ABSTRACT This study aimed to verify the effect of the application of filter cake, via soil and in association with mineral fertilization, on sunflower cultivars (Helianthus annuus L.) and the chemical properties of the soil. The experimental design used was randomized blocks in a 3 × 2 factorial scheme with six replicates, totaling 36 experimental units. Three sunflower cultivars (Sany 66, Nusol 4140, and Nusol 4170) and two fertilization strategies (mineral fertilization with and without filter cake) were used. Resorting to the filter cake increased the oil concentration in Sany 66. Mineral fertilization did not significantly affect achene production, with mean values being 2561.58 kg ha-1. However, including filter cake resulted in a 21% increase in phosphorus (P) content in the 0-0.20 m soil layer in sunflower crops of the Nusol 4140 variety. Therefore, fertilization with filter cake, in combination with chemical fertilizers or alone, can be an effective strategy to increase soil P content and oil concentration in sunflower cultivation.
RESUMO Este estudo teve como objetivo verificar o efeito da aplicação de torta de filtro, via solo e em associação com a fertilização mineral, em cultivares de girassol (Helianthus annuus L.) e nas propriedades químicas do solo. O delineamento experimental utilizado foi em blocos casualizados, em esquema fatorial 3 × 2, com seis repetições, totalizando 36 unidades experimentais. Foram utilizadas três cultivares de girassol (Sany 66, Nusol 4140 e Nusol 4170) e duas estratégias de fertilização (fertilização mineral com e sem torta de filtro). O uso da torta de filtro aumentou a concentração de óleo na cultivar Sany 66. A fertilização mineral não afetou a produção de aquênios, sendo obtido valores médios de 2561,58 kg ha-1. No entanto, a inclusão da torta de filtro resultou em um aumento de 21% no teor de fósforo (P) na camada de solo de 0-0,20 m em cultivos de girassol da variedade Nusol 4140. Portanto, a fertilização com torta de filtro, em combinação com fertilizantes químicos ou isoladamente, pode ser uma estratégia eficaz para aumentar o teor de P no solo e a concentração de óleo no cultivo de girassol.

Abstract The TP53 3’UTR variant rs78378222 A>C has been detected in different tumor types as a somatic alteration that reduces p53 expression through modification of polyadenylation and miRNA regulation. Its prevalence is not yet known in all tumors. Herein, we examine tumor tissue prevalence of rs7837822 in Brazilian cohorts of patients from south and southeast regions diagnosed with lung adenocarcinoma (LUAD, n=586), sarcoma (SARC, n=188) and uterine leiomyoma (ULM, n=41). The minor allele (C) was identified in heterozygosity in 6/586 LUAD tumors (prevalence = 1.02 %) and none of the SARC and ULM samples. Additionally, next generation sequencing analysis revealed that all variant-positive tumors (n=4) with sample availability had additional pathogenic or likely pathogenic somatic variants in the TP53 coding regions. Among them, 3/4 (75 %) had the same pathogenic or likely pathogenic sequence variant (allele frequency <0.05 in tumor DNA) namely c.751A>C (p.Ile251Leu). Our results indicate a low somatic prevalence of rs78378222 in LUAD, ULM and SARC tumors from Brazilian patients, which suggests that no further analysis of this variant in the specific studied regions of Brazil is warranted. However, these findings should not exclude tumor molecular testing of this TP53 3’UTR functional variant for different populations.

RESUMO: Toxoplasma gondii, Neospora caninum e Sarcocystis spp. são parasitas detectados em tecidos de animais domésticos e selvagens. As aves são relevantes no ciclo de vida e epidemiologia dos protozoários devido à grande variedade de espécies de aves, hábitos alimentares e migratórios. O objetivo deste estudo foi a detecção molecular de T. gondii, N. caninum e Sarcocystis spp. em diversas espécies de aves naturalmente infectadas. Portanto, amostras de tecido de cérebro e coração foram coletados de aves recebidas e necropsiadas no Laboratório Central de Diagnóstico de Patologias Aviárias (LCDPA), sendo submetidas a extração de DNA e amplificação pela reação em cadeia da polimerase (PCR) do gene 18S rRNA para Sarcocystis spp., gene NC5 para N. caninum e gene repetitivo 529 pares de bases para T. gondii. N. caninum foi detectado em duas aves (02/65; 3,07%), em amostra de cérebro de Rupornis magnisrostris (número acesso: ON182081, 267pb) e em amostras de cérebro e coração de Dendrocygna bicolor (número acesso: ON211312, 267pb). DNA do genero Sarcocystis spp. foi detectado em três aves (03/65; 4,62%), sendo que no sequenciamento genético foram identificados Sarcocystis spp. (número acesso: MW463929) em cérebro de Nymphicus hollandicus e Sarcocystis speeri (número acesso: MW464125) em cérebro e coração de Amazona aestiva. A análise filogenética revelou que Sarcocystis spp. formou um clado com Sarcocystis spp. que utilizam gambá (Didelphis aurita) como hospedeiro definitivo e S. falcatula que utilizam Lóris-molucano (Trichoglossus moluccanus) como hospedeiro intermediário. S. speeri formou um clado com S. speeri que utilizou Mus musculus como hospedeiro intermediário experimental e formou um clado com Sarcocystis columbae, Sarcocystis corvusi, Sarcocystis halieti e Sarcocystis sp. que afetam espécies de aves. O DNA de T. gondii não foi detectado em nenhum tecido. Este é o primeiro relato de detecção de DNA de N. caninum, Sarcocystis spp. e S. speeri em amostras de tecido para essas espécies de aves estendendo a lista de hospedeiros intermediários.
ABSTRACT: Toxoplasma gondii, Neospora caninum and Sarcocystis spp. are parasites detected in tissues of domestic and wild animals. Birds are relevant in the life cycle and epidemiology of protozoa due to the wide variety of bird species, feeding and migratory habits. The aim of this study was the molecular detection of T. gondii, N. caninum and Sarcocystis spp. in several species of naturally infected birds. Therefore, samples of brain and heart tissue were collected from birds received and necropsied at the Central Laboratory for the Diagnosis of Avian Pathologies (LCDPA), undergoing DNA extraction and amplification by the polymerase chain reaction (PCR) of the 18S rRNA gene to Sarcocystis spp., NC5 gene for N. caninum and repetitive gene 529 base pairs for T. gondii. N. caninum was detected in two birds (02/65, 3.07%), in a brain sample of Rupornis magnisrostris (accession number: ON182081, 267pb) and in a brain and heart sample of Dendrocygna bicolor (accession number: ON211312, 267pb). DNA of the genus Sarcocystis was detected in three birds (03/65, 4.62%), and in the genetic sequencing Sarcocystis spp. (accession number: MW463929) in brain of Nymphicus hollandicus and Sarcocystis speeri (accession number: MW464125) in brain and heart of Amazona aestiva. Phylogenetic analysis revealed that Sarcocystis spp. formed a clade with Sarcocystis spp. that use skunk (Didelphis aurita) as definitive host and Sarcocystis falcatula that use Moluccan loris (Trichoglossus moluccanus) as intermediate host. S. speeri formed a clade with S. speeri that used Mus musculus as an experimental intermediate host and formed a clade with Sarcocystis columbae, Sarcocystis corvusi, Sarcocystis halieti and Sarcocystis sp. that affect bird species. T. gondii DNA was not detected in any tissue. This is the first report of DNA detection of N. caninum, Sarcocystis spp. and S. speeri in tissue samples for these bird species extending the list of intermediate hosts.

Abstract The present work aimed to evaluate the influence of different eucalyptus hybrids on the edaphic macrofauna community. It is hypothesized that the density and diversity of edaphic fauna are influenced by the quantity and quality of the litter deposited by the different hybrids on the soil surface. The study area is in the city of Vitória da Conquista (BA) and consists of seven eucalyptus hybrids, one from sexual reproduction (seminal) and six from asexual (clonal) reproduction. Sampling of the soil macrofauna was performed by the Tropical Soil Biology and Fertility method. All visible invertebrates were collected and identified at the level of large taxonomic groups. The relative density, group richness, and diversity index were calculated. Hybrids did not influence the total density and richness of the soil macrofauna. However, it was possible to observe changes in litter accumulation and composition, and as a result in the diversity indexes and in the presence or absence of macrofauna groups. Among the studied hybrids, those from Eucalyptus urophylla (seminal, 1335 and 1249) promote greater macrofauna diversity and favor the occurrence of detritivores organisms, suggesting the occurrence of better quality and more palatable litter for the edaphic fauna.

In this work, we report the synthesis of three series of carbohydrate-based benzo[f]indole-4,9-diones and amino-1,4-naphthoquinone derivatives and evaluated their cytotoxic activity against eight human cancer cell lines. Several compounds showed a promising cytotoxic activity toward the tumor cell lines (half maximal inhibitory concentration (IC50) < 10.0 μM). The importance of the substitution pattern of the quinone derivatives on the antitumor activity was also discussed. 3-Carboethoxy-2-methyl-benzo[f]indole-4,9-dione derivatives were more cytotoxic than their parent compounds and amino-1,4-naphthoquinones. Unlike clinically useful anticancer agent doxorubicin, the majority of synthesized compounds did not exhibit any lytic effects against erythrocytes or normal human leukocytes.

Novel isatinspirooxazine derivatives were designed and synthesized as potential anti-proliferative agents. The new compounds were obtained from aldol condensation reactions between isatin and 3-(hydroxyimino)butan-2-one in the presence of an organic base in order to generate an aldol adduct, followed by cyclization in trifluoroacetic acid, providing the desired isatinspirooxazines in 30 to 80% yield. All the synthesized compounds, including the starting material and the synthetic intermediates, were tested for in vitro anti-proliferative activity against cell lines MCF-7 and MDA-MB231 (breast cancer) and A549 (lung cancer), highlighting the compound 4-methyl,5'-methyl-spiro[(5-aza-4-eno-3-one-cyclohexane)-1,3'-(1H-indol-one)] with an IC50 (half maximal inhibitory concentration) = 0.34 µM, more potent than the reference drug, doxorubicin (IC50 = 1.88 µM), in breast cancer line MDA-MB231.

A method based on QuEChERS (quick, easy, cheap, effective, rugged, and safe) extraction and gas chromatography-mass spectrometry (GC-MS) detection was described for the pesticides dichlorvos, disulfoton, ethoprophos, parathion methyl, fenchlorphos, chlorpyrifos, azinphos methyl and prothiofos in sapodilla. For all compounds studies, it was observed a strong matrix effect showing the need to use matrix matched calibration strategy. Method was validated, and good linearity (R > 0.99) was obtained for all pesticides studied with limits of detection (LODs) and quantification (LOQs) ranging from 0.01 to 0.06 mg kg-1 and 0.03 to 0.2 mg kg-1, respectively. Recovery studies were performed at different levels (0.08, 0.10, 0.14, 0.20, 0.35 and 1.17 mg kg-1) and showed good results (between 70 and 120% with relative standard deviation (RSD) < 20%). A statistical test was applied to the coefficients of the analytical curves obtained in the sapodilla matrix. Analyses of commercial samples showed chlorpyrifos were detected in about 70 and 33% for fruit and pulps samples, respectively. It should be noted that chlorpyrifos is not permitted in sapodilla crops by ANVISA and EC guidelines.

Through dereplication strategies using gas chromatography-mass spectrometry (GC-MS) and ultra fast liquid chromatography-tandem mass spectrometry (UFLC-MS/MS), the ethanol extract from Psycotria nemorosa leaves (Rubiaceae) showed to be composed of: cinnamic acid, dihydroactinidiolide, 4-hydroxy-β-ionone, phytol, isophytol, 4,8,12,16-tetramethylheptadecan-4-olide, lupeol, a mixture of α/β-amyrin, the keto and acetylated derivatives, besides stigmast-4-en-3-one, campesterol, stigmasterol and γ-sitosterol by GC-MS. Likewise, by UFLC-MS/MS, the main compounds identified were: butin, resveratrol, rutin, kaempferol 7-O-β-D-glucopyranoside, deacetylasperuloside, epiloganin, hordenine, strictosidine, N-methyl-1,2,3,4-tetrahydro-β-carboline and N-formyl-tryptamine. The antinociceptive activity of the crude extract ant its fractions was reported.

Resumo A osteoporose é a principal causa de fraturas na população acima de 50 anos. É uma doença silenciosa que afeta especialmente as mulheres na pós-menopausa e idosos e tem elevada taxa de morbimortalidade. O principal objetivo do tratamento da osteoporose é a prevenção das fraturas. A identificação dessa população de risco através do diagnóstico e tratamento precoces é de fundamental importância. A última diretriz brasileira para tratamento da osteoporose em mulheres na pós-menopausa foi elaborada em 2002. Desde então foram desenvolvidas novas estratégias de diagnóstico da osteoporose, bem como fármacos com novos mecanismos de ação foram adicionados ao arsenal terapêutico. A Comissão de Osteoporose e Doenças Osteometabólicas da Sociedade Brasileira de Reumatologia em conjunto com a Associação Médica Brasileira e sociedades afins desenvolveu esta atualização da diretriz do tratamento da osteoporose em mulheres na pós-menopausa de acordo com as melhores evidências científicas disponíveis. Esta atualização é destinada aos profissionais das várias especialidades médicas e da área da saúde envolvidos no tratamento da osteoporose, médicos em geral e organizações relacionadas à saúde.
Abstract Osteoporosis is the leading cause of fractures in the population older than 50 years. This silent disease affects primarily postmenopausal women and the elderly, and the morbidity and mortality rates are high. The main goal of treating osteoporosis is the prevention of fractures. The identification of populations at risk through early diagnosis and treatment is essential. The last Brazilian guideline for the treatment of postmenopausal osteoporosis was elaborated in 2002. Since then, new strategies for diagnosis and risk stratification have been developed, and drugs with novel action mechanisms have been added to the therapeutic arsenal. The Osteoporosis and Osteometabolic Diseases Committee of the Brazilian Society of Rheumatology, in conjunction with the Brazilian Medical Association and other Societies, has developed this update of the guidelines for the treatment of postmenopausal osteoporosis according to the best scientific evidence available. This update is intended for professionals in many medical and health specialties involved in the treatment of osteoporosis, for physicians in general and for health-related organizations.

The objective of this study was to characterize the clonal profile, virulence factors and antimicrobial resistance, particularly oxacillin resistance, of Staphylococcus aureus isolated from sheep milk. Milk samples were collected from all teats for the California Mastitis Test (CMT), somatic cell count, identification of S. aureus, investigation in these strains of genes encoding toxins (sea, seb, sec, sed, tst), biofilm (icaA, icaC, icaD, bap), leukocidin (luk-PV) oxacillin resistance by mecA gene detection and susceptibility testing (12 antibiotics). Messenger RNA expression was evaluated by RT-PCR in isolates carrying toxin and biofilm genes. Biofilm formation was also evaluated phenotypically by adherence to polystyrene plates. The clonal profile of S. aureus was investigated by pulsed-field gel electrophoresis. A total of 473 milk samples were collected from 242 animals on three farms and 20 S. aureus strains were isolated and none carried the mecA gene. The two sec gene-positive isolates and the isolates carrying the tst and luk-PV genes were positive by RT-PCR. Staphylococcus aureus isolated from the three flocks studied showed high susceptibility to the drugs tested and none was biofilm producer, indicating that biofilm formation was not a virulence factor causing infection by these strains. The typing of 17 S. aureus isolates revealed the presence of a common clone on the three farms studied, and the presence and expression of the sec and tst genes in one strain of this clone suggest the possible acquisition of virulence genes by this clone, a fact that is important for animal health and food hygiene.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Bacterial resistance to commonly used antibiotics has been recognized as a significant global health issue. In this study, we carried out the screening of a family of allylic thiocyanates for their action against a diversity of bacteria and fungi with a view to developing new antimicrobial agents. Allylic thiocyanates bearing halogenated aryl groups, which were readily obtained in two steps from the Morita-Baylis-Hillman adducts, showed moderate-to-high activity against selective pathogens, including a methicillin-resistant S. aureus (MRSA) strain. In particular cases, methyl (Z)-3-(2,4-dichlorophenyl)-2-(thiocyanomethyl)-2-propenoate exhibited antimicrobial activity comparable to the reference antibiotic Imipenem.

OBJECTIVES: to evaluate the antimicrobial, cytotoxic and healing activities of the ethanolic extract of the stems of Z. tuberculosa via topical use and/or oral ingestion. METHOD: antimicrobial assays in vitro using the disk diffusion method, the Artemia salina toxicity test, and in vivo assays with Wistar rats. From these was collected clinical, histological and biochemical data for evaluating the healing process. RESULTS: in vitro antimicrobial testing showed activity in relation to Streptococcus pyogenes, Staphylococcus aureus and Staphylococcus epidermidis, with zones of inhibition of 18, 14 and 10 mm, respectively. The best minimum inhibitory concentration was 62.5 µg/ml for S. aureus, this bacteria being chosen for the in vitro assays. Animals treated with the ointments with the extract of Z. tuberculosa showed the best results in the reduction of the wound diameter, data confirmed by the presence of re-epithelialization in the histological samples. CONCLUSION: the extract was shown to be promising for the continuation of studies which may identify the active ingredients responsible for the pharmacological activity and its mechanism of action in the process of wound healing, so as to develop a product which may be used as an alternate means in the repair of infected cutaneous wounds.

En estos fueron recolectados datos clínicos, histológicos y bioquímicos para evaluación del proceso de cicatrización. RESULTADOS: los ensayos antimicrobianos in vitro mostraron actividad frente a la Streptococcus pyogenes, Staphylococcus aureus y Staphylococcus epidermidis, con halos de inhibición de 18, 14 y 10 mm, respectivamente. La mejor concentración inhibitoria mínima fue 62,5 µg/mL para S. aureus, siendo esta bacteria escogida para los ensayos in vivo. Animales tratados con las pomadas del extracto de la Z. tuberculosa presentaron mejores resultados en la reducción del diámetro de la herida, dato confirmado por la presencia de reepitelización en los cortes histológicos. CONCLUSIÓN: el extracto se mostró promisor para la continuación de estudios que identifiquen los principios activos responsables por la actividad farmacológica y su mecanismo de acción en el proceso de cicatrización de heridas, con la finalidad de desarrollar un producto que pueda ser utilizado de forma alternativa en la reparación de heridas cutáneas infectadas.

From these was collected clinical, histological and biochemical data for evaluating the healing process. RESULTS: in vitro antimicrobial testing showed activity in relation to Streptococcus pyogenes, Staphylococcus aureus and Staphylococcus epidermidis, with zones of inhibition of 18, 14 and 10 mm, respectively. The best minimum inhibitory concentration was 62.5 µg/ml for S. aureus, this bacteria being chosen for the in vitro assays. Animals treated with the ointments with the extract of Z. tuberculosa showed the best results in the reduction of the wound diameter, data confirmed by the presence of re-epithelialization in the histological samples. CONCLUSION: the extract was shown to be promising for the continuation of studies which may identify the active ingredients responsible for the pharmacological activity and its mechanism of action in the process of wound healing, so as to develop a product which may be used as an alternate means in the repair of infected cutaneous wounds.
OBJETIVOS: evaluar las actividades antimicrobianas, citotóxicas y cicatrizantes del extracto etanólico del tallo de la Z. tuberculosa por vía tópica y/o ingestión oral. MÉTODO: ensayos antimicrobianos in vitro por el método de difusión en disco, prueba de toxicidad de la Artemia salina y ensayos in vivo con ratones Wistar. En estos fueron recolectados datos clínicos, histológicos y bioquímicos para evaluación del proceso de cicatrización. RESULTADOS: los ensayos antimicrobianos in vitro mostraron actividad frente a la Streptococcus pyogenes, Staphylococcus aureus y Staphylococcus epidermidis, con halos de inhibición de 18, 14 y 10 mm, respectivamente. La mejor concentración inhibitoria mínima fue 62,5 µg/mL para S. aureus, siendo esta bacteria escogida para los ensayos in vivo. Animales tratados con las pomadas del extracto de la Z. tuberculosa presentaron mejores resultados en la reducción del diámetro de la herida, dato confirmado por la presencia de reepitelización en los cortes histológicos. CONCLUSIÓN: el extracto se mostró promisor para la continuación de estudios que identifiquen los principios activos responsables por la actividad farmacológica y su mecanismo de acción en el proceso de cicatrización de heridas, con la finalidad de desarrollar un producto que pueda ser utilizado de forma alternativa en la reparación de heridas cutáneas infectadas.
