ABSTRACT This study aimed to determine the antibiotic profile of microorganisms isolated from urine samples of patients with community urine tract infections (UTI) admitted to the University Hospital of the Federal University of Sao Carlos to support an appropriate local empirical treatment. A retrospective cross-sectional study was conducted from October 2018 to October 2020. Data from 1,528 positive urine cultures for bacterial pathogens and antibiograms were tabulated. Bacterial species prevalence and their resistance profile were analyzed and compared by sex and age. For Gram-negative fermenting bacteria, resistance rates were compared between patients with previous hospitalization and the total of infections caused by this group. For comparisons, the Chi-square test was performed, using Fisher’s exact test when necessary (BioEstat program, adopting p ≤ 0.05). A multivariate analysis was applied to assess the effect of the studied variables in predicting multidrug resistance. Infections were more prevalent in women and older adults. Gram-negative bacteria represented 90.44% of total cultures. In both sexes, E. coli prevalence was significantly higher in adults compared with older adults (p < 0.0001). For several antibiotics, resistance rates were higher in the older adults compared with other ages and in patients with Gram-negative fermenting infections and previous hospitalization compared with the total of infections by this group of bacteria. The closer to the hospitalization, the higher the number of antibiotics with superior resistance rates. Resistance rates for aminoglycosides, carbapenems, ceftazidime, nitrofurantoin, piperacillin+tazobactam, and fosfomycin were less than 20%, considered adequate for empirical treatment. Only hospitalization in the previous 90 days was statistically significant in predicting infections by multidrug-resistant bacteria.

ABSTRACT Toxoplasmosis is an infection of vast worldwide distribution whose etiologic agent is Toxoplasma gondii. This disease can cause problems ranging from mild symptoms to serious conditions, such as encephalitis, miscarriage and blindness. Therefore, it is of utmost importance to perform a diagnosis with reproducible techniques in order to obtain a good prognosis. The aim of this review was to analyze the efficiency of toxoplasmosis diagnostic techniques based on sensitivity and specificity results. Five research platforms in English language were used (Eric, Elsevier, Google Scholar, PubMed and SciELO), which contained data on the diagnosis of toxoplasmosis. The search and selection were performed for studies published prior to June 2021. The search resulted in the inclusion of 13 articles published from 2005 to 2020. The data revealed the use of different samples in the standardization of techniques such as serum, total blood, colostrum and amniotic fluid. The flow cytometry, lateral flow immunoassay and qPCR techniques showed 100% sensitivity, whereas the ELISA, western blotting, qPCR and RE-LAMP techniques achieved 100% specificity. Significantly, the qPCR and LAMP techniques were more accurate when the likelihood ratio was assessed. The meta-analysis identified that ISAGA and western blotting have low sensitivity values and LIASON, ELFA and ELISA, using a silica bioconjugate, also have low specificity values. It was noted that a wide range of methods have high values of sensitivity and specificity. Therefore, the choice of the method will be based on the conditions and its financial viability.

Abstract In South America, Solenopsis saevissima and S. invicta are the most common fire ants. Nests are founded in areas under anthropic interference like urban or rural areas, but S. invicta is found preferentially in those with the greatest anthropic interference. However, we do not know the rates at which they exist in anthropized areas next to high density of native vegetation. Areas with 60 to 90% of native Atlantic Forest were selected to verify the occurrence of both species in rural and urban areas. We investigated the molecular diversity and applied the reconstruction of the ancestral state analysis for each species. A total of 186 nests were analyzed and we found that the two species had the same proportion in the urban area. However, S. saevissima had a higher rate of prevalence in the rural area, in addition to having a greater number of haplotypes and ancestry associated with this type of habitat for the region. S. invicta had the same number of haplotypes in both rural and urban regions, and less haplotypic diversity. We conclude that S. saevissima is a species typically associated with rural areas and S. invicta, although present, is not dominant in urban areas.

The high prevalence of diabetes and obesity encourages research for the development of α-glucosidase inhibitors from natural sources. This study evaluated the activity of fractions obtained from Protium spruceanum branches. Anti-α-glucosidase activity was investigated in vitro using 4-nitrophenyl-α-D-glucopyranoside as the substrate, while free-radical scavenging activity was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-azinobis-3-ethylbenzotiazoline-6-sulfonic acid (ABTS) assays. Furthermore, a model of oxidative stress promoted by H2O2 in fibroblasts was employed, and cell viability was determined by sulforhodamine B. Fractions inhibited α-glucosidase activity effectively, highlighting the hydromethanolic fraction (HMF). Quercitrin, isolated from the HMF, was identified by spectroscopy and quantified by a validated high performance liquid chromatography with diode array detector (HPLC-DAD) method and exhibited free radical scavenging activity comparable to the HMF. However, this flavonoid showed low anti-α-glucosidase effect, suggesting a synergism effect among several components of the HMF. This proposal is also supported by the docking results obtained by PyRx software. The compounds present in the HMF showed a noncompetitive inhibition at the in silico simulation. Finally, the HMF also protected fibroblasts against cell death induced by oxidative stress. This is the first evidence of the capacity of P. spruceanum branches to inhibit α-glucosidase activity and to counteract oxidative stress. These results encourage the use of this Brazilian plant against hyperglycemia-correlated diseases.

Abstract: Introduction: Cystic nephroma is a rare benign renal tumor of uncertain etiology. In children, it can manifest as a palpable abdominal mass, hematuria, and recurrent urinary infections. Imaging tests such as ultra sound and computed tomography assist in the diagnosis, but confirmation is made through anatomopathological study. Treatment is surgical and may be partial or total nephrectomy, with a good prognosis. Objective: To report a rare case of pediatric cystic nephroma, its clinical manifestations, radiological and histopathological aspects, as well as the treatment used and its evolution. Clinical Case: Pre-school, male, with a history of recurrent urinary infections in the first year of life. At 2 years and 8 months, he presented nodulation in the right hypochondrium with local pain on palpation, associated with urinary disorders and hematuria. An ultrasonography showing enlarged right kidney due to multiseptated cystic formation. Computed tomography showing multiloculated cystic expan sive formation in the right kidney. At 2 years and 10 months, he underwent partial right nephrec tomy for excision and anatomopathological study, which was compatible with Cystic Nephroma. He evolved with regression of hematuria and recurrent episodes of urinary infections, maintaining renal function preserved. Currently, at 4 years and 6 months, asymptomatic. Conclusions: Cystic nephro ma is a rare entity, generally with a good prognosis. The association of clinical findings, radiological images, and anatomopathological study are fundamental for the establishment of diagnosis and a better definition of therapeutic conduct.
Resumen: Introducción: El nefroma quístico es un tumor renal benigno raro, de etiología incierta. En niños puede manifes tarse como una masa abdominal palpable, hematuria e infecciones urinarias recurrentes. Exámenes de imagen como ultrasonografía y tomografía computadorizada ayudan en el diagnóstico, sin embar go, la confirmación se realiza mediante el estudio anatomopatológico. El tratamiento es quirúrgico, pudiendo ser nefrectomía parcial o total, con buen diagnóstico. Objetivo: Describir un caso raro de nefroma quístico pediátrico, sus manifestaciones clínicas, aspectos radiológicos e histopatológicos, así como tratamiento y su evolución. Caso Clínico: Preescolar, sexo masculino, con historia clínica de infecciones urinarias de repetición en el primer año de vida. A los 2 años y 8 meses, presentó nodulación en hipocondrio derecho con dolor local a la palpación, asociado a molestias disúricas y hematuria. La ecografía mostró un riñón derecho aumentado de volumen por formación quística multiseptada. La tomografía computadorizada demostró formación expansiva quística multiocula- da en el riñón derecho. A los 2 años y 10 meses, realizó nefrectomía parcial derecha para exéresis y estudio anatomopatológico, compatible con nefroma quístico. Evolucionó con regresión de la hema turia y de los episodios recurrentes de infecciones urinarias, manteniendo función renal preservada. Actualmente, con 4 años y 6 meses, asintomático. Conclusiones: El nefroma quístico es una entidad rara, en general de buen pronóstico. La asociación de hallazgos clínicos, imágenes radiológicas y es tudio anatomopatológico son fundamentales para el establecimiento diagnóstico y mejor definición de la conducta terapéutica.

A quantitative and confirmatory method for detecting the presence of triphenylmethane dyes in shrimp muscle using ultra-high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) and a quick, easy, cheap, effective, rugged and safe (QuEChERS) extraction approach was validated. The method exhibited linearity and selectivity and the coefficient of determination (R2) was higher than 0.95 for all studied analytes. Limits of detection (LODs) varied from 0.32 to 0.44 µg kg-1 and the limit of quantification (LOQ) was determined to be 0.5 µg kg-1 for all studied analytes. The trueness, precision, decision limits (CCα), detection capability (CCβ) and uncertainty presented adequate performance. In addition to the validation in shrimp muscle, fish and salmon muscle were also satisfactory validated as an extension of scope. The suitability of the proposed method was also evaluated through an interlaboratory proficiency test, in which satisfactory results were obtained. The fully validated method is thus suitable for the analysis of triphenylmethane dyes in shrimp, fish, and salmon muscle.

Trends in suicide rates in Brazil from 1997 to 2015

Objectives: To analyze time trends of suicide rates in Brazil overall and in Brazilian states and compare the estimated suicide rates projected for 2020 with the World Health Organization (WHO) Mental Health Action Plan target. Methods: This was an ecological time-series study from 1997 to 2015, stratified by Brazilian states, specific age groups, and sex. Data were obtained from the Mortality Information System (Sistema de Informações sobre Mortalidade [SIM]) of the Brazilian Ministry of Health. Polynomial regression models were used to analyze the trends in suicide rates and to project suicide rates for 2020. Results: Considering 224 units of analysis, 21 (9.4%) showed a decreasing trend, 108 (48.2%) were stable, and 95 (42.4%) showed an increasing trend. Thus, 67% of units of analysis will not meet the WHO target in 2020. Mean suicide rates were higher in males than in females. People aged 60 years and older presented the highest suicide rates, while 84.7% of total deaths by suicide occurred among 15-to-59-year-olds. Conclusion: Overall, 90.6% of units of analysis had a stable or increasing trend in suicide rates from 1997 to 2015. If these trends remain, most of Brazil will fail to achieve the WHO-recommended reduction in suicide rates by 2020.

Abstract: We investigated the effects of acetylsalicylic acid (ASA) on the total myenteric neuronal population in the descending colon in Trypanosoma cruzi-infected mice. Thirty-five male Swiss mice, 60 days old, were divided into a control group (C group), control group treated with ASA (CA group), infected group (I group), and infected group treated with ASA (IA group). A total of 1300 trypomastigotes of the Y strain of T. cruzi were intraperitoneally inoculated in the IA and I groups. The CA and IA groups were treated with ASA intraperitoneally. At 75 days post-infection (dpi), all of the animals were sacrificed. Neurons in the colon were stained with Giemsa, quantified, and measured. No difference in the course of infection was observed between the IA and I groups, reflected by the parasitemia curve. Acetylsalicylic acid treatment in the CA and IA groups did not alter the total number of myenteric neurons compared with the C and I groups. The CA and IA groups exhibited an increase in the nuclear area, cytoplasmic area, and neuronal body area compared with the C and I groups. Future studies should elucidate the mechanism of action of ASA against Chagas’ disease in the chronic phase.

Abstract Cellulases are efficient enzymes for the conversion of cellulose into glucose. Their use in immobilized form enables them to be reused in successive cycles in many biotechnological processes. Unlike conventional methods of immobilization by covalent bonding, in miniemulsion polymerization the immobilization of enzyme and the synthesis of polymer nanoparticles (support) occur simultaneously. Based on these aspects, the immobilization of cellulose on poly(methyl methacrylate) (PMMA) nanoparticles by miniemulsion polymerization was studied. The surfactant type (non-ionic and ionic) and latex pH showed great influence on cellulase activity. High activity values were obtained only when non-ionic surfactant (Lutensol AT50) and buffering agent (NaHCO3) were used simultaneously. MMA polymerization rate and final monomer conversion were not affected by the presence of cellulase. The maximum immobilization efficiency (60%) was obtained when 6 wt.% of cellulase was used and stable PMMA nanoparticles (133 nm) were obtained. The relative activity profile of immobilized cellulase, for pH as well as temperature, was similar to that reported for the free form. Immobilized enzyme keeps its activity throughout seven days when stored at 4 ºC and phosphate buffer pH 6.0. Based on the results obtained in this work, miniemulsion polymerization as a method for cellulase immobilization on PMMA nanoparticles showed to be a promising technique with high possibility of industrial application.

RESUMO: A distribuição espacial e a abundância de carrapatos em estágio de vida livre foram examinadas em fragmentos de Mata Atlântica no Nordeste do Brasil. O estudo foi realizado em Unidades de Conservação de Proteção Integral Mata do Tapacurá e Mata do Camucim, localizadas no município de São Lourenço da Mata, Pernambuco. Os carrapatos foram capturados pelo método da bandeira de flanela em seis parcelas de 200m2, sendo três parcelas estabelecidas em uma borda de mata adjacente a uma matriz agropastoril e outras três parcelas estabelecidas na borda de mata adjacente ao lago de barragem. Foram capturados 2652 carrapatos do gênero Amblyomma, sendo duas ninfas identificadas como Amblyomma dissimile com distribuição espacialmente agregada, demonstrados pela relação variância/média e o índice de agregação de Lloyd. Houve variação significativa na abundância de carrapatos entre as parcelas (H=26,79, p<0,005) sendo encontrados mais carrapatos na borda da mata adjacente à matriz agropastoril. Quanto as variáveis ambientais, houve correlação positiva entre a cobertura de dossel e a densidade de carrapatos (rs=0,31, p<0,05). No presente estudo observou-se que em estágios de vida livre, notadamente larvas e ninfas, a distribuição espacial e a abundância de carrapatos são fortemente influenciadas por áreas mais fechadas das matas. A maior presença de carrapatos próximos à matriz agropastoril poderia facilitar a migração de carrapatos entre ambientes silvestres e ambientes antrópicos.
ABSTRACT: The spatial distribution and abundance of free-living ticks were examined in Atlantic Forest fragments in northeastern Brazil. The study was conducted on Integral Protection Conservation Units of Mata do Tapacurá and Mata do Camucim, located in the municipality of São Lourenço da Mata, Pernambuco. Ticks were captured by flagging method in six plots of 200m2, three plots set in a forest edge adjacent to an agropastoral matrix and three plots set in the forest edge adjacent to a dam lake. Were captured 2652 ticks of the genus Amblyomma, two nymphs identified as Amblyomma dissimile with spatially aggregated distribution, shown by the variance/mean ratio and Lloyd aggregate index. There was significant variation in the abundance of ticks between plots (H=26.79, p<0.005) and more ticks was found in the woods adjacent to the edge agropastoral matrix. There was a positive correlation between canopy cover and tick density (rs=0.31, p<0.05). In the present study, it was observed that in free-living stages, especially larvae and nymphs, the spatial distribution and abundance of ticks are strongly influenced by more enclosed areas of forests. Most ticks near agropastoral matrix could facilitate the migration of ticks between wild environments and man-made or modified environments.

O testículo encontrava-se localizado no tecido subcutâneo da região inguinal, sem estar aderido aos tecidos circunvizinhos. Após orquiectomia bilateral, ambos os testículos foram mensurados, fixados em formol a 10% e embebidos em parafina para avaliação histopatológica. O testículo esquerdo mediu 1,2cm de largura por 1,7cm de movimento; e o testículo direito mediu 1,5cm de largura por 2,0cm de comprimento. O testículo ectópico apresentou epitélio sem linhagem de células germinativas pós-meióticas. O testículo não ectópico apresentou alterações no epitélio seminífero caracterizando degeneração. Em ambos os epidídimos, o lúmen não continha espermatozoides e as principais alterações estruturais do epitélio foram mais distintas no epidídimo associado ao testículo ectópico. Conclui-se que o testículo ectópico e epidídimo apresentaram lesões características de aumento de temperatura. O testículo e epidídimo não ectópico apesentaram lesões menores mas que puderam ser associadas à infertilidade do quati.
ABSTRACT: This paper reports a case of unilateral extracorporeal ectopic testes in a captive coati (Nasua nasua) in the State Park of Dois Irmãos Zoo, Recife/PE, Brazil. The testicle was located in the subcutaneous tissue of the inguinal region not adhered to the surrounding tissues. After bilateral orchiectomy, both testes were measured, fixed with 10% formalin buffered and embedded in paraffin for histopathological evaluation. The left testis measured 1.2 cm width by 1.7cm length, and the right one measured 1.5 cm width by 2.0 cm length. The ectopic testes had seminiferous epithelium without post-meiotic germ cell lines. The non-ectopic testis had several changes in the seminiferous epithelium that indicated degeneration. In both epididymis, the lumen did not contain sperm and the major epithelial structural alterations were more distinct in the epididymis associated to the ectopic testicle. In conclusion, the ectopic testis and epididymis had lesions compatible with testicular exposition to body temperature. Non-ectopic epididymis and testis had minor lesions but could be related to the infertility of the coati.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

O escopo da presente pesquisa foi desenvolver e validar uma metodologia para extração simultânea de colesterol e óxidos de colesterol utilizando saponificação direta em amostras de leite fluido para posterior análise por cromatografia líquida de alta eficiência em fase reversa com detecção por arranjo de diodos (RP-HPLC-DAD). Foram avaliados os parâmetros analíticos de seletividade, linearidade e faixa de aplicação, precisão inter e intradia, recuperação, limites de detecção e quantificação do método. Os resultados demonstraram que a metodologia pode ser empregada com segurança e confiabilidade para análise do colesterol e óxidos do leite.
The aim of the present work was to develop and validate a reversed-phase highperformance liquid chromatography with diode array detection (RP-HPLC-DAD) method for determination of cholesterol and cholesterol oxides extracted from milk samples. The method consisted of direct samples saponification and analytes extraction, followed by their determination using RP-HPLC-DAD. Suitable analytical selectivity, linearity and range of application, intra-assay and intermediary precision, recovery, detection and quantification limits (LOD and LOQ) were achieved and, the results showed that the developed RP-HPLC-DAD method can be applied to routine determination of cholesterol and cholesterol oxides in milk samples.
