This work describes a green synthesis, the characterization, and biological evaluation of silver nanoparticles (AgNPs). The AgNPs suspension was synthesized using aqueous leaf extract of Eucalyptus grandis, which presented a characteristic band at 407 nm in the UV-Vis spectrum. The AgNPs presented a spherical shape and size of 9.7 ± 0.3 nm. The nanoparticles were stable over a month, indicating that E. grandis’ extract is suitable for their preparation and stabilization. The X-ray analysis showed that the crystallinity of AgNPs corresponded to the centered face phase of silver. The antibacterial and leishmanicidal activities of AgNPs were evaluated. The AgNPs presented antibacterial activity on the Gram-negative bacteria Escherichia coli at 53.9 µg mL-1. The leishmanicidal activity evaluation against promastigote forms of Leishmania infantum, Leishmania amazonensis, and Leishmania braziliensis showed that the biological response is dependent on the volume of AgNP suspension. It was demonstrated that L. infantum was more sensitive to the nanoparticle’s treatment than L. amazonensis and L. braziliensis. The treatment of L infantum promastigotes with 150 µL of AgNP suspension reduced parasite growth by 67.9%, a result which was similar to the treatment with 1 (66.7%) or 2 µL (70.6%) of amphotericin B used as a positive control.

RESUMO Introdução: Um número crescente de mutações causando formas monogênicas de doença de Parkinson (DP) tem sido descrito, principalmente entre pacientes da Europa e da América do Norte. Como a arquitetura genética varia entre diferentes populações, entender os perfis genéticos específicos de pacientes brasileiros é essencial para um melhor aconselhamento genético e para a seleção de participantes para ensaios clínicos. Objetivo: Revisão sistemática para identificar estudos genéticos brasileiros na área e definir o cenário para estudos futuros das formas monogênicas de DP no Brasil. Métodos: Nós pesquisamos as bases de dados MEDLINE, EMBASE e Web of Science desde a criação até dezembro de 2019, usando termos para “Parkinson’s disease”, “genetics” e “Brazil”. A extração de dados foi feita por dois revisores independentes. Para os genes LRRK2 e PRKN, calculamos a prevalência estimada para cada estudo e realizamos uma meta-análise. Resultados: Um total de 32 estudos foram incluídos e 94 pacientes brasileiros com DP com mutações causativas foram identificados em 2872 pacientes avaliados (3.2%). As mutações no PRKN causaram DP em 48 de 576 pacientes (8.3%). As mutações no LRRK2 foram identificadas em 40 de 1566 pacientes (2.5%), sendo a mutação mais comum a p.G2019S (2.2%). Conclusões: As mutações na PRKN são a causa mais comum de DP autossômica recessiva, e as mutações no LRRK2 a causa mais comum de DP autossômica dominante. Nós observamos uma falta de estudos epidemiológicos robustos em genética de DP, especialmente por não levar em conta a diversidade de nossa população.
ABSTRACT Background: Increasing numbers of mutations causing monogenic forms of Parkinson's disease (PD) have been described, mostly among patients in Europe and North America. Since genetic architecture varies between different populations, studying the specific genetic profile of Brazilian patients is essential for improving genetic counseling and for selecting patients for clinical trials. Objective: We conducted a systematic review to identify genetic studies on Brazilian patients and to set a background for future studies on monogenic forms of PD in Brazil. Methods: We searched MEDLINE, EMBASE and Web of Science from inception to December 2019 using terms for "Parkinson's disease", "genetics" and "Brazil". Two independent reviewers extracted the data. For the genes LRRK2 and PRKN, the estimated prevalence was calculated for each study, and a meta-analysis was performed. Results: A total of 32 studies were included, comprising 94 Brazilian patients with PD with a causative mutation, identified from among 2,872 screened patients (3.2%). PRKN mutations were causative of PD in 48 patients out of 576 (8.3%). LRRK2 mutations were identified in 40 out of 1,556 patients (2.5%), and p.G2019S was the most common mutation (2.2%). Conclusions: PRKN is the most common autosomal recessive cause of PD, and LRRK2 is the most common autosomal dominant form. We observed that there was a lack of robust epidemiological studies on PD genetics in Brazil and, especially, that the diversity of Brazil’s population had not been considered.

A catalyst comprised of SnO2 impregnated on ZnO nanowires, which presented remarkable ability to catalyze fatty acid esterification/transesterification reactions, is reported. For optimization of reaction conditions, artificially acidified soybean oil with 10 wt.% oleic acid was used as a model feed. The optimized conditions were: 150 °C, 6 h, 5 g of oil, catalyst concentration of 5%, and methanol:oil molar ratio of 15:1. The catalyst achieved 92% of total fatty acid methyl esters (FAME) content and was used five times without the necessity of catalyst washing from one reaction to the other. Then, such conditions were applied to produce biodiesel from the oil extracted from Scenedesmus sp. microalgae; the system reached 72% of FAME content, without any previous refining or degumming process of the oil. Rietveld refinement, X-ray diffraction, elemental mapping in scanning transmission electron microscopy, X-ray photoelectron spectroscopy, and pyridine-desorption Fourier-transform infrared spectroscopy were used to characterize the material.

The emergence of antibiotic resistance among microbial pathogens is a serious public health problem, resulting in a constant need of discovering new antibiotics. For this purpose, we synthetized and determined the antimicrobial activity of copper (Cu), cobalt (Co), and tin (Sn) complexes coordinated with scorpionate ligand in a tridentate mode against microorganisms of clinical importance. New complexes with the sodium hydrotris(2-mercaptothiazolyl)borate ligand were characterized by physicochemical and spectroscopic techniques (UV-vis, IR, 1H and 13C NMR). Moreover, their antimicrobial activity was evaluated using microdilution broth method. Cytotoxicity was determined in Vero cells and the selectivity index (SI) was calculated. The coordination with metals increased the ligand antimicrobial activity, which showed no activity when evaluated alone (MIC>1,080 µM). Cu-complex (1) had activity against Candida albicans and C. krusei, with MICs of 130 and 66 µM, respectively. The compounds had low toxicity on Vero cells and chemo-informatics assays corroborated a low systemic toxic effect. The SI revealed that the compound 1 was 16 times more selective for C. krusei and 8 times more selective for C. albicans in relation to Vero cells. The Cu-complex stands out as a potential prototype for the development of new antifungal agents.

Abstract: Four novel Cr (III), Co (II) and Ni (II) complexes with 2-thiazoline-2-tiol derivative ligands were synthesized and characterized using elemental, physiochemical, and spectroscopy analytical methods. The results showed the formation of ligands coordinated by sulfur atoms of the C = S bond to the L1 ligand and by the sulfur atom of the C-S bond and/or nitrogen of the intra C-ring bond of the L2 ligand. The activity of the complexes against Gram-negative, Gram-positive bacteria and yeasts of clinical importance was evaluated by broth microdilution method. The best result was obtained to L2-Ni compound against E. cloacae (Minimal Inhibitory Concentration (MIC) = 62.5 µg/mL). Additionally, the compounds showed anti-Candida activity (MIC values ranging from 250 to 500 µg/mL), except the L1-Cr that was not active against any tested microorganism.

ABSTRACT The aim of this study was to optimize the dextranase production by fungus Pochonia chlamydosporia (VC4) and evaluate its activity in dextran reduction in sugarcane juice. The effects, over the P. chlamydosporia dextranase production, of different components from the culture medium were analyzed by Plackett-Burman design and central composite design. The response surface was utilized to determine the levels that, among the variables that influence dextranase production, provide higher production of these enzymes. The enzymatic effect on the removal of dextran present in sugarcane juice was also evaluated. It was observed that only NaNO3 and pH showed significant effect (p<0.05) over dextranase production and was determined that the levels which provided higher enzyme production were, respectively, 5 g/L and 5.5. The dextranases produced by fungus P. chlamydosporia reduced by 75% the dextran content of the sugarcane juice once treated for 12 hours, when compared to the control treatment.

Objective: Associations between parental/caregiver depression and adverse child outcomes are well established and have been described through one or more mechanisms: child psychopathology following exposure to a depressed caregiver, child psychopathology exacerbating a caregiver’s depression, and caregiver and offspring depression sharing the same etiology. Data from low and middle-income countries is scarce. We examined correlations between common symptoms of mental disorders in caregivers and their offspring’s psychopathology in a Brazilian sample. Methods: In this cross-sectional study, adult caregivers were screened for depression during routine home visits by community health workers as part of the Brazilian Family Health Strategy. Caregivers with suspected depression were assessed using the Zung Self-Rating Depression Scale and the Self-Reporting Questionnaire (SRQ-20). Children’s symptoms were evaluated using the Strengths and Difficulties Questionnaire (SDQ). Results: The sample included 68 primary caregivers and 110 children aged 6 to 15 years. Higher caregiver scores on the SRQ-20 correlated significantly with psychiatric symptoms in offspring. Conclusion: These results substantiate our hypothesis that child psychopathology correlates with caregivers’ psychiatric symptoms. This paper adds to the growing literature on community mental health assessment and can help guide future strategies for reducing the burden of common mental disorders in caregivers and children alike in low and middle-income countries.

Resumo Este trabalho objetivou avaliar a contribuição da prolina, do metabolismo oxidativo e dos pigmentos fotossintéticos na propagação in vitro e aclimatização de Pitcairnia encholirioides, uma bromélia criticamente ameaçada de extinção. As análises foram realizadas após 150 dias de cultivo in vitro em meio MS suplementado com 10 µM de GA3 ou 0,2 µM de ANA, 15 ou 30 g L–1 de sacarose, em tubos de ensaio que permitiam trocas gasosas ou em sistema hermeticamente vedado, e também 180 dias após aclimatização. A manutenção in vitro em frascos hermeticamente fechados, com GA3 e 15 g L–1 de sacarose apresentou efeito metabólico adverso, demonstrado pelo menor acúmulo de prolina e pigmentos fotossintéticos e também pelo aumento das atividades de enzimas antioxidantes. Após aclimatização, as diferenças para prolina e pigmentos fotossintéticos não foram mais encontradas e as atividades enzimáticas variaram de maneira desuniforme. Os resultados sugerem que o cultivo in vitro em meio com 0,2 µM de ANA e 30 g L–1 de sacarose, em tubos fechados com tampas que permitem trocas gasosas, é mais adequado para a micropropagação de P. encholirioides, proporcionando uma manutenção prolongada das culturas in vitro e plântulas com qualidade superior para o desenvolvimento ex vitro.
Abstract This study aimed to evaluate the variation in the levels of proline, oxidative metabolism and photosynthetic pigments in plants of Pitcairnia encholirioides grown in vitro under different conditions and after acclimatization. The analyses were performed after 150 days of in vitro cultivation in MS media supplemented with 10 µM GA3 or 0.2 µM NAA, sucrose at 15 or 30 g L–1, in test tubes which allowed gas exchange or in a hermetically sealed system, and 180 days after acclimatization. The in vitro maintenance in hermetically sealed flasks, with GA3 and 15 g L–1 sucrose had adverse metabolic effects, which was demonstrated by the lower proline and photosynthetic pigments accumulation and by the increase in antioxidant enzymes activities. After acclimatization, differences for proline and photosynthetic pigments were no longer found and the enzymatic activities ranged unevenly. The results suggest that the in vitro cultivation in media with 0.2 µM NAA and 30 g L–1 sucrose, in test tubes capped with closures which allowed gas exchange, is more suitable for micropropagation of P. encholirioides, providing a prolonged maintenance of in vitro cultures and plantlets with superior quality for ex vitro development.

RESUMOO trabalho teve como objetivo verificar os efeitos dos estresses hídrico e salino na germinação de sementes de Petiveria alliacea , bem como definir os limites máximos de tolerância da espécie a esses estresses. As sementes foram submetidas aos agentes osmóticos NaCl, CaCl2 e PEG nos potenciais à 0; -0,1; -0,2; -0,3; -0,4; -0,5; -0,6; -0,7; -0,8; -0,9 e -1,0 MPa sob temperatura constante de 25˚C e fotoperíodo de 12 horas, com avaliações diárias durante 30 dias. As variáveis analisadas foram porcentagem de germinação, índice de velocidade de germinação, tempo médio de germinação, índice de sincronização e frequência relativa da germinação. As sementes de P. alliacea sob estresse osmótico apresentam menor porcentagem e velocidade de germinação com a redução dos potencias osmóticos, principalmente com CaCl2. Em potenciais osmóticos mais negativos que -0,4; -0,5 e -0,7 MPa, respectivamente nos agentes CaCl2, NaCl, e PEG, não ocorre germinação. O padrão de distribuição da frequência relativa aumentou a polimodalidade, o tempo médio de germinação e o índice de sincronização da germinação com a redução dos potencias osmóticos.
ABSTRACTThe study aimed to investigate the effects of water and salt stress on seed germination of Petiveria alliacea, as well as to define the limits of tolerance of the species to these stresses. The seeds were subjected to osmotic agents NaCl, CaCl2 and PEG in the potential 0; -0.1; -0.2; -0.3; -0.4; -0.5; -0.6; -0.7; -0.8; -0.9 and -1.0 MPa at a constant temperature of 25˚C and a photoperiod of 12 hours, with daily assessments for 30 days. The variables studied were germination percentage, germination velocity index, middle fear germination, synchronization index and relative frequency of germination. The seeds of P. alliacea under osmotic stress had lower percentage and speed of germination with the reduction of the the osmotic potential, especially with CaCl2. At more negative osmotic potentials than -0.4; -0.5 and -0.7 MPa, respectively in the CaCl2, NaCl, and PEG agents, the germination does not occur. The distribution pattern of the relative frequency increased the several modalities, , the average time of germination and the germination index of synchronization with the reduction of osmotic potential.

Resumo Os efluentes líquidos de algumas indústrias apresentam traços de metais nocivos que possuem propriedade bioacumulativa, o que contribui para o aumento de sua toxicidade. Tendo em vista o crescimento industrial, e o consequente aumento de emissões de contaminantes em efluentes líquidos, a adsorção está entre as operações aplicadas para diminuir as concentrações dos metais a fim de atender a regulamentação ambiental vigente. A biomassa residual vem ganhando destaque no tratamento desses efluentes por se um material renovável e de baixo custo, quando comparado com os adsorventes convencionais. O objetivo deste trabalho é avaliar a eficiência da cinza do bagaço da cana-de-açúcar (CBC) na adsorção dos íons metálicos Cu2+ e Cr3+ por meio de ensaios de isoterma de adsorção. A CBC foi caracterizada quanto a sua microestrutura, composição química, área de superfície específica e morfologia, e o adsorbato foi analisado por absorção atômica de chama. A CBC apresentou-se de maneira eficaz como um bioadsorvente para ambas as soluções sintéticas, contendo os íons Cu2+ e Cr3+, presentes nos efluentes líquidos. As isotermas de adsorção apresentaram um perfil crescente, indicando a eficiência na remoção média de 97,3% para o Cr3+ e 96,4% para o Cu2+, podendo este material ser aplicado como adsorvente em efluentes industriais.
Abstract The liquid effluents of some industries have traces of harmful metals having bioaccumulative property, which contributes to the increase in their toxicity. Given the industrial growth, and the consequent increase in contaminant emissions in wastewater, the adsorption is among the operations applied to decrease the concentrations of metals in order to meet current environmental regulations. The residual biomass has been gaining attention in the treatment of these effluents by a renewable material and cost as compared with conventional adsorbents. The present work aims to evaluate the ashes of the efficiency of sugarcane bagasse (CBC) on the adsorption of metal ions Cu2+ and Cr3+, through isotherm adsorption tests. The CBC was characterized regarding its microstructure, chemical composition, specific surface area and morphology, and the adsorbate was analyzed by flame atomic absorption. The CBC has performed effectively as a bioadsorbent for both synthetic solutions containing Cu2+ and Cr3+ ions present in liquid effluents. The isotherms adsorption showed a growing profile, indicating the average removal efficiency of 97.3% to 96.4% Cr3+, and Cu2+ to, this material can be applied as an adsorbent in industrial effluents.

This study evaluated the metalloproteomic profile of muscle tissue samples from Nile tilapia (Oreochromis niloticus). Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) methods was used for protein fractionation and identification based on image analysis. Determination of Ca, Cu, Fe, Mn and Zn was performed in the protein spots by flame and/or graphite furnace atomic absorption spectrometry (FAAS, GFAAS), the protein spots were characterized by electrospray ionization-tandem mass spectrometry (ESI-MS/MS). FAAS and GFAAS determinations have indicated the presence of calcium in seven protein spots, iron in only one spot, manganese in six spots, and zinc in two spots. Of the seventeen protein spots where the presence of metal ions was detected, ten were characterized by ESI-MS/MS.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A new approach to evaluate immobilization of β-galactosidase on Eupergit® C: structural, kinetic, and thermal characterization

This study aimed to evaluate β-galactosidase immobilization. For this purpose, the ionic strength of the buffer, reaction time, amount of the immobilization support, and pH were evaluated by a central composite design. Assay 8, which consisted of 1.5 mol L-1 phosphate buffer (pH 7.5) and a reaction time of 2 h, produced the maximum yield. Eupergit® C (400 mg) was subsequently used as an immobilization support. Immobilization kinetics wereinvestigated, and a significant increase in the yield was obtained after immobilization compared with that obtained from assay 8 (22.0 U mL-1 vs. 15.6 U mL-1). The enzyme efficiency of actuation was evaluated using o-nitrophenyl-β-D-galactopyranoside and lactose, with lactose providing better results. The reuse of β-galactosidase was evaluated, and more than 50% of the initial enzyme activity was maintained after five cycles of use. Enzyme characterization revealed that immobilization improved some aspects of the thermostability of β-galactosidase.

Ovarian intratumoral 21-hydroxylase deficiency in a postmenopausal hirsute woman

Tumores ovarianos virilizantes são uma causa rara de hiperandrogenismo em mulheres, contabilizando menos de 5% de todos as neoplasias ovarianas. Esses tumores ocorrem mais comumente em mulheres em peri ou pós-menopausa. Relatamos aqui o caso de uma mulher de 64 anos de idade com sintomas de virilização que começaram 3 anos antes. O perfil hormonal revelou níveis aumentados de testosterona e de 17-hidroxiprogesterona (17-OHP). O teste de tetracosactin demonstrou deficiência de 21-hidroxilase. Exames radiológicos mostraram um nódulo no ovário esquerdo. A paciente foi submetida à ooforectomia laparoscópica bilateral e o exame histopatológico revelou um luteoma no ovário esquerdo. A concentração sérica de testosterona e de 17-hidroxiprogesterona após a cirurgia retornou aos níveis normais em um mês. A virilização regrediu em 6 meses. Nossa paciente também revelou uma elevação dos níveis séricos de 17-OHP. A normalização da 17-OHP após a ooforectomia sugere um caso de deficiência de 21-hidroxilase intratumoral. Esta é a primeira descrição de deficiência de 21-hidroxilase intratumoral em uma mulher na pós-menopausa. Arq Bras Endocrinol Metab. 2012;56(9):672-6
Virilising ovarian tumours are a rare cause of hyperandrogenism in women, accounting for less than 5% of all ovarian neoplasms. It occurs most often in - and postmenopausal women. We report a case of a 64 year-old woman with signs of virilisation that had started 3 years before. Blood hormone analysis revealed increased levels of testosterone, and 17-hydroxyprogesterone. The tetracosactin test revealed 21-hydroxylase deficiency. Radiological imaging demonstrated a nodule in her left ovary. The patient was submitted to bilateral laparoscopic oophorectomy, and histopathological examination revealed a luteoma of the left ovary. Postoperative serum testosterone level and 17-hydroxyprogesterone returned to normal levels in one month. Virilism regressed within six months. Our patient also showed an elevation in 17-OHP serum levels. Normalization of 17-OHP after oophorectomy suggests a case of intratumoral 21-hydroxylase deficiency. To our knowledge, this is the first description of ovarian intratumoral 21-hydroxylase deficiency in a postmenopausal woman. Arq Bras Endocrinol Metab. 2012;56(9):672-6
