RESUMO: Os objetivos do presente estudo foram (i) genotipar amostras de Corynebacterium pseudotuberculosis, C. silvaticum e C. auriscanis usando Enterobacterial Repetitive Intergenic Consensus (ERIC-PCR), bem como (ii) analisar as relações epidemiológicas entre os isolados de acordo com biovar (Equi e Ovis), espécie, hospedeiro e origem geográfica das amostras de C. pseudotuberculosis. Sessenta e oito isolados de C. pseudotuberculosis, nove C. silvaticum, um C. auriscanis, C. pseudotuberculosis ATCC® 19410 ™ e a amostra vacinal atenuada C. pseudotuberculosis 1002 foram tipificadas por ERIC 1 + 2-PCR. As amostras de campo foram isoladas de diferentes hospedeiros (bovinos, búfalos, ovinos, caprinos, equinos, cães e suínos) em seis países (México, Portugal, Brasil, Guiné Equatorial, Egito e Israel). Uma alta diversidade genética foi observada entre os isolados de Corynebacterium spp., agrupados em vinte e quatro genótipos com um índice de diversidade Hunter & Gaston (HGDI) de 0,937. A análise da minimal spanning tree (MST) de Corynebacterium spp. revelou três complexos clonais, cada um associado a uma espécie bacteriana. Vinte e dois genótipos foram observados entre isolados de C. pseudotuberculosis, com um HGDI de 0,934. Na análise da MST, três grandes complexos clonais foram formados, agrupando-se em torno da origem geográfica dos isolados de C. pseudotuberculosis. Esses resultados reforçam a alta tipabilidade, concordância epidemiológica e poder discriminatório do ERIC-PCR como método consistente de genotipagem para C. pseudotuberculosis, podendo ser útil como ferramenta epidemiológica no controle da linfadenite caseosa. Além disso, os resultados também indicam o grande potencial de ERIC 1 + 2-PCR para genotipagem de espécies do gênero Corynebacterium além de C. pseudotuberculosis.

ABSTRACT: The aims of the present study were (i) to genotype Corynebacterium pseudotuberculosis, C. silvaticum, and C. auriscanis strains using enterobacterial repetitive intergenic consensus (ERIC-PCR), and (ii) to analyze the epidemiological relationships among isolates according to biovar (Equi and Ovis), species, host, and geographical origin of the C. pseudotuberculosis strains. Sixty-eight C. pseudotuberculosis, nine C. silvaticum, and one C. auriscanis, C. pseudotuberculosis ATCC® 19410™ strain and the attenuated C. pseudotuberculosis 1002 vaccinal strain were fingerprinted by ERIC 1+2-PCR. Field strains were isolated from various hosts (cattle, buffaloes, sheep, goats, horses, dogs, and pigs) in six countries (Mexico, Portugal, Brazil, Equatorial Guinea, Egypt, and Israel). High genetic diversity was found among the studied Corynebacterium spp. isolates, clustering in 24 genotypes with a Hunter & Gaston diversity index (HGDI) of 0.937. The minimal spanning tree of Corynebacterium spp. revealed three clonal complexes, each associated with one bacterial species. Twenty-two genotypes were observed among C. pseudotuberculosis isolates, with an HGDI of 0.934. Three major clonal complexes were formed at the minimal spanning tree, grouped around the geographic origin of C. pseudotuberculosis isolates. These results reinforce the high typeability, epidemiological concordance, and discriminatory power of ERIC-PCR as a consistent genotyping method for C. pseudotuberculosis, which could be useful as an epidemiological tool to control caseous lymphadenitis. Moreover, our results also indicate the potential of ERIC 1+2-PCR for the genotyping of other species of Corynebacterium other than C. pseudotuberculosis.

RESUMO: A mastite influencia negativamente na sobrevivência e no ganho de peso em ovinos com aptidão para produção de carne. O objetivo do presente estudo foi investigar, em ovelhas para produção de carne, a ocorrência de mastite ovina subclínica, no final de lactação e no início da lactação consecutiva, bem como avaliar as características composicionais e celulares do leite em função de diferentes índices pluviométricos. Metades mamárias (821) das raças 'Santa Inês' (479) e 'Morada Nova' (342) foram analisadas. As amostras de leite foram colhidas em dois estágios diferentes de lactação: no desmame e após o parto da lactação consecutiva. Os períodos de colheita de amostras foram denominados "seco" ou "chuvoso", conforme o índice de chuva no mês imediatamente antes ao mês da colheita. A ocorrência de mastite subclínica na desmama nas raças 'Santa Inês' e 'Morada Nova' foi de 16,4 e 12,6% no período seco, e 17,7 e 23,5% no período chuvoso, respectivamente. Na lactação consecutiva, as ocorrências foram 26,7 e 27,7% no período seco e 41,8 e 39,1% no período chuvoso, nas ovelhas 'Santa Inês' e 'Morada Nova', respectivamente. O período pós-parto foi crítico para a ocorrência de mastite subclínica, comparado àquele no final da lactação anterior. A ocorrência da doença teve uma influência negativa na CCS do leite no início da lactação e alterou sua composição, principalmente nos períodos mais chuvosos, provavelmente por dificuldade em manter a higiene no ambiente onde os animais permaneceram.

ABSTRACT: Mastitis negatively influences the survival and weight gain of ovines for meat production. The purpose of this study was to investigate, in sheep for meat production, the occurrence of subclinical mastitis in ewes at the end of lactation and beginning of the consecutive lactation and to assess the composition and cellular characteristics of milk as a function of different rainfall indices. Mammary halves (821) of Santa Ines (479) and Morada Nova (342) ewes were examined. Milk samples were collected in two different moments of lactation: at weaning and postpartum of the consecutive lactation. Sample collection periods were called "dry" or "rainy" according to the rainfall index in the month immediately before the month of collection. The occurrence of subclinical mastitis at weaning in the Santa Ines and Morada Nova ewes were 16.4 and 12.6% in the dry period, and 17.7 and 23.5% in the rainy period, respectively. In the consecutive lactation period, the occurrences were 26.7 and 27.7% in the dry period and 41.8 and 39.1% in the rainy period, for the Santa Ines and Morada Nova ewes, respectively. Postpartum stage was critical for the occurrence of subclinical mastitis, as compared to that at the end of the previous lactation. Occurrence of the disease negatively influenced the SCC in the milk at the beginning of lactation and changed its composition, mainly in the rainiest periods, probably due to a difficulty in maintaining hygiene in the environment where the animals remained.

RESUMO: Maedi-Visna (MV) é uma enfermidade causada por lentivírus de pequenos ruminantes com evolução crônica e em grande parte dos casos sinais clínicos inaparentes. O diagnóstico da doença é baseado em sinais clínicos e dados epidemiológicos, sendo a imunodifusão em gel de ágar (IDGA) o método padrão para a detecção sorológica de anticorpos contra o lentivírus. Sabendo que o estado do Tocantins possui potencial para o desenvolvimento da ovinocultura e que grande parte dos produtores de Colinas do Tocantins, no referido estado, possui interesse em estabelecer criação racional, esta pesquisa teve por objetivo a realização de um estudo acerca da soroprevalência da doença. Foram coletadas 369 amostras de sangue de ovinos, independentemente de raça, sexo e idade, de diferentes propriedades rurais do município para diagnóstico de MV utilizando a técnica de IDGA. Após as análises laboratoriais, para avaliação dos resultados no tocante às categorias, foi utilizado o teste exato de Fisher e também foi calculado o odds ratio , com intervalo de confiança de 95% para verificação da idade como possível fator de risco ou de proteção. Constatou-se que 6 animais (1,62%) se apresentaram positivos no IDGA. Diante desses resultados, foi possível concluir que a frequência de ovinos soropositivos no município é baixa.

ABSTRACT: Maedi-Visna is a disease caused by a small ruminant lentivirus, with a chronic evolution and, in most cases, unapparent signs. Its diagnosis is based on clinical signs and epidemiological data, with the agar gel immunodiffusion (AGID) test being the classical method for detecting antibodies against lentiviruses. Considering that the state of Tocantins has the potential to develop sheep breeding and that the majority of producers from Colinas do Tocantins city has shown an interest in establishing a rational breeding, this study aimed to determine the prevalence of such disease in that region. A total of 369 blood samples was drawn from sheep bred in several farms of that town, regardless of breed, gender, and age. Every sample underwent an AGID to test for Maedi-Visna. After the laboratory analyses were concluded, the Fisher's exact test was used to evaluate the results against the categories. Also, the odds ratio, with 95% confidence interval, was calculated to check whether age played a role as either a risk or protective factor in these results. It was found that six animals (1.62%) were positive in AGID, therefore concluding that the frequency of seropositivity in that region is low.

O objetivo desse estudo foi avaliar a ocorrência da mastite subclínica ovina em duas lactações e a evolução da doença em duas lactações consecutivas. As amostras de leite foram originadas de um rebanho com 160 ovelhas da raça Santa Inês. Na primeira lactação, colheram-se amostras em ovelhas aos 14 dias pós-parto e no período próximo ao desmame. Na segunda lactação, as amostras de leite foram colhidas dos mesmos animais aos 14 dias pós-parto, 52 dias pós-parto e ao desmame. A triagem dos casos de mastite subclínica foi feita preliminarmente por meio do California Mastitis Test (CMT). As amostras de leite para a investigação microbiológica foram obtidas de metades mamárias positivas e negativas ao CMT. Os estafilococos coagulase-negativos foram os micro-organismos de maior ocorrência nas duas lactações. Houve a predominância de ovelhas que permaneceram com mastite subclínica durante a Lactação 1. As curas espontâneas foram significativamente inferiores no período entre 52 dias de lactação e o desmame dos cordeiros. Nenhuma diferença foi observada quanto à evolução dos casos de mastite subclínica infecciosa quando comparadas duas lactações (P>0,05).

The aim of this study was to evaluate the occurrence of ovine subclinical mastitis in two lactations, and the disease evolution in two consecutive lactations. Milk samples originated from a herd with 160 Santa Inês breed sheep. In the first lactation, milk samples were collected from ewes at 14 days post-partum and during the period near weaning. In the second lactation, milk samples were collected from the same animals at 14 days postpartum, at 52 days postpartum and at weaning. Screening of subclinical mastitis was carried out preliminarily by means of California Mastitis Test (CMT). Milk samples for microbiological culture were obtained from CMT-positive and CMT-negative mammary halves. Coagulase-negative Staphylococci were the most prevalent microorganisms isolated in two lactations. Ewes that remained with subclinical mastitis in lactation 1 were predominant. Spontaneous recoveries were significantly lower in the period between 52 days of lactation and weaning of lambs. No difference was observed regarding the progression of subclinical mastitis cases compared the two lactations (P>0.05).

Caseous lymphadenits (CL) is a chronic and subclinical disease that affects goats and sheep and, consequently, causes economic losses, especially to small producers. The purpose of this study, through use of Polymerase Chain Reaction (PCR), was to verify the presence of virulence genes of phospholipase D (PLD), integral membrane protein (FagA), iron enterobactin transporter (FagB), ATP binding cytoplasmic membrane protein (FagC) and iron siderophore binding protein (FagD) in 168 isolates of C. pseudotuberculosis obtained from cases of caseous lymphadenitis in goats and sheep. FagA, FagB and PLD genes were detected in all 145 strains isolated from abscesses in superficial lymph nodes and in 23 strains isolated from viscera. The FagC gene was positive in 167 (99.40%) isolates. The FagD gene was detected in 160 (95.23%) isolates. All virulence factors analyzed were found more frequently among isolates collected in the viscera of animals with CL, indicating a multifactorial nature, as well as variations, in the invasive potential of C. pseudotuberculosis strains.
