RESUMO Os badnavírus (família Caulimoviridae) possuem genoma de dsDNA semicircular encapsidado em partículas baciliformes. Dioscorea bacilliform virus (DBV) é um importante Badnavirus infectando várias espécies de Dioscorea (inhame) no hemisfério sul. No presente estudo, 235 amostras foliares de inhame foram coletadas em diferentes bancos de germoplasma mantidos pela Escola Superior de Agricultura Luiz (ESALQ/USP), Universidade Federal de Viçosa (UFV) e Universidade Federal do Recôncavo Baiano (UFRB). A incidência média de badnavírus nas amostras avaliadas foi de 75,3%. As análises moleculares e filogenéticas, realizadas a partir de 42 sequências nucleotídicas, da região da RT/RNaseH de badnavírus, revelaram a ocorrência do Dioscorea bacilliform AL virus (DBALV) e Dioscorea bacilliform SN virus (DBSNV) em diferentes espécies de inhame. Até o momento, somente o DBALV havia sido relatado em inhame no Brasil, estando restrito à região Nordeste. Portanto, este é o primeiro relato do DBSNV infectando a cultura do inhame, no país. Além das espécies espissomais, também foram detectadas sequências endógenas, com porcentagem de identidade de nucleotídeos variando entre 89% e 100% com endogenous Dioscorea bacilliform virus (eDBV), pertencentes aos grupos 9 e 12. Os resultados obtidos corroboram com a hipótese de que o DBALV e o DBSNV estão presentes em materiais propagativos de inhame e amplamente disseminados no Brasil.

ABSTRACT Badnaviruses (family Caulimoviridae) have semicircular dsDNA genomes encapsidated into bacilliform particles. Dioscorea bacilliform virus (DBV) is an important Badnavirus infecting several species of Dioscorea (yam) in the southern hemisphere. In this study, 235 yam leaf samples were collected from different germplasm banks maintained by “Luiz de Queiroz” College of Agriculture (ESALQ/USP), Federal University of Viçosa (UFV) and Federal University of Recôncavo da Bahia (UFRB). The average incidence of badnavirus in the evaluated samples was 75.3%. Molecular and phylogenetic analyses, carried out from 42 nucleotide sequences, of the badnavirus RT/RNaseH region, revealed the occurrence of Dioscorea bacilliform AL virus (DBALV) and Dioscorea bacilliform SN virus (DBSNV) in different yam species. So far, only DBALV has been reported in yams in Brazil, being restricted to the Northeast region. Therefore, this is the first report of DBSNV infecting yam crop in the country. In addition to the episomal species, endogenous sequences were detected, and the percentage of nucleotide identity varied between 89% and 100% with endogenous Dioscorea bacilliform virus (eDBV), belonging to groups 9 and 12. The obtained results corroborate the hypothesis that DBALV and DBSNV are present in yam propagating materials and are widely disseminated in Brazil.

RESUMO: O manguezal é um ecossistema costeiro, presente em diversas partes do mundo, provedor de diversos serviços ecossistêmicos desde a provisão de alimentos a influência das mudanças climáticas. Devido ao desenvolvimento da sociedade, este ecossistema tem sido submetido a significativos impactos proveniente das atividades antropogênicas. Diante disso, o objetivo deste trabalho foi avaliar os impactos ambientais ocasionados em áreas de mangue que sofreram modificações resultantes de atividades antrópicas (cultivo agrícola, desmatamento, construção civil) comparando com área de mangue conservado. Esta pesquisa se deu através da análise da sequência temporal de imagens aéreas (Google Earth), e análise da qualidade do solo mediante coletas em campo para avaliação dos indicadores químicos e biológicos nos diferentes sistemas de uso do solo. Por se tratarem de alterações permanentes e que afetaram o tipo do solo e a cobertura do mesmo, foram obtidas diferenças significativas entre as características químicas e biológicas dos quatro ambientes, com os diferentes sistemas de uso. O manguezal tem sofrido impactos negativos pelo manejo inadequado e ocupação do solo. A continuidade da intervenção antrópica no mangue dará prosseguimento ao desaparecimento deste ecossistema a longo prazo. Dentre os atributos químicos e biológicos utilizados para as análises realizadas, o alumínio e os organismos edáficos foram os que permitiram maior contribuição para descriminação do grau de perturbação das áreas de cultivo agrícola, desmatada e transição construção civil/manguezal.

ABSTRACT: The mangrove is a coastal ecosystem that is present in different parts of the world. It provides various ecosystem services from food supply to the influence of climate change. Due to the development of society, this ecosystem has been subjected to significant impacts from anthropogenic activities. Therefore, the objective of this research was to evaluate the environmental impacts caused in mangrove areas that have undergone modifications as a result of anthropic activities (agricultural cultivation, deforestation, civil construction) compared with those of conserved mangrove areas. This research took place through the analysis of the temporal sequence of aerial images (Google Earth) and soil quality analysis through field collections to evaluate the chemical and biological indicators in the different land use systems. As these are permanent changes that affect the type of soil and its coverage, significant differences were obtained between the chemical and biological characteristics of the four environments, with different usage systems. The mangrove has been negatively impacted by inadequate management and land occupation. Continuity of anthropic intervention in the mangrove will promote the disappearance of this ecosystem in the long term. Among the chemical and biological attributes used for the analyses that were performed, aluminum and edaphic organisms were the ones that allowed the greatest contribution of distinction from the degree of disturbance in areas of agricultural cultivation, deforestation and civil construction/mangrove transition.

Resumo O estudo de caracteres morfológicos e suas funções biomecânicas na estrutura craniana das aves levanta debates de cunho filogenético, taxonômico e morfofuncional. O objetivo deste trabalho foi descrever e analisar caracteres morfológicos e morfométricos do esqueleto do Ramphastos toco e confrontá-los com a bibliografia disponível. Foram analisados 13 crânios de indivíduos adultos e de um espécime dissecado fixado em formol (4%). As carcaças foram submetidas à técnica de maceração para o preparo dos esqueletos, com a retirada dos tecidos moles, desengorduramento e clareamento em água oxigenada (50%) e hipoclorito de sódio (50%). O formato e amplitude da sutura naso-frontal e zona flexora craniofacial, a presença dos sesamóides intracapsulares rostral e caudal na articulação quadrática e a presença do ligamento pós-orbital foram fatores que desafiaram a bibliografia existente e os modelos de transferência de força propostos durante estudos da cinética craniana da espécie. A discussão que emerge é a possibilidade dessas características limitadoras da cinética craniana descritas nos espécimes possuírem, em conjunto, a função de promover estabilidade funcional para uma estrutura cuja natureza dimensional, em si, gera instabilidade. Os resultados morfométricos também apresentaram divergências significativas quando comparados à bibliografia disponível, o que pode estar relacionado a variáveis como especiação alopátrica, hibridização e amostragem.

Abstract The study of morphological traits and their biomechanical roles in the cranial structure of birds raises phylogenetic, taxonomic and morphofunctional debates. The aim of this study was to describe and analyze morphological and morphometric data of the skull of Ramphastos toco and to compare the results with the available bibliography. Thirteen adult skulls and one folmaldehyde-fixed dissected specimen were analyzed. For skeletal preparation, the carcasses were submitted to maceration with the removal of soft tissues, degreasing and bleaching in hydrogen peroxide (50%) and sodium hypochlorite (50%). The shape and size of the naso-frontal suture, the presence of the rostral and caudal intracapsular sesamoids in the quadratic joint and the presence of the orbital ligament were altogether elements that challenged the available data. It also challenges the strength transferring models proposed in kinetic studies for the species. The discussion that emerges is the possibility of these limiting characteristics of the cranial kinetics described on the specimens have, together, a single role promoting functional stability to a structure that has a dimensional nature that generates instability. The morphometric results also presented significant divergences when compared to the available bibliography, which may be related to variables such as allopatric speciation, hybridization and sampling.

ABSTRACT The litter in tropical ecosystems is one of the compartments responsible for the transference of nutrients to the soil, having great importance for the conservation and maintenance of biodiversity. Thus, this study aimed to quantify the contribution and stock of litter and the leaf decomposition rate of Gardneriana byrsonima A. Juss (murici) in two areas of Caatinga, semiarid region of Alagoas State. Litter production was measured monthly from March of 2012 to February of 2013. The litter decomposition rate and the remaining leaf mass of the murici were also calculated and analyzed for its material chemically. The leaf decomposition was measured in litter bags and the material was chemically analyzed. In the area of Olho D'Água Casado, the litter produced was over Delmiro Gouveia. The leaf fraction contributed to the higher percentage in the litter training in both areas. The leaf material showed slow decomposition to high levels of lignin, cellulose, hemicellulose, and high C:N ratio.

Objective: to verify the association between sociodemographic conditions and the involvement of pregnant women with drugs. Methods: a cross-sectional study was carried out with 268 pregnant women in a public maternity hospital, through an interview. For the bivariate analysis Fisher's Exact Test and odds ratio with 95% confidence interval were used. Results: statistically significant associations were observed between the use of psychoactive substances by pregnant women and schooling (p=0.017), race (p=0.020) and housing condition (p=0.014). Conclusion: it was evidenced that different factors contribute to the occurrence of vulnerability to health among pregnant women, mainly resulting from the integration of individual and social aspects.

Objetivo: verificar a associação entre as condições sociodemográficas e o envolvimento de gestantes com drogas. Métodos: estudo transversal realizado com 268 gestantes em uma maternidade pública, por meio de entrevista. Para a análise bivariada utilizou-se o Teste Exato de Fisher e odds ratio com intervalo de confiança de 95%. Resultados: foram observadas associações estatisticamente significantes entre o uso de substâncias psicoativas pelas gestantes e a escolaridade (p=0,017), raça (p=0,020) e condição de moradia (p=0,014). Conclusão: evidenciou-se que diferentes fatores contribuem para a ocorrência de vulnerabilidade à saúde entre gestantes, sobretudo resultante da integração de aspectos individuais e sociais.

Human papillomavirus (HPV) infections are strongly associated with the development of cervical intraepithelial neoplasias and invasive cervical cancer. Polymorphisms in cytokine-encoding genes and behavioural cofactors could play an important role in protecting an individual against viral infections and cancer. Here, we investigated whether IL-6 -174 G>C, IL-8 +396 G>T, and TGF-β1 +869 G>C and +915 G>C polymorphisms were associated with susceptibility to HPV infection in women from north-east (Pernambuco) Brazil. We analysed 108 healthy uninfected women (HC) and 108 HPV-positive women with cervical lesions. Genetic polymorphisms were assessed using Sanger sequencing and polymerase chain reaction-restriction fragment length polymorphism. Comparison of the distribution of the genotypic and allelic frequencies of the IL-18 +396 T>G polymorphism between HPV infected woman an uninfected controls showed that the GG genotype and G allele were both more frequent in the HC group, and were associated with protection from HPV infection (p = 0.0015; OR = 0.29 CI95% = 0.13-0.61; p = 0.0005; OR = 0.45 CI95% 0.29-0.7, respectively). Individuals from the control group could have previously had HPV infection that was spontaneously eliminated; however, it was undetectable at the time of sample collection. Based on our findings, we hypothesize that the IL-8 +396 G>T polymorphism could interfere with susceptibility to HPV infection, by modulating the ability of immune system to fight the virus.

RESUMO Objetivo Conhecer a rede e o apoio social da pessoa que vivencia o processo de adoecimento e o transplante renal. Métodos Estudo descritivo com abordagem qualitativa, fundamentado nos conceitos de rede e apoio social de Sherbourne e Stewart. Foram realizadas entrevistas semiestruturadas no período de novembro de 2013 a setembro de 2014, com 12 transplantados renais, as quais passaram por análise de conteúdo. Os critérios de seleção foram os seguintes: realização do transplante, há no máximo, cinco anos, ter idade acima de 16 anos, ter cognição e linguagem preservadas. Resultados A partir do processo de análise, foram elaboradas três categorias que apontam a família como o principal componente da rede social: o serviço de saúde oferta apoio em suas várias dimensões, ainda que ocasionalmente desarticulado; demais relações interpessoais são mencionadas como fragilizadas neste processo; a espiritualidade auxilia no enfrentamento. Conclusões Apesar das diferentes formas de apoio aos transplantados, foram observadas fragilidades nessas redes e apoios sociais.

Semi-structured interviews were conducted from November 2013 to September 2014, with 12 kidney transplantees, which passed through content analysis. This study had the following selection criteria: the interviewee must have had a transplant five years ago, at most, be at lease 16 years of age and have preserved cognitive and language skills. Results three categories were constructed from the analysis process that link the family as the main component of the social network, health service offers support in its various dimensions, even if it is occasionally disconnected; other interpersonal relationships are mentioned as vulnerable in this process; spirituality helps in coping. Conclusions despite the different forms of support for transplantees, weaknesses were observed in these networks and social supports.

A grande dificuldade de implantação da semeadura direta no Nordeste é a produção e o acúmulo de fitomassa na superfície do solo. Há necessidade de se desenvolver sistemas de culturas para região Nordeste, que produzam grandes quantidades de fitomassa, para permanecer por maior tempo na superfície, oferecendo proteção ao solo. O presente trabalho objetivou avaliar a capacidade de produção de fitomassa de diferentes sistemas de culturas e sua influencia na produtividade do milho na microrregião de Guarabira - PB. Foram instalados seis sistemas de culturas: semeadura direta com milho (SD); semeadura direta com milho + mucuna (SDM); semeadura direta com milho + guandu (SDG); semeadura direta com milho + guandu + mucuna (SDGM); semeadura direta com milho + fava (SDF) e semeadura direta com milho + fava + mucuna (SDFM), na presença e ausência de adubação mineral com N-P-K, avaliados durante 9 anos. Foram avaliados o percentual de cobertura e a quantidade de fitomassa sobre o solo, bem como a produtividade de grãos de milho. A adubação mineral e as leguminosas proporcionaram acréscimos variando entre 22% e 49% de produtividade de grãos de milho entre os sistemas de cultivo. Com o passar dos anos, os sistemas de culturas promoveram estoque de fitomassa com acréscimos de até 76% na superfície do solo e, embora a precipitação seja um fator limitante para produção de fitomassa, o uso das leguminosas e da adubação mineral com N-P-K nos sistemas de culturas facilitaram a implantação do sistema de semeadura direta na região estudada.

The major problem of implementation of direct seeding in the Northeast is the phytomass production and accumulation on the soil surface. There is a necessity to develop cropping systems for the Northeast region, which produce large quantities of phytomass, to stay for longer time on the soil surface, providing protection to the soil. This study aimed to evaluate the ability of phytomass production of different cropping systems and its influence on corn yield in the microregion of Guarabira - PB. Six cropping systems were installed: tillage with maize (SD); tillage with corn+velvetbean (SDM); tillage with maize+pigeonpea (SDG); tillage with corn+ velvetbean+pigeonpea (SDGM); tillage with corn+broad bean (SDF); and no tillage with corn+ velvetbean+broad bean (SDFM); with and without NPK fertilizer, were evaluated for 9 years. The percentage of crop cover and the phytomass accumulation on the soil and yield of corn were evaluated. The mineral fertilizer and legumes provided increases ranging between 22% and 49% of corn grain yield among tillage systems. Over the years, crop systems promoted stock biomass with increases of up to 76% on the soil surface, and although the precipitation is a limiting factor for biomass production, the use of legumes and fertilization with NPK mineral systems cultures facilitated the implementation of no-till system in this region.

A substituição da vegetação nativa no Brejo Paraibano, inicialmente por cana-de-açúcar e mais recentemente por pastagens, em relevo fortemente ondulado, causaram sérios problemas de degradação do solo. Atualmente, partes dessas pastagens estão dando lugar ao plantio de sabiá (MIMOSA CAESALPINIAEFOLIA: Benth). Objetivou-se, neste trabalho, analisar o efeito dessas mudanças nas propriedades físicas e químicas do solo e no desenvolvimento das raízes. O experimento foi conduzido no município de Areia, PB, em um Argissolo Vermelho-Amarelo distrófico. Os tratamentos foram representados por três diferentes usos: mata secundária, cultivo de sabiá e pastagem com braquiária (BRACHIARIA DECUMBENS: ). As amostras de solo foram coletadas nas diferentes áreas, em três posições da encosta: parte superior, mediana e inferior. Em cada posição, coletou-se solo em três diferentes pontos em quatro profundidades: 0,0-2,5; 2,5-5,0; 5,0-10,0; e 10,0-20,0 cm, para análises físicas e de raízes. Foram feitas amostras compostas para as análises químicas e do conteúdo de argila. O experimento foi conduzido num delineamento inteiramente casualizado, com medidas repetidas no espaço. Observou-se que a agregação, densidade e a porosidade total apresentaram melhores resultados na área de mata nativa secundária. A substituição da área de pasto por sabiá prejudicou a agregação, mas não alterou a densidade e a porosidade total do solo. Os teores de nutrientes foram maiores na área de sabiá, embora sejam significativos apenas para o K e a saturação por bases. As mudanças no uso do solo sempre causam degradação de suas propriedades físicas e químicas, mas, quando essa substituição é feita por um sistema menos agressivo, é difícil perceber modificações em pouco tempo de uso.

The substitution of the native vegetation of marshlands in Paraiba, initially by sugarcane and more recently by pastures, in a strongly undulated relief, has caused serious problems of soil degradation. Currently, some of these pastures are being replaced by plantations of sabiá (Mimosa caesalpiniaefolia Benth). The objective was to analyze the effect of these changes on the soil physical and chemical properties and root development. The experiment was conducted in Areia-PB, in a dystrophic Ultisol. The three treatments were represented by three land uses:secondary native forest, sabiá plantation, and brachiaria pasture grass (Brachiaria decumbens). Soil samples were collected from three slope positions: top, middle, and bottom. For physical properties and root studies, in each position soil was collected at three different points and in four depths (0.0-2.5, 2.5-5.0, 5.0-10.0, and 10.0-20.0 cm). Composite samples were made to assess the chemical properties and clay content. The experiment was conducted in a completely randomized design with repeated measures in space. The results showed better aggregation, density, and porosity in the area of native secondary forest. The replacement of pasture by sabiá lead to a decrease in the aggregation but did not affect bulk density or soil's total porosity. Nutrient levels were higher in the sabiá area, although statistically different just for K and saturation of exchangeable bases. Changes in land use invariably degrade soil physical and chemical properties, but when a less aggressive system is introduced, changes are hardly detectable in the short term.

OBJETIVO: comparar três métodos para detecção do HPV e determinar a prevalência dos genótipos encontrados. MÉTODOS: um total de 120 amostras de raspagem da região cervical de mulheres portadoras de neoplasia intraepitelial cervical foram analisadas pela reação em cadeia da polimerase convencional, usando os sistemas de primers MY09/11, GP05+/06+ e pela Nested-PCR. As amostras foram submetidas à extração de DNA e, logo após, amplificadas com os primers GH20 e PC04 (β-globina) para verificação da qualidade do DNA obtido e pela reação em cadeia da polimerase convencional e Nested-PCR. Os fragmentos amplificados foram visualizados em gel de agarose a 1,2%, corados com Blue Green Loading Dye I. As amostras positivas foram sequenciadas usando o sequenciador automático de DNA "MegaBACE 1000". Para análise estatística foram utilizados os teste do Χ2 e o de Fisher com nível de significância de 5%. RESULTADOS: quinze amostras não se amplificaram para os primers de β-globina, sendo eliminadas do estudo. Das amostras restantes, 40% (42/105) foram positivas para os primers MY09/11, 98% (103/105) para os primers GP05+/06+ e 92% (97/105) para Nested-PCR. Considerado as técnicas MY09/11 e GP05+/06+, foi possível observar 100% de amostras positivas para o HPV. Neste estudo, a prevalência dos genótipos foi de 58, 23, 5, 4 e 3% para HPVs 16, 18, 31, 33 e 56, respectivamente. Os HPV 67 e 83 apresentaram 2% e os HPV 6, 11, 58 e candHPV85, 1% cada. A prevalência dos genótipos neste estudo está de acordo com o reportado em todo o mundo (IC95%=0,4657-0,8976). CONCLUSÕES: para obter resultados mais confiáveis, é necessário o uso de mais que um sistema de primers para detecção do HPV. Acredita-se que as três técnicas estudadas são importantes e adequadas para o diagnóstico clínico do HPV quando apropriadamente combinadas.

PURPOSE: to compare three methods for the detection of HPV infection and to determine the prevalence of the genotypes found. METHODS: a total of 120 cervical scrape samples from patients with cervical intraepithelial neoplasia were analyzed by the conventional polymerase chain reaction using the MY09/11 and GP05+/06+ primers, and by the Nested polymerase chain reaction. The samples were subjected to DNA amplification with the GH20 and PC04 primers (β-globin) to verify DNA quality and also by polymerase chain reaction and Nested polymerase chain reaction. The amplicons were visualized in 1.2% agarose gel stained with Blue Green Loading Dye I. Positive samples also were sequenced using the automatic DNA sequencer "MegaBACE 1000". The Χ2 and Fisher tests were used for statistical analysis with the level of significance set at 5%. RESULTS: fifteen samples were eliminated from the study because they failed to amplify the β-globin gene. Of the remaining samples, 40% (42/105) were positive using primers MY09/11, 98% (103/105) using primers GP05+/06+, and 92% (97/105) using Nested-PCR. With the MY09/11 and GP05+/06+ techniques, it was possible to obtain 100% HPV-positive samples. In this study, the prevalence of the genotypes found was 57, 23, 5, 4 and 3% for HPV genotypes 16, 18, 31, 33 and 56, respectively. HPVs 67 and 83 were present in 2%, and genotypes 6, 11, 58 and candHPV85 were present in 1% each. The prevalence of the more common genotypes (HPV 16 and 18) in this study agrees with that reported worldwide (IC95%=0.4657-0.8976). CONCLUSIONS: to obtain more reliable results, it is necessary the use of more than one primer system to detect HPV infections. We believe that the three techniques studied are important and suitable for the clinical diagnosis of HPV, when they are appropriately combined.
