Resumo O objetivo deste estudo foi avaliar o desempenho de um sistema alagado construído (CWS) na remoção de coliformes totais (TC) e coliformes termotolerantes (ThC) de água residuária da suinocultura, como tratamento complementar a um sistema anaeróbio. Na etapa 1, o sistema experimental foi composto por um sistema alagado construído de escoamento vertical (VFCWS) cultivado com capim Tifton 85 em série com um sistema alagado construído de escoamento subsuperficial horizontal (HFCWS1) cultivado com Taboa. No HFCWS1 os tempos de detenção hidráulica foram de 4,7; 3,1 e 2,3 dias e as taxas de aplicação superficial foram 294, 319 e 397 kg ha-1 d-1 de DQO, nas fases I, II e III, respectivamente. Na etapa 2, o sistema experimental foi composto por um sistema alagado construído de escoamento subsuperficial horizontal (HFCWS2) cultivado com capim Tifton 85, os tempos de detenção hidráulica foram 6,1; 2,0 e 0,5 d e as taxas de aplicação superficial foram 850, 656 e 6,34 kg ha-1 d-1 de DQO, nas fases I, II e III, respectivamente. Na etapa 1, verificou-se que o VFCWS foi mais eficiente na remoção de coliformes quando comparado ao HFCWS1. Quando comparados apenas os HFCWS, verificou-se que a remoção de coliformes na etapa 1 variou de 1 a 2 unidades log no HFCWS1. Na etapa 2, o HFCWS2 mostrou-se mais limitado, apresentando maiores eficiências de remoção na fase I, de 1,6 a 0,8 unidades log para TC e ThC, respectivamente. Em geral, a associação resultou em eficiências que variaram de 96,4 a 99,0% para TC e de 94,2 e 97,6% para ThC, equivalente a remoção média de 1,2 a 2 unidades log, considerada satisfatória.

Abstract This study evaluated the efficiency of a constructed wetland system (CWS) in removing total coliforms (TC) and thermotolerant coliforms (ThC) of swine wastewater, as a complementary treatment to an anaerobic system. At Stage 1, the experimental system was combined using a vertical flow constructed wetland system (VFCWS) cultivated with Tifton 85 grass in series with a horizontal subsurface flow constructed wetland system (HFCWS1) cultivated with Taboa. In HFCWS1, the hydraulic detention times (HDT) were 4.7, 3.1 and 2.3 days and the surface application rates (SAR) were 294, 319 and 397 kg ha-1 d-1 of COD, in Phases I, II and III, respectively. At Stage 2, the experimental system was combined using a horizontal subsurface flow constructed wetland system (HFCWS2) cultivated with Tifton 85 grass, HDT were 6.1, 2.0 and 0.5 days and the SAR were 850, 656 and 6.34 kg ha-1 d-1 of COD, in Phases I, II and III, respectively. In Stage 1, it was verified that the VFCWS was more efficient in coliform removal when compared to HFCWS1. When only HFCWS were compared, coliform removal in Stage 1 was between 1 and 2 log units in HFCWS1. In the stage 2, the HFCWS2 was more limited, with the highest removal efficiencies during Phase I of 1.6 and 0.8 log units for TC and ThC, respectively. In general, the association resulted in efficiencies that ranged from 96.4 to 99.0% for TC, 94.2 and 97.6% for ThC, equivalent to the average removal of 1.2 to 2 log units, considered satisfactory.

RESUMO A seleção de genótipos superiores de soja é um processo complexo; dessa forma, técnicas exploratórias multivariadas podem ser aplicadas para selecionar genótipos, analisando simultaneamente todos os caracteres agronômicos em estudo, aumentando a chance de êxito em um programa de melhoramento. O objetivo do presente estudo consistiu em selecionar genótipos de soja portadores do gene RR com bons caracteres agronômicos por meio de análise multivariada e índice de seleção, e identificar quais caracteres mais influenciam, além de verificar a concordância das técnicas multivariadas e índice de seleção no processo seletivo. O experimento foi conduzido no delineamento experimental de blocos aumentados, sendo avaliados 227 genótipos de soja de geração F5, sendo 85 destes, detectados pela técnica de PCR, como resistentes ao Roundup. Os caracteres avaliados foram: número de dias para maturidade, altura da planta na maturidade, acamamento, valor agronômico, número de ramos por planta, número de vagens por planta, peso de cem sementes e produtividade de grãos. A análise de componentes principais auxiliou na seleção de dezesseis genótipos, com maior produtividade de grãos. Os caracteres relacionados aos componentes de produção exerceram grande influência sobre a produtividade de grãos. Os resultados das análises de agrupamento pelo método de K-means e pelo o método de Ward quando comparados foram semelhantes, pois agruparam os genótipos específicos para os caracteres selecionados na análise de componentes principais, em um mesmo grupo. Houve concordância dos resultados das análises multivariadas com o índice de seleção de Mulamba e Mock quanto aos genótipos selecionados. As metodologias aplicadas são eficientes no processo de seleção de genótipos.

ABSTRACT The selection of superior genotypes of soybean is a complex process, thus exploratory multivariate techniques can be applied to select genotypes analyzing the agronomic traits altogether, increasing the chance of success of a breeding program. Thus, the objective of this study was to select soybean genotypes carrying the RR gene with good agronomical performance through of multivariate analysis and selection index and identify those traits that influence, also verifying the agreement of multivariate techniques and selection index in the selection process. The experiment was conducted in an increased block experimental being evaluated 227 genotypes of F5 generation, which 85 of those were detected to be glyphosate-resistant by PCR. The following traits were evaluated: number of days to maturity, plant height at maturity, lodging, agronomic value, number of branches per plant, number of pods per plant, hundred seeds weight and grain yield. The principal components analysis resulted in the selection of sixteen genotypes with higher grain yield. The traits related to the production of components exerted great influence on grain yield. The clustering by K-means and Ward's methods were similar because they clustered the specific genotypes for the selected traits in the principal components analysis in the same group. There was an agreement on the results of the multivariate analysis in the selection index of Mulamba and Mock in relation to the selected genotypes. The methodologies applied are efficient for selecting genotypes.

RESUMO. O sucesso dos programas de melhoramento genético depende dos procedimentos de seleção e dos métodos de melhoramento adotados para a condução das populações segregantes. O objetivo deste trabalho foi avaliar a eficiência do método Bulk com seleção em F3, (BulkF3) em relação ao método Bulk, bem como comparar qual estratégia de seleção é mais eficiente em termos de ganho genético. Para a realização do trabalho foram utilizadas 20 populações segregantes, conduzidas pelos dois métodos. Foram selecionadas 60 melhores famílias de cada método, de acordo com a performance média agronômica. O delineamento utilizado foi de blocos aumentados. Foram avaliadas as seguintes características agronômicas: altura da inserção da primeira vagem e da planta na maturidade, número de ramos e de vagens por planta, peso de cem sementes e produção de grãos. Para efeito de comparação dos métodos, foram utilizadas as estimativas dos componentes genéticos, ganho genético e predição dos valores genéticos, calculados via modelos mistos (REML e BLUP). Os resultados demonstraram que as famílias obtidas pelo método BulkF3 foram as mais produtivas, com menor porte, maior número de ramos, vagens e peso de cem sementes. O método BulkF3 mostrou-se uma estratégia de condução eficiente para a seleção no melhoramento genético de soja.

ABSTRACT. The success of breeding programs depends on selection procedures and on the breeding methods adopted for selecting segregating populations. The objective of this study was to evaluate the efficiency of the Bulk method with selection in the F3 generation (BulkF3) compared to that of Bulk method as well as determine the most effective selection strategy in terms of genetic gain. Twenty segregating populations were selected by two methods. The 60 best families of each method were selected according to their average agronomic performance. An augmented block design was used. The following agronomic traits were evaluated: insertion height of first pod, plant height at maturity, number of branches and of pods per plant, 100-seed weight, and grain yield. For comparison of the methods, genetic component estimates, genetic gain and predicted breeding values were calculated using mixed models (REML and BLUP). The results showed the families obtained with the BulkF3 method were more productive, showed suitable plant height, a larger number of branches and pods, and higher 100-seed weight. The BulkF3 method was found to be an effective selection strategy for soybean improvement.

RESUMO. O amendoim é uma cultura de grande importância para o agronegócio brasileiro. Um dos principais fatores que afetam sua produção é a incidência de pragas como o tripes-do-prateamento. O objetivo deste trabalho foi avaliar o potencial de resistência ao tripes, em genótipos derivados do cruzamento entre a cultivar IAC 503 e o anfidiploide (A. magna x A. cardenasii)4x, assim como estimar os parâmetros genéticos e fenotípicos em tais genótipos, como auxiliares no processo de seleção. Os experimentos foram conduzidos no esquema de blocos aumentados com testemunhas intercalares em duas gerações (F3 e F4). A resistência ao tripes foi avaliada pela sua infestação natural e pelos sintomas de injúrias causadas pelo inseto. Foram avaliados também, caracteres agronômicos como indicadores da proximidade dos genótipos segregantes ao cultivado. Com base nos resultados, observou-se que as progênies selecionadas apresentaram maior resistência ao tripes quando comparadas aos genótipos cultivados, tendo o anfidiploide como fonte de resistência ao inseto. Algumas progênies selecionadas como resistentes, também apresentaram bons componentes de produção, porém, com grau de adequação aos genótipos A. hypogaea L. ainda pequeno, o que sugere o uso de retrocruzamentos como alternativa na introgressão de genes de resistência e consequente recuperação dos genótipos adaptados dos genitores recorrentes.

ABSTRACT. Peanut is an oilseed crop of great importance for Brazilian agribusiness. A major factor affecting its production is pest incidence, mainly thrips. This study aimed to evaluate the potential for resistance to Enneothrips flavens in genotypes derived from the cross between IAC 503 and the amphidiploid (A. magna x A. cardenasii)4x and to estimate the genetic and phenotype parameters in these genotypes, allowing for better targeting in the selection. The experiments were conducted in a Federer augmented block design with additional checks in two generations (F3 and F4). Resistance to thrips was evaluated by its natural infestation and the symptoms of attacks by the insect. They were also evaluated using agronomic trait indicators of interspecific segregating with cultivated species. The results indicated that the selected progeny exhibited high resistance to thrips compared to commercial genotypes, and they had the amphidiploid as the insect resistance source. Some progenies selected as resistant also had good production traits, but with the degree of suitability to the A. hypogaea L. genotypes still low, the use of a backcross as an alternative for the introgression of resistance genes and the consequent recovery of adapted genotypes of superior recurring parents is suggested.

Devido ao grande aumento da área cultivada com soja geneticamente modificada no Brasil, faz-se necessária a determinação de métodos que sejam rápidos e eficientes na detecção dessas cultivares. O objetivo deste trabalho foi testar a eficiência do método do palito na detecção de plantas de soja RR, bem como distinguir as cultivares quanto à sensibilidade causada pelo herbicida. Foram utilizadas dez cultivares de soja transgênica, resistentes ao ingrediente ativo glifosato, e dez cultivares de soja convencional. Palitos de dente embebidos em glifosato foram aplicados em todas as plantas em estádio V6 e as avaliações foram realizadas aos 2; 4; 6; 8 e 10 dias após a aplicação (DAA). Os efeitos da ação do glifosato nas cultivares e os sintomas de fitotoxicidade, ocasionados nas plantas transgênicas, foram avaliados por meio de escalas de notas. O teste do palito é eficiente para identificar as cultivares de soja RR e, além disso, separálas em grupos de sensibilidade aos sintomas causados pelo glifosato.

Due to the large increase in the area cultivated with genetically modified soybean in Brazil, it has become necessary to determine methods that are fast and efficient for detecting these cultivars. The aim of this work was to test the efficiency of the toothpick method in the detection of RR soybean plants, as well as to distinguish between cultivars, for sensitivity caused by herbicide. Ten transgenic soybean cultivars, resistant to the active ingredient glyphosate, and ten conventional soybean cultivars were used. Toothpicks soaked in glyphosate were applied to all the plants at stage V6 and evaluations were made at 2, 4, 6, 8 and 10 days after application (DAA). The effects of the glyphosate on the cultivars, and the symptoms of phytotoxicity caused in the transgenic plants were evaluated by means of grading scales. The toothpick test is effective in identifying RR soybean cultivars and also in separating them into groups by sensitivity to the symptoms caused by the glyphosate.

BACKGROUND: DNA methylation is commonly linked with the silencing of the gene expression for many tumor suppressor genes. As such, determining DNA methylation patterns should aid, in times to come, in the diagnosis and personal treatment for various types of cancers. Here, we analyzed the methylation pattern from five colorectal cancer patients from the Amazon state in Brazil for four tumor suppressor genes, viz.: DAPK, CDH1, CDKN2A, and TIMP2 by employing a polymerase chain reaction (PCR) specific to methylation. Efforts in the study of colorectal cancer are fundamental as it is the third most of highest incidence in the world. RESULTS: Tumor biopsies were methylated in 1/5 (20 %), 2/5 (40 %), 4/5 (80 %), and 4/5 (80 %) for CDH1, CDKN2A, DAPK, and TIMP2 genes, respectively. The margin biopsies were methylated in 3/7 (43 %), 2/7 (28 %), 7/7 (100 %), and 6/7 (86 %) for CDH1, CDKN2A, DAPK, and TIMP2, respectively. CONCLUSIONS: Our findings showed DAPK and TIMP2 to be methylated in most samples from both tumor tissues and adjacent non-neoplastic margins; thus presenting distinct methylation patterns. This emphasizes the importance of better understanding of the relation of these patterns with cancer in the context of different populations.

Through the Theory of Social Identity our objective was to know the stereotypes present in the rural imaginary related to the gypsy group, and the feelings associated with this ethnic group. Ten women, non-gypsies, residents of a rural Brazilian community, participated in this study. We analyzed the data through the ALCESTE software. The analysis of the information obtained has allowed us to identify the feeling of fear as the main mediator of practices related to the gypsies, confirming characteristics widely present in the social imaginary about this ethnic group. There are present the classic stereotypes of thieves, criminals and the cursed. It discusses the dynamics of identity provoked by the conflict between the rural community and the gypsies, emphasizing the processes that guide the field of identification and differentiation in/out-group.

Através da Teoria da Identidade Social procurou-se conhecer os estereótipos presentes no imaginário rural associados ao grupo cigano, bem como os sentimentos associados a esta etnia. Foram entrevistadas 10 mulheres não-ciganas, moradoras de uma comunidade rural brasileira. Realizou-se a organização dos dados através do software ALCESTE. A análise das informações nos permitiu identificar o sentimento de medo como importante orientador das práticas relacionadas aos ciganos, confirmando características presentes no imaginário social amplamente difundido acerca desta etnia. Estão presentes os clássicos estereótipos de ladrões, malfeitores e amaldiçoados. Discute-se a dinâmica identitária provocada pelo conflito entre a comunidade rural e os ciganos, enfatizando os processos que orientam o campo de identificação e diferenciação endo/exogrupal.
