Anamnestic response against bovine viral diarrhea virus and bovine herpesvirus type 1 in young Holstein heifers vaccinated with four different commercial formulations

ABSTRACT: This study evaluated the vaccine-induced serological response after administering four commercial formulations containing Bovine Viral Diarrhea Virus (BVDV) type-1, BVDV-2, and Bovine Herpesvirus type 1 (BoHV-1) to young heifers with circulating maternal antibody titers. The study also determined the anamnestic response to vaccinations after the complete metabolization of maternal antibodies when the calves reached six months. Eighty-seven Holstein heifers were selected and randomly distributed into four experimental groups based on the vaccine administered. The four vaccine-based experimental groups were as follows: vaccine A (n = 22), vaccine B (n = 22), vaccine C (n = 24), and vaccine D (n = 19), given on the 60th, 90th, and 180th day of life, respectively. Virus-neutralization (VN) tests were performed at baseline (day 60) and 30 days after administering the second (day 120) and third doses (day 210). We analyzed the effect of vaccine group (P ≤ 0.001), day of vaccination (P ≤ 0.001), and group versus vaccine day interaction (P ≤ 0.001) for antibody titers produced against BVDV-1, BVDV-2, and BoHV-1 using the PROC MIXED method (Statistical Analysis System -SAS 9.4). Antibody titers against BVDV-1, BVDV-2, and BoHV-1 were similar at baseline and on day 60 of life. The mean antibody titers were constant and persisted against BVDV-1 in heifers immunized with vaccines A and C. Heifers immunized with vaccine A alone had a similar effect against BVDV-2. Regarding BoHV-1, the antibody titers decreased between days 60 and 210 in groups B, C, and D. The antibody titer for heifers in group A also decreased between days 60 and day 120, and an intense increase in titers was observed on day 210. After being immunized with formulations B, C, and D, the frequency of animals with titers above protective levels for BVDV-1, BVDV-2, and BoHV-1 was very low or null. Indicators of anamnestic response were observed in heifers vaccinated with formulation A only. Therefore, it can be concluded that passive immunity negatively interferes with antibody production after vaccination.
RESUMO: O objetivo desse estudo foi avaliar a resposta sorológica induzida após vacinação com quatro formulações comerciais contendo o BVDV-1, BVDV-2 e BoHV-1 em novilhas jovens com títulos de anticorpos maternos circulantes, assim como analisar uma possível resposta anamnéstica em vacinações, após a completa metabolização dos anticorpos maternais aos seis meses de idade. Foram selecionadas 87 novilhas Holandesas, distribuídas aleatoriamente em quatro grupos experimentais de acordo com as vacinas: vacina A (n = 22), vacina B (n = 22), vacina C (n = 24) e vacina D (n = 19), aplicadas aos dias 60, 90 e 180 de vida. Testes de vírus-neutralização (VN) foram realizados no momento basal (D60) e 30 dias após a aplicação da 2ª dose (D120) e 3ª dose (D210). Observou-se efeito do grupo (P ≤ 0,001), dia da vacinação (P ≤ 0,001), e interação grupo versus dia (P ≤ 0,001) para os títulos de anticorpos produzidos contra o BVDV-1, BVDV-2 e BoHV-1, por meio do comando PROC MIXED (Statistical Analysis System, versão 9,4). Os títulos de anticorpos contra o BVDV-1, BVDV-2 e BoHV-1 eram semelhantes no momento basal aos 60 dias de vida. Os títulos médios de anticorpos foram constantes e persistentes contra o BVDV-1 nas novilhas imunizadas com as vacinas A e C, porém apenas a vacina A teve este mesmo perfil contra o BVDV-2. Em relação ao BoHV-1, os grupos B, C e D apresentaram queda nos títulos de anticorpos do D60 ao D210, enquanto as novilhas do grupo A apresentaram queda do D60 ao D120, com aumento intenso dos títulos no D210. A frequência de animais com títulos acima dos protetores contra o BVDV-1, BVDV-2 e BoHV-1 foram muito baixos ou nulos após as vacinações nos animais imunizados com as formulações B, C e D. Sugere-se que a imunidade passiva interferiu negativamente na indução de anticorpos pelas vacinas, observando-se indicadores de resposta anamnéstica apenas para as novilhas vacinadas com a formulação A.

ABSTRACT High-production cows require facilities providing comfortable environments to enhance production efficiency and sustain milk quality. This study evaluated the impact of climatizing the feeding area post-morning and afternoon milking on environmental variables, thermal comfort indices, and the yield and quality of milk in lactating Girolando cows. The experiment followed a 4 × 4 Latin square design with four treatments across four experimental periods, using four cows per treatment. The post-milking climate control treatments included: shading; shading + ventilation; shading + ventilation + shower; and an evaporative adiabatic cooling system. The evaporative adiabatic cooling system proved most effective in maintaining thermal conditions near the comfort level for dairy cows, as indicated by lower thermal comfort indices. This system also enhanced morning and overall milk production. However, the climate control systems did not significantly affect the contents of protein, fat, or lactose in the milk.
RESUMO As vacas de alta produção necessitam de instalações que ofereçam ambientes amenos, para que ocorra aumento da sua eficiência produtiva e manutenção da qualidade do leite. Dessa forma, esse trabalho objetivou avaliar a influência da climatização da área de alimentação após o período das ordenhas matutina e vespertina sobre as variáveis ambientais, índices de conforto térmico, produtividade e qualidade do leite de vacas girolando em lactação. Foi utilizado o delineamento em quadrado latino 4 × 4, com quatro tratamentos e quatro períodos experimentais, com quatro animais por tratamento, totalizando 16 repetições. Os tratamentos foram os sistemas de climatização na pós-ordenha: sombreamento; sombreamento + ventilação; sombreamento+ ventilação+ ducha e sistema de resfriamento adiabático evaporativo. O sistema de resfriamento adiabático evaporativo aumenta a produção leiteira na ordenha da manhã e na produção total. Os teores de proteína, gordura e lactose não foram influenciados pelos sistemas de climatização estudados.

The objective of the present work was to apply self-organizing feature maps (SOFM) and multilayer perceptron (MLP) to evaluate the protective capacity of tert-butylhydroquinone (TBHQ), butylhydroxyanisole (BHA), and butylhydroxytoluene (BHT) antioxidants against the biodiesel oxidation reaction. For this, the antioxidant concentration and the biodiesel compliance parameters were used as continuous input variables and the type of antioxidant as a categorical. The SOFM proved to be an adequate tool for the classification of biodiesel samples containing antioxidants. The performance of the 5 most active networks of the model ranged from 89 to 99% for training, testing, and validation with an error below 1.42 × 10-4. Statistical tests applied to validate the model showed no significant difference between predicted and experimental values. The global sensitivity analysis showed that the relative protection factor (RPF) contributed with 34.89%, the antioxidant with 31.49%, the induction period with 10.69%, the water content with 6.00%, and the others all together with 16.93% in the construction of the regression models by MLP.

The simultaneous diffusion of inorganic components in the olive pulp in wet brine was modeled based on Fick’s generalized 2nd Law and simulated using the finite element method. The main and crossed diffusion coefficients, the film coefficient and the Biot number were determined, with the application of the simplex optimization method, through the minimization of the percentage errors. The errors between the simulated and experimental data were 5.35% for NaCl and 4.77% for KCl and the adjusted main diffusion coefficients were 0.4358 × 10-12 m2 s-1 for NaCl and 0.5408 × 10-12 m2 s-1 for KCl. The system developed to simulate diffusion allows the control and modulation of the salts content that diffuses through the olive pulp.

Abstract In Compost-Bedded Pack Barn (CBP) systems, air velocity is linked with the thermal comfort of housed dairy cattle and bedding quality and, therefore, assessing ventilation efficiency is essential. In this context, the objective of this study was to evaluate and characterize dependence and spatial distribution of air velocity at the 1.5 m height (vair,M) and at bedding level (vair,B) in an open CBP system with positive pressure ventilation. The study was conducted in 2021, in a facility located in the Zona da Mata region, Minas Gerais, Brazil. The facility area was divided into a mesh composed of 55 equidistant points, where vair,M and vair,B data were collected in the morning (09:00 a.m.) and afternoon (03:00 p.m.) periods, during three weeks in Brazilian winter. Geostatistics techniques were used to assess dependence and spatial distribution. In both periods evaluated, there were a strong occurrence of spatial dependence and non-uniform vair,M and vair,B distributions. The vair,M and vair,B values were lower than recommended (1.8 m∙s–1) in more than 65.0% of the area. Adequate ventilation levels were observed only in the first 20.0 m of the facility, from Southeast to Northwest, because of the fan lines present.

Olive samples were subjected to the salting process in brine containing 1196 mol m−3 of NaCl and 402 mol m−3 of KCl. Samples were collected during 60 h and salt concentration values were determined. With the finite element method (FEM) and the minimization of the errors percentage between the simulated and experimental values, the mass transfer coefficients in the film (hm) were obtained, being 1.0072 × 10−8 and 1.2499 × 10−8 for NaCl and KCl, respectively. The salts concentration at the olive/brine interface was simulated by FEM and analyzed via SOFM. A network with 4 × 4 topology and 10000 training epochs was used. It was observed that the influence of the stationary film formed on the surface of the olives during the salting process depends on the position, the salt involved in the diffusion and that the concentration of the salts, at each point, varies according to the immersion time.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

RESUMO Os hidrogéis favorecem a retenção de umidade no substrato e podem atenuar os efeitos do estresse salino na produção de plantas em vaso. Nesse contexto, buscou-se avaliar o uso do hidrogel para mitigar os danos causados pela salinidade da água de irrigação na emergência, crescimento e índice térmico de mudas de Tagetes patula L. A pesquisa foi realizada em casa-de-vegetação, em setembro de 2020, no município de Fortaleza, estado do Ceará, Brasil. O delineamento utilizado foi em blocos casualizados em parcelas subdivididas. As parcelas corresponderam a sete níveis de condutividades elétricas da água de irrigação - CEa (0,5, 1,0, 2,0, 3,0, 4,0, 5,0 e 6,0 dS m-1) e as subparcelas corresponderam a quatro concentrações de hidrogel (0, 1,0, 2,0 e 3,0 g L-1), com cinco repetições e cada subparcela foi composta por 27 sementes. A aplicação de 3,0 g de hidrogel L-1 de substrato favoreceu a produção de folhas e reduziu o estresse térmico nas plantas associado ao aumento da salinidade do substrato até 6,0 dS m-1. A utilização de 3,0 g L-1 de hidrogel incrementou a altura das plântulas, massa seca da folha e massa seca total em comparação aos tratamentos sem o polímero retentor de água, mesmo em níveis moderados (2,0 a 3,0 dS m-1) e altos de salinidade (4,0 a 6,0 dS m-1), indicando assim possível efeito mitigador dos danos causados pela salinidade da água de irrigação. Entretanto, a intensidade desse efeito mitigador deminui nos maiores níveis de estresse salino.
ABSTRACT Hydrogels favor moisture retention in the substrate and can attenuate the effects of salt stress on the production of potted plants. In this context, this research sought to evaluate the use of hydrogel to mitigate the damage caused by the salinity of irrigation water on the emergence, growth, and thermal index of Tagetes patula L. seedlings. The research was carried out in a greenhouse, in September 2020, in the municipality of Fortaleza, Ceará State, Brazil. The design used was randomized blocks in a split plot. The plots corresponded to seven levels of electrical conductivity of the irrigation water - ECw (0.5, 1.0, 2.0, 3.0, 4.0, 5.0, and 6.0 dS m-1) and the subplots corresponded to four hydrogel concentrations (0, 1.0, 2.0, and 3.0 g L-1), with five replicates and each subplot consisted of 27 seeds. The application of 3.0 g hydrogel L-1 of substrate favored the production of leaves and reduced the thermal index in plants associated with the increase in the salinity of the substrate up to 6.0 dS m-1. The use of 3.0 g L-1 hydrogel increases seedling height, leaf dry mass, and total dry mass compared to treatments without water-retaining polymer, even at moderate (2.0 to 3.0 dS m-1) and high (4.0 to 6.0 dS m-1) salinity levels, thus indicating a possible effect of mitigation of damage caused by the salinity of the irrigation water. However, the intensity of this mitigating effect decreases at higher levels of salt stress.

Chagas disease in the 21st century maintains an epidemiological pattern of endemicity in 21 Latin American countries. Researchers and managers from endemic and non-endemic countries point to the need to adopt comprehensive public health policies to effectively control the interhuman transmission of T. cruzi infection, and to obtain an optimized level of care for already infected individuals, focusing on diagnostic and therapeutic opportunistic opportunities.

Pathogenic and pathophysiological mechanisms of the Cardiomyopathy of Chagas disease were revisited after in-depth updating and the notion that necrosis and fibrosis are stimulated by tissue parasitic persistence and adverse immune reaction, as fundamental mechanisms, assisted by autonomic and microvascular disorders, was well established. Some of them have recently formed potential targets of therapies.nbsp;
The natural history of the acute and chronic phases was reviewed, with enhancement for oral transmission, indeterminate form and chronic syndromes. Recent meta-analyses of observational studies have estimated the risk of evolution from acute and indeterminate forms and mortality after chronic cardiomyopathy. Therapeutic approaches applicable to individuals with Indeterminate form of Chagas disease were specifically addressed. All methods to detect structural and/or functional alterations with various cardiac imaging techniques were also reviewed, with recommendations for use in various clinical scenarios. Mortality risk stratification based on the Rassi score, with recent studies of its application, was complemented by methods that detect myocardial fibrosis.nbsp;
The current methodology for etiological diagnosis and the consequent implications of trypanonomic treatment deserved a comprehensive and in-depth approach. Also the treatment of patients at risk or with heart failure, arrhythmias and thromboembolic events, based on pharmacological and complementary resources, received special attention. Additional chapters supported the conducts applicable to several special contexts, including t. cruzi/HIV co-infection, risk during surgeries, in pregnant women, in the reactivation of infection after heart transplantation, and others.nbsp; nbsp;nbsp;
Finally, two chapters of great social significance, addressing the structuring of specialized services to care for individuals with the Cardiomyopathy of Chagas disease, and reviewing the concepts of severe heart disease and its medical-labor implications completed this guideline.
Esta diretriz teve como objetivo principal atualizar os conceitos e formular as normas de conduta e evidências científicas que as suportam, quanto ao diagnóstico e tratamento da CDC, com especial ênfase na base de racionalidade que a embasou.
A DC no século XXI mantém padrão epidemiológico de endemicidade em 21 países da América Latina. Investigadores e gestores de países endêmicos e não endêmicos indigitam a necessidade de se adotarem políticas abrangentes, de saúde pública, para controle eficaz da transmissão inter-humanos da infecção pelo T. cruzi, e obter-se nível otimizado de atendimento aos indivíduos já infectados, com foco em oportunização diagnóstica e terapêutica.
Mecanismos patogênicos e fisiopatológicos da CDC foram revisitados após atualização aprofundada e ficou bem consolidada a noção de que necrose e fibrose sejam estimuladas pela persistência parasitária tissular e reação imune adversa, como mecanismos fundamentais, coadjuvados por distúrbios autonômicos e microvasculares. Alguns deles recentemente constituíram alvos potenciais de terapêuticas.
A história natural das fases aguda e crônica foi revista, com realce para a transmissão oral, a forma indeterminada e as síndromes crônicas. Metanálises recentes de estudos observacionais estimaram o risco de evolução a partir das formas aguda e indeterminada e de mortalidade após instalação da cardiomiopatia crônica. Condutas terapêuticas aplicáveis aos indivíduos com a FIDC foram abordadas especificamente. Todos os métodos para detectar alterações estruturais e/ou funcionais com variadas técnicas de imageamento cardíaco também foram revisados, com recomendações de uso nos vários cenários clínicos. Estratificação de risco de mortalidade fundamentada no escore de Rassi, com estudos recentes de sua aplicação, foi complementada por métodos que detectam fibrose miocárdica.
A metodologia atual para diagnóstico etiológico e as consequentes implicações do tratamento tripanossomicida mereceram enfoque abrangente e aprofundado. Também o tratamento de pacientes em risco ou com insuficiência cardíaca, arritmias e eventos tromboembólicos, baseado em recursos farmacológicos e complementares, recebeu especial atenção. Capítulos suplementares subsidiaram as condutas aplicáveis a diversos contextos especiais, entre eles o da co-infecção por T. cruzi/HIV, risco durante cirurgias, em grávidas, na reativação da infecção após transplante cardíacos, e outros.nbsp;nbsp;nbsp;
Por fim, dois capítulos de grande significado social, abordando a estruturação de serviços especializados para atendimento aos indivíduos com a CDC, e revisando os conceitos de cardiopatia grave e suas implicações médico-trabalhistas completaram esta diretriz.nbsp;

RESUMO O uso de estratégias de manejo ou alterações no ambiente de cultivo podem reduzir os impactos da salinidade da água de irrigação sobre o desenvolvimento vegetal. Assim, nessa pesquisa foram avaliadas respostas fisiológicas, crescimento inicial e qualidade de plantas de Anadenanthera colubrina (Vell.) Brenan (Angico Branco) sob diferentes níveis de sombreamento e condutividade elétrica da água de irrigação, buscando-se identificar as condições mais favoráveis para o desenvolvimento dessa espécie utilizando-se águas salobras. O experimento foi realizado em blocos casualizados com parcelas subdivididas e quatro repetições. As parcelas foram formadas por quatro ambientes com diferentes níveis de sombreamento: 0 (pleno sol), 30, 50 e 70%, e as subparcelas, formadas por cinco níveis crescentes de condutividade elétrica da água de irrigação: 0,4, 1,6, 2,8, 4,0 e 5,2 dS.m−1. Foram avaliadas as variáveis: condutividade elétrica do substrato, trocas gasosas foliares, altura de plantas, produção de biomassa e índice de qualidade de Dickson aos 40 dias em viveiro. O uso do sombreamento artificial favoreceu o desenvolvimento das plantas e a taxa de fotossíntese, mesmo quando irrigadas com água de moderada e elevada salinidade. O IQD mostrou que as plantas de A. colubrina apresentaram maior índice de qualidade quando produzidas sob sombreamento intermediário (30% e 50%), com perdas no rendimento inferiores a 20% quando irrigadas com água com condutividade elétrica de até 2,8 dS.m−1.
ABSTRACT The use of management strategies or changes in the cultivation environment can reduce the impacts of irrigation water salinity on plant development. Therefore, in this research, physiological responses, initial growth, and plant quality of Anadenanthera colubrina (Vell.) Brenan (Angico Branco) were evaluated under different levels of shading and electrical conductivity of the irrigation water, seeking to identify the most favorable conditions for the development of this species using brackish water. The experiment was carried out in randomized blocks with split plots and four replications. The plots were formed by four environments with different shading levels: 0 (full sun), 30, 50, and 70%, and the subplots, formed by five increasing levels of electrical conductivity of the irrigation water: 0.4, 1.6, 2.8, 4.0, and 5.2 dS.m−1. The following variables were evaluated: substrate electrical conductivity, leaf gas exchange, plant height, biomass production, and Dickson quality index at 40 days in a nursery. The use of artificial shading favored plant development and photosynthesis rate, even when irrigated with moderate and high salinity water. The DQI showed that A. colubrina plants presented higher quality index when produced under intermediate shading (30% and 50%), with yield losses below 20% when irrigated with water with electrical conductivity up to 2.8 dS.m−1.

Abstract Background: Patients using immunosuppressive drugs may have unfavorable results after infections. However, there is a lack of information regarding COVID 19 in these patients, especially in patients with rheumatoid arthritis (RA). Therefore, the aim of this study was to evaluate the risk factors associated with COVID 19 hospitalizations in patients with RA. Methods: This multicenter, prospective cohort study is within the ReumaCoV Brazil registry and included 489 patients with RA. In this context, 269 patients who tested positive for COVID 19 were compared to 220 patients who tested negative for COVID 19 (control group). All patient data were collected from the Research Electronic Data Capture database. Results: The participants were predominantly female (90.6%) with a mean age of 53 ±12 years. Of the patients with COVID 19, 54 (20.1%) required hospitalization. After multiple adjustments, the final regression model showed that heart disease (OR =4.61, 95% CI 1.06–20.02. P < 0.001) and current use of glucocorticoids (OR =20.66, 95% CI 3.09–138. P < 0.002) were the risk factors associated with hospitalization. In addition, anosmia was associated with a lower chance of hospitalization (OR =0.26; 95% CI 0.10–0.67, P < 0.005). Conclusion: Our results demonstrated that heart disease and the use of glucocorticoids were associated with a higher number of hospital admissions for COVID 19 in patients with RA. RESUMO Objetivo: Descrever as práticas de ressuscitação volêmica em unidades de terapia intensiva brasileiras e compará-las com as de outros países participantes do estudo Fluid-TRIPS. Métodos: Este foi um estudo observacional transversal, prospectivo e internacional, de uma amostra de conveniência de unidades de terapia intensiva de 27 países (inclusive o Brasil), com utilização da base de dados Fluid-TRIPS compilada em 2014. Descrevemos os padrões de ressuscitação volêmica utilizados no Brasil em comparação com os de outros países e identificamos os fatores associados com a escolha dos fluidos. Resultados: No dia do estudo, foram incluídos 3.214 pacientes do Brasil e 3.493 pacientes de outros países, dos quais, respectivamente, 16,1% e 26,8% (p < 0,001) receberam fluidos. A principal indicação para ressuscitação volêmica foi comprometimento da perfusão e/ou baixo débito cardíaco (Brasil 71,7% versus outros países 56,4%; p < 0,001). No Brasil, a percentagem de pacientes que receberam soluções cristaloides foi mais elevada (97,7% versus 76,8%; p < 0,001), e solução de cloreto de sódio a 0,9% foi o cristaloide mais comumente utilizado (62,5% versus 27,1%; p < 0,001). A análise multivariada sugeriu que os níveis de albumina se associaram com o uso tanto de cristaloides quanto de coloides, enquanto o tipo de prescritor dos fluidos se associou apenas com o uso de cristaloides. Conclusão: Nossos resultados sugerem que cristaloides são usados mais frequentemente do que coloides para ressuscitação no Brasil, e essa discrepância, em termos de frequências, é mais elevada do que em outros países. A solução de cloreto de sódio 0,9% foi o cristaloide mais frequentemente prescrito. Os níveis de albumina sérica e o tipo de prescritor de fluidos foram os fatores associados com a escolha de cristaloides ou coloides para a prescrição de fluidos.
Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.

Inhibition of Acetylcholinesterase by Coumarin-Linked Amino Acids Synthetized via Triazole Associated with Molecule Partition Coefficient

A previous study for the identification of acetylcholinesterase (AChE) inhibitors demonstrated that the hybrid between tyrosol, the 1,2,3-triazole nucleus, and the coumarin group, namely 7-({1-[2-(4-hydroxyphenyl)ethyl]-1H-1,2,3-triazol-4-yl}methoxy)-4-methyl-2H-chromen-2-one (10), has a high enzyme inhibitory activity. Here, we synthesized analogues of 10 via triazole with pharmacophoric groups represented by tyrosine, phenylalanine, tryptophan, and glycine in addition to evaluating the impact of coumarin-linked amino acids on AChE inhibition. We obtained eight triazoles, six of which are undescribed. In general, the presence of carboxylic acid decreased the inhibitory activity, while aromatic amino acids increased enzymatic inhibition compared to glycine. The derivative containing tyrosine, structurally most similar to 10, presented the lowest inhibition percentage, indicating that phenolic hydroxyl is not the preponderant factor for inhibition. Molecular docking was not enough to explain in vitro experiments. On the other hand, MlogP (logP calculated by the Moriguchi method) was related positively to enzymatic inhibition. To increase the hydrophobicity of the molecules, we tested the esterified triazole derivatives comparatively with the enzyme. The compound ethyl 2-(4-(((4-methyl-2-oxo-2H-chromen-7-yl)oxy)methyl)-1H-1,2,3-triazol-1-yl)acetate (6) presented an increment of inhibitory activity of 46.97 ± 1.75% at 100 μmol L-1. We also associated the best activity with the lowest van der Waals volume and molar mass values.

Benzophenones display several biological activities, including antioxidant, anticancer, and photoprotective. Furthermore, antioxidants can minimize both ultraviolet absorption and tumor development. In the present investigation, a series of twenty-six 1,2,3-triazole-benzophenone derivatives were synthesized and had their antioxidant, anticancer, and photoprotective effects evaluated. For the compounds synthesis, 4,4’-dihydroxybenzophenone (1a) and 2,4-dihydroxybenzophenone (1b) were propargylated, affording the alkynes bis(4-(prop-2-yn-1-yloxy))benzophenone (2a) and (2-hydroxy-4-(prop-2-yn-1-yloxy))benzophenone (2b), respectively. The copper(I)-catalyzed azide-alkyne cycloaddition (CuAAC) reaction between the compounds 2a/2b and several benzyl azides gave the 1,2,3-triazole-benzophenone derivatives with yields ranging from 35 to 95%. The 1,2,3-triazole-benzophenone derivatives at the concentration of 0.2 μg mL-1 (a no-cytotoxic concentration) exhibited a solar protection factor (SPF) comparable to positive control benzophonen-3 (BP-3). Concerning their antioxidant and cytotoxic effects, the derivatives from 1b showed high in vitro antioxidant effects as well as cytotoxicity against A549 (lung carcinoma), MCF-7 (breast carcinoma), and HT-144 (metastatic melanoma) cell lines, without significant cytotoxicity to a non-cancerous cell line. Derivatives 19, 20, and 24 induced cell death and cell cycle arrest at G1/S in HT-144 melanoma cells.
