ABSTRACT Purpose: To investigate the role and mechanism of β1,3-N-acetylglucosaminyltransferase-3 gene (B3GNT3) in esophageal cancer (ESCA). Methods: The starBase database was used to evaluate the expression of B3GNT3. B3GNT3 function was measured using KYSE-30 and KYSE-410 cells of esophageal squamous cell carcinoma (ESCC) cell lines. The mRNA levels were detected by quantitative real-time polymerase chain reaction (qRT-PCR). Cell counting kit-8, clone formation assay and transwell assay were used to detect the changes of proliferation, invasion and migration. Results: B3GNT3 expression was higher in ESCA tissues than in normal tissues. The overall survival rate of ESCA patients with high B3GNT3 expression was lower than that of ESCA patients with low B3GNT3 expression. In vitro functional experiments showed that the proliferation ability, migration and invasion ability of KYSE-30 and KYSE-410 cells with B3GNT3 interference were lower than those of the control, and the overexpression of B3GNT3 had the opposite effect. After silencing B3GNT3 expression in ESCC cell lines, the growth of both cell lines was inhibited and the invasiveness was decreased. Knockdown of B3GNT3 reduced the growth rate and Ki-67 expression level. Conclusions: B3GNT3, as an oncogene, may promote the growth, invasion and migration of ESCC cell.

Abstract Objective To explore the molecular mechanism of neuropathologic damage induced by radiofrequency ablation at different temperatures. Methods This is basic research, and 36 SD rats were used to construct the neuropathological injury model. The rats were subjected to radiofrequency stimulation at different temperatures and were divided into 6 groups according to the temperature injury: 42°, 47°, 52°, 57°, 62°, and 67°C groups. Conduction time, conduction distance, and nerve conduction velocity were recorded after temperature injury. HE-staining was used to observe the histopathological morphology of the sciatic nerve. The expression of SCN9A, SCN3B, and NFASC protein in sciatic nerve tissue were detected by western blot. Results With the increase in temperature, nerve conduction velocity gradually decreased, and neurons were damaged when the temperature was 67°C. HE-staining showed that the degrees of degeneration of neurons in rats at 47°, 52°, 57°, 62°, and 67°C were gradually increased. The expression of SCN9A, SCN3B protein in 57°, 62°, 67°C groups were much higher than that of NC, 42°, 47°, 52°C groups. However, the expression of NFASC protein in 57°, 62°, 67°C groups was much lower than that of the NC, 42°, 47°, 52°C groups. Conclusion There was a positive correlation between temperature caused by the radiofrequency stimulation to neuropathological damage. The mechanism is closely related to the expression of SCN9A, SCN3B, and NFASC protein in nerve tissue caused by heat transfer injury.

Abstract Zinc is one of the most important trace elements and deficiency can lead to significant clinical complications. Microbial transformation is one of the most important methods used to supplement organic zinc and there may be some naturally occurring microorganisms that have both higher zinc tolerances and transformation capabilities. In this study, microorganisms which displayed such characteristics were widely screened, and a strain designated T-A was found. Morphological and molecular determination demonstrated that T-A is closely related to Candida tropicalis with 87% of 18S rDNA homology, with a G+C content measured by RP-HPLC of 88.42%. Candida tropicalis sp. T-A strains were shown to grow and transform zinc best at pH5 in medium with a zinc ion concentration of 8000 μg/mL at 28 °C for 121 hours. Our results showed C. tropicalis sp. T-A’s zinc content was 19.153 mg/g dry weight, with a relative bioavailability in rats of 173% when compared with traditional zinc supplements. Our study suggests that C. tropicalis sp. T-A could be exploited for use as a potential zinc supplement.

ABSTRACT Cabbage is one of the most popular vegetables that is rich in sugars contributing to flavor and consumer acceptance; however, little information is available on the effect of genotypes and growing conditions on sugar accumulation. We assessed the seasonal variation in agronomic characteristics and free sugar content in cabbage (Brassica oleracea L. var. capitata L.) grown under open field conditions. Seventy-five cabbage genotypes were grown in the spring and autumn 2019, and their morphological characteristics and sugar concentrations were evaluated. Six cabbage types produced predominantly round-shaped heads (64 genotypes). Bright green and green were the dominant colors depending on the outer and inner leaves of the cabbage head. The most variable quantitative trait was head weight in both spring (36.9%) and autumn (49.2%). Glucose was the predominant sugar in most genotypes in both seasons. Mean glucose content in spring and autumn was 209.3 and 214.9 mg g-1 with 14.8% and 14.6% variation, respectively. Most genotypes produced higher fructose concentrations in fall than in spring. Sucrose levels showed the highest variation in both spring (36.0%) and autumn (60.5%), followed by fructose and glucose levels. Most agronomic parameters exhibited nonsignificant or negative correlations with sugar content, except the correlation of head height and width with glucose content. Fructose and glucose were positively correlated with total sugar content, while there was nonsignificant correlation with sucrose. Genotypes 160330 and 183701 produced comparably high and stable total sugar content in both seasons and can therefore be used as commercial breeding materials. We revealed a significant effect of genotype and season on cabbage agronomic characteristics and sugar content.

O estudo buscou investigar a associação entre consumo de frutas e verduras e placa carotídea (PC) e espessura íntima-média carotídea (EIMC), dois preditores de aterosclerose entre adultos das áreas urbana e rural com alto risco de desenvolver doenças cardiovasculares (DCVs) em uma região da China. Foram identificados 11.392 adultos com alto risco de DCV, entre 71.511 indivíduos da população geral, em um estudo transversal entre novembro de 2015 e maio de 2016 na Província de Jiangsu. Entre esses 11.392 participantes de alto risco, a prevalência de PC foi de 36,7%. As variáveis independentes, ou seja, frequências de consumo de frutas e verduras, foram avaliadas através de um questionário de frequência alimentar. As variáveis de desfecho, EIMC e PC, foram medidas por ultrassom. A análise ANCOVA não mostrou associação entre valores de EIMC e frequências de consumo de frutas e verduras. Foram introduzidos modelos de regressão logística multivariada para examinar a associação entre consumo de frutas e verduras e PC. Depois de ajustar para potenciais fatores de confusão, as ORs para participantes com consumo eventual e diário de verduras para qualquer PC foram 0,67 (IC95%: 0,58-0,78) e 0,70 (IC95%: 0,62-0,79), respectivamente, comparado com aqueles com consumo raro de verduras. Enquanto isso, as ORs ajustados foram 0,77 (IC95%: 0,64-0,92) e 0,80 (IC95%: 0,68-0,94), separadamente, para adultos com consumo eventual e diário de verduras para desenvolver uma PC única, comparado aos que relatavam consumo raro de verduras. Entretanto, não foi observada uma associação significativa entre consumo de frutas e PC. Entre a população chinesa com alto risco de DCV, o consumo de verduras frescas mostrou associação negativa com o risco de desenvolvimento de placa carotídea.

However, no significant association between fruit consumption and CP was observed. Among the Chinese population at high CVD risk, consumption of fresh vegetables was negatively associated with the risk of developing carotid plaque.

El objetivo de este estudio fue investigar la asociación del consumo de frutas y verduras con la placa carotídea (PC) y el grosor íntima-media carotídeo (GIMC), dos predictores de la aterosclerosis carotídea en adultos urbanos y rurales, con alto riesgo de desarrollar enfermedades cardiovasculares (ECV) en una región de China. The Carbon Tracker system will play a major role in understanding CO2 sinks and sources, gas exchange between the atmosphere and oceans, and gas emissions from forest fires and fossil fuels in Latin America and the Caribbean. This paper discusses the trends in carbon fluxes in the biosphere and ocean, as well as emissions from forest fires and fossil fuel use in the above-mentioned region, using the Carbon Tracker (CT) system. From 2000 to 2009, the mean carbon fluxes for the biosphere, fossil fuel use, wildfires and the ocean in Latin America and the Caribbean were -0.03, 0.41, 0.296, -0.061 Pg C/yr, respectively, and -0.02, 0.117, 0.013, -0.003 Pg C/yr, respectively, in Mexico. The mean net carbon flux for Latin America and the Caribbean was 0.645 Pg C/yr, and 0.126 Pg C/yr for Mexico. The terrestrial sinks in Latin America and the Caribbean are dominated by the forest, agricultural, grass and shrub regions, as well as the Andes mountain range and the net surface-atmosphere fluxes including fossil fuel are dominant in regions around large cities in Mexico, Brazil, Chile, and areas undergoing deforestation along the Amazon River. The results confirm that forest fires are an important source of CO2 in Latin America and the Caribbean. In addition, we can confirm that policies encouraging the use of ethanol in light vehicles in Brazil have helped to decrease carbon emissions from fossil fuel, and assume the effects of the Proárbol program on carbon sinks from the biosphere and from fire emissions sources in Mexico. Based on this analysis, we are confident that the CT system will play a major role in Latin America and the Caribbean as a scientific tool to understand the uptake and release of Co2 from terrestrial ecosystems, fossil fuel use and the oceans, and for long-term monitoring of atmospheric CO2 concentrations.

El sistema rastreador de carbono desempeñará un papel importante para mejorar la comprensión sobre las fuentes y sumideros de CO2, el intercambio de gases entre la atmósfera y el océano, y las emisiones de gases provenientes de incendios forestales y combustibles fósiles en América Latina y el Caribe. El objetivo del presente estudio es analizar las tendencias de los flujos de carbono en la biosfera y el océano, así como las emisiones procedentes de incendios forestales y el uso de combustibles fósiles en dicha región, mediante el sistema rastreador de carbono. De 2000 a 2009 el flujo promedio de carbono para la biosfera, el uso de combustibles fósiles, los incendios y el océano en América Latina y el Caribe fue de -0.03, 0.41, 0.296 y -0.061 Pg C/yr, respectivamente, y en México fue de -0.02, 0.117, 0.013 y -0.003 Pg C/yr, respectivamente. El promedio del flujo neto de carbono en América Latina y el Caribe, por un lado, y México, por el otro, fue de 0.645 y 0.126 Pg C/yr, respectivamente. En América Latina y el Caribe, los sumideros terrestres de carbono se encuentran principalmente en las regiones boscosas y agrícolas, así como en los pastizales, zonas de arbustos y en la cordillera de los Andes. Los flujos netos entre la superficie y la atmósfera que incluyen al combustible fósil son predominantes en los alrededores de las grandes ciudades de México, Brasil y Chile, al igual que en las zonas de deforestación en el río Amazonas. Se ha comprobado que los incendios forestales son una importante fuente de CO2 en América Latina y el Caribe. Por otra parte, podemos confirmar que la política de fomento al uso de etanol en vehículos livianos en Brasil ha contribuido a disminuir las emisiones de carbono generadas a partir de combustible fósiles, y suponer el efecto del programa Proárbol en México como resultado de la absorción de carbono proveniente de la biosfera y de incendios forestales. Este estudio confirma que el sistema rastreador de carbono puede cumplir un papel importante en Latinoamérica y el Caribe como herramienta científica para comprender mejor los procesos de absorción y liberación de carbono derivados de los ecosistemas terrestres, del uso de combustibles fósiles y del océano, además de permitir una vigilancia a largo plazo de las concentraciones de CO2 en la atmósfera.

Establishing an acceptable user-centred electronic health record (EHR) system is a challenging task for healthcare providers due to the need for such systems to meet the requirements of its user population. Concerned nurses are the main end-users of EHR systems. Based on knowledge of evidence-based management (EBM) and the issues (goals and methods) of Health Information Systems (HIS) evaluation, this research was performed in four regional teaching hospitals by adopting a quantitative approach research design to perform "goal-based evaluation" research. The results of Path Analysis indicated that 17 of 21 hypotheses were accepted in this study. In addition, the results of One-way ANOVA with Scheff test comparisons indicated that Age is the most important variable in measuring System Quality, Service Quality, Safety Quality, User Use and User Satisfaction; Education is important in measuring Service Quality; and Seniority is important in measuring System Quality, Service Quality and User Use. Furthermore, the results of Linear regression indicated that only one hypothesis is affected by the demographic variable Education. In summary, this empirical investigation provided evidence-based knowledge to explain nurses’ opinions of EHR systems success, and to distinguish which demographic variables influence their viewpoints of using such systems in southern Taiwan.
