Abstract Pollen grain morphology and the characterization of additional structures related to pollen dispersion can help to understand the strategies presented by species as well as their taxonomic circumscription. This work investigates the morphology of pollen grains and orbicules of Cedrela fissilis Vell. and Cedrela odorata L., two threatened species of the genus Cedrela P. Browne (Meliaceae A. Juss.) in Brazil. Observations of pollen grains and orbicules of the species were carried out pre- and post-chemical treatment of the samples, under light and scanning electron microscopy, aiming at recognizing structures, detailing morphological characterizations and taking measurements, the last of which were submitted to statistical treatment. The results demonstrate that the pollen grains of the two species are medium in size (measuring between 25 and 50µm), radially symmetrical, isopolar, prolate-spheroidal and 4,5-colporate with subcircular to subquadrangular amb and psilate ornamentation; the orbicules also have a smooth surface and measure about 7 to 10µm. Variation was observed in the size of pollen grains, which presented high coefficient of variation and in the orbicules, as well as in the number of apertures.

Biochar from slow pyrolysis under controlled conditions is favorable in the transformation of waste into materials that serve as soil conditioners, and/or environmental remediation. Thus, this work aimed to produce and characterize biochar from cassava waste under different temperature conditions and evaluate interactions with Ca2+ and Pb2+ for agricultural and environmental applications. To investigate the efficiency of the interaction of biochar with nutrients (Ca) and contaminants (Pb), the influence of pH and contact time were evaluated. There was an increase in pH, pHzpc (zero point charge), and condensed aromatic structures in BC, due to the increase in pyrolysis temperature. Also, biochar demonstrated a Pb2+ adsorbed amount of 2.03 to 2.37 mg g-1 and a Ca2+ release capacity of 0.655 to 0.765 mg g-1. In this way, biochar produced from residues of cassava waste can be a viable strategy to enhance agricultural production and reduce environmental contaminants.

This study evaluated the chemical composition, antioxidant and anticholinesterase activities, and the toxicity of hydroethanolic extracts of the Parkia platycephala Benth. Hot extraction with 70% hydroethanolic solution was used to obtain crude extracts of the leaf (LE), bark (BE), flower (FE), and seed (SE). Separation and identification of the compounds were performed by liquid chromatography with diode array detection and gas chromatography coupled with mass spectrometry. Antioxidant and anticholinesterase activities were performed by the 2,2-diphenyl-1-picrylhydrazyl (DPPH•) and Ellman methods, respectively, and the preliminary toxicity by the method with Artemia salina. Chromatography analysis indicated naringin, kaempferol, stigmasterol, and β-sitosterol in all extracts. Gallic acid is prominent in the LE and FE, while ferulic acid is abundant in the BE and lupeol and lupeol acetate are prominent in the SE. The BE antioxidant activity (lethal concentration of 50% (IC50) = 14.72 ± 0.13 µg mL-1) is equivalent to the standard rutin (IC50 = 15.85 ± 0.08 µg mL-1). The SE had the best acetylcholinesterase inhibition potential (IC50 = 5.73 ± 0.68 µg mL-1). Aside from the SE, which is not toxic, the other extracts had low (LE, FE) and moderate (BE) toxicity. The extracts of P. platycephala have potential antioxidant and therapeutic uses, especially against Alzheimer’s disease.

Resumo Fundamento: A vitamina D (VD) tem um importante papel na função cardíaca. No entanto, a vitamina exerce uma curva “dose-resposta” bifásica na fisiopatologia cardiovascular e pode causar efeitos deletérios, mesmo em doses não tóxicas. A VD exerce suas funções celulares ligando-se ao seu receptor. Ainda, a expressão da proteína de interação com a tiorredoxina (TXNIP) é positivamente regulada pela VD. A TXNIP modula diferentes visa de sinalização celular que podem ser importantes para a remodelação cardíaca. Objetivos: Avaliar se a suplementação com VD leva à remodelação cardíaca, e se a TXNIP e a tiorredoxina (Trx) estão associadas com esse processo. Métodos: Duzentos e cinquenta ratos Wistar machos foram alocados em três grupos: controle (C, n=21), sem suplementação com VD; VD3 (n = 22) e VD10 (n=21), suplementados com 3,000 e 10,000 UI de VD/ kg de ração, respectivamente, por dois meses. Os grupos foram comparados por análise de variância (ANOVA) com um fator e teste post hoc de Holm-Sidak (variáveis com distribuição normal), ou pelo teste de Kruskal-Wallis e análise post-hoc de Dunn. O nível de significância para todos os testes foi de 5%. Resultados: A expressão de TXNIP foi mais alta e a atividade do Trx foi mais baixa no grupo VD10. Os animais que receberam suplementação com VD apresentaram aumento de hidroperóxido lipídico e diminuição de superóxido dismutase e glutationa peroxidase. A proteína Bcl-2 foi mais baixa no grupo VD10. Observou-se uma diminuição na β-oxidação de ácidos graxos, no ciclo do ácido tricarboxílico, na cadeia transportadora de elétrons, e um aumento na via glicolítica. Conclusão: A suplementação com VD levou à remodelação cardíaca e esse processo pode ser modulado por TXNIP e Trx, e consequentemente por estresse oxidativo.
Abstract Background: Vitamin D (VD) has been shown to play an important role in cardiac function. However, this vitamin exerts a biphasic “dose response” curve in cardiovascular pathophysiology and may cause deleterious effects, even in non-toxic doses. VD exerts its cellular functions by binding to VD receptor. Additionally, it was identified that the thioredoxin-interacting protein (TXNIP) expression is positively regulated by VD. TXNIP modulate different cell signaling pathways that may be important for cardiac remodeling. Objective: To evaluate whether VD supplementation lead to cardiac remodeling and if TXNIP and thioredoxin (Trx) proteins are associated with the process. Methods: A total of 250 Male Wistar rats were allocated into three groups: control (C, n=21), with no VD supplementation; VD3 (n = 22) and VD10 (n=21), supplemented with 3,000 and 10,000 IU of VD/ kg of chow respectively, for two months. The groups were compared by one-way analysis of variance (ANOVA) and Holm-Sidak post hoc analysis, (variables with normal distribution), or by Kruskal-Wallis test and Dunn's test post hoc analysis. The significance level for all tests was 5%. Results: TXNIP protein expression was higher and Trx activity was lower in VD10. The animals supplemented with VD showed increased lipid hydroperoxide and decreased superoxide dismutase and glutathione peroxidase. The protein Bcl-2 was lower in VD10. There was a decrease in fatty acid β-oxidation, tricarboxylic acid cycle and electron transport chain with shift to increase in glycolytic pathway. Conclusion: VD supplementation led to cardiac remodeling and this process may be modulated by TXNIP and Trx proteins and consequently oxidative stress.

Alkaline extractions from humic material are used to risk assessment in soils. However, there are questions about the type of extraction most suitable for this evaluation. Thus, this work evaluated different soil extractions in search of realistic environmental conditions (simulated rain). The parameters obtained revealed significant differences between the levels and structural characteristics of organic matter, depending on the extractant used, reinforcing the importance of developing methodologies and analytical procedures that minimize structural changes and may be more representative of the molecular structures of organic matter found in soils. The results showed that the material extracted by simulated rain is similar to the fulvic acids of the soil organic matter extracted by alkaline extraction showing the importance of this type of extraction for risk assessment studies in soils.

The suspended particulate material (SPM) present in waters has fundamental importance for a better understanding of environmental behaviors of aquatic pollutants. This work determined the physicochemical parameters (pH, temperature, conductivity, total organic carbon and metals concentration) in water samples from Mundaú-Manguaba estuarine-lagoon system, Alagoas, Brazil. This work extracted and characterized the organic matter present in SPM in this estuary and studied the interaction of this organic matter with potentially toxic metals (Cd2+ and Pb2+). The results of the physical-chemical parameters evaluated showed influence of the tide and also of anthropic contributions close to the estuary. In fact, the lead concentrations determined (0.10-1.32 mg L-1) are well above that allowed by Brazilian legislation (< 0.010 mg L-1). The organic matter present in the SPM showed a high degree of humification being similar to the organic matter extracted from water and showed strong interaction by Pb2+ ions.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

Resumo Objetivo Investigou-se a dieta de Plagioscion squamosissimus presente no reservatório de Santa Cruz, Rio Grande do Norte, Brasil, avaliando as influências das variações espacial e temporal e de fatores abióticos na utilização dos recursos alimentares. Métodos As coletas foram realizadas trimestralmente, entre fevereiro de 2011 e novembro de 2014. Dos 525 exemplares capturados, 375 apresentaram itens alimentares nos estômagos. A dieta foi determinada utilizando: (i) Índice Alimentar (IAi); (ii) e a interpretação gráfica da estratégia alimentar, através de análise de Escalonamento Multidimensional Não Métrico (NMDS), sendo as diferenças na dieta testadas a partir de Análises de Variância Multivariada Permutacional (PERMANOVA). Para verificar se as variáveis ambientais influenciam na dieta foram realizadas Análise de Correspondência Canônica (CCA), usando os dados do IAi e os dados ambientais, sendo a significância dos eixos testada através de simulações de Monte Carlo. Resultados A espécie se alimentou principalmente de itens de origem animal, sendo vegetal um recurso ingerido de forma acidental. A dieta foi composta por camarão, peixe, inseto, molusco e vegetal, este último raramente encontrado. O camarão foi o item principal (92,57%) e os insetos (7,24%) itens acessórios. A ordenação espacial e temporal demonstrou que a espécie não apresentou variação na composição da dieta, assim como apresentou uma baixa associação entre a alimentação e as variáveis ambientais. Conclusão P. squamosissimus caracteriza-se como carcinófaga, em função do predomínio de camarão em sua dieta, evidenciando que a espécie está adaptada às condições oferecidas pelo ambiente do semiárido brasileiro.

Methods The samplings were performed quarterly between February 2011 and November 2014. Of the 525 specimens captured, 375 presented food items in the stomachs. The diet was determined using: (i) Feeding Index (IAi); (ii) and the graphical interpretation of the food strategy through non-metric multidimensional scaling (NMDS) analyzes, the differences in diet being tested from Permutational Multivariate Analyzes of Variance (PERMANOVA). To verify if environmental variables influence the Canonical Correspondence Analyzes (CCA) were used, using the data from the IAi the environmental data, being the significance of the axes tested through Monte Carlo simulations. Results The species feed mainly on items of animal origin, vegetable being a resource ingested accidentally. The diet was composed of shrimp, fish, insect, mollusk and vegetable, the latter rarely found. The shrimp was the main item (92.57%) and the insects (7.24%) accessory items. Spatial and temporal ordering demonstrated that the species didn’t present variation in diet composition, as well as a low association between diet and environmental variables. Conclusion P. squamosissimus is characterized as carcinophagous, due to the predominance of shrimp in its diet, evidencing that the species is adapted to the conditions offered by the Brazilian semi-arid environment.

ABSTRACT Although endophytic bacteria impact the health, and ultimately the fitness, of their hosts, our understanding of the diversity of endophytic species remains limited. Here we report on the endophytic microbiota inhabiting the roots, healthy leaves and leaves attacked by a gall-inducing insect of Baccharis dracunculifolia, a species of major economic relevance in South America, using 16S rRNA gene new generation sequencing. Rhodoplanes and Nitrospira were well represented in the communities of roots and leaves; known to be important for nitrogen cycling. The difference in bacterial diversity between healthy and galled leaves was not pronounced. The leaves seem to harbor specialized bacteria with high tolerances to abiotic stresses such as wide variation in temperature, low humidity, shallow and nutrient-poor soils and high solar irradiation. These findings suggest taxon-specific ecological niches in the leaves and roots, which may be the result of different physicochemical characteristics between these structures. This study provides a basis for further investigations and adds significant new information to the current knowledge of the endophytic bacterial composition in B. dracunculifolia.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Determinação de Mn e Zn em arroz empregando espectrometria de fluorescência de raios x de energia dispersiva

A simple, fast and inexpensive method was developed to determine essential elements in pellets of rice samples using energy dispersive X-ray fluorescence spectrometry (EDXRF). The accuracy and precision were evaluated using Standard Reference Material (rice flour NIST 1568a), and yielding relative standard deviation below 5%. The paired t-test showed good agreement within 95% confidence values. The detection limits (3σ) of Mn and Zn were 5.1 and 2.2 mg kg-1, respectively. The proposed method proved to be effective when used to determine Mn and Zn in commercial samples of rice without go by stage of decomposition.

O gênero Zephyranthes Herb. é um grupo taxonômicamente complexo e cariologicamente variável, compreende cerca de 65 espécies de distribuição predominantemente neotropical. O presente trabalho aborda a variabilidade cariológica em um total de 32 indivíduos de Z. sylvatica obtidas em uma única população do Nordeste do Brasil. Todos os indivíduos apresentaram núcleo interfásico reticulado e complemento cromossômico bimodal, pela ocorrência de um par cromossômico metacêntrico e os demais submetacêntricos. Foram observados três citotipos: 2n = 12 (um par metacêntrico, quatro submetacêntricos e um acrocêntricos) em 24 indivíduos; 2n = 12 + 1B, em cinco, além de um citotipo triplóide com 2n = 18 em três indivíduos. Todos os citotipos diplóides exibiram polimorfismo cromossômico para os pares um e dois, enquanto nos triplóides esse mesmo polimorfismo foi observado nas trincas correspondente aos pares cromossômicos diplóides, geralmente com dois cromossomos homórficos e um cromossomo heteromórfico maior que os dois anteriores. Esses dados confirmam a variabilidade cariológica anteriormente registrada para o gênero. São discutidos os mecanismos de evolução cariotípica envolvidos nessa variabilidade.
Zephyranthes Herb. is a taxonomically complex and cytologically variable group, with about 65 species of Neotropical distribution. Chromosome number variability in 32 individuals of a Zephyranthes sylvatica population from Northeast Brazil was investigated. Three cytotypes were found: 2n = 12 (one metacentric, four submetacentric and one acrocentric pairs), in 24 individuals; 2n = 12 + 1B, in five and three individuals with 2n = 18, a triploid cytotype. All diploid individuals showed chromosomes with polymorphism in pair one and two, while in triploids this polymorphism was observed in all chromosome triplets, generally with two homomorphic chromosomes and a higher or lower heteromorphic chromosome. All individuals had reticulated interfasic nucleus and a slightly asymmetric chromosome complement, with one metacentric chromosome pair and the others more submetacentric to acrocentric. These data confirm the cytological variability previously registered for the genus. Mechanisms involved in karyotypic evolution in this population are discussed.

Severe anemia is the earliest and a frequently fatal complication of Plasmodium falciparum infection. Here we describe Aotus infulatus as a primate model suitable to study this malaria complication. Both non-splenectomized and splenectomized monkeys receiving different inocula of P. falciparum FVO strain presented large (> 50%) decreases in hematocrit values during infection. Non-splenectomized animals were able to control parasite growth (parasitemia did not exceed 4%), but they had to be treated because of severe anemia. Three of 4 splenectomized monkeys did not control parasitemia and were treated, but developed severe anemia after treatment when presenting a negative blood film. Destruction of parasitized red blood cells alone cannot account for the degree of anemia. Non-splenectomized monkeys repeatedly infected with homologous parasites became rapidly and progressively resistant to reinfection and to the development of severe anemia. The data presented here point to A. infulatus as a suitable model for studying the pathogenesis of severe malarial infection.

Foram inoculados por via intradérmica, os coxins plantares traseiros de 64 camundongos Suíços, isogênicos. Os inóculos constituídos de uma suspensão fúngica, obtidos de biópsias de pacientes portadores de Doença de Jorge Lobo, foram submetidos a contagem do número total de fungos em câmara de Neubauer e a determinação da viabilidade segundo técnica do diacetato de fluoresceína-brometo de etídeo (DF-BE). Os animais foram sacrificados em períodos de tempo que variaram de 10 dias a 18 meses pós-inoculação. Um infiltrado celular constituído essencialmente por macrófagos com fungos no seu interior foi aumentando progressivamente até o final da experimentação, entretanto não houve alterações macroscópicas das patas inoculadas. Um pequeno número de células gigantes tipo Langhans começou a aparecer no infiltrado aos 9 meses. Um número considerável de fungos foi observado no período final do experimento, mas quando submetidos a coloração pelo DF-BE somente alguns deles estavam viáveis. Por esse motivo os autores sugerem que existe uma multiplicação de fungos, mas que são destruídos pelos macrófagos e permanecem no tecido um longo período de tempo talvez devido a dificuldade em sua eliminação. Estes achados levam a conclusão que apesar da manutenção da infecção nesses animais, os camundongos Suíços podem não ser considerados um modelo ideal para o estudo da Doença de Jorge Lobo. Entretanto, os autores chamam a atenção para a possibilidade de outras linhagens de camundongos serem mais susceptíveis ao P. loboi.
Sixty-four isogenic Swiss mice were intradermically inoculated in both hind foot pads. The inocula, consisting of fungal suspensions from biopsies obtained from Jorge Lobo’s Disease patients, had the total number of fungi and the viability index determined using a Neubauer chamber and the fluorescein diacetate-ethidium bromide technique (FD-EB), respectively. The animals were sacrificed at times ranging from ten days to eighteen months after inoculation. The cellular infiltrate, mainly consisting of macrophages containing fungi, increased progressively up to end of the study; however, no macroscopic alterations were observed in the inoculated feet. After nine months, small numbers of Langhans’ giant cells started to appear in the infiltrate. A considerable number of fungi was observed at the end of the experimental period, but only a few were viable when stained by the FD-EB technique. This fact suggests that there is a multiplication of fungal cells, which are destroyed by the macrophages but remain in the tissue for a long time due perhaps to the difficulties in their elimination. These findings led us to conclude that in spite of the maintenance of the infection in these animals, Swiss mice cannot be considered an ideal model to study Jorge Lobo’s Disease. However, the authors call attention to the possibility of other mouse strains being more susceptible to Paracoccidioides loboi.
