Crotalaria ochroleuca (Fabaceae) is rich in bioactive compounds used for nematode control. Transformed root culture allows the production of a large amount of hairy root biomass, conditioned by auxin biosynthesis-related genes integrated via the process of Agrobacterium rhizogenes infection, which is known to cause a hairy root phenotype. We transformed A. rhizogenes to increase the biomass of hairy roots and optimized the process of production of bioactive compounds. The transformed nature of hairy roots was confirmed by polymerase chain reaction, which confers kanamycin resistance. The transformed roots were cultured in full-strength in liquid media to realize the growth of hairy roots and the production of bioactive compounds. The transformed roots were grown in a culture medium supplemented with elicitors to produce total phenols, and flavonoids. Chromatographic analysis of transformed roots revealed the presence of flavonoids apigenin-6,8-C-diglucoside and luteolin-6-C-glucoside. The results were obtained by conducting physiological and biochemical studies with the Flavonoids and studying the pathways that led to the production of large amounts of bioactive compounds from the hairy roots of C. ochroleuca. It was observed that the extraction of the compounds significantly affected nematodes and insect larvae, resulting in significantly high levels of economic damage to crops.

Abstract The chemical study of roots from Azadirachta indica A. Juss., Meliaceae, led to the isolation of two new terpenoids, limonoid morenolide and diterpene 17-hydroxy-sandaracopimar-8,15-dien-11-one, in addition to the four well-known limonoids nimbinene, nimbinal, nimbandiol and salannin, and three diterpenoids nimbidiol, ferruginol, and 6,7-dehydroferruginol. Their structural elucidations were based on one and bidimensional Nuclear Magnetic Resonance and Electrospray ionization mass spectrometry spectra data which was compared to the data found in literature. The anti-inflammatory, cytotoxic and antimycobacterial activities of the identified terpenoids were evaluated.

RESUMO Uma nova espécie de Pyrrhulina é descrita baseada em 60 espécimes dos afluentes do rio Amazonas: rio Anapu, rio Capim, rio Guamá e rio Xingu, e drenagens costeiras no estado do Pará, Brasil. A nova espécie distingue-se de todas as congêneres por ter a faixa primária (faixa horizontal de pigmentação escura que se estende posteriormente do focinho) terminando na borda distal do opérculo. Em todas as outras espécies de Pyrrhulina, a faixa primária é restrita ao focinho ou continua além da cabeça (para incluir pelo menos as quatro escamas anteriores da série da linha lateral).

ABSTRACT A new species of Pyrrhulina is described based on 60 specimens from tributaries of the rio Amazonas: rio Anapu, rio Capim, rio Guamá and rio Xingu, and coastal drainages in the state of Pará, Brazil. The new species differs from all congeners by having the primary stripe (the horizontal stripe of dark pigmentation extending posteriorly from the snout) terminating at the distal edge of the opercle. In all other species of Pyrrhulina, the primary stripe is either restricted to the snout or continues beyond the head (i.e. at least the anteriormost four scales of the lateral line series).

Ácidos galoilquínicos, substâncias que têm apresentado atividade leishmanicida, foram isolados do extrato de acetato de etila da planta Byrsonima coccolobifolia. Estes compostos fenólicos juntamente com o ácido gálico demonstraram ser uma nova classe de inibidores não-competitivos potentes em arginase (ARG) de Leishmania amazonensis (Ki variando de 0,10 a 0,68 µmol L-1). O ácido quínico não apresentou atividade inibitória significativa em ARG demonstrando que a unidade galoila tem características importantes que permitem a interação enzima-inibidor. A atividade inibitória significativa do ácido gálico frente à ARG pode ser uma indicação para o entendimento da resposta imune previamente observada em Leishmania donovani, uma vez que a atividade enzimática da arginase está associada à diminuição dos níveis de NO no processo de infecção por Leishmania.

Leishmanicidal galloylquinic acids were isolated from the ethyl acetate extract of Byrsonima coccolobifolia. These phenols and gallic acid showed to be a new class of potent noncompetitive inhibitors of arginase ARG (Ki ranging from 0.10 to 0.68 µmol L-1) from Leishmania amazonensis. Quinic acid did not exhibit significant inhibition of ARG, indicating that galloyl moiety has important features that allows the enzyme-inhibitor interactions. The significant inhibitory activity of gallic acid on ARG can be a clue to understand the immune response previously observed on L. donovani, since ARG activity is associated with the decrease of the levels of NO in Leishmania infection.

Gestational depression is detrimental to the health of the mother and the offspring and contributes to the appearance of depressive and anxiety symptoms during the postnatal period. Traditional antidepressants have undesirable side effects when utilised during gestation, but Hypericum perforatum has been characterised as an efficient and safe antidepressant that prevents the recurrence of symptoms. This study verified the effects of Hypericum perforatumon the behaviour of Wistar rats that were treated during gestation and evaluated 10 and 60 days post-treatment. Pregnant Wistar rats were divided into four groups of ten animals each: one control group that received distilled water and three treatment groups that were treated orally with 36, 72 or 144 mg/kg Hypericum perforatumextract. At 10 and 60 days after parturition and post-treatment, the rats were submitted to the holeboard, the tail suspension, and the forced swim tests. The animals treated with 144 mg/kg Hypericum perforatumexhibited greater head-dipping activity in the hole-board test and reduced immobility in the tail suspension and forced swim tests, suggesting less anxiety and depression 10 and 60 days post-treatment.The results indicated that treating rats with Hypericum perforatumduring the gestational period decreased depressive behaviour and anxiety 10 and 60 days post-treatment.

Solanum paniculatum L., é uma planta com ocorrência em toda a América tropical, especialmente no Cerrado. No Brasil, é utilizada para fins culinários e, na medicina popular, para o tratamento de distúrbios gástricos e hepáticos, além de ressacas. Estudos com extratos de S. paniculatum demonstraram ausência de genotoxicidade em testes com camundongos e bactérias, apesar de promoverem citotoxicidade e antigenotoxicidade em altas dosagens. No intuito de detectar os compostos responsáveis pelas atividades observadas, o extrato etanólico dos frutos de S. paniculatum foi fracionado, os alcalóides esteroidais obtidos foram caracterizados por espectroscopia de ressonância magnética nuclear, e sua ação protetora contra mitomicina C (MMC) foi avaliada utilizando o teste do micronúcleo em medula óssea de camundongos. Camundongos foram tratados via gavage com diferentes concentrações (25, 50 ou 100 mg.kg−1) de cada fração simultaneamente com uma dose intraperitonial de MMC (4 mg.kg−1). A antigenotoxicidade foi avaliada pela frequência de eritrócitos policromáticos micronucleados (EPCMN), enquanto a anticitotoxicidade, pela relação entre eritrócitos policromáticos e normocromáticos (EPC/ENC). Os resultados demonstraram que os alcalóides esteroidais isolados de S. paniculatum protegeram as células contra a ação aneugênica e/ou clastogênica de MMC, assim como, modularam sua ação citotóxica.

Solanum paniculatum L. is a plant species widespread throughout tropical America, especially in the Brazilian Cerrado region. It is used in Brazil for culinary purposes and in folk medicine to treat liver and gastric dysfunctions, as well as hangovers. Previous studies with S. paniculatum ethanolic leaf extract or ethanolic fruit extract demonstrated that they have no genotoxic activity neither in mice nor in bacterial strains, although their cytotoxicity and antigenotoxicity were demonstrated in higher doses. In order to assess the possible compounds responsible for the activities observed, we fractionated the ethanolic fruit extract of S. paniculatum, characterized by 1H and 13C NMR spectra, and evaluated two fractions containing steroidal alkaloids against mitomycin C (MMC) using the mouse bone marrow micronucleus test. Swiss mice were orally treated with different concentrations (25, 50, or 100 mg.kg−1) of each fraction simultaneously with a single intraperitonial dose of MMC (4 mg.kg−1). Antigenotoxicity was evaluated by using the frequency of micronucleated polychromatic erythrocytes (MNPCE), whereas anticytotoxicity was assessed by the polychromatic and normochromatic erythrocytes ratio (PCE/NCE). Our results demonstrated that steroidal alkaloids isolated from S. paniculatum strongly protected cells against MMC aneugenic and/or clastogenic activities as well as modulated MMC cytotoxic action.

OBJETIVO: Avaliar a influência das posições prona e supina em recém-nascidos prematuros pós-síndrome do desconforto respiratório, respirando espontaneamente e em estado de sono ativo, sobre variáveis de padrão respiratório, movimento toracoabdominal e saturação periférica da hemoglobina pelo oxigênio. MÉTODOS: Estudo quase experimental. Doze prematuros com peso > 1.000 g no momento do estudo foram estudados nas duas posições, em ordem randomizada. A pletismografia respiratória por indutância foi utilizada para avaliação do padrão respiratório (volume corrente, frequência respiratória, ventilação minuto, fluxo inspiratório médio) e do movimento toracoabdominal (índice de trabalho respiratório, relação de fase inspiratória, relação de fase expiratória, relação de fase respiratória total e ângulo de fase). A oximetria de pulso registrou a saturação periférica de oxigênio. Para a análise estatística foram realizados os testes t de Student para amostras pareadas ou Wilcoxon. Foi considerado significativo p < 0,05. RESULTADOS: Foram analisados 9.167 ciclos respiratórios. Na posição prona, houve redução significativa do índice de trabalho respiratório (-0,84±0,69; p = 0,001; IC95% -1,29 a -0,40), das relações de fase inspiratória (-27,36±17,55; p = 0,000; IC95% -38,51 a -16,20), expiratória (-32,36±16,20; p = 0,000; IC95% -42,65 a -22,06) e total (-30,20±14,76; p = 0,000; IC95% -39,59 a -20,82). Não houve diferença significativa entre as posições nas demais variáveis analisadas. CONCLUSÃO: A posição prona promoveu diminuição significativa da assincronia toracoabdominal, sem influenciar o padrão respiratório e a saturação periférica de oxigênio.

OBJECTIVE: To assess the effect of prone and supine positions on breathing pattern variables, thoracoabdominal motion and peripheral oxygen saturation of hemoglobin of premature newborn infants recovering from respiratory distress syndrome, while breathing spontaneously and in rapid eye movement sleep. METHODS: This was a quasi-experimental study. Twelve preterms weighing > 1,000 g at enrollment were studied in both positions, in random order. Respiratory inductive plethysmography was used to analyze breathing pattern (tidal volume, respiratory rate, minute ventilation, mean inspiratory flow) and thoracoabdominal motion (labored breathing index, phase relation in inspiration, phase relation in expiration, phase relation in total breath and phase angle). Pulse oximetry was used to evaluate peripheral oxygen saturation. Student's t test for paired samples or the Wilcoxon test were used for statistical analysis. Significance was set at p < 0.05. RESULTS: A total of 9,167 respiratory cycles were analyzed. The prone position was associated with significant reductions in labored breathing index (-0.84±0.69; p = 0.001; 95%CI -1.29 to -0.40), phase relation in inspiration (-27.36±17.55; p = 0.000; 95%CI -38.51 to -16.20), phase relation in expiration (-32.36±16.20; p = 0.000; 95%CI -42.65 to -22.06) and phase relation in total breath (-30.20±14.76; p = 0.000; 95%CI -39.59 to -20.82). There were no significant differences between the two positions in any of the other variables analyzed. CONCLUSION: The prone position resulted in a significant reduction in thoracoabdominal asynchrony, without affecting breathing pattern or peripheral oxygen saturation.

O objetivo deste trabalho foi avaliar o efeito do óleo essencial de Eucalyptus globulus (OEEG) sobre a eclosão e desenvolvimento de larvas de Haemonchus contortus. A determinação da composição química do OEEG foi feita por meio de cromatografia gasosa acoplada à espectrometria de massa. No teste de eclosão de ovos (TEO), utilizaram-se as concentrações 21,75; 17,4; 8,7; 5,43 e 2,71 mg.mL-1; e no de desenvolvimento larvar (TDL) as concentrações foram 43,5; 21,75; 10,87; 5,43 e 2,71 mg.mL-1. Cada ensaio foi acompanhado por um controle positivo, 0,02 mg.mL-1 tiabendazol, no TEO e 0,008 mg.mL-1 ivermectina no TDL, e por um controle negativo com o diluente Tween 80 (3%). A eficácia máxima obtida pelo OEEG sobre ovos foi de 99,3% na concentração de 21,75 mg.mL-1 e, sobre larvas, 98,7% na concentração de 43,5 mg.mL-1. A concentração do OEEG, que inviabilizou 50% dos ovos e das larvas do parasito, foi de 8,3 e 6,92 mg.mL-1, respectivamente. A análise química do óleo identificou como principal componente o monoterpeno 1,8-cineol. O OEEG apresentou atividade ovicida e larvicida in vitro sobre H. contortus, determinando um bom potencial para utilização no controle de nematóides gastrintestinais de ovinos e caprinos.

The objective of this work was to evaluate ovicidal and larvicidal effects of Eucalyptus globulus essential oil (EGEO) on Haemonchus contortus. The chemical composition determination of EGEO was through gas chromatography and mass spectrometry. Egg hatch test (EHT) was performed in concentrations 21.75; 17.4; 8.7; 5.43 e 2.71 mg.mL-1. In larval development test (LDT) were used the concentrations 43.5; 21.75; 10.87; 5.43 e 2.71 mg.mL-1. Each trial was conducted by negative control with Tween 80 (3%) and positive control, 0.02 mg.mL -1 of thiabendazole in EHT and 0.008 mg.mL-1 of ivermectin in LDT. The maximum effectiveness of EGEO on eggs was 99.3% in concentration of 21.75 mg.mL-1 and on larvae was 98.7% in concentration 43.5 mg.mL-1. The concentration of EGEO that inhibits 50% of the eggs and larvae was 8.3 and 6.92 mg.mL-1, respectively. The oil chemical analysis identified as main component the monoterpen 1,8-cineol. EGEO presented ovicidal and larvicidal activities in vitro, revealing a good potential for use in the control of sheep and goat gastrointestinal nematodes.

INTRODUCTION: Burns and electric shock are severe accidents that can cause irreversible results and death. OBJECTIVE: This study intended to identify mothers' opinions about risk factors in the household associated with burns and shock to which children are exposed. We also verified the correlation between demographic variables and risk factors for burns and shock. METHOD: This is a descriptive study, using a quantitative approach. We conducted household visits, involving 87 families with children aged 1 to 6 years old, who had attended the nursery care center, in Fortaleza, state of Ceará, Brazil, during 2004 to 2006, talking to them and filling out a thematic form. RESULTS: Saucepans within the reach of children were the most significant burn risk factor referred by mothers (34) and by the researcher (64). For electric shock, exposed electric outlets were said to be the most important risk factor by mothers (33) and by the researcher (53). CONCLUSION: Nurses should inform family members how to prevent accidents, therefore reducing accidents in childhood.

INTRODUCCIÓN: Las quemaduras y choques eléctricos son accidentes graves que pueden tener consecuencias irreversibles e incluso llevar a la muerte. OBJETIVO: Identificar la presencia de factores de riesgo para quemaduras y choques eléctricos, según las madres y el investigador, en niños de primera infancia y analizar si tales riesgos están vinculados a los datos sociodemográficos. MÉTODO: se trata de un estudio descriptivo con enfoque cuantitativo. Se visitaron 87 familias en sus domicilios con niños entre uno y seis años que iban a una guardería en Fortaleza, Estado de Ceará. El estudio se realizó entre los años 2004 y 2006 con un formulario que trataba del asunto. RESULTADOS: el factor de riesgo más mencionado, tanto por las madres como por el investigador, fue para quemaduras las ollas y para choques eléctricos las tomas eléctricas o enchufes no protegidos al alcance de los niños. CONCLUSIÓN: enfermería debe orientar a los familiares cómo prevenir accidentes, minimizando los riesgos de accidentes en la infancia.

OBJETIVO: Identificar a presença de fatores de riscos para queimaduras e choque elétrico, na opinião das mães e do pesquisador, em crianças da primeira infância, e verificar a associação com os dados sociodemográficos das mães. MÉTODO: Estudo descritivo e abordagem quantitativa. Foram realizadas visitas domiciliares a 87 famílias de crianças de entre 1 e 6 anos que freqüentavam creche, em Fortaleza-CE, no período de 2004-2006, utilizando um formulário sobre o tema. RESULTADOS: O fator de risco mais citado para queimaduras, observado pelas mães (34) e pelo pesquisador (64), foi: panelas ao alcance das crianças, observado pelas mães (34) e pelo pesquisador (64); para choque, o fator de risco mais observado pelas mães (33) e pelo pesquisador (53) foi tomada elétrica não protegida e ao alcance das crianças, observado pelas mães (33) e pelo pesquisador (53). CONCLUSÃO: A enfermagem deve orientar familiares na prevenção de acidentes, minimizando os riscos de acidentes na infância.
