Morphological and molecular characterization of Hysterothylacium spp. parasitizing Pomatomus saltatrix and Pagrus pagrus of the State of São Paulo, Brazil

Abstract Raphidascarid nematodes have been the focus of several studies, mainly due to the zoonotic potential of some species, even though the cases are underreported. Due to the difficulty in identifying their larvae, the use of diagnostic techniques involving morphological and molecular analyses has grown in the last 20 years. The present study had as objective the morphological and molecular characterization of the L3 larval types of Hysterothylacium collected in Pomatomus saltatrix and Pagrus pagrus from the Brazilian coast, close to the municipality of Santos, State of São Paulo. Twenty specimens of P. saltatrix were necropsied and Hysterothylacium type V (n = 257) and Hysterothylacium type X (n = 5) larvae were found. Five specimens of P. pagrus were necropsied and all were parasitized by Hysterothylacium type V larvae. The analyses showed a genetic proximity relationship between Hysterothylacium types V with other Hysterothylacium V and with H. deardorffoverstreetorum, although this is a species inquirenda. Haplotypes for Hysterothylacium type X found in the present study formed a monophyletic group with other Hysterothylacium X, H. amoyense, and H. zhoushanense. Through this study, new hosts and localities were registered for Hysterothylacium type V and Hysterothylacium type X.

RESUMO Alguns fungos fitogênicos sobrevivem nas sementes de cevada, no entanto, o seu período de sobrevivência é desconhecido em condições de armazenamento de sementes cevada no Brasil. O objetivo do trabalho foi quantificar a viabilidade dos fungos Alternaria alternata, Bipolaris sorokiniana e Drechslera teres em sementes de cevada, armazenadas por 315 dias durante o período da entressafra na Região Sul do Brasil. Quatrocentas sementes de cada cultivar foram desinfestadas e submetidas ao teste de sanidade de sementes em meio de cultura batata-dextrose-ágar (BDA), com intervalo de tempo entre avaliação de 35 dias, desde dezembro de 2011 a setembro de 2012. Os dados de incidência e viabilidade dos fungos em função do tempo de armazenamento foram submetidos à análise de regressão. Os três fungos foram detectados nas cinco cultivares em todas as épocas avaliadas, com redução significativa negativa da incidência e viabilidade com o aumento do período de armazenamento. A redução média mensal de viabilidade de A. alternata foi de 8% enquanto de B. sorokiniana e D. teres foi de 10%. No final do período de entressafra a viabilidade dos fungos A. alternata, B. sorokiniana e D. teres, na média das cultivares, manteve-se em 49,8%, 29,6% e 31,0%, respectivamente.
ABSTRACT Some seed-borne fungi can survive in barley seeds; nevertheless, their survival period is unknown in barley seeds under storage conditions in Brazil. The aim of this study was to quantify the viability of the fungi Alternaria alternata, Bipolaris sorokiniana and Drechslera teres in barley seeds stored for 315 days during the off-season in the south region of Brazil. Each cultivar had 400 seeds disinfested and subjected to seed health testing in potato-dextrose-agar (PDA) from December 2011 to September 2012 at 35-day intervals. Data on fungal incidence and viability as a function of the storage time underwent regression analysis. All three fungi were detected in the five cultivars in all evaluated periods, and their incidence and viability significantly reduced with increasing storage periods. Monthly average reduction in the viability of A. alternata was 8%, while that of B. sorokiniana and D. teres was 10%. At the end of the off-season, viability of A. alternata, B. sorokiniana and D. teres, considering the average of cultivars, remained 49.8, 29.6 and 31.0%, respectively.

Molecular characterization of the progenetic metacercariae Crocodilicola pseudostoma parasitizing Rhamdia quelen (Siluriformes, Heptapteridae) in Brazil

Abstract The trematodes have developed several adaptations and strategies to complete their life cycle in the intermediate host, without even reaching the definitive host. Thus, metacercariae through progeny can produce viable eggs by self-fertilization in the second intermediate host. We analyzed 30 specimens of Rhamdia quelen Quoy & Gaimard 1824 (Siluriformes, Heptapteridae) collected in the Jacaré-Pepira River, Ibitinga. Among the specimens analyzed, only one host was parasitized by the progenetic metacercariae of Crocodilicola pseudostoma Willemoes-Suhm 1870 (Digenea: Proterodiplostomidae) presenting prevalence of 3.3%, mean intensity of 68.0 ± 12.4 and mean abundance of 2.3 ± 0.4. This is the first record of progenesis in the metacercariae of C. pseudostoma in the Jacaré-Pepira River, as well as the first partial sequence of COI gene obtained from this species in Brazil.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Decolorization of textile dyes by cyanobacteria

Cianobactérias estão amplamente distribuídas no ambiente e podem ser uma alternativa eficaz e econômica para a remoção de corantes de efluentes de indústrias têxteis. O presente trabalho investigou o potencial de seis linhagens de cianobactérias de descolorir onze tipos de corantes têxteis. A absorção máxima de cada corante foi verificada usando um espectrofotômetro. Espectrometria de massas foi utilizada na verificação da degradação e possível remoção de corantes pelas cianobactérias. Os resultados mostraram que todas as linhagens avaliadas foram capazes de remover indigo, amarelo palanil, amarelo indantreno, azul indantreno, azul dispersol, vermelho indantreno e vermelho dispersol em mais de 50%. O isolado brasileiro Phormidium sp. CENA135 foi capaz de descolorir e remover completamente o índigo azul BANN 30. Este estudo confirmou a capacidade das cianobactérias de descolorir e, possivelmente, degradar corantes têxteis estruturalmente diferentes, sugerindo a possibilidade de sua aplicação em estudos de biorremediação.
Cyanobacteria are widely distributed in the environment and may be an effective and economic alternative for removing dyes from textile industry effluents. The present work investigated the potential of six cyanobacterial strains in decolorizing eleven types of textile dyes. The maximum absorbance of each dye was verified using a spectrophotometer. Mass spectrometry was used to verify the removal and possible degradation of dyes by the cyanobacteria. The results showed that all of the evaluated cyanobacteria were able to remove indigo, palanil yellow, indanthrene yellow, indanthrene blue, dispersol blue, indanthrene red and dispersol red by more than 50%. The Brazilian isolate Phormidium sp. CENA135 was able to decolorize and completely remove indigo blue BANN 30. This study confirmed the capacity of cyanobacteria to decolorize and possibly to structurally degrade different textile dyes, suggesting the possibility of their application in bioremediation studies.

The diversity, extracellular enzymatic activities and photoprotective compounds of yeasts isolated in Antarctica

The diversity of yeasts collected from different sites in Antarctica (Admiralty Bay, King George Island and Port Foster Bay and Deception Island) and their ability to produce extracellular enzymes and mycosporines were studied. Samples were collected during the austral summer season, between November 2006 and January 2007, from the rhizosphere of Deschampsia antarctica, ornithogenic (penguin guano) soil, soil, marine and lake sediments, marine water and freshwater from lakes. A total of 89 isolates belonging to the following genera were recovered: Bensingtonia, Candida, Cryptococcus, Debaryomyces, Dioszegia, Exophiala, Filobasidium, Issatchenkia (Pichia), Kodamaea, Leucosporidium, Leucosporidiella, Metschnikowia, Nadsonia, Pichia, Rhodotorula, and Sporidiobolus, and the yeast-like fungi Aureobasidium, Leuconeurospora and Microglossum. Cryptococcus victoriae was the most frequently identified species. Several species isolated in our study have been previously reported to be Antarctic psychophilic yeasts, including Cr. antarcticus, Cr. victoriae, Dioszegia hungarica and Leucosporidium scottii. The cosmopolitan yeast species A. pullulans, C. zeylanoides, D. hansenii, I. orientalis, K. ohmeri, P. guilliermondii, Rh. mucilaginosa, and S. salmonicolor were also isolated. Five possible new species were identified. Sixty percent of the yeasts had at least one detectable extracellular enzymatic activity. Cryptococcus antarcticus, D. aurantiaca, D. crocea, D. hungarica, Dioszegia sp., E. xenobiotica, Rh. glaciales, Rh. laryngis, Microglossum sp. 1 and Microglossum sp. 2 produced mycosporines. Of the yeast isolates, 41.7% produced pigments and/or mycosporines and could be considered adapted to survive in Antarctica. Most of the yeasts had extracellular enzymatic activities at 4ºC and 20ºC, indicating that they could be metabolically active in the sampled substrates.
