ABSTRACT Two experiments were conducted to evaluate the effects of β-mannanase on egg production performance, egg quality, intestinal microbiota, viscosity, and ammonia concentration in laying hens. In Exp. 1, two hundred and seventy 30-wk-old Hy-Line Brown laying hens were assigned to 6 diets arranged in a 3 × 2 factorial of three levels of MEn and CP [(a corn-soybean meal based diet (HEHP), a diet containing 50 kcal of MEn/kg and 1.0% less energy and CP than the HEHP (MEMP), and a diet containing 80 kcal of MEn/kg and 1.5% less energy and CP than HEHP (LELP)], and β-mannanase supplementation (0 or 0.04%). In Exp. 2, A total of two hundred and sixteen62-wk-old Hy-Line Brown laying hens were assigned to 6 dietary treatments in a 3 × 2 factorial arrangement. In Exp. 1, β-mannanase supplementation increased egg production rate in hens fed LELP diet, but not in those fed HEHP or MEMP diet (interaction, p<0.01), and the interaction was significant (p<0.01) for egg mass. β-mannanase supplementation decreased (p<0.05) ammonia concentration. In Exp. 2, the supplementation of β-mannanase increased egg production rate and egg mass in hens fed LELP diet, whereas no differences were found in those fed HEHP or MELP diet (interaction, p<0.01). The supplementation of β-mannanase in a lower energy and protein diet resulted in similar production performance when compared to high-energy and high-protein diet during early and late stages of egg production.

ABSTRACT Salmonella enterica subspecies enterica serotype Enteritidis (SE) has caused foodborne infections over decades. It is transmitted mainly from contaminated eggs to humans. SE is commonly present in layer houses, and closely interacts with environmental factors. The objective of the present study was to develop a viable PCR method to identify SE in environmental samples collected in layer farms of different sizes, and to evaluate SE contamination status in four main egg-production provinces of northern China. After specificity retrieval using Primer-BLAST against the NCBI database, three SE specific oligonucleotide primers were selected as candidate primers. The primers targeting Prot6e gene were adopted and primers targeting Sdf I were also selected to validate the results, after testing eight different types of pooled poultry environmental samples (overshoe, air, drinking nipple, feed, egg collection belt, eggshell, air inlet, and air outlet) by PCR. A PCR detection limit of 1 CFU/mL was determined using cell lysates from pure cultures. Testing time was less than 48 h. On-farm samples were collected from two layer farm sizes (one housing more than 50,000 layers, and the other, less than 50,000 layers) in each province. The applied PCR method was shown to be simple, inexpensive and effective for screening SE in a large amount of farm samples. The study identified only one SE-positive farm, which a large farm and where nine samples were found to be contaminated with SE: drinking nipples (3), egg collection belt (1), air inlet (1), air (1), overshoe (1) and eggshell (2).

In previous studies, we showed that the C/N ratio of the first residue added influences soil respiration, microbial biomass and nutrient availability after the second residue addition which we refer to as legacy effect. The aim of the study was to investigate the effect of residue addition frequency on microbial activity and nutrient availability, and the legacy effect. In Part A, the effect of high C/N residue (H) addition frequency in the first 16 days (d) was studied by adding H to soil once (d0), twice (d0, 8) or four times (d0, 4, 8, 12) followed by 10 g kg-1 low C/N residue (L) on day 16. The influence of L addition rate was assessed in Part B by adding H once on day 0 followed by L added once (d16), twice (d16, 24) or four times (d16, 20, 24, 24, 28). Cumulative respiration, microbial biomass C, N and P, N and P availability from d0 to d15 were higher with L than H. Particularly with L, cumulative respiration, microbial biomass and nutrient availability compared to single addition were lower on d7 with residue added twice or four times, but higher on d15. In Part A, H added twice or four times from d0 to d15 compared to a single H addition followed by a single L addition on d16 increased cumulative respiration, microbial nutrient uptake and reduced N and P availability on d23, but had no effect on d32. In Part B, frequent L addition after d16 enhanced microbial nutrient uptake on d23 but reducing nutrient availability compared to single L addition. It can be concluded that residue addition frequency strongly influences soil respiration, microbial biomass and nutrient availability after addition of low C/N residue.

ABSTRACTThis experiment was conducted to evaluate the combination effect of low dietary non-phytate phosphorus (NPP) concentrations, phytase (PHY) levels, and 25-hydroxycholecalciferol (25-OH-D3) levels on the growth performance and meat quality of broilers. Two levels of NPP, two levels of PHY, and two levels of 25-OH-D3 resulted in a 2�2�2 factorial arrangements, with eight treatments (TRT). The birds on TRT 1-4 were fed diet 1 (NRC NPP was reduced by 0.1) and the birds on TRT 5-8 were fed with diet 2 (NRC NPP was reduced by 0.2). Each diet was mixed with different levels PHY and 25-OH-D3. Performance and meat quality parameters were measured. Results showed that during entire experiment the most advantageous effects were obtained with TRT 3 (NRC NPP reduced by 0.1 + 600 U/kg phytase + 34.5�g/kg 25-OH-D3) and TRT 4 (NRC NPP reduced by 0.1 + 600 U/kg phytase + 69�g/kg 25-OH-D3). The lowest body weight gain (BWG) and feed intake(FI) were observed with TRT 5 (NRC NPP reduced by 0.2 + 300 U/kg phytase + 34.5�g/kg 25-OH-D3). Lowering NRC NPP by 0.1 to 0.2 significantly reduced weight gain (WG) (p< 0.05) and FI (p< 0.05) during the starter phase (ST), while during grower phase (GF) lowering NRC NPP by 0.1 to 0.2 did not affect WG (p>0.05) and produced small decrease in FI. BWG, FI and feed conversion ratio were not influenced (p>0.05) by different PHY or 25-OH-D3 levels. In addition, the meat color, pH, and shear force were not affected by the different NPP, PHY or 25-OH-D3levels.

The genetic diversity and phylogenetic relationships in the genus Secale L. (rye) was evaluated using 24 Secale cereale microsatellite (SCM) markers. The average polymorphism information content (PIC) value of each microsatellite locus in 30 Secale accessions evaluated was higher than that in 47 cultivated ryes (Secale cereale ssp. cereale). The mean genetic similarity (GS) index in Secale was lower than that in cultivated rye. The highest within-species GS index was observed for S. sylvestre and the lowest for S. strictum, whereas the highest between-species GS index was found between S. cereale and S. vavilovii and the lowest between S. sylvestre and S. cereale. There was no obvious difference in GS levels in the cultivated rye accessions from Asia, Europe, North America or South America. Cluster analysis indicated that all the Secale accessions could be distinguished by the 24 microsatellite loci. We also found that the S. sylvestre accessions were obviously divergent from the accessions of other species and that the S. vavilovii accessions were closely related to the S. cereale accessions. Our results also showed that S. strictum was heterogeneous and showed great within-species differences. The microsatellite-derived dendrogram faithfully reflected the phylogenetic relationships between Secale species but did not indicate a possible domestication process of the cultivated rye based on the geographical sources of the accessions.
