A chemical investigation of Anaxagorea dolichocarpa Sprague & Sandwith, a member of Annonaceae family, was carried out. The ethanolic extract from the roots of this plant led, by chromatography tecniches, to isolation of the new azaphenanthrene alkaloids dolichocarpine (1) and 9-methoxyeupolauramine (2), besides the known alkaloids eupolauramine (3), 3-methoxyeupolauridine (4), eupolauramine (5) and 4-methylsampangine (6). The structures of isolated compounds were established by 1D and 2D NMR, HRESIMS, tandem MSn and IR data. The cytotoxicity of compounds 1 - 5 was evaluated against HCT-116 (human colorectal carcinoma) and L929 (murine fibroblast) cell lines.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The phytochemical investigation of Erythroxylum pulchrum St. Hil. (Erythroxylaceae) led to the isolation of three known flavonoid glycosides quercetin-3-O-α-L-rhaminoside, ombuin-3-ruthinoside and ombuin-3-ruthinoside-5-glucoside. These flavonoids are being described for the first time in this E. pulchrum. The structures of the compounds were determined by analysis of IR, MS and NMR data, as well as by comparison with literature data. The methanolic extract of leaves from E. pulchrum inhibited the growth of the Bacillus subtilis CCT 0516, Escherichia coli ATCC 2536, Pseudomonas aeruginosa ATCC 8027, P. aeruginosa ATCC 25619, Staphylococcus aureus ATCC 6538, S. aureus ATCC 25925, Streptococcus sanguinis ATCC 15300, S. salivarius ATCC 7073, S. mutans ATCC 25175 and Streptococcus ATCC. S. aureus ATCC 25925 was the most sensitive among the other S. sanguinis while S. salivarius proved the most resistant.

Sidastrum paniculatum (L.) Fryxell belongs to the family Malvaceae and is popularly known as "malva roxa" or "malvavisco". The phytochemical study of the hexane, CHCl3 and EtOAc phases from the crude ethanol extract of S. paniculatum led to the isolation of six compounds: a mixture of β-sitosterol and stigmasterol, 4-methoxy-3-hydroxybenzoic acid, 4-methoxy-3-hydroxybenzaldehyde, N-trans-feruloyltyramine and kaempferol-3-O-β-D-(6''-E-p -coumaroyl) glucoside. The structural identification of the compounds was made on the basis of spectroscopic methods such as IR, ¹H and 13C NMR with the aid of including two-dimensional techniques, besides comparison with literature data. The β-sitosterol and stigmasterol mixture showed a significant anti-inflammatory activity.

3',8"-biisokaempferide, a cytotoxic biflavonoid and other chemical constituents of Nanuza plicata (Velloziaceae)

Foi isolado das folhas de Nanuza plicata um novo biflavonóide, chamado 3',8"-biisocampferideo (1), juntamente com os conhecidos compostos amentoflavona (2), ácido patagônico(3),(4aR,5S,6R,8aR)-5-[2-(2,5-dihidro-5-metóxi-2-oxofuran-3-il)etil]-3,4,4a,5,6,7,8, 8a-octahidro-5,6,8a-trimetilnaftaleno-1-ácido carboxílico (4), cafeoilquinato de metila (5), ácido 3,5-di-cafeoilquínico (6) e luteolina (7). Os compostos 3, 4, 5 e 6 são relatados pela primeira vez em Velloziaceae. As estruturas dos compostos foram elucidadas com base em métodos espectroscópicos, especialmente RMN e EM. A citotoxicidade de 3',8"-biisocampferideo foi estudada em células de glioblastoma humano (GL-15). A concentração efetiva, que produziu morte em 50% das células após 72 h foi 36,5 µmol L-1. Alterações na morfologia celular, incluindo retração e degradação de citoplasma, foram observadas quando as células foram tratadas com concentrações a partir de 20 µmol L-1 de 3',8"-biisocampferideo por 72h.
A novel biflavonoid, named 3',8"-biisokaempferide (1), along with the known compounds amentoflavone (2), patagonic acid (3), (4aR,5S,6R,8aR)-5-[2-(2,5-Dihydro-5-methoxy-2-oxofuran-3-yl)ethyl]-3,4,4a,5,6,7,8,8a-octahydro-5,6,8a-trimethylnaphthalene-1-carboxylic acid (4), 5-caffeoylquinic acid methyl ester (5), 3,5-di-caffeoylquinic acid (6) and luteolin (7), were isolated of the leaves from Nanuza plicata. The compounds 3, 4, 5 and 6 are reported for the first time in Velloziaceae. The structures of the compounds were elucidated on the basis of spectroscopic methods, especially NMR and MS analyses. The cytotoxicity of 3',8"-biisokaempferide was studied in cultures of human glioblastoma GL-15 cells. The effective concentration, which killed 50 % of cells after 72 h was 36.5 µmol L-1. Changes in cellular morphology, including retraction and degradation of cytoplasm, were observed when cells were treated with concentrations from 20 µmol L-1 of 3',8"-biisokaempferide for 72 h.

Racional - A hemorragia digestiva alta é complicação comum e grave da maioria das doenças do trato gastrointestinal superior. A endoscopia tem papel fundamental no diagnóstico e no tratamento da hemorragia digestiva, mas os dados epidemiológicos a esse respeito ainda são precários em nosso meio. Objetivos - Determinar as características clínicas, a acurácia endoscópica, a eficácia do tratamento e a evolução clínica dos pacientes admitidos no setor de endoscopia digestiva do Hospital Universitário da Universidade Federal do Rio de Janeiro, Rio de Janeiro, RJ, com sangramento digestivo alto. Pacientes e Métodos - Estudo retrospectivo de registros consecutivos de pacientes com hemorragia digestiva alta submetidos a endoscopia de urgência, em período de 2 anos. Resultados - A maioria dos pacientes era do sexo masculino (68.7%) com idade média de 54,5 ± 17,5 anos. O sítio de sangramento foi identificado em 75.6% dos pacientes. A acurácia diagnóstica da endoscopia foi maior quando realizada nas primeiras 24 horas do início do sangramento e quando da presença de hematêmese. A úlcera péptica foi a principal causa de hemorragia digestiva alta (35%). A prevalência de sangramento varicoso (20,45%) indica freqüência alta de hepatopatia subjacente. O tratamento endoscópico foi realizado em 23,86% dos pacientes. Hemostasia permanente foi alcançada em 86% dos pacientes após a primeira intervenção endoscópica e em 62,5% dos pacientes após ressangramento. Cirurgia de emergência foi raramente necessária. A quantidade de unidades de sangue utilizada foi em média 1,44 ± 1,99 por paciente. O tempo médio de internação hospitalar foi de 7,71 ± 12,2 dias. Ressangramento foi observado em 9,1% dos pacientes. A taxa de mortalidade de 15,34% foi significativamente relacionada com a presença de doença hepática prévia. Conclusões - A acurácia diagnóstica foi correlacionada tanto com o intervalo de tempo entre o episódio de sangramento e a realização da endoscopia, como com a forma de apresentação clínica. A terapêutica endoscópica mostrou-se eficaz num grupo selecionado de pacientes. O aumento do tempo de hospitalização e a maior taxa de mortalidade nos pacientes submetidos a terapêutica endoscópica foram atribuídos a maior prevalência de varizes esofagianas e à presença de hepatopatia subjacente.
Background - Upper gastrointestinal bleeding is a frequent and potentially severe complication of most digestive diseases of the upper gastrointestinal tract. Upper endoscopy has a crucial role in the diagnosis and treatment of upper gastrointestinal bleeding, however epidemiological studies are still limited in our country. Aims - To assess the clinical characteristics, endoscopic accuracy, treatment efficiency and clinical outcome of patients admitted to the endoscopic unit with upper gastrointestinal bleeding. Methods - A retrospective study of consecutive records from patients who underwent emergency endoscopy for upper gastrointestinal bleeding was performed during a period of 2 years. Results - Most patients were male 68.7%, with a mean age of 54.5 ± 17.5 years. A bleeding site could be detected in 75.6% of the patients. Diagnostic accuracy was greater within the first 24 hours of the bleeding onset, and in the presence of hematemesis. Peptic ulcer was the main cause of upper gastrointestinal bleeding (35%). The prevalence of variceal bleeding (20.45%) indicates a high rate of underlying liver disease. Endoscopic treatment was performed in 23.86% of the patients. Permanent hemostasis was achieved in 86% of the patients at the first endoscopic intervention, and in 62.5% of the patients after rebleeding. Emergency surgery was seldom necessary. The average number of blood units was 1.44 ± 1.99 per patient. The average length of hospital stay was 7.71 ± 12.2 days. Rebleeding was reported in 9.1% of the patients. The overall mortality rate of 15.34% was significantly correlated with previous liver disease. Conclusions - Diagnostic accuracy was related to the time interval between the bleeding episode and endoscopy, and to clinical presentation. Endoscopic therapy was an effective tool for selected patients. The resulting increased duration of hospitalization and higher mortality rate in the patients submitted to therapeutic endoscopy were attributed to a higher prevalence of variceal bleeding and underlying liver disease.

Phytochemical investigation of the aerial parts of S. paludosum afforded 3,4,7,8-tetramethyl gossypetinr, 3,7- dimethyl kaempferol, 3-methyl kaempferol, 3-methyl apigenin, and 3-methyl quercetin ethers, besides N-p-transcoumaroyltiramine, and protocatecuid acid. The structures were established from spectral data of the natural substances and the permethyl and acetyl derivatives of tetramethyl gossypetin.
