ABSTRACT Pacas (Cuniculus paca) are highly hunted animals because of the flavor of their meat, and commercial breeding is recommended. However, this species has a relatively low reproductive rate. This study aimed to collect semen from pacas through electroejaculation and obtain the sperm parameters of this species for the first time. Seven male pacas were used, submitted to an anesthetic protocol before stimulation by an electroejaculator appropriate for the species. The stimulus protocol was performed in three series: series I, 10 stimuli with 1 and 2 V; series II, 10 stimuli with 3 and 4 V; and series III, 10 stimuli with 5 V and interval between series of 2 s. The collected material was evaluated for color, volume, motility, vigor, and concentration. The sperm parameters collected showed a mean volume of 0.43±0.33 mL, concentration of 45.5±42.44×106 sperm/mL, motility of 33.33±32.14%, and mean vigor of 2.6±1.15. In this study, the anesthetic protocol did not seem to favor semen collection by electroejaculation in the pacas. The electrical stimulation protocol was able to stimulate all animals in the study; however, there were few samples with sperm cells and a low rate of motility and vigor in most ejaculates.

RESUMO Pacas (Cuniculus paca) são animais altamente caçados devido ao sabor de sua carne, sendo a criação comercial delas, recomendada. Entretanto, é uma espécie com baixa taxa reprodutiva. O objetivo deste estudo foi efetuar coleta de sêmen em pacas por meio da eletroejaculação e da obtenção, pela primeira vez, dos parâmetros espermáticos dessa espécie. Foram utilizadas sete pacas machos, as quais, antes dos estímulos por eletroejaculador apropriado para a espécie, foram submetidas a protocolo anestésico. O protocolo de estímulo foi realizado em três séries: série I, 10 estímulos com 1 e 2V; série II, 10 estímulos com 3 e 4V; série III, 10 estímulos com 5V, e intervalo entre as séries de dois segundos. O material coletado foi avaliado quanto à cor, ao volume, à motilidade, ao vigor e à concentração. Os parâmetros espermáticos coletados demonstraram volume médio de 0,43±0,33mL, concentração de 45,5±42,44x106 espermatozoides/mL, motilidade de 33,33±32,14% e vigor médio de 2,6±1,15. Neste estudo, o protocolo anestésico parece não ter favorecido a coleta de sêmen por eletroejaculação em pacas. Entretanto, o protocolo de estímulos elétricos foi capaz de estimular todos os animais do estudo. Assim, chegou-se ao resultado de poucas amostras com a presença de células espermáticas, bem como baixo índice de motilidade e vigor na maioria dos ejaculados.

RESUMO: Na micropropagação, o nitrato de potássio (KNO3), reagente puro para análise (P.A.), utilizado no preparo dos meios de cultura, possui custo elevado e a sua aquisição depende de trâmites burocráticos, por se tratar de substância controlada pelo Exército Brasileiro. O objetivo deste trabalho foi avaliar o efeito da substituição do KNO3 P.A. por fertilizante comercial (com fonte de KNO3), encontrado livremente no comércio, na micropropagação de palma (Opuntia stricta (Haw.) Haw. cv Orelha de Elefante. Os tratamentos foram de seis concentrações do fertilizante (0; 0,5; 1; 1,5; 2 e 2,5 g L-1) e um controle constituído de 1,9 g L-1 de reagente KNO3, conforme mostrado nos sais MS. Avaliou-se a sobrevivência, tamanho e número de brotações do explante, e o valor da biomassa fresca. Após a aclimatização das mudas avaliou-se a sobrevivência, número de brotações, comprimento da parte aérea, número de raízes, formação da raquete, massa média da biomassa fresca, absorção de macronutrientes e alterações morfológicas das mudas. Os explantes inoculados em meio com fertilizantes nas concentrações de 0,0; 2,0 e 2,5 g L-¹ não se desenvolveram. A resposta dos explantes nas concentrações de 0,5 e 1,5 g L-1 do fertilizante foram iguais aos desenvolvidos em meio contendo KNO3, e na concentração de 1,0 g L-1, em todas as variáveis, as médias foram superiores em relação as do controle. Dessa forma, constatou-se a viabilidade do uso do fertilizante no cultivo in vitro da palma, o que propiciou a eliminação dos entraves burocráticos e redução no custo de 99,12% na compra do produto.

ABSTRACT: In micropropagation, potassium nitrate (KNO3), an ACS reagent grade chemical, used in the preparation of growing mediums is expensive and its procurement depends on bureaucratic procedures, as it is controlled by the Brazilian Army. This research to assessed the effect of replacing the ACS KNO3 for a commercially available fertilizer (KNO3- based) on the micropropagation of the prickly pear cactus (Opuntia stricta (Haw.) Haw. cv. Elephant Ear. Treatments used six different fertilizer concentrations (0, 0.5, 1, 1.5, 2 and 2.5 g L-1) and a control consisting of 1.9 g L-1 KNO3, as shown in the MS salts. The survival, size and number of sprouts and the value of fresh biomass were evaluated. After seedling acclimation, we assessed the survival, number of sprouts, length, and number of roots, racket formation, average fresh biomass mass, macronutrient absorption and morphological changes of the seedlings. Explants inoculated with fertilizers at concentrations of 0.0; 2.0 and 2.5 g L-¹ did not grow. The response of explants at concentrations of 0.5 and 1.5 g L-1 of the fertilizer were the same as those developed in a KNO3 medium, and at a concentration of 1.0 g L-1, in all variables, the means were higher than those of the control medium. Therefore, it showed the feasibility of using fertilizers in the in vitro cultivation of the prickly pear cactus, which may remove bureaucratic barriers and reduce product costs by 99.12%.

Abstract The objective of this research was to evaluate the inoculation and co-inoculation of bacteria with biotechnological potential, isolated from Brachiaria decumbens Stapf. and Brachiaria humidicola (Rendle) Schweickerdt, under germination and seedling growth of B. decumbens cv. Basilisk, as well as, to verify the influence of the co-inoculum in the soil indigenous bacterial community. For this, two assays in a completely randomized design were set up. The experimental period was 21 days. In a germination chamber, 25 treatments were evaluated (24 bacterial inoculants and a control – without inoculation). In greenhouse assay, were used five co-inoculations (bacterial consortium). The bacterial consortium was obtained based on the bacterial strain performance in the germination test. In addition, the control and one treatment with mineral fertilizer (NPK) were tested. In germination test, the seed inoculation promoted increases of 61, 40, 144, 82, 6, 96, 91 and 52% in germination vigor, speed germination index, number of absorbent hairs, number of plumules, primary root length, hypocotyl length, total length, and dry matter of seedlings, respectively, when compared to control. The co-inoculation also increased the growth parameters of B. decumbens plants when compared to the control treatment. In addition, promoted changes in the soil bacterial community structure. Becoming an important strategy to increase the germination rate and germination speed of B. decumbens plants.

Abstract Mesenchymal stem cells present clinical potential to recover and regenerate injured tissues in diverse pathologies. The in vitro expansion and characterization of these cells contribute to elucidation of the mechanisms of senescence and strategies involving cell therapies. This study aimed to compare specific characteristics between initial and advanced passages of mesenchymal stem cells derived from adipose tissue and bone marrow. Both cell types were characterized according to immunophenotype, osteogenic differentiation, genomic instability, migration assay, doubling population time and colony forming ability. Our results demonstrated that both cell types were able to maintain an immunophenotypic profile typical of mesenchymal stem cells during increasing passages. Adipose stem cells at initial passage presented greater migration capacity compared to advanced passage cells, and advanced passage cells proliferated faster than initial passage cells. Bone marrow stem cells at early passages presented higher osteogenic potential than advanced. At advanced passages they presented higher colony forming capacity and genetic damage than those at initial passage. These results suggest that mesenchymal stem cells maintained in culture presented characteristics of senescence that should be monitored prior the use in regenerative medicine and cells derived from bone marrow at initial passage have better potential for therapeutic use in bone tissue engineering.

RESUMO Diante da transição na estrutura energética mundial, com o deslocamento do consumo dos combustíveis fósseis para fonte renováveis e limpas, registra-se um aumento da demanda por biomassa florestal para fins energéticos, em especial os pellets de madeira, e as importações têm crescido nos últimos anos. Logo, este artigo analisou a concentração mundial das importações de pellets, de 2012 a 2018. Os dados de importações de pellets foram obtidos da Food and Agriculture Organization of the United Nations – FAO e para mensurar a concentração foram utilizados os indicadores: Razão de Concentração [CR(k)], Índice de Hirschman-Herfindal (HHI) Índice de Entropia de Theil (E), Coeficiente de desigualdade de Gini (G) e o Índice de Hall-Tideman (HTI). Os resultados apontaram um crescimento de 16,67% a.a. da importação mundial de pellets, saindo de 8,76 milhões toneladas (t), em 2012, para 22,15 milhões de t em 2018. Quanto aos índices de concentração, o CR(k) indicou concentração muito alta para os países e alta nos subcontinentes. O HHI apresentou concentração alta para os continentes e subcontinentes e uma concentração moderada para os países. A Entropia e o HTI corroboraram as análises encontradas no HHI. O G apontou forte desigualdade para todos os níveis territoriais e apresentou tendências de redução na desigualdade a partir de 2015. A redução da concentração, nos últimos anos de estudo, encontra relação com a expansão e a difusão tecnológica de conversão energética da biomassa densificada, o que torna o combustível mais acessível.

ABSTRACT Faced with the transition in the global energy structure with the shift in consumption of fossil fuels to renewable and clean sources, there has been an increase in the demand for forest biomass for energy purposes, especially wood pellets, and imports have grown in recent years. Therefore, this study analyzed the world concentration of pellet imports from 2012 to 2018. Data on pellet imports were obtained from the Food and Agriculture Organization of the United Nations (FAO), and the following indicators were used to measure the concentration: Concentration Ratio [CR(k)], Hirschman-Herfindal Index (HHI), Theil Entropy Index (E), Gini Inequality Coefficient (G) and the Hall-Tideman Index (HTI). The results showed a growth of 16.67% p.a. of global pellet imports, from 8.76 million tons (t) in 2012 to 22.15 million tons in 2018. The CR(k) indicated very high concentration for countries and high in the subcontinents. The HHI showed a high concentration for continents and subcontinents and a moderate concentration for countries. Entropy and HTI corroborated the analyzes found in the HHI. The G pointed out strong inequality for all territorial levels and showed trends towards a reduction in inequality as of 2015. The reduction in the concentration in the final years of study is related to the expansion and technological diffusion of energy conversion of the densified biomass, which makes this fuel more affordable.

This study evaluated the effects on the internal quality of eggs of various storage environments through which eggs may pass between being laid and being consumed. Commercial eggs (N = 648) from Dekalb White hens were used. Treatments consisted of T1: 28 days at 4 °C; T2: 28 days at 20 °C; T3: 7 days at room temperature (27 °C ± 2 °C) (humidity 55%) and 21 days at 4 °C; T4: 7 days at room temperature and 21 days at 20 °C; T5: 14 days at room temperature and 14 days at 4 °C; T6: 14 days at room temperature and 14 days at 20 °C; T7: 21 days at room temperature and 7 days at 4 °C; T8: 21 days at room temperature and 7 days at 20 °C; and T9: 28 days at room temperature. The characteristics that were evaluated consisted of Haugh unit (HU), yolk index (YI), colour (L*, a* and b*), albumen pH, yolk pH and lipid oxidation. Eggs stored 28 days were darker (L*), and had greater yolk pH and lipid oxidation than fresh eggs. Eggs stored under T1 and T3 conditions had greater HU and YI than eggs stored in the other environments. The albumin pH of eggs stored at room temperature (T9) was highest of the treatments. Yellowness was increased in eggs stored under T4, T6, T8, and T9 conditions. Eggs should be stored under refrigeration as this promotes maintenance of internal quality and mitigates negative effects of previous storage conditions.

Abstract Plant growth-promoting bacteria (PGPB) have received great interest in recent decades. However, PGPB mechanisms remain poorly understood in forage species. We aimed to evaluate roots endophytic and rhizospheric bacteria strains from Brachiaria humidicola and Brachiaria decumbens. The strains were evaluated for biological nitrogen-fixing in saline stress (0 to 10.0 g L-1 of NaCl), N-acyl homoserine lactones and indole-like compounds (ILC) production, the activity of hydrolytic enzymes, and inorganic phosphate solubilization (IPS) under different C sources. The diversity of strains was assessed by BOX-PCR. About 58% of strains were positive for BNF. High salinity levels reduced the growth and BNF. About 58% produced N-acyl homoserine lactones. The ILC was present in 39% of strains. Cellulase, polygalacturonase, pectate lyase, and amylase production were observed in 77, 14, 22, and 25% of strains, respectively. The IPS was observed in 44, 81, and 87% of isolates when glucose, mannitol and sucrose were used, respectively. Comparing two plant species and niches, the strains associated with B. humidicola and root endophytic presented more PGPB mechanisms than others. We found high strain diversity, of which 64% showed similarity lower than 70%. These results can be supporting the bioproducts development to increase forage grasses production in tropical soils.

The oxygen uptake (V˙O2) kinetics during onset of and recovery from exercise have been shown to provide valuable parameters regarding functional capacity of both chronic obstructive pulmonary disease (COPD) and chronic heart failure (CHF) patients. To investigate the influence of comorbidity of COPD in patients with CHF with reduced ejection fraction on recovery from submaximal exercise, 9 CHF-COPD male patients and 10 age-, gender-, and left ventricle ejection fraction (LVEF)-matched CHF patients underwent constant-load exercise tests (CLET) at moderate and high loads. The V˙O2, heart rate (HR), and cardiac output (CO) recovery kinetics were determined for the monoexponential relationship between these variables and time. Within-group analysis showed that the recovery time constant of HR (P<0.05, d=1.19 for CHF and 0.85 for CHF-COPD) and CO (P<0.05, d=1.68 for CHF and 0.69 for CHF-COPD) and the mean response time (MRT) of CO (P<0.05, d=1.84 for CHF and 0.73 for CHF-COPD) were slower when moderate and high loads were compared. CHF-COPD patients showed smaller amplitude of CO recovery kinetics (P<0.05) for both moderate (d=2.15) and high (d=1.07) CLET. Although the recovery time constant and MRT means were greater in CHF-COPD, CHF and CHF-COPD groups were not differently affected by load (P>0.05 in group vs load analysis). The ventilatory efficiency was related to MRT of V˙O2 during high CLET (r=0.71). Our results suggested that the combination of CHF and COPD may further impair the recovery kinetics compared to CHF alone.

RESUMO O hipoparatireoidismo, quer seja primário ou secundário, é uma doença rara em cães, causada pela diminuição da secreção de paratormônio pelas paratireoides, que leva a sinais clínicos resultantes da hipocalcemia. O omeprazol vem sendo cada vez mais utilizado na medicina veterinária visando à diminuição na produção de líquor, mas existem poucos estudos sobre os efeitos colaterais relacionados ao uso crônico dessa medicação. Relata-se o caso de um cão macho da raça Yorkshire Terrier, com quatro anos de idade, com sinais clínicos de dor, sendo verificada calcificação em pelve e divertículo renal. Segundo o proprietário, o cão fazia uso de omeprazol há mais de dois anos devido ao histórico de hidrocefalia. Os exames laboratoriais evidenciaram anemia microcítica hipocrômica, hipocalemia, hiperfosfatemia, hipocalcemia, hipomagnesemia e hipercalciúria. A dosagem do paratormônio sérico confirmou o hipoparatireoidismo. Após a suspensão do omeprazol, as alterações encontradas nos exames se normalizaram, confirmando que a causa do hipoparatireoidismo era o uso crônico da medicação.

ABSTRACT Primary or secondary hypoparathyroidism is a rare disease in dogs caused by the decreased secretion of parathormone from the parathyroid glands, leading to clinical signs of hypocalcemia. Omeprazole has been increasingly used in veterinary medicine in order to reduce the production of cerebrospinal fluid, but there are few reports of side effects related to its chronic use. We report a case of a four-year-old male Yorkshire terrier with clinical signs of pain, calcification in the pelvis and renal diverticulum. According to the owner, the dog had been receiving omeprazole for over 2 years because of the history of hydrocephalus. Hematological exams revealed hypochromic microcytic anemia, hypokalemia, hyperphosphatemia, hypocalcemia, hypomagnesemia besides hypercalciuria. The determination of serum parathyroid hormone concentrations confirmed hypoparathyroidism. After interrupting omeprazole, the altered features on the exams returned to normal values, confirming that the cause of hypoparathyroidism was the chronic use of the drug.

ABSTRACT Introduction and objective: In this study, we evaluated the influence of the transcript type on hematological and clinical parameters, as well as the event-free survival of 50 patients in the Chronic myeloid leukemia chronic phase. Methods: We reviewed the medical records of 55 patients with Chronic myeloid leukemia. The eligibility criteria were based on the availability of hematological and clinical baseline data in the medical records. Data on BCR-ABL transcripts were obtained from medical records. Results: Eighteen patients (36%) had the b2a2 transcript, 24 (48%) had b3a2 and 8 (16%) had b2a2/b3a2. The median platelet count for transcripts b2a2, b3a2 and b2a2/b3a2 was 320.65 × 103/L, 396 × 103/L, and 327.05 × 103/L, respectively (p = 0.896). We could not find any differences in relation to the other hematological parameters, when compared to the transcript type. Comparison between spleen and liver size and type of transcript did not differ inside the groups (p = 0.395 and p = 0.647, respectively) and the association between risk scores and transcript type did not show statistical significance (p > 0.05). The 21-month probability for event-free survival was 21%, 48% and 66% for the transcripts b2a2, b3a2 and b2a2/b3a2 respectively (p = 0.226) Conclusion: We conclude that the expression BCR-ABL transcripts have no influence on hematological, clinical and event-free survival parameters of patients in the Chronic myeloid leukemia chronic phase.

Resumo Concluiu-se que, pela regra do fator previdenciário, as alíquotas vigentes são insuficientes para cobertura dos benefícios previdenciários, uma vez que as alíquotas atuarialmente justas são 30,69% e 35,27% para homens e mulheres, respectivamente. Contudo, caso a reforma da Previdência fosse aprovada em seu texto original, os percentuais justos seriam reduzidos para 22,25%, e 21,60%, respectivamente. Além da idade mínima, grande parte dessa redução deve-se às regras propostas para cálculo da pensão, que admitem valor abaixo do salário mínimo. Esses resultados atenderam ao objetivo deste trabalho, que foi comparar as alíquotas previdenciárias atuarialmente justas para o Regime Geral de Previdência Social, tendo como base as regras do fator previdenciário e da proposta de idade mínima, presente na Proposta de Emenda à Constituição n. 287/2016. As mudanças demográficas ocorridas no Brasil nos últimos anos chamam a atenção para a questão da sustentabilidade do sistema previdenciário nacional, e aprovar a reforma previdenciária tem sido prioridade do governo. Logo, é indiscutível a necessidade de um estudo atuarial que viesse calcular alíquotas atuarialmente justas e comparar o atual cenário com as propostas de reforma. Utilizaram-se modelos atuariais com múltiplos decrementos para o cálculo das alíquotas justas, considerando uma família padrão [funcionário(a) de 25 anos, cônjuge e dois filhos], sendo o homem três anos mais velho que a mulher. Adotaram-se como premissas biométricas as tábuas IBGE 2015 - Extrapolada (mortalidade) e Álvaro Vindas (invalidez), a taxa real de crescimento salarial de 2% a.a. e a taxa real de juros de 3% a.a.

Abstract It has been shown that under the social security factor rule current contribution rates are insufficient to cover social security benefits, since the actuarially fair rates are 30.69% and 35.27% for men and women, respectively. However, if the social security reform were approved as submitted, the fair rates would be reduced to 22.25% and 21.60%, respectively. Besides the minimum age, part of this reduction is due to the proposed rules allowing pension values lower than the minimum wage. These results served the objective of this work, which was to compare the actuarially fair social security rates for the General Social Welfare Policy (GSWP), based on the social security factor rules and the minimum age proposal present in Proposed Constitutional Amendment n. 287/2016. The demographic changes that have taken place in Brazil in recent years raise questions about the sustainability of the national social security system and approving social security reform has been a government priority. Therefore, there is an undisputed need for an actuarial study that calculates actuarially fair rates and compares the current scenario with the reform proposals. Multiple decrement actuarial models were used to calculate the fair rates considering a standard family (25-year-old worker, spouse, and two children), in which the man is three years older than the woman. The IBGE 2015 Extrapolated (mortality) and Álvaro Vindas (disability) tables were adopted as biometric assumptions, and a real wage growth rate of 2% p.a. and real interest rate of 3% p.a. were used.

RESUMO Este trabalho analisou a produção mundial de MDF e o seu grau de concentração, entre os anos de 1995 e 2016. Para mensurar e analisar esta concentração foram usados a Razão de Concentração [CR(k)], o Índice de Herfindahl-Hirschman (HHI), o Índice de Entropia de Theil (E), o Índice de Hall e Tideman (HTI), o Índice de Concentração Compreensível (CCI) e o Índice de Gini (G). A produção de mundial de MDF cresceu 12,81% a.a., no período de 1995 a 2016, chegando a 99 milhões de m3 no final da série temporal. A China assumiu a hegemonia em 2001 e chegou em 2016 com cerca de 60% da oferta mundial de MDF. O CR(k) da produção mundial de MDF inferiu uma alta concentração, principalmente, no CR(4) a partir de 2009. O CR(8) manteve-se com uma média de concentração moderadamente alto. Nesse período mais de 90% da oferta ficaram retidas no CR(20). Os índices HHI, E e HTI corroboram de que há alta concentração na produção mundial de MDF, assim como o CCI no período estudado. A desigualdade indicada pelo G também apresentou comportamento crescente e foi classificada em forte e muito forte. Os indicadores de concentração se mostraram eficientes em avaliar o grau de concentração da oferta mundial de MDF.

ABSTRACT This papaer analyzed the global MDF production and its concentration degree between 1995 and 2016. In order to measure and analyze this concentration, we used the Concentration Ratio [CR(k)], the Herfindahl-Hirschman Index (HHI), Theil Entropy Index (E), Hall and Tideman Index (HTI), the Comprehensive Concentration Index (CCI), and the Gini Index (G). Global MDF production grew 12.81% p.a. from 1995 to 2016, reaching 99 million m3 at the end of the time series. China took over the hegemony in 2001 and arrived in 2016 with about 60% of the world’s MDF supply. The CR(k) of the global MDF production inferred a high concentration, mainly in the CR(4) from 2009. The CR(8) remained with a moderately high average concentration. During this period more than 90% of the offer was retained in the CR(20). The HHI, E and HTI indices corroborate that there is high concentration in global MDF production, as well as the CCI in the studied period. The inequality indicated by G also presented increasing behavior and was classified as strong and very strong. The concentration indicators were efficient in evaluating the concentration degree of the world MDF supply.

RESUMO A fauna de anfíbios do estado de Mato Grosso do Sul foi compilada e elaborada pela primeira vez, totalizando 97 espécies, o que representa pouco mais de 10% das espécies que ocorrem no Brasil. Nove famílias e 25 gêneros de Anura e apenas uma espécie de Gymnophiona foram registradas. As áreas de Cerrado apresentaram a maior riqueza, seguidas pelas florestas estacionais semideciduais, Chaco e veredas. Em relação às macrorregiões consideradas, foram registradas 56 espécies para a planície de inundação (majoritariamente representada pelo Pantanal) e 92 para o planalto. As regiões mais amostradas em Mato Grosso do Sul são o Parque Nacional da Serra da Bodoquena e algumas áreas do Pantanal, o que resulta em lacunas de amostragens por todo o estado, principalmente nas regiões norte, leste/sudeste e parte do oeste/sudoeste. Foram identificadas áreas consideradas importantes para amostragens em decorrência da complexidade de fitofisionomias e relevos encontrados no estado: Maciço do rio Apa, Serra de Maracaju, grande parte do Chaco brasileiro, leque do rio Taquari, planalto do Taquari-Itiquira, escarpas do planalto Central, áreas úmidas/florestas estacionais da planície do rio Paraná e áreas interioranas do Pantanal, onde o acesso é difícil.

ABSTRACT Amphibian species from Mato Grosso do Sul state were compiled and checked for the first time. A total of 97 species were recorded, representing more than 10% of the known Brazilian amphibians. Twenty five anuran genera in nine families and only one caecilian were recorded. Cerrado was the richest area followed by semideciduous forests, Chaco, and veredas (palm swamps). It was recorded 56 species in floodplain areas and 92 in the surrounding cerrados. The Serra da Bodoquena National Park and some Pantanal areas were better sampled than other regions in Mato Grosso do Sul, resulting in sampling gaps across the state. Important areas for future inventories were identified: mountainous regions in Serra de Maracaju and those bordering the Apa river, Brazilian chacoan areas, the Taquari river alluvial fan, the broad Taquari-Itiquira flat area, the Central Plateau scarps, wet areas from Parana river bordering São Paulo state, and inland areas from Pantanal.

AbstractThe aim of this study was to evaluate the influences of variables of preparation on total flavonoids content from extractive solution of Lippia sidoides Cham., Verbenaceae. Thus a 23 factorial design was used to study the importance of plant proportion, the extraction method and solvent on the extraction of flavonoid. The methodology of determination of chemicals in factorial design was validated according to the parameters required by Brazilian Health Agency. The extraction solution was selected through a full factorial design where the best conditions to achieve the highest content of flavonoids were: 7.5% (w/v) of plant with ethanol 50% (v/v) as solvent. The polyphenols content was determined by LC method and its relationship with the antioxidant and free radical scavenging activities was evaluated. The free radical scavenging activities and antioxidant potentials were determined for different concentrations using various in vitro models. Our results indicate that extracts exhibited a significant dose-dependent antioxidant effect as evaluated by TRAP/TAR assays. Besides, we observed an antioxidant activity against hydroxyl radicals and nitric oxide, and protection against lipid peroxidation in vitro. Our results suggest that the extract presents significant in vitro antioxidant potential indicating promising perspectives for its use as pharmaceutical/or food additive.

Foi avaliada a ação de pastas formuladas com hidróxido de cálcio P.A. e diferentes veículos (solução fisiológica - pasta A e óleo de Copaifera langsdorffii Desfon - pasta B) sobre microrganismos bucais e formação de ponte dentinária em cães. A ação antimicrobiana das pastas e de seus componentes individuais foi avaliada pela técnica da concentração inibitória mínima pela diluição em ágar. Os materiais foram diluídos e avaliados contra quinze cepas padrão de microrganismos relacionados a doenças endodônticas. Os microrganismos foram cultivados e, após a incubação, os dados foram analisados (Anova One-Way, Tukey, P≤0,05). Para a avaliação da formação de ponte dentinária, quatro incisivos superiores de dez animais foram tratados, sendo dois capeados com pasta A (GA) e dois com a pasta B (GB). Após 90 dias, os dentes foram extraídos para obtenção de cortes histológicos, com o objetivo de se avaliar o grau de formação de ponte dentinária. Os dados obtidos foram submetidos à análise estatística (Kruskal-Wallis, P<0,05). Segundo a ação antimicrobiana, os materiais foram classificados em ordem decrescente: hidróxido de cálcio P.A., pasta A, pasta B e óleo de Copaifera langsdorffii Desfon, com diferenças estatisticamente significantes entre o hidróxido de cálcio P.A., as pastas e os veículos. Ao final do estudo, observou-se que as duas pastas avaliadas foram semelhantes quanto à formação de ponte dentinária. Considerando-se os microrganismos selecionados, conclui-se que as pastas avaliadas foram semelhantes entre si em termos de potencial antimicrobiano, diferindo dos componentes individuais. Em relação à formação de ponte dentinária, não foram observadas diferenças significantes entre os dentes tratados com as diferentes pastas.

This study analysed the effect of pastes formulated with calcium hydroxide P.A. and different vehicles (saline solution - paste A and Copaifera langsdorffii Desfon oil - paste B) on oral microorganisms and dentin bridge formation in dogs. The antimicrobial action of the pastes and their components was analysed by the minimum inhibitory concentration in agar gel technique. The components were diluted and tested on fifteen standard strains of microorganisms associated with endodontic diseases. The microorganisms were cultivated and after incubation data was analysed using One-Way ANOVA and Turkey's test (P≤0.05). Four superior incisors of ten animals were used to evaluate dentin bridge formation. Two incisors were capped with paste A (GA) and two with paste B (GB). After 90 days, the teeth were extracted for histological analysis and the degree of dentin bridge formation evaluated. Data was analysed by the Kruskal-Wallis test (P<0.05). The pastes and their components were classified in the following decreasing order of antimicrobial action: calcium hydroxide P.A., paste A, paste B and Copaifera langsdorffii Desfon oil. Calcium hydroxide P.A. showed significantly higher antimicrobial action than the pastes or their vehicles. No significant difference was observed between the two pastes in dentin bridge formation. Based on the microorganisms studied, it can be concluded that the pastes analysed showed similar antimicrobial potential but differed significantly from their individual components. No significant difference was observed in dentin bridge formation between the different pastes tested.
