RESUMO Com o intuito de desenvolver novas medidas de proteção de plantas contra doenças, o objetivo deste trabalho foi verificar o controle da pinta preta (Alternaria solani) e da mancha bacteriana (Xanthomonas vesicatoria) no tomateiro, e a atividade antimicrobiana contra seus agentes causais utilizando pó solúvel de alecrim (Rosmarinus officinalis). Para o teste de estabilidade, o pó solúvel foi armazenado a 40 oC por 0, 60, 120 e 180 dias e testado em cinco concentrações (0, 50, 100, 150 e 200 mg L-1). Definidos o tempo de prateleira e a melhor concentração, avaliaram-se os intervalos de aplicação de sete, 14 e 21 dias. Como tratamentos controle foram utilizados os fungicidas Azoxystrobina (200 mg do ingrediente ativo – i.a. mL-1) para pinta preta e Oxicloreto de cobre (400 mg i.a. mL-1) para mancha bacteriana. Não houve atividade antimicrobiana in vitro do extrato contra A. solani e X. vesicatoria. A concentração calculada de extrato de alecrim que resultou em menor severidade das doenças foi de 130 mg L-1, com tempo de prateleira do pó solúvel de 24 meses. O extrato proporcionou o mesmo nível de controle da pinta preta e da mancha bacteriana que os fungicidas utilizados, com eficiência de até 75%, em intervalo de aplicação de sete dias. Estes resultados demonstram a eficiência do pó solúvel de alecrim para controle da pinta preta e da mancha bacteriana do tomateiro.

ABSTRACT Aiming to develop new measures of plant protection against diseases, the objective of this study was to verify the control of early blight (Alternaria solani) and bacterial spot (Xanthomonas vesicatoria) in tomato plants, as well as the antimicrobial activity against their causal agents, using soluble powder of rosemary (Rosmarinus officinalis). For the stability test, the soluble powder was kept at 40 oC during 0, 60, 120 and 180 days and assayed at five concentrations (0, 50, 100, 150 and 200 mg L-1). When the shelf life and the best concentration were defined, the application intervals of seven, 14 and 21 days were evaluated. As control treatments, the following fungicides were used: Azoxystrobin (200 mg active ingredient – a.i mL-1) for early blight and Copper Oxychloride (400 mg a.i. mL-1) for bacterial spot. There was no in vitro antimicrobial activity of the extract against A. solani and X. vesicatoria. The calculated concentration of rosemary extract that resulted in the least severity of diseases was 130 mg L-1, with 24 months of shelf life of the soluble powder. The extract provided the same control level for early blight and bacterial spot as that of the used fungicides, showing efficiency of up to 75% at a seven-day application interval. These results show the efficiency of rosemary soluble powder in controlling early blight and bacterial spot in tomato plants.

O feijoeiro (Phaseolus vulgaris L.) está entre as principais culturas da agricultura nacional e pode ser afetada por várias doenças, como a mancha angular causada por Pseudocercospora griseola. Com o objetivo de desenvolver métodos alternativos ao químico para o controle desta doença, verificou-se o potencial de extratos de micélio de Pycnoporus sanguineus para atividade antimicrobiana in vitro contra P. griseola e para indução de resistência e ativação de enzimas de defesa em feijoeiro, como peroxidase, polifenoloxidase e β-1,3-glucanase, além de sua influência nos teores de proteínas e clorofilas. Os experimentos in vitro e em casa de vegetação foram constituídos pelos extratos de micélio de P. sanguineus e dos controles água, acibenzolar-S-metil (ASM: 75 mg i.a./L) e fungicida azoxystrobin (40 mg i.a./L). In vitro o extrato de micélio apresentou efeito sobre o crescimento micelial, a esporulação e a germinação de esporos de P. griseola. In vivo a severidade foi reduzida em 93% e 50% em casa de vegetação e a campo respectivamente, em relação ao controle água. A atividade das enzimas peroxidase e polifenoloxidase e os teores de proteínas e clorofilas foram maiores nas plantas tratadas com o extrato. Estes resultados indicam a eficiência de P. sanguineus para o controle alternativo da mancha angular do feijoeiro.

Angular leaf spot, caused by the fungus Pseudocercospora griseola, is a major disease of common bean (Phaseolus vulgaris L.) in Brazil. The objective of this research was to develop an alternative method for controlling this disease based on an aqueous extract of Pycnoporus sanguineus mycelium. It was evaluated the antimicrobial activity of the aqueous extract against P. griseola as well as its activity of resistance induction against angular leaf spot. The role of the plant defense enzymes peroxidase, polyphenol oxidase and β-1,3-glucanase, and the proteins and chlorophyll content was investigated. The experiments in vitro and in greenhouse used extracts from mycelium of P. sanguineus and water, Acibenzolar-S-methyl (ASM: 75 mg/L) and the fungicide azoxystrobin (40 mg/L) were the control treatments. In vitro, the extract of mycelium inhibited the mycelial growth as well as the sporulation and conidia germination of P. griseola. However severity was reduced in 93% and 50% in greenhouse and field respectively. The activities of peroxidase, polyphenol oxidase and the content of proteins and chlorophylls had an increment in plants treated with extract. These results indicate the potential of P. sanguineus for alternative control of the angular leaf spot in bean.

Com o objetivo de desenvolver métodos alternativos para o controle da mancha angular do feijoeiro, causada por Pseudocercospora griseola, filtrados de cultura de Pycnoporus sanguineus foram testados contra P. griseola. Foi investigado ainda o potencial desses filtrados em ativar enzimas de defesa do feijoeiro, como peroxidase, polifenoloxidase e β-1,3 glucanase, bem como alterar teores de proteína total e clorofilas. Para os experimentos in vitro e em casa de vegetação foram utilizados extratos de filtrado de cultura de P. sanguineus em concentrações variando de 1 a 20% e os controles água, acibenzolar-S-metil (ASM: 75 mg i.a. L-1) e fungicida (azoxystrobin: 40 mg i.a. L-1). No experimento in vitro, o extrato não apresentou efeito inibitório sobre o crescimento micelial, na esporulação e na germinação de conídios de P. griseola. A área abaixo da curva de progresso da mancha angular foi reduzida em 82 e 49% respectivamente, nas plantas cultivadas em casa de vegetação e no campo em relação ao controle água. A atividade das enzimas peroxidase e polifenoloxidase e os teores de proteínas e clorofilas foram maiores nas plantas tratadas com o extrato. Estes resultados indicam a eficiência do filtrado de cultura de P. sanguineus para o controle alternativo da mancha angular do feijoeiro.

With the aim of developing an alternative method for the control of angular leaf spot of common bean, caused by Pseudocercospora griseola, aqueous extracts of Pycnoporus sanguineus culture filtrates were evaluated in vitro for antimicrobial activity against P. griseola, and possible induction of resistance in bean against P. griseola. The role of the defense enzymes peroxidase, polyphenol oxidase, and β-1,3-glucanase, and the content of proteins and chlorophyll, were also investigated. For the in vitro and greenhouse experiments the culture filtrate of P. sanguineus at the concentrations of 1 to 20% was used, and water, acibenzolar-S-methyl (ASM: 75 mg i.a. L-1) and fungicide (azoxystrobin: 40 mg i.a. L-1) served as control treatments. In the in vitro study, culture filtrate did not inhibit mycelial growth, sporulation and conidia germination of P. griseola. The total area under the angular leaf spot progress curve was reduced by 82 and 49% respectively, for greenhouse and field experiments in relation to the water control. The activities of peroxidase and polyphenol oxidase, and the content of proteins and chlorophylls, increased in plants treated with the extract. The results from this study indicate the potential of P. sanguineus culture filtrate as an alternative control for angular leaf spot on beans.
