RESUMO. A eficiência do uso da água (UEA) foi proposta como uma alternativa para mitigar os efeitos das mudanças climáticas na agricultura e reduzir a pressão sobre os recursos hídricos, principalmente em espécies com altos requerimentos de água que são cultivadas sob condições de sequeiro. O objetivo deste estudo foi caracterizar o UEA instantâneo, integral e molecular de quatro cultivares de feijão, contrastando sua resposta em condições de água limitadas para comparar os mecanismos componentes desta característica entre cultivares tolerantes à seca. Os resultados indicaram que os cultivares tolerantes aumentaram seu UEA instantâneo em comparação aos suscetíveis; entretanto, houve diferença entre as cultivares, uma vez que Pinto Villa apresentou maior condutância estomática e taxa de transpiração, levando a um custo maior de água para produção de sementes do que Pinto Saltillo. Além disso, os genes EUA de ycf2, rrn16, rpoC2, hardy, ndhK, erecta e ycf1 foram apenas sobreexpressos em Pinto Saltillo sob condições de água limitadas, o que se revelou ser a cultivar mais eficiente do UEA. Portanto, os mecanismos componentes da UEA são diferentes mesmo entre as cultivares tolerantes à seca e os mecanismos pelos quais as cultivares tolerantes aumentaram sua UEA instantânea e integral foram diferentes.

ABSTRACT. The water-use efficiency (WUE) has been proposed as an alternative to mitigate the effects of climate change in agriculture and to reduce pressure on water resources, mainly in species with high water requirements which are grown under rainfed conditions. The aim of this study was to characterize the instantaneous, integral and molecular WUE of four bean cultivars contrasting in their response under limiting water conditions to compare the component mechanisms of this trait between drought tolerant and susceptible cultivars. Results indicated that tolerant cultivars increased their instantaneous WUE in comparison to susceptible ones; however, there was a difference between cultivars since Pinto Villa had a higher stomatal conductance and transpiration rate, leading to a higher water cost to produce seed than Pinto Saltillo. Furthermore, ycf2, rrn16, rpoC2, hardy, ndhK, erecta and ycf1 WUE genes were only overexpressed in Pinto Saltillo under limited water conditions, which turned out to be the most WUE efficient cultivar. Therefore, the component mechanisms of WUE are different even between drought tolerant cultivars and the mechanisms by which the tolerant cultivars increased their instantaneous and integral WUE were different.

Among the main factors affecting bean production is the wrong varietal distribution over diverse environmental conditions under which it is grown. The research work was aimed at evaluating the agro-morphological behavior based on the variability characterization of promising common bean lines for late sowings. Thus, seeding was performed on January 20, 2014, using a randomized block design and four repetitions in “El Mulato” farm, belonging to "Orlando Cuellar" Strengthened Credit and Service Cooperative (SCSC), San José de las Lajas municipality, Mayabeque province. The evaluation was carried out on 14 agro-morphological variables that included phenological and morphological parameters, yield and its components, as well as rust (Uromyces appendiculatus) resistance. Results from analyzing statistical parameters and main components allowed detecting genetic variability among the evaluated lines. It was also detected that pod number per plant and 100-grain weight were the highest correlated variables with yield. In general, six out of the evaluated lines combined a high-yielding potential and from mid to high rust resistant reaction, standing out SCR 15 line, with a greater yield potential than those so far reported in Cuba. This suggests assessing these lines in regional trials for a further generalization-extension of the possible cultivars that contribute to focus an appropriate management of these genotypes and their use as rust resistant sources in bean breeding programs

Entre los principales factores que afectan la producción de frijol se encuentra la mala distribución de variedades para las diversas condiciones ambientales en que se cultiva. El objetivo de este trabajo fue evaluar el comportamiento agro-morfológico, considerando la caracterización de la variabilidad de líneas de frijol común promisorias para siembras tardías. La siembra se realizó el 20 de enero de 2014 con un diseño de bloques al azar y cuatro repeticiones en la finca “El Mulato”, perteneciente a la Cooperativa de Créditos y Servicios Fortalecida (CCSF) “Orlando Cuellar”, en el municipio San José de las Lajas, Mayabeque. La evaluación se realizó usando 14 variables agro-morfológicos que incluyeron parámetros fenológicos, morfológicos, rendimiento y sus componentes, y resistencia a la roya (Uromyces appendiculatus). Los resultados del análisis de parámetros estadísticos y componentes principales, permitió detectar la variabilidad genética entre las líneas evaluadas. Además, se detectó que las variables de mayor correlación con el rendimiento fueron el número de vainas por plantas y la masa de 100 granos. De forma general, seis de las líneas evaluadas combinan alto potencial de rendimiento y reacción entre intermedia y altamente resistente ante la roya, destacándose la línea SCR 15, con rendimiento potencial muy superior a los reportados en Cuba hasta la fecha. Esto sugiere evaluar estas líneas en ensayos regionales para una posterior generalización-extensión de los posibles cultivares que contribuyan a enfocar un manejo adecuado de estos genotipos y su empleo como fuentes de resistencia a la roya en los programas de mejoramiento genético del frijol

Abstract The pinto bean type is in high demand by consumers in the North-Central Mexico. The new variety 'San Rafael' derived from crosses of the two-parent Pinto Bayacora by Pinto Saltillo conducted in 2004 in the Bajio Experimental Station of the National Institute of Forestry, Agriculture and Livestock described. The selection process 'San Rafael' was carried out under irrigated conditions and time in the Bajio region and in a highland village in northern Guanajuato. The plant 'San Rafael' is indeterminate habit prostrate short guide, short crop cycle photoperiod neutral reaction (can be planted any time of year and latitude without its cycle is changed). San Rafael presents resistance to major leaf diseases that occur in the plateau, such as halo blight, anthracnose and rust. The grain is of type pinto, oval, medium size, with tolerance dimming (long shelf life), grain type of high commercial value. For its neutral response to photoperiod, 'San Rafael' allow their exploitation in different time windows, expanding the supply of fresh bean in the year. 'San Rafael' suits production systems under irrigation and temporary in the highlands and the northwest region. It defined its crop cycle and plant size, this variety can be used in closed systems furrows in double rows.

Resumen El frijol de tipo pinto es de alta demanda por los consumidores del Centro-Norte de México. Se describe la nueva variedad ‘San Rafael’derivada de la cruza biparental de Pinto Bayacora por Pinto Saltillo realizada en 2004 en el Campo Experimental Bajío del Instituto Nacional de Investigaciones Forestales, Agrícolas y Pecuarias. El proceso de selección de ‘San Rafael’ se llevó a cabo bajo condiciones de riego y temporal en la región del Bajío y en una localidad del altiplano en el Norte de Guanajuato. La planta de ‘San Rafael’ es de hábito indeterminado postrado de guía corta, ciclo de cultivo corto de reacción neutral al fotoperiodo (puede sembrarse en cualquier época del año y latitud sin que se modifique su ciclo). San Rafael presenta resistencia a las principales enfermedades foliares que ocurren en el Altiplano, como son tizón de halo, antracnosis y roya. El grano es de tipo pinto, forma ovalada, tamaño mediano, con tolerancia al oscurecimiento (larga vida de anaquel), tipo de grano de alto valor comercial. Por su respuesta neutral al fotoperíodo, ‘San Rafael’ permitirá su explotación en diferentes ventanas de tiempo, ampliando así la oferta de frijol fresco en el año. ‘San Rafael’ se adapta a los sistemas de producción bajo riego y temporal en el altiplano y la región del noroeste. Por lo definido de su ciclo de cultivo y tamaño de planta, esta variedad se puede utilizar en sistemas de surcos cerrados a doble hilera.

The responsible agent of rust in common bean (Phaseolus vulgaris L.) is U. appendiculatus, a pathogen that reduces bean production in temperate localities with humidity periods of 10 h at least. In México rust causes yield losses up to 80 %. Resistance of common bean to U. appendiculatus is regulated by at least 11 genes, namely Ur-3, Ur-4, Ur-5, Ur-7, Ur-9, Ur-11, Ur-12, Ur-Ouro Negro, plus two more unnamed genes. These genes confer resistance to several rust races depending on the combination present, which provide evidence that genes are arranged into clusters and that resistance is race-specific. The aim of our research was to identify the rust resistance genes contained in a rust-resistant common bean genotype ('PTB 08005') and in two rust-susceptible varieties ('Pinto Maverick' and 'Pinto Fresnillo'), in order to determine the specific gen combination conferring resistance to the U. appendiculatus races prevalent in the state of Guanajuato, México. The genes identified in the resistant variety, but not in the susceptible ones, were Ur-3, Ur-4, Ur-7 and Ur-11. In addition, six experimental lines of Pinto bean type were also tested to determinate their potential resistance to the fungus. The experimental line 117 had the same resistance genes as genotype 'PTB 08005'.

U. appendiculatus es el agente causal de la roya del frijol (Phaseolus vulgaris L.), enfermedad que limita su producción en climas templados y con periodos húmedos de al menos 10 h. En México la roya ha provocado disminuciones de hasta 80 % en el rendimiento. La resistencia del frijol a la infección por U. appendiculatus está regulada por al menos 11 genes denominados Ur-3, Ur-4, Ur-5, Ur-6, Ur-7, Ur-9, Ur-11, Ur-12, Ur-Ouro Negro, más otros dos aún no denominados, los cuales confieren resistencia a múltiples razas en función de la combinación presente, lo cual indica que están organizados en grupos y que el tipo de resistencia es específica de razas. En esta investigación se identificaron los genes de resistencia a U. appendiculatus en un genotipo de frijol resistente ('PTB 08005') y dos susceptibles ('Pinto Maverick' y 'Pinto Fresnillo') con el fin de determinar la combinación específica de genes que confiere resistencia a las razas de este hongo presentes en el Estado de Guanajuato, México. Los genes presentes en la variedad resistente, pero no en las susceptibles, fueron Ur-3, Ur-4, Ur-7 y Ur-11. Adicionalmente se identificó la presencia de genes de resistencia en seis líneas experimentales de frijol Pinto con el fin de determinar su potencial de resistencia al patógeno. La línea 117 tuvo el mismo potencial de resistencia que el genotipo 'PTB 08005'.

The fungi Colletotrichum gloeosporioides, is the agent that causes anthracnose and the main limitation to the harvest of commercial quality Hass avocado (Persea americana). The genetic diversity of the population of this fungus has been studied in several parts of the world, yet in Mexico, its population structure, and especially the components of its diversity, are unknown. The genetic, pathogenic and morphological diversity was determined for 21 monoconidial strains of Colletotrichum gloeosporioides from Michoacán, Mexico. The genetic diversity was estimated by means of random amplified DNA polymorphism, and by determining the profiles of isoenzymes of each monoconidial strain. Pathogenic diversity was measured with the invasive capacity of avocado pulp and the growth rate in vitro was determined by comparing the characteristics of the mycelia of each monoconidial strain. Results showed a high diversity of Colletotrichum gloeosporioides with a genetic variability index of 0.3031, and the most accurate estimator was the random amplified DNA polymorphism (genetic variability index= 0.344) and isoenzyme profile (genetic variability index= 0.35). The typical characteristics were white mycelia, cotton-like consistency, white colony, random mycelia growth and short hyphae on the growth border. There was no relation between the pathogenicity measured as the invasive capacity of the pulp and this growth rate in vitro. The group analyses determined that the relations between the strains are established based mainly on the origin and, in a secondary manner, on the symptom in the fruit.

El hongo Colletotrichum gloeosporioides, es el agente causal de antracnosis y representa la principal limitante para la cosecha de frutos con calidad comercial de aguacate Hass (Persea americana). La diversidad genética poblacional de este hongo se ha estudiado en diversas partes del mundo, pero en México se desconoce su estructura poblacional y particularmente los componentes de su diversidad. Se determinó la diversidad genética, patogénica y morfológica de 21 cepas monoconidiales de Colletotrichum gloeosporioides de Michoacán, México. La diversidad genética se estimó por medio del polimorfismo de ADN amplificado al azar y por la determinación de los perfiles de isoenzimas de cada cepa monoconidiales. La diversidad patogénica se midió mediante la capacidad invasiva de pulpa de aguacate y por la tasa de crecimiento in vitro. La diversidad morfológica in vitro se determinó por la comparación de caracteres del micelio de cada cepa monoconidiales. Los resultados mostraron alta diversidad de Colletotrichum gloeosporioides con un índice de variabilidad genética de 0.3031 y los estimadores más precisos fue el polimorfismo de ADN amplificado al azar (índice de variabilidad genética igual a 0.344) y perfiles de isoenzimas (índice de variabilidad genética igual a 0.35). Las características típicas fueron micelio blanco, consistencia algodonosa, colonia blanca, crecimiento de micelio al azar e hifas cortas en el borde de crecimiento. No se encontró relación entre la patogenicidad medida como capacidad invasiva de pulpa y la velocidad de crecimiento in vitro. Los análisis de agrupamiento establecieron que las relaciones entre cepas se dan principalmente con base en la localidad de origen y de manera secundaria, en el síntoma producido en el fruto.

Maize landraces from three regions: Sierra Norte, Valles Centrales and Costa in the state Oaxaca, were sampled looking for transgenic sequences. The strategies to accomplish this goal were: 1) search for 35 S promoter of the caulifl ower mosaic virus by qualitative PCR, 2) immunological detection of phosphinotricin acetyl transferase (PAT) enzyme activity, which confers resistance to the herbicides glufosinate, 3) detection of resistant plants to glufosinate, 4) detection of the transgene bar by PCR, 5) assessment of transgenic content by quantitative PCR and 6) sequencing of specific amplified fragments. The use of the first technique allowed for the detection of the 35S promoter sequence in plants from five plots from the Sierra Norte region, in the district of Ixtlan. The 35S promoter was not detected in samples from the other two regions. Immunological detection for the phosphinotricin acetyl transferase protein was negative for all samples analyzed. Twenty-three plants out of 1435 were resistant to the herbicide glufosinate; however, none showed amplification of the bar transgene. Out of fi ve initial samples only three were confi rmed positive for 35S promoter by quantitative PCR and sequencing. The 35S positive samples did not test positive for the bar transgen; therefore, the detected 35S promoter sequence might have a different origin as well as the herbicide resistance present in these maize landraces from the region Sierra Norte.

Se analizó maíz criollo de tres regiones representativas de producción del estado de Oaxaca (Sierra Norte, Valles Centrales y Costa), con el fin de establecer la presencia de secuencias transgénicas. Las estrategias empleadas fueron: 1) búsqueda del promotor 35S del virus del mosaico de la coliflor por medio de PCR cualitativo, 2) detección inmunológica de la actividad de la enzima fosfinotricin acetil transferasa (PAT) que confiere resistencia a herbicida 3) detección de resistencia a la aplicación de glufosinato en plantas, 4) detección molecular del transgen bar, mismo que codifica la proteína fosfinotricin acetil transferasa, 5) determinación del contenido transgénico de las muestras positivas por PCR en tiempo real y 6) secuenciación de las bandas específicas de amplificación. Como resultado de la primera estrategia se detectó la secuencia del promotor 35 S en plantas de cinco parcelas de maíz de la Sierra Norte, pertenecientes al distrito de Ixtlán. Este promotor no se detectó en el resto de las muestras colectadas de las regiones Valles Centrales y Costa. La detección inmunológica de la proteína PAT fue negativa en todas las muestras analizadas. De 143 5 plantas sometidas a la aplicación de glufosinato, 23 resistieron la actividad del herbicida; sin embargo, ninguna evidenció amplificación del fragmento del transgen bar. Las cinco plantas inicialmente positivas para PCR cualitativo se redujeron a tres muestras positivas por PCR cuantitativo y secuenciación. En conclusión, las muestras amplifi cadas para el promotor 35 S no evidenciaron presencia ni actividad del gen bar; sin embargo, podría ser otro el origen del promotor 3 5 S y la naturaleza de la resistencia al glufosinato en los maíces criollos de la Sierra Norte.
