NMR-Based Metabolomic Screening for Metabolites Associated with Resistance to Meloidogyne javanica in Annona muricata Roots

The resistance of Annona muricata roots to the nematode Meloidogyne javanica was investigated by nuclear magnetic resonance (NMR)-based metabolomics in combination with principal component analysis (PCA). Metabolic changes in roots exposed and not exposed to the nematode were evaluated and compared. In addition, the presence of nematicidal compounds in the root extracts was investigated through in vitro assay against Meloidogyne javanica second-stage juveniles. Plants exposed to nematodes showed significant changes in their metabolism after 24 h. Several resistance-related metabolites, including dopamine, xanthine and aromatic compounds, could be identified in the roots with the joint analysis of 1D/2D NMR and the loadings of PC3 (17.8%). A.muricata root chloroform extract, containing mainly acetogenins, has shown nematostatic activity against M. javanica, suggesting that a pre-formed defense mechanism can support the reported resistance. For the first time, metabolomic studies allowed to identify induced and pre-formed defense mechanisms and their related metabolites in Annona muricata.

Abstract: Here we model and describe the wood volume of Cerrado Sensu Stricto, a highly heterogeneous vegetation type in the Savanna biome, in the state of Minas Gerais, Brazil, integrating forest inventory data with spatial-environmental variables, multivariate regression, and regression kriging. Our study contributes to a better understanding of the factors that affect the spatial distribution of the wood volume of this vegetation type as well as allowing better representation of the spatial heterogeneity of this biome. Wood volume estimates were obtained through regression models using different environmental variables as independent variables. Using the best fitted model, spatial analysis of the residuals was carried out by selecting a semivariogram model for generating an ordinary kriging map, which in turn was used with the fitted regression model in the regression kriging technique. Seasonality of both temperature and precipitation, along with the density of deforestation, explained the variations of wood volume throughout Minas Gerais. The spatial distribution of predicted wood volume of Cerrado Sensu Stricto in Minas Gerais revealed the high variability of this variable (15.32 to 98.38 m3 ha-1) and the decreasing gradient in the southeast-northwest direction.

Biomass smoke COPD has less tomographic abnormalities but worse hypoxemia compared with tobacco COPD

Special attention has emerged towards biomass smoke-induced chronic obstructive pulmonary disease (COPD), providing new knowledge for prevention and therapeutic approach of non-smoker COPD patients. However, the understanding of biomass smoke COPD is still limited and somewhat controversial. The aim of the present study was to compare COPD exclusively caused by tobacco smoking with COPD exclusively caused by environmental or occupational exposures. For this cross-sectional study, COPD patients were recruited from outpatient clinics and formed two groups: non-smoker COPD group (n=16) with exposure to biomass smoke who did not smoke cigarette and tobacco smoker COPD group (n=15) with people who did not report biomass smoke exposure. Subjects underwent pulmonary function tests, thoracic high-resolution computed tomography, 6-min walk test, and sputum induction. The non-smoker COPD group had biomass smoke exposure of 133.3±86 hour-years. The tobacco COPD group smoked 48.5±27.4 pack-years. Women were 62.5 and 66.7%, respectively, of non-smokers and smokers. The non-smoker COPD group showed higher prevalence of dyspnea, lower arterial oxygen tension (PaO2), and lower arterial oxygen saturation (SaO2%) with similar spirometry results, lung volumes, and diffusion capacity. Regarding inflammatory biomarkers, differences were detected in sputum number of lymphomononuclear cells and in sputum concentrations of interleukin (IL)-6 and IL-8 with higher values in the smoker group. Emphysema was more prevalent in the tobacco smoker group, which also showed higher relative bronchial wall thickness and lower lung density by quantitative analysis. Biomass smoke induced more hypoxemia compared to tobacco in COPD patients with similar severity.

RESUMO O linfoma é uma neoplasia maligna caracterizada pela proliferação neoplásica de células originadas de tecido linfoide e corresponde a cerca de 90% das neoplasias hematopoiéticas em cães. Células T reguladoras (Tregs) têm sido alvo de diversas investigações na área da oncologia devido ao potencial de regulação negativa da resposta do sistema imune e à manutenção ativa do mecanismo de imunossupressão tumoral. O objetivo do presente estudo foi a comparação da porcentagem de Tregs no sangue periférico entre cães com linfoma multicêntrico e animais saudáveis e a porcentagem de Tregs no sangue periférico e nos linfonodos de cães com linfoma multicêntrico. Foram utilizados 26 animais: 10 cães saudáveis, como grupo controle (CG), e 16 cães com linfoma multicêntrico, como grupo linfoma (LG). Observou-se maior expressão de Tregs no sangue periférico de cães do LG em comparação ao CG. Entretanto, não foi observada diferença significativa entre as expressões de Treg nos linfonodos e no sangue periférico do LG. Com esses resultados, foi possível concluir que o linfoma multicêntrico apresenta alta expressão de Tregs, tornando-se condição interessante para o estudo de tratamentos capazes de suprimir as células T reguladoras.
ABSTRACT Lymphoma is a malignant tumor characterized by cell proliferation of lymphoid origin and corresponds to 90% of all hematopoietic neoplasms of dogs. Regulatory T cells (Tregs) have been the target of many investigations in oncology due to their potential of down-regulating immune responses, as well as ensuring the maintenance of active mechanisms of tumor suppression. The aims of the present study were to compare the percentage of Tregs in peripheral blood between dogs with multicentric lymphoma and healthy animals, together with the percentage of Tregs in peripheral blood and lymph nodes of dogs with multicentric lymphoma. Twenty-six animals were enrolled in the study: 10 healthy dogs comprised the control group (CG) and 16 dogs with multicentric lymphoma comprised the Lymphoma Group (LG). We observed that dogs in the LG showed a significantly higher Tregs expression in peripheral blood compared to the CG. No significant difference was observed between Tregs expression in lymph nodes and peripheral blood of the LG, however. With these results, it is possible to conclude that multicentric lymphoma is a neoplasm with high Tregs expression, which poses this as a condition of interest when investigating treatments that can suppress Regulatory T cells.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

(Nota sobre a germinação de sementes de Vochysia tucanorum tratadas com reguladores vegetais). O objetivo deste trabalho foi avaliar a resposta germinativa sob luz branca e escuro de sementes de Vochysia tucanorum Mart. tratadas com GA3 e CEPA. Sementes recém-coletadas de uma área de Cerrado foram armazenadas durante 14 dias em duas temperaturas (25 ± 2 °C e 7 ± 1 °C). Após o período de armazenamento, as sementes foram pré-tratadas com água destilada (controle), ácido giberélico (GA3), ácido 2-cloroetilfosfônico (CEPA) e uma mistura de GA3 + CEPA; em seguida, as sementes foram semeadas em placas de Petri sobre papel filtro umedecido com água destilada e colocadas para germinar em escuro e sob luz branca. Os resultados sugerem que as sementes não são fotoblásticas nem dormentes, no entanto um comportamento fotoblástico emerge quando as sementes foram previamente armazenadas em baixa temperatura e embebidas em soluções de GA3 e CEPA. Em geral, não houve diferença entre as temperaturas de armazenamento de 7 °C e 25 °C. A germinação sob luz branca de sementes pré-tratadas com CEPA + GA3 e CEPA foi antecipada, em comparação com o controle de água destilada, sendo que o efeito da mistura CEPA + GA3 foi mais pronunciado do que o de CEPA sozinho. Assim, a taxa de germinação de sementes de V. tucanorum, sob luz branca, pode ser melhorada pela embebição das mesmas em soluções de CEPA ou de CEPA + GA3.
(Note on the germination of Vochysia tucanorum seeds treated with growth regulators). The aim of this work was to evaluate the germination response of Vochysia tucanorum Mart. seeds treated with GA3 and CEPA and germinated under white light or darkness. Newly collected seeds from a Cerrado area were stored for 14 days at two temperatures (25 °C ± 2 and 7 °C ± 1). After the storage period the seeds were pre-treated with distilled water (control), gibberellic acid (GA3), 2-chloroethylphosphonic acid (CEPA) and a mixture of GA3 + CEPA. Following this, the seeds were sown in Petri dishes on filter paper moistened with distilled water and germinated in either darkness or white light. The results suggest that seeds are non-photoblastic and non-dormant, however a photoblastic behavior emerges when the seeds were previously stored at low temperature and imbibed in CEPA and GA3 solutions. In general, there is no difference between the 7 °C and 25 °C storage temperatures. The germination of seeds pre-treated with CEPA and CEPA + GA3 under white light was faster as compared to the distilled water control, and the effect of the CEPA + GA3 mixture was more pronounced than CEPA alone. Thus, the germination rate of V. tucanorum seeds can be improved by treatment with CEPA or CEPA + GA3 under white light.

INTRODUÇÃO: A dispnéia é um sintoma de difícil avaliação, principalmente nas doenças ocupacionais. OBJETIVO: Avaliar a relação entre presença e intensidade de dispnéia crônica, e sua repercussão funcional em ex-trabalhadores com asbestose na avaliação de disfunção e incapacidade. MÉTODO: Escores de dispnéia pelas escalas Medical Research Council modificada, American Medical Association de 1984 e 1993 e Baseline Dyspnea Index foram obtidos em 40 ex-trabalhadores com diagnóstico de asbestose, os quais foram também submetidos a espirometria, medidas da capacidade de difusão pulmonar do monóxido de carbono e testes de exercício cardiopulmonar incremental e submáximo. RESULTADO: Dispnéia esteve presente em 72,5% e 67,5% dos indíviduos de acordo com as escalas do Medical Research Council e American Medical Association de 1984, respectivamente e em apenas 37,5% e 31,6% dos pacientes de acordo com as escalas American Medical Association de 1.993 e Baseline Dyspnea Index. Houve melhor concordância entre as escalas Medical Research Council e American Medical Association de 1993, e American Medical Association de 1984 e American Medical Association de 1993 quando as graduações "ausente" e "leve" foram agrupadas. Não foi observada relação significativa entre dispnéia de acordo com cada uma das escalas e presença de anormalidades funcionais no repouso e/ou exercício. CONCLUSÃO: O nível de concordância entre as escalas de dispnéia varia significativamente em indivíduos com asbestose. Há falta de relação dos índices de dispnéia com variáveis que avaliam disfunção respiratória em repouso e exercício.
BACKGROUND: Dyspnea is a symptom that is difficult to evaluate, especially in occupational diseases. OBJECTIVE: To evaluate the relationship between chronic dyspnea, in its varying degrees of severity, and the functional repercussions for dysfunction or incapacitation in former workers with asbestosis. METHOD: A total of 40 former workers diagnosed with asbestosis were evaluated. Dyspnea scores were determined using the modified Medical Research Council scale, the 1984 and 1993 American Medical Association scales, and the Baseline Dyspnea Index. Spirometry, measurement of diffusion capacity for carbon monoxide and cardiopulmonary exercise tests (incremental and submaximal) were also performed. RESULTS: Based on scores obtained using the Medical Research Council and 1984 American Medical Association scales, respectively, 72.5% and 67.5% of the subjects were classified as dyspneic, compared with 37.5% and 31.6%, respectively, using the 1993 American Medical Association and Baseline Dyspnea Index scales. There was greater concordance between the Medical Research Council and 1993 American Medical Association scales, as well as between the 1984 and 1993 American Medical Association scales, when the categories of "absent" and "mild" were grouped. No significant relation was found between dyspnea, as determined by each of the scales, and functional abnormalities - either at rest or during exercise. CONCLUSION: In individuals with asbestosis, the degree of concordance among the available dyspnea scales varies significantly. There is a real need for dyspnea indices that evaluate respiratory dysfunction at rest and during exercise.

Rounded atelectasis (RA) is a benign and unusual form of subpleural lung collapse that has been described mostly in asbestos-exposed workers. This form of atelectasis manifests as a lung nodule and can be confused with bronchogenic carcinoma upon conventional radiologic examination. The objective of the present study was to evaluate the variation in contrast uptake in computed tomography for the identification of asbestos-related RA in Brazil. Between January 1998 and December 2000, high-resolution computed tomography (HRCT) was performed in 1658 asbestos-exposed workers. The diagnosis was made in nine patients based on a history of prior asbestos exposure, the presence of characteristic (HRCT) findings and lesions unchanged in size over 2 years or more. In three of them the diagnosis was confirmed during surgery. The dynamic contrast enhancement study was modified to evaluate nodules and pulmonary masses. All nine patients with RA received iodide contrast according to weight. The average enhancement after iodide contrast was infused, reported as Hounsfield units (HU), increased from 62.5 ± 9.7 to 125.4 ± 20.7 (P < 0.05), with a mean enhancement of 62.5 ± 19.7 (range 40 to 89) and with a uniform dense opacification. In conclusion, in this study all patients with RA showed contrast enhancement with uniform dense opacification. The main clinical implication of this finding is that this procedure does not permit differentiation between RA and malignant pulmonary neoplasm.

Introdução: O tabagismo é o principal fator de risco prevenível de morbidade e mortalidade em países desenvolvidos e está em ascensão nos países em desenvolvimento. Apesar deste fato, e do maior conhecimento sobre seus efeitos, a prevalência de tabagistas continua elevada. Com o objetivo de comparar o valor de monóxido de carbono no ar exalado (COex) entre indivíduos fumantes e não fumantes, avaliar fatores que influenciam estes valores entre os que fumam e também avaliar a possível influência do tabagismo passivo, foram medidos níveis de COex em funcionários e pacientes do Instituto do Coração HC-FMUSP. Materiais e métodos: Este estudo transversal incluiu 256 voluntários que responderam a um questionário e foram submetidos à mensuração de COex em aparelho micromedidor de CO. Resultados: Dos entrevistados, 106 eram do sexo masculino e 150 do feminino e a idade média foi de 43,4 anos (Vmin-max: 15-83). 107 informaram ser tabagistas, 118 não fumantes e 31 fumantes passivos. A média de COex dos fumantes foi de 14,01ppm (Vmin-max: 1-44), dos fumantes passivos 2,03ppm (Vmin-max: 0-5) e, dos não fumantes, 2,50ppm (Vmin-max: 0-9). Houve diferença estatisticamente significante ente o grupo de fumantes e os demais (p < 0,001), mas não entre os fumantes passivos e os não fumantes. Foi encontrada correlação positiva entre número de cigarros fumados por dia e valores de COex e negativa entre o intervalo após ter fumado o último cigarro e o valor de COex. Para um valor de corte de COex igual a 6ppm, foram encontradas sensibilidade de 77% e especificidade de 96%. Conclusão: A mensuração de COex constitui-se um indicador de fácil emprego, baixo custo, não invasivo e que permite a obtenção de resultado imediato, com o valor de corte de COex de 6ppm apresentando boa especificidade para aferir o hábito tabágico.
Introduction: Smoking is the major preventable risk of morbidity and mortality. However, its prevalence is high in developed countries and increasing in developing countries, even though its effects are now better known. The purpose of this study was to compare the exhaled carbon monoxide concentration (exCO) between smokers and nonsmokers, evaluate the factors that influence this parameter among smokers and the potential influence of passive smoking by measuring exCO in workers and patients of Instituto do Coração HC-FMUSP. Materials and methods: This cross study included 256 volunteers who responded to a questionnaire and were submitted to exCO measuring with the MicroCo meter device. Results: There were 106 males and 150 females. Mean age was 43.4 years (Vmin-max: 15-83). There were 107 smokers, 118 nonsmokers and 31 passive smokers. Mean exCO was 14.01 ppm (Vmin-max: 1-44) among smokers, 2.03 ppm (Vmin-max: 0-5) among passive smokers and 2.50 ppm (Vmin-max: 0-9) among nonsmokers. Significant statistical difference was observed between smokers and the other groups (p < 0.001), but not between nonsmokers and passive smokers. A positive correlation was found between the number of cigarettes smoked per day and exCO values while there was a negative correlation between the exCO values and the timing of the last cigarette. For a reference limit value of 6 ppm, sensitivity was 70% and specificity was 96%. Conclusion: exCO metering is easy to perform, low-cost, noninvasive and allows the obtention of immediate results and the reference limit value of 6 ppm has good specificity to evaluate the smoking habit.

Dois modelos experimentais foram usados para estudar o efeito de uma carga hiperosmolar nos fluxos bidirecionais de sódio e cloro no intestino de animais em restrição energética. No modelo I, 1 mL de NaCl 900 mOsm foi introduzido no intestino delgado e deixado de 5 a 70 minutos, a fim de determinar o tempo para ocorrer a maior secreção de sódio, o que se observou aos 10 minutos. No modelo II, os fluxos birecionais de sódio e cloro foram determinados em ratos em restrição energética após carga hipertônica, aos 10 minutos. Foram estudados efeitos de: inibidor metabólico (2-4 dinitrofenol) e farmacológicos. Sugere-se que na má nutrição ocorre impedimento da reabsorção intestinal, favorecendo aumento resultante de secreção.
Two experimental models were tried in young malnourished rats in order to study effect of an hyperosmolar challenge in the small intestine on the bi-directional fluxes of sodium. Weanling rats were fed with energy restricted diets. In model I 1 mL of NaCl 900 mOsm/kg was introduced in the small intestine of the rats and left from 5 up to 70 min, in order to determine the moment of higher net Na+ secretion, which occurred at 10 min. In model II, the bi-directional fluxes of Na+ and Cl- were studied using NaCl or mannitol 900 mOsm/kg under the effect of mecholil, atropine or 2-4 dinitrophenol, for 10 min. Mecholil decreased the Na+ absorption enhancing the net secretion. Control rats were used as reference. In the restricted diets animals occurred an increase of the net secretion stimulated by NaCl 900 mOsm/kg, and this effect was enhanced by mecholil. It is suggested that in malnutrition there is an impairment in Na- intestinal absorption.
