RESUMO A família Callichthyidae compreende um grupo monofilético bem corroborado, dividida em duas subfamílias: Corydoradinae e Callichthyinae. Uma proposta recente, baseada em dados moleculares, sugeriu que a subfamília Corydoradinae é composta por nove linhagens, possivelmente gêneros. As espécies pertencentes a cada uma destas linhagens possuem extensivas modificações no tamanho do genoma, incluindo espécies diplóides, tetraplóides e octaplóides. Considerando a ocorrência dessas diferentes linhagens e que as extremas variações em conteúdo de DNA podem implicar em alterações significativas na estrutura dos espermatozoides, o presente estudo analisou a morfologia do sistema reprodutor masculino e a morfometria da cabeça dos espermatozoides de representantes das nove linhagens de Corydoradinae, procurando características particulares em cada uma. Os dados morfológicos revelaram a ocorrência de grande variação dentro das linhagens, maior que aquela observada entre espécies de diferentes linhagens. Diferentemente, os dados morfométricos obtidos para oito das nove linhagens revelaram grande congruência com a atual proposta para Corydoradinae. Estes resultados demonstram que há correlação entre as variações em conteúdo de DNA e o tamanho da cabeça dos espermatozoides, fornecendo, assim, subsídio adicional para a definição das nove linhagens de Corydoradinae.

ABSTRACT Callichthyidae comprises a well-corroborated monophyletic group divided into two subfamilies: Corydoradinae and Callichthyinae. A recent proposal, based on molecular data, suggests that Corydoradinae is composed by nine monophyletic lineages, possibly genera. The species pertaining to those lineages have extensive modification in the size of genome, including diploid, tetraploid and octoploid species. Considering the occurrence of these monophyletic lineages and that the variations in DNA content may imply in significant alterations on the structure of spermatozoa, this study analyzed the morphology of the male reproductive system and the morphometry of the head of the spermatozoa of representatives of the nine lineages of Corydoradinae, seeking for particular characteristics of each lineage. Morphological data revealed a high intra-lineage variation, larger than that observed among species of different lineages. In contrast, morphometric data obtained for eight out of the nine lineages, revealed large congruency with the hypothesis that Corydoradinae is composed by different lineages. These results demonstrate that there is a correlation among variations in DNA content and the size of the spermatozoon head, thus providing additional subsides for the definition of the Corydoradinae lineages.

Em Corydoradinae, a presença de espermátides junto com espermatozóides no lúmen dos túbulos testiculares sugere uma espermatogênese do tipo semicística, enquanto que em Callichthyinae a produção do esperma ocorre inteiramente dentro dos espermatocistos no epitélio germinativo, caracterizando a espermatogênese cística. A espermiogênese em Callichthyinae é caracterizada por um desenvolvimento inicial lateral do flagelo, pela presença de rotação nuclear em diferentes graus, formação de uma fossa nuclear excêntrica ou medial, formação de um canal citoplasmático, e presença de migração centriolar, sendo mais similar à espermiogênese do tipo I. Em Corydoradinae, a espermiogênese é caracterizada pelo desenvolvimento excêntrico do flagelo, ausência de rotação nuclear, fossa nuclear excêntrica, formação de um canal citoplasmático, e ausência de migração centriolar, diferindo dos tipos descritos previamente. O processo de espermatogênese e espermiogênese em Corydoradinae e Callichthyinae revelaram caracteres únicos para cada subfamília, corroborando a hipótese de que as mesmas constituem grupos monofiléticos. Em relação à ultraestrutura do esperma, a análise comparativa das espécies de Callichthyidae mostra que as características gerais encontradas nos espermatozóides foram similares, reforçando a hipótese de monofilia da família.

In Corydoradinae, the presence of spermatids in the lumen of the testicular tubules together with spermatozoa suggests that spermatogenesis is of the semicystic type, whereas in Callichthyinae, sperm production occurs entirely within spermatocysts in the germinal epithelium, characterizing cystic spermatogenesis. Spermiogenesis in Callichthyinae is characterized by an initial lateral development of the flagellum, the presence of nuclear rotation to different degrees, an eccentric or medial formation of a nuclear fossa, formation of a cytoplasmic channel, and presence of centriolar migration, being more similar to type I spermiogenesis. In Corydoradinae, spermiogenesis is characterized by eccentric development of the flagellum, the absence of nuclear rotation, an eccentric nuclear fossa formation, formation of a cytoplasmic channel, and absence of centriolar migration, differing from the types previously described. The process of spermatogenesis and spermiogenesis in Corydoradinae and Callichthyinae revealed unique characters for each of these subfamilies, corroborating the hypotheses that they constitute monophyletic groups. In relation to sperm ultrastructure, the comparative analysis of the callichthyid species shows that the general characteristics found in the spermatozoa were similar, thus, reinforcing the hypothesis that the family is monophyletic.

A proposição foi realizada, no período pós-operatório imediato, em 40 ratos Wistar, distribuídos por sorteio em doi grupos: grupo NC, vinte ratos correspondentes ao grupo controle, não diabético, submetidos a operação simulada e o grupo PT, 20 ratos correspondentes ao grupo diabético que recebeu transplante de pâncreas heterotópico de ratos Wistar normais. Durante sete dias, antes do transplante, e 1, 3, 6, 12, 24, 48, 72, 96 horas após, determinava-se a glicose sanguínea, a insulina plasnática e o glucagon. Estes parâmetros eram obtidos também do grupo NC. Diabetes mellitus experimental era induzida pela administração intravenosa de aloxana. O grupo PT era imunosuprimido com ciclosporina A. O grupo NC apresentou níveis normais de glicose sanguínea, de insulina plásmica e de glucagon, durante todo o experimento. Foi encontrada nítida hiperinsulinemia no sangue venoso periférico do grupo PT. A insulina plasmática era significantemente maior no grupo PT comparada ao grupo NC começando 72 horas após o transplante. O glucagon plasmático, elevado no período pré-transplante, não se alterou após o transplante. Apesar de hiperinsulinemia e hiperglucagonemia, os níveis de glicose sanguínea eram elevados 6 horas após o transplante e mantiveram-se normais após este período. Considerando-se os níveis de glicose sanguínea 12 horas pós-transplante, não houve diferença estatisticamente significante entre os grupos PT e NC, até o sacrifício.

The consequences of the bypass of the portal venous effluent into the systemic circulation on the blood glucose, plasma insulin, and glucagon in immediate postoperative period of rats submitted to pancreas transplantation were studied. Forty outbred male Wistar rats were randomly assigned to two experimental groups: group NC included 20 non-diabetic control rats, submitted to simulated operating (sham-operated), and group PT included 20 diabetic rats that received heterotopic pancreas transplantation from normal donor Wistar rats. For 7 days prior and 1, 3, 6, 12, 24, 48, 72 and 96 hours after transplantation blood glucose, plasma insulin, and glucagon were recorded. These parameters were also concurrently recorded for NC rats. Diabetes was induced by i.v. alloxan administration; PT rats were immunosuppressed with cyclosporin A. NC rats presented normal values of blood glucose, plasma insulin, and glucagon over the course of experiment. A marked hyperinsulinemia was found in peripheral venous blood of PT rats, being plasma insulin significantly higher than that for NC rats (P<0.01) beginning 72- h after transplant. The plasma glucagon, elevated in pre-transplant period, did not change after transplant. Despite hyperinsulinemia and hyperglucagonemia, the blood glucose levels were elevated up to 6- h after transplant, but were within normal levels following this period. Beginning at 12- h after transplant the blood glucose levels observed in PT rats did not differ significantly to NC rats until the sacrifice.
