ABSTRACT: Rhipicephalus (Boophilus) microplus is a hard tick endemic in livestock-growing regions and causes economic losses in the largest beef-producing countries, including Brazil, Mexico, Argentina, Australia and Uruguay. The use of chemical acaricides is still the main strategy to control R. microplusinfestations. Nevertheless, immunological control of R. microplus with an anti-tick vaccine is a suitable alternative and has manifold advantages because it can avoid drug-resistance and the presence of acaricide residues in milk, beef and in the environment. Indeed, vaccines based on the Bm86 antigen have had relative commercial and technical success to control R. microplus in some regions. Although, the efficacy of such vaccines varies among tick populations and is insufficient to provide an acceptable level of protection. Therefore, the need to search for better antigens is impelling. This review focused on the restrictions imposed on the use of acaricides in Brazil and in the European Union, as well as on the impacts of Bm86-based vaccines on R. microplus control. The efficacy of experimental anti-tick vaccines (based on subolesin, glutathione S-transferase, ferritin 2; voltage-dependent anion channel; aquaporin, 60 S acidic ribosomal protein, metalloprotease and trypsin) that can elicit an immune response against the physiological functions of various ticks is discussed.

RESUMO: O Rhipicephalus (Boophilus) microplus é um carrapato duro que é endêmico de regiões de pecuária e causa perdas econômicas nos maiores países produtores de carne bovina, incluindo Brasil, México, Argentina, Austrália e Uruguai. O uso de acaricidas ainda é a principal estratégia para controlar infestações por R. microplus. No entanto, o controle imunológico do R. microplus com uma vacina contra carrapatos é uma alternativa adequada e possui diversas vantagens, por evitar a seleção de populações de carrapato resistentes a drogas, evitar a presença de resíduos de acaricidas no leite, na carne e no ambiente. As vacinas baseadas no antígeno Bm86 tiveram relativo sucesso comercial e técnico no controle do R. microplus em diversas regiões. No entanto, a eficácia dessas vacinas varia entre as populações de carrapatos e é insuficiente para fornecer um nível aceitável de proteção. Portanto, há uma necessidade de procurar novos antígenos. Esta revisão foca nas restrições impostas ao uso de acaricidas no Brasil e na União Europeia, bem como nos impactos das vacinas baseadas em Bm86 no controle do R. microplus. Também é discutida a eficácia de vacinas anti-carrapatos experimentais (baseadas em subolesina, glutationa S-transferase, ferritina 2; canal aniônico dependente de voltagem; aquaporina, proteína ribossômica ácida 60S, metaloprotease, tripsina) que podem elicitar uma resposta imune contra as funções fisiológicas de vários carrapatos.

Esta revisão tem como objetivo apresentar os anticoagulantes e inibidores da agregação plaquetária que foram encontrados em animais hematófagos. Esses animais precisam inibir as reações hemostáticas no local onde se alimentam no hospedeiro para realizar a refeição sangüínea e também para manter o sangue fluido nos seus próprios tratos digestivos. Devido a essa necessidade, eles desenvolveram ao longo da evolução uma grande diversidade de substâncias que são injetadas no hospedeiro através da saliva e que permitiram o sucesso de seu parasitismo. Tais recursos farmacológicos podem ser utilizados como ferramentas em pesquisa da fisiologia vascular e hemostática, e têm potencial uso terapêutico em doenças cardiovasculares.

In this review, we present anticoagulants and inhibitors of platelet aggregation isolated from hematophagous animals. These animals have to inhibit, at the site of injury, the host hemostasis in order to blood-feed and maintain the blood fluid inside their digestive tract. During evolution, hematophagous animals developed a diversity of anti-homeostatic substances that are injected into the host through their saliva and that are crucial to successful parasitism. These anti-homeostatic substances could be used as tools in vascular physiology investigation and they also have potential therapeutic applications.

Amostras de contrafilé (músculo L. dorsi ) provenientes de 26 bovinos, sendo 14 Polled Hereford (HH), sete 3/4Hereford 1/4Nelore (3/4H1/4N) e cinco 5/8Hereford 3/8Nelore (5/8H3/8N), machos castrados, abatidos aos dois anos de idade, foram coletadas 24 h após o abate e analisadas quanto à atividade de calpastatina e textura, tanto no 1o dia post mortem quanto após um período de maturação de 10 dias a 2o C. A atividade de calpastatina foi determinada pelo ensaio de inibição da m-calpaína e a textura através da força de cisalhamento (Warner-Bratzler). A carne de novilhos 5/8H3/8N apresentou, no 1o dia, maiores (p<0,05) atividade de calpastatina e força de cisalhamento do que a carne de novilhos HH e 3/4H1/4N. Não foram detectadas diferenças (p>0,05) entre os grupos HH e 3/4H1/4N para as mesmas características. Após 10 dias, houve uma diferença na atividade de calpastatina, porém não significativa (p>0,05), entre o grupo 5/8H3/8N (1,57U/g) e os demais (HH=1,23U/g; 3/4H1/4N=1,35U/g), e diferença significativa entre os grupos HH e 5/8H3/8N para força de cisalhamento (3,67 e 5,00kg, respectivamente). Conclui-se que a atividade de calpastatina determinada 24 h post mortem pode ser útil para a previsão da textura da carne, maturada ou não, em programas de melhoramento genético, e que a participação crescente do genótipo Bos indicus nos rebanhos da Região Sul, a par das conhecidas vantagens zootécnicas, poderá resultar em carne de pior textura.

Boneless rib steaks (L. dorsi muscle) from 26 two years old steers, 14 Polled Hereford, seven 3/4Hereford 1/4Nelore (3/4H1/4N) and five 5/8Hereford 3/8Nelore (5/8H3/8N), were collected 24 hs after slaughter and analysed for calpastatin activity and texture at the 1st day post mortem and at the 10th day of aging at 2o C. Calpastatin activity was determined by m-calpain inhibition assay and texture by shear force (Warner-Bratzler). Beef from 5/8H3/8N steers showed higher (p<0.05) calpastatin activity and shear force values than beef from HH and 3/4H1/4N steers in the 1st day. No differences (p>0.05) were detected in the same traits between groups HH and 3/4H1/4N. After 10 days of aging, there was a difference in calpastatin activity, although non-significant (p>0.05), amongst group 5/8H3/8N (1.57U/g) and the others (HH=1.23U/g; 3/4H1/4N=1.35U/g), and a significant difference in shear force between groups HH and 5/8H3/8N (3.67 and 5.00kg, respectively). It was concluded that the calpastatin activity determined 24 hs post mortem can be useful to predict the texture of beef, aged or not, in genetic improvement programs. Also, the increasing participation of Bos indicus genotype in the South Region herds of cattle, besides the productivity advantages, may result in beef of lower texture quality.
