Abstract Background Most of the evidence about the impact of the post-acute COVID-19 Syndrome (PACS) reports individual symptoms without correlations with related imaging. Objectives To evaluate cardiopulmonary symptoms, their predictors and related images in COVID-19 patients discharged from hospital. Methods Consecutive patients who survived COVID-19 were contacted 90 days after discharge. The Clinic Outcome Team structured a questionnaire evaluating symptoms and clinical status (blinded for hospitalization data). A multivariate analysis was performed to address the course of COVID-19, comorbidities, anxiety, depression, and post-traumatic stress during hospitalization, and cardiac rehabilitation after discharge. The significance level was set at 5%. Results A total of 480 discharged patients with COVID-19 (age: 59±14 years, 67.5% males) were included; 22.3% required mechanical ventilation. The prevalence of patients with PACS-related cardiopulmonary symptoms (dyspnea, tiredness/fatigue, cough, and chest discomfort) was 16.3%. Several parameters of chest computed tomography and echocardiogram were similar in patients with and without cardiopulmonary symptoms. The multivariate analysis showed that PACS-related cardiopulmonary-symptoms were independently related to female sex (OR 3.023; 95% CI 1.319-6.929), in-hospital deep venous thrombosis (OR 13.689; 95% CI 1.069-175.304), elevated troponin I (OR 1.355; 95% CI 1.048-1.751) and C-reactive protein during hospitalization (OR 1.060; 95% CI 1.023-1.097) and depression (OR 6.110; 95% CI 2.254-16.558). Conclusion PACS-related cardiopulmonary symptoms 90 days post-discharge are common and multifactorial. Beyond thrombotic and markers of inflammation/myocardial injury during hospitalization, female sex and depression were independently associated with cardiopulmonary-related PACS. These results highlighted the need for a multifaceted approach targeting susceptible patients.
Resumo Fundamento A maioria da evidência sobre o impacto da síndrome COVID pós-aguda (PACS, do inglês, post-acute COVID-19 syndrome) descreve sintomas individuais sem correlacioná-los com exames de imagens. Objetivos Avaliar sintomas cardiopulmonares, seus preditores e imagens relacionadas em pacientes com COVID-19 após alta hospitalar. Métodos Pacientes consecutivos, que sobreviveram à COVID-19, foram contatados 90 dias após a alta hospitalar. A equipe de desfechos clínicos (cega quanto aos dados durante a internação) elaborou um questionário estruturado avaliando sintomas e estado clínico. Uma análise multivariada foi realizada abordando a evolução da COVID-19, comorbidades, ansiedade, depressão, e estresse pós-traumático durante a internação, e reabilitação cardíaca após a alta. O nível de significância usado nas análises foi de 5%. Resultados Foram incluídos 480 pacientes (idade 59±14 anos, 67,5% do sexo masculino) que receberam alta hospitalar por COVID-19; 22,3% necessitaram de ventilação mecânica. A prevalência de pacientes com sintomas cardiopulmonares relacionados à PACS (dispneia, cansaço/fadiga, tosse e desconforto no peito) foi de 16,3%. Vários parâmetros de tomografia computadorizada do tórax e de ecocardiograma foram similares entre os pacientes com e sem sintomas cardiopulmonares. A análise multivariada mostrou que sintomas cardiopulmonares foram relacionados de maneira independente com sexo feminino (OR 3,023; IC95% 1,319-6,929), trombose venosa profunda durante a internação (OR 13,689; IC95% 1,069-175,304), nível elevado de troponina (OR 1,355; IC95% 1,048-1,751) e de proteína C reativa durante a internação (OR 1,060; IC95% 1,023-1,097) e depressão (OR 6,110; IC95% 2,254-16,558). Conclusão Os sintomas cardiopulmonares relacionados à PACS 90 dias após a alta hospitalar são comuns e multifatoriais. Além dos marcadores trombóticos, inflamatórios e de lesão miocárdica durante a internação, sexo feminino e depressão foram associados independentemente com sintomas cardiopulmonares relacionados à PACS. Esses resultados destacaram a necessidade de uma abordagem multifacetada direcionada a pacientes susceptíveis.

Synthesis and Evaluation of the Antileishmanial Activity of Novel Eugenol Analogs Containing 1,2,3-Triazole Fragments against Intracellular Leishmania braziliensis

This investigation describes the synthesis of eugenol analogs presenting 1,2,3-triazole fragments and evaluation of their antileishmanial activity. The alkylation of guaiacol (1) with allyl bromide afforded 1-(allyloxy)-2-methoxybenzene (2) (93% yield). The Claisen rearrangement conducted with 1 gave ortho eugenol (3) (82% yield). Alkylation procedures performed with 3 produced 1-allyl-3-methoxy-2-(prop-2-yn-1-yloxy)benzene (4) (73% yield) and 1-allyl-3-methoxy-2-(pent-4-yn-1-yloxy)benzene (6) (53% yield). The copper(I)-catalyzed alkyne-azide cycloaddition (CuAAC) reactions involving alkynes 4 and 6 with different benzylic azides afforded twenty-two eugenol analogs with 1,2,3-triazole functionalities (48-93% yield). We screened the compounds at 10 μmol L 1 against Leishmania braziliensis intracellular amastigotes during macrophage infection. The action of these compounds was compared with the known leishmanicidal drug amphotericin B. None of the analogs were toxic to macrophages at 10 μmol L-1. The cytotoxic concentration at 50% (CC50), effective concentration at 50% (EC50), and selectivity index (SI) were determined to the best compounds 4-((2-allyl-6-methoxy)phenoxymethyl)-1-(4-chlorobenzyl)-1 H-1,2,3-triazole (8c) and 4-((2-allyl-6-methoxy)phenoxymethyl)-1-(4-trifluoromethoxybenzyl)-1 H-1,2,3-triazole (8h). They showed a significant leishmanicidal effect, with EC50 of 28.09 µmol L-1 (8c) and 52.03 µmol L-1 (8h). The SIs were 9.7 for 8c and > 5.7 for 8h. These compounds have the potential as new leishmanicidal agents against L. braziliensis and may represent a starting point for the development of alternative treatments for cutaneous leishmaniasis.

Resumo Barragens são umas das principais ameaças para peixes potamódromos Neotropicais, causando bloqueios de rotas migratórias e perdas de áreas de reprodução em um número crescente de sistemas de drenagem. Neste cenário, dados atuais defendem que a manutenção de tributários de fluxo livre em bacias hidrográficas fragmentadas seria uma importante estratégia na conservação de peixes migradores, tanto em situações de panmixia quanto em cenários com diferentes estoques coexistindo, tal como já relatado para Prochilodus lineatus. Este estudo analisou locos microssatélites e haplótipos mitocondriais em amostras de P. lineatus obtidas em diferentes anos ao longo da bacia do rio das Cinzas, no reservatório de Capivara (maior reservatório na calha principal do rio Paranapanema, alto rio Paraná) e no rio Tibagi, todos conectados por trechos livres, visando investigar as contribuições da bacia do rio das Cinzas na dinâmica populacional de peixes migradores na região do reservatório de Capivara. Ambos os marcadores detectaram altos níveis de diversidade genética e indicaram diferentes estoques de P. lineatus ao longo da bacia do rio das Cinzas, sugerindo alguma estruturação genética temporal, bem como a mistura destes estoques na amostra do reservatório de Capivara e de outros pontos, corroborando as relações de estoques neste reservatório e seus tributários. Abstract Dams are one of the main threats to Neotropical potamodromous fish, causing blockages of migratory routes and loss of breeding areas in an increasing number of drainage systems. In this scenario, current data argue that the maintenance of free-flowing tributaries along fragmented watersheds would be an important strategy to the migratory fish conservation, both in panmixia situations and in scenarios with different coexisting stocks, as already reported for Prochilodus lineatus. This study analyzed microsatellite loci and mitochondrial haplotypes in samples of P. lineatus obtained in different years along the Cinzas River basin, as well as in the Capivara Reservoir (largest Reservoir in the Paranapanema main channel, upper Paraná River) and in the Tibagi River, all connected by free stretches, aiming to investigate the contributions of the Cinzas River basin to the population dynamics of migratory fish in the Capivara reservoir region. Both markers detected high genetic diversity levels and indicated different genetic stocks of P. lineatus along the Cinzas River basin, suggesting some temporal genetic structuring, as well as the mixture of these stocks in the sample from the Capivara reservoir and from other points, corroborating the relationships among stocks in this reservoir and its tributaries. systems scenario freeflowing flowing conservation P largest channel stretches region structuring points

Abstract ZnO-NPs can be obtained through various methods, resulting in nanoparticles with different size and morphology, which directly influences their antimicrobial potential. The objective of this work was to evaluate the antibacterial activity of ZnO-NPs obtained by a solochemical process against important human foodborne pathogens: Staphylococcus aureus, Salmonella Typhimurium, Bacillus cereus and Pseudomonas aeruginosa. ZnO-NPs were identified as nanorods with the length between 90.1 and 100 nm (10.5 % frequency), the diameter between 80.1 and 90 nm (21 % frequency), and wurtzite type crystalline structure. The Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) were equal to 0.05 mg mL-1 and 0.5 mg mL-1 for S. aureus and S. Typhimurium, respectively, lower than previous results related in the literature. ZnO-NPs produced by solochemical method had a superior antibacterial activity. For instance, they can be incorporated in packaging materials for increasing microbial safety and food shelf-life by inhibiting bacterial growth.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

OBJETIVO: Uma elevada incidência de enteroparasitoses é encontrada em regiões urbanas do nordeste brasileiro. As infecções por Giardia lamblia têm sido relacionadas com aumento da prevalência de alergias cutâneas e gastrointestinais. Contudo, ainda existe pouca informação sobre a associação entre a giardíase e doenças alérgicas das vias aéreas. Diante disso, o presente estudo se propôs a verificar a relação entre a infecção por G. lamblia em crianças de área urbana e reatividade alérgica respiratória. MÉTODOS: O estudo incluiu 110 pacientes, de ambos os sexos e idades, entre 5 e 15 anos. Os pacientes foram submetidos a questionários de avaliação dos sintomas clínicos, testes cutâneos de leitura imediata e exames coproparasitológicos e sorológicos. RESULTADOS: Foi verificada uma frequência elevada de crianças infectadas por G. lamblia (45%, 50/110). A infecção pelo protozoário não foi associada com maior risco de alergias respiratórias (p = 0,075), elevação de IgE total (p = 0,701), IgE específica (p = 0,250) ou teste cutâneo positivo para diferentes alérgenos ambientais (p = 0,239). CONCLUSÃO: O estudo demonstrou que a presença dos sintomas de asma, atopia cutânea e marcadores sorológicos não foram associados com a presença de infecção pela G. lamblia nessa amostra de crianças.
OBJECTIVES: There is a high incidence of intestinal parasite infection in urban areas in the Northeast of Brazil. Giardia lamblia infections have been associated with increased prevalence of cutaneous allergies and gastrointestinal disorders. However, little is known about the relationship between giardiasis and allergic diseases of the airways. The present study aimed to investigate the possible association between respiratory allergic diseases and infections by G. lamblia in children from urban areas. METHODS: This study recruited 110 patients of both sexes aged 5-15 years. Patients were administered a questionnaire evaluating clinical symptoms and were given skin tests, parasite tests and serum tests. RESULTS: A high incidence of G. lamblia was observed (45%, 50/110). Infections by this protozoan were not associated with increased risk of respiratory allergy (p = 0.075), high total IgE levels (p = 0.701), positive specific IgE tests (p = 0.250), or positive skin tests for a range of environmental allergens (p = 0.239). CONCLUSION: This study demonstrated that symptoms of asthma, skin allergy and serum markers were not associated with G. lamblia infections in this sample of children from urban areas.

Multimetric index for assessing ecological condition of running waters in the upper reaches of the Piabanha-Paquequer-Preto Basin, Rio de Janeiro, Brazil

The objective of this study was to develop a rapid multimetric index using benthic macrofauna as bioindicators of ecological conditions for the upper reaches of the Piabanha-Paquequer-Preto Basin located in the state of Rio de Janeiro, Brazil. A total of 33698 macroinvertebrates were collected in 27 sites. Benthic macroinvertebrates were sampled using the multi-habitat method that consists in sampling 20 m² of substrate collected in approximate proportion to the representation of all major habitat types in the reach. A subsampling procedure was used. The main steps followed to develop and test the index were: exclusion of unsuitable metrics using box-whisker plots, considering the degree of overlapping among interquartile limits (1º-3º) and confirmed by Mann-Whitney U test (p < 0.05) using six reference rivers and six impaired rivers. From all the candidate potential metrics, 36 were considered suitable. After identifying redundant metrics, through Spearman correlation analysis, and considering practical application criteria, six core metrics were selected to compose the Piabanha-Paquequer-Preto Multimetric Index (PPPMI) using the continuous method: Family richness, Shannon-Wiener family diversity, EPT family richness, %Diptera, %MOLD (Mollusca + Diptera) and %Collectors. The sensitivity of the index was tested in 15 rivers using a Principal Components Analysis (PCA) of the six environmental variables. The first axis of the PCA was highly correlated to the PPPMI scores (r = 0.703, p < 0.001). The PPPMI responded to a set of environmental variables associated to a gradient of human disturbance affecting the ecological condition of the waterbodies. This indicates that the PPPMI is an effective tool for biological monitoring and decision making in the hydrographic region of the Piabanha-Paquequer-Preto rivers.

OBJETIVO: Realizar uma análise comparativa do teste de caminhada de seis minutos em crianças e adolescentes saudáveis em pistas de diferentes metragens (30,5 e 20 metros). MÉTODOS: Foram avaliados 67 voluntários de escolas públicas de uma cidade de uma região metropolitana, com idades de 7 a 14 anos, sendo 36 meninos e 31 meninas. Todos foram submetidos a quatro testes de caminhada, sendo dois em cada uma das pistas. As variáveis analisadas foram: distância caminhada, trabalho de caminhada, pressão arterial média (PAM), frequência cardíaca (FC) e saturação de oxigênio (SaO2). Para análise estatística, foi utilizado ANOVA One-Way para medidas repetidas para um p<0,05. RESULTADOS: Na comparação entre os testes realizados em cada pista e na comparação do melhor teste entre pistas, não foram observadas diferenças significativas para a PAM, FC e SaO2. A distância caminhada não foi estatisticamente diferente entre os dois testes realizados em cada pista. Entretanto, na comparação entre pistas nos testes de melhor desempenho, os voluntários caminharam distâncias significativamente maiores na pista de 30,5 metros (p<0,05). Porém, esse aumento foi inferior a 10%. Quanto à sobrecarga cardíaca e ao trabalho de caminhada, não foram observadas diferenças significativas entre pistas. CONCLUSÕES: Apesar das diferenças encontradas na distância caminhada entre pistas, ela foi menor que 10% sem variações importantes nos outros parâmetros avaliados. Portanto, a pista de 20 metros se adequou aos critérios de reprodutibilidade descritos na literatura para a população do presente estudo.
OBJECTIVE: To perform a comparative analysis of the six-minute walk test in healthy children and adolescents in corridors of 30.5m (100 feet) 20m (65.6 feet) in length. METHODS: We evaluated 67 participants (36 boys and 31 girls), aged 7 to 14 years old, from public schools of a city in a metropolitan area. All were submitted to four walking tests, two in each of the corridors. The variables analyzed were: walked distance, work rate, mean blood pressure, heart rate and oxygen saturation. Statistical analysis was performed using one-way ANOVA for repeated measures and significance level at p<0.05. RESULTS: The comparison between the tests in each corridor and between the best tests in the different corridors did not show significant differences in the blood pressure, heart rate and oxygen saturation. The walked distance was not statistically different in the two tests on each corridor. However, the participants covered greater distances on the 30.5m corridor (p<0.05) compared to the best test between corridors. However, this increase was less than 10%. Regarding the cardiac overload and the work rate, there were no significant differences between the corridors. CONCLUSIONS: There were differences in walked distance between the corridors, however they were less than 10% with no significant changes in the other measured parameters. Therefore, the 20m corridor had a good reproducibility for the population of this study.
