ABSTRACT Reserva Ecológica de Guapiaçu (REGUA) is a private, protected area inside a remnant of the Atlantic Forest with high biodiversity. Although the bats of the reserve have been sampled for more than two decades, few studies have been published about them. Based on a compilation of data from several surveys, we present an updated list of the bat species there and compare it with surveys from other locations in the Atlantic Forest. From August 1998 to September 2021, at least 194 sampling nights were carried out at REGUA by different research groups from various institutions, totaling 448,092 m2.h of sampling. A total of 4,069 individuals were captured, belonging to 47 species and six families. Additionally, our results indicate that it is possible that some species that occur at REGUA have not been recorded yet. REGUA has the greatest number of bat species known for the Atlantic Forest. This most likely results from the fact that the reserve includes large areas of mature, continuous forest connected with other protected areas in the State of Rio de Janeiro. Another factor contributing to the high diversity of bat species at REGUA is that the area has been intensely sampled for many years. Given that the bat assemblage there appears to be a good proxy to ascertain the ecological patterns of biodiversity in well-preserved forests, we consider REGUA to be an important area for long-term ecological research. The basic knowledge about the ecological interactions of bats with different food resources and zoonotic microorganisms offers a unique opportunity to carry out research in several areas of knowledge, making it possible to address questions about bat assemblage structure, bat-parasite ecology, competition, niche partitioning, and other related studies.

Resumo Introdução: A audiometria de altas frequências pode contribuir para a detecção precoce de alterações auditivas causadas por medicações ototóxicas. No tratamento dos pacientes com fibrose cística, existem muitos fármacos ototóxicos amplamente utilizados. A detecção precoce de alterações auditivas deve permitir que sejam identificadas antes que o dano atinja as frequências da fala. A lesão causada pela ototoxicidade é irreversível, traz importantes consequências sociais e psicológicas. Nas crianças, a perda auditiva, mesmo restrita às altas frequências, pode afetar o desenvolvimento da linguagem. Objetivo: Investigar a eficácia e a efetividade do monitoramento da audição por meio da audiometria de altas frequências em pacientes pediátricos com fibrose cística. Método: Foram consultadas as bases de dados eletrônicas PubMed, Medline, Web of Science e Lilacs, de janeiro a novembro de 2015. Foram selecionados os estudos em que foi feita audiometria de altas frequências em pacientes com fibrose cística em tratamento com medicamentos ototóxicos e publicados em português, inglês e espanhol. Para a avaliação da qualidade metodológica dos artigos optou-se pelo uso do sistema Grade. Resultados: No processo de busca feito de janeiro de 2015 a novembro de 2015 foram encontradas 512 publicações, 250 na PubMed, 118 na Medline, 142 na Web of Science e dois na Lilacs. Desses, foram selecionados nove artigos. Conclusão: Identificou-se a ocorrência de perda auditiva em altas frequências em pacientes com fibrose cística sem queixas auditivas. Admite-se que audiometria em altas frequências possa ser um método de diagnóstico precoce a ser recomendado para investigação auditiva de pacientes em risco de ototoxicidade.

Abstract Introduction: High-frequency audiometry may contribute to the early detection of hearing loss caused by ototoxic medications. Many ototoxic drugs are widely used in the treatment of patients with cystic fibrosis. Early detection of hearing loss should allow known harmful drugs to be identified before the damage affects speech frequencies. The damage caused by ototoxicity is irreversible, resulting in important social and psychological consequences. In children, hearing loss, even when restricted to high frequencies, can affect the development of language. Objective: To investigate the efficacy and effectiveness of hearing monitoring through high-frequency audiometry in pediatric patients with cystic fibrosis. Methods: Electronic databases PubMed, MedLine, Web of Science and LILACS were searched, from January to November 2015. The selected studies included those in which high-frequency audiometry was performed in patients with cystic fibrosis, undergoing treatment with ototoxic drugs and published in Portuguese, English and Spanish. The GRADE system was chosen for the evaluation of the methodological quality of the articles. Results: During the search process carried out from January 2015 to November 2015, 512 publications were identified, of which 250 were found in PubMed, 118 in MedLine, 142 in Web of Science and 2 in LILACS. Of these, nine articles were selected. Conclusion: The incidence of hearing loss was identified at high frequencies in cystic fibrosis patients without hearing complaints. It is assumed that high-frequency audiometry can be an early diagnostic method to be recommended for hearing investigation of patients at risk of ototoxicity.

A artrite-encefalite caprina (CAE) é diagnosticada rotineiramente pela técnica de imunodifusão em gel de agarose (IDGA), que é considerada pouco sensível. Objetivou-se com este estudo padronizar testes de Elisa-i e Western Blot (WB) para diagnóstico precoce de anticorpos em caprinos contra CAEV e comparar os resultados obtidos nesses testes com a prova de IDGA. Para a padronização dos testes Elisa-i e WB, utilizaram-se diferentes concentrações e diluições de antígeno, soros e conjugado. No Elisa-i, adotaram-se microplacas rígidas com 96 poços, sendo a combinação de concentração de 0,5µg/poço de antígeno e diluições de 1:100 de soro e 1:1500 de conjugado a que apresentou melhor resultado. No WB foram utilizadas membranas de nitrocelulose, definindo-se as diluições de 1:50 de soro e 1:15000 de conjugado. Para avaliar o desempenho das técnicas, 222 amostras de soro caprino foram testadas e os dados obtidos foram comparados com o IDGA. A sensibilidade e a especificidade do Elisa-i/IDGA, WB/IDGA e WB/Elisa-i foram de 70% e 91%, 100% e 72,6%, 84,6% e 76,5%, concomitantemente. O índice Kappa desses testes foi de 0,35, 0,2 e 0,36, respectivamente. As técnicas de Elisa-i e WB apresentaram-se mais sensíveis que a IDGA, podendo ser utilizadas como ferramentas para o diagnóstico precoce da CAE.

Caprine arthritis-encephalitis (CAE) is routinely diagnosed with the Agarose Gel Immunodiffusion (AGID) technique, which is considered to have low sensitivity. The objective of this study was to standardize testing i-Elisa and Western Blot for early detection of antibodies against CAEV in goats and compare the results obtained in these tests with proof of AGID. For standardization of i-Elisa and WB, different concentrations and dilutions of antigen, sera and conjugate were used. In the i-Elisa, rigid microplate with 96 wells was adopted, and the combination that showed the best result was a concentration of 0.5µg/ well of antigen and dilutions of the serum of 1:100 and conjugate of 1:1500. In the WB nitrocellulose membranes were used, and the dilutions of the serum were defined at 1:50 and conjugate at 1:15000. To evaluate the performance of the techniques, 222 goat serum samples were tested and the data were compared with the AGID. The sensitivity and specificity of Elisa-i/IDGA, WB/AGID and WB/Elisa-i were 70% and 91%, 100% and 72.6%, 84.6% and 76.5%, concomitantly. The Kappa index of these tests was 0.35, 0.2 and 0.36, respectively. The i-Elisa and WB techniques were more sensitive than the AGID and can be used as tools for early diagnosis of CAE.

RESUMO O Vírus da Artrite Encefalite Caprina (CAEV) e Vírus Maedi-visna (MVV) pertencem ao gênero Lentivírus da família Retroviridae. São considerados geneticamente distintos, mas antigenicamente relacionados. O objetivo desde trabalho foi isolar o vírus da CAE de um animal oriundo de um rebanho do Rio Grande do Norte e positivo pelo teste de Imunodifusão em Gel de Agarose (IDGA) através do co-cultivo de leucócitos infectados do sangue periférico em Membrana Sinovial Caprina (MSC). Dezesseis caprinos da raça Saanen, com suspeitas clínicas para CAE foi testado por IDGA e Western Blotting. Para o isolamento viral, os leucócitos do sangue periférico foram isolados por co-cultivo em MSC. Monócitos/Macrófagos coletados foram inoculados em monocamadas pré-formadas em garrafas A25. O resultado do IDGA foi positivo para um animal, confirmado por Western Blotting. Após 50 dias de co-cultivo, foi realizada a coloração da monocamada com cristal de violeta a 0,1% para visualização do ECP e realizada nested-PCR do sobrenadante do co-cultivo, com confirmação do efeito citopático viral. A cepa isolada, denominada BrRN-CNPC.G1 foi considerada o primeiro isolamento do CAEV no Estado do Rio Grande do Norte. Esse estudo permitirá em breve, realizar a caracterização molecular do genoma do vírus isolado, através da análise de seus diferentes genes estruturais e comparar com outras sequencias virais isoladas para identificar a provável origem da infecção desse animal e estabelecer as possíveis divergências entre cepas padrões de Lentivírus e cepas regionais circulantes.

ABSTRACT Caprine Arthritis Encephalitis virus (CAEV) and Visna/maedi virus (VISNA), belong to the genus Lentivirus family Retroviridae. The aim of the present study was to isolate the CAE virus from a positive goat when tested by agarose gel immunodifusion (AGID) by cocultivation techniques of infected peripheral blood leukocytes in goat synovial membrane (GSM). In this study, one flock of 16 goats, obtained from a flock of Rio Grande do Norte, Brazil, with clinical suspicions for CAEV, was screened using agar-gel immunodiffusion (agid) and Western blotting. The result was positive for one animal, confirmed by Western blotting. For virus isolation, peripheral blood leukocytes (PBL) were isolated from the blood for co-culture on goat sinovial membrane (GSM) cells. Monocytes/macrophages collected were inoculated in a monolayer of 90% semiconfluent cells in the A25 culture bottles. After 50 days of co-culture, the monolayer was stained with crystal violet 0.1% for viewing viral cytopathic effects (CPE) characteristic of viruses, and nested-PCR was performed with the supernatant of co-culture for confirmation of CPE. The isolate, named BrRN-CNPC.G1, was considered the first isolation of small ruminant lentivirus from a naturally infected goat in a flock in Rio Grande do Norte, Brazil. This study will soon allow for the molecular characterization of the isolated virus genome, by analysis of its different structural genes and their comparison with other isolated viral sequences to identify the likely source of infection of that animal and establish the possible differences between strains of lentiviruses and circulating regional strains.

Toxoplasmose é uma das mais comuns zoonoses parasitárias do mundo. Infecções em seres humanos e em animais variam nas diferentes áreas geográficas influenciadas pelas condições ambientais. A soroprevalência da infecção por Toxoplasma gondii em bovinos no Brasil varia de 1,03 a 71,0%. O estudo transversal foi realizado em 58 de um total de 453 propriedades na região Sul Fluminense do estado do Rio de Janeiro. Vacas leiteiras acima de 3 anos de idade (n=589) foram selecionadas para coleta de sangue e a detecção de anticorpos anti-T. gondii foi feita pelo teste de imunofluorescência indireta (IFI) com titulação inicial de 1:16 e títulos > 64 foram considerados positivos. Após análise univariada dos fatores de risco, gatos em contato com bovinos, em contato com a água de beber dos animais e o número de gatos foram associados com a soroprevalência de T. gondii. A regressão logística demonstrou que o número maior de gatos (>3) teve um risco duas vezes maior (p=0,0138) que propriedades que tinham um número menor de gatos (1-2). Em contraste, a presença de galinhas foi considerada um fator de proteção (p=0,025).

Toxoplasmosis is one of the most common parasitic zoonoses throughout the world. Infection in man and animals varies in different geographical areas influenced by many environmental conditions. Seroprevalence of Toxoplasma gondii infection in cattle in Brazil ranges from 1.03 to 71%. A cross-sectional survey was carried out in 58 out of 453 farms in the South Fluminense Paraiba Valley, State of Rio de Janeiro, Brazil. Over 3-year-old cattle (n=589) from dairy herds were selected for blood collection and detection of anti-T. gondii antibodies by indirect immunofluorescence reaction (IFA) with initial titration of 1:16; titers > 64 were considered positive. Univariate analysis of risk factors showed that cats in contact with cattle, cats in contact with drinking water, and number of cats were associated with T. gondii seroprevalence. Logistic regression revealed a two-fold increased risk for infection of cattle (p=0.0138) through larger number of cats (>3) compared with low numbers of cats (1-2) on the farm. In contrast, the presence of chickens was considered a protective factor (p=0.025).

Foram utilizados 112 leitões com peso inicial de 28,68kg, distribuídos em blocos ao acaso, em esquema fatorial 8×2 (oito tratamentos x dois sexos), sete repetições e dois animais por baia. As dietas foram formuladas com base no fósforo total, sendo estas à base de milho e de farelo de soja. O fósforo foi suplementado utilizando-se os fosfatos: bicálcico (FBC); monobicálcico (MBC); supertriplo (ST); supersimples (SS); rocha Catalão (ROCHA); mistura de fontes (MIST) e ácido fosfórico (AcF); e dieta-controle, sem o fósforo suplementar (CONT). Avaliou-se previamente a composição química de amostras de fontes de fósforo disponíveis no mercado. Os fosfatos MBC, FBC e AcF apresentaram baixo nível de contaminação por metais pesados. Observou-se variação no teor de metais pesados entre as marcas comerciais de supertriplo. O consumo do fosfato resultou em acúmulo de cobre nos músculos. A dieta-controle provocou acúmulo de chumbo e zinco no fígado, e a mistura de fontes e supertriplo ocasionou acúmulo de cádmio no fígado. A utilização de fontes alternativas de fósforo menos elaboradas influenciou a deposição de minerais no fígado e nos músculos dos suínos.

One hundred twelve pigs averaging weight of 28.68kg, were allotted to a completely randomized blocks with seven replicates of two animals per box. The treatments consisted in factorial 8×2 (eight phosphorus sources x two sexes). The experimental diets were formulated based on corn and soybean meal and the phosphorus was supplemented through phosphate dicalcium (FBC), monodicalcium (MBC), triple superphosphate (ST), simple superphosphate (SS), Catalão rock (ROCHA), mixture of sources (MIST), and phosphoric acid (AcF) in addition to a control diet without supplemental phosphous (CONT). Samples of phosphorus sources available in the market were evaluated for particle size and chemical composition. The MBC, FBC and AcF showed a low level of heavy metal contamination, but a variation in the heavy metal composition for ST was observed. Swines fed on CONT diet showed lead and zinc accumulation in the liver. Swines fed on triple superphosphate and mixture of sources diets showed cadmium accumulation in the liver. Swine fed on ST diet showed copper accumulation in the muscles. Swine diets formulated with alternative and less elaborated sources of phosphorus caused liver and muscles deposition of minerals.

Hydroxyurea is used for sickle-cell disease patients in order to increase fetal hemoglobin synthesis and consequently decrease the severity of pain episodes. Fetal hemoglobin, which is formed by gamma-globin chains A and G, is present in a constant composition throughout fetal development: about 75% of Ggamma and 25% of Agamma. In contrast, adult red cells contain about 40% of Ggamma and 60% of Agamma. In the present study, we analyzed the effect of hydroxyurea induction on the gamma chain composition of fetal hemoglobin in 31 sickle-cell disease patients treated with hydroxyurea. The control group was composed of 30 sickle-cell disease patients not treated with hydroxyurea in clinical steady state. The patients were older than 13 years and were not matched for age. All patients were seen at Hemocentro/UNICAMP and Boldrini Infantile Center, Campinas, SP, Brazil. The levels of total hemoglobin were significantly higher in patients treated with hydroxyurea (mean ± SD, 9.6 ± 2.16 g/dl) than in untreated patients (8.07 ± 0.91 g/dl). Fetal hemoglobin levels were also higher in treated patients (14.16 ± 8.31%) than in untreated patients (8.8 ± 4.09%), as was the Ggamma/Agamma ratio (1.45 ± 0.78 vs 0.98 ± 0.4, P < 0.005). The increase in the Ggamma/Agamma ratio in patients treated with hydroxyurea suggests the prevalence of a pattern of fetal hemoglobin synthesis, whereas patients not treated with hydroxyurea maintain the adult pattern of fetal hemoglobin synthesis. Because no correlation was observed between the Ggamma/Agamma ratio and total hemoglobin or fetal hemoglobin levels, the increase in Ggamma chain synthesis may not imply a higher production of hemoglobin.

Indivíduos imunocomprometidos infectados com Strongyloides stercoralis podem desenvolver quadros severos de hiperinfecção ou doença disseminada com elevada mortalidade. Pacientes portadores de neoplasias hematológicas tem imunodepressão causada pela doença básica ou pelo seu tratamento. Um estudo prospectivo foi desenvolvido no Hospital de Clínicas de Porto Alegre, entre julho 1994 e julho de 1995. Setenta e dois pacientes (HIV negativos), tiveram 3 amostras de fezes coletadas em diferentes dias e não tinham recebido medicação antihelmíntica recentemente. Larvas, isoladas por método de Baermann modificado, foram encontradas em 6 pacientes, resultando em prevalência de 8,3%. Não foi documentado nenhum episódio de estrongiloidose complicada. O resultado positivo para pesquisa de S. stercoralis estava significativamente associado (p < 0,001) com eosinofilia. O conhecimento das taxas de prevalência e incidência de doença grave é importante para adequar as recomendações para tratamento empírico além da rigorosa pesquisa da estrongiloidose nos pacientes com neoplasias hematológicas.

Immunocompromised individuals infected with Strongyloides stercoralis may develop severe hyperinfection or disseminated disease with high mortality. Patients with hematological malignancies are at risk because of immunodepression produced either by the disease or its treatment. A prospective study was undertaken at the Hospital de Clínicas de Porto Alegre, from July 1994 to July 1995. Seventy-two (HIV negative), had 3 stool samples collected at different days and had not received recent anti-helmintic therapy. Larvae, isolated in a modified Baermann method, were found in 6 patients, with a resultant prevalence of 8.3%. No complicated strongyloidosis was documented. The positive result for S. stercoralis larvae was significantly associated (p < 0.001) with eosinophilia. Knowledge of prevalence figures and incidence of severe disease is important to adequate guidelines for empirical treatment besides the rigorous search for strongyloidosis in patients with hematological malignancies.
