RESUMO A fim de avaliar o efeito de diferentes doses da rbST sobre a dinâmica folicular, a produção e a maturação in vitro de oócitos, 20 vacas Sindi, divididas em três grupos, receberam um dispositivo de progesterona intravaginal, estradiol e PGF2α, além de 2mL de solução salina (grupo controle), 250 (grupo rbST 250) ou 500mg de rbST (grupo rbST 500). Cinco dias depois, realizou-se a ovum pick up, e os complexos cumulus-oócitos (CCOs) recuperados foram selecionados, classificados e maturados in vitro. Os dados de contagem foram comparados pelo procedimento glht (General Linear Hypothesis Test), e os dados em porcentagem foram submetidos ao qui-quadrado, no programa estatístico R, onde as diferenças foram consideradas significativas quando P<0,05. Não houve diferença (P>0,05) entre os grupos quanto à quantidade de folículos e à taxa de maturação. Os grupos rbST 250 e rbST 500 foram significativamente superiores (P<0,05) ao grupo controle em relação ao número de folículos grandes (0,42±0,20 vs. 0). O grupo rbST 500 apresentou maior (P<0,05) porcentagem de oócitos viáveis (91,52%) do que os grupos controle (67,85%) e rbST 250 (53,33%). A rbST aumenta o número de folículos grandes, e 500mg de rbST aumentam a porcentagem de oócitos viáveis em vacas Sindi.

ABSTRACT In order to evaluate the effect of different doses of rbST on the follicular dynamics, production, and in vitro maturation of oocytes, 20 Sindhi cows were divided into three groups, receiving an intravaginal progesterone device, estradiol and PGF2α, and 2mL of solution saline (Control Group), 250 (rbST 250 Group) or 500mg rbST (rbST 500 Group). Five days later, the ovum pick up was performed, and the cumulus-oocyte (CCO) complexes recovered were selected, classified, and matured in vitro. The counting data were compared by the glht (General Linear Hypothesis Test) procedure, and the percentage data were submitted to Qui- square, in the statistical program R, where differences were considered significant when P< 0.05. There was no difference (P> 0.05) between the groups regarding follicle quantity and maturation rate. The rbST 250 and rbST 500 groups were significantly higher (P< 0.05) than the Control group in relation to the number of large follicles (0.42±0.20 versus 0). The rbST 500 group presented higher (P< 0.05) percentage of viable oocytes (91.52%) than the Control (67.85%) and rbST 250 (53.33%) groups. rbST increases the number of large follicles and 500mg rbST increases the percentage of viable oocytes in Sindhi cows.

The behavior of cows at different stocking rates (2, 4 and 6 UA), in rotational grazing was studied. The experimental design was randomized blocks in an split plot arrangement with repeated measures and three repetitions. Observations of feeding behavior of the animals were held every 10 minutes during the daytime. Grazing, rumination, and other activities of the animals were affected by period of grazing, days of grazing and stocking rate. There were negative correlations between grazing time of animals and the variables: rumination time, leisure, other activities, temperature and humidity index (ITU) and black globe-humidity index (ITGU). During periods when rainfall occurred the animals showed similar behavior of grazing (grazing time average of 434 minutes), which allowed the use of both stocking rates, without much damage to the behavior of the animals in the rainy season.

Objetivou-se verificar o comportamento de vacas em diferentes taxas de lotação (2, 4 e 6 UA), em pastejo rotacionado. O delineamento experimental utilizado foi blocos ao acaso em arranjo de parcelas subdivididas com medidas repetidas no tempo, com três repetições. As observações do comportamento ingestivo dos animais foram realizadas de 10 em 10 minutos durante o período diurno. O tempo de pastejo, ruminação, ócio e de outras atividades dos animais foi afetado pelo período de avaliação, dia de pastejo e taxa de lotação. Verificou-se a presença de correlação negativa entre o tempo de pastejo e tempo de ruminação, de ócio, outras atividades, índice de temperatura e umidade (ITU) e índice de temperatura globo e umidade (ITGU). Durante os períodos em que ocorreu precipitação pluviométrica os animais apresentaram comportamento de pastejo semelhante (tempo de pastejo médio de 434 minutos), o que permitiu o uso de ambas as lotações, sem maiores danos ao comportamento dos animais, no período chuvoso do ano.

O objetivo deste trabalho foi a clonagem das glicoproteínas transmembrana G e F de um isolado de vírus respiratório sincicial bovino (BRSV) - um isolado brasileiro denominado BRSV-25-BR- que já demonstrou possuir mutações em regiões altamente conservadas do gene da proteína G - em sistema procariótico, com o intuito de sequenciar fragmentos genômicos maiores. As substituições de nucleotídeos foram confirmadas e tais clones podem ser utilizados em futuros estudos sobre os efeitos biológicos destas proteínas tanto in vitro como in vivo.

The aim of this work was the cloning of those transmembrane glycoproteins G and F from an isolate bovine respiratory syncytial viruses (BRSV) - a Brazilian isolate of BRSV, named BRSV-25-BR in previous studies, in a prokaryotic system to proceed the sequencing of larger genomic fragments. The nucleotide substitutions were confirmed and these clones may also be used in further studies regarding the biological effects of those proteins in vitro and in vivo.

Our aim was to determine the frequencies of the angiotensin-converting enzyme (ACE) gene alleles D and I and any associations to cardiovascular risk factors in a population sample from Rio de Janeiro, Brazil. Eighty-four adults were selected consecutively during a 6-month period from a cohort subgroup of a previous large cross-sectional survey in Rio de Janeiro. Anthropometric data and blood pressure measurements, echocardiogram, albuminuria, glycemia, lipid profile, and ACE genotype and serum enzyme activity were determined. The frequency of the ACE*D and I alleles in the population under study, determined by PCR, was 0.59 and 0.41, respectively, and the frequencies of the DD, DI, and II genotypes were 0.33, 0.51, and 0.16, respectively. No association between hypertension and genotype was detected using the Kruskal-Wallis method. Mean plasma ACE activity (U/mL) in the DD (N = 28), DI (N = 45) and II (N = 13) groups was 43 (in males) and 52 (in females), 37 and 39, and 22 and 27, respectively; mean microalbuminuria (mg/dL) was 1.41 and 1.6, 0.85 and 0.9, and 0.6 and 0.63, respectively; mean HDL cholesterol (mg/dL) was 40 and 43, 37 and 45, and 41 and 49, respectively, and mean glucose (mg/dL) was 93 and 108, 107 and 98, and 85 and 124, respectively. A high level of ACE activity and albuminuria, and a low level of HDL cholesterol and glucose, were found to be associated with the DD genotype. Finally, the II genotype was found to be associated with variables related to glucose intolerance.
