effects are considered random, obtaining the Empirical Best Linear Unbiased Predictor (EBLUP) from the general (GCA) and the specific (SCA) combining ability effects. The results showed that validation of the genetic makeup imputation based on body weight can be used because of the strong correlation between the observed and imputed genotype. The fish classified as advanced hybrids had a genetic composition with a high probability of belonging to known genotypes and there was consistency in genotype imputation according to the different characteristics used.

RESUMO A conservação dos recursos genéticos vegetais é uma medida importante para prevenção do processo de erosão genética. Bancos de sementes são as formas mais comuns de conservação ex situ, entretanto, sementes de cafeeiro não podem ser armazenadas por métodos convencionais. A criopreservação é uma alternativa viável para conservação em longo prazo de espécies, como o cafeeiro, que produzem sementes intermediárias ou recalcitrantes. O objetivo deste trabalho foi criopreservar embriões zigóticos de Coffea arabica L. cv Catuaí Vermelho avaliando o efeito da desidratação antes da criopreservação, e da reidratação osmótica após o descongelamento. Antes da criopreservação foram testados e determinados os teores de umidade em diferentes tempos de desidratação (0, 15, 30, 60 e 120 min). Após a desidratação os embriões foram criopreservados por uma hora em nitrogênio líquido, e após o descongelamento foram reidratados utilizando solução osmótica. Para avaliação da viabilidade de embriões desidratados e/ou criopreservados, utilizou-se o teste com o cloreto de 2,3,5-trifenil tetrazólio. Embriões não desidratados não sobreviveram ao congelamento. Embriões desidratados até cerca de 20% de umidade que não passaram por uma reidratação osmocondicionada, não germinaram. Embriões desidratados até cerca de 20% de umidade, criopreservados e reidratados lentamente após o descongelamento, apresentaram 98% de germinação. A máxima viabilidade obtida através do teste de tetrazólio foi 75% após 60 minutos de desidratação. Portanto, embriões zigóticos de cafeeiro podem ser criopreservados utilizando desidratação física em sílica gel por 60 minutos (23% de teor de umidade), seguida de reidratação com solução osmótica após o descongelamento. Esse método permitiu a germinação de 98% dos embriões criopreservados.

ABSTRACT Conservation of plant genetic resources is important to prevent genetic erosion. Seed banks are the most common method of ex situ conservation; however, coffee seeds can not be stored by conventional methods. Cryopreservation is a viable alternative for long-term conservation of species that produce intermediate or recalcitrant seeds, as coffee. The aim of this work was to cryopreserve Coffea arabica L. cv Catuaí Vermelho IAC 144 zygotic embryos, and analyse the effects of dehydration prior cryopreservation and osmotic rehydration after thawing, in embryos germination and seedlings formation after cryopreservation. Prior to cryopreservation, different dehydration times (0, 15, 30, 60 and 120 min) were tested. Dehydrated embryos were cryopreserved in liquid nitrogen for 1 hour, and after thawing were rehydrated by osmotic solutions. Dehydrated and non-cryopreserved embryos were also analysed. The test with 2,3,5 triphenyl tetrazolium chloride (TTC) was used to evaluate the embryos viability. Non-dehydrated embryos did not survive after freezing. Embryos that were dehydrated until 20% of the moisture content did not germinate when osmotic rehydration was not performed. In contrast, cryopreserved embryos with the same moisture content presented 98% germination when they were rehydrated slowly in osmotic solution. According to tetrazolium tests, embryos presented maximum viability (75%) after dehydration for 60 minutes (23% moisture content). Therefore, coffee zygotic embryos (Coffea arabica L. cv. Catuaí Vermelho) can be successfully cryopreserved using physical dehydration in silica gel for 60 minutes (23% moisture content), followed by osmotic rehydration after thawing. This method allowed a germination of 98% of cryopreserved zygotic embryos.
