Resistance determinants of emerging pathogens isolated from an intensive care unit as a parameter of population health conditions of the Legal Amazon microregion

Abstract Bacteria responsible for causing infections are common in hospital environments, water, soil, and food products. The infection risk is intensified by the absence of public sanitation, poor quality of life, and food scarcity. These external factors promote the dissemination of pathogens by direct contamination or biofilm formation. In this work, we identified bacterial isolates obtained from intensive care units in the southern region of Tocantins, Brazil. We compared matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) techniques and 16S ribosomal ribonucleic acid (rRNA) molecular analysis; we also performed phenotypic characterization. Fifty-six isolates characterized using morphotinctorial tests were classified as gram-positive (80.4%; n = 45) and gram-negative (19.6%; n = 11) and were resistant to several antibiotic classes; notably, we identified the blaOXA-23 resistance gene in the ILH10 isolate. Microbial identification using MALDI-TOF MS resulted in the identification of Sphingomonas paucimobilis and Bacillus circulans. 16S rRNA sequencing revealed four isolates belonging to the genera Bacillus and Acinetobacter. The similarity was superior to 99% for Acinetobacter schindleri in the Basic Local Alignment Search Tool (BLAST), grouped in the clade superior to 90%. Several strains isolated from intensive care units (ICU) were resistant to various antibiotic classes. These techniques allowed for the identification of several microorganisms of importance in public health, enabling improvements in human infection control and proving the quality of inputs, food, and water.
Resumo As bactérias responsáveis por causar infecções são comuns em ambientes hospitalares, água, solo e produtos alimentícios. O risco de infecção é intensificado pela ausência de saneamento público, má qualidade de vida e escassez de alimentos. Esses fatores externos promovem a disseminação de patógenos por contaminação direta ou formação de biofilme. Neste trabalho, identificamos isolados bacterianos obtidos de unidades de terapia intensiva na região sul do Tocantins, Brasil. Comparamos técnicas de espectrometria de massa de tempo de voo com ionização por dessorção a laser assistida por matriz (MALDI-TOF MS) e análise molecular de ácido ribonucleico ribossômico 16S (rRNA); também realizamos caracterização fenotípica. Cinquenta e seis isolados caracterizados por testes morfotintoriais foram classificados como gram-positivos (80,3%; n = 45) e gram-negativos (19,6%; n = 11) e foram resistentes a várias classes de antibióticos; notavelmente, identificamos o gene de resistência blaOXA-23 no isolado de ILH10. A identificação microbiana usando MALDI-TOF MS resultou na identificação de Sphingomonas paucimobilis e Bacillus circulans. O sequenciamento do 16S rRNA revelou quatro isolados pertencentes aos gêneros Bacillus e Acinetobacter. A similaridade foi superior a 99% para Acinetobacter schindleri no BLAST, agrupado no clado superior a 90%. Várias cepas isoladas de ICU foram resistentes a várias classes de antibióticos. Essas técnicas permitiram a identificação de diversos microrganismos de importância em saúde pública, possibilitando melhorias no controle de infecções humanas e comprovando a qualidade dos insumos, alimentos e água.

Fatty Acid Content and Physicalchemical Properties of Cagaite Seed Oil (Eugenia dysenterica DC) Obtained by Different Extraction Methods

Cagaiteira, the fruit popularly known as cagaite, belongs to the Myrtaceae family. Seed oil cagaite (Eugenia dysenterica DC) was obtained by three methods of extraction, extraction using an ultrasound (OCU), extraction by mechanical pressing (OCP) and extraction with Soxhlet extractor (OCS) in 3 different times (3, 6 and 9 h) and the content of fatty acids and physicalchemical properties were compared. The rate of saturated fatty acids ranged from 19.46% (OCP03) to 31.18% (OCS09), while the amount of unsaturated fatty acids ranged from 54.72% (OCP03) to 67.64% (OCS09). Linoleic and linolenic acids, important in nutrition food, varied between 32.22-38.11 and 5.55-8.78%, respectively. For oxidative stability, OCUAq (heated ultrasound extraction) samples showed the highest induction periods, showing a positive correlation with antioxidant activity and phenol content, demonstrating the efficiency of the use of ultrasonic extraction to obtain quality oils from cagaite seeds.

Resumo Este trabalho teve como objetivo descrever a biologia reprodutiva de Hypoptopoma inexspectatum na região do Pantanal Sul. Foram amostrados e analisados entre fevereiro de 2009 a janeiro de 2011 um total de 538 indivíduos. Não foi observada diferença na proporção sexual e ambos os sexos apresentam relação alométrica de peso/comprimento positiva. A reprodução ocorreu principalmente entre os meses de janeiro e fevereiro. Fêmeas atingiram o tamanho de primeira maturação (L50) com 37,80 mm e machos com 45,80 mm. A fecundidade absoluta média foi 177,43 (dp±127,06) ovócitos e a fecundidade relativa foi de 3,12 (dp±2,23) ovócitos/mg. A fecundidade foi positivamente correlacionada com o comprimento padrão (Spearman r= 0,43; p=0,013) e a desova parcelada foi a estratégia de desova utilizada por H. inexspectatum.
Abstract This work aimed to describe the reproductive biology of Hypoptopoma inexspectatum in southern Pantanal. A total of 538 individuals were sampled and analyzed from February 2009 through January 2011 in southern Pantanal. We did not observe differences in sex-ratio, and both sexes presented positive allometric length/weight relationship (LWR). Reproduction occurs mainly from January to February. Females reached first maturation (L50) with 37.80 mm and males with 45.80 mm. Absolute fecundity is estimated in mean=177.43 (sd±127.06) oocytes and relative fecundity in 3.12 (sd±2.23) oocytes/mg. Fecundity is positively correlated with standard length (Spearman r= 0.43; p=0.013), and total spawning is a used spawning strategy for H. inexspectatum.

Resumo Objetivo Comparar por meio de ultrassom tridimensional (US-3D) a posição e o ângulo entre os braços da faixa, em diferentes técnicas de inserção de sling de uretra média, para tratamento de incontinência urinária de esforço, 3 anos após a cirurgia, correlacionando os achados ultrassonográficos aos resultados clínicos pós-operatórios. Métodos Este é umestudo de coorte transversal prospectivo de 170 pacientes que se submeteram a um procedimento de sling entremaio de 2009 e dezembro de 2011. Foi possível avaliar as imagens de US em 105 pacientes: 26 com tension-free vaginal tape (TVT), 42 com tension-free vaginal tape-obturator (TVT-O) e 37 com tension-free vaginal tape-Secur (TVT-S). As imagens (em repouso, em manobra de Valsalva e em contração perineal) foram analisadas por dois observadores diferentes, que desconheciam o tipo de sling utilizado na cirurgia, assimcomo as queixas da paciente. A análise estatística foi realizada por meio dos testes t de Student, qui-quadrado, Kruskal-Wallis, e análise de variância com comparações múltiplas de Tukey. Resultados As médias dos ângulos entre os braços da faixa foram: TVT = 119,94°, TVT-O = 141,93°, TVT-S = 121,06° (p < 0,001). As médias das distâncias entre o colo vesical e a faixa, em repouso, foram: TVT = 1,65 cm, TVT-O = 1,93 cm, TVTS = 1,95 cm (p = 0,010). A taxa de cura objetiva dos slings foi de 87,8% (TVT = 88,5%, TVT-O = 90,5% e TVT-S = 83,8%; p = 0,701). A taxa de cura subjetiva foi de 83,8% (TVT = 88,5%, TVT-O = 88,5% e TVT-S = 78,4%; p = 0,514). Os slings estavam na uretra média em 85,7% (TVT = 100%, TVT-O = 73,8% e TVT-S = 89,2%; p = 0,001) dos pacientes, e a localização mais distal foi associada a obesidade (distal: 66,7% obesas; uretra média: 34% obesas; p = 0.003). Os sintomas de urgência foram observados em 23,8% das pacientes (TVT = 30,8%, TVT-O = 21,4%, TVT-S = 21,6%; p = 0,630). Não houve diferenças significativas quando se comparam os achados ultrassonográficos e os grupos de pacientes com sintomas de urgência, cura subjetiva e objetiva. Conclusão O ângulo formado pelos braços da faixa foi mais obtuso noTVT-Oquando comparado com o TVT ou o TVT-S. Os TVTs foram localizados mais frequentemente na uretra média quando comparados com os outros dois grupos, mesmo em pacientes obesas. Entretanto, as medidas ultrassonográficas não tiveram correlação com os sintomas urinários três anos após a cirurgia.
Abstract Objective Using three-dimensional ultrasound (3D-US), we aimed to compare the tape position and the angle formed by the sling arms in different techniques of midurethral sling insertion for the surgical treatment of stress urinary incontinence, three years after surgery. In addition, we examined the correlations between the US findings and the clinical late postoperative results. Methods A prospective cross-sectional cohort study of 170 patients who underwent a sling procedure between May 2009 and December 2011 was performed. The final sample, with US images of sufficient quality, included 26 retropubic slings (tension-free vaginal tape, TVT), 42 transobturator slings (tension-free vaginal tape-obturator, TVTO), and 37 single-incision slings (tension-free vaginal tape-Secur, TVT-S). The images (at rest, during the Valsalva maneuver, and during pelvic floor contraction) were analyzed offline by 2 different observers blinded against the surgical and urinary continence status. Group comparisons were performed using the Student t-test, the chi-squared and the Kruskal-Wallis tests, and analyses of variance with Tukey multiple comparisons. Results Differences among the groups were found in themean angle of the tape arms (TVT = 119.94°, TVT-O = 141.93°, TVT-S = 121.06°; p < 0.001) and in the distance between the bladder neck and the tape at rest (TVT = 1.65 cm, TVT-O = 1.93 cm, TVTS = 1.95 cm; p = 0.010). The global objective cure rate was of 87.8% (TVT = 88.5%, TVT-O = 90.5%, TVT-S = 83.8%; p = 0.701). The overall subjective cure rate was of 83.8% (TVT = 88.5%, TVT-O = 88.5% and TVT-S = 78.4%; p = 0.514). The slings were located in the mid-urethra in 85.7% of the patients (TVT = 100%, TVT-O = 73.8%, TVTS = 89.2%; p = 0.001), with a more distal location associated with obesity (distal: 66.7% obese; mid-urethra: 34% obese; p = 0.003). Urgency-related symptoms were observed in 23.8% of the patients (TVT = 30.8%, TVT-O = 21.4%, TVT-S = 21.6%; p = 0.630). Conclusions The angle formed by the arms of the sling tape was more obtuse for the transobturator slings compared with the angles for the retropubic or single-incision slings. Retropubic slings were more frequently located in the mid-urethra compared with the other slings, regardless of obesity. However, the analyzed sonographic measures did not correlate with the urinary symptoms three years after the surgery.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Foram estudados os efeitos da incorporação de quatro diferentes aditivos poliméricos nas propriedades reológicas e de filtração de fluidos de perfuração à base de água e argilas bentoníticas de Boa Vista, PB. Os resultados mostraram que estas propriedades são fortemente influenciadas pela presença dos polímeros e que o tamanho da cadeia polimérica é um fator determinante.
The effects of addition of four different polymeric additives in the rheological and water loss properties of clay dispersions as drilling fluids based on water were studied. The results show that these properties are strongly dependent on the polymeric additives as well as on polymer chain length.

Efeitos de ciclos de umedecimento e secagem na reorganização da estrutura microgranular de latossolos

Um experimento foi realizado em casa de vegetação com o objetivo de examinar as modificações provocadas por ciclos de umedecimento e secagem em amostras de Latossolos com diferentes mineralogias, em agregados integrais ou quando destruídos por moagem. Amostras de terra fina seca ao ar foram passadas em peneira de 1,0 mm e subdivididas em grupos: solo integral não moído e solo moído em almofariz e passado em peneira de 0,105 mm. O solo moído foi ainda subdividido em solo moído puro ou adicionado de ácido húmico purificado. Os materiais foram montados em cilindro de alumínio e submetidos a dez ciclos sucessivos de umedecimento e secagem, realizados por ascensão capilar, seguidos por secagem ao ar, com intervalo de sete dias entre cada ciclo. Após o término dos ciclos, os solos foram impregnados com resina de poliéster, montando-se seções finas para microscopia, as quais foram levadas para observação em microscópio ótico e fotografadas. As imagens obtidas foram digitalizadas, analisadas e quantificadas por meio do programa QUANTIPORO, desenvolvido no Departamento de Solos/Universidade Federal de Viçosa. Os resultados mostraram completa modificação na forma e no padrão da estrutura após a aplicação dos ciclos nos tratamentos que sofreram destruição de agregados por moagem. Essas mudanças foram atribuídas a uma reacomodação dos agregados fragmentados com a retração do plasma, que se seguiu ao processo de secagem. Todos os materiais estudados mostraram uma estrutura bem diferente da estrutura primária original destes solos. Não se observou qualquer tendência à reversão da estrutura após os dez ciclos, demonstrando que outros fatores, além dos físico-químicos e mineralógicos, devem ser invocados para explicar a gênese de microagregados em Latossolos.
A greenhouse experiment was conducted to examine the structural modifications engendered by wetting-drying cycles in Latosol samples (Oxisols) with different mineralogical properties submitted to dry grinding, compared with bulk samples. Air-dried soil samples were sieved through a 1.00 mm sieve and separated in two groups. One was kept as control group and the other ground in a mortar and sieved through a 0.105 mm sieve. This second was also split in two groups, one of which received particulate humic acid treatment. The materials were placed in aluminum columns, subjected to 10 weekly wetting cycles by capillarity followed by air-drying. At the end of the cycles, the samples were impregnated with polyester resin, cut in blocks and thin-sectioned for microscopy analysis. The sections were scrutinized under optical microscope, photographed, and the images digitalized, analyzed, and quantified with the Software QUANTIPORO. After the wetting and drying cycles, important alterations in both shape and structural pattern were observed in the ground samples. These changes were attributed to a re-organization of soil particles during the plasma shrinkage after the drying process. All materials presented a different structural pattern in relation to the original soil structure. No trend for reversion to the original structure was observed after the cycles. This indicates that other factors besides the physical-chemical and mineralogical should be considered to explain the genesis of Latosol microaggregates.

Foram avaliadas características morfométricas da porosidade de um Argissolo Vermelho-Amarelo, cultivado por dez anos sob os tratamentos: semeadura direta; arado de aiveca + grade destorroadora; arado de discos + grade destorroadora; grade pesada + arado de aiveca + grade destorroadora; grade pesada + arado de discos + grade destorroadora; grade pesada + grade destorroadora. O delineamento experimental foi em blocos ao acaso, com seis tratamentos e quatro repetições. Blocos de solo com estrutura natural, com dimensões de 15 cm de largura por 15 cm de profundidade, foram amostrados para análises morfométricas da porosidade. As análises dos blocos polidos mostraram descontinuidade e diminuição da porosidade nos sistemas de preparo do solo que envolveram o uso da grade pesada. O sistema de semeadura direta mostrou boa conexão entre os macroporos, sem revelar descontinuidade entre a superfície e as camadas inferiores.
The porosity characteristics of a Red-Yellow Podzolic was evaluated after 10 years of cultivation, under the following soil management treatments: no-tillage, moldboard plowing + disking, disk plowing + disking, off-set disk harrowing followed by moldboard plowing + disking, off-set disk harrowing followed by disk plowing + disking and off-set disk harrowing followed by disking. Undisturbed 15 cm high and 15 cm deep soil blocks, were sampled for morphometric analysis of the soil porosity. Results indicated the presence of surface sealing and discontinuity between top and subsurface in treatments where offset disk harrowing was employed. The no-tillage system showed higher connection between macropores, without discontinuities between the topsoil and deeper layers.

Estrutura e dinâmica de populações arbóreas de um fragmento de floresta estacional semidecidual na região de Piracicaba, SP

Para verificar a hipótese de que a fragmentação das florestas tropicais associada às perturbações antrópicas podem resultar numa maior perda de espécies arbóreas e na mudança na composição florística ao longo do tempo foi analisado um fragmento de floresta estacional semidecidual de 9,5 ha localizado na região de Piracicaba, Estado de São Paulo, onde houve dois incêndios em passado recente. Neste fragmento, denominado de Mata do Pomar, o recrutamento, a mortalidade e a mudança do tamanho populacional de espécies arbóreas foram obtidos num intervalo de quatro anos (1991 e 1995). Padrões de recrutamento, mortalidade e conseqüente mudança populacional variaram entre as espécies e os grupos ecológicos. As dez espécies mais abundantes no ano de 1991, que representam a classe 1 de densidade ( > ou = 15 indivíduos/ha), foram responsáveis por 79,9% do recrutamento, contra apenas 7,5% da classe 2 (5,2 a 13,8 indivíduos/ha) e 9,0% da classe 3 (1,7 e 3,4 indivíduos/ha). Particularmente, as três espécies mais abundantes em 1991, Bauhinia forficata, Aloysia virgata e Urera baccifera, todas pioneiras, e uma espécie oportunista, Croton floribundus, foram responsáveis pelo alto recrutamento apresentado pela classe 1 de densidade. Apenas essas espécies perfizeram 75% do recrutamento e, conjuntamente com uma baixa mortalidade, experimentaram um aumento pronunciado em suas populações. Além do baixo recrutamento obtido pelas classes 2 e 3, as espécies pioneiras e oportunistas foram responsáveis por 74,2% do recrutamento nessas duas classes e apresentaram maior ganho em densidade que as espécies tolerantes e reprodutoras à sombra. Além desses resultados, foi também realizada uma análise comparativa deste fragmento com outros dois: um fragmento de mesmo tamanho (fragmento Santa Rita - 9,5 ha) e um fragmento com tamanho superior (fragmento Capuava - 86 ha), ambos também localizados no mesmo município. O fragmento Mata do Pomar apresenta menos da metade do número de espécies encontrado nos fragmentos Santa Rita e Capuava, embora tenha o mesmo tamanho do fragmento Santa Rita. Além disso, ocorre uma alta dominância local de poucas espécies no fragmento Mata do Pomar: as quatro espécies mais abundantes, B. forficata, A. virgata, U. baccifera e Trema micrantha, todas pioneiras, são responsáveis por 62,0% do total de indivíduos, ao passo que as quatro espécies mais abundantes nos fragmentos Santa Rita e Capuava perfazem apenas 30,6% e 23,2% dos indivíduos, respectivamente. Este estudo demonstra que as estratégias de conservação da biodiversidade em fragmentos de floresta estacional semidecidual no interior do Estado de São Paulo devem considerar o histórico de perturbações na identificação de prioridades.
Fragmentation of tropical forests associated with anthropogenic disturbance can result in a loss of tree species and floristic composition change throughout the time in a forest fragment. To gather quantitative data regarding this hypothesis, this study was conducted in a 9.5 ha semideciduous seazonal forest fragment located in Piracicaba region, São Paulo state, Brazil. This fragment is named "Mata do Pomar", where two fires occurred in the recent past. In this fragment, recruitment, mortality and populational change of tree species were obtained in 4-year period (1991 and 1995). Different patterns of recruitment, mortality and populational change were found among tree species and ecological groups. The ten species more abundant in 1991 (class 1 - > or = 15 individuals/ha ) presented 79.9% of recruitament, against only 7.5% and 9.0% of recruitment presented by density classes 2 (5.2 to 13.8 individuals/ha) and 3 (1.7 and 3.4 individuals/ha), respectively. In particular, the three most abundant tree species in 1991, Bauhinia forficata, Aloysia virgata and Urera baccifera, all pioneer species, and Croton floribundus, an opportunistic species, were responsible for 75% of recruitment. Coupled with the high recruitment, these species also had low mortality, which resulted in a pronunciated population increasing. In the two lower density classes, 2 and 3, beyond the low recruitment, the pioneer and the opportunistic species presented 74.2% of recruitment. These last groups also presented a greater density gain than shade-tolerant canopy species and shade-tolerant understory species in the density classes 2 and 3. Beyond these results were also done a comparative analysis of this fragment with a fragment of same size ("Santa Rita" fragment -9.5 ha) and a fragment of 86 ha ("Capuava" fragment), both located in Piracicaba region. The "Mata do Pomar" fragment presents less of the half of tree species than "Santa Rita" and "Capuava" fragments, although with the same size of "Santa Rita" fragment. Further, the four most abundant tree species in the "Mata do Pomar" fragment, B. forficata, A. virgata, U. baccifera and T. micrantha are responsible for 62% of individuals, whereas the four most abundant tree species in "Santa Rita" and "Capuava" fragments are responsibles for only 30.6% and 23.2% of individuals, respectively, indicating a high local dominance in the "Mata do Pomar" fragment. This study demonstrates that strategies of conservation biodirversity in semideciduous seazonal forest fragments in the São Paulo state should consider the anthropogenic disturbance in the identification of conservation priorities.
