Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Genotoxicity and mutagenicity of Echinodorus macrophyllus (chapéu-de-couro) extracts

Echinodorus macrophyllus, commonly known as chapéu-de-couro, is a medicinal plant used in folk medicine to treat inflammation and rheumatic diseases. In this work, we used short-term bacterial assays based on the induction of SOS functions to examine the genotoxicity and mutagenicity of an aqueous extract of E. macrophyllus leaves. Whole extract and an ethyl acetate fraction showed similar genotoxicity and caused an ~70-fold increase in lysogenic induction. The extract also gave a positive result in the SOS chromotest with an increase of 12-fold in β-Galactosidase enzymatic units. There was a strong trend towards base substitutions and frameshifts at purine sites in the mutations induced by the extract in Escherichia coli (CC103 and CC104 strains) and Salmonella typhimurium test strains (22-fold increase in histidine revertants in TA98 strain). Since reactive oxygen species may be implicated in aging process and in degenerative diseases, we used antioxidant compounds as catalase, thiourea and dipyridyl in the lysogenic induction test. All this compounds were able to reduce the induction factor observed in the treatment with chapéu-de-couro, thus suggesting that the genotoxicity and mutagenicity were attributable to the production of reactive oxygen species that targeted DNA purines.

Durante as últimas décadas, a disseminação de ações governamentais e os mecanismos criados para proteger os direitos dos consumidores se associaram em crescentes esforços para garantir a qualidade e confiabilidade dos produtos. As amostras de preservativos (látex), dispositivos médicos e bolsas de sangue (PVC - cloreto polivinílico) foram testadas utilizando o ensaio de difusão em ágar. O teste avalia a citotoxicidade induzida por biomateriais, medindo a reatividade biológica de culturas de células de mamífero em contato com tais materiais. PVC é utilizado na produção de dispositivos médicos, devido às suas propriedades específicas, incluindo flexibilidade, obtida após a adição de plastificantes (ftalatos) que podem apresentar toxicidade mesmo em doses baixas. Látex é um elastômero natural utilizado em luvas cirúrgicas e preservativos que na sua formulação inclui a dispersão do látex líquido e de substâncias químicas, como antioxidantes e um acelerador de vulcanização, ambos capazes de induzir citotoxicidade. As amostras foram analisadas pelo Instituto Nacional de Controle de Qualidade em Saúde - INCQS da Fundação Oswaldo Cruz (Fiocruz) em atendimento às ações governamentais de vigilância sanitária no tocante ao controle de qualidade. Um aumento na qualidade dos produtos em relação aos resultados no ensaio de difusão em ágar foi observado no período compreendido entre 2000-2007. Esta situação, dentre outras ações, reflete uma melhoria na qualidade dos produtos que pode ser traduzida em saúde para a população.
Over the last decades, governmental actions and mechanisms created to protect consumer rights have been linked to a growing effort to guarantee the quality and reliability of products. Samples of condoms (latex), medical devices and blood bags (PVC - polyvinyl chloride) have been tested using the agar diffusion assay. This assay evaluates the cytotoxicity induced by biomaterials by measuring the biological reactivity of mammalian cell cultures in contact with these materials. PVC is used in the production of medical devices because of its specific properties, such as flexibility, obtained after the addition of plasticizers (phthalates), which can cause toxicity even at low doses. Latex is a natural elastomer used for surgical gloves and condoms with a formulation that includes dispersion of liquid latex and chemicals, such as antioxidants and a vulcanizing accelerator, both of which are able to induce cytotoxicity. Samples were analyzed by the National Institute of Quality Control in Health - INCQS of the Oswaldo Cruz Foundation (Fiocruz) in accordance with the governmental sanitary surveillance actions on respect to the quality control. We observed an increase in the quality of the products in relation to the results of the agar diffusion assay during the period between 2000 and 2007. This situation, together with other actions, reflects in an improvement in the quality of products that can be translated in the health of the population.

Ligation of the abdominal esophagus decreases scorpion toxin-induced gastric secretion in rats

OBJETIVO: A toxina de escorpião purificada do veneno do escorpião Tityus serrulatus (Tx) induz um aumento no volume, acidez e secreção de pepsina no suco gástrico de ratos. A ligadura do esôfago diminui a secreção ácida do estômago em ratos. O objetivo deste trabalho foi determinar a influência da ligadura do esôfago sobre a secreção gástrica induzida pela Tx em ratos. MÉTODOS: 44 ratos machos, brancos foram administrados água ad libitum, mas não alimentados por 20 a 24 horas, anestesiados com uretana e canulados a traquéia e a veia jugular. Foram realizadas as ligaduras do esôfago cervical ou abdominal ou operações simuladas antes e após a administração na veia jugular de 0,25 mg/kg de toxina de escorpião (fração T1). Uma hora após foram determinados o volume, acidez, pH e atividade péptica do suco gástrico. RESULTADOS: A toxina de escorpião induziu um aumento do volume do suco gástrico, da acidez gástrica e da produção de pepsina e uma diminuição do pH quando injetada na veia de animais não operados ou com operação simulada. A ligadura do esôfago cervical não interferiu nos efeitos da toxina, enquanto a ligadura do esôfago abdominal diminuiu os efeitos da toxina no estômago do rato. CONCLUSÃO: A ligadura do esôfago abdominal diminui a secreção gástrica estimulada pela toxina de escorpião.
PURPOSE: Scorpion toxin purified from Tityus serrulatus venom (Tx) induces an increase in volume, acidity and pepsin secretion in the gastric juice of rats. Ligation of oesophagus has been shown to reduce the acid gastric secretion in rats. The aim of this paper was to determine the influence of the esophageal ligation on gastric secretion induced by Tx in rats METHODS: Forty-four male albino rats were given water ad libitum, but no food for 20 to 24 hours, anesthetized with urethane and the trachea and jugular vein cannulated. Cervical or abdominal esophageal ligation or sham-operations were performed before and after the injection of 0.25 mg/kg of scorpion toxin (fraction T1) into the jugular vein. One hour later, the volume, acidity, pH and peptic activity of gastric juice were determined. RESULTS: The scorpion toxin induced an increase in gastric juice volume, acidity and pepsin output and a decrease in pH when injected into the vein of intact animals or in sham-operated animals. Cervical esophagus ligation did not interfere with the effects of toxin, however, ligation of the abdominal esophageal decreased the toxin effect on the rat stomach. CONCLUSION: Ligation of the abdominal esophagus decreases the gastric secretion induced by scorpion toxin.

Provided its universal susceptibility to penicillin, there was little interest about antimicrobial resistance in Streptococcus pyogenes before 1989. However, the knowledge about some outbreaks of erythromycin-resistant streptococci in Japan and Australia prompted us to begin a surveillance in the hospital and then as multicentre studies involving all the country. The results of such eleven-year surveillance (1989-2000) are being presented. Penicillin-resistant or intermediately-penicillin-susceptible group A streptococci were not found. Resistance to cloramphenicol or tetracycline was not detected. Erythromycin resistance changed from 0 - 2.0% to 9.9% along this period. These rates were due to the increasing isolation of streptococci showing the M phenotype (probably active efflux), some of them showing the mefA genotype. Among isolates obtained from invasive infections erythromycin resistance was not found. Comparing the evolution of erythromycin resistance with the national consumption of macrolides, a similar trend as that published by finnish authors was appreciated.
En 1989 era escasa la preocupación acerca de la resistencia de Streptococcus pyogenes a los antibióticos habida cuenta de su sensibilidad universal a la penicilina. No obstante, algunos brotes puntuales en Japón y Australia de cepas resistentes a macrólidos en las décadas anteriores impulsaron a comenzar con la vigilancia de la resistencia de S. pyogenes a los antibióticos, primero en este hospital y luego en forma multicéntrica junto a otros centros de todo el país. En este trabajo se presentan los resultados de esta vigilancia, realizada durante once años (período 1989-2000). No se encontraron cepas resistentes ni con sensibilidad intermedia a penicilina en estreptococos del grupo A. Tampoco fue detectada resistencia a cloranfenicol ni a tetraciclina. La resistencia a eritromicina ascendió de un 0 - 2,0% a un 9,9% durante el período citado y ese aumento se debió principalmente a aislamientos que presentaban el fenotipo M (probable eflujo), algunos de los cuales demostraron poseer el genotipo mefA. En aislamientos de infecciones invasivas no se detectó resistencia a eritromicina. Al comparar la evolución de la resistencia a eritromicina entre 1990 y 2000 con el consumo nacional de macrólidos se pudo observar una tendencia similar a la publicada previamente por investigadores finlandeses.

Erythromycin (ERY) resistance in Streptococcus pyogenes has recently emerged as a problem of growing concern all through the world. We are presenting the comparison of results of the continuous surveillance of erythromycin resistance in S. pyogenes performed since 1989 in the Hospital de Pediatría J.P.Garrahan of Buenos Aires City, with independently observed rates in other five centers of Buenos Aires and seven centers of six other Argentinian cities, obtained between 1999 and 2001. A significant increase of erythromycin resistance was observed among S. pyogenes isolated in the Hospital Garrahan (6.6% in 1998-1999 to 9.9% in 2000). Similar trends were also detected in other centers of other Argentinian cities when recent data were compared to results of a multicenter study performed in 1995. However, lower rates of resistance were recorded in Mendoza, Cipolletti and Neuquén in comparison with data of 1995, 1998 and 1998 respectively. The reason of such decreasing resistance rates deserves to be investigated. The average of ERY-resistance rates obtained in the surveyed centers was 6.7% (range 0.5 - 14.1%). Control of antimicrobial use should be performed to warrant the future effectiveness of macrolide antibiotics regarding the positive association between use and resistance. These results also suggest that susceptibility tests for macrolides should be performed whenever S. pyogenes is isolated in Argentina.
La resistencia a la eritromicina en Streptococcus pyogenes ha emergido en los últimos tiempos como un problema creciente en todo el mundo. En este trabajo se presenta la comparación de los resultados de la vigilancia continua de la resistencia a la eritromicina que se viene realizando en el Hospital de Pediatría J.P.Garrahan de Buenos Aires desde 1989, con resultados independientes de otros cinco centros de la ciudad de Buenos Aires y siete de otras seis ciudades argentinas, obtenidos entre 1999 y 2001. Se observó un aumento significativo en el Hospital Garrahan (6.6% en 1998-1999 a 9.9% en el año 2000) y una tendencia similar en otros centros de diversas ciudades argentinas si se comparan estos datos con los de un estudio multicéntrico realizado en 1995. No obstante, se registraron menores porcentajes de resistencia en Mendoza, Neuquén y Cipolletti, en relación a lo hallado en 1995, 1998 y 1998 respectivamente. La razón de esta disminución merece ser investigada. El porcentaje promedio de resistencia a eritromicina obtenido en los distintos centros participantes de este estudio fue de 6.7% (rango 0.5-14.1%). Debe efectuarse un control en el uso de estos antibióticos para garantizar la efectividad futura de los macrólidos, teniendo en cuenta la asociación estrecha entre uso y resistencia. Estos resultados sugieren que deberían realizarse pruebas de sensibilidad a los macrólidos para todos los aislamientos de S. pyogenes en la Argentina.
