Abstract The present study sought to evaluate the protective effect of Epigallocatechin-3-gallate (EGCG) and commercial green tea (GT) on eroded dentin using in vitro and in situ experimental models. For the in vitro experiment, matrix metalloproteinases (MMPs) were extracted from demineralized human coronary dentin powder (citric acid, pH 2.3) and assessed via a colorimetric assay and electrophoresis in gelatin. The gels were exposed to buffers with: control (no treatment), 0.05% sodium fluoride (NaF), 0.12% chlorhexidine digluconate (CHX), GT infusion, and 0.1% EGCG, and their respective activity was analyzed by zymography. For the in situ experiment, 20 healthy volunteers (aged 20-32 years) participated in this single-center, blind, crossover study. The subjects wore upper removable devices containing four human dentin blocks. Erosive challenge (coke-1 min) was performed four times/day/5 days. Blocks were treated for 1 min with: control (No treatment), 0.05% NaF, 0.1% EGCG, and GT. Thereafter, the specimens were subjected to stylus profilometry and SEM. ANOVA was used to evaluate dentin roughness and wear, with a significance level of 5%. In the zymography analysis, 0.12% CHX, GT, and 0.1% EGCG were found to inhibit the action of MMPs; however, in the colorimetric assay, only green tea inhibited the activity of MMPs. There were no significant differences observed in dentin roughness or wear (p > 0.05). Herein, EGCG and GT inhibited the activity of endogenous proteases, resulting in protection against erosion-induced dentin damage; however, they could not prevent tooth tissue loss in situ.

Chagasina pode ser considerada como uma proteína com potencial para protetor incorporado a planta (PPI), devido aos seus efeitos deletérios sobre insetos praga. No entanto, estudos extensivos de segurança com PPI são necessários antes de introduzi-las na planta alvo. Assim, um experimento de alimentação de curto prazo em ratos com doses elevadas de r-chagasina foi conduzido para fornecer evidências sobre a sua segurança. Três dietas teste contendo caseína + r-chagasina (0,25; 0,5 e 1% de proteína total) foram oferecidas aos ratos (10 dias). As dietas teste não apresentaram efeitos adversos sobre o desenvolvimento, o peso de órgãos, parâmetros hematológicos e perfis de proteínas séricas dos ratos, fornecendo informações preliminares sobre a segurança da r-chagasina.

Chagasin may be considered a potential plant-incorporated protectant (PIP) protein due to its deleterious effects on insect pests. However, extensive safety studies with PIP's are necessary before introducing them into the target plant. Thus, a short-term feeding trial in rats with high doses of r-chagasin was conducted to provide evidences about its safety. Three test diets containing casein + r-chagasin (0.25, 0.5 and 1% of total protein) were offered to rats (10 days). The test diets did not show adverse effects upon the development, organ weight, hematological parameters and serum protein profiles of rats, providing preliminary information on the safety of r-chagasin.

The new iridoid glucoside 10-O-vanilloyl-geniposidic acid has been isolated from the aerial parts of Alibertia myrciifolia along with hydroxyhopanone, 3α,22-dihydroxyhopane, ursolic acid, luteolin-3´,4´-dimethyl ether, caffeic acid and geniposidic acid. The structures of the isolated compounds were determined by means of mass spectrometry and nuclear magnetic resonance spectral analyses. The antifungal activities of the iridoids 10-O-vanilloyl-geniposidic acid and geniposidic acid were evaluated against the phytopathogenic fungi strains Colletotrichum gloeosporioides, Fusarium solani and Aspergillus niger.

Eletroforese em gel de poliacrilamida das proteínas solúveis extraídas de sementes de doze espécies da tribo Diocleae, apresenta padrão qualitativo semelhante no que diz respeito às bandas correspondentes às lectinas, sugerindo a possibilidade de seu uso como marcadores taxonômicos.

Polyacrylamide gel electrophoresis of the soluble proteins extracted from seeds of twelve species belonging to the tribe Diocleae showed substantive qualitative pattern similiarities between the bands correspondent to the lectins.
