The fungi Fusarium oxysporum f. sp. vasinfectum (Fov) and F. oxysporum f. sp. phaseoli (Fop) are important pathogens that cause wilt on cotton and common bean, respectively. In the present study, phylogenetic trees constructed with partial sequences of the translation elongation factor gene and ribosomal intergenic spacer region of Brazilian Fov and Fop strains revealed polyphyletic origin of strains within both formae speciales. This creates an obstacle for the development of specific molecular diagnostic methods and explains the failure of supposedly specific methods described in the literature to correctly discriminate formae speciales of F. oxysporum.

Objetivou-se avaliar a incidência de fungos e nematoides em sementes de Brachiaria sp. e Panicum maximum produzidas nos estados de Mato Grosso do Sul (MS), Mato Grosso (MT), Goiás (GO), Minas Gerais (MG) e São Paulo (SP). Os principais fungos encontrados nas sementes foram Bipolaris sp., Curvularia sp. e Phoma sp.. As menores incidências destes fungos foram encontradas nas sementes das cultivares BRS Piatã e Xaraés de Brachiaria brizantha e Brachiaria decumbens cv. Basilisk, oriundas dos estados de GO, MG e MS, respectivamente. As cultivares Marandu e BRS Piatã, provenientes das várias regiões, apresentaram elevada ocorrência de Aphelenchoides sp. e Ditylenchus sp.. Sementes da cultivar Humidicola, de Brachiaria humidicola, produzidas em MS e SP, não apresentaram associação com nematoides. As sementes de Panicum maximum cv. Massai e cv. Mombaça apresentaram maiores incidências de Bipolaris sp., Cladosporium sp., Curvularia sp., Fusarium sp. e Phoma sp., bem como de Aphelenchoides sp. e Ditylenchus sp., especialmente nas sementes produzidas em MT. Alguns dos patógenos encontrados são agentes causais de doenças de grande importância em forrageiras, a exemplo de Bipolaris sp., causando a mancha foliar do Panicum, de alta severidade em Tanzânia, proporcionando sérios comprometimentos da sustentabilidade das pastagens.

This study aimed to assess the incidence of fungi and nematodes in Brachiaria sp. and Panicum maximum seeds produced in the Brazilian states of Mato Grosso do Sul (MS), Mato Grosso (MT), Goiás (GO), Minas Gerais (MG) and São Paulo (SP). The main fungi found in the seeds were Bipolaris sp., Curvularia sp. and Phoma sp.. The lowest incidence of these fungi was found for seeds of Brachiaria brizantha cultivars BRS Piatã and Xaraés, and Brachiaria decumbens cv. Basilisk, from the states of GO, MG and MS, respectively. The cultivars Marandu and BRS Piatã, from several regions, exhibited high occurrence of Aphelenchoides sp. and Ditylenchus sp.. Seeds of B. humidicola cultivar Humidicola, produced in MS and SP, did not show association with nematodes. The seeds of Panicum maximum cv. Massai and cv. Mombaça showed higher incidence of Bipolaris sp., Cladosporium sp., Curvularia sp., Fusarium sp. and Phoma sp., as well as Aphelenchoides sp. and Ditylenchus sp., especially for seeds produced in MT. Some of the detected pathogens are causative agents of diseases of major importance in forage plants, such as Bipolaris sp., causing leaf spot in Panicum, of high severity in Tanzânia, which provides serious compromising of the pasture sustainability.

Visando avaliar o efeito do tratamento químico na incidência de Fusarium graminearum, bem como na germinação, emergência e altura de plântulas, sementes de trigo da cultivar BR 18 Terena foram tratadas com os seguintes fungicidas nas respectivas doses de i.a./ 100 kg de sementes: captana (150,0 g), tiofanato metílico (75,0 mL), triflumizole (45,0 g), triticonazole (45,0 mL), triadimenol (13,5 mL), tolyfluanida (75,0 g), tebuconazole (5,0 mL), fludioxonil (5,0 mL), difeconazole (30,0 mL) e thiabendazole (30,0 mL). Em laboratório os fungicidas triflumizol, triadimenol, triticonazole, thiabendazole e tiofanato metílico reduziram significativamente a incidência de F. graminearum nas sementes em relação à testemunha. Thiabendazole e tiofanato metílico foram superiores ao triflumizol e triadimenol; os demais fungicidas comportaram-se de maneira semelhantes à testemunha. Não houve influência dos fungicidas na germinação das sementes. Em casa de vegetação a emergência e velocidade de emergência de plântulas, também não foram afetadas. Por outro lado, triadimenol interferiu negativamente na altura de plântulas aos sete DAS e captana, trticonazole, tebuconazole e triadimenol aos 14 DAS.

The effect of chemical treatment of wheat seeds (cv. "BR 18 Terena") was studied under laboratory and greenhouse conditions using the following fungicides with the respective doses of i.a./100 kg of seeds: captan (150 g), methilic tioafanate (75 mL), triflumizole (45 g), triticonazole (45 mL) triadimenol (13.5 mL), tolyfluanid (75 g), tebuconazole (5 mL), fludioxonil (5 mL), difeconazole (30 mL) and thiabendazole (30 mL). In laboratory, seeds health and germination were evaluated while under greenhouse conditions were evaluated emergency, velocity of emergency and the height of seedlings. The fungicides triflumizole, triadimenol, triticonazole, thiabendazole and methilic tioafanate reduced significantly the incidence of F. graminearum in the seeds with thiabendazole and methilic tioafanate being the most efficient ones. Seeds germination, emergence and velocity of the seedlings were not affected by chemical treatment with the fungicides. On the other hand, triadimenol affect negativity the height of seedlings to 7 days after sowing and captan, triticonazole, tebuconazole and triadimenol to 14 days.

O objetivo deste trabalho foi comparar diferentes métodos utilizados para a detecção de Fusarium graminearum em sementes de trigo. Foram empregados 22 tratamentos: papel de filtro (PFC) com congelamento; papel de filtro (PF); PF mais 0,02% de 2,4 - D; meio semi-seletivo (MSS); MSS mais 0,02% de 2,4 - D; MSS mais KCl (-0,8 MPa); KCl, NaCl, manitol e sacarose nos potenciais osmóticos de -0,4, -0,6, -0,8 e -1,0 MPa. O delineamento estatístico empregado foi o inteiramente casualizado, com quatro repetições (duas placas mais substrato com 25 sementes cada/repetição). Não houve diferenças significativas entre os diferentes métodos empregados e o método do PF com congelamento das sementes, considerado o método padrão para o teste de sanidade de sementes de gramíneas, na detecção de F. graminearum em sementes de trigo.

The objective of this work was to compare different methods used in seed health testing for detection of Fusarium graminearum in wheat seeds. Twenty two treatments had been used: blotter test (BTDF) deep freezing; blotter test (BT); BT more 0.02% of 2,4- D; media half-selective (MSS); MSS more 0.02% of 2,4 - D ; MSS more KCl (-0,8 MPa); KCl, NaCl, manitol and sacarose in the osmotic potentials of -0,4, -0,2, -0,6 and -0,8 MPa. The used statistical delineation was entirely casualized, with four repetitions (two plates more substratum with 25 seeds each/repetition). There were no significant differences between of the different employed methods and the blotter test deep freezing, considered the method standard for seed health testing of grassy, for detection of the Fusarium graminearum in wheat seeds.

Considerando a grande variabilidade apresentada pelo complexo Diaporthe/Phomopsis, os métodos rotineiros de sanidade de sementes, empregados na detecção do gênero Diaporthe, não são confiáveis visto que, para sua identificação são utilizadas as características morfológicas das colônias que se desenvolvem sobre as sementes. Diante disso, este trabalho teve como objetivo desenvolver iniciadores específicos, bem como um método confiável para detecção e identificação de Diaporthe phaseolorum var. meridionalis (Dphm), em sementes de soja. Amostra de sementes da cultivar IAS5, analisada previamente para sanidade, não portadoras de Diaporthe (SS) foram inoculadas com Dphm (SI), obtendo-se amostras com as seguintes proporções de SI:SS: 1:10, 1:50, 1:100, 1:200 e 1:400. Para a extração de DNA do micélio do patógeno das sementes foi necessário primeiro incubar as sementes por 7 dias, utilizando-se o método do papel de filtro modificado com 2,4 D e, em seguida submetê-las à lavagem em tampão de extração (1% de SDS, 10 mM de TRIS/HCL e 25 mM de EDTA) por 20 min, sob agitação a 100 rpm. Em seguida, alíquotas de 350 miL do sobrenadante foram transferidas para novos microtubos contendo 350 miL de resina de sílica gel da Wizard/Promega permanecendo em contacto com a mesma por 1 minuto. Os DNAs extraídos foram utilizados como molde na reação de PCR com os iniciadores: DphLe (TCG GCC TTG GAA GTA GAA AG) e DphRi (ACT GAA TGC GTT GCG ATT CT) Utilizando-se estes iniciadores, foi possível detectar a presença de D. phaseolorum var meridionalis em sementes de soja, na proporção de uma semente infectada com o patógeno em amostras de 400 sementes (0,25% de incidência), utilizando-se o método do papel de filtro modificado com 2,4 D associado à técnica de PCR.

In spite of the great variability of the Diaporthe/Phomopsis complex, the routine seed health testing, used for detection of the Diaporthe, is not reliable, taking consideration into that for its identification morphological characteristics of the colonies are used. By considering, this work had as objective to develop an accurate, fast and reliable method for Diaporthe phaseolorum var. meridionalis (Dphm) detection and identification in soybean seeds. A sample of seeds from cultivar IAS5, analyzed previously for sanity, without Diaporthe (SS) was inoculated with Dphm (S.I.). Samples were collected with the following SI:SSrations: 1:10, 1:50, 1:100, 1:200 and 1:400. DNA mycelium extraction was necessary first to incubate, the seeds, during 3 to7 days, using blotter test modified with 2,4 D and washing them with buffer extraction (1% of SDS, 10 mM of TRIS/HCl and 25 mM of EDTA) for 20 min, on an orbital shaker (100 rpm). An aliquot of 350 muL of the supernatants was transferred to microcentrifuge tubes with 350 muL of silica resin gel of the Wizard kit (Promega) remaining in contact during one minute. DNA extracted was used as template in the reaction of PCR by using the following primers: DphLe (TCG GCC TTG GAA GTA GAA GAC) and DphRi (ACT GAA TGC GTT GCG ATT CT). Using these primers, it was possible to detect the presence of D.phaseolorum var meridionalis in soybean seeds, in the ratio of one infected seed with Dphm in samples of 400 seeds (0.25% of incidence), using blotter test modified with 2,4 D associated with the PCR technique.
