Compararam-se a sensibilidade dos métodos padrão de sedimentação fecal (MPSF) e modificado da tamisação e coloração das fezes (MTCF)-quatro tamises, ambos empregados no diagnóstico de infecção por Fasciola hepatica. Amostras de fezes foram obtidas de 51 bovinos oriundos de uma área endêmica por fasciolose no Sudoeste da Argentina. Cada amostra foi colocada em um recipiente contendo formalina 5%. Oito mililitros da suspensão, equivalente a 2g de fezes, foram usados em cada método testado, registrando-se o número de ovos por amostra. A porcentagem de amostras positivas pelo MTCF (27/51) foi maior que a apresentada pelo MPSF (11/51), o que representou 60% de amostras falso-negativas. A porcentagem de concordância pelos dois métodos foi 41%. A complexidade do MPSF pode diminuir sua sensibilidade pela perda de ovos durante o processamento. Estes resultados confirmaram que o MPSF subestima a contagem de ovos e que o método MTCF é mais confiável.

The sensitivity and utility of a standard faecal sedimentation method (FSM) and a modified stool sieving staining method (FSSM), both currently employed for the diagnosis of Fasciola hepatica infection were compared. Faecal samples were obtained from 51 bovines of an endemic area for fasciolosis in Southwestern Argentina. Each sample was placed in a recipient containing 5% formalin. Eight millilitres of the suspension, equivalent to 2g of faeces, were used for each of the two methods tested. The number of eggs found per sample was recorded. The proportion of positive samples obtained by the FSSM (27/51) was significantly higher than that by the FSM (11/51) (P<0.05). The percent of agreement between methods was 41%. Over a total of 27 positive samples detected by the FSSM, the FSM missed 16, yielding 60% false negative samples. The FSSM enhanced 2.5 times the sensitivity of diagnosis. The complexity of the FSM may decrease its sensitivity through missing and loss of eggs during sample processing. These results confirmed that the commonly used FSM underestimates the prevalence and the egg output in cattle and that the FSSM is a more reliable diagnostic method.

This paper reports on the standardization of four serological reactions currently used in human serodiagnosis for the detection of anti-Trypanosoma cruzi antibodies in naturally and experimentally infected dogs. Indirect immunofluorescence test (IFAT) and hemagglutination test (IHAT) were standardized, and complement fixation test (CFT) and direct agglutination test (DAT) were used for diagnostic confirmation. Four hundred and eighty one mongrel dogs that were studied by xenodiagnosis were used: (1) parasitemic dogs of two localities of endemic area (EA) of Santiago del Estero province in Argentina (n = 134); (2) non-parasitemic dogs of the same area (n = 285); (3) dogs experimentally infected with T. cruzi in the patent period (n = 6); (4) non-infected dogs (n = 56) which were born in the city of Buenos Aires (BA), one non-EA for Chagas' disease. For IFAT, parasitemic dogs EA showed 95% of reactive sera. Non parasitemic dogs EA showed 77% of non reactive sera. None sera from BA were reactive for dilutions higher than four. For IHAT, 84% of sera of parasitemic dogs EA showed serological reactivity and among non parasitemic dogs BA, 61% were non reactive, while the remainder showed at most titres of 1/16. The cut-off titres for IFAT and IHAT were 1/16 and 1/32 respectively, and for CFT and DAT 1/1 and 1/128 respectively. Sensitivity for IFAT, IHAT, CF and DAT were 95%, 84%, 97% and 95% respectively.

Treze comunidades de 7 províncias argentinas foram escolhidas para avaliação de sorologia como indicador da transmissão da Doença de Chagas. Das comunidades mencionadas, seis não tinham história prévia de tratamento com inseticidas e sete tinham recebido tratamento esporádico ou continuado. Vinte por cento dos moradores das casas de cada localidade foram estudados por sorologia. As amostras foram obtidas por punção da ponta do dedo e 50 microlitros de sangue foram misturadas com 150 microlitros de uma solução conservadora de glicerina a 50% em meio de cultivo, para serem estudados por hemaglutinação indireta, e imunofluorescência indireta. Nas áreas não tratadas a prevalência da infecção em crianças de 0-4 anos foi de 17,5% chegando a mais de 22% para as de 5-9 anos e a 33,3% no grupo 10-14 anos. A prevalência nas áreas tratadas e sob vigilância foi de 2.6% em crianças de 0-4 anos, 5,4% anos de 5-9 anos e de 6,2% em jovens de 10-14 sendo as diferenças entre os dois tipos de áreas estatisticamente significativas (< 0,005). Este estudo propõe a sorologia como um indicador válido para a avaliação da transmissão da Doença de Chagas em áreas rurais.

Thirteen communities from 7 Argentinian provinces were selected for the evaluation of serology as an indicator of transmission of Chagas disease. Of the communities appraised, 6 did not have a history of previous treatment with insecticides and 7 had received sporadic or continuous insecticide treatment. The inhabitants of 20% of the houses of each locality were studied by serology. The samples were obtained byfinger pricking and 50 fil of blood were mixed with 150μl of 50% glycerine solution in tissue culture media to be assayed by Indirect Hemagglutination and Indirect Immunofluorescence tests. In untreated areas, the prevalence of infection in infants 0-4 years old was 17.5%, reaching to over 22% for the 5-9 year old group, and to 33.3% in 10-14 year old individuals. The prevalence in treated and surveyed areas was 2.6% in 0-4 year old children, 5.4% in 5-9 year old and 6,2% in 10-14 year old youngsters. The differences between both areas were statistically significant (p < 0.005). This study favors serology as a valid indicator for the evaluation of transmission of Chagas disease in rural areas.
